Recording the height of the hypocotyl at 2016, 4 and 16 days (time A), recording the height of the seedling and the growth condition of the leaves at 2017, 2 and 1 days (B), detecting the SOD activity of the leaves, and calculating the survival rate. When the corresponding data of the acclimated group and the control group are marked with different letters, the acclimated group and the control group are shown to have obvious difference on the indexes corresponding to the data.
The test results shown in table 1 indicate that the domesticated composition activity rate is significantly higher than that of the control group. The SOD activity of the leaf of the control group 1 is obviously higher than that of the domesticated group, because the active oxygen of the leaf is increased in the process of resisting low temperature of the control group seedling, and the SOD activity is enhanced in order to eliminate redundant oxygen free radicals. The increase in SOD activity has been generally recognized as a physiological response of plants against cold stress. Therefore, the results show that the mangrove plant north-shift domestication method provided by the invention obviously improves the growth quality and survival rate of mangrove plants. Compared with the survival rate of the seedlings in the domesticated group and the survival rate of the seedlings in the control group 2 are about 8 percent, and the domestication method provided by the invention has the advantage that the cold-proof protection solution has a better domestication effect.
Examples 2 to 5
Examples 2 to 5, the same conditions as in example 1 were used except that the temperature acclimation conditions in the acclimation in step (3) were different; specifically, see table 2 for examples 2-5, T in table 2 indicates the minimum outdoor temperature, and table 3 for the results of the acclimatization test.

The present embodiment is the same as embodiment 1 except that the specific steps of alternately acclimatizing and culturing the high and low temperature of the hypocotyl of the kandelia candel planted in the planting field are different from embodiment 1, and the specific steps of alternately acclimatizing and culturing the high and low temperature are as follows:
a first stage acclimatization step: when the minimum outdoor temperature reaches 6 ℃, rolling up the roller shutter for low-temperature domestication for 10 days, wherein the minimum outdoor temperature T is ensured to be higher than 2 ℃; closing the roller shutter to raise the temperature in the shed to over 10 deg.c, and domesticating for 10 days;
a second stage domestication step: when the minimum outdoor temperature reaches 2 ℃, rolling up the roller shutter for low-temperature domestication for 10 days, wherein the minimum outdoor temperature T is ensured to be higher than 0 ℃; closing the roller shutter to raise the temperature in the shed to over 8 deg.c, and domesticating for 10 days;
a third stage acclimatization step: when the minimum outdoor temperature reaches 0 ℃, rolling up the roller shutter for low-temperature domestication for 5 days, wherein the minimum outdoor temperature T is ensured to be higher than-4 ℃; closing the roller shutter to raise the temperature in the shed to over 6 deg.c, and domesticating for 5 days; and finally, stabilizing the temperature of the greenhouse to be more than 6 ℃ until the spring of the next year. The results of the acclimatization test are shown in Table 4.
Example 7
The present embodiment is the same as embodiment 1 except that the specific steps of alternately acclimatizing and culturing the high and low temperature of the hypocotyl of the kandelia candel planted in the planting field are different from embodiment 1, and the specific steps of alternately acclimatizing and culturing the high and low temperature are as follows:
a first stage acclimatization step: when the outdoor minimum temperature reaches 10 ℃, rolling up the roller shutter for low-temperature domestication for 15 days, wherein the outdoor minimum temperature T is ensured to be higher than 6 ℃; closing the roller shutter to raise the temperature in the shed to over 15 deg.c, and domesticating for 15 days;
a second stage domestication step: when the minimum outdoor temperature reaches 6 ℃, rolling up the roller shutter for low-temperature domestication for 10 days, wherein the minimum outdoor temperature T is ensured to be higher than 2 ℃; closing the roller shutter to raise the temperature in the shed to over 10 deg.c, and domesticating for 10 days;
a third stage acclimatization step: when the minimum outdoor temperature reaches 2 ℃, rolling up the roller shutter for low-temperature domestication for 10 days, wherein the minimum outdoor temperature T is ensured to be higher than-4 ℃; closing the roller shutter to raise the temperature in the shed to over 8 deg.c, and domesticating for 10 days; and finally, stabilizing the temperature of the greenhouse to be more than 6 ℃ until the spring of the next year. The results of the acclimatization test are shown in Table 4.
Example 8
The present embodiment is the same as embodiment 1 except that the specific steps of alternately acclimatizing and culturing the high and low temperature of the hypocotyl of the kandelia candel planted in the planting field are different from embodiment 1, and the specific steps of alternately acclimatizing and culturing the high and low temperature are as follows:
a first stage acclimatization step: when the outdoor minimum temperature reaches 10 ℃, rolling up the roller shutter for low-temperature domestication for 15 days, wherein the outdoor minimum temperature T is ensured to be higher than 2 ℃; closing the roller shutter to raise the temperature in the shed to over 15 deg.c, and domesticating for 15 days;
a second stage domestication step: when the minimum outdoor temperature reaches 2 ℃, rolling up the roller shutter for low-temperature domestication for 10 days, wherein the minimum outdoor temperature T is ensured to be higher than 0 ℃; closing the roller shutter to raise the temperature in the shed to over 10 deg.c, and domesticating for 10 days;
a third stage acclimatization step: when the minimum outdoor temperature reaches 0 ℃, rolling up the roller shutter for low-temperature domestication for 5 days, wherein the minimum outdoor temperature T is ensured to be higher than-4 ℃; closing the roller shutter to raise the temperature in the shed to over 6 deg.c, and domesticating for 5 days; and finally, stabilizing the temperature of the greenhouse to be more than 6 ℃ until the spring of the next year. The results of the acclimatization test are shown in Table 4.

