Abstract
本发明属于家禽养殖技术,具体涉及一种抗肉鸡应激的富硒中草药添加剂、制备方法及使用方法,本发明的富硒中草药添加剂由中草药提取物和磨成粉末的酵母硒片按照1000:1‑1.5的质量比例混合而成;所述中草药提取物由如下重量份的中药原料制备而成:构树叶20‑35份、青蒿10‑20份、柴胡10‑15份、小地松12‑15份、银杏叶15‑35份、刺五加5‑8份、霍香6‑12份、山楂8‑15份。本发明的抗肉鸡应激的富硒中草药添加剂能够有效降低应激的不良影响,提高调节的免疫功能,改善肉鸡的消化吸收功能和生长性能,发挥了营养物质和药物的双重作用。
Description
技术领域
本发明属于饲料添加剂领域,具体涉及一种抗肉鸡应激的富硒中草药添加剂、制备方法及使用方法。
背景技术
肉鸡生长快、经济效益高,因此,饲养量大。但是肉鸡体型大、皮厚、皮下及腹部脂肪多的特点也造成其对应激的抵抗力差,尤其是高温导致的热应激的造成肉鸡大量死亡;而且,高密度的养殖模式、狭小的活动空间、饲料的改变及骤变的气温等因素都可以引起肉鸡应激的发生,严重降低了肉鸡养殖的经济效益。应激导致肉鸡抗氧化酶活性下降、免疫器官发育异常、免疫功能低下、精神沉郁;消化酶活性降低,采食量减少,饲料实际转化效率下降,生长速度减慢,发育不良。缓解应激的策略包括改善饲养环境、添加维生素、矿物质、电解质及抗菌药等。随着人们食品安全意识的增强,化学物质、抗菌药、激素及无机质的残留与副作用日益引起人们的警惕。天然的有机中草药成分和有机微量元素受到人们的青睐。中草药成分丰富,而且基本无毒副作用,作用广泛,资源丰富;具有镇静安神、抗应激、抗惊厥、增强免疫、健胃消食和促进生长的功效,并且通过提高抗氧化酶活性改善肉品质。提高养殖效益,具有巨大的发掘潜力和市场前景。
综上,现有技术存在的问题是抗菌药和激素等物质添加的饲料,饲喂肉鸡,这些物质残留在肉鸡体内,人食用肉鸡后会带来毒副作用,现在亟需一种天然、无毒副作用,而且能够有效抗肉鸡应激的饲料添加剂。
发明内容
为解决上述问题,本发明提供了一种抗肉鸡应激的富硒中草药添加剂、制备方法及使用方法,添加剂中没有使用抗菌药和激素等物质,有效提高肉鸡的抗氧化酶活性、抗应激能力、改善肉鸡的生长性能和肉品质。
本发明提供的一种抗肉鸡应激的富硒中草药添加剂,其特征在于,所述富硒中草药添加剂由中草药提取物和磨成粉末的酵母硒片按照1000:1-1.5的质量比混合而成;所述中草药提取物由如下重量份的中药原料制备而成:构树叶20-35份、青蒿10-20份、柴胡10-15份、小地松12-15份、银杏叶15-35份、刺五加5-8份、霍香6-12份、山楂8-15份。
优选的,所述抗肉鸡应激的富硒中草药添加剂在饲料中的使用方法,其特征在于,所述添加剂的加入量占饲料总重量的0.5-1%。
同时,本发明还提供了一种抗肉鸡应激的富硒中草药添加剂的制备方法,包括以下步骤:
S1,按照以下重量份称取各中草药原料:构树叶20-35份、青蒿10-20份、柴胡10-15份、小地松12-15份、银杏叶15-35份、刺五加5-8份、霍香6-12份、山楂8-15份;并将称取的各中草药原料于50℃烘箱内烘干,然后用中药超微粉碎机超微粉碎,过100目筛,制得中草药超微细粉,于4-10℃、干燥处保存;
S2,将中草药超微细粉与复合酶解液按1:20-30的质量比混合,在55℃-65℃恒温水浴锅中酶解25-40min,回流法除去溶剂,在50℃鼓风干燥箱干燥1h,得中草药酶解粉,备用;所述复合酶解液是将纤维素酶和果胶酶溶于体积分数60%-75%的乙醇溶液中制成的,且纤维素酶的浓度为10-25μg/g、果胶酶的浓度为80-110μg/g;
