Abstract
本发明公开了一种具有抗三高和减肥效果的SOD保健组合物,包括如下重量百分比的原料:超氧化物歧化酶35‑60%,洋葱提取液1‑8%,芹菜梗提取液1‑8%,黑芝麻提取液1‑8%,核桃提取液1‑8%和余量三七提取液,该保健组合物具有减肥、降血脂、降血压和降血糖的效果。本发明还公开了该具有抗三高和减肥效果的SOD保健组合物的制备方法和应用。

本发明涉及保健品领域,特别涉及一种具有抗三高和减肥效果的SOD保健组合物及其制备方法和应用。
背景技术
随着人们生活水平的提高,人均体重得到普遍增加,同时相应出现一些过度肥胖的人群,而由肥胖所带来的一系列疾病发生率也相应出现逐渐上升的趋势。肥胖是危害人类健康的一种以脂肪代谢失衡为主的营养性代谢紊乱疾病,是多种因素综合作用的结果。肥胖对人的健康危害性极大,主要表现在肥胖与心血管疾病、II型糖尿病、高血压、血脂异常、高尿酸血症、某些肿瘤以及睡眠呼吸紊乱等多种疾病密切相关。
而药物或手术进行减肥副作用较大,因此需要研发无副作用的天然保健品或药品的组分。
发明内容
本发明的第一目的是提供已有具有抗三高和减肥效果的SOD保健组合物,其具有可减肥和抗三高作用,且无副作用的优点。
本发明的上述技术目的是通过以下技术方案得以实现的:
一种具有抗三高和减肥效果的SOD保健组合物,包括如下重量百分比的原料:
进一步设置:所述超氧化物歧化酶的制备方法包括如下步骤:
原料预处理,获得待处理原料,所述原料中包含玉米籽粒;
培养诱导,将待处理原料在25-30℃中浸泡10-20h,在温度25-30℃,相对湿度10-40RH%的条件下使玉米籽粒发芽,所述玉米籽粒的芽长1-2mm,并加入辅料,辅料包括芦荟、大豆和刺梨,获得初处理原料,初处理原料中包括发芽的玉米籽粒90-100份,芦荟0.2-1份,大豆0.2-1份,刺梨0.5-2份;
破碎溶解,将初处理原料破碎处理,然后用磷酸盐缓冲液溶解;
色谱分离,将经破碎溶解处理的溶液进行色谱分离,获得SOD酶液。

SOD在抗衰老、抗疲劳、抗心血管系统疾病上具有较好的效果;超氧化物歧化酶是生物体内催化氧自由基发生歧化反应的重要酶,可以通过阻断自由基的链反应,清除机体代谢过程中产生的超氧阴离子自由基,延缓由于自由基侵害而出现的衰老现象;提高人体对由于自由基侵害而诱发疾病的抵抗力;提高人体对自由基外界氧化因子的抵抗力,增强机体对外界环境的适应力。
黑芝麻能够提高的的SOD的活力,同时能明显降低MDA的活力,表明黑芝麻具有抗氧化衰老的作用。黑芝麻油因为富含的不饱和脂肪酸的降血脂和其他活性成分的抗氧化作用,能够达到预防并减轻动脉粥样硬化的发生和发展。黑芝麻黑色素具有保肝护肝作用,其作用机制与抗氧化作用有关。还有很多研究表明,芝麻素对于慢性肝损伤有显著的保护作用,并能够改善四氯化碳引起的慢性肝损伤大鼠肝细胞的损害及坏死状况,对肝功能有一定的保护作用,而且芝麻素具有调节血脂和防治脂肪肝的作用。
芹菜是一种常见蔬菜,不仅具有很高的营养价值,还具有较高的药理作用。芹菜味甘性凉,具有降压,降糖,降血脂,镇静安神等功效,可用于治疗高血压,神经衰弱,食欲不振等症。
洋葱的叶子比其他种蔬菜中总黄酮的含量要高,包括花色素类、黄酮醇类以及黄酮类。其中,包括糖基化衍生物、异鼠李素、槲皮素、山奈酚的糖苷配基等在内的黄酮醇已经有十六种从洋葱中提取出来。另外,洋葱中还含有的甾体皂苷类化合物、含氮化合物、前列腺素类等有效成分,具有抗菌、预防心血管疾病、降血糖、降低胆固醇、抗癌防癌功能、抗氧化作用等。
核桃具有抗氧化、延缓衰老作用,预防心脑血管疾病作用,抑制癌细胞增殖的作用,降血压作用。核桃多肽具有一定的体外降血压活性。
核桃仁中富含丰富的不饱和脂肪酸、多酚类成分及优质的蛋白质,不饱和脂肪酸在预防动脉粥样硬化、降低血脂等方面起着非常重要的作用。
山核桃仁提取液具有较强的抗氧化与清除羟自由基的作用。山核桃仁的总抗氧化能力、还原力和对羟自由基清除率的变化与核桃仁中总多酚含量的变化一致,总多酚含量越高,总抗氧化能力和还原力越强,对羟自由基的清除能力也越强。山核桃中的多酚类物质是自由基的有效清除剂,是阻止山核桃氧化劣变的重要因子。
三七提取物中包括三七总皂苷,对血糖具有明显的调节作用,能使葡萄糖性高血糖降低,同时拮抗胰高血糖素升高血糖的作用,因而具有双向自动调节血糖的功能。三七还具有降低血中胆固醇和血脂类的作用,在血脂代谢中,能降低总脂质的水平,尤其使三酸甘油酯含量明显降低。
本发明的第二目的是提供上述具有抗三高和减肥效果的SOD保健组合物的制备方法,其具有操作简单的优点。