Claims (25)
1. A mangrove plant north-shift domestication method is characterized by sequentially comprising the following steps:
firstly, collecting well-developed mangrove plant embryonic axis, soaking the mangrove plant embryonic axis in seawater with the salinity of 12-30 per mill, and then transporting the mangrove plant embryonic axis to a planting place as soon as possible;
step two, inserting the embryonic axis of the mangrove plant into a planting field; selecting a mudflat with the latitude of 28-31 degrees N at the planting place; the salinity of seawater on the tidal flat is 12-30 per mill;
step three, building a greenhouse in the planting field before winter, ensuring that the tidal water can normally rise and fall into the greenhouse, and performing high-low temperature alternative domestication cultivation on the embryonic axis of the mangrove plant inserted in the planting field;
in the third step, the alternately domesticating and culturing at high and low temperature comprises:
a first stage domestication step, when the outdoor minimum temperature T is more than 9 ℃ and less than or equal to 10 ℃, rolling up the greenhouse roller shutter to start low-temperature domestication, wherein in the low-temperature domestication process, the outdoor minimum temperature T is ensured to be higher than 6 ℃, when the outdoor minimum temperature T is below 6 ℃, closing the greenhouse, and when the outdoor minimum temperature T rises back to be higher than 6 ℃, rolling up the greenhouse roller shutter to continue low-temperature domestication, wherein the total time of the low-temperature domestication is at least 5 days; closing the greenhouse after the low-temperature domestication is finished, and starting heat preservation domestication when the temperature in the greenhouse is raised to be above 15 ℃, wherein the total time of the heat preservation domestication is at least 5 days;
a second stage domestication step, when the outdoor minimum temperature T is more than 5 ℃ and less than or equal to 6 ℃, rolling up the greenhouse roller shutter to start low-temperature domestication, wherein in the low-temperature domestication process, the outdoor minimum temperature T is higher than 2 ℃, when the outdoor minimum temperature T is lower than 2 ℃, the greenhouse roller shutter is required to be closed, when the outdoor minimum temperature T rises back to be higher than 2 ℃, the greenhouse roller shutter is rolled up again to continue the low-temperature domestication, and the total time of the low-temperature domestication is at least 5 days; closing the greenhouse after the low-temperature domestication is finished, and starting heat preservation domestication when the temperature in the greenhouse is raised to be above 10 ℃, wherein the total time of the heat preservation domestication is at least 5 days;
a third stage acclimatization step, namely rolling up the greenhouse roller shutter to start low-temperature acclimatization when the outdoor minimum temperature T reaches more than 1 ℃ and less than or equal to 2 ℃, wherein in the low-temperature acclimatization process, the outdoor minimum temperature T is higher than 0 ℃, the greenhouse is required to be closed when the outdoor minimum temperature T is lower than 0 ℃, the greenhouse roller shutter is rolled up again to continue the low-temperature acclimatization when the outdoor minimum temperature T rises back to be higher than 0 ℃, and the total time of the low-temperature acclimatization is at least 3 days; closing the greenhouse after the low-temperature domestication is finished, and starting heat preservation domestication when the temperature in the greenhouse is raised to be more than 8 ℃, wherein the total time of the heat preservation domestication is at least 3 days;
a fourth stage of domestication, wherein when the outdoor minimum temperature T is greater than-1 ℃ and less than or equal to 0 ℃, the greenhouse roller blind is rolled up to start low-temperature domestication, in the low-temperature domestication process, the outdoor minimum temperature T is guaranteed to be higher than-4 ℃, when the outdoor minimum temperature T is lower than-4 ℃, the greenhouse is required to be closed, when the outdoor minimum temperature T is raised back to be higher than-4 ℃, the greenhouse roller blind is rolled up to continue low-temperature domestication, and the total time of the low-temperature domestication is at least 2 days; closing the greenhouse after the low-temperature domestication is finished, and starting heat preservation domestication when the temperature in the greenhouse is raised to be higher than 6 ℃, wherein the total time of the heat preservation domestication is at least 2 days; finally, stabilizing the temperature in the greenhouse to be more than 6 ℃ until the greenhouse is opened in the spring of the next year to ensure that the mangrove plants grow normally under natural conditions;
spraying cold-proof protective liquid to the mangrove when the minimum outdoor temperature is reduced to 10 ℃;
the cold-proof protective liquid comprises the following raw materials in parts by weight: 10-80 parts of micro-nano shell powder, 10-90 parts of sodium alginate, 1-15 parts of glucose and 3-25 parts of glycerol.
