本发明公开了基于马面鱼鱼鳞活性物质的复配物,复配物的成分及其重量份为马面鱼鱼鳞活性物1~5份、绿豆籽提取物5~10份、葡萄糖8~13份、甲基纤维素10~16份、维生素C 0.7~1.5份、水果粉5~12份、柠檬酸3~9份、水30~40份。有益效果为:本发明复配物是一种保健口服液,配方合理,营养丰富全面,无腥苦异味,酸甜可口,口感润滑,未添加任何化学防腐剂,安全性高,符合国家倡导的绿色食品理念;该复配物的有效成分易于被机体吸收利用,能清除机体内的羟自由基,减缓机体的老化、疾病的产生和发展,具有良好的开发利用潜力。

技术领域
本发明涉及鱼类产品深加工技术领域,尤其是涉及基于马面鱼鱼鳞活性物质的复配物。
背景技术
鱼鳞是大多数鱼类体表的皮肤衍生物,鱼鳞占鱼体重2~3%。鳞片外表上看是透明的,像花瓣,且边缘呈微小的卷曲,白色光泽,略带有鱼腥味。鱼鳞含水量为16.4~17.8%,平均为17.5%。鱼鳞是特殊的保健食品,它含有较多的卵鳞脂,有増强大脑记忆力、缓延细胞衰老的作用。鱼鳞含有丰富的蛋白质、脂肪和多种维生素,还有铁、锌、钙和多种人体必需的微量营养素,其中钙、磷的含量很高,能预防小儿佝偻病及中老年人骨质疏松与胥骨折。鱼鳞中胶原蛋白含量达到45%,其中含有大量的甘氨酸、脯氨酸、羟脯氨酸、羟赖氨酸等。而羟脯氨酸和羟赖氨酸在其他蛋白中很少被发现。胶原蛋白是当今经常出现在人们嘴中的一个美容热点话题,从外用化妆品,到口服液、胶囊,到处都能见到它的影子。胶原蛋白也称骨胶原,是一种丝状的胶原蛋白纤维,其主要生理机能是做结缔组织的粘合物质,能使皮肤结实富有弹性,它存在于人体皮肤、骨骼、肌腱等部位。
我国是水产大国,2010年我国水产品总产量达到5373.0万吨,水产品出口居世界首位,达203.64亿美元,继续位居大宗农产品出口首位。随着水产品产量的提高,我国水产品加工业得到了迅速的发展,同时在水产品的加工过程中产生了大量的下脚料。我国每年生产的鱼副产物中,鱼鳞大约可达到65万吨,然而这些鱼鳞并没有得到很好的利用。每年约有30万吨鱼鳞作为废弃物被丢弃。这不仅造成了资源的浪费,也对环境造成了污染。目前对鱼鳞的利用主要有:提取鱼鳞胶原蛋白,制备明胶,制备胶原蛋白肽,提取卵鳞脂,提取羟基磷灰石,提取鱼银等。
现有技术如授权公告号为CN 102613383 B的中国发明专利,公开了一种鱼鳞蛋白酶解液及其制备方法,该酶解液由如下原料制备得到:鱼鳞、蛋白酶和蒸馏水;其中鱼鳞和蒸馏水的重量份数比为:70~90份:10~30份,其中蛋白酶的添加量为鱼鳞和蒸馏水总重量的0.05~0.2%。该酶解液香气香味浓郁,口感醇厚,提升了鱼鳞蛋白酶解液的风味强度,但该制备方法所得酶解液有腥苦味,色泽发暗,影响其推广利用。
发明内容
本发明的目的在于提供一种营养丰富全面,无腥苦异味,酸甜可口,易于被机体吸收利用,能清除机体内的的羟自由基的基于马面鱼鱼鳞活性物质的复配物。
