本发明涉及新型牙科器具清洁(cleansing)组合物,特别是无醇组合物。本发明还涉及浸渍清洁组合物的牙科器具擦巾(dental appliance wipe)及其在清理(cleaning)可拆卸牙科器具中的用途。
发明背景
许多可拆卸牙科器具佩戴者需要频繁的全天清理。清理在进食后特别普遍,此时佩戴者常常经历食物颗粒卡在牙科器具下,这可造成刺激和不适。为了除去食物颗粒,佩戴者通常使用牙刷或用水冲洗牙科器具。这两种方法都具有需要水并且可能需要取下牙科器具的缺点,这在离家时不方便。此外,已经发现用牙膏反复刷洗义齿(dentures)造成义齿丙烯酸树脂(denture acrylic)的划伤(Kiesow等人, Journal of Prosthetic Dentistry,2016, Vol. 115, No. 2, 第189-198页)。

定义
术语“清理(clean)”、“清洁(cleansing)”或“清理(cleaning)”在本文中可互换使用以指通过本发明的制剂的擦拭和/或清理作用加上口气清新功能去除食物颗粒、污渍、微生物(microbes/microorganisms)和其它口腔碎片。
本文所用的术语“牙科器具”是指义齿或局部义齿、假牙(artificial teeth)、可拆卸的正畸桥和托牙板(上牙和下牙类型)、正畸保持器和器具、保护性的护牙托(protective mouthguards)、用于防止夜磨牙症和/或颞下颌关节(TMJ)病症的夜用护板(nightguards)等。
本文所用的术语“牙科器具清洁剂”是指在口腔外用于清理牙科器具的制剂。
本文所用的术语“微乳液”是指与助表面活性剂结合的油和表面活性剂的澄清、热力学稳定的均质液体混合物。微乳液可不含水或为稀释形式。一旦稀释,微乳液具有均一的亚微米尺寸粒子。
本文所用的术语“擦巾”是指不溶性基质,包括纺织和无纺基质以及海绵,例如纤维素基质。无纺基质是优选的。无纺是指该层由没有编织成织物而是成型为通过机械、热或化学缠结纤维或长丝而结合在一起的片或网结构的纤维构成。该不溶性基质优选是粘胶聚酯混纺物、粘胶聚丙烯混纺物或粘胶聚酯聚丙烯混纺物;更优选粘胶聚丙烯混纺物;更优选65%粘胶-35%聚丙烯混纺物。不可溶基质可获自多种多样的商业来源。根据本发明的优选擦巾是NOVONETTE™ Thermal Bond,一种可获自Suominen的擦巾。不可溶基质不限于任何形式、特定模式(pattern)、设计或几何。其可以是平纹、图案化或压花的并且其也可以是任何尺寸或形状。本发明的擦巾优选为11cm x 15cm或15cm x 20 cm。该不可溶基质绝不能撕裂或钩破并且打算在一次性使用后处置或丢弃。
本文所用的术语“湿擦巾”是指不溶性基材,将根据本发明的牙科器具清洁组合物加载到其中和其上。一旦加载,该湿擦巾含有足量的清洁组合物以使其湿(wet)、潮湿(damp)或湿润(moistened)。也就是说,该擦巾具有每克干基质至少2.0克清洁组合物的负载系数。
本文所用的术语“脂肪酸异丙酯”是指异丙醇与合适的脂肪酸,如肉豆蔻酸或棕榈酸的酯。肉豆蔻酸异丙酯是异丙醇与肉豆蔻酸(十四烷酸)的酯,并且也被称为十四烷酸丙-2-基酯、十四烷酸1-甲基乙基酯或肉豆蔻酸异丙酯。棕榈酸异丙酯是异丙醇与棕榈酸(十六烷酸)的酯,并且也被称为十六烷酸丙-2-基酯、十六烷酸异丙酯、十六烷酸异丙基酯或十六烷酸1-甲基乙基酯。

