Abstract
本发明涉及一种富硒香菇多肽的制备方法,包括粉剂制备、酶解、醇沉、浓缩干燥等步骤。本发明使用以硒酸精氨酸为硒源培育而得的富硒香菇为原料,无机硒残留量少,安全性高;利用酶解技术将香菇子实体中的大分子蛋白质降解为小分子多肽,与其他组分分离;通过醇沉除去可溶性杂质,提高多肽含量。本发明得到的富硒香菇多肽硒含量高,无机硒残留量低,香气浓郁,具有易消化吸收、抗原性低、抗氧化、增强人体免疫力等多种生理功能,可作为硒营养强化剂添加到食品中,也可应用于保健品和药品的开发,前景广阔。

技术领域
本发明涉及一种含硒多肽的制备方法,具体地,本发明涉及一种富硒香菇多肽的制备方法。
背景技术
多肽是生物体内的有效活性成分,具有多种营养保健作用。香菇多肽是香菇蛋白的水解产物,氨基酸含量丰富且种类齐全,还富含在谷物蛋白和蔬菜蛋白中缺乏的亮氨酸和赖氨酸。富硒香菇多肽,富含有机硒,从富硒香菇中提取而得。富硒香菇与蛋白相比在体内更易消化,抗原性低,具有许多独特的生理功能特性和理化性质,更易添加到食品中,用途更广泛。富硒香菇多肽进一步增加了香菇多肽的营养保健价值,是一种安全、高效的有机硒保健品。
硒是人体必需的微量元素之一,许多疾病的发生与硒的摄入量偏低有关,在膳食中适当增加硒的摄入,可以提高人体对某些疾病的抵御能力。硒是谷胱甘肽过氧化物酶的活性中心,是体内重要的抗氧化酶,能清除体内自由基和确保细胞膜的完整性,因此在预防肿瘤、抗衰老和预防心血管等疾病方面起着非常重要的作用。
目前,食用菌活性肽提取的研究较少。含硒多肽与普通的多肽提取不同,一方面要减少硒的损失,另一方面要保证多肽的活性。水提醇沉法是多肽的制备方法之一,这种方法多肽得率不高。酶解法也常用于多肽制备,这种方法的杂质较多。本方法,先将香菇蛋白酶解增加多肽含量,再利用醇沉法除去可溶性杂质提高多肽含量,不仅多肽的纯度较高,而且硒的损失少,许多含硒物质保留了下来。
发明内容
本发明提供了一种富硒香菇多肽的制备方法,利用酶解技术将蛋白质与其他组分分离降解为小分子多肽,通过醇沉进一步纯化多肽,利用真空冷冻技术进行干燥。
本发明实现目的的技术方案是:一种富硒香菇多肽的制备方法,包括以下步骤:
(1)取新鲜的富硒香菇子实体,在温度60℃条件下,鼓风干燥10~16h,然后粉碎成粒径为小于75μm的富硒香菇粉剂;
(2)酶解:取重量份数为100~200份的步骤(1)中富硒香菇粉剂加入到重量份数为1000份的pH8.0稀氢氧化钠溶液中,混合均匀,然后加入重量份数为10份的碱性蛋白酶搅拌酶解,酶解温度55℃,搅拌速率50~70rpm,酶解4~8h后,过滤除杂,得滤液;
(3)醇沉:向步骤(2)中的滤液中加入3倍~4倍体积的无水乙醇,在温度4℃条件下,沉淀10~14h后,5000g冷冻离心20min,除去可溶性杂质,取上清液;
(4)浓缩干燥:将步骤(3)中的上清液在温度50~60℃条件下,以30~40rpm的转速,旋转蒸发浓缩8~10倍,浓缩液在温度-20℃冰柜冷冻24h,然后在真空度20Pa,温度-75℃的条件下,连续真空冷冻干燥24h,得富硒香菇多肽冻干粉。
其中富硒香菇粉剂的硒含量为20~80mg/kg。
其中富硒香菇多肽冻干粉硒含量为50~200mg/kg,小分子多肽的含量超过80%。
本发明的优点和积极效果是:本发明使用以硒酸精氨酸为硒源培育而得的富硒香菇为原料,无机硒残留量少,安全性高;本发明所得富硒香菇多肽硒含量高,杂质少,且香气浓郁,具有易消化吸收、抗原性低、抗氧化、增强人体免疫力等多种生理功能,可作为硒营养强化剂添加到食品中,也应用于保健品和药品的开发,前景广阔。