本发明针对上述技术中提到的问题,采取的技术方案为:基于马面鱼鱼鳞活性物质的复配物,成分及其重量份为:马面鱼鱼鳞活性物1~5份、绿豆籽提取物5~10份、葡萄糖8~13份、甲基纤维素10~16份、维生素C 0.7~1.5份、水果粉5~12份、柠檬酸3~9份、水30~40份。上述复配物是一种保健口服液,配方合理,营养丰富全面,无腥苦异味,酸甜可口,口感润滑,未添加任何化学防腐剂,安全性高,符合国家倡导的绿色食品理念;有效成分易于被机体吸收利用,能清除机体内的的羟自由基,减缓机体的老化、疾病的产生和发展,具有良好的开发利用潜力。
基于马面鱼鱼鳞活性物质的复配物的制备方法,具体操作步骤为:
脱钙处理:将鱼鳞粉以料液比为1:23~30加入浓度为0.15~0.2mol/L的EDTA溶液,在温度为35~40℃、功率为400~450W下微波脱钙80~100min,清水洗涤即可,EDTA具有鳌合大部分金属离子的作用,能除去钙离子,溶液的pH偏中性,对鱼鳞的组织结构、胶原蛋白的生物活性等保存较好,但脱钙时间长,而由于微波可起到快速脱钙的效果,但是随着功率的增大,胶原蛋白损失较严重,两者相结合可大大缩短脱钙时间,提高脱钙效率,且对胶原蛋白的影响不大;
脱杂蛋白:将脱钙后的鱼鳞粉以料液比为1:8~12加入质量浓度为2~5%的NaCl溶液,低温浸泡22~26h,期间换一次NaCl溶液,清水冲洗,过滤即可,利用NaCl能有效地脱除杂蛋白,使得胶原蛋白的流失率相对较低,降低杂质对活性提取物浓度的影响,提高活性提取物的纯度和得率,且该处理方法无毒无害,对活性提取物的成分及功效无任何影响;
酶法提取:将脱脂后鱼粉按料液比为1:7~10加水,混合均匀,调节pH值至6.5~7.5,加入胰蛋白酶800~1000U/g、中性蛋白酶600~800U/g,在50~60℃下酶解3~5h,灭酶,离心,备用,该步骤采用双酶酶解法提取马面鱼鱼鳞活性物质,利用两种蛋白酶作用的专一性,使原料蛋白质充分水解成活性多肽,提高马面鱼鱼鳞蛋白质的转化率,使蛋白质尽可能的降解为活性多肽,且可使活性肽具有味道鲜美、营养丰富的优点;
脱腥去苦:将酶解液中加入0.5~1wt%的β-环糊精,在20~30℃下搅拌1~2h,微滤,β-环糊精(β-CD)是环糊精葡萄糖基转移酶作用于淀粉产生的一组环状低聚糖,具有“内疏水、外亲水”特殊分子结构,使得CD能作为“宿主”包络不同“客体”化合物,达到脱腥去苦的目的;
超滤:在压力为0.3~0.8MPa下经截取分子量为3~8kDa的超滤膜超滤,喷雾干燥,即得基于马面鱼鱼鳞活性物质,该活性物质中含有一种抗氧化活性肽,其氨基酸序列为HSHAFCCGAAKCYLCRVLHPGKLCVCNCSK,具有较好的清除自由基的能力,阻止质子化超氧阴离子和羟基自由基对细胞成分和连接组织的有害进攻,减缓机体的老化、疾病的产生和发展,可加工成营养补充剂,也可以作为制备抗氧化、抗衰老的保健品或化妆品的原料,具备良好的开发利用潜力;
复配物配制:按配方称取马面鱼鱼鳞活性物、绿豆籽提取物、葡萄糖、甲基纤维素、维生素C、水果粉、柠檬酸、水,混合均匀,灌装,杀菌,即得基于马鲛鱼活性多肽的复配物,该复配物是一种多肽-亚铁螯合物,可制成医用食品,能容易的透过肠粘膜而被肠道有效吸收,促进和提高了人体对铁元素的吸收利用,利用率高,对人体无毒副作用,无金属异味,性质稳定,具有改善缺铁性贫血,促进生长、延缓衰老及增强机体免疫力等生物及营养功能活性。
