本发明属于微生物发酵应用领域,尤其涉及一种温经驱寒的益生菌发酵功能食品及其制备方法。
背景技术
痛经是指女性在经期或经期前后出现下腹疼痛,可伴随手脚冰凉、腰骶部酸痛、恶心呕吐、晕厥等,是妇科常见病。随着现代社会饮食文化以及观念的改变,很多女性喜食寒食或感受寒邪等多种原因,导致寒客冲任,与血相搏,致子宫冲任感寒,气血失畅,不通则痛。其临床表现为经行少腹绞痛且有定处,发冷或按之有积块,经色黑夹血块,块下则痛减,常伴月经愆期,舌黯淡、边有瘀斑,苔白润,脉紧或涩;虚寒则表现腹胀甚于痛,喜按喜揉喜暖。实寒证治以温经止痛、活血祛瘀。传统中医治疗方法主要包括灌肠、针刺、穴位贴敷、中药内服、中药熏蒸等方式,而针对温经驱寒、活血化瘀食补调理的方法并不多,主要有以下几种:

上述现有技术公开的针对女性温经驱寒、活血化瘀的食品多是各种天然食品提取物和各营养素复配而成,制备方法一般为简单的加工混合,采用益生菌固态发酵制备此类功能食品未见报道。
发明内容
本发明的目的在于针对现有技术的不足,提供一种利用益生菌固态分段发酵小扁豆、大豆、当归、肉桂、干姜、小茴香等原料生产的功能性食品,所述功能性食品主要具有温经驱寒、活血化瘀、美容养颜等功效。
为实现上述目的,本发明的技术方案如下:
一种温经驱寒的益生菌发酵功能食品,是以小扁豆、大豆、当归、肉桂、干姜、小茴香的混合物作为发酵基质,采用罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌进行固态分段发酵后真空冷冻干燥而得。
所述温经驱寒的益生菌发酵功能食品的制备方法,包括如下步骤:
(1)选取优质的小扁豆、大豆、当归、肉桂、干姜、小茴香分别磨粉过100目筛;
(2)配制活化培养基:每100mL水中添加10g大豆粉;灭菌条件:121℃,20min;
(3)配制发酵培养基:将小扁豆粉、大豆粉、当归粉、肉桂粉、干姜粉、小茴香粉混合均匀,混合物与水按照质量比1:1.5-2.0配制固态发酵培养基,置于固态发酵罐中;灭菌条件:121℃,20min;
(4)菌种活化:罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌的甘油管分别按1%(v/v)接种于活化培养基进行活化,pH自然,控制温度为37℃;分别活化24h后,按5%(v/v)的接菌量再次转接活化培养基进行二次活化,再活化24h后得二级种子液;
(5)益生菌固态分段发酵:
①将枯草芽孢杆菌与凝结芽孢杆菌的二级种子液接入固态发酵罐中,维持温度37℃,通气量60L/min,发酵10-18h;
②待第①步发酵完成后,将乳酸片球菌与植物乳杆菌的二级种子液接入固态发酵罐中,维持温度37℃,密闭发酵12-24h;
③待第②步发酵完成后,将动物双歧杆菌、青春双歧杆菌、长双岐杆菌的二级种子液接入固态发酵罐中,维持温度37℃,密闭发酵18-36h;
④待第③步发酵完成后,将副干酪乳杆菌、罗伊氏乳杆菌的二级种子液接入固态发酵罐中,维持温度37℃,密闭发酵24-48h;
(6)发酵结束后经真空冷冻干燥,即得益生菌发酵功能食品。
所述小扁豆、大豆、当归、肉桂、干姜、小茴香均为市售。
优选的,所述小扁豆粉:大豆粉:当归粉:肉桂粉:干姜粉:小茴香粉的质量比为3:6:6:3:2:1。
优选的,配制固态发酵培养基时所述混合物与水的质量比为1:1.8。
优选的,所述枯草芽孢杆菌、植物乳杆菌、副干酪乳杆菌、罗伊氏乳杆菌、乳酸片球菌、长双歧杆菌、青春双歧杆菌、动物双歧杆菌、凝结芽孢杆菌的接种量分别为106-107CFU/g固态发酵培养基。
优选的,所述真空冷冻干燥的条件为真空冷冻干燥24h,至含水量不大于12%。
优选的,所述植物乳杆菌为IOB602,已于2018年6月29日保藏于中国微生物菌种保藏管理委员会普通微生物中心,地址:北京市朝阳区北辰西路1号院3号中国科学院微生物研究所,邮编100101,分类命名:Lactobacillus plantarum,保藏编号为CGMCC No.16021。此株植物乳杆菌具有较好的抗胃酸、抗胆盐的能力,在pH 3时2h存活率86.05%,0.3%胆盐4h存活率78.04%,并且具有广泛的抗真菌性,不需要添加额外的防腐剂,也可以延长饮料和食品保藏时间。

