Abstract
本发明提供了一种具有增强人体免疫力的罗汉果饮品及制备方法,其包括如下原料:罗汉果浓缩液,雪梨浓缩液、低聚果糖,酵素,人参根提取物、β‑葡聚糖,天然功能性浓缩液,所述天然功能性浓缩液选自橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液中的至少一种。本发明所得罗汉果饮品,外观颜色澄清透亮,长时间贮存放置稳定性好,口感柔和平衡,避免了一般罗汉果饮品发苦发涩等问题。本发明提供的饮品具有显著的润喉清肺止咳效果。而且,本发明通过枸杞浓缩液,β‑葡聚糖和人参根提取物三者发挥了协同增效的作用,极大提升了生物机体的免疫力。

技术领域
本发明涉及一种罗汉果制取物,还涉及含有这种制取物的罗汉果饮品及其制备方法。
背景技术
罗汉果属葫芦科植物,是一种多用途的药用和保健果品。据《中药大辞典》记载,罗汉果味甜、性凉、无毒,入脾肺,具有清肺止咳,润肠通便、清热解毒、祛痰补血、益肝、健脾、镇静、止渴、促进肠胃机能、抑制哮喘以及降压等功效。1987年,卫生部将罗汉果列为既是药物也是食物的中药之一,罗汉果的饮用一般均是用开水冲泡后饮用,但这种饮用方式不够便捷,为解决这一问题,已有诸多厂家将罗汉果制成饮用方便的颗粒冲剂或直接饮用的饮料。
目前市售的罗汉果冲剂或饮料越来越多,多为和其他功能性提取物复配,大多针对如何改善口感,功能性较为单一。
CN201810765590.5公开了一种罗汉果花茶拼饮品,将罗汉果和鲜花按照比例组合压片,然后利用微波真空干燥,控制工艺参数等条件,可以最大程度保留罗汉果的大部分活性营养成分以及鲜花的清香,从而制备得到口感鲜甜的罗汉果花茶。
CN200910114442.8公开了一种罗汉果饮品及其制备方法,按以下方法制得:取干罗汉果于100~120℃温度下焙烤2~3小时,破碎后进行水提,水提液真空浓缩得到浓缩液,取浓缩液于10~35℃环境下密封静置存放30~300天,将所得料液干燥即得。
CN201810450997.9公开了一种罗汉果速溶饮品,罗汉果速溶饮品由罗汉果甙Ⅳ、甘露醇、可可粉、玉米淀粉、麦芽糊精、栀子粉组成。本发明的有益效果是:本发明的一种罗汉果速溶饮品的制备方法的原料来源广,生产成本低,所制得的罗汉果速溶饮品口感好、味甘、清甜爽口、清热解渴,有清热解毒,利咽化痰,生津止渴,治咽炎喉咙嗓子不适等问题。
现有技术中大部分关于罗汉果饮品的专利,主要聚焦于改善罗汉果饮品的口感问题,还有少量专利关注罗汉果中营养物质在提取过程中流失的问题,很少对罗汉果饮品的生理功能进行研究的文献。部分专利的罗汉果饮品,关注罗汉果饮品中热量的问题,期望提供一种甜度与蔗糖接近,但热量远小于蔗糖的饮品。还有部分专利的罗汉果饮品,具有舒缓喉咙,改善多痰、咳嗽症状,适用于肺热或肺燥咳嗽、百日咳及暑热伤津口渴等,此外还有润肠通便的功效。但上述专利中的功能,都是由罗汉果提取物本身具有的特性所带来,并没有和其他组分发挥协同配合的作用。
发明内容
本发明要解决的技术问题是提供一种具有增强人体免疫力的罗汉果饮品及制备方法。所述罗汉果饮品外观澄清透亮,口感柔和,气味清香,长时间放置保持稳定,不产生沉淀絮凝,而且除了市面上常见罗汉果饮品的的润喉清肺生理基础上,本发明提供的罗汉果饮品组分发挥协同配合作用,能够增强人体的免疫力。
