背景技术
流行病学研究表明,食用十字花科芸苔属蔬菜,如西兰花、羽衣甘蓝、抱子甘蓝、花椰菜、白菜等可预防胰腺癌、肺癌、直肠癌、乳腺癌及前列腺癌等多种癌症的发生,这主要与芸苔属蔬菜体内的硫代葡萄糖苷(硫苷)的水解产物—萝卜硫素有关。
萝卜硫素,(1-异硫氰基-4-甲基亚磺酰基丁烷,l-isothiocyanato-4-methylsulfiny butane,sulfbraphane),分子量177.3,分子式为C6H11S2NO。它是葡萄糖莱菔子苷(硫苷的一种)经酶解后产生的,通过调控致癌物的代谢而作用,通过作用于人体内的“Phase two enzymes”激发人体的自我抗癌机制,中和体内某些致癌化学物质,造成癌细胞阻滞,抑制其繁殖并诱导其死亡,可有效预防胰腺癌、肺癌、直肠癌、乳腺癌及前列腺癌等多种癌症的发生。因此,它被认为是迄今为止蔬菜中发现的抗癌效果最好、效力最强的天然活性物质。作为药品或食品的功效成分,具有很高的研究价值和广阔的市场前景。

发明内容
本发明的主要目的为了弥补上述问题,提供一种能全面提高免疫力、同时预防大多数癌症的含萝卜硫素的保健品及其制备方法。
本发明提出一种含萝卜硫素的保健品,按照质量百分比计算,包括30%-60%的萝卜硫素提取物、28%-35%的灵芝孢子粉、10%-18%的人参皂苷RH2干粉、3%-8%的淀粉。
优选地,按照质量百分比计算,包括48%的萝卜硫素提取物、30%的灵芝孢子粉、18%的人参皂苷RH2干粉、4%的淀粉。
本发明还提出一种含萝卜硫素的保健品的制备方法,包括如下步骤:
S10,分别称取设定比例范围内的萝卜硫素提取物、灵芝孢子粉、人参皂苷RH2干粉,并混合均匀;
S20,称取设定比例范围内的淀粉,加少量水制成混合浆后,再加入到步骤S10处理后的混合物中,混匀制成软材;
S30,将所述软材制成颗粒、胶囊或压片。
优选地,步骤S10中所述萝卜硫素提取物通过下述方法制备:
S11,取新鲜的西兰花种子于75—80℃下脱水0.5—2小时,然后在55—65℃下烘干至含水量低于8%,用粉碎机粉碎至30—60目得到西兰花种子粉末,并作为原材料加入过量正己烷脱脂,用高速匀浆机混合均匀,控制温度20—40℃,时间30—50分钟,过滤后得滤渣备用;
S12,向滤渣中加入0.05mol/mL的磷酸缓冲液调节PH至4.5—5.5,加入粗芥子硫苷酸酶,搅拌均匀后置于30—45℃条件下酶解0.5—1.5小时,所述滤渣与粗芥子硫苷酸酶的比例为1g:3mL;然后加入质量分数为95%的乙醇,经超声波振荡提取10—60分钟,接着静置30—45分钟后过30目筛,除去提取物残渣,得到滤液;所述滤液经真空抽滤除去杂质后,得到抽提液;
S13,步骤S20得到的抽提液加水溶解后,用二氯甲烷萃取3次,合并萃取液,过滤并收集滤液,将滤液置于35—40℃条件下真空浓缩得到萝卜硫素粗品;所述加水溶解的量为抽提液的10倍量,萃取所用二氯甲烷的量为加水抽提液的2倍量;
S14,将上述制备得到的萝卜硫素粗品采用硅胶柱层析,以体积分数为10%的乙酸乙酯为洗脱剂进行洗脱,收集萝卜硫素馏分,35—40℃温度下真空浓缩即得萝卜硫素提取物。
优选地,步骤S11中所述原材料的质量与正己烷体积之比为1g:20—30mL。

优选地,按照质量百分比计算,其包括48%的萝卜硫素提取物、30%的灵芝孢子粉、18%的人参皂苷RH2干粉、4%的淀粉。
