Abstract
本发明提供了一种式(I)所示的作为口服液抗氧添加剂的11‑O‑罗汉果醇肟醚衍生物以及一种以所述式(I)化合物为抗氧剂的抗氧化抗衰老口服液。所述口服液包括以下重量份的原料:0.2‑0.5份式(I)所示的11‑O‑罗汉果醇肟醚衍生物,20‑30份罗汉果脱色浓缩汁,3‑6份秋葵汁,100‑150份水。本发明提供的口服液,口感醇正,保留了罗汉果的特殊风味,颜色澄清透亮,气味果味浓郁。同时通过小鼠试验验证,口服液能够显著提高小鼠的抗氧化能力。说明本发明提供的口服液能够有效缓解,防止生物有机体内自由基对器官产生的氧化损伤。

背景技术
有研究认为,人体的衰老部分原因是由于自由基引发脂质过氧化,产生过氧化脂质(LPO) 等代谢产物,形成褐色物质沉积于各个器官中,最终导致细胞的生理功能和损伤,可能导致免疫力下降,记忆力衰退等。这些自由基包括氧自由基、氢自由基和过氧化氢等,它们广泛存在于机体组织中,攻击细胞膜、线粒体膜中的不饱和脂肪酸,造成脂质过氧化增强,脂质过氧化产物又可分解为更多的自由基,引起自由基的连锁反应,引起脑细胞、肌肉细胞等广泛损伤。正常情况下,人体内的自由基是处于不断产生与清除的动态平衡之中。自由基产生过多或清除过慢,它通过攻击生命大分子物质及各种细胞,会造成机体在分子水平、细胞水平及组织器官水平的各种损伤,加速机体的衰老进程并诱发各种疾病。人体在不可避免地产生自由基的同时,也在自然产生着抵抗自由基的抗氧化物质,以抵消自由基对人体细胞的氧化攻击。研究证明,人体的抗氧化系统和人体的免疫系统都是对任一健康的重要系统,有机体抗氧化的能力和健康的关系愈显重要。随着科学技术的发展,生产生活中可能产生各种过氧化物;手机、电视、电脑等产生的辐射等也会产生自由基,此外,现代人压力大,生活节奏加快,饮食习惯胡奥,睡眠不足造成的亚健康状态,以及强力劳动及大运动量者、在缺氧环境中作业者、抵抗力低下者、血脂偏高者容易产生缺氧都可能导致人体的抗氧化系统失调,从而破坏人体细胞的完整性、稳定性,它对人体的伤害医学上称之为“氧化损伤”。如何增强免疫力,提高人体抗氧化能力,成为全社会都关注的问题。
因此,在食品生产中,为了消除自由基带来的影响,人们常常使用化学合成的抗氧化剂,虽然效果好,但是本身存在一定的安全性。因此,人们往往更倾向于使用天然的、可靠的、安全的抗氧化产品。现有技术有大量的采用天然物质作为抗氧化剂组分,配制口服液的技术。比如以黑蒜,虫草等为原料的口服组合物。但一般存在制备工艺复杂,成本高昂等缺点。
罗汉果是我国独有的药食同源植物,富含各种营养元素和黄酮,多糖,多酚等生物活性物质。罗汉果苷,特别是罗汉果甜苷V是目前最受欢迎的代替蔗糖的甜味剂,以高甜度,低热量的特点在欧美市场备受欢迎。有文献对罗汉果苷的生物活性特别是抗氧化作用进行了研究,发现罗汉果苷V具有一定的抗氧化能力,对羟基和超氧阴离子自由基均有一定的清除作用。一般认为罗汉果苷具有抗氧化活性是因为苷元上氢原子有一定活性,可以和羟基自由基结合,皂苷母核上的碳原子可以形成碳自由基,进一步氧化形成过氧自由基。四环三萜类皂苷的苷元以及羟基是具有抗氧化活性的关键集团。但罗汉果皂苷类化合物的抗氧化机制还没有深入研究,有待进一步探究。而且罗汉果甜苷作为抗氧剂,抗氧化能力较弱,所以目前罗汉果易提取物的产品主要还是以甜味剂为主。罗汉果醇(Mogrol)是罗汉果甜苷ⅠA1、ⅠE1、ⅡE、Ⅲ、Ⅳ、ⅣE、Ⅴ和赛门苷I等的苷元;11-O-罗汉果醇(11-O-Mogrol)则是11-O-罗汉果苷V的苷元,据报道,11-O-罗汉果醇的抗氧化能力较低,不适宜作为食品或者护肤品中的抗氧剂使用。