要解决上述技术问题,本发明的第一个目的是提供一种具有增强人体免疫力的罗汉果饮品,其包括如下原料:罗汉果浓缩液,雪梨浓缩液、低聚果糖,酵素,人参根提取物、β-葡聚糖,天然功能性浓缩液,所述天然功能性浓缩液选自橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液中的至少一种。
优选地,本发明上述具有增强人体免疫力的罗汉果饮品,包括如下重量份的原料:20-30份罗汉果浓缩液,20-30份雪梨浓缩液、40-50份低聚果糖,2-5份酵素,0.3-0.5份人参根提取物、1-2份β-葡聚糖,2-4份天然功能性浓缩液,所述天然功能性浓缩液选自橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液中的至少一种,条件是所述天然功能性浓缩液中至少包括枸杞浓缩液。
优选地,所述功能性浓缩液为橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液的复配。
进一步优选地,所述功能性浓缩液为橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液按照1-2:2-4:2-4:2-4:2-4:2-4:3-5的复配。
本发明配制罗汉果饮品饮品的原料中,罗汉果浓缩液通过本厂自制,具体是通过如下制备方法得到:
(1)将罗汉果用破碎机适度破碎,将破碎后的罗汉果榨汁,果渣再用水淋洗,合并果汁和淋洗液,得到提取液;
(2)将步骤(1)得到的提取液离心,得离心液;
(3)在步骤(2)得到的离心液中加入蛋白酶试剂进行酶解,得到酶解液,将所述酶解液加热至90℃进行灭酶;
(4)将步骤(3)得到的灭酶后的酶解液通过前后串联的氧化铝层析柱和活性炭层析柱,收集流出液;
(5)将步骤(4)所得的流出液先过超滤膜,再加水透析超滤膜;
(6)将步骤(5)所得的超滤滤出液用纳滤膜进行分离纯化,浓缩,加水透析,;
(7)将步骤(6)纳滤截留液浓缩,得到罗汉果甜浓缩液。
其中步骤(1)所述适度破碎是指每个鲜果破碎至6-10瓣为标准。
所述步骤(3)中蛋白酶和提取液的质量体积比为所述蛋白酶制剂的用量为鲜果重量的0.1‰~0.3‰。
所述步骤(4)中氧化铝层析柱或活性炭层析柱的流速为0.5~2.0BV/h,高径比为2~6:1。
所述步骤(5)中超滤膜的截留分子量为50000~10000Da;
步骤(5)和步骤(6)加水透析直到滤出液电导率小于200μS/cm为符合标准;
所述步骤(7)中浓缩为真空干燥浓缩,浓缩至固形物含量为25~40%。
按照上述方法得到的罗汉果浓缩液,无其它化学添加物,天然绿色,澄清透亮,口感好,风味纯正,清爽醇厚。同时保留了罗汉果中的具有润喉清肺功效的有效成分。
本发明原料所用酵素,口感酸甜,具有良好的润喉清肺、排毒的功效。以玉竹、百合、罗汉果、胖大海、芦根、银耳、菊花这几种药食同源草本为原料,采用恒温多段式发酵工艺,富集多重清肺排毒因子。
本品中使用的酵素口感酸甜,具有良好的润喉清肺、排毒的功效。具体是以玉竹、百合、罗汉果、胖大海、芦根、桑叶、银耳中的至少一种为原料,采用益生菌进行发酵得到,富集多重清肺排毒因子,并且通过益生菌发酵后,原料中的有效成分更容易被机体吸收,使得本品的清肺利咽效果更加显著。
