Abstract
本发明属于保健食品领域,公开了一种预防并改善脂肪肝的竹荪面条及其制备方法,该面条由以下重量份组成:竹荪粉15‑20份、富铬发芽糙米粉5‑10份、荜茇提取物0.5‑1份、葫芦巴总皂苷0.5‑1份、芹菜5‑10份、山楂5‑8份、木瓜10‑12份、食盐0.5‑1份、小麦粉60‑70份。该面条以竹荪粉、富铬发芽糙米粉、荜茇提取物和葫芦巴总皂苷为主要原料,再配以芹菜、山楂、木瓜等果蔬添加至面粉中,使得制成的竹荪面条具有降酶保肝作用,能够降低肝细胞纤维化程度,减少脂肪在肝脏中的聚集,从而对脂肪肝具有一定的预防及治疗作用。

本发明涉及保健食品技术领域,具体涉及一种预防并改善脂肪肝的竹荪面条及其制备方法。
背景技术
脂肪肝是指由于各种原因引起的肝细胞内脂肪堆积过多的病变,是一种常见的肝脏病理改变,而非一种独立的疾病,脂肪性肝病正严重威胁国人的健康,成为仅次于病毒性肝炎的第二大肝病,发病率在不断升高,且发病年龄日趋年轻化。正常人肝组织中含有少量的脂肪,如甘油三酯、磷脂、糖脂和胆固醇等,其重量约为肝重量的3-5%,如果肝内脂肪蓄积太多,超过肝重量的5%或在组织学上肝细胞50%以上有脂肪变性时,就可称为脂肪肝。研究表明,高脂血症是导致脂肪肝的重要因素之一,长期大量进食高脂类的食物,在小肠生成和吸收的游离脂肪酸和甘油增加,进入血液的脂肪乳糜微粒增多,最终使肝脏摄取大量的脂肪酸合成脂肪,当脂肪在肝脏合成的量超过肝脏的转运能力时就造成脂肪的蓄积,形成脂肪肝。
目前大多数人选择采用食疗法控制血糖血脂,但在市面上对于预防及治疗脂肪肝的加工产品较少,且中国专利《一种山药高粱米面条及其制作方法》(CN201310524299.6)、《一种鲜山药面条及其制作方法》(CN20081019271.5)、《一种山药芋头玉米面条及其制作方法》(CN201310526645.4)这些发明所制备的面条均能减少皮下脂肪的沉积,防止肝结缔组织的萎缩,具有预防胶原性病及脂肪肝发生的作用。但这些发明所含预防及治疗脂肪肝的活性成分单一,功能性广泛,不能够很好的针对脂肪肝提出改进。
发明内容
为了解决上述问题,本发明提供了一种预防并改善脂肪肝面条及其制备方法,以竹荪粉、富铬发芽糙米粉、荜茇提取物和葫芦巴总皂苷为主要原料,再配以芹菜、山楂、木瓜等果蔬添加至面粉中,使得制成的竹荪面条具有降酶保肝作用,能够降低肝细胞纤维化程度,减少脂肪在肝脏中的聚集,从而对脂肪肝具有一定的预防及治疗作用。
本发明可以通过以下技术方案来实现:
一种预防并改善脂肪肝的竹荪面条,由以下重量份组成:竹荪粉15-20份、富铬发芽糙米粉5-10份、荜茇提取物0.5-1份、葫芦巴总皂苷0.5-1份、芹菜5-10份、山楂5-8份、木瓜10-12份、食盐0.5-1份、小麦粉60-70份。
优选地,一种预防并改善脂肪肝的竹荪面条,由以下重量份组成:竹荪粉17.5份、富铬发芽糙米粉7.5份、荜茇提取物0.75份、葫芦巴总皂苷0.75份、芹菜7.5份、山楂6.5份、木瓜11份、食盐0.75份、小麦粉65份。
优选地,所述竹荪粉的制备方法为:
a.将竹荪母种在无菌条件下接种到PDA斜面上,置于25℃恒温箱中培养7-10天,得到活化竹荪菌丝;
