本发明提供一种植物发酵物。提供一种新型有效成分,其能够配合至皮肤外用组合物、饮料食品用组合物中,兼具抗氧化作用和美白作用,对于通过以这两种作用的组合为基础的协同效应来改善皮肤的老化和肌肤粗糙、改善色斑、雀斑、以及改善细胞的不健全化而言是有用的。本发明是利用酵母、乳酸菌、曲霉或枯草杆菌使属于景天科(Crassulaceae)红景天属(Rhodiola)的植物和/或其提取物发酵而得到的发酵物、以及配合有该发酵物的皮肤外用组合物、饮料食品用组合物。

背景技术
以往,作为配合至皮肤外用剂、饮料食品中的有效成分,研究开发了源自天然物的成分。但是,将这些源自天然物的成分用作皮肤外用剂、饮料食品的有效成分时,在有效性、稳定性等的方面存在课题。

发明内容
发明要解决的问题
本发明人等为了解决上述课题而进行了深入研究,结果发现:属于景天科(Crassulaceae)红景天属(Rhodiola)的植物的发酵物具有格外优异的抗氧化效果和美白效果。
以往,通过例如专利文献1~3而已知将属于红景天属的植物的提取物用于皮肤外用剂等。但是,将属于红景天属的植物或其提取物的发酵物用作皮肤外用剂、饮料食品用途的有效成分尚属未知。
用于解决问题的方法
本发明是利用乳酸菌、酵母、曲霉或枯草杆菌使属于景天科红景天属的植物发酵而得到的发酵物。
另外,本发明的特征在于,上述乳酸菌为乳酸杆菌或乳酸球菌。
另外,本发明中,发酵物是利用酵母使属于景天科(Crassulaceae)红景天属(Rhodiola)的植物发酵而得到的。

具体实施方式
以下,针对本发明进行详细说明。
作为本发明中使用的景天科红景天属的植物,可列举出例如大花红景天(Rhodiola Crenulata)、红景天(Rhodiola Rosea)、Rhodiola ishidae、高山红景天(Rhodiola.sachalinensis)、玫瑰红景天(Rhodiola Rosea)等。
利用微生物(酵母、乳酸菌、曲霉或枯草杆菌)使景天科红景天属的植物或其提取物发酵时,对该植物的发酵部位没有特别限定,可以使用全草、花、叶、根、茎等适当的部位,优选使用根或者包括根在内的全草。
红景天属植物的发酵例如可如下那样地进行。首先,作为发酵的同化源,可以使用红景天属的植物自身(以下有时称为植物体),或者,也可以使用将植物体用适当的介质提取而得到的提取物。此外,使用提取物时,还可以不通过固液分离来去除被提取物的植物体,而在包含植物体的状态下直接进行发酵。此处,红景天属植物可以是存活的状态,也可以是预先经干燥或半干燥而得到的产物。此外,作为形状,也可以直接使用采取后的形状。
将植物体作为同化源来进行发酵时,首先使植物体悬浮在溶剂中。作为用于使植物体悬浮的溶剂,可以使用水或者水与低级醇类(甲醇、乙醇、异丙醇等)或二醇类(乙二醇、1,3-丙二醇、1,3-丁二醇、1,2-戊二醇、甘油等)的混合液。此处,植物体与溶剂的混合比以植物体的干燥重量换算通常为1:1~1:100、优选为1:10~1:50的范围。
将植物体的提取物用作发酵同化源时,作为提取物的制备方法,可以采取下述方法等:将提取对象部位、例如全草或根部根据需要预先进行水洗、干燥,优选进一步细切或粉碎,并在此基础上通过浸渍法、对流提取法等适当的手段使其与提取溶剂相接触。此外,也可以使用超临界提取法。
