本发明涉及多糖高效提取和饮料加工技术领域,具体涉及桦褐孔菌多糖的高效提取方法及其杂粮复合饮料的调配,特别是对桦褐孔菌多糖的提取方法。
二、背景技术
桦褐孔菌,又名桦树茸、白桦茸、桦树泪等,是一种生长在白桦树上的木腐类真菌,主要分布于中国黑龙江、吉林,俄罗斯西西伯利亚,北美,芬兰,波兰等北部寒冷地带。因桦褐孔菌中富含多糖、多酚、三萜、皂苷、黄酮等化合物,所以其具有肿瘤防治、“三高”防治、抗糖尿防治、调节人体免疫力的作用。
桦褐孔菌水提取物中,多糖含量最高,但是现在桦褐孔菌多糖提取技术仍不完善,无法高效率获取桦褐孔菌内多糖,造成资源浪费。
三、发明内容
本发明的目的是提供一种桦褐孔菌杂粮复合饮料,这种桦褐孔菌杂粮复合饮料用于制备具有抗糖尿防治、调节人体免疫力的杂粮复合饮料。
为了实现上述目的,本发明采用的具体技术方案如下:这种桦褐孔菌杂粮复合饮料由桦褐孔菌提取液、青稞提取液、甘草提取液、木糖醇调配而成,按重量份数计:桦褐孔菌提取液40-50份,青稞提取液20-30份,甘草提取液10-20份,木糖醇5-6份。

步骤一、制备桦褐孔菌提取液;
步骤二、制备青稞提取液;
步骤三、制备甘草提取液;

有益效果:
1、本发明杂粮复合饮料有抗糖尿防治、调节人体免疫力的作用,属于功能型保健饮料。
2、本发明中桦褐孔菌多糖的提取方法为球磨研磨与气流超微粉碎联用预处理技术。由于球磨粉碎会使原料粒径出现先减小后增大的现象,所以会导致球磨粉碎的局限性比较大。实验发现,使用球磨粉碎桦褐孔菌时,研磨1h时桦褐孔菌的粒径达到最小值,粒径约为3.15μm。气流超微粉碎仪粉碎桦褐孔菌,当时间为25min后,粒径基本处于平稳状态,粒径约为2.72μm。本发明选择两者联用技术,即先使用球磨仪进行1h研磨,在经过气流超微粉碎仪粉碎25min,最终得到桦褐孔菌粉末的粒径约为2.63μm。
3、对于桦褐孔菌中的多糖活性成分,传统提取方法为超声波辅助、微波辅助等热水浸提制备,本发明采用球磨研磨与气流超微粉碎联合预处理桦褐孔菌原料,即使用球磨将桦褐孔菌粒径研磨至最小而后放入至气流超微粉碎仪中,从而可以减小桦褐孔菌粒径,增大多糖等活性成分得率,缩短萃取时间,提高生产效益,此法操作简便,破碎强度大,适宜工业化生产。
4、本发明提供了一种以桦褐孔菌、青稞、甘草为原料制成杂粮复合饮料,纯天然、健康营养等特点。本发明杂粮复合饮料,颜色为暗红色,有青稞香气、口味清甜、且该杂粮复合饮料中添加了天然的木糖醇,适宜糖尿病人饮用,具有抗糖尿防治、调节人体免疫力的作用。
四、具体实施方式
下面对本发明做进一步的说明:
这种桦褐孔菌杂粮复合饮料由桦褐孔菌提取液、青稞提取液、甘草提取液、木糖醇调配而成,按重量份数计:桦褐孔菌提取液40-50份,青稞提取液20-30份,甘草提取液10-20份,木糖醇5-6份。

这种桦褐孔菌杂粮复合饮料通过以下方法制备:
1、原料预处理,将桦褐孔菌子实体粉碎,过60目筛,置于球磨仪中,而后在置于气流超微粉碎仪中,得到桦褐孔菌粉末,备用。青稞清洗灰尘,去除不良籽粒,烘干后置于烤箱中,上下火130℃,烘烤30-45min,备用。甘草清洗灰尘,备用。
2、浸提,将处理好的三种原料进行水浴加热,目的是得到三种原料浸提液,具体如下:
1)取桦褐孔菌10g粉末,按照料液比1:60加入蒸馏水,85℃水浴加热1.5h,反复三次,收集上清液。
2)取青稞籽粒10g,按照料液比1:40加入蒸馏水,85℃水浴加热1h,反复两次,收集上清液。
3)取青稞10g,按照料液比1:40加入蒸馏水,95℃水浴加热2h,收集上清液。
3、过滤,离心机5000 r/min离心5min,收集上清液。
4、吸附,将三种上清液中分别加入适量硅藻土吸附,除去上清液内杂质。
5、离心,离心机5000 r/min离心5min,收集上清液。
6、调配,将处理好的上清液以桦褐孔菌:青稞:甘草=5:3:2的比例进行调配,木糖醇以6%添加量加入至其中。
7、杀菌,调配好的上清液缓慢加热至85℃,保持15min,自然条件下降温至60摄氏度,保持不变。
8、灌装,取玻璃瓶,高温蒸汽灭菌20min。
9、包装,将调配好的饮料灌入至玻璃瓶中,将玻璃瓶灌满,盖严瓶盖。