有益效果:
小扁豆籽粒含蛋白质约25%,脂肪0.7%,碳水化合物60%及多种维生素和矿物质营养元素。可以提供蛋白质和能够降低胆固醇的可溶纤维,它的含铁量是其他豆类的两倍;小扁豆中维生素B和叶酸的含量也较高,叶酸对女性非常重要,可以降低胎儿畸形率;深色扁豆里的色素有抗氧化剂的作用,可以预防心脏病和癌症,抗衰老。
大豆含有丰富的蛋白质、大豆异黄酮,植物固醇、大豆低聚糖等,可增强机体免疫、防止血管硬化、具有降血糖、抗氧化等作用。
当归既能补血,又能活血,既可通经,又能活络。凡妇女月经不调,痛经,血虚闭经,面色萎黄,衰弱贫血,子宫出血,产后瘀血,例经(月经来潮时,出现口鼻流血)等妇女的常见病,都可以用当归治疗。
肉桂有温中补肾、散寒止痛功能,治腰膝冷痛,虚寒胃痛,慢性消化不良,腹痛吐泻,受寒经闭。
干姜味辛性热,可温中散寒,回阳通脉,温肺化饮。
小茴香味辛性温,小茴香可散寒止痛,理气和胃。

本发明所述一种益生菌固态分段发酵的温经驱寒、活血化瘀、美容养颜功能性食品,既保存了天然药食同源原有营养价值,又具有益生菌发酵制品的营养保健作用,口感独特。该食品通过益生菌固态分段发酵既使不同特性益生菌彼此协调,共同繁殖,达到微生态制剂的标准,又能将天然原料中含有的大分子物质通过益生菌降解为更易吸收的小分子物质。更重要的是,该食品生产过程节能环保、无废水废气废料产生,易于实现大规模的生产。
具体实施方式
下面通过具体的实施方案叙述本发明。除非特别说明,本发明中所用的技术手段均为本领域技术人员所公知的方法。另外,实施方案应理解为说明性的,而非限制本发明的范围,本发明的实质和范围仅由权利要求书所限定。对于本领域技术人员而言,在不背离本发明实质和范围的前提下,对这些实施方案中的物料成分和用量进行的各种改变或改动也属于本发明的保护范围。
实施例1:一种益生菌固态分段发酵的温经驱寒功能性食品及其制备方法
1、原料配比的确定
(1)选取优质的小扁豆、大豆、当归、肉桂、干姜、小茴香分别磨粉过100目筛;
(2)配制活化培养基:每100mL水中添加10g大豆粉;灭菌条件:121℃,20min;
(3)配制发酵培养基:将小扁豆粉、大豆粉、当归粉、肉桂粉、干姜粉、小茴香粉按一定比例混合均匀,混合物与水按照质量比1:1.5配制固态发酵培养基,置于6T固态发酵罐中灭菌备用;灭菌条件:121℃,20min;
(4)菌种活化:罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌的甘油管分别按1%(v/v)接种于活化培养基进行活化,pH自然,控制温度为37℃;分别活化24h后,按5%(v/v)的接菌量再次转接活化培养基进行二次活化,再活化24h后得二级种子液;
(5)益生菌固态分段发酵:
①将活化完成的枯草芽孢杆菌与凝结芽孢杆菌通过接种管路接入6T固态发酵罐中,使每克发酵基质中枯草芽孢杆菌的接种量为106-107CFU/g,凝结芽孢杆菌的接种量为106-107CFU/g,维持温度37℃,通气量60L/min,发酵10h;