作为提取溶剂,可列举出水;甲醇、乙醇、丙醇等低级醇类、油醇、硬脂醇、辛基十二醇等高级醇类;乙二醇、1,3-丙二醇、1,2-戊二醇、1,3-丁二醇、甘油等多元醇类;乙酸乙酯、乙酸丁酯、丙酸甲酯、三辛酸甘油酯等酯类;丙酮、甲乙酮等酮类;乙醚、异丙醚等醚类;正己烷、甲苯、氯仿等烃系溶剂等,它们可以单独使用或者混合使用两种以上。
关于使用混合溶剂时的混合比,例如,如果为水与乙醇的混合溶剂,则以容量比(以下相同)计优选设为1:1~25:1的范围;如果为水与甘油的混合溶剂,则以容量比计优选设为1:1~15:1的范围;如果为水与1,3-丙二醇或1,3-丁二醇的混合溶剂,则以容量比计优选设为1:1~15:1的范围。作为提取溶剂,使用除了水之外的溶剂或者水与除了水之外的溶剂的混合溶剂时,可以在转移至发酵工序之前,通过浓缩等操作先将除了水之外的溶剂蒸馏去除,再以固体成分达到0.01~10%左右的方式再次溶解于水或者用水稀释来作为发酵的同化源。
作为如上那样调整的悬浮液或提取物溶液的发酵处理中使用的微生物,可列举出下述微生物。
本发明中,酵母可列举出例如酿酒酵母(Saccharomyces cerevisiae)、泡盛酵母(Saccharomyces awamori)、Saccharomyces chevalieri、卡尔斯伯酵母(Saccharomycescarlsbergensis)、贝酵母(Saccharomyces bayonus)等酵母属的酵母;耐碱酵母(Galactomyces)属的酵母;德尔布有孢圆酵母(Torulaspora delbruekii)、Torulasporafermentati、Torulaspora rosei等有孢圆酵母属的酵母;鲁氏接合酵母(Zygosaccharomyces rouxii)、Zygosacchar omyces soya、Zygosaccharomyces sake、Zygosaccharomyces miso、乳酸克鲁维酵母(Zygosaccharomyces lactis)等接合酵母属的酵母;耐盐假丝酵母(Candida versatilis)、埃切假丝酵母(Candida etchellsii)、乳酒假丝酵母(Candida kefyr)、清酒假丝酵母(Candida sake)、斯考特假丝酵母(Candidascottii)等假丝酵母属的酵母;出芽短梗霉菌(Aureobasidium Pullulans)、Aureobasidium mansonii、Aureobasideium microstictum等出芽短梗属的酵母。此外,作为本发明所述的酵母,可以是清酒酵母、葡萄酒酵母、啤酒酵母、源自植物的酵母、源自海洋的酵母中的任一者。源自植物或源自海洋的酵母是指,从存在于自然界的植物或海水等原材料中分离酵母而得到的酵母。这些酵母可从自然界的原材料中通过分離微生物的常规方法得到。例如,可以在放入原材料的培养液中使酵母在其最适温度及pH下增殖后,将培养液涂布于培养基后进行分离。可列举出源自属于百合科(Liliaceae)的植物的花的酵母、或者源自属于蔷薇科(Rosaceae)的植物的花的酵母。作为百合科的植物,优选为属于百合属(Lilium)的植物,可列举出例如日本百合(Lilium japonicum)、天香百合(Lilium auratumLindl.)、卷丹(Lilium lancifolium Thunb.)、野百合(Lilium brownii F.E.Br.)、高贵百合(Lilium nobilissimum Makino)、含百花百合(Lilium candidum L.)、渥丹(Liliumconcolor Salisb.)