b.用2g蔗糖、1.5g黄豆饼粉、2g玉米粉、0.4g酵母粉、0.03g磷酸二氢钾、0.08g硫酸镁、0.01g维生素B1、水1000ml制成pH为6的液体培养基,取80ml液体培养基装入500ml三角瓶内,以接入量为1cm3将活化好的竹荪菌丝接入液体培养基中,置于25℃恒温摇床中,以120r/min的转速进行以及培养5天;
c.将一级摇瓶中的菌丝以接种量为6%接入二级摇瓶中,置于25℃恒温摇床中,以120r/min的转速进行二级培养4天;
d.取出二级培养液,用0.088mm目筛过滤,将菌丝体置于80℃烘箱中烘干至恒重,粉碎过100目筛,得到竹荪粉。
优选地,所述富铬发芽糙米粉的制备方法为:
a.取经挑选、除杂、均匀的糙米100g,漂洗干净,用1%的次氯酸钠溶液浸泡30min,然后用去离子水冲洗5次,并用70mg/L三氯化铬水溶液在25℃下浸泡10h,然后置于35℃下通气培养18h,得到富铬发芽糙米;
b.将富铬发芽糙米用清水清洗沥干后,置于50℃的热风温度下干燥至富铬发芽糙米含水量为14%,然后粉碎,得到富铬发芽糙米粉。
优选地,所述荜茇提取物的制备方法为:取荜茇粗粉250g,置于1000ml的圆底烧瓶中,加入330ml的去离子水浸泡12h,然后加热回流提取4h,过滤,取滤液减压浓缩,干燥至恒重,得到荜茇提取物。
优选地,所述葫芦巴总皂苷的制备方法为:取葫芦巴粉末100g,按料液比为1:8加入70%乙醇加热回流提取3次,每次1h,过滤,合并3次滤液,减压回收乙醇后,加入蒸馏水,调节溶液浓度为30%,然后加入D-101型大孔树脂柱上,用蒸馏水洗脱至无色后,再用4BV的70%乙醇洗脱,收集洗脱液,取洗脱液经超滤器超滤,得到超滤液,然后减压回收乙醇,浓缩干燥,得到葫芦巴总皂苷。
本发明还提供一种预防并改善脂肪肝的竹荪面条的制备方法,包括以下步骤:
(1)取山楂去核,木瓜削皮去核后,与芹菜混合,加入500ml蒸馏水,用组织匀浆器破碎后,放入榨汁机中榨汁,取榨汁液经3000r/min离心15min,取上清液即为果蔬汁;
(2)取竹笋粉、富铬发芽糙米粉、小麦粉混合过筛,放入和面机中,再加入荜茇提取物、葫芦巴总皂苷和步骤(1)的果蔬汁,搅拌均匀,和面15-20min,得到面团;
(3)将步骤(2)的面团放入熟化机中,以5-10r/min的转速熟化15-30min;
(4)将步骤(3)熟化好的面团经多级锟压和切条机切条,得到2mm的面条;
(5)将步骤(4)切好的面条放入烘房中,烘干至面条含水量14%,得到本发明预防并改善脂肪肝的竹荪面条。
本发明所述各主要功效成分作用机理如下:
竹荪具有滋补强壮、益气补脑、宁神健体、补气养阴、润肺止咳以及清热利湿等功效,能够保护肝脏,减少腹壁脂肪的积存,从而能预防脂肪肝的发生。
荜茇提取物中所含荜茇宁是调节脂类代谢的主要成分,能通过显著降低血清中的转氨酶活力,调节肝功能指标,从而有效的预防了酒精性脂肪肝的发生和发展。
铬对酯类代谢的主要作用是维持血液的正常胆固醇水平,人体如果缺铬,胰岛素的生物学活性会降低,糖耐量受损,且会引发脂类代谢紊乱,但补充铬元素后,胰岛素的活性增强,能降低主动脉上胆固醇的沉积,帮助调节脂类代谢,从而改善血脂状况;铬还能够增强脂蛋白酶和卵磷脂胆固醇酰基转移酶的活性,从而增强了高密度脂蛋白的合成作用。