本发明原料所用β-葡聚糖,其是一种由葡萄糖单位组成的多聚糖是由葡萄糖单位组成的多聚糖,能调节血糖水平。
本发明原料用低聚果糖替代了蔗糖等糖浆的加入,不仅降低了产品的热量,更能够有效促进肠道有益菌群的增值,促进肠道健康。
本发明原料所用人参根提取物,具有提高免疫力,缓解疲劳,改善虚弱体质,调节亚健康状态的功能。所用人参根提取物的提取制备方法包括如下步骤:
(1)原料前处理:将除杂除霉的人参根适当破碎;
(2)水提取:将破碎后的原料加入80℃的热水浸提,提取3-5次,每次2-4h;
(3)过滤:将浸提液经过绢布初滤后,过精滤或者离心后,得清液;
(4)浓缩:将清液进行真空减压浓缩至固形物含量15~40%。
本发明原料所用天然功能性浓缩液的原料优选为橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液中的复配。其能够各自发挥功能,其中橘红浓缩液具有理气宽中,燥湿化痰的功效;桑叶浓缩液具有具有疏散风热、清肺润燥、清肝明目的功效;桔梗浓缩液具有具有宣肺祛痰、利咽排脓的功效;金银花浓缩液具有具有清热解毒、疏散风热的功效;玉竹浓缩液具有具有养阴润燥、生津止渴的功效;陈皮浓缩液具有具有理气开胃,燥湿化痰的功效;枸杞浓缩液具有滋补肝肾,益精明目的功效。并且最终所得罗汉果饮品,颜色澄清透亮,口感柔和,平衡。不会出现一般罗汉果浓缩液中发苦发涩,有金属味,酸味等情况。并且产品稳定性好,长时间贮存放置也不会发生絮凝或沉淀现象。
本发明所用原料雪梨浓缩液具有具有润肺清燥、止咳化痰、养血生肌的功效。
上述天然功能性浓缩液以及雪梨浓缩液的制备方法是本领域所熟知的,水提或者醇提皆可。具体是以重量为原料20-40倍,温度为30-70℃的热水为溶媒、分2-5次浸提,经过固液分离后而得到的粗提液再次浓缩而制得。所述固液分离优选为先过筛网后再过孔径为0.45-0.7μm陶瓷膜。所述浓缩的方法为真空浓缩。
人参根提取物本身具有一定的增强人体免疫力的生理功能,发明人预料不到地发现,本发明罗汉果饮品中,枸杞浓缩液,β-葡聚糖和人参根提取物三者发挥了协同增效的作用,使得所得罗汉果饮品增强人体免疫力的生理功能显著增加。
本发明的第二个目的在于提供所述罗汉果饮品的制备方法,包括如下步骤:
(S1)将人参提取物和β-葡聚糖加入热水,搅拌溶解溶,备用;
(S2)将罗汉果浓缩汁、酵素、雪梨浓缩汁、低聚果糖和天然功能性浓缩液与步骤(S1)准备好的人参提取物溶液及β-葡聚糖溶液混合到一起,搅拌混匀,灭菌即得。
步骤(S1)总所述热水的温度为60-80℃,热水和待溶解物(人参提取物和/或β-葡聚糖)的质量比为1-2:1。所述灭菌的方法是本领域所熟知的,本发明具体是先将物料升温到100℃保持5min,再降温至85℃热水中水浴15min,起到消毒杀菌作用,即得成品。
本发明方法的有益效果如下:
一、本发明所得罗汉果饮品,外观颜色澄清透亮,长时间贮存放置稳定性好,口感柔和平衡,避免了一般罗汉果饮品发苦发涩等问题。
二、本发明所得罗汉果饮品,通过枸杞浓缩液,β-葡聚糖和人参根提取物三者发挥了协同增效的作用,极大提升了生物机体的免疫力。

本发明实施例所用酵素为外购,厂家为一阳生(厦门)生物科技有限公司,型号为清菲酵素液。
本发明实施例中罗汉果饮品的感官评价方法为:
根据GB/T 16291.2-2010的规定来筛选感官评定人员,经过口味敏感性培训后,最终筛选20名(男女各半)的感官评价人员,组成感官评定小组。感官分析人员在评定实验开始前1h内限制饮食,尤其是能严重影响味觉的食物。针对不同味感实验从20名中选取10名最合适的感官分析人员。实验过程中,分别取10mL待评定溶液于一次性纸杯中,由感官评定人员含于口中,停留数秒吐出,评定时间间隔为20min,评定结果取10名评定人员打分的平均值。
通过调查问卷的形式,针对放置一个月后的罗汉果饮品进行打分,分别从外观、口感做问卷调查和打分,每项满意度最高10分,每次调查问卷测试人员为12人,去掉一最高分值,去掉一最低分值,最后求其整体的平均值。
实施例1
(1)称取0.3份人参提取物后,加入配方量50%的70℃热水,搅拌使其充分溶解,冷却后备用;
(2)称取1份β-葡聚糖,加入配方量50%的70℃热水搅拌,使其充分溶解,冷却后备用;
(3)将25份罗汉果浓缩汁、3份酵素、0.15份橘红浓缩液、0.3份桑叶浓缩液、0.3份桔梗浓缩液、0.3份金银花浓缩液、0.3份玉竹浓缩液、0.3份陈皮浓缩液、0.4份枸杞浓缩液、23份雪梨浓缩液、48份低聚果糖与上述步骤准备好的人参提取物溶液及β-葡聚糖溶液混合到一起,搅拌10min充分混匀;
(4)分装:在包装机器上,将准备好的罗汉雪梨植物饮品分装至指定的包装容器中;
(5)灭菌:将分装好的罗汉雪梨植物饮品放入85℃热水中水浴15min,起到消毒杀菌作用,即得成品。
所得罗汉果饮品外观为深褐色,流动性好,澄清无杂质;在室温下贮存2个月,无沉淀,状态均匀,稳定性好,够干柔和,清香,回甘。具体的感官评价见表1所示。

注:a表示其中吸光度是将样品用纯水稀释100倍后,用1cm比色皿在620nm处测定,吸光度越低,表示该样品的透明度越高,状态越澄清透亮。
沉淀量是采用加速破坏实验原理,将每个实施例分为5组平行样品,每组重量50g,重量严格一致,放在55℃培养箱中7天,之后将样品稀释10倍后进行离心,测量沉淀重量。沉淀越少,说明越稳定。
应用例2
对本发明部分实施例以及对比例所得饮品,按照《保健食品检验与评价技术规范》进行测试,进行了免疫力的测试。
对小鼠NK细胞活性的影响
实验动物:清洁级Wistar大鼠,购自长沙疾病防治与控制中心。
正常对照组:每只小鼠每天胃饲生理盐水0.3mL,连续30天。
产品组:分为4组,分别为实施例1,对比例1,对比例2,对比例3,实验动物数量为10只,每只小鼠每天胃饲本发明产品0.3mL,连续30天。
具体实验操作如下:采用LDH释放法,小鼠脾细胞悬液,用5%小牛血清RPMI1640培养液稀释细胞浓度至3x107cells/mL。靶细胞采用小鼠白血病细胞株YAC-1细胞(5x105cells/mL),加至96孔板,离心5min,37℃5%CO2培养3h,加入预冷的RPMI1640培养液50μL,离心5min,吸取100μL LDH底液/孔,3min后加入1M HCl(50μL/孔)终止反应,于自动酶标仪读取波长490nm的OD值。
NK细胞活性按照[(反应孔OD-自然释放孔OD)/(最大释放孔OD-自然释放孔
OD)]×100%计算得到。结果以means±s,即平均值±标准偏差给出,测试结果如下表2所示。

