Abstract

Description
本发明涉及对织物的洗涤,公开了洗衣用洗涤剂组合物,该洗涤剂组合物专门设计用于洗涤有色织物。本发明的洗涤剂组合物可保持色彩的清晰度,尤其是经过多次洗涤之后。
洗衣用洗涤剂组合物在现有技术中是已知的。已知在这些组合物中使用了乙烯基吡咯烷酮聚合物,如EP262897和EP256696中所述。还已知在专门设计用于有色织物洗涤的洗涤剂组合物中使用了这些聚合物,如EP-A-265 257中公开的组合物含有乙烯基吡咯烷酮聚合物、羧酸酯聚合物和羧甲基纤维素;EP372291描述的洗涤剂组合物包括特殊的表面活性剂混合物和乙烯基吡咯烷酮聚合物。更确切地,正如DE2814287和DE2814329中所述,已知乙烯基吡咯烷酮聚合物在洗衣过程中起到染料转移抑制剂的作用。
还已知在洗衣用洗涤剂组合物中使用酶,包括使用纤维素酶。已知纤维素酶对保持织物颜色具有一定的作用,因为纤维素酶能够控制织物起球。这一内容在如EP177165中已有描述。
现已发现通过使用包括聚乙烯吡咯烷酮和纤维素酶的洗涤剂组合物,洗衣时可以获得优良的织物色彩保持;还惊奇地发现,这两种组分的协同作用优于二者单独使用时其作用的加和。
本发明的组合物是包含常规去污组分的洗衣用洗涤剂组合物,其特征在于该组合物包括碱性纤维素酶和分子量5000至1000000的聚乙烯吡咯烷酮,前者在成品中的含量使得在每升洗涤溶液中释放出0.005至40mg的所述纤维素酶,后者在成品中的含量使得在每升洗涤溶液中释放出5至500mg的所述聚乙烯吡咯烷酮。
本发明的洗衣用洗涤剂组合物包含常规的去污组分,包括表面活性剂、助洗剂和微量组分。
用于本发明组合物的适宜表面活性剂包括阴离子表面活性剂,如下列物质的水溶性盐:烷基苯磺酸盐、烷基硫酸盐、烷基聚乙氧基醚硫酸盐、链烷磺酸盐、α-烯烃磺酸盐、α-磺基烷基羧酸盐或其酯、烷基甘油基醚磺酸盐、脂肪酸甘油单酯硫酸盐和磺酸盐、烷基酚聚乙氧基醚硫酸盐、2-酸基链烷-1-磺酸盐以及β-烷氧基磺酸盐。
特别优选的烷基苯磺酸盐在直链或支链烷基链上具有9-15个碳原子,优选11-13个碳原子。适宜的烷基硫酸盐在烷基链上具有10-22个碳原子,优选具有12-18个碳原子。适宜的烷基聚乙氧基醚硫酸盐在烷基链上具有10-18个碳原子,并且平均每个分子中含有1-23个-CH2CH2O-基团;优选在烷基链上具有10-16个碳原子,并且平均每个分子中含有1-6个-CH2CH2O-基团。
适宜的链烷磺酸盐基本上是直链的并且含有8-24个碳原子,优选含有14-18个碳原子。适宜的α-烯烃磺酸盐具有10-24个碳原子,优选14-16个碳原子;α-烯烃磺酸盐可通过与三氧化硫反应再接着中和而制备,中和的条件是使所有的磺酸内酯都水解成相应的羟基链烷磺酸盐。适宜的α-磺基羧酸盐含有6-20个碳原子:这里不仅包括α-磺化脂肪酸的盐,还包括由含有1-14个碳原子的醇所制得的酯。
适宜的烷基甘油基醚硫酸盐是具有10-18个碳原子醇的醚,尤其是由椰子油和牛脂衍生的醚。适宜的烷基酚聚乙氧基醚硫酸盐在烷基链上具有8-12个碳原子,并且平均每个分子中含有1-6个-CH2CH2O-基团。适宜的2-酸基链烷-1-磺酸盐在酰基中含有2-9个碳原子,在链烷部分含有9-23个碳原子。适宜的β-烷氧基链烷磺酸盐在烷基中含有1-3个碳原子,在链烷部分含有8-20个碳原子。