S3,将乳酸杆菌、双歧杆菌、酵母菌和嗜热链球菌的菌粉按照质量比为1-3:2-5:3-5:3-6的比例混合,得到的发酵复合菌溶于蒸馏水,制得发酵复合菌液;将中草药酶解粉与发酵复合菌液按1-1.5:1的质量比例混匀,在35-40℃的发酵罐中发酵2-3d,得到中草药发酵液;
S4,将中草药发酵液在超声波提取仪中提取1-2h,然后经-80℃的超低温冷冻干燥机进行干燥,得到提取物干粉;
S5,将提取物干粉与磨成粉末的酵母硒片按1000:1-1.5的比例混匀,制得抗肉鸡应激的富硒中草药添加剂。
优选的,所述抗肉鸡应激的富硒中草药添加剂的制备方法,S3中,发酵复合菌液质量百分比浓度为0.2%-0.5%。
优选的,所述抗肉鸡应激的富硒中草药添加剂的制备方法,S4中,超声提取仪的参数为温度40-50℃,功率为300-400W,频率25-30kH。
本发明的有益效果为:
1)本添加剂组方具有镇静解痉、抗惊厥、抗菌、增强免疫功能、健脾益气及健胃消食等功效。其中,构树叶含蛋白、黄酮甙、酚类、有机酸和鞣质,具有清热、凉血、利湿及杀虫等功效。青蒿含有青蒿素、青蒿内酯、山山柰酚和槲皮黄素,具有解暑清热,除湿杀虫及凉血截疟等功效。柴胡含有黄酮、多元醇、油酸、多糖、香豆素及微量元素等成份。小地松含有蛋白、有机酚、维生素,具有镇静安神、止咳平喘、驱虫止泻功效。银杏叶含有黄酮、银杏内酯及白果内酯等成分。具有强大的抗氧化、抗炎和抗过敏功效。刺五加含有刺五加甙B、刺五加甙E、挥发油、鞣质、棕桐酸、亚麻仁油酸、维生素,具有补肾健脾、益气安神及强筋骨等功效。霍香含有甲基胡椒酚、茴香醛、黄酮及藿香甙的成分,具有藿香酮和广藿香醇。其它成分有苯甲醛、丁香油酚、桂皮的功效与作用醛等,具有抗真菌作用、杀虫、清热利胆等功效。山楂含有多糖、黄酮、柠檬酸、皂苷、硒及维生素等营养成分,具有健胃消化、抗氧化、增强免疫及抗癌的功效。成分丰富,功效全面,有效提高肉鸡抗应激能力。
2)本添加剂富含易于肉鸡吸收的酵母硒,能够协同锌、铜及维生素清除自由基;发挥抗病毒作用;调节机体体液免疫和细胞免疫功能;并且具有抗衰老、抗疲劳、抗辐射及抗癌等功效。
具体实施方式
下面结合具体实施例对本发明进行详细说明,但应当理解本发明的保护范围并不受具体实施方式的限制。下列实施例中未注明具体条件的方法,通常按照常规条件操作,未注明的材料来源均为市售,由于不涉及发明点,故不对其步骤进行详细描述。
实施例1
一种抗肉鸡应激的富硒中草药添加剂,由中草药提取物和磨成粉末的酵母硒片按照1000:1.3的质量比混合而成;所述中草药提取物由如下重量份的中药制备而成:构树叶30份、青蒿15份、柴胡12份、小地松14份、银杏叶25份、刺五加7份、霍香10份、山楂12份。
一种抗肉鸡应激的富硒中草药添加剂的制备方法,包括如下步骤:
S1,按照实施例1的配方称取各中草药,在50℃烘箱内烘干,然后用中药超微粉碎机超微粉碎,过120目筛,制得中草药超微细粉,于6℃、干燥处保存;
S2,将中草药超微细粉与复合酶解液按1:25的质量比混合,在60℃恒温水浴锅中酶解30min,回流法除去溶剂(至固体表面无明显液体或液珠为止),在50℃鼓风干燥箱干燥1h,得中草药酶解粉,备用;所述复合酶解液是将纤维素酶和果胶酶溶于体积分数60%的乙醇溶液中制成的,且纤维素酶的浓度为15μg/g、果胶酶的浓度为90μg/g;
