The cream of the present comparative example is the same as the cream of example 2, except that the mass percentages of acetyl hexapeptide-8, palmitoyl pentapeptide-4, and the plant extract in the cream of the present comparative example are 1%, 13%, and 0.5%, respectively.
Physical and chemical index test one
130 healthy skin volunteers were randomly selected for testing, and a group of example samples was tested every 10 persons, with each group being a unit, and the test results were averaged.
The moisturizing creams of examples 1-8 and the creams of comparative examples 1-5 were tested for use by applying 3 times daily for 30 consecutive days in the morning, at noon and at night.
1. The moisture content before and after use was measured using a skin moisture tester, and the% moisture content increase before and after use was calculated by the following method: skin moisture lifting rate (skin moisture content measured after use-skin moisture content measured before use)/skin moisture content before use × 100%.
2. The skin fineness and wrinkle change are measured by a skin detector, specifically a Visia skin detector; the fineness raising rate is obtained by the following calculation method: the fineness increase rate is (skin fineness measured after use-skin fineness measured before use)/skin fineness measured before use × 100%.
3. The total score of experience score is 10 points, 8-10 points are good, 6-8 points are general, and 6 points and below are poor experience.
The test results are shown in tables 2, 3 and 4.
TABLE 2
Regarding the wrinkle-reducing effect of the cream, it can be seen from the data of table 2 that: (1) the effects of examples 1 to 7 in reducing wrinkles are overall superior to the comparative example. This demonstrates that the rational arrangement of acetyl hexapeptide-8, palmitoyl pentapeptide-4 and plant extracts in the cream formulation of the present invention is of paramount importance. (2) In the whole example, the effect of the example 2 is the best, namely the example 1, the example 3 and the example 6, and meanwhile, the effects of the examples 1 to 6 are better compared with the example 7 on the whole, which shows that the cream formula provided by the invention has a preferable formula.
TABLE 3
Regarding the improvement of moisture content, fineness and use experience of the skin by the face cream, the data in table 3 show that: (1) the effects of examples 1 to 7 are overall superior to those of the comparative example.
After the use, the test product is stopped from being used and other moisturizing products are not used, and a return visit test is carried out after 2 weeks to test the moisturizing retention rate. The retention was calculated by: moisture retention rate is skin moisture content measured after 2 weeks/skin moisture content measured immediately after use × 100%.
TABLE 4
Test items | Moisture retention after 2 weeks/%) |
Example 1 | 88.1 |
Example 2 | 93.2 |
Example 3 | 86.4 |
Example 4 | 85.8 |
Example 5 | 86.3 |
Example 6 | 87.5 |
Example 7 | 83.8 |
Comparative example 1 | 82.6 |
Comparative example 2 | 26.3 |
Comparative example 3 | 33.1 |
Comparative example 4 | 79.2 |
Comparative example 5 | 80.3 |
Regarding the long-lasting moisturizing effect of the cream, it can be seen from the data in table 4 that: (1) the effects of examples 1 to 7 are overall superior to those of the comparative example.
Physical and chemical index test II and permeability test experiment
In the using process of the cream, the palmitoyl pentapeptide-4 penetrates deeply into the dermis tissue, acts on skin fibroblasts, promotes the synthesis of collagen, effectively reverses the aging and reduces wrinkles, so the palmitoyl pentapeptide-4 is used as a test object of permeability.
(1) Taking 8 Kunming mice with similar body weight (within the range of 30-35 g), numbering 1-8, and depilating the skin of the chest and abdomen of the mice with 6 wt% of sodium sulfide;
(2) three random positions (2.0mm multiplied by 1.5mm) are confirmed at the chest and abdomen of the No. 1-10 mice, wherein, 1.0g of the corresponding cream sample is coated at two positions, and 1.0g of the palmitoyl pentapeptide-4 sample is directly coated at the other position;
(3)1 hour later, the neck of the mouse is cut off and the mouse is killed, the chest and abdomen skin coated with the cream sample and the palmitoyl pentapeptide-4 sample is taken down, and the chest and abdomen skin which is not coated with the cream sample and has the same size is taken as a blank control group;
(4) removing epidermis from the chest and abdomen skin of the mouse, grinding the dermal tissue to be chyle, adding 5mL of absolute ethyl alcohol, mixing, centrifuging at 8000r/min for 15min, collecting the supernatant, and purifying with LC-9A high performance liquid chromatograph (Shimadzu, Japan) [ C18 column as chromatographic column, at 25 deg.C, with ethanol as mobile phase: the content of palmitoyl pentapeptide-4 was determined at a flow rate of 1.0mL/min and a detection wavelength of 224nm with water of 10: 90.
The results of the permeability test are shown in table 5, wherein the content of the blank control group is 0%, the content of the sample coated with palmitoyl pentapeptide-4 is 100%, and the results are the average value of the test results of dermal tissues of two different parts.
TABLE 5
Test items | Permeability of |
Example 1 | 63.6% |
Example 2 | 68.2% |
Example 3 | 62.4% |
Example 4 | 64.5% |
Example 5 | 66.8% |
Example 6 | 58.6% |
Example 7 | 60.3% |
Example 8 | 56.1% |
Regarding the penetration effect of the cream, it can be seen from the data in table 5: the permeation effect of examples 1 to 8 is overall superior. Also, the permeability of examples 1-7 is generally better than that of example 8, indicating that there is a preference for the choice of emulsifier and emollient in the formulations of the present invention.