我国罗汉果资源丰富,在生产罗汉干甜苷作为主要产品的同时,会产生大量副产品,如何对这些副产物进行有效利用,也是对罗汉果资源综合,全面的利用,进一步提高罗汉果经济价值,具有重要意义。现有技术中还未有以11-O-罗汉果醇衍生物为抗氧剂的口服液产品。

本发明的再一个目的是提供一种抗氧化抗衰老口服液,期中抗氧化剂包括上述式(I)化合物。
进一步地,本发明提供的口服液,包括以下原料:式(I)所示的11-O-罗汉果醇肟醚衍生物,罗汉果脱色浓缩汁,秋葵汁。
更进一步地,所述口服液,包括以下重量份的原料:0.2-0.5份式(I)所示的11-O-罗汉果醇肟醚衍生物,20-30份罗汉果脱色浓缩汁,3-6份秋葵汁,100-150份水。
所述(I)所示的11-O-罗汉果醇肟醚衍生物通过将11-O-罗汉果醇肟化后,在以卤化试剂反应后得到的肟醚化产物。
进一步地,所述罗汉果脱色浓缩汁的制备方法参考申请人在前的专利申请201810969469.4,包括如下步骤:
a、鲜果榨汁、冷提:将罗汉果鲜果洗净后,压榨,果渣再用冰水浸提,将榨汁与冰水提取液合并,得罗汉果果汁;
b、酶解、熬煮、离心:罗汉果果汁的pH值至碱性,再加入蛋白酶,酶解,然后,将酶解液加热熬煮,快速冷却,离心,得酶解离心液;
c、脱盐、脱碱:将酶解离心液上阳离子交换树脂柱进行脱盐、脱碱,收集流出液;
d、脱色、脱苦:将阳离子交换树脂流出液上阴离子交换树脂柱进行脱色、脱苦,收集阴离子交换树脂柱流出液,浓缩,得罗汉果脱色浓缩汁。
罗汉果脱色浓缩汁最大程度了保留了罗汉果的天然有益成分,具有甜度高、低热量、口感最接近蔗糖等特点,功效如下:清热润肺、利咽开音、润肠通便、提高免疫功能、保护肝脏、抑菌、降血糖、抗肿瘤等。
所述秋葵汁是将秋葵打浆,过滤,酶解得到,制备方法包括以下步骤:
a、提取:新鲜的黄秋葵用水清洗干净,用打浆机破碎,加入纯水,加热提取,过滤,得秋葵提取液;
b、纯化:将秋葵提取液冷却至室温,加入复合酶(果胶酶、蛋白酶和纤维素酶),酶解,高温灭酶。加入活性炭搅拌脱色,陶瓷膜过滤,将陶瓷膜滤液减压浓缩,得秋葵汁。
步骤S1)中复合酶的用量为黄秋葵的0.3-0.5wt%;步骤S2)中,酶解温度为35-40℃,酶解时间4-6h,减压浓缩至糖度30-40brix。
高温灭酶的温度是本领域所公知的,一般为80-90℃。
秋葵中富含多种营养物质,含有阿拉伯聚糖、半乳聚糖、鼠李聚糖、蛋白质,维生素A 和β—胡萝卜素等,还富含铁、等人体必须元素,能够增强体力、提高免疫力,有益于视网膜健康、维护视力。富含有锌和硒等微量元素,能增强人体免疫力,增强体质,减少疾病风险。发明人预料不到地发现,本发明提供的口服液中,式(I)的11-O-罗汉果醇肟醚衍生物和秋葵汁能够起到协同增效的作用,经过试验验证,能够显著提高免疫力。
可选地,本发明提供的口服液中,还可以加入各种助剂,比如酸味调节剂,甜味剂,稳定剂,氨基酸,植物提取液,食用色素。这些助剂的种类和用量是本领域所熟知的。
举例而言,所述如酸味调节剂包括但不限于柠檬酸和苹果酸;所述甜味剂包括但不限于蜂蜜、甜菊糖甙或罗汉果糖甙;所述稳定剂包括但不限于羧甲基纤维素钠或聚葡萄糖;所述植物提取液包括但不限于金银花提取液、枸杞提取液、桑葚提取液、芦笋提取液或葡萄籽提取液;所述食用色素包括但不限于番茄红素。