②待第①步发酵完成后,将活化好的乳酸片球菌与植物乳杆菌通过接种管路接入6T固态发酵罐中,使每克发酵基质中乳酸片球菌的接种量为106-107CFU/g,植物乳杆菌的接种量为106-107CFU/g,维持温度37℃,密闭发酵12h;
③待第②步发酵完成后,将活化好的动物双歧杆菌、青春双歧杆菌、长双岐杆菌通过接种管路接入6T固态发酵罐中,使每克发酵基质中动物双岐杆菌的接种量为106-107CFU/g,青春双歧杆菌的接种量为106-107CFU/g,长双岐杆菌的接种量为106-107CFU/g,维持温度37℃,密闭发酵18h;
④待第③步发酵完成后,将活化好的罗伊氏乳杆菌、副干酪乳杆菌通过接种管路接入6T固态发酵罐中,使每克发酵基质中罗伊氏乳杆菌的接种量为106-107CFU/g,副干酪乳杆菌的接种量为106-107CFU/g,维持温度37℃,密闭发酵24h;
(6)发酵结束后,分别对发酵样品中的益生菌活菌数进行检测,结果见表1;
所述小扁豆粉、大豆粉、当归粉、肉桂粉、干姜粉、小茴香粉的原料配比标号:组合一:6:5:4:3:2:1,组合二:6:5:6:5:3:1,组合三:3:6:6:3:2:1,组合四:6:6:4:1:2:1,组合五:3:5:5:4:2:2,均为质量比。

由表1可知,原料的最优配比为组合三,即为小扁豆粉:大豆粉:当归粉:肉桂粉、干姜粉:小茴香粉质量比为3:6:6:3:2:1,发酵后总活菌数达到1.9×1010CFU/g。
2、接种量的确定
(1)选取优质的小扁豆、大豆、当归、肉桂、干姜、小茴香分别磨粉过100目筛;
(2)配制活化培养基:每100mL水中添加10g大豆粉;灭菌条件:121℃,20min;
(3)配制发酵培养基:将小扁豆粉、大豆粉、当归粉、肉桂粉、干姜粉、小茴香粉按一定比例混合均匀,混合物与水按照质量比1:1.5配制固态发酵培养基,置于6T固态发酵罐中灭菌备用;灭菌条件:121℃,20min;
(4)菌种活化:罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌的甘油管分别按1%(v/v活化培养基的体积比)接种于活化培养基进行活化,pH自然,控制温度为37℃;分别活化24h后,按5%(v/v)的接菌量再次转接活化培养基进行二次活化,再活化24h后得二级种子液;
(5)益生菌固态分段发酵:
①将活化完成的枯草芽孢杆菌与凝结芽孢杆菌通过接种管路接入6T固态发酵罐中,维持温度37℃,通气量60L/min,发酵10h;
②待第①步发酵完成后,将活化好的乳酸片球菌与植物乳杆菌通过接种管路接入6T固态发酵罐中,维持温度37℃,密闭发酵12h;
③待第②步发酵完成后,将活化好的动物双歧杆菌、青春双歧杆菌、长双岐杆菌通过接种管路接入6T固态发酵罐中,维持温度37℃,密闭发酵18h;
④待第③步发酵完成后,将活化好的罗伊氏乳杆菌、副干酪乳杆菌通过接种管路接入6T固态发酵罐中,维持温度37℃,密闭发酵24h;
(6)发酵完毕后,分别对发酵样品中的益生菌活菌数进行检测;
所述益生菌固态分段发酵时各菌的接种量分别均为:1×106CFU/g、5×106CFU/g、1×107CFU/g、5×107CFU/g四组,结果见表2。