由表2数据可以看出,给与小鼠产品30天后,本发明优选实施例(实施例1,2,8)能够显著增加小鼠的NK细胞活性,与对照组有显著差异(P<0.01)。而对比例1-3可以表明,缺失人参提取物、β-葡聚糖和枸杞浓缩液中的任意一种,虽然NK细胞活性相对于对照组有一定提升,但提升幅度大为下降,说明本发明提供的罗汉果饮品中,人参提取物、β-葡聚糖和枸杞浓缩液共同发挥了协同增效的作用,显著地提高了生物机体的免疫力。实施例12,13中人参提取物、β-葡聚糖的用量不在本发明优选范围内,但同样起到了协同增效提升免疫力的作用。
对小鼠血清溶血素的影响
实验动物:清洁级Wistar大鼠,购自长沙疾病防治与控制中心。
正常对照组:每只小鼠每天胃饲生理盐水0.3mL,连续30天。
产品组:分为4组,分别为实施例1,对比例1,对比例2,对比例3,实验动物数量为10只,每只小鼠每天胃饲本发明产品0.3mL,连续30天。
结果见表3所示:

由表3可见,在连续胃饲30天后,本发明优选实施例(实施例1,2,8)三个产品组的抗体积数与阴性对照组相比,经统计学处理,有显著性差异(P<0.01)。对比例1-3与对照组相比,无显著性差异(P>0.05)。说明本发明提供的罗汉果饮品可明显提高小鼠血清溶血素生成。而对比例1-3可以表明,缺失人参提取物、β-葡聚糖和枸杞浓缩液中的任意一种,虽然小鼠血清溶血素的生成对于对照组有一定提升,但提升幅度大为下降,说明本发明提供的罗汉果饮品中,人参提取物、β-葡聚糖和枸杞浓缩液共同发挥了协同增效的作用,显著地提高了生物机体的免疫力。实施例12,13中人参提取物、β-葡聚糖的用量不在本发明优选范围内,但同样起到了协同增效提升免疫力的作用。
本发明提供的罗汉果饮品,对小鼠胃饲30天后,对小鼠的NK细胞活性试验结果为阳性,对小鼠的体液免疫试验结果为阳性,说明本发明提供的罗汉果饮品对生物机体具有增强免疫力的生理功效。
应用例3毒性试验
具体是小鼠饲养环境为室温23-25℃,相对湿度55-60%,分别进行小鼠30天喂养试验,每天胃饲0.3mL本发明实施例1所得饮品。
经小鼠急性经口毒性试验,表明本发明饮品为无毒级物质;经Ames试验、小鼠骨髓细胞微核试验和小鼠精子畸形试验,表明本发明药物未呈现有致突变性;经小鼠30天喂养试验,表明本发明药物对动物的活动、进食、体重增长、红细胞数、血红蛋白含量、白细胞数及分类、肝肾功能、蛋白代谢、脂代谢、糖代谢、脏器湿重及脏体比均无不良影响,各主要脏器的大体及镜下检查均未见有特异性病理改变。
应用例4清肺润喉止咳
案例1:李某,男,45岁,20年以上烟龄,平均每天消耗香烟1~1.5包,长期干咳,咳嗽时喉咙处有明显的淤痰。服用本发明实施例1制备的罗汉果饮品,每次10ml,每日3次,连续服用半个月后,咳嗽次数显著减少,喉咙干痒现象显著改善,咳嗽时淤痰现象减弱。
案例2:张某,男,35岁,某重点高中教师,教授科目数学,平均每天3~4节课,说话声音嘶哑,经常感觉咽喉不适。服用上述实施例8制备的本发明,每次10ml,每日2次,一周后声音音质清亮,喉咙不适症状显著改善。
上述详细说明是针对本发明其中之一可行实施例的具体说明,该实施例并非用以限制本发明的专利范围,凡未脱离本发明所为的等效实施或变更,均应包含于本发明技术方案的范围内。
Claims (9)