灵芝孢子粉,即灵芝的种子,它是灵芝在生长成熟期,从灵芝菌褶中弹射出来的极其微小的卵形生殖细胞。味甘,性温,无毒,温补不燥,能够促进机体正常代谢,增强机体抵抗力,抑制肿瘤生长,是目前癌症患者康复治疗的首选药。
人参皂苷RH2干粉是人参中提取的天然活性成份,经众多国际权威专家论证具有良好防癌、抗癌活性功能,能抑制癌细胞的生长,它也可有效增强单核巨噬细胞吞噬功能,可使癌细胞死亡,同时,它可以提高免疫力降低放化疗的毒性。
本发明还提出一种含萝卜硫素的保健品的制备方法,包括如下步骤:
S10,分别称取设定比例范围内的萝卜硫素提取物、灵芝孢子粉、人参皂苷RH2干粉,并混合均匀;
S20,称取设定比例范围内的淀粉,加少量水制成混合浆后,再加入到步骤S10处理后的混合物中,制成软材;
S30,将所述软材制成颗粒、胶囊或压片。
具体地,步骤S10中所述萝卜硫素提取物通过下述方法制备:
S11,取新鲜的西兰花种子于75—80℃下脱水0.5—2小时,然后在55—65℃下烘干至含水量低于8%,用粉碎机粉碎至30—60目得到西兰花种子粉末,并作为原材料加入过量正己烷脱脂,用高速匀浆机混合均匀,控制温度20—40℃,时间30—50分钟,过滤后得滤渣备用;
优选地,所述原材料的质量与正己烷体积之比为1g:20—30mL。
本步骤中,西兰花种子粉末需加入过量正己烷脱脂后再进行提取。这是由于西兰花种子中含有大量油脂,阻碍了硫代葡萄糖苷与酶的接触,无法使萝卜硫素完全释放;而且由于混合液中含有大量的水溶性蛋白和多糖,沉淀物质较多,溶液黏度很大,萃取和过滤都比较困难,导致部分萝卜硫素留在滤渣中不容易被洗出来,从而降低提取率。优选地,原材料质量与正己烷体积之比为1g:20—30mL。
为充分利用资源,本步骤中,过滤后,滤液为溶解有西兰花种子油脂的正己烷,将其送至脱溶剂釜进行脱正己烷处理,回收正己烷并获得粗制西兰花种子籽油,获得的粗制西兰花种子籽油可以进行精炼处理,制备出精炼食用西兰花种子籽油。
S12,向滤渣中加入0.05mol/mL的磷酸缓冲液调节PH至4.5—5.5,加入粗芥子硫苷酸酶,搅拌均匀后置于30—45℃条件下酶解0.5—1.5小时,所述滤渣与粗芥子硫苷酸酶的比例为1g:3mL;然后加入质量分数为95%的乙醇,经超声波振荡提取10~60min,接着静置30~45min后过30目筛,除去提取物残渣,得到滤液;所述滤液经真空抽滤除去杂质后,得到抽提液;
添加外源粗芥子硫苷酸酶,有利于促进细胞内相关酶促反应的发生,从而提高萝卜硫素含量,提高生产效率。
优选地,本步骤中,加入乙醇的质量是混合物质量的8—8.5倍;所述超声震荡频率40kHz,功率200w。抽真空时,真空度为-0.06MPa至-0.08MPa,温度为30℃—40℃。
S13,步骤S12得到的抽提液加水溶解后,用二氯甲烷萃取3次,合并萃取液,过滤并收集滤液,将滤液置于35—40℃条件下真空浓缩得到萝卜硫素粗品;所述加水溶解的量为抽提液的10倍量,萃取所用二氯甲烷的量为水层料液的2倍量;
二氯甲烷提取率最高,而且容易挥发,在旋转蒸发时很容易除去,因此采用二氯甲烷作为萃取剂。需注意的是,二氯甲烷具有较大的毒性,因此当提取的萝卜硫素用作食品或药品时,还需要适量添加乙酸乙酯作为萃取剂。得到的萝卜硫素粗品呈油状。
S14,将上述制备得到的萝卜硫素粗品采用硅胶柱层析,以体积分数为10%的乙酸乙酯为洗脱剂进行洗脱,收集萝卜硫素馏分,35—40℃温度下真空浓缩即得萝卜硫素提取物。
实施例1:
S10,按质量称取萝卜硫素提取物300g、灵芝孢子粉280g、人参皂苷RH2干粉100g,并混合均匀;
