技术领域
本申请主张2011年04月18日提交的韩国专利申请第10-2011-0035613号的优先权,上述说明书全体为本申请的参考文献。
本发明涉及使活性多肽的结合亲和力或结合特异性增强的新型蛋白质骨架模块。更详细地说,本发明涉及下述的蛋白质骨架模块及其制造方法,该蛋白质骨架模块的特征在于,其包含:由序列编号1所示氨基酸序列的第1位~第19位氨基酸序列构成的多肽;包含活性多肽的多肽;和由序列编号1所示氨基酸序列的第29位~第86位氨基酸序列构成的多肽。
背景技术
若对蛋白质氨基酸序列的2级、3级结构进行分析,则大部分蛋白质由独立的结构域(或者模块)构成。结构域为结构上、功能上独立的单元。同一结构域在不同蛋白质中有一个以上的分布,一个蛋白质由不同的结构域构成。结构域的具体信息可在Prosite(Hulo N等,Nucleic Acids Res,36:D245-249,2008;网址:http://kr.expasy.org/prosite/),SMART(Letunic I等,Nucleic Acids Res,34:D257-D260,2006;网址:http://smart.embl-heidelberg.de/)等生物信息学网站上查询。
生物体分子间的相互作用(例如蛋白质-蛋白质、蛋白质-核酸等)在细胞的生长、分化、发育、细胞间/内的信号传递、物质转运等各种生命现象的产生中具有重要功能。作为现有的与目标分子特异性结合来调节生物学活性的分子,主要开发了抗体(全长或片段)。但是,抗体的问题在于其具有下述多种问题:其表达量少,溶解度低,必须要使用动物细胞的表达细胞株,精制费用高,在还原性细胞内环境中的稳定性降低等。因此,迫切需要开发出可克服抗体的问题同时还具有抗体这样的与目标分子(targeted molecules)特异性结合的特性的抗体以外的蛋白质。
为了在癌症或动脉硬化的诊断和治疗中进行使用,利用噬菌体展示等方法发掘出的肽具有低结合力、非稳定性、高免疫性等限制。有必要开发出可克服这样的问题、提高活性肽在人体内的结合力与稳定性、同时降低免疫性的新技术。
发明内容

本发明人发现,在选择人体来源的蛋白质中的一部分进行最佳变形、插入活性肽的情况下,显示出了可使活性多肽的结合亲和力或结合特异性增强等可代替现有肽制剂和抗体制剂的优异的性质,从而完成了本发明。
本发明的其他目的在于提供一种内置有活性多肽的蛋白质骨架模块,其特征在于,其包含下述多肽依次连接而成的多肽:
a)由序列编号1所示氨基酸序列的第1位~第19位氨基酸序列构成的多肽;
b)包含活性多肽的多肽;以及
c)由序列编号1所示氨基酸序列的第29位~第86位氨基酸序列构成的多肽。
本发明的其他目的在于提供一种多聚核苷酸,其编码上述内置有活性多肽的蛋白质骨架模块。
本发明的其他目的在于提供一种表达载体,其包含上述多聚核苷酸。

以下详细说明本发明。
本发明提供了内置有(包含有)活性多肽的蛋白质骨架模块和编码上述内置有活性多肽的蛋白质骨架模块(蛋白支架(protein scaffold),模块化蛋白支架(modularizedprotein scaffold))的多聚核苷酸,该内置有活性多肽的蛋白质骨架模块的特征在于,其包含下述多肽依次连接而成的多肽:a)由序列编号1所示氨基酸序列的第1位~第19位氨基酸序列构成的多肽;b)包含活性多肽的多肽;以及c)由序列编号1所示氨基酸序列的第29位~第86位氨基酸序列构成的多肽。
本发明的蛋白质骨架模块具有使所内置的活性多肽的结合亲和力或结合特异性增强的特征。
活性多肽是指通过与对象蛋白质的接触而发挥出信号传递、对象蛋白质的活化(activation)、失活(deactivation)、磷酸化(phosphorylation)等各种作用的多肽。本发明的蛋白质骨架模块通过使内置的活性多肽的结合亲和力或结合特异性增加的方法来增加活性多肽的活性。
