Abstract
本发明公开了一种中药微生态制剂,属于兽用中医药技术领域及饲料添加剂技术领域。该中药微生态制剂主要由以下原料制备而成:原料药以重量份数计为:党参2~10份、黄芪2~10份、茯苓2~8份、六神曲1~5份、麦芽1~6份、山楂1~5份、甘草1~3份、槟榔1~3份,菌液以体积分数计为:嗜酸乳杆菌菌液1~3份、粪肠球菌菌液1~6份、枯草芽孢杆菌菌液1~8份,菌液的总重量与原料药的总重量的比值为(1~5):(45~55)。本发明将上述8味纯天然中草药与三种益生菌复配制备微生态制剂,可以在动物体内产生多种水解酶,促进蛋白、淀粉、脂肪和半纤维素的分解,有利于饲料中营养物质的消化吸收。

背景技术
随着畜牧业的迅速发展,抗生素、化学合成药物、激素类药物作为饲料添加剂的种种弊端逐渐被人们所认识。如抗生素在杀死有害病菌的同时也杀死了肠道内的有益生物,造成肠道内微生物菌群不平衡。另外,畜禽产品中残留的药物会严重影响人类疾病的治疗和遗传,直接影响人类的健康。因此,寻找安全可靠、效果显著的禽类饲料添加剂成为亟待解决的问题。
在我国中草药以其天然、功能多样、安全可靠、经济环保的优势作为饲料添加剂的历史由来已久。中草药饲料添加剂,顾名思义是以中草药为原料制成的饲料添加剂,虽然有些学者将其归入非营养性饲料添加剂,按国家审批和管理也归入药物类饲料添加剂。然而,由于中药既是药物也是天然产物,含有多种有效成分,基本具有饲料添加剂的所有作用,可作为独立的一类饲料添加剂。我国应用中药作为饲料添加剂的历史悠久,早在两千多年前就开始用来促进动物生长,增重和防治疾病。

一种中药微生态制剂,主要由以下原料制备而成:原料药以重量份数计为:党参2~10份、黄芪2~10份、茯苓2~8份、六神曲1~5份、麦芽1~6份、山楂1~5份、甘草1~3份、槟榔1~3份,菌液以体积分数计为:嗜酸乳杆菌菌液1~3份、粪肠球菌菌液1~6份、枯草芽孢杆菌菌液1~8份,菌液的总重量与原料药的总重量的比值为3:50(1~5):(45~55)。
优选的,一种中药微生态制剂,主要由以下原料制备而成:原料药以重量份数计为:党参4份、黄芪4份、茯苓4份、六神曲2份、麦芽2份、山楂2份、甘草1份、槟榔1份,菌液以体积分数计为:嗜酸乳杆菌菌液2份、粪肠球菌菌液3份、枯草芽孢杆菌菌液5份,菌液的总重量与原料药的总重量的比值为3:50。
所述的嗜酸乳杆菌菌液的浓度为0.5×108~1.0×108CFU/ml。
所述的粪肠球菌菌液的浓度为0.5×108~1.0×108CFU/ml。
所述的枯草芽孢杆菌菌液的浓度为0.5×108~1.0×108CFU/ml。
各原料药功效简述如下:
党参:为桔梗科党参属植物党参、素花党参、川党参、管花党参、球花党参、灰毛党参的根;含有糖类如果糖、菊糖、多糖和4种杂多糖CP1、CP2、CP3、CP4,苷类如丁香苷、正己基-β-吡喃葡萄糖苷、乙基-α-D-呋喃果糖苷、党参苷,生物碱和含氮成分如胆碱、黑麦草碱等,还含有丁香醛、香草酸、2-呋喃羧酸、苍术内酯Ⅰ、Ⅱ等多种活性成分;党参性味甘,平,归脾、肺经;能健脾补肺、益气生津;主治脾胃虚弱、食少便溏、四肢乏力、肺虚喘咳、气短自汗、气血两亏诸证;现代药理学证明党参具有增强机体应激能力、增强机体免疫功能、延缓衰老、抗溃疡、抗肿瘤辅助作用等。
