Description
Technical Field
The invention discloses a composition, in particular to an anti-aging composition, a preparation method and application thereof, belonging to the technical field of cosmetics.
Background
The skin is the outermost tissue of the human body and plays a role in protecting the human body from external injury and resisting various external mechanical, chemical, physical and biological stimuli. Human skin aging is classified into endogenous aging, which is an aging process occurring with the increase of age under the influence of genetic factors, and exogenous aging, which is aging caused by the influence of the environment on the skin. The main factor of environmental impact is the damage of free radicals, which are abundant in active oxygen free radicals in our living environment. The damage of free radicals to human skin is mainly caused by lipid peroxidation, protein peroxidation and DNA damage, which continuously attack and destroy the outer cellular tissues of the human body through strong oxidative reactions. Therefore, signal transduction of human cells is disturbed, apoptosis is accelerated, skin aging is accelerated, and a series of chronic skin health problems such as dry skin, laxity, desquamation, melanin increase, and even skin spots are caused, which afflict many consumers.
Scavengers of free radicals, known as antioxidants, achieve antioxidant action by inhibiting or terminating the oxygen radical chain reaction. In recent years, with the increasing demands of people on cosmetics, such as safety, high efficiency, no stimulation and the like, the natural antioxidant of plant source is increasingly emphasized due to the characteristics of wide source, safety, effectiveness, natural, no toxicity and the like. Natural antioxidants are of various types and generally include superoxide dismutase, catalase, glutathione, carnosine, vitamin C, vitamin E, plant polyphenols, flavonoids, carotenoids, etc.
At present, many anti-aging and anti-wrinkle products on the market supplement collagen and elastin in the skin, or use a single antioxidant to reduce the generation of free radicals to achieve the anti-aging purpose. However, most of substances contained in the composition are macromolecules which are difficult to absorb by skin, and in addition, antioxidants are often irritant to a certain extent, and the risk of easy inactivation exists, so that the antioxidant and anti-aging effects are not ideal.
In summary, the antioxidant and anti-aging products which can effectively remove free radicals and promote the production of collagen and elastin are developed through the combination of plant extracts at present, and the antioxidant and anti-aging products have wide market application prospects.

And (3) testing 2: oxidation resistance test
This test was conducted to examine the radical scavenging ability of the skin care compositions of examples 1 to 5 and comparative examples 1 to 3 prepared by the sample preparation example.
The test method comprises the following steps:
1) preparing 0.1mmol/L DPPH solution with anhydrous ethanol (1mg DPPH dissolved in 24ml anhydrous ethanol), performing ultrasonic treatment for 5min, shaking thoroughly, mixing well, storing in dark, and using up within 5 h.
2) Adding 2mL of test sample solution (the sample is dissolved by absolute ethyl alcohol) and 2mL of LDPPH solution into the same test tube, shaking uniformly, standing at room temperature in the dark for 30min, and then measuring absorbance Asample, and simultaneously measuring absorbance A0 after the 2mL of LDPPH solution and 2mL of solvent are mixed.
3) Representation of radical scavenging capacity: percent DPPH clearance (a0-Asample)/a0 x 100. A0 is the absorbance at 520nm of DPPH without sample; asample is the absorbance at 520nm of DPPH added to the sample. The test results are shown in Table 4.

A proper amount of samples are taken and applied on the face, the samples are used for 1 time in the morning and at night every day, products or preparations with the same efficacy types are forbidden to be used on the tested parts, the test period is 0 day, 14 days and 28 days, before the test, a subject firstly cleans the face uniformly, the rest is carried out for 30min in a constant temperature and humidity room, and the data acquisition of the face is carried out by using various devices.
Water content of skin stratum corneum: the moisture content of the horny layer of the face skin is tested by a skin moisture content test probe Corneometer (Courage & Khazaka, Germany), the numerical unit is a.u., the moisture content is measured for 3 times, and the average value is taken, wherein the higher the value is, the better the value is, the moisture content of the horny layer of the face skin is represented to be high, and the moisture content test probe is mainly used for judging the moisturizing effect of the product on the skin.
Skin wrinkle length: images before and after use are photographed by an Antera3D facial image analyzer Miravex Limited (Ireland), wrinkle parameters are analyzed through image comparison, a wrinkle length value is obtained, a numerical unit is mm, the smaller the wrinkle parameter is, the lighter the wrinkle is, and the wrinkle effect of the product on skin is mainly judged.
Skin firmness: the skin compactness is measured through a skin compactness inspection probe cutometer (Courage & Khazaka, Germany), 10 cycles are measured each time, an average value is taken, the compactness F4 parameter represents the resilience speed of the skin, the faster the better the resilience speed is, the smaller the numerical value is, the better the resilience speed is, the more compact the skin is, and the skin compactness is mainly used for judging the skin compactness effect of the product.
Skin glossiness: skin gloss was measured by a skin gloss test probe GlossimeterGL 200(Courage & Khazaka, Germany) in a.u. for 3 measurements, with an average value indicating that the higher the number the better the skin is, the glossier the skin, and was used primarily to evaluate the effect of the product on improving skin gloss.
The specific statistical results of the test parameters are shown in table 5, the statistical analysis software of the data is SPSS, the data improvement value normal distribution significance test is carried out by adopting Shapiro-WilkTest, and if Sig (on both sides) > 0.01, the data are normally distributed, and the pairing t test is carried out. (bilateral) < 0.01, the distribution is abnormal, and Wilcoxon test is performed.
TABLE 5 descriptive statistical results table for each test index
After 28 days of use compared to before use: the water content of the skin stratum corneum is increased by 30.69%, and the significance is realized, so that the test sample has the effect of improving the water content of the skin; the skin wrinkle length value is reduced by 43.59%, and the significance is realized, so that the test sample has the wrinkle lightening effect; 50.05% reduction in skin firmness F4, with significance, suggests that the test sample has an effect of improving skin firmness; the skin glossiness increased by 7.69%, and was significant, suggesting that the test sample had the effect of improving skin glossiness.
In conclusion, the skin care product containing the anti-aging composition has the advantages that the skin is improved in all aspects, the moisture content of the stratum corneum of the skin is increased, the wrinkle length is reduced, the firmness F4 is reduced, the skin glossiness is improved, and the anti-aging effect on the skin is obvious. The results are schematically shown in FIG. 1.