、台湾百合(Lilium formosanum Wallace)、大花卷丹(Liliumleichtlinii Hook.f.var.tigrinum(Regel)Nichols.)、麝香百合(Lilium longiflorumThunb.)等。此外,作为蔷薇科植物,优选为属于蔷薇科蔷薇属(Rosa)的蔷薇、或者属于蔷薇科樱属(Prunus,Cerasus)的樱树。本发明中确认了源自百合科植物的花和蔷薇科植物的花的酵母具有后述的抗氧化效果和美白效果。
作为发酵中使用的乳酸菌,可列举出例如植物乳杆菌(Lactobacillusplantarum)、短乳杆菌(Lactobacillus brevis)、干酪乳杆菌(Lactobacillus casei)、德氏乳杆菌(Lactobacillus delbrueckii)等乳杆菌(Lactobacillus)属的乳酸菌;广布肉杆菌(Carnobacterium divergens)、居鱼肉杆菌(Carnobacterium piscicola)等肉杆菌(Carnobacterium)属的乳酸菌;肠膜状明串珠菌(Leuconostoc mesenteroides)、乳明串珠菌(Leuconostoc lactis)、柠檬明串珠菌(Leuconostoc citreum)等明串珠菌(Leuconostoc)属的乳酸菌;粪链球菌(Streptococcus faecalis)、酿脓链球菌(Streptococcus pyogenes)等链球菌属的乳酸菌;钻黄肠球菌(Enterococcuscaseliflavus)、Enterococcus sulfreus等肠球菌(Enterococcus)属的乳酸菌;植物乳球菌(Lactococcus plantarum)、乳乳球菌(Lactococcus lactis)、棉籽糖乳球菌(Lactococcus rafinolactis)等乳球菌属的乳酸菌;融合魏斯氏菌(Weissella confusa)、坎式魏斯氏菌(Weissella kandleri)等魏斯氏菌属的乳酸菌;小阿托波氏菌(Atopobiumminutum)、极小阿托波氏菌(Atopobium parvulus)等阿托波氏菌(Atopobium)属的乳酸菌;河流漫游球菌(Vagococcus fluvialis)、沙氏漫游球菌(Vagococcus salmoninarum)等漫游球菌(Vagococcus)属的乳酸菌;有害片球菌(Pediococcus damnosus)、戊糖片球菌(Pediococcus pentosaceus)等片球菌(Pediococcus)属的乳酸菌。
作为发酵中使用的曲霉,可列举出例如米曲霉(Aspergillus oryzae)、黄曲霉(Aspergillus flavus)、Aspergillus polyoxogenes、酱油曲霉(Aspergillus sojae)等黄曲霉;泡盛曲霉(Aspergillus awamori)、川内曲霉(Aspergillus kawauchii)、宇佐美曲霉(Aspergillus usami)、黑曲霉(Aspergillus niger)等黑曲霉;安卡红曲霉(Monascusanka)、丛毛红曲霉(Monascus pilosus)等红曲霉。
作为发酵中使用的枯草杆菌(Bacillus subtilis),可列举出例如纳豆芽孢杆菌(Bacillus natto)、枯草芽孢杆菌(Bacillus subtilis)、环状芽胞杆菌(Bacilluscirculans)。