S20,称取淀粉30g,加少量水将所述淀粉制成混合浆后,再加入S10所述的混合物中,制成软材;
S30,将所述软材制成颗粒。
实施例2:
S10,按质量称取萝卜硫素提取物480g、灵芝孢子粉300g、人参皂苷RH2干粉180g,并混合均匀;
S20,称取淀粉40g,加少量水将所述淀粉制成混合浆后,再加入S10所述的混合物中,制成软材;
S30,将所述软材制成胶囊。
实施例3:
S10,按质量称取萝卜硫素提取物600g、灵芝孢子粉350g、人参皂苷RH2干粉120g,并混合均匀;
S20,称取淀粉80g,加少量水将所述淀粉制成混合浆后,再加入S10所述的混合物中,制成软材;
S30,将所述软材制成压片。
本发明以从粉碎好的西兰花种子中提取的萝卜硫素为原料,并配加人参皂苷RH2,灵芝孢子粉,淀粉等几种生物活性物质,与萝卜硫素优势互补,具有良好的防癌抗癌作用,在提高机体免疫力、抑制肿瘤细胞作用方面亦显示出积极的效果。
以上所述仅为本发明的较佳实施例,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (5)
1.一种含萝卜硫素的保健品,其特征在于,按照质量百分比计算,包括30%-60%的萝卜硫素提取物、28%-35%的灵芝孢子粉、10%-18%的人参皂苷RH2干粉、3%-8%的淀粉。
2.如权利要求1所述的含萝卜硫素的保健品,其特征在于,按照质量百分比计算,包括48%的萝卜硫素提取物、30%的灵芝孢子粉、18%的人参皂苷RH2干粉、4%的淀粉。
3.一种含萝卜硫素的保健品的制备方法,其特征在于,包括如下步骤:
S10,分别称取设定比例范围内的萝卜硫素提取物、灵芝孢子粉、人参皂苷RH2干粉,并混合均匀;
S20,称取设定比例范围内的淀粉,加少量水制成混合浆后,再加入到步骤S10处理后的混合物中,混匀制成软材;
S30,将所述软材制成颗粒、胶囊或压片。
4.如权利要求3所述的含萝卜硫素的保健品的制备方法,其特征在于,步骤S10中所述的萝卜硫素提取物通过下述方法制备:
S11,取新鲜的西兰花种子于75—80℃下脱水0.5—2小时,然后在55—65℃下烘干至含水量低于8%,用粉碎机粉碎至30—60目得到西兰花种子粉末,并作为原材料加入过量正己烷脱脂,用高速匀浆机混合均匀,控制温度20—40℃,时间30—50分钟,过滤后得滤渣备用;
S12,向滤渣中加入0.05mol/mL的磷酸缓冲液调节PH至4.5—5.5,加入粗芥子硫苷酸酶,搅拌均匀后置于30—45℃条件下酶解0.5—1.5小时,所述滤渣与粗芥子硫苷酸酶的比例为1g:3mL;然后加入质量分数为95%的乙醇,经超声波振荡提取10—60分钟,接着静置30—45分钟后过30目筛,除去提取物残渣,得到滤液;所述滤液经真空抽滤除去杂质后,得到抽提液;
S13,步骤S20得到的抽提液加水溶解后,用二氯甲烷萃取3次,合并萃取液,过滤并收集滤液,将滤液置于35—40℃条件下真空浓缩得到萝卜硫素粗品;其中,加水溶解的量为抽提液的10倍量,萃取所用二氯甲烷的量为加水抽提液的2倍量;
S14,将上述制备得到的萝卜硫素粗品采用硅胶柱层析,以体积分数为10%的乙酸乙酯为洗脱剂进行洗脱,收集萝卜硫素馏分,置于35—40℃温度下真空浓缩即得萝卜硫素提取物。
5.如权利要求4所述的含萝卜硫素的保健品的制备方法,其特征在于,步骤S11中所述原材料的质量与正己烷体积之比为1g:20—30mL。