结合亲和力(binding affinity)是指使生物体分子间结合的性质的强度。
结合特异性(binding specificity)是指特定生物体分子仅与结合对象分子特异性结合的性质。
本发明的蛋白质骨架模块意味着不会对生物体内免疫体系带来影响、可内置活性多肽的蛋白质的结构体。本发明蛋白质骨架模块的分子量小、生物体内活性优异,不会对内置的活性多肽的活性带来影响、不会对生物体内免疫体系带来影响,来自在生物体内丰富存在的蛋白质,不会产生个体间效果的差异或产生个体特异性效果。本发明人首先开发出了内置活性多肽且使活性多肽的稳定性和特性提高的蛋白质骨架模块,将其命名为DMID(Designed modular Immuno Diagnostics,免疫诊断设计模块)。
本发明的活性多肽可具有药理活性,内置有活性多肽的蛋白质骨架模块可通过活性多肽的作用显示出内在的药理活性。内置有活性多肽的蛋白质骨架模块还具有活性多肽的药理活性、特别具有效果得到增强的药理活性。
本发明的蛋白质骨架模块优选可基于稳定素(stabilin)-2来源的多肽片段进行设计,更优选为在序列编号1所示的氨基酸序列中在不会对骨架带来影响的部分进行活性多肽的取代或插入而成的蛋白质骨架模块。
本发明一个实施例的蛋白质骨架模块是基于在人体中丰富存在的稳定素-2来源的多肽片段而设计的。稳定素-2与利用跨膜受体的淋巴细胞诱导、细胞附着、受体清除、血管生成相关。该蛋白质具有7个成束蛋白(fasciclin)结构域、16个表皮生长因子(epidermal growth factor)(EGF)样结构域、2个层粘蛋白(laminin)型EGF样结构域、一个C型凝集素(lectin)样透明质烷(hyaluronan)结合样模块等多种结构域。
另一方面,本发明提供编码上述蛋白质骨架模块的多聚核苷酸。
本发明的多聚核苷酸具有的特征是,对使活性多肽的结合亲和力或结合特异性增强的蛋白质骨架模块进行编码。
本发明的多聚核苷酸意味着编码下述蛋白质骨架模块的物质,该蛋白质骨架模块具有序列编号1所表示的氨基酸序列、或具有与上述氨基酸序列具有至少70%以上同源性(homology)的氨基酸序列,上述多聚核苷酸为DNA或RNA,优选为序列编号2所表示的DNA。
本发明的特征在于提供插入有上述本发明的多聚核苷酸的表达载体。
上述表达载体意味着如下制造的表达载体:本领域技术人员通过本领域已知的任意方法将本发明的多聚核苷酸插入到载体中,利用适当的转录/解码调节序列表达本发明的蛋白质骨架模块,由此制造出该表达载体。
本发明的“表达载体”意味着能够插入或导入编码本发明蛋白质的多聚核苷酸序列的本领域已知的质粒、病毒或其他介质。本发明的多聚核苷酸序列与表达调节序列能够可操纵地联接,上述可操纵联接的基因序列与表达调节序列可同时含有选择标记和复制起点(replication origin)、并包含在一个表达载体内。“可操纵联接(operablylinked)”是指适当的分子与表达调节序列结合时,以能够进行基因表达的方式联接而成的基因和表达调节序列。“表达调节序列(expression control sequence)”意味着利用特定的宿主细胞可操纵地联接而成的用于调节多聚核苷酸序列表达的DNA序列。这样的调节序列包括:用于实施转录的启动子、用于调节转录的任意的操纵序列、编码适宜的mRNA核糖体结合部位的序列和用于调节转录和解码(解読)的终结的序列。