利用微生物使上述悬浮液或提取物发酵时,在发酵工序之前,需要进行杀菌来去除阻碍发酵的杂菌。作为该杂菌的杀菌去除方法,可以使用将发酵原材料预先用杀菌用乙醇等清洗后再悬浮至无菌水等无菌溶剂中的方法,此外,也可以将发酵原材料悬浮至溶剂后,通过加热杀菌等对悬浮液进行杀菌。作为加热杀菌处理,通常使用下述方法:将悬浮液以120~130℃加热10~20分钟的高压釜杀菌法;以80~90℃保持60~120分钟且1天保持1次、在2~3天内反复进行的间断杀菌法之类的加热杀菌法。
此外,在发酵处理时,作为碳源,可以添加葡萄糖、半乳糖、果糖、蔗糖、麦芽糖、乳糖等糖类。
将无菌化的悬浮液与培养液装入发酵罐中,并向其中接种微生物而使其发酵。微生物的接种量适合为107~108个/mL。即使接种量大于上述范围,发酵的进行时间也基本不变,另一方面,如果小于上述范围,则至发酵结束为止需要较长时间,故不优选。
发酵温度通常为5~50℃的范围、优选为各微生物的最佳生长温度的范围。发酵天数在最佳温度下通常为1~10天、优选为2~5天的范围。如果发酵天数短于上述的通常范围,则存在不会充分进行发酵、发酵物的有效性降低的倾向,另一方面,如果延长至超过10天,则不仅观察不到有效性的进一步上升,还会出现着色、发酵异味的加剧,均不优选。
进行上述发酵处理时,为了使红景天属植物的成分更有效地用作微生物的同化源,可以在接种微生物之前或者接种的同时向前述悬浮液或提取物溶液中添加酶来进行水解处理。
此时,作为酶,可以使用选自蛋白分解酶、淀粉分解酶、果胶质分解酶和纤维素分解酶中的至少1种酶。
作为蛋白分解酶,可以使用例如肌动蛋白酶(アクチナーゼ)等肌动蛋白酶类、胃蛋白酶等胃蛋白酶类、胰蛋白酶、胰凝乳蛋白酶等胰蛋白酶类、木瓜蛋白酶、木瓜凝乳蛋白酶等木瓜蛋白酶类、甘氨酰甘氨酸肽酶、羧肽酶、氨肽酶等肽酶类、菠萝蛋白酶等。
作为淀粉分解酶,可以使用例如α-淀粉酶、β-淀粉酶、葡糖淀粉酶、β-半乳糖苷酶等。
作为果胶质分解酶,可以使用例如果胶解聚酶、果胶脱甲氧酶、果胶裂解酶、果胶酯酶、聚半乳糖醛酸酶等。
作为纤维素分解酶,可以使用例如纤维素酶、半纤维素酶、琼脂酶、甘露聚糖酶、壳多糖酶、脱乙酰壳多糖酶、卡拉胶酶、精氨酸酶、岩藻多糖酶(フコイダナーゼ)、菊粉酶、木聚糖酶、木质素酶等。
酶的用量相对于悬浮液中的红景天属植物的固体成分合计优选为0.001~10重量%。此外,相对于红景天属植物的提取物的固体成分,合计优选为0.0001~5重量%。作为温度、时间等处理条件,如果在发酵前进行酶处理,则适合在各酶的最佳温度附近进行1~24小时的处理,另一方面,如果在发酵的同时进行酶处理,则应用与基于前述微生物的发酵同样的条件。
在上述发酵结束后,为了进行微生物杀菌,此外,如果是组合使用酶处理的情况,则兼作微生物的杀菌和酶的失活,对发酵液以70~100℃实施10~120分钟左右的加热杀菌处理后,将其直接供作皮肤外用组合物或饮食用组合物的配合原料,或者通常且适合通过过滤或离心分离等固液分离手段来分取液相,并根据需要将pH调整至一般化妆品的pH区域、即pH6~8,进而根据需要通过稀释或浓缩而制成适当的浓度后,供作皮肤外用组合物或饮食用组合物的配合原料。此外,根据情况,也可以将通过喷雾干燥法、冷冻干燥法等常规方法将固液分离后的液相制成粉末而得到的产物配合至上述组合物中。