黄芪:为豆科黄芪属植物蒙古黄芪和膜荚黄芪的根;含有皂苷类成分如黄芪苷,异黄酮成分如毛蕊异黄酮,脂肪酸类如棕榈酸和亚油酸,多糖类如黄芪多糖、杂多糖等,以及近二十多种微量元素;黄芪性味甘,温,归肺、脾经;能益气升阳,固表止汗,利水消肿,托毒生肌;主治一切气虚血亏之症,如脾虚泄泻,肺虚咳嗽,脱肛,子宫下垂,自汗,盗汗,水肿,血痹,痈疽难溃或久溃不敛;现代药理学证明黄芪能影响机体代谢具有抗氧化作用,还能抗血栓、抗病毒、抗关节炎、抗癌、抗炎镇痛,保护心脑血管系统和促进造血功能。
茯苓:为多孔菌科卧孔属真菌茯苓的菌核;含有三萜类如茯苓酸、16α-羟基齿孔酸、β-对羟基苯酰基去氢齿孔酸等,多糖类如茯苓聚糖、茯苓次聚糖及高度、分支的β-D-葡聚糖等,还含有麦角甾醇、辛酸、十一烷酸、月桂酸、十二碳烯酸、棕榈酸、十二碳烯酸脂、辛酸脂及无机元素等多种活性成分;性味甘、淡,平,归心、脾、肺、肾经;能利水渗湿、健脾和胃、宁心安神;主治小便不利、水肿胀满、痰饮咳逆、呕吐、脾虚食少、泄泻、心悸不安、失眠健忘等;现代药理学证明茯苓具有利尿、抗癌及免疫增强作用等。

甘草:为豆科甘草属植物甘草、光皮甘草、胀果甘草的根及根茎;含有三萜皂苷如甘草甜素、乌拉尔甘草皂苷A、B等,黄酮类化合物如甘草苷元、甘草苷等,香豆素类化合物如甘草香豆素、甘草酚等,生物碱类如喹啉类、异喹啉类等,多糖如甘草葡聚糖GBW、甘草多糖UA、UB、UC等,还含有甘草苯丙呋喃、正二十三烷等多种有效成分;甘草性味甘、平,归脾、胃、心、肺经;能和中缓急、润肺、解毒、调和诸药;炙用治脾胃虚弱、倦怠食少、腹痛便溏、四肢挛急疼痛、心悸、脏躁、肺痿咳嗽,生用治咽喉肿痛、痈疮肿毒、小儿胎毒及药物、食物中毒等;现代药理学证明甘草具有抗微生物、抗炎、镇咳、祛痰、抗肿瘤、抗突变、抗氧化等功效,对消化系统、心血管系统、生殖系统及脑、肾功能等具有调节作用。
槟榔:为棕榈科槟榔属植物槟榔的种子;含有生物碱类如槟榔碱、槟榔次碱、去甲基槟榔碱、去甲基槟榔次碱、槟榔副碱、高槟榔碱等,鞣质如右旋儿茶素、左旋表儿茶素、原矢车菊素A-1、B-1和B-2以及槟榔鞣质A、B两类化合物,还含有脂肪酸如月桂酸、肉豆蔻酸、棕榈酸、硬脂酸、右旋等以及氨基酸类如脯氨酸、色氨酸、甲硫氨酸、酪氨酸等多种活性成分;性味苦、辛,温,归胃、大肠经;能驱虫消积、下气行水、截疟;主治虫积、食滞、脘腹胀痛、泻痢后重等;现代药理学证明槟榔具有驱虫、抗病原微生物、灭螺等作用。
一种中药微生态制剂,可以采用常规方法制备。本发明仅提供一种具体的制备方法,包括以下步骤:
(1)按照重量份数准确取党参、黄芪、茯苓、六神曲、麦芽、山楂、甘草和槟榔,烘干后粉碎、过筛,再加水煎煮3次,合并3次煎液,滤过得药液备用;