能够插入编码上述本发明的蛋白质骨架模块的多聚核苷酸的表达载体有大肠杆菌来源的质粒(pBR322、pBR325、pUC118、pUC119、pET30a、pET30c和pGEX-GST)、枯草杆菌来源的质粒(pUB110和pTP5)、酵母来源的质粒(YEp13、YEp24和YCp50)和Ti质粒等,可以使用逆转录病毒、腺病毒或痘苗病毒之类的动物病毒、杆状病毒之类的昆虫病毒和植物病毒,可以使用pPZP、pGA和pCAMBIA系之类的双元载体。只要为本领域技术人员即可选择适于导入本发明多聚核苷酸序列的载体,在本发明中,只要是能够将本发明的多聚核苷酸序列导入到宿主细胞内的载体,任意载体均可使用。但是,可优选使用按照容易诱导蛋白质表达和容易分离出所表达的蛋白质的方式而设计出的载体,更可优选使用作为含有本发明多聚核苷酸的重组载体的pET32a载体。
本发明提供经上述本发明的重组载体转化的宿主细胞。

本发明的诊断用组合物能够发现活性多肽的靶物质的有无、分布、量等,可提供能够对与此相关的疾病的有无、进展状态进行判断的信息。与包括未内置在本发明的蛋白质骨架模块中的活性多肽的诊断用组合物相比,本发明组合物的诊断灵敏度、精度、准确度优异,诊断能力优异。
本发明提供含有上述内置有活性多肽的蛋白质骨架模块作为有效成分的诊断用试剂盒。

活性多肽意味着通过与对象蛋白质的接触而发挥出信号传递、对象蛋白质的活化(activation)、失活(deactivation)、磷酸化(phosphorylation)等各种作用的多肽。由于AP-1具有与细胞因子IL-4受体结合的氨基酸序列,因而其自身与IL-4受体具有结合力。已知IL-4受体在人类的多种癌症(大肠癌、肺癌、子宫癌和乳腺癌)中过表达,因而与IL-4受体选择性结合的活性多肽的开发可在癌症的诊断和治疗中应用。由于IL-4发挥出利用增加细胞附着的细胞因子来诱发动脉硬化的作用,因而AP-1肽可开发成动脉硬化的诊断和治疗剂。
本发明提供含有以内置有AP-1的蛋白质骨架模块作为有效成分的癌症或动脉硬化诊断用组合物、以及含有以内置有AP-1的蛋白质骨架模块作为有效成分的癌症或动脉硬化诊断用试剂盒。
本发明提供一种癌症或动脉硬化诊断方法,其包括将内置有AP-1的蛋白质骨架模块以有效量对于所需要的个体进行给药的工序。
本发明提供内置有AP-1的蛋白质骨架模块的用途,其用于制造癌症或动脉硬化诊断用制剂。
对于上述癌症,只要是随着其发生、进展、退化或消失会使带有AP-1具有亲和性的结构的生物体物质的水平发生变化的癌,即可为任意癌症,优选为大肠癌、肺癌、子宫癌或乳腺癌。
本发明的药物组合物可单独含有本发明的蛋白质骨架模块、或者可追加含有2种以上的药学上容许的载体、赋形剂或稀释剂。药学上容许的载体中,例如可追加含有口服给药用载体或非口服给药用载体。口服给药用载体可以包含乳糖、淀粉、纤维素衍生物、硬脂酸镁、硬脂酸等。可以包含在肽制剂的口服给药中使用的各种药物传递物质。进一步地,非口服给药用载体可以包含水、适宜的油、食盐水、水性葡萄糖等,可以追加含有稳定化剂和保存剂。适宜的稳定化剂有亚硫酸氢钠、亚硫酸钠或抗坏血酸之类的抗氧化剂。适宜的保存剂有苯扎氯铵、羟苯甲酯或羟苯丙酯和氯丁醇。其他药学上容许的载体可参考下述文献中的记载(Remington's PharmaceuticalSciences,19th ed.,Mack Publishing Company,Easton,PA,1995)。
本发明的组合物可通过任意方法对以人为代表的哺乳动物进行给药。例如,可进行口服给药或非口服给药。非口服的给药方法没有限定,可进行静脉内、肌肉内、动脉内、骨髓内、隔膜内、心脏内、经皮、皮下、腹腔内、鼻腔内、肠管、局部、舌下或直肠内给药。
本发明的药物组合物可根据如上所述的给药途径进行口服给药用或非口服给药用制剂的制剂化。
