Abstract
The invention discloses a method for preparing nutrient solution for seedling-stage germination of bletilla striata, belonging to the field of traditional Chinese pharmacology, wherein the method for preparing the nutrient solution for seedling-stage germination of the bletilla striata breaks through only one layer of cell seed coat in embryo water absorption and swelling to generate false roots; protocorm and leaf primordium formation phase; the protocorm continues to expand, and three nutrient solution formulas are respectively prepared during three periods of the true root growth period. The bletilla striata seeds treated by the nutrient solution have high germination rate and short germination time, wherein the germination rate is 95 percent, and the germination time is 15 days.
Description
Technical Field
The invention relates to the field of traditional Chinese pharmacology, in particular to a preparation method of a nutrient solution for seedling-stage germination of bletilla striata in the Dabie mountain.
Background
Bletilla striata is a perennial herb bulbous plant, and the plant height is 18-60 cm. Mainly distributed in china, japan and northern burma. The main flowering period is in spring, but depending on the climate in each area, flowers may appear from late winter to early summer. Bletilla striata has wide medicinal value and garden value. It is mainly used for astringing to stop bleeding, eliminating swelling and promoting granulation. The flower has purple red, white, blue, yellow and pink colors, can be viewed in a potted plant room, and can also be embellished on a shaded flower table, a flower environment or a corner of a courtyard. Many wild bletilla striata grow in the Dabie mountain region, and the wild bletilla striata is gradually cultivated artificially. However, some nutrient solution needs to be filled to help the growth of the bletilla striata during seedling germination, and the existing bletilla striata nutrient solution is not suitable for cultivating the bletilla striata in the Dabieshan mountain area. Bletilla striata in Dabieshan area has fine seeds without endosperm, nutrition needs to be added in the seed germination process, so that the contradiction between matrix configuration and nutritional ingredients is caused, and meanwhile, mixed bacteria needs to be controlled due to the addition of the nutritional ingredients, so that the problem that the conventional nutrient solution cannot be well solved.
Disclosure of Invention
The invention aims to solve the problem that the contradiction between matrix configuration and nutrient components caused by the addition of nutrition in the bletilla striata seed germination process and the problem that mixed bacteria need to be controlled due to the addition of the nutrient components are solved by providing a method for preparing nutrient solution for the seedling-stage germination of the wild bletilla striata in the Dabie mountain.
In order to achieve the purpose, the invention adopts the technical scheme that:
the preparation method of the nutrient solution for the seedling-stage germination of the bletilla striata in the Dabie mountain comprises the following steps:
(1) sowing and seed treatment: seed dressing agent and bletilla striata seeds are needed for seed dressing in seed sowing, a 40-mesh sieve is used for uniformly dressing the seeds, and naphthylacetic acid is used for treating the seeds, wherein the using concentration of the naphthylacetic acid is 2 mg/L;
(2) embryo water absorption and expansion break through only one layer of cell seed coat, rhizoid appears, the service time of the nutrient solution formula 1 is 15 days, and the nutrient solution formula 1: 1/4MS and 6-BA (1mg/L), with ppm antibacterial agent, at a concentration of 2 mg/L;
(3) protocorm and leaf primordium formation period, the using time of the nutrient solution formula 2 is 30 days, and the nutrient solution formula 2: 1/2MS and 6-BA (1mg/L), with ppm antibacterial agent, at a concentration of 2 mg/L;
(4) the protocorm continues to expand, the growth period of the true root is long, the service time of the nutrient solution formula 3 is 105 days, and the nutrient solution formula 3 is as follows: 1/2MS and 6-BA (1mg/L), and adding phosphoric diamine water and urea water in the middle at a volume ratio of 30: 1.
Preferably, the raw material of the seed dressing agent in the step (1) is lime powder and activated carbon, and the volume ratio of the lime powder to the activated carbon is 50: 1.
Has the advantages that: the preparation method of the nutrient solution for seedling-stage germination of the bletilla striata in the Dabie mountain wild species solves the problems that nutrition needs to be added in the germination process of the bletilla striata seeds, so that the contradiction between the configuration of a matrix and the nutrient components is caused, and meanwhile, the mixed bacteria needs to be controlled due to the addition of the nutrient components. The bletilla striata seeds treated by the nutrient solution have high germination rate and short germination time, wherein the germination rate is 95 percent, and the germination time is 15 days.