擦巾
通过用每克干基质至少1克组合物润湿干基质,制造根据本发明的湿擦巾。优选用每克干基质至少2.0克,更优选至少3.0克且甚至更优选至少2.7克或3.5克组合物润湿干基质。
根据本发明的擦巾可独立地作为单擦巾或作为合装包(multi-pack)包装。在擦巾作为单擦巾包装时,各擦巾可包装在独立小袋中,在一盒中包括许多这样的小袋,例如在一盒中1、14、15、20、28、30或40个小袋。在擦巾作为合装包(multi-pack)包装时,许多擦巾(例如20、25、30、40、50或70个)可包装在可重复密封容器,如可使用胶粘标签或塑料盖重新关闭或密封的包装或容器内。
根据本发明的湿擦巾的包装外壳必须保护该产品以防香味和/或溶剂损失。其绝不能与该产品相互作用以对清洁组合物或包装性质造成不利影响。根据本发明的湿擦巾的包装可为任何合适的形式。该包装外壳优选包括铝阻隔层压材料和硬质可关闭盖(hard-closable cap)。该包装可为任何合适的尺寸和形状,其优选应该小并且易于携带。
实验
根据本发明的组合物的下列实施例仅意在举例说明而非限制本发明的范围。对所公开的实施方案的各种变动和修改是本领域技术人员显而易见的,并且可以不背离本发明的精神和所附权利要求书的范围而作出这样的变动和修改。
制造方法
可通过以任何方便的顺序以适当的相对量混合成分来制备根据本发明的组合物(参见下文的实施例)。优选分开形成油相和水相,且然后合并这两个相。
可通过制备合适尺寸的干不溶性基材、然后通过例如喷雾分散使清洁组合物与所述基材接触来制备根据本发明的湿擦巾。一旦制成,将湿擦巾转移到合适尺寸的可重复气密密封容器中,然后密封。

试验数据
义齿的丙烯酸树脂表面上的生物膜形成最常在不好好遵守义齿清洁规范后发生。定殖的生物膜细胞,特别是真菌属——念珠菌属的那些,被认为是念珠菌属相关的义齿性口炎的主要成因。这种感染是普遍的,发生在>65%的义齿佩戴者中。所见临床表现是与义齿的安装面接触的腭的红斑区域。
显然,良好遵守适当的义齿清洁规范是防止念珠菌属相关的义齿性口炎的关键。通常,这样的清洁方案涉及将义齿浸泡在适当的清洁剂中。另外,可以使用牙刷与专卖性的牙膏或凝胶结合刷洗义齿表面。
这一研究的目的是使用体外方法评估浸渍实施例6和实施例8的组合物的牙科擦巾对从义齿基托丙烯酸树脂上去除微生物的效力。
材料和方法
丙烯酸树脂表面上的微生物型生物膜的形成
将之前通过在37℃下在无菌人工唾液中浸泡24小时预调节的义齿丙烯酸树脂的方块(1 cm2)在37℃下浸在已制备和标准化的微生物菌剂(microbial inoculum)中72小时。在培养后,将被污染的丙烯酸树脂浸在无菌水中5分钟以除去松散附着/浮游的微生物。
使用清洁擦巾的微生物去除
为了进行标准化的“擦拭”程序,将清洁擦巾附着到牙科材料抛光仪器的表面上。这一装置具有能以50-600 rev/min的速度运行的旋转转盘。转盘的直径为300 mm,因此理想地适合大或小的试样。在这一研究中,将清洁擦巾安置在旋转转盘的表面上以使其随转盘的运动旋转。将包含压重(pressure weight)的静态臂(static arm)安装在转盘上方,并将丙烯酸树脂方块保持在固定位置并与清洁擦巾直接接触。通过保持恒速旋转和改变时间,实现不同的擦拭程度。在这些研究中,评估清洁擦巾通过在100 rev/min下擦拭60 s和/或在100 rev/min下擦拭10 s或在标准化流动下的温水冲洗去除微生物定殖的能力。对照物是未暴露于擦拭过程的丙烯酸树脂表面。
在擦拭后,将丙烯酸树脂表面上的微生物在4% (v/v)福尔马林中固定24小时。通过共焦激光扫描显微术(CLSM)检查来自各试验组的碘化丙啶染色图像。将图像转化成二进制黑和白以使用ImageJ软件测定覆盖百分比(Rasband, W.S., ImageJ, U. S. NationalInstitutes of Health, Bethesda, Maryland, USA, http://imagej.nih.gov/ij/,1997-2014)。使用SPSS Statistics (v20) (IBM)进行定量数据分析。使用Kruskal-Wallis检验和Mann-Whitney-Wilcoxon检验进行多元分析以比较成对处理组之间的定殖面积。在0.05水平下测定显著性差异。
使用不同处理进行两个研究。