3、料水比的确定

(4)菌种活化:罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌的甘油管分别按1%(v/v)接种于活化培养基进行活化,pH自然,控制温度为37℃;分别活化24h后,按5%(v/v)的接菌量再次转接活化培养基进行二次活化,再活化24h后得二级种子液;

(6)发酵结束后,分别对发酵样品中的益生菌活菌数进行检测,结果见表3。

由表3可知,配制固态发酵培养基的最佳料水比为1:1.8,发酵后益生菌活菌数最高为6.9×109CFU/g。
4、发酵时间的确定
(1)选取优质的小扁豆、大豆、当归、肉桂、干姜、小茴香分别磨粉过100目筛;
(2)配制活化培养基:每100mL水中添加10g大豆粉;灭菌条件:121℃,20min;
(3)将小扁豆粉、大豆粉、当归粉、肉桂粉、干姜粉、小茴香粉按质量比3:6:6:3:2:1均匀混合,得到混合物与水按质量比1:1.8配制固态发酵培养基,置于6T固态发酵罐中灭菌备用;灭菌条件:121℃,20min;
(4)菌种活化:罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌的甘油管分别按1%(v/v)接种于活化培养基进行活化,pH自然,控制温度为37℃;分别活化24h后,按5%(v/v)的接菌量再次转接活化培养基进行二次活化,再活化24h后得二级种子液;
(5)益生菌固态分段发酵:

④待第③步发酵完成后,将活化好的罗伊氏乳杆菌、副干酪乳杆菌通过接种管路接入6T固态发酵罐中,维持温度37℃,密闭发酵24-48h;
以上①-④步各菌接种量均为每克固态培养基中1×107CFU/g;
(6)具体发酵时间的组合方案见表4;发酵结束后,分别对发酵样品中的益生菌活菌数进行检测,结果见表5。

