人类皮肤包括三个主要层:表皮、真皮和皮下脂肪层。表皮包括四层(从顶部到底部):角质层、颗粒层、棘层和基底层。角质层和颗粒层之间可存在单独的第五层透明层。基底层产生细胞,所述细胞逐渐向上迁移以形成其它表皮层。随着这些细胞向上迁移,它们失去它们的中央核,并且开始产生皮肤蛋白(角蛋白)和脂肪(脂质)。当存在于表皮的上层时,这些细胞被称为角质细胞。黑素细胞是位于表皮的基底层中的另一类细胞。黑素细胞造成黑素的产生,黑素是皮肤外观的主要因素颜料。
参见图1、2和3,黑素是在黑素细胞中由一组复杂的反应产生的,这些反应以基本水平涉及酪氨酸酶以及作为底物的L-酪氨酸。酪氨酸酶催化L-酪氨酸转化成DOPA(L-3,4-二羟基苯丙氨酸)以及DOPA转化成多巴醌。多巴醌进一步经历转化以形成黑素。黑素聚集于被称为黑素体的细胞器官中,所述细胞器官沿着被称为枝状体的黑素细胞细长丝转移至角质细胞。酪氨酸酶及其活性的产生部分地决定所产生黑素的量。就角质细胞而言,转移至角质细胞的黑素量和类型决定人皮肤的可见色素沉着程度。
黑素的产生周期的一个机理是通过细胞的吞噬作用将黑素体从黑素细胞中转移到角质细胞。研究已发现,在角质细胞上表达的蛋白酶-活化的受体2(PAR-2)涉及黑素体的转移,因此PAR-2可调控色素沉着。参见例如Seiberg,M.等人的The Protease-Activated Receptor2RegulatesPigmentation via Keratinocyte-Melanocyte Interactions,Experimental CellResearch,254,25-32(2000)。用胰蛋白酶(或胰蛋白酶样蛋白酶)(或用肽激动剂SLIGRL)活化PAR-2,引发黑素体转移,从而有助于黑素在角质细胞内的分布。在一些情况下,黑素的分布可表现为老年斑或不均匀的皮肤色调。据信,抑制PAR-2的胰蛋白酶(或胰蛋白酶样蛋白酶)活化的化合物通过角质细胞来破坏或降低黑素细胞的吞噬作用。通过抑制胰蛋白酶活性或通过抑制SLIGRL肽结合而抑制PAR-2的化合物可调控色素沉着过度和黑素过度产生。参见例如Seiberg,M.等人的The Protease-Activated Receptor2Regulates Pigmentation via Keratinocyte-MelanocyteInteractions,Experimental Cell Research,254,25-32(2000)。
在年轻皮肤中,黑素是均匀分布的,并且黑素细胞活性低,仅限于产生基本色素沉着。在老化皮肤中,过度的黑素产生和随后的黑素转移可最终产生永久性局部变色,其具有足够的尺寸和对比度,而表现为老年斑(雀斑)或扩散性色素沉着过度或不均匀的皮肤色调。化妆品领域中持续需要最小化、降低、改善、或治疗老年斑的尺寸和/或对比度和/或改善总体皮肤色调的治疗组合物和方法。
发明内容
提供了配制用于向皮肤局部施用的化妆品组合物。所述组合物包含安全且有效量的环己-1,2,3,4,5,6-六醇;安全且有效量的N-酰基氨基酸化合物;和皮肤病学可接受的载体。
在另一个实施例中,所述化妆品组合物包含安全且有效量的环己-1,2,3,4,5,6-六醇;安全且有效量的N-酰基氨基酸化合物;安全且有效量的维生素B3化合物;安全且有效量的2-己基癸-1-醇;和皮肤病学可接受的载体。
还提供了用于改善、减少或治疗不均匀皮肤色调或老年斑的美容方法。所述方法包括鉴定需要此类治疗的皮肤表面;向皮肤表面局部施用化妆品组合物,所述组合物包含安全且有效量的环己-1,2,3,4,5,6-六醇、安全且有效量的N-酰基氨基酸化合物;和皮肤病学可接受的载体。在治疗阶段期间可将所述方法重复足够的次数,以改善、减少、或治疗皮肤表面的不均匀皮肤色调或老年斑。