适用于本发明组合物的非离子表面活性剂是HLB为11.5-17.0的水溶性乙氧基化物质,包括C10-20伯醇和仲醇乙氧基化物以及C6-10烷基酚乙氧基化物。与7-30摩尔环氧乙烷(每摩尔醇)缩合的C14-18直链伯醇是优选的实例,它们是C14-C15(EO)7、C16-18(EO)25、尤其是C16-18(EO)11。
另一类非离子表面活性剂包括通式为
RO(CnH2nO)tZx的烷基聚葡糖苷化合物,其中Z是衍生自葡萄糖的部分;R是含有12-18个碳原子的饱和疏水烷基;t为0至10,n是2或3;x为1.3至4,该化合物包括少于10%的未反应的脂肪醇和少于50%的短链烷基聚葡糖苷。这类化合物及其在洗涤剂中的应用公开在EP-B0070077、0075996和0094118中。
适宜作为非离子表面活性剂的还有通式为的多羟基脂肪酸酰胺表面活性剂,其中R1是H、C1-4烃基、2-羟乙基、2-羟丙基或这些基团的组合,R2是C5-31烃基,以及Z是具有直链烃基链的多羟基烃基,至少有三个羟基与该直链烃基链直接相连。优选地,R1是甲基,R2是直链C11-15烷基或链烯基如椰子烷基或这些基团的基合,以及Z在还原胺化条件下衍生自还原性糖,如葡萄糖、果糖、麦芽糖、乳糖。
也可使用其他类型的表面活性剂,如两性离子及两性表面活性剂以及阳离子表面活性剂。
可用于本发明的阳离子辅助表面活性剂包括R4R5R6R7N+X-类型的水溶性季铵盐化合物,其中R4是具有10-20、优选12-18个碳原子的烷基,R5、R6和R7分别是C1-C7的烷基,优选甲基;X-是阴离子,如氯离子。这些三甲基铵化合物的实例包括氯化C12-C14烷基三甲基铵和甲硫酸盐椰子烷基三甲基铵。本发明的组合物包括1-70%wt表面活性剂,优选10%-30%,最优选15%-25%。助洗剂
本发明使用的适宜助洗剂包括次氮基三乙酸盐、多羧酸盐、柠檬酸盐、水溶性磷酸盐如三聚磷酸盐及正和焦磷酸钠、及其混合物。金属离子螯合剂包括上面的所有物质,以及象乙二胺四乙酸盐、氨基多膦酸盐和各种其他多官能有机酸和盐的物质,其数目太多,在此不能一一列举。作为这些物质用于各种洗涤组合物中的具体例子,参见US3,579,454。用于本发明的优选的多官能有机酸种类有柠檬酸、乙二胺四亚甲基膦酸和二亚乙基三胺五亚甲基膦酸。
另一类在本发明中有用的去污助洗物质是不溶性硅铝酸钠。德国专利2422655公开的1-10微米粒径沸石(例如沸石A)助洗剂特别优选用于低磷酸盐组合物中。
本发明的组合物还可包含饱和或不饱和的脂肪酸,以及相应的皂。适宜的饱和或不饱和脂肪酸在烷基链中含有10-18个碳原子。优选在烷基链中含有14-18个碳原子的不饱和脂肪酸,最优选油酸。也可使用其相应的皂。
本发明的组合物还可含有通式为R-CH(COOH)CH2(COOH)的化合物,即琥珀酸的衍生物,其中R是C10-20的烷基或链烯基,优选C12-16的烷基或链烯基,或者其中的R可被羟基、磺基、亚砜或砜取代基取代。
琥珀酸盐助洗剂优选以其水溶性盐形式使用,包括钠、钾、铵和醇铵盐。
琥珀酸盐助洗剂的具体例子包括:月桂基琥珀酸盐、肉豆蔻基琥珀酸盐、棕榈基琥珀酸盐、2-十二碳烯基琥珀酸盐(优选)、2-十五碳烯基琥珀酸盐等等。
还可在本文中用作助洗剂的是US4,663,071中描述的化合物,即单琥珀酸酒石酸盐和二琥珀酸酒石酸盐的混合物,单琥珀酸与二琥珀酸的重量比为97∶3至20∶80,优选95∶5至40∶60。
本发明的组合物包括1%至70%、优选30%至60%、最优选40%至50%的助洗剂。
