Description

Dendrobium nobile polysaccharide herbal face cream and preparation method thereof
Technical Field
The invention relates to the technical field of application of traditional Chinese medicinal materials, and in particular relates to dendrobium stem polysaccharide herbal face cream and a preparation method thereof.
Background
With the development of the cosmetic industry and the pursuit of people for beauty, the quality and safety of cosmetics are emphasized more while the effect of skin care products on skin is emphasized, more and more people favor the cosmetics based on natural products, and in order to effectively delay skin aging, the counter measures which can be taken in the field of cosmetics are as follows: supplying water and oil to maintain water-oil balance, inhibiting excessive free radical production (antioxidation), supplying skin collagen and elastin, inhibiting tyrosinase activity and melanin production, inhibiting non-enzymatic glycosylation reaction, etc. The oxidation resistance is an important consideration factor of the quality of cosmetics, but at present, some products based on natural products on the market only add certain components in a targeted manner, and cannot play an oxidation resistance role while effectively resisting wrinkles.
Disclosure of Invention
Aiming at the defects of the prior art, the invention provides the face cream which is prepared from natural products, has the functions of effectively resisting wrinkles and resisting oxidation.
One of the purposes of the invention is to provide application of dendrobium nobile lindl polysaccharide in cosmetics or skin care products with anti-wrinkle effect.
The invention also aims to provide application of the dendrobium nobile lindl polysaccharide in cosmetics or skin care products with anti-wrinkle and anti-oxidation effects.
The invention aims to provide dendrobium nobile polysaccharide herbal face cream, which comprises 2-10% of white vaseline, 1-2% of glyceryl monostearate, 1-2% of stearyl cetyl alcohol, 1-2% of cetyl alcohol, 3-10% of shea butter, 0.05-1% of dendrobium nobile polysaccharide, 60-80% of ultrapure water, 10-20% of propylene glycol and 5-10% of polyglycerol-10 stearate by mass.
The working principle and the beneficial effects of the invention are as follows: dendrobium nobile is a commonly used rare Chinese medicinal material belonging to the genus Dendrobium of the family Orchidaceae, which contains effective components such as alkaloid, polysaccharide, flavonoid and phenols, and substances extracted from the dendrobium nobile are widely used in medicines, foods and cosmetics.
In the invention, the white vaseline has the main functions of: a lubricant, an ointment base; the glyceryl monostearate has oleophilic and hydrophilic groups, and has the functions of moistening, emulsifying and the like; the stearic acid: octadecanoic acid is widely applied to preparation of vanishing cream and cold cream in cosmetics to achieve the emulsification effect. Generally, alkali is used for neutralization when the stearic acid is added, but the addition of an alkali agent is not generally considered, so that the absorption of acid by the skin can be adjusted by adding a small amount of stearic acid; the stearic cetyl alcohol: white solid crystals, particles or wax blocks, and has the effects of inhibiting greasy feeling, reducing viscosity of wax raw materials, stabilizing emulsion of cosmetics, and the like; in various cosmetics, as a base; the effect is better when the composition is matched with hexadecanol; the cetyl alcohol: the emulsion is suitable for cream and emulsification, is used as a matrix, an emulsifier cream, an ointment matrix and the like, has the effects of inhibiting greasy feeling and reducing viscosity of wax raw materials, has similar effect with stearyl cetyl alcohol, and has better matching use effect; the shea butter comprises the following components: the skin care product is rich in unsaturated fatty acid, is easy to absorb, can enhance the oxidation resistance of the skin, is solid oil, can promote cell regeneration, and has a skin nutrition function; the polyglycerol-10 stearate: the high polymer emulsifier has good hydrophilicity, can form o/w type emulsified liquid, and has the functions of stabilizing and thickening; the used water is ultrapure water subjected to impurity removal, so that the influence of impurities on the face cream production is avoided; the propylene glycol is used as a safe antioxidant, has an antioxidant effect on the face cream and has an antioxidant water-locking capacity on the skin.

Cell dosing: after the cells grow in an adherent manner, a control group and an additive group are arranged, a DMEM culture medium is used as the control group, the dendrobium nobile polysaccharide is prepared by a DMEM high-sugar culture medium to have final concentrations of 0.1mg/mL, 0.3mg/mL and 1mg/mL respectively as the additive group, and the dendrobium nobile polysaccharide is put into a cell culture box for culture for 48 hours.
MTT assay to detect 3T3L1 cell viability: and adding 20 mu L of 5mg/mL MTT solution into all the wells respectively, placing the wells in an incubator for incubation for 4h, removing the culture solution by suction, adding 150 mu L of DMSO into each well, keeping out of the light for 20min to fully dissolve crystals, and measuring the light absorption value at 490nm by using an enzyme-linked immunosorbent assay.
The fibroblast is an important cell on the surface of the skin, and has an important effect in the aspect of aging, and as shown in figure 2, the dendrobium nobile polysaccharide has a good effect of promoting the growth of the fibroblast, can be used as a main active ingredient of dendrobium nobile face cream, and can improve the anti-aging activity of the face cream.
Determination of ability of dendrobium nobile polysaccharide to eliminate DPPH free radical
Preparation of DPPH free radical solution: the DPPH was dissolved in 70% methanol to prepare a 0.2mmol/L DPPH solution.
Preparing a dendrobium nobile polysaccharide test solution: precisely weighing 96mg of dendrobium nobile polysaccharide, adding 30ml of ultrapure water to dissolve overnight, centrifuging at 1100rpm for 10min, and taking supernatant to obtain 3.2mg/ml sample mother liquor. Setting the concentration gradient of dendrobium nobile polysaccharide drug as 1.6mg/ml, 0.8mg/ml, 0.4mg/ml, 0mg/ml, 4 concentration gradients. 3 sets of replicates were run at each concentration.
Absorbance of test solution and control solution: 2ml of sample solutions with different concentrations were taken, 2ml of 0.2mmol/L DPPH solution was added, mixed well, reacted in a dark room for 30min, and the absorbance (AX1) was measured at 517nm using a spectrophotometer. Meanwhile, 2ml of sample solutions with different concentrations were taken, 2ml of 70% methanol was added, mixed well, reacted in a dark room for 30min, and then the absorbance (AX2) was measured at 517 mm. 2ml of 70% methanol was added to 2ml of 0.2mmol/L DPPH solution, mixed well, and reacted in the dark for 30min as a blank (A0).
The DPPH radical clearance rate of polysaccharides can be expressed as:
DPPH clearance (100%) (1-AX/A0) 100%
In the formula: AX is the absorbance of the sample after reaction with DPPH solution, and A0 is the absorbance of 70% methanol after reaction with DPPH solution.
The ability of dendrobium nobile polysaccharides to scavenge DPPH free radicals is plotted with the experimental concentration of a sample as the abscissa and the corresponding free radical scavenging rate as the ordinate. The results are shown in fig. 3, and the dendrobium nobile polysaccharide has good activity of eliminating DPPH free radicals.