本发明所述的发酵物可以配合至皮肤外用组合物(化妆品、医药外用品、外用药品)、美容用或健康促进用的饮食用组合物中。作为皮肤外用组合物,可列举出例如乳液、乳霜、洗剂、精华、润肤膏、口红、粉底、液体粉底、彩妆压粉、腮红、粉霜(白粉)、洁面产品、沐浴露、头皮用洗发水、头发用洗发水、头发用护发素、生发用、护发用的洗发水或营养剂、肥皂等清洁用化妆品、以及入浴剂等,但本发明不限定于它们。此外,作为美容用或健康促进用的饮料食品用组合物,可以配合至美容饮料、营养饮料、运动饮料、类水饮料(near water)、维生素饮料、矿物质饮料、酒精饮料等饮料;各种汤类(也包括粉末汤)、奶制品、果冻、糖果、片状点心、口香糖等食品;片剂、液状、颗粒状或胶体状的健康食品/饮料等,但本发明不限定于它们,可以配合至能够口服摄取的饮料食品等
关于本发明的发酵物的配合量,以其固体成分计,在基础化妆品的情况下为0.002~1.0重量%(固体成分重量%、以下相同)的范围,在彩妆化妆品的情况下为0.002~1.0重量%的范围,此外,在清洁用化妆品的情况下为0.002~10.0重量%的范围。此外,在毛发用化妆品的情况下,以发酵物的固体成分量计,为0.0001~5.0重量%的范围。此外,饮食用组合物中的本发明的发酵物的配合量以固体成分量计优选为0.1~15重量%的范围。
将本发明所述的发酵物用于皮肤外用组合物、饮食用组合物时,可以在发酵物的基础上根据需要适当配合皮肤外用剂、饮料食品中通用的成分、例如油性成分、表面活性剂(合成系、天然物系)、乳化剂、保湿剂、增稠剂、防腐/杀菌剂、粉体成分、抗氧化剂、螯合剂、pH调节剂、色素、香料等。此外,在不损害本发明发酵物的有效性、优点的情况下,也可以与其它生理活性成分组合来配合。
此处,作为油性成分,可列举出例如莲花油、橄榄油、荷荷巴油、蓖麻油、大豆油、米油、米糠油、米胚油、椰子油、甘菊油、棕榈油、可可油、池花籽油、香柠檬油、蔷薇果油、薰衣草油、牛油树脂、茶树油、鳄梨油、澳洲坚果油、源自植物的角鲨烷等源自植物的油脂类;貂油、海龟油等源自动物的油脂类;蜜蜡、巴西棕榈蜡、米糠蜡、羊毛脂等蜡类;液态石蜡、凡士林、石蜡、角鲨烷等烃类;肉豆蔻酸、棕榈酸、硬脂酸、油酸、异硬脂酸、二十碳烯酸等脂肪酸类;月桂醇、鲸蜡醇、硬脂醇等高级醇类;肉豆蔻酸异丙酯、棕榈酸异丙酯、油酸丁酯、2-乙基己基甘油酯、高级脂肪酸辛基十二烷基酯(硬脂酸辛基十二烷基酯等)等合成酯类和合成三甘油酯类等。

作为乳化剂或乳化助剂,也可以配合酶处理甜菊等甜菊衍生物、皂苷或其衍生物、酪朊或其盐(钠等)、糖与蛋白质的复合体、蔗糖或其酯、乳糖、源自大豆的水溶性多糖、源自大豆的蛋白质与多糖的复合体、羊毛脂或其衍生物、胆固醇、甜菊衍生物(甜菊酶处理物等)、硅酸盐(铝、镁等)、碳酸盐(钙、钠等)、皂苷及其衍生物、卵磷脂及其衍生物(氢化卵磷脂等)、乳酸菌发酵米、乳酸菌发酵发芽米、乳酸菌发酵谷类(麦类、豆类、杂谷等)等。
作为保湿剂,有例如甘油、丙二醇、二丙二醇、1,3-丁二醇、聚乙二醇、山梨糖醇、木糖醇、吡咯烷酮羧酸钠等,进而可列举出海藻糖等糖类、粘多糖类(例如透明质酸及其衍生物、软骨素及其衍生物、肝素及其衍生物等)、弹性蛋白及其衍生物、胶原蛋白及其衍生物、NMF相关物质、乳酸、脲、高级脂肪酸辛基十二烷基酯、海藻提取物、白芨根(白及)提取物、各种氨基酸和它们的衍生物。