由表5可知,最佳的分段发酵时间为组合三,发酵后活菌数为4.9×1010CFU/g,即枯草芽孢杆菌与凝结芽孢杆菌发酵12h;乳酸片球菌与植物乳杆菌密闭发酵24h;动物双歧杆菌、青春双歧杆菌、长双岐杆菌密闭发酵36h;罗伊氏乳杆菌、副干酪乳杆菌密闭发酵24h。
实施例2:益生菌固态分段发酵的温经驱寒功能性食品生物活性成分测定
利用益生菌固态分段发酵的温经驱寒功能性食品根据接入菌种的种类、顺序不同,产生了不同的代谢产物,代谢产物的种类更多、活性更高,可以产生协同效应,更好的发挥功效。
①益生菌分段固态发酵接入纳豆芽孢杆菌和凝结芽孢杆菌后,在37℃的环境中,通气发酵12h后,利用高效液相法测定金雀异黄酮、大豆异黄酮苷元、雌马酚。色谱柱为Hypersil ODS2(C18)200mm×4.6mm,5μm,有机相为甲醇+乙酸0.1%(v/v),水相为过膜水+乙酸0.1%(v/v),柱温为27℃,检测波长为260nm,流速为1mL/min进样量为20μL,结果见表6。利用液相色谱法测定γ-生育酚。采用Agilent 1100系列正相液相色谱(NP-HPLC)进行分析,使用荧光检测器(FLD),检测器设置为λexc=290nm,λemis=330nm,10μL微量进样。液相色谱分析柱为silica 5μL柱(250×4.6mm,Phenomenex,Torrance,CA,USA)。流动相为MTBE-正己烷(10:90,v/v),洗脱流速为1mL/min,洗脱时间25min,结果见表6。
②接入乳酸片球菌和植物乳杆菌后,在37℃的环境中,密闭发酵24h后,用比色法测定总类胡萝卜素含量。参照Ndolo等报道的方法,稍加改动后用96孔酶标板快速测定。在96孔板的每孔中,依次加入稀释至合适倍数的小扁豆脂溶性提取液,用UV-Vis多功能酶标仪在450nm下测定吸光度(EL 340,Bio-Tek Instruments,Inc.Winooski,VT,USA),结果见表6。利用高效液相色谱法测定铁离子的含量。以试剂2-2-喹啉-5-二乙氨基酚为柱前衍生试剂,内含2%醋酸甲醇溶液和PH4.0醋酸-醋酸钠缓冲溶液梯度洗脱为流动相,WatersNova-Pak-C18液相色谱柱,二极管矩阵检测器铁元素含量,结果见表6。参照Li等报道的方法测定DPPH清除率,将其稍加改动后应用于96孔酶标板中。Trolox和DPPH用7%的甲基化-β-环糊精溶液(randomly-methylated-β-cyclodextrin,RMCD)溶解。在酶标板的每孔中加入280μL浓度为65μM的DPPH溶液,加入20μL Trolox标准溶液或发酵样品提取物。室温避光反应6h后,于540nm下用UV-Vis可见多功能酶标仪测定其吸光度(EL 340,Bio-TekInstruments,Inc.,Winooski,VT,USA),结果见表6。
③接入长双歧杆菌、青春双歧杆菌、动物双歧杆菌,在37℃的环境中,密闭发酵36h,利用高效液相色谱法测定6-姜酚。色谱柱:Agilent ZORBAX SB-C18柱(4.6mm×150.0mm,5μm);流动相:乙腈-甲醇-水(40∶5∶55);检测波长:280nm;流速:1.0m L/min;柱温:25℃;进样量:10μL。利用高效液相色谱法测定阿魏酸。Waters X-Bridge C18(250mm×4.6mm,5μm)色谱柱,以乙腈-0.1%磷酸水溶液为流动相,流速为0.5m L·min-1,检测波长为280nm,柱温25℃,进样体积为10μL,结果见表6。
④接入罗伊氏乳杆菌和副干酪乳杆菌,在37℃的环境中,密闭发酵12h后,利用火焰原子吸收法测定矿物元素。利用苯酚-硫酸法测定多糖含量:采用紫外分光光度计于487nm处测其吸光度值A,根据回归方程计算当归多糖含量(以葡萄糖量计算),结果见表6。利用AACC32-07法测定总膳食纤维。称10g样品溶于1000ml蒸馏水中,离心干燥后,在每份样品中,加入预热至60℃的95%乙醇225mL,乙醇与样品的体积比为4:1。室温下沉淀1h。用15mL78%乙醇将硅藻土湿润和重新分布在已称重的坩埚中。用适度的抽力把坩埚中的硅藻土吸到玻板上。酶解过滤,用78%乙醇和刮勺移所有内容物微粒到坩埚中。抽真空,分别用15mL的78%乙醇,95%乙醇和丙酮冲洗残渣各2次,将坩埚内的残渣抽干后在105℃烘干过夜。将坩埚置干燥器中冷却至室温。坩埚重量,包括膳食纤维残渣和硅藻土,精确称至0.1mg。减去坩埚和硅藻土的干重,计算残渣重。结果见表6。

