Abstract
本发明公开了一种低致敏牛乳水解蛋白婴幼儿食品,涉及婴幼儿食品技术领域,该食品包括以下重量份原料:脱敏牛乳粉21‑27份、酶解米粉8.5‑12.7份、微量元素粉5.1‑7.3份、植物油4.1‑4.7份、低聚半乳糖1.6‑2.2份,将上述原料混合制得该种低致敏牛乳水解蛋白婴幼儿食品,本发明的脱敏牛乳粉的水解度高于现有技术中的部分水解奶粉产品,通过使用胰蛋白酶和复合风味蛋白酶复合使用、合理的酶解参数设计,降低水解苦味,同时酶解装置加酶均匀,避免局部酶浓度过高,换热效率高,避免局部高温,从而避免局部水解度过高导致大量苦味肽生成;本婴幼儿食品采用酶解米粉为主要原料,口感细腻、易于吸收,深受婴幼儿喜爱。

技术领域
本发明属于婴幼儿食品技术领域,具体地,涉及一种低致敏牛乳水解蛋白婴幼儿食品及其制备方法。
背景技术
婴儿阶段除了母乳和婴儿配方奶粉之外,还应给予宝宝一些补充营养的辅助食品,在婴儿阶段,母乳是宝宝最理想的食品,但随着宝宝一天天长大,大约四个月开始光吃母乳或者婴儿配方奶粉已经无法满足宝宝的营养需求。所以这段时间,除了原先母乳或婴儿配方奶粉之外,还应该给宝宝添加易消化的食物,婴儿米粉是一种常见的婴儿食品。
中国专利CN106666713B提出一种婴幼儿辅食及其制备方法,该专利中以山药、小米、莲藕、糙米等为原料制成浆料,再干燥制成粉料食品,以达到营养均衡、食用方便的目的,但是该配方中淀粉含量高,婴幼儿的消化能力不强,难以消化吸收,另外,婴幼儿习惯了母乳的味道,难以接受该类食品,为此,本发明提出在婴儿食品中添加牛乳粉。牛乳中含有大量αs-酪蛋白和β-乳球蛋白,这两种蛋白质以未被消化的形式进入人体,就会引起过敏反应,婴幼儿的消化能力不强,直接食用会引起湿疹、过敏性鼻炎等症状,现有技术中,对牛乳进行酶解,将大分子的αs-酪蛋白和β-乳球蛋白分解为小分子或者多肽,使其致敏性丧失,但是蛋白质的水解物中含有疏水性多肽,其与人的味蕾接触会产生苦味,随着水解程度的加深,越来越多的疏水多肽暴露出来使苦味加剧。中国专利CN104757138A公开一种以乳清浓缩蛋白和益生元(低聚果糖,低聚半乳糖,低聚果糖和低聚半乳糖的混合物)为壁材,以蛋白酶水解乳清浓缩蛋白获得的乳清蛋白水解物为芯材,制备微胶囊化乳清蛋白水解物,解决蛋白水解物苦味严重问题,该种方法制备工艺复杂,且制备条件难以控制。本申请人经长期试验工作发现,通过控制牛乳水解参数,可以有效减少苦味产生,本发明基于上述问题,提供一种适用于均匀水解的酶解装置,且提供一种低致敏牛乳水解蛋白婴幼儿食品及其制备方法。
发明内容
本发明的目的在于提供一种低致敏牛乳水解蛋白婴幼儿食品及其制备方法。
本发明要解决的技术问题:酶解牛乳产生苦味和现有婴幼儿食品不易吸收问题。
本发明的目的可以通过以下技术方案实现:
一种低致敏牛乳水解蛋白婴幼儿食品,包括以下重量份原料:脱敏牛乳粉21-27份、酶解米粉8.5-12.7份、微量元素粉5.1-7.3份植物油4.1-4.7份、低聚半乳糖1.6-2.2份;
所述脱敏牛乳粉,由以下步骤制备:
步骤A1:取鲜牛乳至脱脂机中,升温至55℃,保温12-15min,脱脂处理至牛乳中含脂率低于0.12%,再升温至82℃,保持10-15s,升温时间小于8-11min,对牛乳进行杀菌,再降温至30-35℃,降温时间小于5-7min,制得脱脂牛乳;
步骤A2:将脱脂牛乳泵入酶解装置中进行酶解,首先将脱脂牛乳泵入酶解釜内,水解蛋白酶分散于无菌水中得到混合液,再将混合液泵入投料箱内,混合液从传动轴管底部的通孔进入脱脂牛乳中,电热丝对脱脂牛乳升温至40-45℃,电机驱动搅拌叶对脱脂牛乳进行搅拌换热,酶解35-42min,再升温至85-93℃,保温2.5-3.3min灭酶处理后,立即从进水管通入冷却水,降温至30℃,制得酶解牛乳;
步骤A3:将牛磺酸、DHA藻油、乳化剂、无菌水投入乳化机中,在转速为1500-1700r/min条件下,乳化反应7-10min,制得乳化液,再将乳化液通过投料箱加入酶解牛乳中,再加入植物油和低聚半乳糖,在转速为220-310r/min转下,搅拌10min,制得复合牛乳;
步骤A4:将复合牛乳泵入真空浓缩机中,浓缩至复合牛乳浓度为51-53%,再将复合牛乳泵入喷雾干燥机,喷雾干燥制得脱敏牛乳粉。
进一步地,所述微量元素粉包括:蛋黄粉、猪肝粉、胡萝卜粉、核桃粉、豌豆粉。
进一步地,步骤A2中所述水解蛋白酶,包括胰蛋白酶、复合风味蛋白酶,胰蛋白酶、复合风味蛋白酶的用量质量比为1.9:0.85,水解蛋白酶、脱脂牛乳的用量比为1.15-1.27g:1000g。
进一步地,步骤A3中,乳化剂为大豆卵磷脂,且酶解牛乳、牛磺酸、DHA藻油、乳化剂、无菌水用量比为1000mL:0.21g:1.7mL:0.25mL:10mL。
进一步地,一种低致敏牛乳水解蛋白婴幼儿食品的制备方法:称取酶解米粉、脱敏牛乳粉、微量元素粉混合。
进一步地,该种低致敏牛乳水解蛋白婴幼儿食品中添加有营养强化剂,营养强化剂为维生素A、维生素D、维生素E、维生素K、维生素B1、维生素B2、维生素B6、维生素B12、烟酸、叶酸、泛酸、维生素C、胆碱、钙、铁、锌、镁、锰、硒、铜、碘和钾中的一种或多种。
进一步地,步骤A2中所述酶解装置,包括酶解釜,酶解釜的底部中心设置有电机,酶解釜的顶部中心设置有投料箱,且电机和投料箱均通过密封轴承座与酶解釜固定连接,酶解釜的中部设置有传动轴管,传动轴管的底端与电机的输出端固定连接,传动轴管的顶端穿过密封轴承座的轴套与投料箱内部连通,传动轴管的底部周侧壁上开设有若干通孔,传动轴管的周侧壁上固定有搅拌叶,酶解釜的内侧壁和底壁上固定有换热板,换热板靠近传动轴管的侧壁上卡装有冷却管,冷却管的一端连通有出水管,冷却管的另一端连通有进水管,且出水管和进水管均贯穿酶解釜的侧壁与外界连通,酶解釜的侧壁和底壁均设置有夹层,且夹层内部安装有电热丝,酶解釜底部的一侧设置有出料管,酶解釜底部的另一侧设置有温度计。
进一步地,所述搅拌叶为螺旋状叶片。
进一步地,所述换热板靠近传动轴管的侧壁上纵向开设有若干换热槽。
本发明的有益效果:
1、本低致敏牛乳水解蛋白婴幼儿食品,添加有酶解米粉,米粉先经淀粉酶酶解,弥补婴幼儿肠胃淀粉酶少的问题,使得本食品易于被吸收;添加脱敏牛乳粉,将鲜牛乳中的αs-酪蛋白和β-乳球蛋白分解为小分子或者多肽,使其致敏性丧失,降低婴幼儿食用过敏风险,此外在脱敏牛乳粉的制备过程中添加DHA藻油和牛磺酸,有助于婴幼儿的智力发育,提升婴幼儿的免疫力,并且以乳化液的方式加入,使其加入更加均匀;添加微量元素粉,其富含维生素和膳食纤维,有利于营养均衡,促进肠道蠕动。
2、本发明提供的脱敏牛乳粉,采用胰蛋白酶对牛乳蛋白进行酶解,可有效对αs-酪蛋白和β-乳球蛋白水解,本申请人对脱敏牛乳粉进行测定,其牛乳蛋白水解度为5.7%,αs-酪蛋白和β-乳球蛋白的分子量在3000-4000Da之间,致敏性几乎丧失,复合风味蛋白酶的加入,进一步从多肽链的末端切断释放一个氨基酸,从而把苦肽彻底降解为氨基酸,减轻脱敏牛乳粉的苦味。
3、本发明提供牛乳蛋白酶解装置,水解蛋白酶分散于无菌水中,从投料箱顶部泵入,混有水解蛋白酶的混合液从传动轴管底部的通孔进入待酶解牛乳中,同时,电机驱动螺旋状的搅拌叶,使得牛乳自下而上被搅动,使得水解蛋白酶均匀混入牛乳中,避免局部酶浓度过高,导致水解过度,进而生产大量的苦味肽;酶解釜的内壁上固定有换热板,在换热板与牛乳的接触面上开设纵向的换热槽,增加换热面积,提升换热效率,便于控制酶解温度与酶解时间,另一方面,防止升温时靠近加热端形成局部高温,导致水解过度;换热板外侧设计冷却管,高温灭菌后,及时进行强制降温,避免蛋白质变性,导致营养成分降低。

请参阅图1-2所示,本发明提供脱敏牛乳粉制备用酶解装置,该酶解装置包括酶解釜10,酶解釜10的底部中心设置有电机15,酶解釜10的顶部中心设置有投料箱11,且电机15和投料箱11均通过密封轴承座与酶解釜10固定连接,酶解釜10顶部的一侧设置有进料管12,酶解釜10的中部设置有传动轴管17,传动轴管17的底端与电机15的输出端固定连接,传动轴管17的顶端穿过密封轴承座的轴套与投料箱11的内部连通,传动轴管17的底部周侧壁上开设有若干通孔,传动轴管17的周侧壁上固定有搅拌叶18,且搅拌叶18呈螺旋状叶片结构,酶解釜10的内侧壁和底壁上固定有换热板20,换热板20靠近传动轴管17的侧壁上纵向开设有若干换热槽,换热板20开设换热槽的侧壁上卡装有冷却管19,冷却管19的一端连通有出水管13,冷却管19的另一端连通有进水管14,且出水管13和进水管14均贯穿酶解釜10的侧壁与外界连通,酶解釜10的侧壁和底壁均设置有夹层,且夹层内部安装有电热丝22,酶解釜10底部的一侧设置有出料管16,酶解釜10底部的另一侧设置有温度计21。
该酶解装置的使用方法如下:
将脱脂牛乳从进料管12泵入酶解釜10中,水解蛋白酶分散于无菌水中泵入投料箱11,混有水解蛋白酶的混合液经传动轴管17底部的通孔进入脱脂牛乳中,同时启动电机15,通过传动轴管17带动搅拌叶18转动,螺旋状的搅拌叶18使得脱脂牛乳从下往上翻动,将水解蛋白酶与脱脂牛乳均匀混合,避免水解蛋白酶局部浓度偏高,使得该区域酶解度过高,导致大量苦味肽产生;酶解时,启动电热丝22,对酶解釜10内部进行升温,换热板