作为增稠剂,可列举出例如藻酸、琼脂、角叉菜胶、墨角藻聚糖等源自褐藻、绿藻或红藻的成分;白芨根(白及)提取物;果胶、刺槐豆胶、芦荟多糖体、产碱菌多醣等多糖类;黄原胶、黄蓍胶、刺槐豆胶、瓜尔胶等胶类;羧甲基纤维素、羟乙基纤维素等纤维素衍生物;聚乙烯醇、聚乙烯基吡咯烷酮、羧基乙烯基聚合物、丙烯酸/甲基丙烯酸共聚物等合成高分子类;透明质酸及其衍生物;聚谷氨酸及其衍生物等。
作为防腐/杀菌剂,有例如脲;对羟基苯甲酸甲酯、对羟基苯甲酸乙酯、对羟基苯甲酸丙酯、对羟基苯甲酸丁酯等对羟基苯甲酸酯类;苯氧基乙醇、二氯酚、六氯酚、盐酸氯己啶(塩酸クロルヘキシジン)、苯扎氯铵、水杨酸、乙醇、十一碳烯酸、酚类、Jamal(咪唑啉脲(イミダゾデイニールウレア))、多磷酸、丙二醇、1,2-戊二醇、各种精油类、树皮干馏物、人参发酵液、源自甘蔗等植物的乙醇或1,3-丁二醇等。
作为粉体成分,有例如绢云母、氧化钛、滑石、高岭土、膨润土、氧化锌、碳酸镁、氧化镁、氧化锆、硫酸钡、无水硅酸、云母、尼龙粉末、聚乙烯粉末、丝粉、纤维素系粉末、谷类(米、麦、玉米、粟等)的粉末、豆类(大豆、小豆等)的粉末等。
作为紫外线吸收剂,有例如对氨基苯甲酸乙酯、对二甲基氨基苯甲酸乙基己酯、水杨酸戊酯及其衍生物、对甲氧基肉桂酸2-乙基己酯、肉桂酸辛酯、氧苯酮、2,4-二羟基二苯甲酮、2-羟基-4-甲氧基二苯甲酮-5-磺酸盐、4-叔丁基-4-甲氧基苯甲酰基甲烷、2-(2-羟基-5-甲基苯基)苯并三唑、尿刊酸、尿刊酸乙酯、芦荟提取物等。
作为抗氧化剂,有例如丁基羟基茴香醚、丁基羟基甲苯、没食子酸丙酯、日本紫珠(ムラサキシキブ)的提取物、白芨根的提取物、芍药提取物、维生素E及其衍生物(例如维生素E烟酸盐、维生素E亚油酸盐等)等。
作为螯合剂,有例如乙二胺羟基乙基三乙酸三钠、依地酸或其盐类、葡糖酸、植酸、多聚磷酸钠、偏磷酸钠、羟基乙烷二膦酸四钠等。

使用红景天(Rhodiola Rosea)来代替制造例1的大花红景天,并使用源自樱树的花的酵母(酿酒酵母),除此之外,与制造例1同样操作,得到酵母发酵物溶液400g(固体成分浓度为1.01%)。
制造例3.发酵物的制备(3)
除了使用Rhodiola ishidae来代替制造例1的大花红景天之外,与制造例1同样操作,得到酵母发酵物溶液400g(固体成分浓度为1.00%)。
制造例4.发酵物的制备(4)
将大花红景天的根干燥,向干燥物10g中添加纯化水100g,进行加热杀菌。向该提取悬浮液中接种乳酸菌(乳乳球菌)108个/mL,在氮气气流下以37℃静置培养3天。在培养结束后进行加热杀菌,过滤培养液,得到大花红景天的乳酸菌发酵物溶液400g(固体成分浓度为1.05%)。
制造例5.发酵物的制备(5)
除了使用红景天来代替制造例4的大花红景天之外,与制造例4同样操作,得到乳酸菌发酵物溶液400g(固体成分浓度为1.04%)。
制造例6.发酵物的制备(6)
除了使用植物乳杆菌(Lactobacillus plantarum)来代替制造例4的乳酸菌(乳乳球菌)之外,与制造例4同样操作,得到乳酸菌发酵物溶液400g(固体成分浓度为1.02%)。
制造例7.发酵物的制备(7)
将大花红景天的根干燥,向干燥物10g中添加纯化水100g和葡萄糖1g,进行加热杀菌。向该提取悬浮液中接种源自日本百合的花的酵母(酿酒酵母)108个/mL,在氮气气流下以30℃静置培养3天。在培养结束后进行加热杀菌,添加1,3-丁二醇100g,以80℃进行静置。将其过滤,得到大花红景天的酵母发酵物溶液400g(固体成分浓度为1.07%)。
制造例8.发酵物的制备(8)