(2)按照体积份数准确取嗜酸乳杆菌菌液、粪肠球菌菌液和枯草芽孢杆菌菌液,混匀后根据菌液总量与原料药总量之间的比值,将混合菌液加入到步骤(1)的药液中,在密闭条件下厌氧发酵,发酵产物经冷冻干燥即得。
所述步骤(1)中的烘干温度为60~80℃。
所述步骤(1)中的过筛为过60~100目筛。
所述步骤(1)中加水煎煮为每次煎煮加5~6倍量的水,煎煮1~2小时。
所述步骤(2)中嗜酸乳杆菌菌液的制备方法为:先将嗜酸乳杆菌接种于斜面培养基中,将斜面培养基上产生的悬浮液加入到MRS液体培养基中扩大培养,至对数生长期时菌液的浓度为0.5×108~1.0×108CFU/ml即得。
所述步骤(2)中粪肠球菌菌液的制备方法为:先将粪肠球菌接种于斜面培养基中,将斜面培养基上产生的悬浮液加入到MRS液体培养基中扩大培养,至对数生长期时菌液的浓度为0.5×108~1.0×108CFU/ml即得。
所述步骤(2)中枯草芽孢杆菌菌液的制备方法为:先将枯草芽孢杆菌接种于斜面培养基中,将斜面培养基上产生的悬浮液加入到LB液体培养基中扩大培养,至对数生长期时菌液的浓度为0.5×108~1.0×108CFU/ml即得。
所述步骤(2)中厌氧发酵的温度为30~37℃,发酵时间为36~72小时。
一种中药微生态制剂作为饲料添加剂的应用,包括以下步骤:以重量百分比计,将中药微生态制剂以0.2~0.4%的量加入肉鸡饲料中即可。
本发明的有益效果:
本发明是在中兽医学卫气营血辩证理论方法的指导下,筛选出党参、黄芪、茯苓、六神曲、麦芽、山楂、甘草、槟榔共8味纯天然中草药,按照君臣佐使原则配伍组方。方中党参、黄芪健脾益气为君;辅以六神曲、山楂、麦芽消积开胃;佐以茯苓利水渗湿、健胃脾补中,槟榔消积杀虫;使以甘草调和诸药。诸药合用能增强机体免疫力,促进机体生长。
本发明采用嗜酸乳杆菌、粪肠球菌和枯草芽孢杆菌制备微生态制剂,其中嗜酸乳杆菌能调整肠道菌群平衡,抑制肠道不良微生物的增殖,分泌抗生物素类物质如嗜酸乳菌素、嗜酸杆菌素、乳酸菌素等,对肠道致病菌产生拮抗作用;粪肠球菌有利于改善肠道内微生态平衡,防治动物肠道菌丛区系紊乱,还能分解蛋白质为小肽,合成B族维生素,增强巨嗜细胞的活性,促进动物的免疫反应,提高抗体水平等;枯草芽孢杆菌能迅速消耗环境中的游离氧,造成肠道低氧,促进有益厌氧菌生长,并产生乳酸等有机酸类,降低肠道PH值,间接抑制其它致病菌生长。刺激动物免疫器官的生长发育,激活T、B淋巴细胞,提高免疫球蛋白和抗体水平,增强细胞免疫和体液免疫功能,提高群体免疫力。三种益生菌联用可通过动物消化道生物的竞争性排斥作用,抑制病原菌繁殖,阻止病原菌吸附于肠壁,促进其排出体外;补充有益菌群,调整动物肠道菌群微生态平衡;发挥营养助消化作用,提高饲料转化率,合成营养物质,促进动物生长;还可通过增强非特异性免疫功能来预防疾病,提高机体免疫力;抑制肠道内氨及胺等毒性物质的产生,改善环境卫生,净化环境。