在口服给药用制剂的情况下,本发明的组合物可利用本领域公知的方法制剂化成粉末、颗粒、片剂、丸剂、糖衣片剂、胶囊剂、液剂、凝胶剂、糖浆、浆料剂、悬浮液等。例如,对于口服用制剂,可将活性成分与固体赋形剂混配,将其粉碎,添加适宜的辅助剂,以颗粒混合物的形式进行加工,从而得到片剂或糖衣片剂。适宜的赋形剂的示例包括:包括乳糖、葡萄糖、蔗糖、山梨糖醇、甘露醇、木糖醇、红藓醇和麦芽糖醇等的糖类以及包括玉米淀粉、小麦淀粉、米淀粉和马铃薯淀粉等的淀粉类;包括纤维素、甲基纤维素、羧甲基纤维素钠和羟丙基甲基纤维素等的纤维素类;明胶、聚乙烯吡咯烷酮等之类的填充剂。进一步地,根据情况可添加交联接合聚乙烯吡咯烷酮、琼脂、藻酸或海藻酸钠等作为崩解剂。本发明的药物组合物可以追加含有抗凝聚剂、润滑剂、湿润剂、香料、乳化剂和防腐剂等。
在非口服给药用制剂的情况下,可利用本领域公知的方法制剂化成注射剂、乳膏剂、洗剂、外用软膏剂、油剂、保湿剂、凝胶剂、气溶胶和鼻腔吸入剂的形态。这些剂型全部以制药化学中通常公知的处方的形式记载在文献(Remington'sPharmaceutical Science,15th Edition,1975,Mack Publishing Company,Easton,Pennsylvania18042,Chapter87:Blaug,Seymour)中。
本发明组合物的蛋白质骨架模块的总有效量可以以单剂量(single dose)对患者进行给药,也可以利用通过多剂量(multiple dose)长期给药的分次治疗方法(fractionatedtreatment protocol)进行给药。本发明的药物组合物中,可根据疾病程度的不同使有效成分的含量不同。优选的是,本发明组合物的蛋白质骨架模块的用量为每1天每1kg患者体重为约0.01ug~1,000mg、最优选为0.1ug~100mg。但是,对于上述蛋白质骨架模块的用量,不仅要考虑药物组合物的给药途径和治疗次数,而且要考虑患者的年龄、体重、健康状态、性别、疾病的严重程度、饮食和排泄率等各种要因等来确定对于患者的有效给药量,因而考虑到这些方面,只要为具有本领域常规知识的人,即可根据特定用途确定上述蛋白质骨架模块的适当的有效给药量。本发明的药物组合物只要可表现出本发明的效果即可,对其剂型、给药途径和给药方法没有特别限制。
本发明的这样的效果在本发明说明书的实施例中示出。
本发明的实施例中,对于多种模块结构域中的亲和力高、分子量和免疫原性低、在人体丰富存在、能够进行三维结构检证的模块结构域进行筛选,选择出最佳结构域即稳定素(Stamiin)-2的EGFL结构域。对所选择的序列进行三维模型化,利用电脑模拟(in silico)诱发突变,确立最佳模型。
在本发明另一实施例中,制作对所选择的蛋白质骨架模块进行编码的多聚核苷酸序列,将其导入到能够进行表达的表达载体中,利用其转化大肠杆菌。对转化后的菌株进行培养,使本发明的蛋白质骨架模块进行表达,之后对其进行提取精制。
在本发明的另一实施例中,利用表面等离子共振分析对于本发明的内置有作为活性多肽的AP-1的蛋白质骨架模块的结合能力进行测定。由测定结果确认到,在AP-1内置于本发明的蛋白质骨架模块中的情况下,亲和力比未内置的单独AP-1肽(nakedAP-1)高174倍。
在本发明的另一实施例中,对于活性多肽由本发明的蛋白质骨架模块所致的检测灵敏度的变化进行测定。结果确认到,在AP-1内置于本发明的蛋白质骨架模块中的情况下,对IL-4活性的抑制为未内置的单独AP-1肽的10倍以上。
在本发明的另一实施例中,对于本发明的内置有活性多肽的蛋白质骨架模块所致的诊断灵敏度进行测定。由细胞成像(Cellular imaging)或动物成像(Animal imaging)实验结果确认到,在AP-1内置于本发明的蛋白质骨架模块中的情况下,肿瘤靶向性功能高于未内置的单独AP-1肽。

其中,下述实施例仅为本发明的示例,本发明的内容并不限定于下述实施例。
实施例1.