由表6可知,一种益生菌固态分段发酵的温经驱寒功能性食品具有活血化於、驱寒止痛的功效,分析原因如下:与未发酵的食品相比,经过益生菌固态分段发酵的小扁豆、大豆、干姜、当归、肉桂、小茴香等原料中的大分子物质被益生菌降解代谢为具有更高生物活性的小分子物质,例如各种糖苷类物质被代谢为苷元类物质,不同种类的多酚含量明显增加,多糖类物质含量有效提高等等,这些物质构成了该发酵食品的基本物质基础。大豆中金雀异黄酮、大豆异黄酮苷元的增加和雌马酚产生,可有效促进女性雌激素的产生,调节女性因雌激素下降造成的各种内分泌问题,如绝经、月经不规律、更年期综合征等;小扁豆中γ-生育酚、总类胡萝卜素含量的增加,可帮助女性合成维生素E,降低缺乏维生素E的风险;小扁豆中铁离子含量的提高可增加机体的造血功能,预防女性缺铁性细胞贫血,有效改善女性经期量少的问题;干姜中有效成分6-姜酚的增加,可以镇痛抗炎,暖宫散寒、缓解痛经;当归中阿魏酸的增加能够刺激造血干细胞的增殖分化能力,增强骨骼造血功能;当归多糖为益生菌产促进因子,同时可调节免疫,提升机体造血功能;小茴香中小茴香酮的增加,可增强输卵管、子宫内膜、子宫肌层、皮层及卵巢功能,降低妇科疾病的发生概率;同时,上述所有原料中的膳食纤维均可润肠通便,促进肠道微生态平衡,排除身体毒素,达到美容养颜的功效,多酚类物质还兼具美白功效。
实施例3:益生菌固态分段发酵的温经驱寒功能性食品的补血作用
采用环磷酰胺(CTX)与乙酰苯肼(APH)合用建立小鼠血虚模型,以胸腺指数(TI)、脾指数(SI)、红细胞数(RBC)、白细胞数(WBC)、板数(PLT)、血红蛋白数(HCT)、红细胞压积(HGB)的变化为观察指标,比较益生菌固态分段发酵的温经驱寒功能性食品的补血作用,结果见表7。

由表7可知,益生菌固态分段发酵的温经驱寒功能性食品具有补血作用。外周血常规是观察血细胞变化最常用的检测指标,也是血虚证动物的观察指标,血液中的红细胞数、白细胞数、血小板数、血红蛋白数以及红细胞比容变化可以反映动物模型造血功能状态和药物影响。胸腺、脾是体内重要的免疫器官,当感染或损伤时脾指数、胸腺指数随之发生明显的变化,胸腺、脾指数通常可以作为反映免疫功能变化的指标。本实验中,造模后小鼠胸腺萎缩,脾脏出现代偿性地肿大,血液中的红细胞数、白细胞数、血小板数、血红蛋白数以及红细胞比容均降低,而益生菌固态分段发酵的温经驱寒功能性食品能明显改善以上状况。综合分析结果得知,益生菌固态分段发酵的温经驱寒功能性食品具有补血作用。
实施例4:益生菌固态分段发酵的温经驱寒功能性食品的活血作用
以皮下注射盐酸肾上腺素(Adr)结合冰水浴建立急性大鼠血瘀模型,以血浆粘度(PV)、全血粘度(WBV)、凝血酶原时间(PT)、凝血酶时间(TT)、活化部分凝血活酶时间(APTT)、纤维蛋白原含量(FIB)为观察指标,比较产品活血的作用,结果见表8和表9。

由表8和表9综合分析可知,血液粘度是衡量血液流动性的综合指标,是决定血液流变的重要因素,凝血四项是反应凝血系统最常用的筛选试验,本实验中,造模后,大鼠全血高切、低切、中切粘度及血浆粘度都增大,APTT、TT、PT减小,益生菌固态分段发酵的温经驱寒功能性食品均能降低血液粘度,延长APTT、TT、PT时间,改善血液循环,具有活血效果。
实施例5:益生菌固态分段发酵的温经驱寒功能性食品对痛经的调理作用
将118例寒凝血瘀型原发性痛经患者随机分为实验组60例和对照组58例。对照组予温经散寒汤,实验组采用益生菌固态分段发酵的温经驱寒功能性食品,一日3次,每次10g,随三餐食用,食用3个月。食疗后观察痛经持续时间改善情况,对比分析作用,如表10所示。