本发明组合物的特征在于它们包括碱性纤维素酶和聚乙烯吡咯烷酮。正是组分的这一特定组合提供了本发明组合物的优良的织物色彩保护性质。当织物用本发明的组合物重复洗涤时,较好地获得了织物色彩保护效果。因此本发明还包括使用本发明的组合物对织物进行多次洗涤的洗涤织物的方法。纤维素酶
用于本发明的纤维素酶可以是任何细菌或真菌纤维素酶,具有的最佳pH介于5和9.5之间。
适宜的纤维素酶公开在GB-A-2 075 028;GB-A-2 095 275和DE-OS-2 447 832中。
这些纤维素酶的例子是由Humicola insolens(Humicola griseavar.thermoidea)菌株、特别是Humicola菌株DSM1800产生的纤维素酶,由杆菌属N真菌或属于气单胞菌属的产生纤维素酶212的真菌产生的纤维素酶,以及从海洋软体动物(Dolabella AuriculaSolander)的肝胰腺提取出的纤维素酶。
加入本发明组合物的纤维素酶可以以非粉末状颗粒的形式,如″丸″或″小球″,或以液体形式,在液体形式中纤维素酶是以悬浮于(例如)非离子表面活性剂或溶于水介质中的纤维素酶浓缩物提供的。
优选用于本发明的纤维素酶的特征在于,根据C14CMC方法,当在洗衣试验溶液中纤维素酶蛋白质的浓度是25×10-6%wt时,所述纤维素酶能够除去至少10%的固定的放射性标记羧甲基纤维素。
其他适宜的水溶性有机盐是均聚或共聚酸或其盐,其中聚羧酸包括至少两个彼此被不多于两个碳原子分开的羧基。
这种类型的聚合物公开在GB-A-1,596,756中。这些盐的例子是MW2000-5000的聚丙烯酸盐和其与马来酸酐的共聚物,该共聚物具有20,000至70,000、尤其是约40,000的分子量。纤维素酶
通过不同的分析方法确定了对于各种应用的酶的活性、尤其是纤维素酶的活性。这些方法都是试图提供对所预期的使用性能的真实估价,或至少提供与使用性能有关的测量结果。正如欧洲专利申请EP-A-350098中详细说明的那样,许多方法,尤其是被纤维素酶生产者所经常使用的那些方法,与洗衣用洗涤剂组合物中纤维素酶的使用性能没有充分的联系;原因在于这些活性测量方法所赖于产生的各种其他使用条件。
EP-A-350098描述的方法被发展成为对洗衣用洗涤剂组合物中的纤维素酶活性等级具有预测关系的方法。
因此,本发明使用EP-A-350098公开的方法筛选纤维素酶,以区分能用于本发明的纤维素酶和那些不能达到本发明目的的纤维素酶。下文称之为C14CMC方法的筛选方法是从EP-A-350098公开的方法沿用而来,可叙述如下:原理:
C14CMC方法筛选的原理是,在给定的洗涤溶液中纤维素酶浓度下,测量从衣服基质上除去的固定羧甲基纤维素(CMC)。除去的CMC通过用C14放射性碳对一些CMC进行放射性标记来测定。对于纤维素酶处理前后衣服基质上放射性C14量的简单计数便可评价出纤维素酶活性。
样品制备:
CMC的制备:根据表I制备放射性CMC储液。参照EP-A-350098的方法可获得放射性CMC。
织物基质:织物基质是尺寸为5cm×5cm的小块细棉布,在其中心灌注0.35ml放射性标记的CMC储液,然后将小块细棉布晾干。
CMC的固定:为了将放射性标记的CMC固定在小块细棉布上,使用了德国Original Haunau制造的洗衣测量仪器″Linitest OriginalHaunau″。在测量仪的金属缸中注入400ml硬水(Ca++离子浓度为4mmol/l);每个金属缸最多使用13块布条。然后在洗衣测量仪器中给金属缸加热保温,在40分钟内经过20℃-60℃的升温循环。保温过程结束后,小布条用自来水冲洗1分钟,挤干,空气干燥至少30分钟。