发芽糙米含有GABA、膳食纤维及抗脂质氧化物质等化学成分,能够降低血清中甘油三酯和总胆固醇的含量,促进血液微循环,预防动脉粥样硬化、内脏功能障碍和癌症等,且糙米中含有丰富的镁、钾、钙和铁等人体所必需的微量元素,能提高人体的免疫功能,促进新陈代谢,防治脂肪在肝内沉积,从而具有预防脂肪肝形成的作用。
葫芦巴总皂苷能通过减轻乙醇及其代谢产物乙醛,经CYP2E1代谢过程诱导的脂质过氧化状况,从而提高机体的抗氧化能力,改善肝细胞线粒体功能,葫芦巴总皂苷还能通过升高脂联素水平,激活磷酸腺苷依赖蛋白激酶和过氧化物酶体增殖物受体-α信号转到通路,从而具有降血脂,降酶保肝,显著减少肝脏中沉积的脂质的作用,对酒精性脂肪肝具有一定的治疗作用。
芹菜、山楂、木瓜等果蔬,含有调节血脂及胆固醇含量的成分,具有一定的护肝降酶作用,能够减轻肝组织炎性病变,降低肝细胞纤维化程度,减轻脂肪在肝脏的聚集,从而具有预防脂肪肝的作用。
本发明具备以下有益效果:
本发明将竹荪添加至面粉中,能够降低面团的粘稠度,改善面团的操作性能,增强面团的筋力和面团的弹韧性,还能够将竹荪粉中的营养物质添加至面粉中,提高了面条的营养价值及功效作用。
本发明利用三氯化铬制备发芽糙米,得到富铬发芽糙米粉,能够降低血清中胆固醇和甘油三酯的含量,提高血清中高密度脂蛋白,从而能够促进TC的逆运转,减少胆固醇在外周组织和血管的沉积,抑制细胞摄入LDL,从而具有抵抗血脂升高的作用。
本发明将竹荪粉、富铬发芽糙米粉、荜茇提取物和葫芦巴总皂苷添加至面条中,使面条具有降酶保肝,减少肝脏中沉积的脂质的作用,本发明还添加了芹菜、山楂、木瓜等含调节血脂、胆固醇成分的营养果蔬,使得本发明制备的竹荪面条能够减轻肝组织炎性病变,降低肝细胞纤维化程度,减少脂肪在肝脏的聚集,从而具有预防脂肪肝形成的作用。
具体实施方式
为了使本发明实现的技术手段、创作特征、达成目的与功效易于明白了解,下面结合具体实施例,进一步阐述本发明。
实施例1
一种预防并改善脂肪肝的竹荪面条,由以下重量份组成:竹荪粉15份、富铬发芽糙米粉5份、荜茇提取物0.5份、葫芦巴总皂苷0.5份、芹菜5份、山楂5份、木瓜10份、食盐0.5份、小麦粉60份。
所述竹荪粉的制备方法为:
a.将竹荪母种在无菌条件下接种到PDA斜面上,置于25℃恒温箱中培养7天,得到活化竹荪菌丝;
b.用2g蔗糖、1.5g黄豆饼粉、2g玉米粉、0.4g酵母粉、0.03g磷酸二氢钾、0.08g硫酸镁、0.01g维生素B1、水1000ml制成pH为6的液体培养基,取80ml液体培养基装入500ml三角瓶内,以接入量为1cm3将活化好的竹荪菌丝接入液体培养基中,置于25℃恒温摇床中,以120r/min的转速进行以及培养5天;
c.将一级摇瓶中的菌丝以接种量为6%接入二级摇瓶中,置于25℃恒温摇床中,以120r/min的转速进行二级培养4天;
d.取出二级培养液,用0.088mm目筛过滤,将菌丝体置于80℃烘箱中烘干至恒重,粉碎过100目筛,得到竹荪粉。
所述富铬发芽糙米粉的制备方法为:
a.取经挑选、除杂、均匀的糙米100g,漂洗干净,用1%的次氯酸钠溶液浸泡30min,然后用去离子水冲洗5次,并用70mg/L三氯化铬水溶液在25℃下浸泡10h,然后置于35℃下通气培养18h,得到富铬发芽糙米;