由表10可知,食用益生菌固态分段发酵的温经驱寒功能性食品对痛经时间有显著改善,这是因为中医学认为,经期产后,感受寒邪,或过食寒凉生冷,寒客冲任,与血相搏,以致子宫、冲任气血失畅。经前、经期气血下注冲任,子宫气血更加壅滞,“不通则痛”。若经前、经期冒雨、涉水,或久居湿地,则发为寒凝血瘀证。因此,寒凝血瘀型原发性痛经的治疗之法主要在于温经散寒,化瘀止痛,使冲任气血运行流畅,所以益生菌固态分段发酵的温经驱寒功能性食品对凝血瘀型原发性痛经有显著的改善作用。

1.一种温经驱寒的益生菌发酵功能食品,其特征在于,所述温经驱寒的益生菌发酵功能食品是以小扁豆、大豆、当归、肉桂、干姜、小茴香的混合物作为发酵基质,采用罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌进行固态分段发酵后真空冷冻干燥而得。
2.如权利要求1所述温经驱寒的益生菌发酵功能食品,其特征在于,所述植物乳杆菌的保藏编号为CGMCC No.16021;所述动物双歧杆菌的保藏编号为CGMCC No.16028;所述副干酪乳杆菌的保藏编号为CGMCC No.16022;所述青春双歧杆菌的保藏编号为CGMCCNo.16026;所述凝结芽孢杆菌的保藏编号为:CGMCC No.16025;所述枯草芽孢杆菌的保藏编号为CGMCC No.16024;所述罗伊氏乳杆菌的保藏编号为CGMCC No.16023;所述长双歧杆菌的保藏编号为CGMCC No.16027;所述乳酸片球菌的保藏编号为CGMCC No.16077。
3.如权利要求2所述温经驱寒的益生菌发酵功能食品,其特征在于,所述小扁豆:大豆:当归:肉桂:干姜粉:小茴香粉的质量比为3:6:6:3:2:1。
4.权利要求3所述温经驱寒的益生菌发酵功能食品的制备方法,包括如下步骤:
(1)选取优质的小扁豆、大豆、当归、肉桂、干姜、小茴香分别磨粉过100目筛;
(2)配制活化培养基:每100mL水中添加10g大豆粉;灭菌条件:121℃,20min;
(3)配制发酵培养基:将小扁豆粉、大豆粉、当归粉、肉桂粉、干姜粉、小茴香粉混合均匀,混合物与水按照质量比1:1.5-2.0配制固态发酵培养基,置于固态发酵罐中;灭菌条件:121℃,20min;
(4)菌种活化:罗伊氏乳杆菌、长双歧杆菌、乳酸片球菌、植物乳杆菌、动物双歧杆菌、副干酪乳杆菌、青春双歧杆菌、凝结芽孢杆菌和枯草芽孢杆菌的甘油管分别按1%(v/v)接种于活化培养基进行活化,pH自然,控制温度为37℃;分别活化24h后,按5%(v/v)的接菌量再次转接活化培养基进行二次活化,再活化24h后得二级种子液;

③待第②步发酵完成后,将动物双歧杆菌、青春双歧杆菌、长双岐杆菌的二级种子液接入固态发酵罐中,维持温度37℃,密闭发酵18-36h;
④待第③步发酵完成后,将副干酪乳杆菌、罗伊氏乳杆菌接入固态发酵罐中,维持温度37℃,密闭发酵24-48h;
(6)发酵结束后经真空冷冻干燥,即得益生菌发酵功能食品。
5.如权利要求4所述温经驱寒的益生菌发酵功能食品的制备方法,其特征在于,配制固态发酵培养基时所述混合物与水的质量比为1:1.8。
6.如权利要求4所述温经驱寒的益生菌发酵功能食品的制备方法,其特征在于,所述枯草芽孢杆菌、植物乳杆菌、副干酪乳杆菌、罗伊氏乳杆菌、乳酸片球菌、长双歧杆菌、青春双歧杆菌、动物双歧杆菌、凝结芽孢杆菌的接种量分别为106-107CFU/g固态发酵培养基。
7.如权利要求4所述温经驱寒的益生菌发酵功能食品的制备方法,其特征在于,所述真空冷冻干燥的条件为:真空冷冻干燥24h,至含水量不大于12%。