1.一种具有增强人体免疫力的罗汉果饮品,其包括如下原料:罗汉果浓缩液,雪梨浓缩液、低聚果糖,酵素,人参根提取物、β-葡聚糖,天然功能性浓缩液,所述天然功能性浓缩液选自橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液中的至少一种。
2.如权利要求1所述的罗汉果饮品,其特征在于,包括如下重量份的原料:20-30份罗汉果浓缩液,20-30份雪梨浓缩液、40-50份低聚果糖,2-5份酵素,0.3-0.5份人参根提取物、1-2份β-葡聚糖,2-4份天然功能性浓缩液,所述天然功能性浓缩液选自橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液中的至少一种,条件是所述天然功能性浓缩液中至少包括枸杞浓缩液。
3.如权利要求2所述的罗汉果饮品,其特征在于,所述功能性浓缩液为橘红浓缩液、桑叶浓缩液、桔梗浓缩液、金银花浓缩液、玉竹浓缩液、陈皮浓缩液、枸杞浓缩液按照1-2:2-4:2-4:2-4:2-4:2-4:3-5的复配。
4.如权利要求1所述的罗汉果饮品,其特征在于,所述罗汉果浓缩液通过包括如下步骤的制备方法得到:
(1)将罗汉果用破碎机适度破碎,将破碎后的罗汉果榨汁,果渣再用水淋洗,合并果汁和淋洗液,得到提取液;
(2)将步骤(1)得到的提取液离心,得离心液;
(3)在步骤(2)得到的离心液中加入蛋白酶试剂进行酶解,得到酶解液,将所述酶解液加热至90℃进行灭酶;
(4)将步骤(3)得到的灭酶后的酶解液通过前后串联的氧化铝层析柱和活性炭层析柱,收集流出液;
(5)将步骤(4)所得的流出液先过超滤膜,再加水透析;
(6)将步骤(5)所得的超滤滤出液用纳滤膜进行分离纯化,浓缩,加水透析;
(7)将步骤(6)纳滤截留液浓缩,得到罗汉果甜浓缩液。
5.如权利要求4所述的罗汉果饮品,其特征在于,其中步骤(1)所述适度破碎是指每个鲜果破碎至6-10瓣为标准;所述蛋白酶制剂的用量为鲜果重量的0.1‰~0.3‰;和/或所述步骤(4)中氧化铝层析柱或活性炭层析柱中上柱的流速为0.5~2.0BV/h,高径比为2~6:1;和/或所述步骤(5)中超滤膜的截留分子量为50000~10000Da,步骤(7)中所述浓缩为真空浓缩,浓缩至固形物含量为25~40%。
6.如权利要求1所述的罗汉果饮品,其特征在于,所述酵素是以玉竹、百合、罗汉果、胖大海、芦根、银耳、菊花中的至少一种为原料,采用益生菌进行发酵得到。
7.如权利要求1所述的罗汉果饮品,其特征在于,所述人参根提取物的提取制备方法包括如下步骤:
(1)原料前处理:将除杂除霉的人参根适当破碎;
(2)水提取:将破碎后的原料加入80℃的热水浸提,提取3-5次,每次2-4h;
(3)过滤:将浸提液经过绢布初滤后,过精滤或者离心后,得清液;
(4)浓缩:将清液进行真空减压浓缩至固形物含量为15~40%。
8.权利要求1-7任一项所述罗汉果饮品的制备方法,包括如下步骤:
(S1)将人参提取物和β-葡聚糖加入热水,搅拌溶解,备用;
(S2)将罗汉果浓缩汁、酵素、雪梨浓缩汁、低聚果糖和天然功能性浓缩液与步骤(S1)准备好的人参提取物溶液及β-葡聚糖溶液混合到一起,搅拌混匀,灭菌即得。
9.如权利要求8所述的方法,其特征在于,步骤(S 1)总所述热水的温度为60-80℃,热水和待溶解物(人参提取物和/或β-葡聚糖)的质量比为1-2:1。