20采用铜材制成,迅速导热,酶解釜10侧壁的脱脂牛乳在搅拌叶18的作用下,从上向下流动循环,与换热槽接触时,快速换热,避免酶解釜10内壁侧形成局部高温,使得该区域酶活性较高,导致过度水解产生苦味肽;灭酶时,提升电热丝22加热功率,提升电机15转速,对酶解后的牛乳快速加热灭酶,灭酶完成后,立即从进水管14通入冷却水,持续搅拌对灭酶后的牛乳进行降温,避免蛋白质长时间处于高温导致变性,减少牛乳营养成分失效,冷却后的牛乳从出料管16排出,即可脱脂牛乳的酶解。
实施例1
脱敏牛乳粉,由以下步骤制备:

步骤A3:将牛磺酸、DHA藻油、大豆卵磷脂、无菌水投入乳化机中,在转速为1500r/min条件下,乳化反应7min,制得乳化液,再将乳化液通过投料箱11加入酶解牛乳中,其中,酶解牛乳、牛磺酸、DHA藻油、大豆卵磷脂、无菌水用量比为1000mL:0.21g:1.7mL:0.25mL:10mL,再加入植物油和低聚半乳糖,在转速为220r/min转下,搅拌10min,制得复合牛乳;
步骤A4:将复合牛乳泵入真空浓缩机中,浓缩至复合牛乳浓度为51%,再将复合牛乳泵入喷雾干燥机,控制干燥温度为155℃,制得脱敏牛乳粉。
实施例2
脱敏牛乳粉,由以下步骤制备:
步骤A1:取鲜牛乳至脱脂机中,升温至55℃,保温13min,脱脂处理至牛乳中含脂率低于0.12%,再升温至82℃,保持12s,设置升温时间为10min,对牛乳进行杀菌,使用冷却装置降温至33℃,设置降温时间为6min,制得脱脂牛乳;
步骤A2:将脱脂牛乳泵入酶解装置中进行酶解,首先将脱脂牛乳泵入酶解釜10内,取胰蛋白酶和复合风味蛋白酶,按照用量质量比为1.9:0.85,制得水解蛋白酶,再按照水解蛋白酶、脱脂牛乳的用量比为1.21g:1000g,取水解蛋白酶分散于无菌水中震荡3min得到混合液,再将混合液泵入投料箱11内,混合液从传动轴管17底部的通孔进入脱脂牛乳中,电热丝22对脱脂牛乳升温至42℃,电机15驱动搅拌叶18对脱脂牛乳进行搅拌换热,酶解38min,再升温至90℃,保温3min灭酶处理,立即从进水管通入冷却水,降温至30℃,制得酶解牛乳;
步骤A3:将牛磺酸、DHA藻油、大豆卵磷脂、无菌水投入乳化机中,在转速为1600r/min条件下,乳化反应8min,制得乳化液,再将乳化液通过投料箱11加入酶解牛乳中,其中,酶解牛乳、牛磺酸、DHA藻油、大豆卵磷脂、无菌水用量比为1000mL:0.21g:1.7mL:0.25mL:10mL,再加入植物油和低聚半乳糖,在转速为280r/min转下,搅拌10min,制得复合牛乳;
步骤A4:将复合牛乳泵入真空浓缩机中,浓缩至复合牛乳浓度为51%,再将复合牛乳泵入喷雾干燥机,控制干燥温度为155℃,制得脱敏牛乳粉。
实施例3
脱敏牛乳粉,由以下步骤制备:
步骤A1:取鲜牛乳至脱脂机中,升温至55℃,保温15min,脱脂处理至牛乳中含脂率低于0.12%,再升温至82℃,保持15s,设置升温时间为11min,对牛乳进行杀菌,使用冷却装置降温至35℃,设置降温时间为7min,制得脱脂牛乳;