b.将富铬发芽糙米用清水清洗沥干后,置于50℃的热风温度下干燥至富铬发芽糙米含水量为14%,然后粉碎,得到富铬发芽糙米粉。
所述荜茇提取物的制备方法为:取荜茇粗粉250g,置于1000ml的圆底烧瓶中,加入330ml的去离子水浸泡12h,然后加热回流提取4h,过滤,取滤液减压浓缩,干燥至恒重,得到荜茇提取物。
所述葫芦巴总皂苷的制备方法为:取葫芦巴粉末100g,按料液比为1:8加入70%乙醇加热回流提取3次,每次1h,过滤,合并3次滤液,减压回收乙醇后,加入蒸馏水,调节溶液浓度为30%,然后加入D-101型大孔树脂柱上,用蒸馏水洗脱至无色后,再用4BV的70%乙醇洗脱,收集洗脱液,取洗脱液经超滤器超滤,得到超滤液,然后减压回收乙醇,浓缩干燥,得到葫芦巴总皂苷。
上述一种预防并改善脂肪肝的竹荪面条的制备方法,包括以下步骤:
(1)取山楂去核,木瓜削皮去核后,与芹菜混合,加入500ml蒸馏水,用组织匀浆器破碎后,放入榨汁机中榨汁,取榨汁液经3000r/min离心15min,取上清液即为果蔬汁;
(2)取竹笋粉、富铬发芽糙米粉、小麦粉混合过筛,放入和面机中,再加入荜茇提取物、葫芦巴总皂苷和步骤(1)的果蔬汁,搅拌均匀,和面17.5min,得到面团;
(3)将步骤(2)的面团放入熟化机中,以7.5r/min的转速熟化22.5min;
(4)将步骤(3)熟化好的面团经多级锟压和切条机切条,得到2mm的面条;
(5)将步骤(4)切好的面条放入烘房中,烘干至面条含水量为14%,得到本发明预防并改善脂肪肝的竹荪面条。
实施例2
一种预防并改善脂肪肝的竹荪面条,由以下重量份组成:竹荪粉17.5份、富铬发芽糙米粉7.5份、荜茇提取物0.75份、葫芦巴总皂苷0.75份、芹菜7.5份、山楂6.5份、木瓜11份、食盐0.75份、小麦粉65份。
实施例3
一种预防并改善脂肪肝的竹荪面条,由以下重量份组成:竹荪粉20份、富铬发芽糙米粉10份、荜茇提取物1份、葫芦巴总皂苷1份、芹菜10份、山楂8份、木瓜12份、食盐1份、小麦粉70份。

取80只小鼠,随机分为8组,每组10只小鼠,其中一组每天灌喂基础日粮作为正常对照组,一组灌喂高脂饲料作为模型组,将高脂饲料与实施例1-3或对比例1-3制备的面条混合,分别喂食剩余6组,连续喂食2周后,将所有动物禁食12h,称量体重,摘除眼球取血,用全自动生化分析仪测量血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量,其结果如表1所示:

由表1可知,模型组血清的TC、HDL-C、LDL-C含量及TC/HDL-C较正常对照组明显升高,TG含量与正常对照组无明显差异,而实施例1-3能显著降低TG含量,对比例1-3则与正常对照组、模型组无显著差异。
取血后迅速剖腹,将完整的肝脏取出,用滤纸吸干表面水分后称重,测定其肝指数,肝指数=肝重/体重*100%;取肝组织100mg,按1:9的比例加入异丙醇,在冰水中制成10%匀浆,提取上清液,用全自动生化分析仪测定TG、TC含量,其结果如表2所示:

由表2可知,与正常对照组相比较,模型组的肝指数及TC、TG水平明显升高,而实施例1-3组的肝指数明显低于模型组,对比例1-3组则无显著差异,实施例1-3还能显著降低TG含量,TC含量则无显著差异。