根据EP-A-350098,固定放射性CMC的布条样品在不经洗涤时也可当作″空白样品″测试。样品处理:
洗衣试验溶液:根据表II的组成制备洗衣试验溶液,平衡至pH7.5。洗衣试验溶液是将纤维素酶试验样品加入的基质。在确定加入纤维素酶的量之前通过加水平衡至100%时,应当小心不要稀释洗衣试验溶液。在该筛选试验中使用的纤维素酶的量,应当能在洗衣试验溶液中提供25×10-6%wt的纤维素酶蛋白质(相当于14.5℃时0.25mg/l)。
洗涤方法:在洗衣模拟过程中处理用放射性标记CMC灌注的布条。洗衣模拟过程是在由德国Haunau,Original Haunau制造的洗衣测量型仪器″Linitest Original Haunau″中进行。将单块布条放入20cm3玻璃瓶中,将瓶中装入10ml洗衣试验溶液,然后密封液体。每个洗衣测量仪的缸中至多放5只瓶子,在缸中灌入水作为洗衣模拟的热传递介质。在40分钟内随着从20℃至60℃的升温循环进行洗衣模拟。
样品的模拟过程结束后,将玻璃瓶没入冷水中,然后将每块布条从各自的瓶子中取出,在烧杯中用软自来水冲洗,挤干,并空气干燥至少30分钟。测量:
为了测量除去的放射性标记CMC,使用了闪烁计数器,如LKB1210 Ultrabeta闪烁计数器。为了获得最准确的结果,应该遵循特定闪烁计量器的最佳操作规程。例如,对于LKB 1210 Ultrabeta闪烁计数器,应按下列步骤操作。将待测布条放入装有12ml闪烁器液体(如Packard生产的闪烁器299)的塑料瓶中。接着使布条稳定至少30分钟,再将塑料瓶放入LKB 1210 Ultrabeta闪烁计数器,即可获得布条的相应放射性读数。
为了测量仅由纤维素酶导致的CMC除去量,必须测量用不含纤维素酶的洗衣试验溶液处理而同时灌注的布条。那么,纤维素酶活性可用放射性标CMC除去百分数表示,该百分数通过下式计算:其中:XO是用不含纤维素酶的洗衣试验溶液处理的布条的放射性闪烁计数:

统计学考虑,方法校准:
为了提供统计学上可靠的结果,应当引入标准化统计分析。对于给出的使用LKB1210 Ultrabeta闪烁计数器的实例,已发现对每个放射性闪烁计数可采用3个布条样品。
为了通过内部相互校核而对测量方法进行校准,推荐使用EP-A-350098的″空白样品″的测量和计算,这将能发现并减少误差。结果分析:
这里叙述的筛选试验确实提供了一个快速、独特和可信的方法,该方法可用于鉴别符合本发明活性标准的纤维素酶与不是本发明组成部分的纤维素酶。
已经发现,根据上述C14CMC方法,能除去10%或更多固定的放射性标记CMC即表明相应的纤维素酶满足了本发明的要求。
对本领域技术人员显而易见的是,除去百分数大于10%表明相应的纤维素酶具有更高的活性。因此可以预料,根据C14CMC方法在洗衣试验溶液中的蛋白质浓度下,纤维素酶提供大于25%或优选大于50%的放射性标记CMC除去量,将表明该纤维素酶用于洗衣用洗涤剂时具有更好的性能。
还可以预料,C14CMC方法中使用的纤维素酶浓度愈高,提供的除去百分率也愈高。然而,在纤维素酶浓度和由此获得的除去百分率之间不存在被证实的线性关系。

根据本发明,优选的纤维素酶是那些在国际专利申请WO91/17243中叙述的酶。例如,用于本发明组合物的纤维素酶制剂主要由同源内葡聚糖酶组分构成,该组分与对高纯43KD纤维素酶(衍生自Humicolainsolens,DSM1800)产生的抗体起免疫反应,或者该组分与所述的43KD内葡聚糖酶同源。