将大花红景天的根干燥,向干燥物10g中添加纯化水100g和葡萄糖1g,进行加热杀菌。向该提取悬浮液中接种乳酸菌(乳乳球菌)108个/mL,在氮气气流下以37℃静置培养3天。在培养结束后进行加热杀菌,添加1,3-丁二醇100g,以80℃进行静置。将其过滤,得到大花红景天的乳酸菌发酵物溶液400g(固体成分浓度为1.02%)。
制造例9.发酵物的制备(9)
将大花红景天的根干燥,向干燥物10g中添加纯化水100g和葡萄糖4g,进行加热杀菌。向该提取悬浮液中接种曲霉(米曲霉)108个/mL,在氮气气流下以30℃静置培养3天。在培养结束后进行加热杀菌,过滤培养液,得到大花红景天的曲霉发酵物溶液400g(固体成分浓度为0.91%)。
制造例10.发酵物的制备(10)
将大花红景天的根干燥,向干燥物10g中添加纯化水100g和葡萄糖4g,进行加热杀菌。向该提取悬浮液中接种枯草杆菌(纳豆芽孢杆菌)108个/mL,在氮气气流下以30℃静置培养3天。在培养结束后进行加热杀菌,过滤培养液,得到大花红景天的枯草杆菌发酵物溶液400g(固体成分浓度为0.85%)。
制造例11.发酵物的制备(11)
除了使用源自樱树的花的酵母(酿酒酵母)来代替制造例7的酵母之外,通过与制造例7同样的操作,得到大花红景天的酵母发酵物溶液400g(固体成分浓度为1.05%)。
制造例12.发酵物的制备(12)
除了使用源自蔷薇的花的酵母(酿酒酵母)来代替制造例7的酵母之外,通过与制造例7同样的操作,得到大花红景天的酵母发酵物溶液400g(固体成分浓度为1.03%)。
比较制造例1.提取物的制备(1)
将大花红景天的根干燥,向干燥物5g中添加纯化水50g,进行加热杀菌。将该提取悬浮液在氮气气流下以30℃静置培养3天。将提取悬浮液进行加热杀菌并过滤,得到大花红景天的提取溶液200g(固体成分浓度为1.02%)。

试验例1.SOD状活性的评价试验
将0.2M Tris-盐酸缓冲液50μL、1mM乙二胺四乙酸(EDTA)二钠溶液20μL、0.75mM四唑硝基蓝氯化物(NBT)溶液10μL、1mM黄嘌呤溶液20μL、0.06U/mL黄嘌呤氧化酶溶液50μL、制造例1~12的发酵物1~12和比较制造例1的提取物50μL(以各自的最终浓度达到0.1%的方式进行制备)混合来制备试样溶液。将该试样溶液以37℃温育5分钟,使其产生超氧化物。并且,测定NBT被超氧化物还原而生成的甲臜量、即560nm下的吸光度。此外,作为对照,使用50%的1,3-丁二醇水溶液,进行与上述相同的操作,求出添加各试样时的吸光度相对于此处得到的吸光度的相对值,作为超氧化物残留率(%)。此外,为了确认试验体系是否正常发挥功能,作为阳性对照,针对添加有超氧化物歧化酶(SOD)8.75units/mL来代替试样溶液的情况,也进行相同的试验。
将试验例1的结果示于表1。

如表1所示可确认:本发明所述的发酵物具有格外优异的SOD状活性。
试验例2.DPPH自由基消除作用的评价试验
将DPPH(1,1-二苯基-2-苦基肼)2.4份溶解于乙醇20份,向其中添加纯化水20份来制备DPPH溶液。相对于该DPPH溶液24份,添加18v/v%乙醇溶液19.2份、2M乙酸-乙酸钠缓冲液(pH5.5)4.8份,以DPPH添加溶液的形式进行制备。此外,为了除去提取液自身的色调对试验造成的影响,使用50v/v%乙醇溶液来代替DPPH溶液,将18v/v%乙醇溶液与2M乙酸-乙酸钠缓冲液的混合液作为对照液。接着,将制造例1~12的发酵物1~12和比较制造例1的提取物分别用纯化水进行稀释,从而制备12种试样溶液。