本发明采用纯天然中草药,原料药易得,价格低廉,安全、无有害残留,对畜禽机体的毒副作用小,不易产生抗药性,主要通过整体作用、双向调节、组合效应、扶正祛邪发挥药物功效。中草药在养殖过程中起到治疗疾病和预防保健的作用,其进入机体后不仅直接作用于细菌和病毒,还能作用于机体的免疫器官,帮助提高抵抗力,诱导机体产生内源性干扰素,利用机体身的排异功能,驱逐细菌和病毒。中草药治病不但病愈,而且还帮助机体恢复体质,排出毒素,一举两得。经过微生物发酵过的中药,其中的中药成分经过微生物转化,由大分子变成小分子,起效快、药效高、100%被机体吸收;同时发酵过程可以分解中草药的毒性物质,使中药制剂真正无毒副作用。
本发明将上述8味纯天然中草药与三种益生菌复配制备微生态制剂,可以在动物体内产生多种水解酶,促进蛋白、淀粉、脂肪和半纤维素的分解,有利于饲料中营养物质的消化吸收,同时可合成营养物质以及一些未知促生长因子参与新陈代谢,促进动物生长;微生态制剂作为饲料添加剂进行保健和治疗,可以达到事半功倍的效果。
本发明制备微生态制剂的方法简单、易操作,使用方便,成本低,适于大规模生产及推广应用。
具体实施方式
下述实施例仅对本发明作进一步详细说明,但不构成对本发明的任何限制。
实施例1
本实施例中中药微生态制剂,由以下原料制备而成:原料药以重量份数计为:党参2份、黄芪10份、茯苓2份、六神曲5份、麦芽1份、山楂5份、甘草1份、槟榔3份,菌液以体积分数计为:嗜酸乳杆菌菌液1份、粪肠球菌菌液6份、枯草芽孢杆菌菌液1份,菌液的总重量与原料药的总重量的比值为1:55。
本实施例中中药微生态制剂的制备方法,包括以下步骤:
(1)按照重量份数准确取党参、黄芪、茯苓、六神曲、麦芽、山楂、甘草和槟榔,60℃烘干后粉碎,过60目筛,再加水煎煮3次,每次煎煮加5倍量的水,煎煮1.5小时,合并3次煎液,滤过得药液备用;
(2)按照体积份数准确取嗜酸乳杆菌菌液、粪肠球菌菌液和枯草芽孢杆菌菌液,混匀后根据菌液总量与原料药总量之间的比值,将混合菌液加入到步骤(1)的药液中,在密闭条件下厌氧发酵,发酵的温度为30℃,发酵时间为72小时,将发酵产物冷冻干燥即得。
所述步骤(2)中嗜酸乳杆菌菌液、粪肠球菌菌液、枯草芽孢杆菌菌液的制备方法为:先将嗜酸乳杆菌、粪肠球菌、粪肠球菌分别接种于各自的斜面培养基中,将各个斜面培养基上产生的悬浮液依次加入到MRS液体培养基、MRS液体培养基、LB液体培养基中扩大培养,至对数生长期时菌液的浓度分别为0.5×108~1.0×108CFU/ml即得。
本实施例中中药微生态制剂作为饲料添加剂的应用,包括以下步骤:以重量百分比计,将上述制备的中药微生态制剂以0.3%的量加入肉鸡饲料中即可。
实施例2
本实施例中中药微生态制剂,由以下原料制备而成:原料药以重量份数计为:党参4份、黄芪4份、茯苓4份、六神曲2份、麦芽2份、山楂2份、甘草1份、槟榔1份,菌液以体积分数计为:嗜酸乳杆菌菌液2份、粪肠球菌菌液3份、枯草芽孢杆菌菌液5份,菌液的总重量与原料药的总重量的比值为3:50。
本实施例中中药微生态制剂的制备方法,包括以下步骤:
(1)按照重量份数准确取党参、黄芪、茯苓、六神曲、麦芽、山楂、甘草和槟榔,80℃烘干后粉碎,过100目筛,再加水煎煮3次,每次煎煮加6倍量的水,煎煮2小时,合并3次煎液,滤过得药液备用;