应当强调指出,本发明的所有纤维素酶都应满足上述筛选试验的标准。然而,在丹麦专利申请1159/90中建立了附加标准,该标准可以与本发明的筛选试验相结合,从而鉴别优选的纤维素酶。
对本发明组合物特别有用的纤维素酶制剂是那些酶,它们除了通过筛选试验以外,其内葡聚糖酶组分具有的CMC内酶活性为每毫克总蛋白至少50,优选至少约60,特别是至少约90CMC内酶单位。特别是,优选内葡聚糖组分具有的CMC内酶活性为每毫克总蛋白至少100CMC内酶单位。
本文中术语″CMC内酶活性″是指根据内葡聚糖酶将纤维素降解为葡萄糖、纤维素二糖和丙糖的能力而确定的内葡聚糖酶组分的内葡聚糖酶活性,该活性是通过注入本发明的纤维素酶制剂之后羧甲基纤维素(CMC)溶液的粘度的降低来确定的,下文将详细叙述。
CMC内酶(内葡聚糖酶)活性可通过CMC粘度的降低测定如下:配制底物溶液,在pH9.0的0.1M缓血酸胺缓冲液中含有35g/lCMC(Hercules 7LFD)。将待分析的酶样品溶解于相同的缓冲溶液中。混合10ml底物溶液和0.5ml酶溶液,并转移至粘度汁(如Haake VT181,NV传感器,181rpm)中,恒温在40℃。混合后再等30分钟尽快读出粘度读数。在这些条件下将粘度减至一半时酶的用量被定义为1单位CMC内酶活性。
本领域技术人员熟知的SDS聚丙烯酰胺凝胶电泳(SDS-PAGE)和用蛋白质标记的等电点聚焦被用来分别测定用于本发明的纤维素酶制剂中的内葡聚糖酶组分的分子量和等电点(PI)。这样,该内葡聚糖酶组分的分子量被测定为43KD。该内葡聚糖酶的等电点被测定为约5.1。
纤维素二糖水解酶的活性可定义为,对于对硝基苯基纤维素二糖的活性。该活性被确定为在37℃和pH7.0时每分钟释放出的硝基苯的微摩尔数。已发现本发明的内葡聚糖酶组分基本没有纤维素二糖水解酶活性。
在本发明纤维素酶制剂中的内葡聚糖酶组分最初是通过彻底纯化方法分离得到的,即根据US4,435,307的方法,包括粗H.inSolens纤维素酶混合物的反相HPLC纯化。该纯化方法令人惊奇地分离出作为单一组分的43KD内葡聚糖酶,该组分因其极高的内葡聚糖酶活性而具有意想不到的优良性质。
而且,除了筛选试验,在本发明组合物中有用的纤维素酶还可进一步定义为显示内葡聚糖酶活性的酶(下文称作″内葡聚糖酶″),这些酶具有下面所附顺序表ID#2所示的氨基酸顺序,或者定义为显示内葡聚糖酶活性的酶的同源酶。
在本文中,术语″同源″用来表示被DNA编码的多肽,该多肽在某一特定条件(如在5xSSC中预浸,并在20%甲酰胺、5xDenhardt氏溶液、50mM磷酸钠(pH6.8)和50ug变性声处理的小牛胸腺DNA的溶液中于40℃预杂交1小时,接着在补充有100uM ATP的相同溶液中于40℃杂交18小时)下,杂交成与为带有该氨基酸顺序的内葡聚糖酶编码的DNA相同的探测物。该术语包括上述顺序的衍生物,这些衍生物的得到是通过在原顺序的C-和N-端的一端或两端加上一个或多个氨基酸残基,在原顺序的一个或多个部位取代一个或多个氨基酸残基,在原氨基酸顺序的一端或两端或在原顺序内的一个或多个部位删除一个或多个氨基酸残基,或在原顺序的一个或多个部位嵌入一个或多个氨基酸残基。
本文的内葡聚糖酶可通过Humicola种如Humicola insolens(例如DSM1800菌株)生产,根据国际承认用于专利程序的微生物保藏布达佩斯条约(布达佩斯条约)的条款,DSM1800菌株于1981年10月1日保藏在德国微生物收藏馆,Nascheroder Weg 1B,D-3300 Braunschweig,FRG。