S3,将乳酸杆菌、双歧杆菌、酵母菌和嗜热链球菌的菌粉按照质量比为2:3:4:5的比例混合,得到的发酵复合菌溶于蒸馏水,制得发酵复合菌液,菌液质量百分比浓度为0.3%;将中草药酶解粉与发酵复合菌液按1.3:1的质量比例混匀,在37℃的发酵罐中发酵3d,得到中草药发酵液;
S4,将中草药发酵液在温度45℃,功率为350W,频率28kHz的超声波提取仪中提取1.5h,然后经-80℃的超低温冷冻干燥机进行干燥,得到提取物干粉;
S5,将中草药提取物与磨成粉末的酵母硒片按1000:1.3的质量比例混匀,制得抗肉鸡应激的富硒中草药添加剂。
实施例2
一种抗肉鸡应激的富硒中草药添加剂,由中草药提取物和磨成粉末的酵母硒片按照1000:1的质量比例混合而成;所述中草药提取物由如下重量份的中药原料制备而成:构树叶20份、青蒿10份、柴胡10份、小地松12份、银杏叶15份、刺五加5份、霍香6份、山楂8份。
一种抗肉鸡应激的富硒中草药添加剂的制备方法,包括如下步骤:
S1,按照实施例2的配方称取各中草药原料,在50℃烘箱内烘干,然后用中药超微粉碎机超微粉碎,过100目筛,制得中草药超微细粉,于4℃、干燥处保存;

S4,将中草药发酵液在温度40℃,功率为300W,频率25kHz的超声波提取仪中提取1h,然后经-80℃的超低温冷冻干燥机进行干燥,得到提取物干粉;
S5,将提取物干粉与磨成粉末的酵母硒片按1000:1的质量比例混匀,制得抗肉鸡应激的富硒中草药添加剂。
实施例3
一种抗肉鸡应激的富硒中草药添加剂,由中草药提取物和磨成粉末的酵母硒片按照1000:1.5的质量比例混合而成;所述中草药提取物由如下重量份的中药制备而成:构树叶35份、青蒿20份、柴胡15份、小地松15份、银杏叶35份、刺五加8份、霍香12份、山楂15份。
一种抗肉鸡应激的富硒中草药添加剂的制备方法,包括如下步骤:
S1,按照实施例3配方称取各中草药,在50℃烘箱内烘干,然后用中药超微粉碎机超微粉碎,过100目筛,制得中草药超微细粉,于10℃、干燥处保存;
S2,将中草药超微细粉与复合酶解液按1:30的质量比混合,在65℃恒温水浴锅中酶解40min,回流法除去溶剂(至固体表面无明显液体或液珠为止),在50℃鼓风干燥箱干燥1h,得中草药酶解粉,备用;所述复合酶解液是将纤维素酶和果胶酶溶于体积分数75%的乙醇溶液中制成的,且纤维素酶的浓度为25μg/g、果胶酶的浓度为110μg/g;
S3,将乳酸杆菌、双歧杆菌、酵母菌和嗜热链球菌的菌粉按照质量比为3:5:5:6的比例混合,溶于蒸馏水,制得发酵复合菌液,菌液质量百分比浓度为0.5%;将中草药酶解粉与发酵复合菌液按1.5:1的比例混匀,在40℃的发酵罐中发酵3d,得到中草药发酵液;
S4,将中草药发酵液在温度50℃,功率为400W,频率30kHz的超声波提取仪中提取2h,然后经-80℃的超低温冷冻干燥机进行干燥,得到提取物干粉;
S5,将提取物干粉与磨成粉末的酵母硒片按1000:1.5的质量比例混匀,制得抗肉鸡应激的富硒中草药添加剂。
在合肥某畜牧公司对本饲料添加剂的效果进行对比试验,选600只20日龄肉鸡,平均分成五组,除了饲料不同,其他条件完全相同的环境中让肉鸡尽可能正常饲养,饲养周期为30天。对照组的处理方式为:饲喂添加普通添加剂的饲料;空白组的处理方式为:饲喂不加添加剂的饲料;实验组1的处理方式为:饲喂添加实施例1制得的添加剂;实验组2的处理方式为:饲喂添加实施例2制得的添加剂;实验组3的处理方式为:饲喂添加实施例3制得的添加剂。并且对照组和实验组添加剂的加入量同为饲料总重量的0.3%。