具体实施方式
下面通过具体实施例对本发明作进一步详述,以下实施例只是描述性的,不是限定性的,不能以此限定本发明的保护范围。
实施例1:选择以硒酸精氨酸为硒源培育得到的新鲜富硒香菇,在温度60℃条件下,鼓风干燥12h,然后粉碎成粒径小于75μm的富硒香菇粉剂;称取重量份数为100份的富硒香菇粉剂加入到重量份数为1000份的pH8.0稀氢氧化钠溶液中,混合均匀,然后加入重量份数为10份的碱性蛋白酶搅拌酶解,酶解温度55℃,搅拌速率60rpm,酶解5h后,过滤除杂,得滤液;向滤液中加入4倍体积的无水乙醇,4℃沉淀12h,5000g冷冻离心20min,除去可溶性杂质,取上清。将上清在55℃,以40rpm的转速,旋转蒸发浓缩10倍,浓缩液在-20冰柜冷冻24h后,在真空度20Pa,温度-75℃的条件下,连续真空冷冻干燥24h,得富硒香菇多肽冻干粉。
其中富硒香菇粉剂的硒含量为50mg/kg。
其中富硒香菇多肽冻干粉硒含量为105mg/kg,小分子多肽的含量83%。
实施例2:选择以硒酸精氨酸为硒源培育得到的新鲜富硒香菇,在温度60℃条件下,鼓风干燥12h,然后粉碎成粒径小于38μm的富硒香菇粉剂;称取重量份数为150份的富硒香菇粉剂加入到重量份数为1000份的pH8.0稀氢氧化钠溶液中,混合均匀,然后加入重量份数为10份的碱性蛋白酶搅拌酶解,酶解温度55℃,搅拌速率60rpm,酶解6h后,过滤除杂,得滤液;向滤液中加入4倍体积的无水乙醇,4℃沉淀12h,5000g冷冻离心20min,除去可溶性杂质,取上清。将上清在55℃,以40rpm的转速,旋转蒸发浓缩10倍,浓缩液在-20冰柜冷冻24h后,在真空度20Pa,温度-75℃的条件下,连续真空冷冻干燥24h,得富硒香菇多肽冻干粉。
其中富硒香菇粉剂的硒含量为69mg/kg。
其中富硒香菇多肽冻干粉硒含量为146mg/kg,小分子多肽的含量89%。
Claims (5)
1.一种富硒香菇多肽的制备方法,其特征在于包括以下步骤:
(1)取新鲜的富硒香菇子实体,在温度60℃条件下,鼓风干燥10~16h,然后粉碎成粒径为小于75μm的富硒香菇粉剂;
(2)酶解:取重量份数为100~200份的步骤(1)中富硒富硒香菇粉剂加入到重量份数为1000份的pH8.0稀氢氧化钠溶液中,混合均匀,然后加入重量份数为10份的碱性蛋白酶搅拌酶解,酶解温度55℃,搅拌速率50~70rpm,酶解4~8h后,过滤除杂,得滤液;
(3)醇沉:向步骤(2)中的滤液中加入3倍~4倍体积的无水乙醇,在温度4℃条件下,沉淀10~14h后,5000g冷冻离心20min,除去可溶性杂质,取上清液;
(4)浓缩干燥:将步骤(3)中的上清液在温度50~60℃条件下,以30~40rpm的转速,旋转蒸发浓缩8~10倍,浓缩液在温度-20℃冰柜冷冻24h,然后在真空度20Pa,温度-75℃的条件下,连续真空冷冻干燥24h,得富硒香菇多肽冻干粉。
2.根据权利要求1所述的富硒香菇多肽的制备方法,其特征在于所述富硒香菇以硒酸精氨酸为硒源培育而得。
3.根据权利要求1所述的富硒香菇多肽的制备方法,其特征在于所述富硒香菇粉剂硒含量为20~80mg/kg。
4.根据权利要求1所述的富硒香菇多肽的制备方法,其特征在于所述富硒香菇多肽冻干粉硒含量为50~200mg/kg。
5.根据权利要求1所述的富硒香菇多肽的制备方法,其特征在于所述富硒香菇多肽冻干粉中小分子多肽的含量超过80%。