所述水没有特别限定,一般饮品中常规用水即可,一般所用水经过杀菌处理。
本发明的另一目的是提供上述口服液制备方法,包括以下步骤:
(1)按重量份称取式(I)所示的11-O-罗汉果醇肟醚衍生物、秋葵汁、罗汉果脱色浓缩汁,可选地,加入助剂,加入无菌纯化水搅拌均匀,加热溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到含有11-O-罗汉果醇肟醚衍生物和秋葵汁的口服液。
步骤(1)中所述加热的温度为60-80℃,步骤(2)中所述超滤膜的截留分子量为2000-3500Da。
按照本发明上述方法制备得到的含有式(I)所示11-O-罗汉果醇肟醚衍生物和秋葵汁的口服液,口感醇正,保留了罗汉果的特殊风味,颜色澄清透亮,气味果味浓郁。
发明人将式(I)化合物和秋葵汁复配后,预料不到底发现,能够显著提高口服液抗氧化能力,在小鼠试验中,过氧化脂质降解产物丙二醛(MDA)含量明显降低,超氧化物歧化酶(SOD) 和谷胱甘肽过氧化物酶(GSH-Px)活力明显提高,和空白对照组差异显著(P<0.01)。说明本发明提供的口服液能够有效缓解有机体内自由基等对器官氧化产生的损伤。
具体实施方式
下面结合实施例对本发明作进一步说明。
若无特别说明,实施例中所述“份”均指质量份,%均指质量百分比。
本发明实施例所用超滤膜采购自陶氏DOW,截留分子量分为3000Da。
制备例1式(I)化合物的制备
(1)肟化、后处理:取11-O-罗汉果醇23.8g,溶解于150ml无水甲醇,加入硫酸羟胺4.5g(含羟胺0.055mol)以及三乙胺20.2g,70℃回流反应5小时,硅胶TLC检验反应结束后,减压浓缩至浓缩液固形物含量25%,将浓缩液冷却至温度10℃,以10r/min的速度搅拌,析晶18小时,抽滤,干燥,得11-肟-罗汉果醇21.6g。
(2)醚化、后处理:取步骤(1)所得11-肟-罗汉果醇20.2g(0.04mol)溶解于110mlDMF,加入氯丙烯3.14g以及叔丁醇钠0.08g,5℃反应5小时,硅胶TLC检验反应结束后,将反应液倒入1000ml冰水中,用800ml石油醚萃取,收集石油醚层,用水将石油醚层洗至中性,将石油醚层以1BV/小时的流速通过硅胶层析柱(柱层析硅胶的规格为80~100目,硅胶的用量为400g,硅胶层析柱的高径比为5:1),用3BV洗脱剂(石油醚:二氯甲烷=7:1,V/V) 洗脱,洗脱的流速为1.5BV/小时,收集洗脱液,浓缩,挥干溶剂,得11-肟丙烯醚-罗汉果醇17.4g,纯度99.1%,产率65.7%,其结构如式(I-2)所示。
1H NMR(CDCl3):1.05(d,3H),1.13-1.21(m,12H),1.24-1.26(m,11H),1.29(q,2H),1,43-1.47(m,5H),1.65(t,1H),1.92(d,2H)1.97(t,1H),3.23(t,1H),3.28(t,1H),4.22(d,2H), 5.23(d,2H),,5,37(s,1H),5.92(s,1H)。Anal.Calcd.for C33H55NO4:C.74.81,H.10.45,N. 2.64,O.12.09。m/z:529.38,Found:529.56。
制备例2罗汉果脱色浓缩汁的制备
(1)鲜果榨汁、冷提:将200kg罗汉果鲜果洗净后,用榨汁机压榨,果渣再用600kg,8℃的冰水浸提2次,每次0.5h,将榨汁与冰水提取液合并,得罗汉果果汁;