培养诱导,将待处理原料在25-30℃中浸泡10-20h,在温度25-30℃,相对湿度10-40RH%的条件下使玉米籽粒发芽,所述玉米籽粒的芽长1-2mm,并加入辅料,辅料包括芦荟、大豆和刺梨,获得初处理原料,初处理原料中包括发芽的玉米籽粒90-100份,芦荟0.2-1份,大豆0.2-1份,刺梨0.5-2份;
破碎溶解,将初处理原料破碎处理,然后用磷酸盐缓冲液溶解;
色谱分离,将经破碎溶解处理的溶液进行色谱分离,获得SOD酶液。生物试验:
用60只小鼠,随机分为10四组,每组6只,第1组服用普通饲料,第2组服用高脂高热量饲料,第3组至第7组在服用的高脂高热量饲料中加入实施例1-5的保健组合物,第8组至第10组在服用的高脂高热量饲料中加入对比例1-3的保健组合物,并充分混合,其中保健组合物用量为7mg/g体重。每日测进食量、每周测体重,饲养、观察4周,最后解剖、观察内脏脂肪,并取腹主动脉血液检测血糖,检测结果见表1。

其中:TC是指血液中各脂蛋白所含胆固醇之总和,FBG是大鼠空腹血糖。
根据上表,可见第3组至第7组的小鼠具有良好的减肥的效果,能够较好的降低的血糖和血脂;第8组中减少了SOD酶的含量,可见SOD酶对减肥和抗三高具有较好的效果;第9组和第10组分别取消了洋葱提取液和黑芝麻提取液的添加,可见洋葱提取物和黑芝麻提取物对保健组合物的效果具有促进效果。

Claims (9)
2.根据权利要求1所述的具有抗三高和减肥效果的SOD保健组合物,其特征在于,所述超氧化物歧化酶的制备方法包括如下步骤:
原料预处理,获得待处理原料,所述原料中包含玉米籽粒;
培养诱导,将待处理原料在25-30℃中浸泡10-20h,在温度25-30℃,相对湿度10-40RH%的条件下使玉米籽粒发芽,所述玉米籽粒的芽长1-2mm,并加入辅料,辅料包括芦荟、大豆和刺梨,获得初处理原料,初处理原料中包括发芽的玉米籽粒90-100份,芦荟0.2-1份,大豆0.2-1份,刺梨0.5-2份;
破碎溶解,将初处理原料破碎处理,然后用磷酸盐缓冲液溶解;
色谱分离,将经破碎溶解处理的溶液进行色谱分离,获得SOD酶液。
3.根据权利要求2所述的具有抗三高和减肥效果的SOD保健组合物,其特征在于,所述超氧化物歧化酶的活性单位为5000-15000U/g。
4.根据权利要求1所述的具有抗三高和减肥效果的SOD保健组合物,其特征在于,所述洋葱提取物采用酶提取法提取,其提取步骤包括:
Sp1、新鲜洋葱去皮、切碎烘干,粉碎;
Sp2、取洋葱粉,用乙醇回流脱脂两次,每次3h,然后抽虑,收集滤渣烘干备用;
Sp3、将脱脂洋葱粉与40倍体积的磷酸盐缓冲溶液和纤维素酶搅拌均匀,53℃恒温酶解100,然后灭酶10min,将酶解液冷却离心,过滤得洋葱提取液。
5.根据权利要求1所述的具有抗三高和减肥效果的SOD保健组合物,其特征在于,所述核桃提取液采用山核桃作为原料。
7.一种权利要求1-6任一所述具有抗三高和减肥效果的SOD保健组合物的制备方法,其特征在于,包括如下步骤:
步骤一、分别制备超氧化物歧化酶、洋葱提取液、芹菜梗提取液、黑芝麻提取液、核桃提取液和三七提取液分别加入搅拌釜,在25℃条件下,搅拌35min;
步骤二、然后将搅拌釜内混合液加热至40摄氏度保温6h,然后进行过滤,得到保健组合物。
8.如权利要求1-6任一所述的具有抗三高和减肥效果的SOD保健组合物在减肥或抗三高保健品中的应用。
9.如权利要求1-6任一所述的具有抗三高和减肥效果的SOD保健组合物在减肥或抗三高药品中的应用。