综上所述,模型组血脂、肝脂、肝指数显著高于正常对照组,肝细胞出现明显的脂肪变性,而实施例1-3能降低血清TG的含量,并降低肝组织TG含量,抑制肝指数升高,减轻肝细胞脂肪变性,具有防治脂肪肝的作用,对比例1-3对脂肪肝无明显改善作用,实施例1-3对脂肪肝的防治效果比较明显,尤其实施例2效果较好,因此,在竹荪面条对小鼠脂肪肝治疗作用的实验中采用实施例2。
试验例2竹荪面条治疗脂肪肝的功效验证
取51只小鼠,实验前小鼠自由饮水进食,在实验环境中适应6天,随机分成2组,一组12只小鼠作为正常对照组(NC),喂食基础日粮,一组39只小鼠灌胃高脂饲料作为模型组(HF),12周后,将模型组小鼠随机分成3组,一组12只小鼠继续喂食高脂饲料,一组13只小鼠喂食基础日料作为ND组,一组14只小鼠喂食本发明制备的实施例2作为ED组,连续喂食2周后,所有动物禁食12h,称量体重,摘除眼球取血,用全自动生化分析仪测量血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量,其结果如表3所示:

HF组血清TC、HDL-C、LDL-C含量及TC/HDL-C叫NC组明显升高,而TG含量与正常对照组无显著差异,ND组血清TC含量及TC/HDL-C显著高于NC组,HDL-C、LDL-C及TG含量恢复至NC组水平,ED组能明显降低血清TG含量。
取血后迅速剖腹,将完整的肝脏取出,滤纸吸干表面水分后称重,测定其肝指数,肝指数=肝重/体重*100%;称取适量肝组织,加生理盐水,在冰水中制成10%匀浆,提取上清液,用全自动生化分析仪测定TG、TC含量,按试剂盒说明检测SOD活性及MDA水平,其结果如表4所示:

与NC组比较,HF组肝指数及TC、TG水平明显高于NC组,ND组肝指数及TC、TG水平也显著高于NC组,但低于HF组,ED组与HF组和ND组比较,其肝指数和TG含量均明显降低,而TC水平则无显著差异。HF组肝组织SOD活性显著低于NC组,MDA含量明显高于NC组,ND组SOD活性也显著低于NC组,却明显高于HF组,而MDA含量也高于NC组,但低于HF组,ED组与HF组和ND组比较,SOD活性显著上升,MDA含量明显下降。
总体而言,本发明制备的预防并改善脂肪肝的竹荪面条能够显著降低肝组织中的TG水平,有效改善脂肪肝变性程度,抑制过氧化反应,提高机体清除氧自由基的能力,从而具有治疗脂肪肝的作用。
综上所述,本发明制备的预防并改善脂肪肝的竹荪面条,能够调控脂质代谢,抑制肝脏脂质沉积,抑制氧化应激反应,改善脂肪肝变性程度,保护肝细胞,从而具有预防并改善脂肪肝的作用。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对本发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。
Claims (7)
1.一种预防并改善脂肪肝的竹荪面条,其特征在于,由以下重量份组成:竹荪粉15-20份、富铬发芽糙米粉5-10份、荜茇提取物0.5-1份、葫芦巴总皂苷0.5-1份、芹菜5-10份、山楂5-8份、木瓜10-12份、食盐0.5-1份、小麦粉60-70份。