DMID(免疫诊断设计模块)互补组筛选和模型化
1-1.筛选
对于在人表达的蛋白质结构域内大量存在的模块结构域进行对人基因数据库比较分析的后基因组筛选,从而选定20个互补组,之后以a)高亲和力、b)低分子量、c)低免疫原性(low immunogenicity)、d)在人体丰富存在、以及e)能够进行三维结构检证的条件为基准,选择EGFL(EGF样)结构域。
EGFL为在多种血浆蛋白质中存在的由40个氨基酸构成的模块,具有低分子、高安全性、低免疫性的特性。其为由2个beta-片层(beta-sheet)和3个二硫键构成的简单结构,其能够进行三维结构检证,存在有不会对结构带来影响的loop,容易插入活性多肽,因而适宜作为DMID互补。
1-2.三维模型化
为了进行EGFL结构域的三维模型化,使用BLAST与AlignMaster程序。人EGFL结构域的结构由Protein DataBank提供。
理论基准的突变和二维模型化利用nnPredic((www.cmpharm.ucsf.edu/~nomi/nnpredict.html)和jppred(http://www.copmbio.dundee.ac.uk/~www-jpred/submit.html)来进行。理论基准的三维模型化(Knowledge based3-dimensional models)由Swiss Model(http://www.expasy.ch/)提供,此外在in silico突变诱发后利用Chimera/Modeller6检证。最后对DMID模型利用PyMOL分析,进行可视化。
实施例2.
重组DMID-AP1表达和精制
2-1.转化菌株制作-DMID-SalI-HindIII盒的制作
为了制作本发明的蛋白质骨架模块(DMID),将稳定素2蛋白质所存在的EGFL结构域(4621-4869)克隆到作为E-coli表达载体的pET32a(Novagen)的BamH1与Xhol位点,制造表达蛋白质。将EGFL结构域的GCTGAC(4639-4644)基因变异为GTCGAC来制造限制酶Sall位点,将AAAGCA(4703-4708)变异为AAGCTT来制造限制酶HindIII位点,制造能够插入活性多肽的DMID-Sall-HindIII盒。
2-2.转化菌株的培养
将转化后的Origami菌株在含有50ug/ml氨苄青霉素与25ug/ml卡那霉素的20ml的磷酸胰蛋白胨(TP)培养基(2%细菌用胰蛋白胨,0.2%Na2HPO4,0.1%KH2PO4,0.8%NaCl,1.5%酵母提取液,以及0.2%葡萄糖)中培养一夜。
将培养液以4000×g进行5分钟离心分离,回收沉淀物,利用含有50ug/ml氨苄青霉素的同体积TP进行再悬浮。将其利用TP培养基稀释至100倍,在37℃进行培养,直至OD600值为0.5。将培养终止的培养液冷却至20℃后,利用0.2mM的IPTG(IsopropyI-β-D-thiogalactoside)进行12小时的蛋白质表达的诱导,之后回收培养细胞,保存在-80℃直至进行使用时为止。
2-3.蛋白质提取和分离精制
将上述保存的细胞再悬浮在10ml含有1mM PMSF、0.5mM DTT的2×PBS中,在负载率(Duty cycle):30%、输出:2.5、5分钟的条件下进行超声波粉碎。将粉碎的细胞在15,000g进行10分钟离心分离,采集上清液。
将回收的上清液按照制造商的操作规范装在经2×PBS缓冲液平衡后的Ni-NTN柱(amersham pharmacia biotech AB,Vt=0.5ml)中进行上样。对于带有6(his)tag的DMID,使柱带有含有30mM咪唑的2×PBS,利用含有30mM咪唑的2×PBS对柱进行水洗(柱体积的10倍),之后利用含有250mM咪唑的2×PBS缓冲液(洗脱缓冲液)对柱进行蛋白质回收。
实施例3.