(2)酶解、熬煮、离心:用氢氧化钙的饱和水溶液调节步骤(1)所得罗汉果果汁的pH值至9,再加入0.1kg碱性蛋白酶制剂,在25℃下,酶解1.5h,然后,将酶解液在90℃下,加热熬煮0.5h,以2℃/min的速度,快速冷却至30℃,离心,得酶解离心液;
(3)脱盐、脱碱:将步骤(2)所得酶解离心液,以上柱的流速为1.5BV/h,上001×7型阳离子交换树脂柱(001×7型阳离子交换树脂的用量为50kg,阳离子交换树脂柱的高径比为3:1)进行脱盐、脱碱,收集流出液;
(4)脱色、脱苦:将步骤(3)所得流出液,以上柱的流速为1.5BV/h,上LX-T5型阴离子交换树脂柱(LX-T5型阴离子交换树脂的用量为50kg,阴离子交换树脂柱的高径比为 3:1)进行脱色、脱苦,收集流出液,在50℃,-0.09MPa下,薄膜浓缩至糖度为67brix,得无蛋白罗汉果纯味浓缩汁21.39kg。
所得无蛋白罗汉果浓缩汁中,经凯氏定氮法检测,蛋白质的质量含量<0.1%,;经高效液相色谱外标法检测,罗汉果甜苷V的质量含量为4.53%,罗汉果甜苷V的收率为95%;所得无蛋白罗汉果浓缩汁呈淡黄色透明、浓稠液体,气味清香,滋味甘甜、无异味。
制备例3秋葵汁的制备
a、提取:新鲜的黄秋葵50kg,用水清洗干净,用打浆机破碎,加入300kg纯水,加热至85℃,搅拌提取2h,过滤,得秋葵提取液;
b、纯化:将秋葵提取液冷却至室温,加入复合酶(果胶酶、蛋白酶和纤维素酶各0.06kg),35℃酶解4h,90℃高温灭酶。加入0.1kg活性炭,搅拌脱色1h,陶瓷膜过滤,将陶瓷膜滤液在70℃、-0.08MPa条件下减压浓缩至糖度为40brix,得秋葵汁9.6kg。
实施例1
(1)按重量份称取如下组分:0.2份式(I)所示的11-O-罗汉果醇肟醚衍生物、3份秋葵汁、 20份罗汉果脱色浓缩汁,1份柠檬酸,1.3份聚葡萄糖,再加入100份无菌纯化水搅拌均匀,加热70℃溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到含有11-O-罗汉果醇肟醚衍生物和秋葵汁的口服液。
实施例2
(1)按重量份称取如下组分:0.5份式(I)所示的11-O-罗汉果醇肟醚衍生物、6份秋葵汁、 30份罗汉果脱色浓缩汁,2份苹果酸,1.7份聚葡萄糖,再加入150份无菌纯化水搅拌均匀,加热70℃溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到含有11-O-罗汉果醇肟醚衍生物和秋葵汁的口服液。
实施例3
(1)按重量份称取如下组分:0.4份式(I)所示的11-O-罗汉果醇肟醚衍生物、4份秋葵汁、 25份罗汉果脱色浓缩汁,2份苹果酸,1.7份聚葡萄糖,再加入120份无菌纯化水搅拌均匀,加热60℃溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到含有11-O-罗汉果醇肟醚衍生物和秋葵汁的口服液。
对比例1
(1)按重量份称取如下组分:4份秋葵汁、25份罗汉果脱色浓缩汁,2份苹果酸,1.7份聚葡萄糖,再加入120份无菌纯化水搅拌均匀,加热70℃溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到含有秋葵汁的口服液。
对比例2
