可按照以下方法制备已知通式(Ⅰ)的有效成分:
a)使通式(Ⅱ)的醛与吗啉和硫反应
(其中R和R′的定义同上);或
b)使通式(Ⅲ)的苯甲酸或其活性衍生物与吗啉反应
(其中R和R′的定义同上);或
c)用五硫化二磷使通式(Ⅳ)所示的氧基化合物硫化
(其中R和R′的定义用上);
并根据需要,将由此获得的通式(Ⅰ)化合物中的羟基(其中R代表氢)甲基化或乙基化。
从文献〔如,EUR.J.Med.Chem.-Chim.Ther.14,27-31(1979)〕中可查到以上反应。
通式(Ⅰ)化合物促进体重增加和提高饲料利用率的作用可通过以下试验加以证实:
用猪作为试验动物。每一组动物由6头猪组成,并且用6头猪进行的每一次试验均重复三次。所用饲料含有50mg/kg的通式(Ⅰ)试验化合物。动物在同样条件下饲养,并且所有的动物组均接受具有相同组成和相同数量的饲料。给参考组动物喂养的是同样的饲料,所不同的是,该饲料中含有的是黄霉素而不是通式(Ⅰ)化合物。而喂养对照组动物的饲料中则既不加通式(Ⅰ)试验化合物也不加黄霉素。由此获得的试验结果见表(Ⅰ)。

从以上数据可以明显指出,用含有通式(Ⅰ)化合物的饲料喂养的动物其体重增加明显地高于参考组。同时,当动物饲料加入式(Ⅰ)化合物时,增加相同体重所需的饲料量明显减少。这证明饲料的利用率得到提高。
另一方面,本发明提供了制备饲料添加剂、预混合物和待用饲料的方法,它包括将通式(Ⅰ)化合物(其中R和R′的定义同上)与适宜的可食用固体或液体载体或稀释剂混合,并且可任意地与饲料添加剂和饲料生产中通常使用的添加剂混合。
作为载体,可使用来源于植物或动物且可用于喂养动物的任何物质。为此,例如可应用合适形式(粗碾谷类、细碾谷类、粗磨谷粉等)的小麦、大麦、麸、玉米、大豆、燕麦、黑麦以及苜蓿等,另外,也可使用鱼粉、肉粉、脱脂乳粉或木薯淀粉及其混合物等。使用高蛋白含量的无纤维绿色植物精饲料比较有利。
可使用的添加剂包括硅酸、抗氧化剂、淀粉、磷酸二钙、碳酸钙、山梨酸等。还可使用各种亚烷基二醇。所使用的淀粉可以是小麦、玉米或马铃薯淀粉。
组合物中有效成分的含量可在很宽的范围内变化。饲料添加剂和精饲料中可含有约5-80%(按重量计),优选约10-80%(按重量计)的式(Ⅰ)有效成分。待用的动物饲料中有效成分的含量可为约0.001-0.04%(按重量计),优选约0.001-0.01%(按重量计)。饲料添加剂和精饲料中也可含有常用的维生素(如维生素A、B1、B2、B3、B6、B12、E、K)和微量元素(如Mn、Fe、Zn、Cu、I)。
将饲料添加剂和精饲料用合适的饲料组分稀释或加到合适的动物饲料中可制成待用的动物饲料。
本发明的饲料添加剂、预混合物和待用饲料可用于促进猪、羊、牛、家禽等各种家畜(尤其是猪)的体重增加并提高饲料利用率。
再一方面,本发明提供了一种促进动物体重增加并提高饲料利用率的方法,它包括用本发明饲料或饲料添加剂喂养动物。
再一方面,本发明提供了通式(Ⅰ)化合物在促进家畜体重增加和提高饲料利用率方面的应用。
下面的非限制性实施例用于进一步说明本发明。实施例中所出现的熔点未经过校正。
实施例1
4-(4′-羟基-3′-甲氧基苯基)-硫代甲基吗啉
将104.5g(1.5mole)吗啉、32.0g(1.0mole)粉状硫和152.2g(1.0mole)4-羟基-3-甲氧基-苯甲醛于该反应混合物的沸点(115℃)下反应4小时。然后用乙醇结晶产物。
产率:226.8g(89.%)黄色晶体。
M.P.:167-168℃。
元素分析:C12H15NO3S(253.4)
计算值:C%=56.87 H%=5.97 N%=5.56 S%=12.65
实测值:C%=57.02 H%=6.05 N%=5.49 S%=12.66
UV:λmax=281nm(=15442)
实施例2
4-(4′-乙氧基-3′-甲氧基苯基)-硫代甲基吗啉
将87.1g(1.0mole)吗啉、17.6g(0.55mole)粉状硫和90.1g(0.5mole)4-乙氧基-3-甲氧基-苯甲醛于该反应混合物的沸点下反应。当检测不出醛成分时,冷却深红色油状物,产品用甲醇结晶。
产率:134.4g(95.5%)黄色层状结晶
M.P.:127-129℃
元素分析:C14H19NO3S(281.4)
计算值:C%=59.75 H%=6.81 N%=4.98 S%=11.39
实测值:C%=59.46 H%=6.75 N%=5.07 S%=11.45
UV:λmax=281nm(=15232)
实施例3
4-(3′,4′,5′-三甲氧基苯基)-硫代甲基吗啉
将28.1g(0.1mole)4-(3′,4′,5′-三甲氧基苯基)-氧代-甲基吗啉、100ml吡啶和22.3g(0.1mole)五硫化二磷于该反应混合物的沸点下反应5小时。然后蒸除溶剂,用水使产物析出。
产率:25.9g(87.5%)黄色结晶
M.P.:140-142℃
元素分析:C14H19NO4S(297.3)
计算值:C%=56.55 H%=6.44 N%=4.71 S%=10.78
实测值:C%=56.69 H%=6.40 N%=4.72 S%=10.65
UV λmax=245and280nm(=8810和13434)
实施例4
4-(3′,4′-二甲氧基苯基)-硫代甲基吗啉
将25.1g(0.1mole)4-(3′,4′-二甲氧基苯基)-氧代甲基吗啉、100ml吡啶和22.3g(0.1mole)五硫化二磷的混合物煮沸8小时。然后真空除去溶剂,将残余物例入300g冰水中,产物用乙醇结晶。
产率:23.79g(89.0%)黄色结晶
元素分析:C13H17NO3S(267.37)
计算值:C%=58.4 H%=6.41 N%=5.24 S%=11.99
实测值:C%=58.27 H%=6.47 N%=5.32 S%=12.17