重组DMID-AP1结合能力提高检证-表面等离子共振分析
为了研究本发明的内置有AP1的蛋白质骨架模块(DMID-AP1)的结合能力,使用Biacore2000,利用表面等离子共振分析法对于未内置的单独AP1与DMID-AP1的结合常数进行测定。将利用昆虫细胞(SF21)表达和精制的人IL-4受体的胞外域(IL-4Ra)利用HBR缓冲液(10mM HEPES,pH7.4;150mM NaCl;3.4mM EDTA;以及0.005%表面活性剂P20)稀释后,在Biacore CM5chip表面按照制造商的方法进行包被(2500RU)。使利用同一缓冲液稀释的AP-1肽或DMID-AP1流过(50ul/min),测定RU值的变化,之后使用Biaevaluation2.0程序求出结合常数。
如图2和图3所示进行了表面等离子共振分析,结果确认到,与未内置的单独肽相比,包埋肽DMID显示出了高174倍的亲和力。
实施例4.
重组DMID-API检测能力提高检证1—基于IL-4的IL-4受体的活性抑制度的测定
将5×105的THP1(NIH AIDS Research and Reagent Program,NCI9949)细胞接种在6孔培养板中,之后利用100ng/ml的佛波醇12-十四酸酯13-乙酸酯(Phorbol12-myristate13-acetate)(PMA)(Sigma,P8139-1MG)进行3天处理使其活化。在无血清条件下对细胞进行18小时培养后,同时进行0.67nM(1×)的单独IL-4或多种浓度[10n×(0.67NM)]AP-1肽或重组DMID-AP1的处理。通过IL-4与IL-4受体间的结合而诱导的Stat蛋白质的磷酸化使用兔多克隆抗体抗STAT6(Cell Signaling,#9362)与兔多克隆抗体抗pTyr641-STAT6(Cell Signaling,#9361)利用Western法进行测定。结果确认到,如图4所示,与AP1相比,本发明的DMID-AP1将对Stat蛋白质进行磷酸化的IL-4活性抑制了10倍以上。
实施例5.
重组DMID-AP1检测能力提高检证2—细胞成像、动物成像
5-1.细胞影像(细胞成像)
为了研究本发明的内置有AP1的蛋白质骨架模块(DMID-AP1)与在细胞中表达的IL-4受体的竞争结合能力,使用荧光标记蛋白质对于H226细胞系中的未内置的单独AP1与DMID-AP1相对于IL-4受体的结合能力进行测定。将AP-1肽或精制的DMID-AP1利用透析缓冲液(50mM硼酸,150mM NaCl,1mM EDTA,pH9.0)在4℃进行透析后,与荧光物质异硫氰酸荧光素(FITC)(sigma)在37℃反应1小时进行标记。在37℃加入50mM甘氨酸5分钟使反应停止后,利用透析缓冲液(20mM Tris HCl,pH8.0)于4℃进行透析。向H226细胞中分别添加相同量的对照组肽、AP-1、DMID-AP1使其结合,洗涤后利用4%多聚甲醛(PFA)进行固定,细胞核利用4',6-二脒基-2-苯基吲哚(DAPI)进行染色,之后利用荧光显微镜(Zeiss,Oberkochen,Germany)进行观察。
结果确认到,如图5所示,对于AP1-DMID-FITC,与不同的AP-1或对照组肽相比,表达IL-4受体的细胞标记能力优异。
5-2.动物影像(动物成像)