(1)按重量份称取如下组分:0.4份式(I)所示的11-O-罗汉果醇肟醚衍生物、25份罗汉果脱色浓缩汁,2份苹果酸,1.7份聚葡萄糖,再加入120份无菌纯化水搅拌均匀,加热70℃溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到含有11-O-罗汉果醇肟醚衍生物口服液。
对照组
(1)按重量份称取如下组分:25份罗汉果脱色浓缩汁,2份苹果酸,1.7份聚葡萄糖,再加入120份无菌纯化水搅拌均匀,加热70℃溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到罗汉果浓缩汁稀释后的口服液。
效果例1电子舌口感测试
采用TS-5000Z智能味觉分析系统即电子舌进行品评,采用同人舌头味觉细胞工作原理相类似的人工脂膜传感器技术,可以客观数字化的评价食品的口感指标,本申请从苦味、涩味、酸味、甜味、原料本味等基本味觉感官指标,同时还可以分析苦的回味、涩的回味和鲜的回味(丰富度)8个口感对上述实施例和对比例,以及对照组进行进行评价,以10分为单项满分,80分为总计满分,分数越高,说明饮品的口感越佳。结果如下表1所示:

效果例2主观感官评价
对所得口服液饮品评价分为四个维度,即色泽、气味、口感和溶液澄清度。其中色泽主要通过肉眼和420nm处吸光度确认;口感和风味通过通过申请人在前的专利CN108497248A中记载的方法进行。将实施例,对比例以及对照组所得饮品各配制10等份,请10位评审人品尝,进行感官评价,并按照表2的标准进行打分,总分为100份,评分越高,说明饮品的主观感官评价越佳。结果如下表3所示:

通过表1和表3数据可以得知,本发明提供的口服液饮品口感佳,香味纯正,颜色澄清透明,(I)所示的11-O-罗汉果醇肟醚衍生物以及秋葵汁加入没有影响罗汉果提浓缩汁的天然独特风味,无论在电子舌还是主观感官评价中,本发明提供的口服液饮品和对照组基本无差别。
效果例3毒性实验
(1)口服给药
用昆明种小鼠,体重20g±2g,雌雄各半,每实验组共20只。将实施例1-3制备得到的口服液,一日3次灌胃给予禁食小鼠、每次0.5mL,连续观察7天,未见死亡,未见毒副反应。
(2)长期毒性试验
用SD大鼠,体重200±20g,雌雄各半,共。分别灌胃给药。将实施例1-3口服液,剂量按照每两周按照1mL/kg/d、2mL/kg/d、5mL/kg/d、10mL/kg/d,20mL/kg/d五个浓度递增,每天连续给药,持续12周。血象和肝,肾功能及血糖等生化指标均无明显差异;对重要脏器未引起明显的病理,表明20mL/kg/d时没有毒副反应。
效果例4抗氧化性的测试
按照国家食品药品监督管理总局颁发的《抗氧化功能评价方法》以及《保健食品功能学评价程序和检验方法规范》,测试本发明所得口服液饮品的抗氧化性能。具体是选择20±2g 健康小鼠,适应性喂养5天,分为实验组(实施例1-3),对比组(对比例1-2),空白对照组,一共6组,每组16只。
造模:小鼠用D-半乳糖以0.1mg/10g的注射量腹腔注射,每日一次,连续造模6周。每组取一半小鼠(8只),断头处死小鼠,取血液制备血清,按照TBA荧光显色法测试过氧化脂质降解产物丙二醛(MDA),邻苯三酚自氧化法测试超氧化物歧化酶(SOD),DTNB直接法测试谷胱甘肽过氧化物酶(GSH-Px)。
剩余小鼠按照1mL/kg/d给与受试样品,口服液(实施例,对比例和空白对照组共6组)按照灌胃给予小鼠,,一日一次,每次0.2mL,持续1个月后断头处死小鼠,取血液制备血清,测试MDA、SOD、GSH-Px。结果如下表4所示:

从表4结果可知,对注射D-半乳糖的小鼠造模6周后,给予本发明实施例口服液后,小鼠体内的抗氧化能力恢复,或者有所增强。血清中MDA含量明显降低,而SOD和GSH-Px 的活力明显提高。并且与对照组差异显著(P<0.01)。而对比例2,仅含有式(I)所示的11-O-罗汉果醇肟醚衍生物,给予小鼠后,虽然抗氧化能力也有提高,但显著性不如实施例。说明式(I)所示的11-O-罗汉果醇肟醚衍生物和秋葵汁,两者发挥了协同增效的作用,秋葵汁的存在,更为显著地提高了口服液对于小鼠的抗氧化能力的增强。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于此。在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,包括各个技术特征以任何其它的合适方式进行组合,这些简单变型和组合同样应当视为本发明所公开的内容,均属于本发明的保护范围。
Claims (10)
2.一种抗氧化抗衰老口服液,其抗氧剂包括权利要求1所述的式(I)化合物。
3.如权利要求2所述的口服液,其特征在于,包括以下原料:式(I)所示的11-O-罗汉果醇肟醚衍生物,罗汉果脱色浓缩汁,秋葵汁。
4.如权利要求3所述的口服液,其特征在于,包括以下重量份的原料:0.2-0.5份式(I)所示的11-O-罗汉果醇肟醚衍生物,20-30份罗汉果脱色浓缩汁,3-6份秋葵汁,100-150份水。
5.如权利要求2或3所述的口服液,其特征在于,所述罗汉果脱色浓缩汁的制备方法包括如下步骤:
a、鲜果榨汁、冷提:将罗汉果鲜果洗净后,压榨,果渣再用冰水浸提,将榨汁与冰水提取液合并,得罗汉果果汁;
b、酶解、熬煮、离心:罗汉果果汁的pH值至碱性,再加入蛋白酶,酶解,然后,将酶解液加热熬煮,快速冷却,离心,得酶解离心液;
c、脱盐、脱碱:将酶解离心液上阳离子交换树脂柱进行脱盐、脱碱,收集流出液;
d、脱色、脱苦:将阳离子交换树脂流出液上阴离子交换树脂柱进行脱色、脱苦,收集阴离子交换树脂柱流出液,浓缩,得罗汉果脱色浓缩汁。
6.如权利要求2或3所述的口服液,其特征在于,所述秋葵汁是将黄秋葵打浆,过滤,酶解得到。
7.如权利要求6所述的口服液,其特征在于,所述秋葵汁的制备方法包括以下步骤:
S1)、提取:新鲜的黄秋葵用水清洗干净,用打浆机破碎,加入纯水,加热提取,过滤,得秋葵提取液;
S2)、纯化:将秋葵提取液冷却至室温,加入复合酶酶解,高温灭酶,加入活性炭搅拌脱色,陶瓷膜过滤,将陶瓷膜滤液减压浓缩,得秋葵汁,所述高温灭酶的温度是80-90℃。
8.如权利要求7所述的口服液,其特征在于,所述复合酶为果胶酶、蛋白酶和纤维素酶按照质量比为1-2:1-2:1-2的复配。
9.如权利要求8所述的口服液,其特征在于,步骤S2)中复合酶的用量为黄秋葵的0.3-0.5wt%;步骤S2)中,酶解温度为35-40℃,酶解时间4-6h,减压浓缩至糖度30-40brix。
10.权利要求2-9任一项所述口服液的制备方法,包括以下步骤:
(1)按重量份称取式(I)所示的11-O-罗汉果醇肟醚衍生物、秋葵汁、罗汉果脱色浓缩汁,可选地,加入助剂,加入无菌纯化水搅拌均匀,加热溶解,保温;
(2)保温结束后,将步骤(1)中的物料用超滤膜过滤,UHT灭菌,无菌灌装,得到含有11-O-罗汉果醇肟醚衍生物和秋葵汁的口服液。