2.根据权利要求1所述的一种预防并改善脂肪肝的竹荪面条,其特征在于,由以下重量份组成:竹荪粉17.5份、富铬发芽糙米粉7.5份、荜茇提取物0.75份、葫芦巴总皂苷0.75份、芹菜7.5份、山楂6.5份、木瓜11份、食盐0.75份、小麦粉65份。
3.根据权利要求1或2所述的一种预防并改善脂肪肝的竹荪面条,其特征在于,所述竹荪粉的制备方法为:
a.将竹荪母种在无菌条件下接种到PDA斜面上,置于25℃恒温箱中培养7-10天,得到活化竹荪菌丝;
b.用2g蔗糖、1.5g黄豆饼粉、2g玉米粉、0.4g酵母粉、0.03g磷酸二氢钾、0.08g硫酸镁、0.01g维生素B1、水1000ml制成pH为6的液体培养基,取80ml液体培养基装入500ml三角瓶内,以接入量为1cm3将活化好的竹荪菌丝接入液体培养基中,置于25℃恒温摇床中,以120r/min的转速进行以及培养5天;
c.将一级摇瓶中的菌丝以接种量为6%接入二级摇瓶中,置于25℃恒温摇床中,以120r/min的转速进行二级培养4天;
d.取出二级培养液,用0.088mm目筛过滤,将菌丝体置于80℃烘箱中烘干至恒重,粉碎过100目筛,得到竹荪粉。
4.根据权利要求1或2所述的一种预防并改善脂肪肝的竹荪面条,其特征在于,所述富铬发芽糙米粉的制备方法为:
a.取经挑选、除杂、均匀的糙米100g,漂洗干净,用1%的次氯酸钠溶液浸泡30min,然后用去离子水冲洗5次,并用70mg/L三氯化铬水溶液在25℃下浸泡10h,然后置于35℃下通气培养18h,得到富铬发芽糙米;
b.将富铬发芽糙米用清水清洗沥干后,置于50℃的热风温度下干燥至富铬发芽糙米含水量为14%,然后粉碎,得到富铬发芽糙米粉。
5.根据权利要求1或2所述的一种预防并改善脂肪肝的竹荪面条,其特征在于,所述荜茇提取物的制备方法为:取荜茇粗粉250g,置于1000ml的圆底烧瓶中,加入330ml的去离子水浸泡12h,然后加热回流提取4h,过滤,取滤液减压浓缩,干燥至恒重,得到荜茇提取物。
6.根据权利要求1或2所述的一种预防并改善脂肪肝的竹荪面条,其特征在于,所述葫芦巴总皂苷的制备方法为:取葫芦巴粉末100g,按料液比为1:8加入70%乙醇加热回流提取3次,每次1h,过滤,合并3次滤液,减压回收乙醇后,加入蒸馏水,调节溶液浓度为30%,然后加入D-101型大孔树脂柱上,用蒸馏水洗脱至无色后,再用4BV的70%乙醇洗脱,收集洗脱液,取洗脱液经超滤器超滤,得到超滤液,然后减压回收乙醇,浓缩干燥,得到葫芦巴总皂苷。
7.一种制备如权利要求1-6任一项所述的一种预防并改善脂肪肝的竹荪面条的方法,其特征在于,包括以下步骤:
(1)取山楂去核,木瓜削皮去核后,与芹菜混合,加入500ml蒸馏水,用组织匀浆器破碎后,放入榨汁机中榨汁,取榨汁液经3000r/min离心15min,取上清液即为果蔬汁;
(2)取竹笋粉、富铬发芽糙米粉、小麦粉混合过筛,放入和面机中,再加入荜茇提取物、葫芦巴总皂苷和步骤(1)的果蔬汁,搅拌均匀,和面15-20min,得到面团;
(3)将步骤(2)的面团放入熟化机中,以5-10r/min的转速熟化15-30min;
(4)将步骤(3)熟化好的面团经多级锟压和切条机切条,得到2mm的面条;
(5)将步骤(4)切好的面条放入烘房中,烘干至面条含水量为14%,得到本发明预防并改善脂肪肝的竹荪面条。