除非另外指明,本文所用的所有百分比和比率均按总体组合物的重量计,并且所有测量均在25℃下进行。所有数值范围是包括端值在内的较窄的范围;描述的范围上限和下限是可互换的,以进一步形成没有明确描述的范围。
本发明的组合物和方法可包含、基本上由或由本文所描述的基本组分以及任选成分组成。如本文所用,“基本上由...组成”是指所述组合物或组分可包含附加成分,只要附加成分不在本质上改变本发明权利要求保护的组合物或方法的基本特性和新型特性。

I.组合物
本发明的实施例包含N-酰基氨基酸化合物和环己-1,2,3,4,5,6-六醇的组合,并且还可任选包含一种或多种维生素B3化合物或2-己基癸-1-醇。本发明人已惊奇地发现,N-酰基氨基酸化合物与环己-1,2,3,4,5,6-六醇的组合一定程度地抑制胰蛋白酶活性,这对化合物个体是出乎意料地有利或优异的。发明人还惊奇地发现,N-酰基氨基酸化合物、环己-1,2,3,4,5,6-六醇、维生素B3化合物和2-己基癸-1-醇的组合一定程度地抑制胰蛋白酶活性,这对化合物个体是出乎意料地有利或优异的。
本发明的组合物可被制成多种产品形式,包括但不限于溶液、悬浮液、乳液、霜膏、凝胶、调色剂、棒状物、笔剂、喷剂、气溶胶、膏剂、糊剂、泡沫、粉末、摩丝、剃刮霜膏、擦拭物、试纸、遮瑕膏、粘合剂绷带、水凝胶、成膜产品、面部和皮肤面膜(具有和不具有不溶薄片)、化妆品如粉底、眼线和眼影等。组合物形式可符合所选的具体皮肤病学可接受的载体(如果存在于组合物中)。
A.N-酰基氨基酸化合物
本发明的组合物包含安全且有效量的一种或多种N-酰基氨基酸化合物。氨基酸可为本领域已知氨基酸中的任何一种。本发明的N-酰基氨基酸化合物符合下式:
其中R可为氢、烷基(取代或未取代的、支化或直链的)、或烷基和芳基的组合。本领域已知的氨基酸的可能侧链的列表描述于1981年由W.H.Freeman and Company公布的Stryer,Biochemistry中。R1可为饱和或不饱和的、直链或支化的、取代或未取代的C1-C30烷基;取代或未取代的芳族基团;或它们的混合物。
N-酰基氨基酸化合物优选选自N-酰基苯丙氨酸、N-酰基酪氨酸、它们的异构体、它们的盐、以及它们的衍生物。氨基酸可为D或L异构体、或它们的混合物。N-酰基苯丙氨酸符合下式:

C.维生素B3化合物
本发明的组合物可任选包含安全且有效量的一种或多种维生素B3化合物,所述化合物具有式:

前述维生素B3化合物的示例性衍生物包括烟酸酯,包括烟酸的非血管舒张性酯(如生育酚烟酸酯、十四烷基烟酸酯)。适宜维生素B3化合物的例子是本领域熟知的,并且可从多个来源(如Sigma Chemical Company,ICN Biomedicals,Inc.和Aldrich Chemical Company)商购获得。