2.根据权利要求1所述的桦褐孔菌杂粮复合饮料,其特征在于:所述的桦褐孔菌杂粮复合饮料的制备方法:
步骤一、制备桦褐孔菌提取液;
步骤二、制备青稞提取液;
步骤三、制备甘草提取液;
步骤四、调配,将桦褐孔菌提取液、青稞提取液、甘草提取液按照所述重量份数进行调配,再将木糖醇按照所述份数加入至其中;
步骤五、杀菌,将步骤四调配好的上清液缓慢加热至85℃,保持15min,自然条件下降温至60摄氏度,保持不变;
步骤六、灌装,将玻璃瓶高温蒸汽灭菌20min,将调配好的饮料灌入至玻璃瓶中,将玻璃瓶灌满,盖严瓶盖。
3.根据权利要求2所述的桦褐孔菌杂粮复合饮料,其特征在于:所述的桦褐孔菌提取液的制备方法:
步骤一、原料预处理,将桦褐孔菌子实体粉碎,过60目筛,先使用球磨仪进行1h研磨,再经过气流超微粉碎仪粉碎25min,最终得到桦褐孔菌粉末的粒径约为2.63μm;
步骤二、浸提,将桦褐孔菌粉末进行水浴加热,取桦褐孔菌粉末,按照料液比1:60加入蒸馏水,85℃水浴加热1.5h,反复三次,收集上清液;
步骤三、离心除杂,将步骤二收集的上清液利用离心机5000 r/min离心5min,收集上清液;
步骤四、吸附,将步骤三收集的上清液中加入适量硅藻土吸附,除去上清液内杂质;
步骤五、离心,将步骤四处理后的上清液利用离心机5000 r/min离心5min,收集上清液,得到桦褐孔菌提取液。
4.根据权利要求3所述的桦褐孔菌杂粮复合饮料,其特征在于:所述的青稞提取液的制备方法:
步骤一、原料预处理,青稞清洗灰尘,去除不良籽粒,烘干后置于烤箱中,烘烤30-45min;
步骤二、浸提,将处理好青稞进行水浴加热,按照料液比1:40加入蒸馏水,85℃水浴加热1h,反复两次,收集上清液;
步骤三、离心除杂,将步骤二收集的上清液利用离心机5000 r/min离心5min,收集上清液;
步骤四、吸附,将步骤三收集的上清液中加入适量硅藻土吸附,除去上清液内杂质;
步骤五、离心,将步骤四处理后的上清液利用离心机5000 r/min离心5min,收集上清液,得到青稞提取液。
5.根据权利要求4所述的桦褐孔菌杂粮复合饮料,其特征在于:所述的甘草提取液的制备方法:
步骤一、原料预处理,甘草清洗灰尘;
步骤二、浸提,将处理好的甘草进行水浴加热,按照料液比1:40加入蒸馏水,95℃水浴加热2h,反复两次,收集上清液;
步骤三、 离心除杂,将步骤二收集的上清液利用离心机5000 r/min离心5min,收集上清液;
步骤四、吸附,将步骤三收集的上清液中加入适量硅藻土吸附,除去上清液内杂质;
步骤五、离心,将步骤四处理后的上清液利用离心机5000 r/min离心5min,收集上清液,得到甘草提取液。
6.根据权利要求5所述的桦褐孔菌杂粮复合饮料,其特征在于:所述的桦褐孔菌选自大兴安岭的桦褐孔菌。
7.根据权利要求6所述的桦褐孔菌杂粮复合饮料,其特征在于:所述的青稞为产自西藏的青稞。
8.根据权利要求7所述的桦褐孔菌杂粮复合饮料,其特征在于:所述的甘草产自内蒙古的甘草。