与现有技术相比,本发明的优点在于:1)本发明复配物是一种保健口服液,配方合理,营养丰富全面,无腥苦异味,酸甜可口,口感润滑,未添加任何化学防腐剂,安全性高,符合国家倡导的绿色食品理念;2)该复配物的有效成分易于被机体吸收利用,能清除机体内的的羟自由基,减缓机体的老化、疾病的产生和发展,具有良好的开发利用潜力;3)该制备方法采用双酶酶解法提取马面鱼鱼鳞活性物质,使原料蛋白质充分水解,提高活性提取物质的得率,且该产品无任何毒副作用和不良反应,是理想的保健口服液。
具体实施方式
下面通过实施例对本发明方案作进一步说明:
实施例1:
基于马面鱼鱼鳞活性物质的复配物,成分及其重量份为:马面鱼鱼鳞活性物4份、绿豆籽提取物8份、葡萄糖10份、甲基纤维素16份、维生素C 1.2份、水果粉10份、柠檬酸8份、水38份。
基于马面鱼鱼鳞活性物质的复配物的制备方法,具体操作步骤为:
1)脱钙处理:将鱼鳞粉以料液比为1:26加入浓度为0.2mol/L的EDTA溶液,在温度为38℃、功率为420W下微波脱钙100min,清水洗涤即可,EDTA具有鳌合大部分金属离子的作用,能除去钙离子,溶液的pH偏中性,对鱼鳞的组织结构、胶原蛋白的生物活性等保存较好,但脱钙时间长,而由于微波可起到快速脱钙的效果,但是随着功率的增大,胶原蛋白损失较严重,两者相结合可大大缩短脱钙时间,提高脱钙效率,且对胶原蛋白的影响不大;
2)脱杂蛋白:将脱钙后的鱼鳞粉以料液比为1:10加入质量浓度为4%的NaCl溶液,低温浸泡24h,期间换一次NaCl溶液,清水冲洗,过滤即可,利用NaCl能有效地脱除杂蛋白,使得胶原蛋白的流失率相对较低,降低杂质对活性提取物浓度的影响,提高活性提取物的纯度和得率,且该处理方法无毒无害,对活性提取物的成分及功效无任何影响;
3)酶法提取:将脱脂后鱼粉按料液比为1:8加水,混合均匀,调节pH值至7,加入胰蛋白酶1000U/g、中性蛋白酶600U/g,在60℃下酶解4h,灭酶,离心,备用,该步骤采用双酶酶解法提取马面鱼鱼鳞活性物质,利用两种蛋白酶作用的专一性,使原料蛋白质充分水解成活性多肽,提高马面鱼鱼鳞蛋白质的转化率,使蛋白质尽可能的降解为活性多肽,且可使活性肽具有味道鲜美、营养丰富的优点;
4)脱腥去苦:将酶解液中加入0.8wt%的β-环糊精,在28℃下搅拌2h,微滤,β-环糊精(β-CD)是环糊精葡萄糖基转移酶作用于淀粉产生的一组环状低聚糖,具有“内疏水、外亲水”特殊分子结构,使得CD能作为“宿主”包络不同“客体”化合物,达到脱腥去苦的目的;
5)超滤:在压力为0.8MPa下经截取分子量为6kDa的超滤膜超滤,喷雾干燥,即得基于马面鱼鱼鳞活性物质,该活性物质中含有一种抗氧化活性肽,其氨基酸序列为HSHAFCCGAAKCYLCRVLHPGKLCVCNCSK,具有较好的清除自由基的能力,阻止质子化超氧阴离子和羟基自由基对细胞成分和连接组织的有害进攻,减缓机体的老化、疾病的产生和发展,可加工成营养补充剂,也可以作为制备抗氧化、抗衰老的保健品或化妆品的原料,具备良好的开发利用潜力;

实施例2:
基于马面鱼鱼鳞活性物质的复配物,成分及其重量份为:马面鱼鱼鳞活性物5份、绿豆籽提取物7份、葡萄糖11份、甲基纤维素15份、维生素C 1.5份、水果粉9份、柠檬酸7份、水35份。
基于马面鱼鱼鳞活性物质的复配物的制备方法,具体操作步骤为:
1)脱钙处理:将鱼鳞粉以料液比为1:25加入浓度为0.17mol/L的EDTA溶液,在温度为37℃、功率为450W下微波脱90min,清水洗涤即可,EDTA具有鳌合大部分金属离子的作用,能除去钙离子,溶液的pH偏中性,对鱼鳞的组织结构、胶原蛋白的生物活性等保存较好,但脱钙时间长,而由于微波可起到快速脱钙的效果,但是随着功率的增大,胶原蛋白损失较严重,两者相结合可大大缩短脱钙时间,提高脱钙效率,且对胶原蛋白的影响不大;
2)脱杂蛋白:将脱钙后的鱼鳞粉以料液比为1:9加入质量浓度为5%的NaCl溶液,低温浸泡25h,期间换一次NaCl溶液,清水冲洗,过滤即可,利用NaCl能有效地脱除杂蛋白,使得胶原蛋白的流失率相对较低,降低杂质对活性提取物浓度的影响,提高活性提取物的纯度和得率,且该处理方法无毒无害,对活性提取物的成分及功效无任何影响;
3)酶法提取:将脱脂后鱼粉按料液比为1:9加水,混合均匀,调节pH值至7,加入胰蛋白酶900U/g、中性蛋白酶700U/g,在55℃下酶解4h,灭酶,离心,备用,该步骤采用双酶酶解法提取马面鱼鱼鳞活性物质,利用两种蛋白酶作用的专一性,使原料蛋白质充分水解成活性多肽,提高马面鱼鱼鳞蛋白质的转化率,使蛋白质尽可能的降解为活性多肽,且可使活性肽具有味道鲜美、营养丰富的优点;
4)脱腥去苦:将酶解液中加入1wt%的β-环糊精,在25℃下搅拌1.5h,微滤,β-环糊精(β-CD)是环糊精葡萄糖基转移酶作用于淀粉产生的一组环状低聚糖,具有“内疏水、外亲水”特殊分子结构,使得CD能作为“宿主”包络不同“客体”化合物,达到脱腥去苦的目的;

1)脱钙处理:将鱼鳞粉中加入EDTA溶液,微波脱钙,清水洗涤;
2)脱杂蛋白:将脱钙后的鱼鳞粉中加入NaCl溶液,浸泡,清水冲洗,过滤;
3)酶法提取:将脱脂后鱼粉加水混合均匀,加入胰蛋白酶、中性蛋白酶,酶解,灭酶,离心;

6.根据权利要求3所述的基于马面鱼鱼鳞活性物质的复配物的制备方法,其特征在于:所述的脱杂蛋白步骤中鱼鳞粉与NaCl溶液的料液比为1:8~12,NaCl溶液的浓度为2~5%,浸泡时间为22~26h。
7.根据权利要求3所述的基于马面鱼鱼鳞活性物质的复配物的制备方法,其特征在于:所述的酶法提取步骤中脱脂后鱼粉与水的料液比为1:7~10。
8.根据权利要求3所述的基于马面鱼鱼鳞活性物质的复配物的制备方法,其特征在于:所述的酶法提取步骤中胰蛋白酶的添加量为800~1000U/g,中性蛋白酶的添加量为600~800U/g,酶解pH值为6.5~7.5,温度为50~60℃,时间为3~5h。
9.根据权利要求3所述的基于马面鱼鱼鳞活性物质的复配物的制备方法,其特征在于:所述的脱腥去苦步骤中β-环糊精的添加量为0.5~1wt%,处理温度为20~30℃,时间为1~2h。
10.根据权利要求3所述的基于马面鱼鱼鳞活性物质的复配物的制备方法,其特征在于:所述的超滤步骤中超滤的压力为0.3~0.8MPa,超滤膜的截取分子量为3~8kDa。