步骤A2:将脱脂牛乳泵入酶解装置中进行酶解,首先将脱脂牛乳泵入酶解釜10内,取胰蛋白酶和复合风味蛋白酶,按照用量质量比为1.9:0.85,制得水解蛋白酶,再按照水解蛋白酶、脱脂牛乳的用量比为1.27g:1000g,取水解蛋白酶分散于无菌水中,在15KHz频率下超声分散3min得到混合液,再将混合液泵入投料箱11内,混合液从传动轴管17底部的通孔进入脱脂牛乳中,电热丝22对脱脂牛乳升温至45℃,电机15驱动搅拌叶18对脱脂牛乳进行搅拌换热,酶解42min,再升温至93℃,保温3min灭酶处理,立即从进水管通入冷却水,降温至30℃,制得酶解牛乳;
步骤A3:将牛磺酸、DHA藻油、大豆卵磷脂、无菌水投入乳化机中,在转速为1700r/min条件下,乳化反应10min,制得乳化液,再将乳化液通过投料箱11加入酶解牛乳中,其中,酶解牛乳、牛磺酸、DHA藻油、大豆卵磷脂、无菌水用量比为1000mL:0.21g:1.7mL:0.25mL:10mL,再加入植物油和低聚半乳糖,在转速为310r/min转下,搅拌10min,制得复合牛乳;
步骤A4:将复合牛乳泵入真空浓缩机中,浓缩至复合牛乳浓度为53%,再将复合牛乳泵入喷雾干燥机,控制干燥温度为155℃,制得脱敏牛乳粉。
对比例1
本对比例为市售脱脂水解奶粉(雀巢-超启能恩)
取实施例1-3和对比例1,进行氨基酸含量测定,取实施例1-3和对比例1配置成浓度为15%的乳液,邀请100名育儿师对乳液的苦味接受度进行评分(评分范围为0-100分,100分即为完全接受),取平均分。具体测试如表1;

由表1可知,实施例1-3中的氨基酸的含量高于对比例1,可知实施例1-3制得的脱敏牛乳粉的水解度高于对比例1,大分子的致敏蛋白被酶解的更加充分,使得本发明制得的脱敏牛乳粉的致敏性更低,复合风味蛋白酶的加入,进一步从多肽链的末端切断释放一个氨基酸,从而把苦肽彻底降解为氨基酸,减轻脱敏牛乳粉的苦味,加入的牛磺酸有掩盖苦味的作用,酶解装置使得酶加入均匀,避免形成局部高温,使得蛋白质的水解状态可控性高,减少局部水解度过高生产大量苦味肽,由表1可知,本发明制得的脱敏牛乳粉的苦味接受度高。
实施例4
一种低致敏牛乳水解蛋白婴幼儿食品,包括以下重量份原料:脱敏牛乳粉21份、酶解米粉8.5份、微量元素粉5.1份,按上述重量份称取原料混合制得低致敏牛乳水解蛋白婴幼儿食品;
所述脱敏牛乳粉采用实施例1制得的脱敏牛乳粉,其中植物油的重量份为4.1份,低聚半乳糖的重量份为1.6份;
所述微量元素粉包括蛋黄粉、猪肝粉、胡萝卜粉、核桃粉、豌豆粉,且其质量分数分别为22.5wt%、18.5wt%、21.3wt%、23wt%、14.7wt%。
实施例5
一种低致敏牛乳水解蛋白婴幼儿食品,包括以下重量份原料:脱敏牛乳粉25份、酶解米粉10.4份、微量元素粉6.2份,按上述重量份称取原料混合制得低致敏牛乳水解蛋白婴幼儿食品;
所述脱敏牛乳粉采用实施例2制得的脱敏牛乳粉,其中植物油的重量份为4.5份,低聚半乳糖的重量份为1.9份;