再一方面,除了筛选试验外,用于本发明的纤维素酶还可定义为具有附录顺序表ID#4所示的氨基酸顺序的内葡聚糖酶或其(如上述定义)显示内葡聚糖酶活性的同源酶。所述内葡聚糖酶可通过镰孢属(Fusarium)菌如Fusorium oxysporum(例如DSM2672菌株)生产。根据布达佩斯条约的条款DSM 2672菌株于1983年6月6日保藏在德国微生物收藏馆,Mascheroder Weg 1B,D-3300 Braunschweig,FRG。
而且,可以预料从其他产生纤维素溶解酶的微生物,如木霉属(Trichoderma)、Myceliophthora、Phanerochaete、Schizophyllum、青霉属(Penicillium)、曲霉属(Aspergillus)和地丝菌属(Geotrichum)菌可衍生出同源内葡聚糖酶。
然而,对本发明纤维素酶制剂的工业生产来说,为保证产生过多的所需酶活性,优选使用DNA重组技术或其他技术,包括所涉及的微生物的发酵或变异调整。这些方法和技术在本领域是已知的,而且也容易被本领域的技术人员所实施。
内葡聚糖酶组分是可通过一种方法生产的组分,该方法包括培养用重组DNA载体转化的宿主细胞,DNA载体带有编码所述内葡聚糖酶组分或所述内葡聚糖酶组分前体的DNA顺序以及便编码内葡聚糖酶组分或其前体的DNA顺序得以表达的编码功能DNA顺序,所述方法在培养基上并在使内葡聚糖酶组分或其前体得以表达和使内葡聚糖酶组分从培养基得以回收的条件下进行。
包含上述编码内葡聚糖酶或该酶前体形式的DNA顺序的DNA结构,包括具有附录顺序表ID#1或ID#3所示的DNA顺序或其变体的DNA结构。适宜的DNA顺序变体的实例是这样的核苷酸替代物,该核苷酸替代物不能产生内葡聚糖酶的另一种氨基酸顺序,但相应于所用宿主微生物的密码子,在该宿主微生物内部引入了DNA结构;或是这样的核苷酸替代物,该核苷酸替代物确实产生了不同的氨基酸顺序,并因此可能产生不同的蛋白质结构,该蛋白质结构可能产生与原来的酶具有不同性质的内葡聚糖酶突变体。可能性变体另外的实例是在顺序的一端引入一个或多个核苷酸或者是在顺序的一端或内部删除一个或多个核苷酸。
在本发明中有用的编码内葡聚糖酶的DNA结构可通过已确定的标准方法合成制备,例如由S.L.Beaucage和M.H.Caruthers在Tetrahedron Letters 22,1981,PP.1859-1869中叙述的磷酰胺法,或由Matthes等人在EMBO Journal3,1984,PP.801-805中叙述的方法。根据磷酰胺法,低聚核苷酸可在(例如)自动DNA合成器中合成,再纯化、韧化、连接并在适宜载体上无性繁殖。
编码内葡聚糖酶或其前体的DNA结构可被分离出来,例如,通过建立cDNA或产生纤维素酶微生物的基因库,如Humicola insolens,DSM1800,并采用常规方法筛选正性克隆,根据标准技术(参看Sambrook等人,Molecular Cloning:A Laboratory Manual,2nd.Ed.Cold Spring Harbor.1989),利用在内葡聚糖酶的全部或部分氨基酸顺序的基础上合成的低聚核苷酸探测物杂交;或通过选择表达适宜酶活性(也就是上面定义的CMC内酶活性)的克隆;或通过选择产生能与原纤维素酶(内葡聚糖酶)的抗体起反应的蛋白质的克隆。