统计试验生长过程中肉鸡的平均日采食量、平均日增重和平均生病率,其中平均发病率为所有生病次数与肉鸡数量的比值。经统计,结果如表1所示:
由表1可知,与对照组和空白组相比,饲喂本发明制备的富硒中草药提取物添加剂的肉鸡平均日采食量和平均日增重均有所提高,发病率明显下降;抗氧化酶SOD活性有所升高,氧化应激产物MDA有所降低;表明本发明的富硒中草药添加剂有效降低应激的不良影响,提高调节的免疫功能,改善肉鸡的消化吸收功能和生长性能,发挥了营养物质和药物的双重作用。

本饲料添加剂营养成分和药用成分也可以应用于其他畜禽养殖。
需要说明的是,本发明采用的酵母硒片为市售的牡丹江灵泰药业股份有限公司的西维尔酵母硒片,对照组的普通添加剂为山东龙昌动物保健品有限公司的龙昌胆汁酸。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。

1.一种抗肉鸡应激的富硒中草药添加剂,其特征在于,所述富硒中草药添加剂由中草药提取物和磨成粉末的酵母硒片按照1000:1-1.5的质量比混合而成;所述中草药提取物由如下重量份的中药原料制备而成:构树叶20-35份、青蒿10-20份、柴胡10-15份、小地松12-15份、银杏叶15-35份、刺五加5-8份、霍香6-12份、山楂8-15份。
2.根据权利要求1所述的抗肉鸡应激的富硒中草药添加剂在饲料中的使用方法,其特征在于,所述添加剂的加入量占饲料总重量的0.5-1%。
3.一种如权利要求1所述的抗肉鸡应激的富硒中草药添加剂的制备方法,其特征在于,包括如下步骤:
S1,按照以下重量份称取各中草药原料:构树叶20-35份、青蒿10-20份、柴胡10-15份、小地松12-15份、银杏叶15-35份、刺五加5-8份、霍香6-12份、山楂8-15份;并将称取的各中草药原料在50℃下烘干,将烘干的中草药原料混合,然后用中药超微粉碎机超微粉碎,过100目筛,制得中草药超微细粉,于4-10℃、干燥处保存;
S2,将中草药超微细粉与复合酶解液按1:20-30的质量比混合,在55℃-65℃下酶解25-40min,回流法除去溶剂,50℃烘干1h,得中草药酶解粉,备用;所述复合酶解液是将纤维素酶和果胶酶溶于体积分数60%-75%的乙醇溶液中制成的,且纤维素酶的浓度为10-25μg/g、果胶酶的浓度为80-110μg/g;
S3,将乳酸杆菌、双歧杆菌、酵母菌和嗜热链球菌的菌粉按照质量比为1-3:2-5:3-5:3-6的比例混合,得到的发酵复合菌溶于蒸馏水,制得发酵复合菌液;将中草药酶解粉与发酵复合菌液按1-1.5:1的质量比例混匀,在35-40℃下发酵2-3d,得到中草药发酵液;
S4,将中草药发酵液在超声波提取仪中提取1-2h,然后经-80℃干燥,得到提取物干粉,即中草药提取物;
S5,将中草药提取物与磨成粉末的酵母硒片按1000:1-1.5的质量比例混匀,制得抗肉鸡应激的富硒中草药添加剂。
4.根据权利要求2所述的抗肉鸡应激的富硒中草药添加剂的制备方法,其特征在于,S3中,发酵复合菌液中发酵复合菌的质量百分比浓度为0.2%-0.5%。
5.根据权利要求2所述的抗肉鸡应激的富硒中草药添加剂的制备方法,其特征在于,S4中,超声提取仪的参数为温度40-50℃,功率为300-400W,频率25-30kH。