2. The method for domesticating mangrove plants in the north according to claim 1, wherein in the first step,
the embryonic axis of the mangrove plant is the first premature embryonic axis or the eight-nine mature embryonic axis.
3. The method for domesticating mangrove plants in the north according to claim 1, wherein in the first step,
the mangrove plant hypocotyl is from Fujian, and the collection time is 4-5 months per year.
4. The method for domesticating mangrove plants in the north according to claim 1, wherein in the first step,
the soaking time is 1-10 h.
5. The method for domesticating mangrove plants in the north according to claim 4, wherein in the first step,
the soaking time is 60-120 min.
6. The method for domesticating mangrove plants in the north according to claim 1, wherein in the first step,
the soaking treatment is to soak the mangrove plant hypocotyls in seawater collected from the land;
the transportation days in the process of transporting the hypocotyls of the mangrove plants from the collection land to the planting land are not more than 4 days.
7. The method for domesticating mangrove plants in the north according to claim 1, wherein in the first step,
during transportation, the seedling tops of the embryonic axes of the mangrove plants are upwards placed in the ventilating container, and straws are placed at the bottom layer, so that good ventilation is ensured.
8. The method for domesticating mangrove plants in the north according to any one of claims 1 to 7, wherein in the second step, the embryonic axis of the mangrove plants is sterilized and disinfected before the transplanting.
9. The mangrove plant north-shift domestication method according to claim 8, characterized in that the sterilization and disinfection treatment is soaking for 10-15h with 0.1-0.2 wt% potassium permanganate solution.
10. The mangrove plant north-shift domestication method according to any one of claims 1-7, characterized in that in the second step, the transplanting mode is as follows: and (3) transplanting a mangrove plant hypocotyl in each hole, wherein the transplanting depth is 6-10cm, and the plant spacing and the row spacing are not less than 30 cm.
11. The method for domesticating mangrove plants in the north according to claim 10, wherein in the second step, the transplanting mode is as follows: and (3) punching by using a wood bar with the diameter of 2-3cm, and then performing the transplanting.
12. The method for domesticating mangrove plants in the north according to claim 10, wherein in the second step, the transplanting mode is as follows: the transplanting selection is carried out 1-2 days after the current month climax, and the transplanting selection is carried out after the ebb tide in the cloudy day.
13. The mangrove plant north-shift domestication method according to any one of claims 1 to 7, characterized in that in the third step, the planting field adopts a mixture of soil and sand as a transplanting medium.
14. The mangrove plant north-shift domestication method of claim 13, wherein the soil is sea mud of the planting field.
15. The mangrove plant north-shift domestication method according to claim 13, wherein the sand has a particle size of 0.2-2 mm.
16. The mangrove plant north-shift domestication method according to claim 13, wherein the weight ratio of soil to sand in the transplanting culture medium is 10: 1-8: 1.
17. The method for domesticating mangrove plants in the north according to claim 1, wherein the seawater salinity of the tidal flat is 14-19 per mill.
18. The mangrove plant north-shift domestication method according to claim 13, wherein the greenhouse is a roller shutter greenhouse.
19. The mangrove plant north-shift acclimatization method according to any one of claims 1 to 7, wherein the step of alternately acclimatizing at high and low temperatures is repeated in winter of the second year.
20. The mangrove plant north-shift acclimatization method according to any one of claims 1 to 7, wherein in the third step,
in the first stage domestication step, the total time of the low-temperature domestication is 12-18 days, and the total time of the heat preservation domestication is 12-18 days;
in the second stage domestication step, the total time of the low-temperature domestication is 8-12 days, and the total time of the heat preservation domestication is 8-12 days;
in the third stage acclimation step, the total time of the low-temperature acclimation is 8-12 days, and the total time of the heat preservation acclimation is 8-12 days;
in the fourth stage acclimation step, the total time of the low-temperature acclimation is 3-8 days, and the total time of the heat preservation acclimation is 3-8 days.
21. The mangrove plant north-shift domestication method according to claim 1, characterized in that holes are periodically punched on the surface of the beach.
22. The mangrove plant north-shift domestication method according to claim 1, characterized in that the cold-proof protection liquid comprises the following raw materials by weight: 20-50 parts of micro-nano shell powder, 30-60 parts of sodium alginate, 2-10 parts of glucose and 5-20 parts of glycerol.
23. The mangrove plant north-shift domestication method according to claim 1, wherein in the cold-proof protection solution, the particle size of the micro-nano shell powder is 0.1-100 μm.
24. The mangrove plant north-shift domestication method according to claim 23, wherein in the cold-proof protection solution, the micro-nano shell powder has a particle size of 1-100 μm.
25. The mangrove plant north-shift domestication method according to claim 1, characterized in that the micro-nano shell powder is selected from one or a mixture of several of mussel shell, oyster shell, pearl shell and scallop shell.