Shapiro-Wilk检验显示,数据不是正态分布的(P<0.0001)。ANOVA揭示,在试验组之间存在定殖水平的显著差异(P=0.02)。使用Tukey post-hoc检验的单因素ANOVA显示,与未擦拭的对照物相比,用对比实施例A擦拭60s(P=0.012)、实施例6擦拭10 s(P=0.037)和实施例6擦拭60 s(P=0.002)发生残留定殖的显著减少。在未擦拭对照物和用参比实施例A擦拭10 s之间没有显著减少的迹象(P=0.304)。
结论
显而易见,与未擦拭对照物相比,用对比实施例A和实施例6擦巾擦拭义齿丙烯酸树脂表面60 s能够显著减少微生物。此外,使用实施例6擦巾10 s也测定微生物去除的显著减少。
这一去除研究表明,用浸渍根据本发明的组合物的擦巾擦拭义齿丙烯酸树脂10秒的物理作用提供与浸渍代表性漱口水溶液的擦巾相比改进的去除。

结论
这一研究揭示,使用实施例8擦拭被微生物污染的丙烯酸树脂10s与未擦拭对照物和水冲洗样品相比都导致显著去除微生物。

3.根据权利要求1或权利要求2的牙科器具清洁组合物,其中所述聚氧乙烯山梨糖醇酐酯选自聚山梨酯20、聚山梨酯21、聚山梨酯40、聚山梨酯60、聚山梨酯65、聚山梨酯80、聚山梨酯81、聚山梨酯85及其组合。
4.根据权利要求1至3任一项的牙科器具清洁组合物,其中第二山梨糖醇酐酯选自山梨糖醇酐单硬脂酸酯、山梨糖醇酐三硬脂酸酯、山梨糖醇酐单月桂酸酯、山梨糖醇酐棕榈酸酯、山梨糖醇酐油酸酯、山梨糖醇酐倍半油酸酯、山梨糖醇酐三油酸酯、山梨糖醇酐异硬脂酸酯及其组合。
5.根据权利要求1至4任一项的牙科器具清洁组合物,其中所述组合物进一步包含选自丙二醇、水及其混合物的溶剂。
6. 根据权利要求1至5任一项的牙科器具清洁组合物,其中当所述清洁组合物不含水作为溶剂时,脂肪酸异丙酯以10至65 % w/w的量存在。
7. 根据权利要求1至6任一项的牙科器具清洁组合物,其中当所述清洁组合物含有水作为溶剂时,脂肪酸异丙酯以3至15 % w/w的量存在。
8. 根据权利要求1至7任一项的牙科器具清洁组合物,其中当所述清洁组合物不含水作为溶剂时,聚氧乙烯山梨糖醇酐酯以10至45 % w/w的量存在。
9. 根据权利要求1至8任一项的牙科器具清洁组合物,其中当所述清洁组合物含有水作为溶剂时,聚氧乙烯山梨糖醇酐酯以0.5至4 % w/w的量存在。
10. 根据权利要求1至9任一项的牙科器具清洁组合物,其中当所述清洁组合物不含水作为溶剂时,第二山梨糖醇酐酯以10至65 % w/w的量存在。
11. 根据权利要求1至10任一项的牙科器具清洁组合物,其中当所述清洁组合物含有水作为溶剂时,第二山梨糖醇酐酯以0.5至4 % w/w的量存在。
12.根据权利要求1至11任一项的牙科器具清洁组合物,其中所述组合物进一步包含选自山梨酸钾、苯甲酸钠、苯氧基乙醇、氯化十六烷基吡啶及其组合的至少一种防腐剂。
13.浸渍如权利要求1至12任一项中所定义的组合物的牙科器具清洁擦巾。
14.如权利要求1至12任一项中所定义的牙科器具清洁组合物或如权利要求13中所定义的牙科器具清洁擦巾用于清理牙科器具的用途。
15.如权利要求1至12任一项中所定义的牙科器具清洁组合物或如权利要求13中所定义的牙科器具清洁擦巾用于清理义齿的用途。