需要说明的是,各组合物按照相对于试样溶液总量的溶液的最终浓度分别达到0.1%的方式用纯化水稀释。将该试样溶液与DPPH添加溶液或对照液以1:3的比例进行混合,并以37℃静置20分钟后,测定将各试样溶液和DPPH添加溶液混合时的550nm下的吸光度与将各试样溶液和对照液混合时的550nm下的吸光度之差,确认DPPH自由基的残留量。此外,作为对照(Control),针对使用50%的1,3-丁二醇水溶液的情况,也进行与上述相同的操作,求出将未添加试样时的DPPH自由基残留率设为100时的添加各试样时的DPPH自由基残留率的相对值。此外,为了确认试验体系是否正常发挥功能,作为阳性对照,针对添加有水溶性维生素E[Trolox](最终浓度为25μM)来代替试样溶液的情况,也进行相同的试验。
将试验例2的结果示于表2。

如表2所示可确认:本发明所述的发酵物具有格外优异的DPPH自由基消除作用。
试验例3.酪氨酸酶活性抑制的评价试验
使用McIlvaine缓冲液(pH6.8)来制备125U/mL的酪氨酸酶的酶液。接着,将制造例7、8、11、12的发酵物7、8、11、12和比较制造例1的提取物用纯化水制备的试样溶液以100μL/孔的量添加至96孔的微孔板中。此处,作为试样溶液,使用了按照制造例1的提取物溶液的最终浓度(溶液形式的浓度)相对于其总量分别达到0.5%的方式制备的产物。接着,同样地以100μL/孔的量添加0.03%L-酪氨酸溶液,并充分混合。向其中以100μL/孔的量添加上述酪氨酸酶的酶液并充分混合,在室温下静置10分钟。并且,测定波长490nm下的反应溶液的吸光值。此外,使用纯化水来代替试样溶液并同样操作后,作为对照,通过将对照吸光值设为100时的相对值来求出酪氨酸酶活性率。

Claims (10)
1.一种发酵物,其是利用乳酸菌、曲霉或枯草杆菌使属于景天科Crassulaceae红景天属Rhodiola的植物发酵而得到的。
2.根据权利要求1所述的发酵物,其特征在于,乳酸菌为乳酸杆菌或乳酸球菌。
3.一种发酵物,其是利用酵母使属于景天科Crassulaceae红景天属Rhodiola的植物发酵而得到的。
4.根据权利要求3所述的发酵物,其特征在于,酵母源自属于百合科Liliaceae的植物或者属于蔷薇科Rosaceae的植物。
5.根据权利要求4所述的发酵物,其特征在于,百合科植物是属于百合属Lilium的植物,蔷薇科植物是属于蔷薇属Rosa或樱属Prunus,Cerasus的植物。
6.根据权利要求1或2所述的发酵物,其中,发酵物是在糖的存在下利用乳酸菌、曲霉或枯草杆菌使属于景天科Crassulaceae红景天属Rhodiola的植物发酵而得到的。
7.根据权利要求3~5中任一项所述的发酵物,其中,发酵物是在糖的存在下利用酵母使属于景天科Crassulaceae红景天属Rhodiola的植物发酵而得到的产物。
8.根据权利要求1~7中任一项所述的发酵物,其特征在于,属于景天科Crassulaceae红景天属Rhodiola的植物为大花红景天Rhodiola Crenulata、红景天Rhodiola Rosea、Rhodiola ishidae、高山红景天Rhodiola.sachalinensis和玫瑰红景天Rhodiola Rosea中的任一种。
9.一种皮肤外用组合物,其包含权利要求1~8中任一项所述的发酵物作为有效成分。
10.一种饮食用组合物,其包含权利要求1~8中任一项所述的发酵物作为有效成分。