(2)按照体积份数准确取嗜酸乳杆菌菌液、粪肠球菌菌液和枯草芽孢杆菌菌液,混匀后根据菌液总量与原料药总量之间的比值,将混合菌液加入到步骤(1)的药液中,在密闭条件下厌氧发酵,发酵的温度为37℃,发酵时间为36小时,将发酵产物冷冻干燥即得。
所述嗜酸乳杆菌菌液、粪肠球菌菌液、枯草芽孢杆菌菌液的制备方法同上。
本实施例中中药微生态制剂作为饲料添加剂的应用,包括以下步骤:以重量百分比计,将上述制备的中药微生态制剂以0.3%的量加入肉鸡饲料中即可。
实施例3
本实施例中中药微生态制剂,由以下原料制备而成:原料药以重量份数计为:党参10份、黄芪2份、茯苓8份、六神曲1份、麦芽6份、山楂1份、甘草3份、槟榔1份,菌液以体积分数计为:嗜酸乳杆菌菌液3份、粪肠球菌菌液1份、枯草芽孢杆菌菌液8份,菌液的总重量与原料药的总重量的比值为1:9。
本实施例中中药微生态制剂的制备方法,包括以下步骤:
(1)按照重量份数准确取党参、黄芪、茯苓、六神曲、麦芽、山楂、甘草和槟榔,70℃烘干后粉碎,过80目筛,再加水煎煮3次,每次煎煮加5倍量的水,煎煮1.5小时,合并3次煎液,滤过得药液备用;
(2)按照体积份数准确取嗜酸乳杆菌菌液、粪肠球菌菌液和枯草芽孢杆菌菌液,混匀后根据菌液总量与原料药总量之间的比值,将混合菌液加入到步骤(1)的药液中,在密闭条件下厌氧发酵,发酵的温度为35℃,发酵时间为50小时,将发酵产物冷冻干燥即得。
所述嗜酸乳杆菌菌液、粪肠球菌菌液、枯草芽孢杆菌菌液的制备方法同上。
本实施例中中药微生态制剂作为饲料添加剂的应用,包括以下步骤:以重量百分比计,将上述制备的中药微生态制剂以0.3%的量加入肉鸡饲料中即可。
对比例1
本对比例中肉鸡用中药组合物,由以下重量份数的原料药组成:党参4份、黄芪4份、茯苓4份、六神曲2份、麦芽2份、山楂2份、甘草1份、槟榔1份。制备方法包括以下步骤:按照重量份数准确取各味原料药,粉碎后混匀、过60目筛即得。
本对比例中肉鸡用中药组合物作为饲料添加剂的应用,包括以下步骤:以重量百分比计,将上述制备的粉剂以0.3%的量加入肉鸡饲料中即可。
试验例
试验动物:从某养鸡场中随机选取150羽1日龄肉鸡,随机分为5组,每组30羽。
试验组别:试验组1~3依次饲喂实施例1~3制备的肉鸡饲料,对照组饲喂对比例1制备的肉鸡饲料;空白对照组饲喂普通肉鸡饲料(普通肉鸡饲料中不含有任何药物成分)。
试验方法:取实施例1~3制备的肉鸡饲料和普通肉鸡饲料,连续45天定时定量饲喂1日龄肉鸡,记录肉鸡的初始重量和试验末重量,统计日增重量和料重比,结果详见下表1。
表1各处理组中肉鸡增重及料重比
结论:从表1可以看出,试验组的日增重量约为20g/羽,料重比为1.68~1.87;对照组的日增重量约为18.61/羽,料重比1.98;空白对照组的日增重量约为17.10g/羽,料重比2.2。统计分析表明,试验组的日增重量显著高于对照组及空白对照组(P<0.01),料重比显著低于对照组及空白对照组(P<0.01)。