最后,DNA结构可以来源于混合的合成与基因源、混合的合成与cDNA源或混合的基因与cDNA源,其制备是根据标准技术连接合成、基因或cDNA源(合适的)片段,而这些片段则相应于整个DHA结构的各个部分。DNA结构也可采用特定的底物通过聚合酶链反应进行制备,如US4,683,202或R.k.Saiki等人在Science 239.1988,PP.487-491中所述。
在其上嵌入上述DNA结构的重组表达载体包括任何能方便地进行重组DNA方法的载体,而且载体的选择常常取决于将其引入其中的宿主细胞。因此,载体可以是自动复制的载体,也就是能作为染色体外实体存在的载体,其复制不依赖于染色体的复制,例如质粒。另一方面,载体可以是这样的载体,当它被引入宿主细胞时,它就结合到宿主细胞基因组中,并与将其结合的染色体一起复制。
在载体中,编码内葡聚糖酶的DNA顺序应该能够连到适宜的启动基因和终止基因顺序上。启动基因可以是任何在所选择的宿主细胞中显示转录活性的DNA顺序,并可衍生自编码与宿主细胞同源或异源蛋白质的基因。用于分别连接编码内葡聚糖酶的DNA顺序、启动基因和终止基因,并将它们引入适宜的载体中的方法是本领域的技术人员所熟知的(参看,例如,Sambrook等人的op.cit.)。
用上述DNA结构或上述表达载体转换的宿主细胞可属于(例如)Aspergitlus种、最优选Aspergillys oryzae或Aspergillusniger。通过包括原生质体形成和原生质体转换、接着再生细胞壁的方法,可进行真菌细胞转换;该方法本身为已知的。用Aspergillus作为宿主微生物记载在EP238 023(属于Novo Industri A/S)中,该专利的内容在此引入作为参考。宿主细胞也可以是酵母细胞,如Saccharomyces cerevisiae菌株。
另一方面,宿主生物可以是细菌(尤其是Streptomyces和Bacillus菌株)和大肠埃希杆菌。细菌细胞的转换可按照常规方法进行,例如Sambrook等人在Molecular Cloning:A Laboratory Manual(Cold Spring Harbor,1989)中叙述的方法。
适宜DNA顺序的筛选以及载体的构成也可按标准方法进行,参考Sambrook等人的op.cit.。
用于培养转换的宿主细胞的培养基可以是任何适于生长所述宿主细胞的普通培养基。表达的内葡聚糖酶可方便地隐入培养基并通过已知方法从培养基中回收;该已知方法包括通过离心或过滤将细胞与培养基分离,利用盐如硫酸铵沉淀培养基的蛋白质或分,接着利用色谱方法,如离子交换色谱、亲合色谱等。
通过使用上面指出的重组DNA技术、蛋白质纯化技术、发酵和变异技术或本领域已知的其他技术,就有可能提供高纯度的内葡聚糖酶。
上述纤维素酶在本发明组合物中的含量,应当使洗涤溶液中释放出的酶蛋白质的量为每升洗涤溶液0.005-40mg,优选每升洗涤溶液0.01-10mg。乙烯吡咯烷酮聚合物
本发明组合物还包括具有分子量5000-1000000、优选5000-50000、最优选8000-15000的聚乙烯吡咯烷酮。聚乙烯吡咯烷酮在本发明组合物中的含量,应当使洗涤溶液中释放出的聚乙烯吡咯烷酮的量为5-500mg/l、优选15-100mg/l、最优选25mg/l-75mg/l。任选组分
本发明组合物一般包括通常构成洗涤剂组合物任 选组分。抗再沉积和污垢悬浮剂、荧光增白剂、白土、漂白剂、漂白活化剂、抑泡剂、防结块剂、染料、香料和颜料是这些任选组分的例子,并且它们可按需要以不同的量加入。
