本发明公开了一株链格孢(Alternaria sp)菌株GW61 CGMCC No.21070及其在制备交链孢酚中的应用。该菌株已于2020年11月27日作为专利菌株保藏于中国微生物菌种保藏管理委员会普通微生物中心(简称为CGMCC,地址为:北京市朝阳区北辰西路1号院3号),保藏中心登记入册编号CGMCC No.21070。本发明有效解决了制备高纯度的交链孢酚标准品这一行业技术难题。
Description
技术领域
本发明涉及链格孢菌生产交链孢酚的技术领域。
背景技术
链格孢(Alternaria sp.)广泛分布于蔬菜、水果、谷物等农产品中。在农产品生产和运输中,污染的交链孢霉可产生多种交链孢霉毒素,主要包括包括交链孢酚、交链孢菌酮酸、交链孢毒素I、II、III、腾毒素等。交链孢霉毒素对生殖和发育系统有很强的毒性,并且具有致畸和致癌性风险。其中交链孢酚是可诱发胎儿食管鳞状细胞癌。尽管对交链孢酚对动物的毒性进行了大量研究,但对于食物和饲料中限量标准目前仍未出台相关政策和官方规定,因此该毒素标准品研发技术发展较为缓慢。如何制备高纯度的交链孢酚标准品用于今后农产品检测一直是本领域没有解决的技术难题。
发明内容
本发明的第一目的是提供一株链格孢(Alternaria sp)菌株GW61,属于链格孢属真菌(Alternaria sp),该菌株已于2020年11月27日作为专利菌株保藏于中国微生物菌种保藏管理委员会普通微生物中心(简称为CGMCC,地址为:北京市朝阳区北辰西路1号院3号),保藏中心登记入册编号CGMCC No.21070。
本发明的第二目的是提供利用该菌株发酵并纯化制得交链孢酚的应用。

本发明有效解决了制备高纯度的交链孢酚标准品这一行业技术难题。

具体实施方式
实施例1
1.菌株GW61的采集
2019年9月从陕西省咸阳市收集的玉米中分离到一株菌,命名为菌株GW61。
2.菌株GW61的鉴定
菌株GW61的BT部分序列见序列表的序列1(SEQ ID NO.1),与GENBANKACCESSIONNO.JN854007.1的序列相似性最高,相似性为99.5%。
根据以上鉴定结果,菌株GW61属于链格孢(Alternaria sp)。
3.菌株GW61的保藏
菌株GW61,属于链格孢(Alternaria sp),已于2020年11月27日保藏于中国微生物菌种保藏管理委员会普通微生物中心(简称为CGMCC,地址为:北京市朝阳区北辰西路1号院3号),保藏中心登记入册编号CGMCC No.21070。
4.菌株GW61制备交链孢酚
1)取在-80℃甘油管中保藏的菌株GW61,用接种环接种于平皿马铃薯葡萄糖琼脂(PDA)培养基上,28℃静置培养5-10天。
所述PDA培养基为:40g Potato Dextrose Agar干粉(美国Gibco公司),蒸馏水定容至1000mL,115℃灭菌30min。
2)用无菌水冲洗步骤1中的平皿PDA培养基收集真菌孢子液,将所收集的孢子液接种到含有发酵培养基的1000mL三角瓶中,28℃静置培养20天得到发酵物;所述发酵物是由孢子、菌丝及所述固体培养基培养基组成。
所述步骤2中,所述PDA培养基上的孢子液的接种量为所述发酵培养基(由大米、玉米、小麦和糙米等和水组成的培养基)重量优选5%;所述由大米、玉米、小麦和水组成的培养基中,所述大米、玉米、小麦和水的重量份数比为100g:100mL;
所述发酵的条件可为20-35℃,如28℃固体静置培养,7-35天(具体可为28天)。
发酵培养基:玉米、大米、小麦、糙米等,并不限于这四种培养基。玉米培养基:将200g大米在200mL水中浸泡过夜,121℃高压蒸汽灭菌20min。大米培养基:将200g玉米渣在200mL水中浸泡过夜,121℃高压蒸汽灭菌20min。小麦培养基:将200g小麦在200mL水中浸泡过夜,121℃高压蒸汽灭菌20min。糙米培养基:将200g糙米在200mL水中浸泡过夜,121℃高压蒸汽灭菌20min。
3)在步骤2得到的三角瓶(含固体发酵物)中加入2.0L有机试剂(乙酸乙酯:甲醇=4:1),室温浸提24h,收集上清液,并重复提取3次,合并上清液。上清液利用旋转蒸发仪减压蒸馏至干燥,得到粗提物。
4)将步骤3中得到的粗提物利用二氯甲烷和甲醇混合溶剂(1:1)溶解并与硅胶混合(粗浸膏与硅胶的质量比为1:2),待溶剂挥发后,进行吸附层析分离(吸附层析分离为薄层层析硅胶柱分离,所述凝胶柱的型号为Sephadex LH-20),先用二氯甲烷洗脱两次,再用二氯甲烷与甲醇的混合液洗脱五次,二氯甲烷与甲醇的体积比依次为:99:1、99:1、98:2、98:2和97:3依次得到7个馏分。
5)将步骤4依次得到的7个馏分中第7个馏分用体积比为2:1的二氯甲烷和甲醇的混合液作为洗脱液吸附层析(凝胶柱层析)分离,每50ml收集1份,依次得到5个馏分,将所述5个馏分中第5个馏分纯化(用反相色谱柱进行纯化,流动相为体积百分浓度为65%的甲醇水溶液;该反相色谱柱可为型号为RP-18的Agilent反相色谱柱,色谱柱的柱长为250mm,内径为9.4mm,检测波长254纳米)后,得到如下式I所述化合物:

式I化合物的HPLC纯度色谱图如图1所示,峰面积分析其中式I含量为99.84%。