试验结果表明,本发明制备的微生态制剂能显著提高肉鸡日增重,降低料重比,对肉鸡增重具有显著的促进作用,适于实际饲养应用。整个用药期间试验组鸡群未发现任何异常,表明本发明实施例制备的微生态制剂的毒副作用小。
Claims (9)
1.一种中药微生态制剂,其特征在于:主要由以下原料制备而成:原料药以 重量份数计为:党参2~10份、黄芪2~10份、茯苓2~8份、六神曲1~5份、麦芽1~6 份、山楂1~5份、甘草1~3份、槟榔1~3份,菌液以体积分数计为:嗜酸乳杆菌菌液1~ 3份、粪肠球菌菌液1~6份、枯草芽孢杆菌菌液1~8份,菌液的总重量与原料药的总重 量的比值为(1~5):(45~55)。
2.根据权利要求1所述的中药微生态制剂,其特征在于:主要由以下原料制 备而成:原料药以重量份数计为:党参4份、黄芪4份、茯苓4份、六神曲2份、麦芽2 份、山楂2份、甘草1份、槟榔1份,菌液以体积分数计为:嗜酸乳杆菌菌液2份、粪肠 球菌菌液3份、枯草芽孢杆菌菌液5份,菌液的总重量与原料药的总重量的比值为3:50。
3.一种如权利要求1或2所述的中药微生态制剂的制备方法,其特征在于: 包括以下步骤:
(1)按照重量份数准确取党参、黄芪、茯苓、六神曲、麦芽、山楂、甘草和槟榔, 烘干后粉碎、过筛,再加水煎煮3次,合并3次煎液,滤过得药液备用;
(2)按照体积份数准确取嗜酸乳杆菌菌液、粪肠球菌菌液和枯草芽孢杆菌菌液,混 匀后根据菌液总量与原料药总量之间的比值,将混合菌液加入到步骤(1)的药液中,在 密闭条件下厌氧发酵,发酵产物经冷冻干燥即得。
4.根据权利要求3所述的中药微生态制剂的制备方法,其特征在于:所述步 骤(1)中加水煎煮为每次煎煮加5~6倍量的水,煎煮1~2小时。
5.根据权利要求3所述的中药微生态制剂的制备方法,其特征在于:所述步 骤(2)中嗜酸乳杆菌菌液的制备方法为:先将嗜酸乳杆菌接种于斜面培养基中,将斜面 培养基上产生的悬浮液加入到MRS液体培养基中扩大培养,至对数生长期时菌液的浓度 为0.5×10 8~1.0×10 8CFU/ml即得。
6.根据权利要求3所述的中药微生态制剂的制备方法,其特征在于:所述步 骤(2)中粪肠球菌菌液的制备方法为:先将粪肠球菌接种于斜面培养基中,将斜面培养 基上产生的悬浮液加入到MRS液体培养基中扩大培养,至对数生长期时菌液的浓度为 0.5×10 8~1.0×10 8CFU/ml即得。
7.根据权利要求3所述的中药微生态制剂的制备方法,其特征在于:所述步 骤(2)中枯草芽孢杆菌菌液的制备方法为:先将枯草芽孢杆菌接种于斜面培养基中,将 斜面培养基上产生的悬浮液加入到LB液体培养基中扩大培养,至对数生长期时菌液的浓 度为0.5×10 8~1.0×10 8CFU/ml即得。
8.根据权利要求3所述的中药微生态制剂的制备方法,其特征在于:所述步 骤(2)中厌氧发酵的温度为30~37℃,发酵时间为36~72小时。
9.一种如权利要求1或2所述的中药微生态制剂作为饲料添加剂的应用,其 特征在于:包括以下步骤:以重量百分比计,将中药微生态制剂以0.2~0.4%的量 加入肉鸡饲料中即可。