按以下组成配制用作补充猪饲料的预混合物:

实施例9
将400Kg粗磨的大豆粉装入混合器中,并在搅拌下加入3.1Kg大豆油,然后搅拌混合物直至固体被油包裹为止。再加入9.1Kg实例2的有效成分,搅拌混合物直至得到一种均质的混合物。最后,加入9.0Kg大豆油,并再次均化混合物。
实施例10
将0.5Kg实施例2的有效成分在搅拌下加到10Kg玉米粉中,同时向该混合物中喷撒丙二醇,然后加入1.4Kg磷酸二钙,并均化混合物。
实施例11
将10Kg苜蓿粉和15Kg具有高蛋白质含量的无纤维苜蓿精饲料搅拌20小时。然后以均匀的速度向混合物中喷撒1Kg玉米油,从而保证在加入以下另外成分时喷撒继续进行:2.5Kg实施例1的有效成分、10Kg玉米淀粉、0.3Kg二氧化硅、0.6Kg维生素C、9Kg玉米淀粉和2.5Kg上述有效成分。将由此获得的混合物进一步搅拌5分钟。
实施例12
重复实施例9的步骤,所不同的是,用1,4-丁二醇代替大豆油作为湿润剂。
实施例13
A)将3.5Kg马铃薯淀粉与2.9Kg实施例2的有效成分混合。向该混合物中喷撒0.05Kg矿物油。然后加入0.2Kg山梨酸、0.4Kg二氧化硅和0.1Kg丙酸钙,将混合物再搅拌2分钟。
B)将4.2Kg鱼粉与22Kg黑麦麸混合,并向混合物中喷撒0.6Kg矿物油。然后在搅拌条件下加入4Kg按方法A)制得的混合物、10Kg玉米粉、4Kg按照方法A)制得的混合物以及9Kg玉米粉。最后向混合物中喷撒0.6Kg矿物油。
实施例14
将100Kg小麦麸、10Kg实施3的有效成分、2.5Kg碳酸钙、0.15Kg维生素E和0.4Kg丙酸钙与4Kg丙二醇一起均化。
实施例15
将10Kg大豆粉和0.6Kg实施例3的有效成分与2.5Kg1,4-丁二醇一起均化。
实施例16
将50Kg大豆粉、6Kg实例4的有效成分、0.5Kg二氧化硅、1.6Kg大豆油以及0.2Kg丙酸钙一起均化。
Claims (9)
6、根据权利要求1-5中任何一项所述的组合物,它包括作为载体的可用于喂养动物的植物或动物来源的物质,优选以粗碾谷物、细碾谷物、粗磨谷粉等形式的小麦、大麦、麸皮、玉米、大豆、燕麦、黑麦或苜蓿,以及鱼粉、肉粉、脱脂乳粉、木薯粉或其混合物。
7、制备权利要求1-6中任何一项所述组合物的方法,它包括将通式(Ⅰ)的化合物(其中R和R′的定义同上)与合适的可食用固体或液体载体或稀释剂混合,并且可任意地与饲料添加剂和饲料生产中通常使用的添加剂混合。
8、一种促进动物体重增加并提高饲料利用率的方法,它包括用权利要求1-6中任何一项所述的饲料或饲料添加剂喂养动物。
9、通式(Ⅰ)化合物在促进家畜体重增加和提高饲料利用率方面的应用。