将1×107个H226细胞悬浮在培养基中,皮下注射至5~6周龄的雌性BALB/c裸鼠的右侧大腿部,制造皮下肿瘤模型。约3周后肿瘤的尺寸为0.1cm~1cm,此时用于实验中。将肿瘤异种移植小鼠麻醉,尾静脉注射结合有荧光物质的AP-1肽与DMID-AP1150ug。利用Optics分析荧光标记肽的肿瘤寻靶作用功能。
结果确认到,如图6所示,与AP-1相比,DMID-AP1的肿瘤寻靶作用功能得到了提高。
工业实用性
本发明涉及活性多肽的结合亲和力或结合特异性得到增强的新颖的蛋白质骨架模块,详细地说,本发明提供下述蛋白质骨架模块和其制造方法,该蛋白质骨架模块的特征在于,其包含下述多肽:由序列编号1所示氨基酸序列的第1位~第19位氨基酸序列构成的多肽;包含活性多肽的多肽;和由序列编号1所示氨基酸序列的第29位~第86位氨基酸序列构成的多肽。本发明的蛋白质骨架模块使内置的活性多肽的结合亲和力或结合特异性增强,从而在疾病的诊断和治疗中是有效的,因而工业实用性大。
Claims (17)
1.一种内置有活性多肽的蛋白质骨架模块,其特征在于,其包含下述多肽依次连接而成的多肽:
a)由序列编号1所示氨基酸序列的第1位~第19位氨基酸序列构成的多肽;
b)包含活性多肽的多肽;以及
c)由序列编号1所示氨基酸序列的第29位~第86位氨基酸序列构成的多肽。
2.一种多聚核苷酸,其编码权利要求1所述的内置有活性多肽的蛋白质骨架模块。
3.一种表达载体,其包含权利要求2所述的多聚核苷酸。
4.一种宿主细胞,其经权利要求3所述的表达载体所转化。
5.一种内置有活性多肽的蛋白质骨架模块的制造方法,其包括对权利要求4所述的宿主细胞进行培养的工序。
6.一种诊断用组合物,其包含权利要求1所述的内置有活性多肽的蛋白质骨架模块作为有效成分。
7.一种药物组合物,其包含权利要求1所述的内置有活性多肽的蛋白质骨架模块作为有效成分。
8.一种诊断用试剂盒,其包含权利要求1所述的内置有活性多肽的蛋白质骨架模块作为有效成分。
9.一种内置有活性多肽的蛋白质骨架模块,其特征在于,其包含下述成分依次连接而成的多肽:
a)由序列编号1所示氨基酸序列的第1位~第19位氨基酸序列构成的多肽;
b)包含序列编号3所示的激活蛋白-1也即AP-1的氨基酸序列的多聚核苷酸;以及
c)由序列编号1所示氨基酸序列的第29位~第86位氨基酸序列构成的多肽。
10.一种多聚核苷酸,其编码权利要求9所述的内置有活性多肽的蛋白质骨架模块。
11.如权利要求10所述的多聚核苷酸,其特征在于,所述多聚核苷酸为选自由序列编号4~序列编号6组成的组中的任意一种。
12.一种癌症或动脉硬化诊断用组合物,其包含权利要求9所述的内置有活性多肽的蛋白质骨架模块作为有效成分。
13.一种癌症或动脉硬化诊断用试剂盒,其包含权利要求9所述的内置有活性多肽的蛋白质骨架模块作为有效成分。
14.一种诊断方法,其包括将权利要求1所述的内置有活性多肽的蛋白质骨架模块以有效量对于所需要的个体进行给药的工序。
15.权利要求1所述的内置有活性多肽的蛋白质骨架模块的用途,其用于制造诊断用制剂。
16.一种癌症或动脉硬化诊断方法,其包括将权利要求9所述的内置有活性多肽的蛋白质骨架模块以有效量对于所需要的个体进行给药的工序。
17.权利要求9所述的内置有活性多肽的蛋白质骨架模块的用途,其用于制造癌症或动脉硬化诊断用制剂。