所述微量元素粉包括蛋黄粉、猪肝粉、胡萝卜粉、核桃粉、豌豆粉,且其质量分数分别为27.4wt%、7.5wt%、37.8wt%、19.3wt%、7.9wt%。
实施例6
一种低致敏牛乳水解蛋白婴幼儿食品,包括以下重量份原料:脱敏牛乳粉27份、酶解米粉12.7份、微量元素粉7.3份,按上述重量份称取原料混合制得低致敏牛乳水解蛋白婴幼儿食品;
所述脱敏牛乳粉采用实施例3制得的脱敏牛乳粉,其中植物油的重量份为4.7份,低聚半乳糖的重量份为2.2份;
所述微量元素粉包括蛋黄粉、猪肝粉、胡萝卜粉、核桃粉、豌豆粉,且其质量分数分别为17.1wt%、27.6wt%、32.7wt%、11.5wt%、11.1wt%。
对比例2
本对比例为市售米粉(冠军宝贝铁锌钙营养米粉)
取实施例4-6和对比例2,进行营养成分测定,邀请1000位8-12个月的婴幼儿,采用实施例4-6和对比例2进行喂食1个月,测试结束后,由婴幼儿的监护人投票,每位限投一票,得票数计为喜爱度评分,具体测定结果如表2;

由表2可知,实施例4-6制得的婴幼儿食品,含有均衡的营养成分,其中加入脱敏牛乳粉,提升食品中的蛋白质含量、钙含量,有利于婴幼儿的成长,同时使得食品中具有一定的乳香味,更为婴幼儿所接受,加入酶解米粉,较现有米粉类食品,口感更为细腻,更受婴幼儿喜爱。

Claims (5)
1.一种低致敏牛乳水解蛋白婴幼儿食品,其特征在于,包括以下重量份原料:脱敏牛乳粉21-27份、酶解米粉8.5-12.7份、微量元素粉5.1-7.3份、植物油4.1-4.7份、低聚半乳糖1.6-2.2份;
所述脱敏牛乳粉,由以下步骤制备:
步骤A1:取鲜牛乳脱脂处理至含脂率低于0.12%,升温至82℃,对牛乳进行杀菌,冷却至30-35℃,制得脱脂牛乳;
步骤A2:将脱脂牛乳泵入酶解装置中,水解蛋白酶分散于无菌水中后均匀加入脱脂牛乳中,升温至40-45℃,酶解35-42min,再升温至85-93℃,保温2.5-3.3min灭酶处理,降温至30℃,制得酶解牛乳;
步骤A3:将牛磺酸、DHA藻油、乳化剂、无菌水投入乳化机中,制得乳化液,将乳化液加入酶解牛乳中,再加入植物油和低聚半乳糖搅拌制得复合牛乳;
步骤A4:将复合牛乳浓缩至浓度为51-53%,再喷雾干燥制得脱敏牛乳粉。
2.根据权利要求1所述的一种低致敏牛乳水解蛋白婴幼儿食品,其特征在于,所述微量元素粉包括:蛋黄粉、猪肝粉、胡萝卜粉、核桃粉、南瓜粉、豌豆粉。
3.根据权利要求1所述的一种低致敏牛乳水解蛋白婴幼儿食品,其特征在于,步骤A2中,水解蛋白酶包括胰蛋白酶、复合风味蛋白酶,胰蛋白酶、复合风味蛋白酶的用量质量比为1.9:0.85,水解蛋白酶、脱脂牛乳的用量比为1.15-1.27g:1000g。
4.根据权利要求1所述的一种低致敏牛乳水解蛋白婴幼儿食品,其特征在于,步骤A3中,乳化剂为大豆卵磷脂,且酶解牛乳、牛磺酸、DHA藻油、乳化剂、无菌水用量比为1000mL:0.21g:1.7mL:0.25mL:12mL。
5.根据权利要求1所述的一种低致敏牛乳水解蛋白婴幼儿食品的制备方法,其特征在于,称取酶解米粉、脱敏牛乳粉、微量元素粉混合,制得低致敏牛乳水解蛋白婴幼儿食品。