本发明组合物可以呈液态、糊状或粒状,优选呈粒状。本发明的粒状组合物也可以是″浓缩型″,即该粒状组合物比普通粒状洗涤剂具有相对更高的密度,即550-950g/l;在这种情况下,本发明的粒状洗涤剂组合物与普通粒状洗涤剂相比含有较低量的″无机盐填料″;典型的填料盐是碱性醚金属硫酸盐和氯化物,特别是硫酸钠;″浓缩″洗涤剂一般含有不超过10%的填料盐。液态和普通粒状洗涤剂的一般使用浓度为洗涤液体的1-2%wt,优选1.5%wt;而浓缩粒状洗涤剂的使用浓度为0.5-1.5%wt,优选1%wt。
下列实施例说明本发明和由本发明获得的意想不到的优良色彩保护效果。

用该组合物,其在洗涤液体中的浓度为0.7%,将由两种有色织物(91%棉/9%聚酯蓝色编织物和桃红色100%纯棉法兰绒材料)组成的待洗物洗涤十次。在第十个洗涤循环,另加将颜色扩散到洗涤液中的织物。在一个实验中,此另加的织物是红色棉织品;在另一个实验中,此另加的织物是棕色棉织品。然后使用具有下列标度的评审标记评价被洗物的颜色外观:
1、我认为我看到了与对比物的区别
2、我肯定看到了与对比物的区别
3、我看到了与对比物的很大的区别

接着在上述洗涤剂组合物中或者补加1%PVP,或者补加0.025%纤维素酶,或者两者0.025%纤维素酶和1%PVP均加入。按上述的方法再次评价颜色外观,结果总结在下面的表中。
这些结果表明PVP和纤维素酶一起的作用出人意料地优于这两种组分单独作用的加和。

Claims (11)
1、包括普通去污组分的洗衣用洗涤剂组合物,其持在于它包括碱性纤维素酶和分子量为5000-100000的聚乙烯吡咯烷酮,碱性纤维素酶在成品中的含量使得在洗涤溶液中释放出0.005-40mg/l的所述纤维素酶,聚乙烯吡咯烷酮在成品中的含量使得在洗涤溶液中释放出5-500mg/l的所述聚乙烯吡咯烷酮,其中,根据C14CMC方法,在洗衣试验溶液中纤维素酶蛋白质为25×10-6%wt时,所述纤维素酶能够除去至少10%的固定的放射性标记羧甲基纤维素,和纤维素酶基本上由同源内葡聚糖酶组分构成,该组分与对部分纯化的约≈43KD纤维素酶(衍生自Humicola insolens)产生的单克隆抗体起免疫反应,或该组分与所述的≈43KD内葡聚糖酶同源。
2、根据权利要求1的洗衣用洗涤剂组合物,其中所述洗涤剂组合物含有不超过15%wt的无机盐填料,以及
所述洗涤剂组合物具有550-950g/l(组合物)的密度。
3、根据权利要求1或2的洗衣用洗涤剂组合物,其中纤维素酶在成品中的含量使得在洗涤液体中释放出0.01mg/l-10mg/l的纤维素酶。
4、根据权利要求1或2的洗衣用洗涤剂组合物,其中聚乙烯吡咯烷酮具有5000-50000的分子量。
5、根据权利要求1或2的洗涤剂组合物,其中聚乙烯吡咯烷酮的含量使得在洗涤溶液中释放出15mg/l-100mg/l、的聚乙烯吡咯烷酮。
6、根据权利要求4的洗衣用洗涤剂组合物,其中聚乙烯吡咯烷酮具有8000-15000的分子量。
7、根据权利要求5的洗涤剂组合物,其中聚乙烯吡咯烷酮的含量使得在洗涤溶液中释放出25mg/L-75mg/L的聚乙烯吡咯烷酮。
8、根据权利要求1或2的洗衣用洗涤剂组合物,其中所述的普通去污组分由表面活性剂和助洗剂构成。
9、根据权利要求1的洗衣用洗涤剂组合物,它呈粒状形式。
10、根据权利要求8的洗衣用洗涤剂组合物,它具有550g/l-950g/l的密度。
11、洗涤织物的方法,其中所述织物用权利要求1或2的洗涤剂组合物多次洗涤。