D.2-己基癸-1-醇
本发明的组合物包含安全且有效量的2-己基癸-1-醇,常称为己基癸醇。在本发明的组合物中,2-己基癸-1-醇可构成按所述组合物的重量计大于约0.001%、0.01%、或2.5%,和/或小于约10%或5%。
在一些实施例中,化妆品组合中N-酰基氨基酸化合物与环己-1,2,3,4,5,6-六醇化合物的比率介于约1:100至约10:1之间;或约1:60至约2:1之间,或约1:30至约1:1之间,或约1:3。
在一些实施例中,化妆品组合中N-酰基氨基酸化合物与环己-1,2,3,4,5,6-六醇化合物与维生素B3化合物与2-己基癸-1-醇的比率介于约0.2:2:5:5和约1:3:5:5之间,或介于约1:2:5:5和约1:3:5:5之间,或介于约0.2:3:5:5和约1:3:5:5之间。
E.皮肤病学可接受的载体
本发明的组合物也可包含所述组合物的皮肤病学可接受的载体(“载体”)。如本文所用,短语“皮肤病学可接受的载体”是指载体适于局部施用到角质组织,具有良好的美学性质,与本发明的活性物质相容,并且不引起任何安全或毒性问题。在一个实施例中,所述载体的含量按所述组合物的重量计为约50%至约99%,约60%至约98%,约70%至约98%,或约80%至约95%。
所述载体可以多种形式提供。非限制性例子包括简单溶液(含水的或油基的)、乳液和固体形式(凝胶、棒状物、可流动的固体、无定形材料)。在某些实施例中,皮肤病学可接受的载体为乳液形式。乳液一般可分类为具有连续水相(如水包油和水包油包水)或连续油相(如油包水和油包水包油)。本发明的油相可包含有机硅油、非有机硅油(诸如烃油、酯、醚等)、以及它们的混合物。
水相通常包含水。然而,在其它实施例中,水相可包含不是水的组分(非水组分),包括但不限于水溶性保湿剂、调理剂、抗微生物剂、湿润剂和/或其它水溶性护肤活性物质。在一个实施例中,所述组合物中的非水组分包括湿润剂,诸如甘油和/或其它多元醇。然而,应当认识到,所述组合物可以是基本上(即小于1%的水)或完全无水的。
选择适宜的载体以获得所需的产品形式。此外,组合物组分(例如N-酰基氨基酸化合物、环己-1,2,3,4,5,6-六醇、维生素B3化合物和2-己基癸-1-醇)的溶解度或可分散度可决定载体的形式和组成。在一个实施例中,水包油或油包水的乳液是优选的。
乳液还可包含乳化剂。所述组合物可包含任何适宜百分比的乳化剂以充分乳化所述载体。适宜的重量范围包括按所述组合物的重量计约0.1%至约10%或约0.2%至约5%的乳化剂。乳化剂可为非离子的、阴离子的或阳离子的。适宜的乳化剂公开于例如美国专利3,755,560、美国专利4,421,769和McCutcheon的Detergents and Emulsifiers北美版第317-324页(1986)中。根据所需的产品形式,合适的乳液可具有宽范围的粘度。
如本领域所熟知,所述载体还可包含增稠剂以提供具有适宜粘度和流变特征的组合物。
II.胰蛋白酶测定实验
提供下列测试方法以示出本发明多个实施例的某些特征和优点,并且不应理解为限制其范围。
将得自牛胰腺的胰蛋白酶(购自Sigma-Aldrich,USA,目录号T8802)稀释于磷酸盐缓冲的盐溶液(PBS)中,以制得具有约0.001%胰蛋白酶的原液。在具有透明底部的96孔白色板(例如Corning Costar#3904,购自_VWR International,LLC(Pennsylvania,USA))中,将约90uL的0.001%胰蛋白酶原液与约10uL的测试化合物(例如N-十一碳烯酰-L-苯丙氨酸)或载体对照物如二甲基亚砜(DMSO)或水混合。通常,每板具有8个对照物孔。通过加入100uL基质Z-苯丙氨酸-精氨酸-7-酰氨基-4-甲基香豆素(例如Calbiochem目录号03-32-1501-10mM于PBS中,购自LifeTechnologies,California,USA)开始测定,并且在具有360nm激发波长和460nm发射波长的荧光读板器(例如Gemini读板器)中监测所述孔板。以动力学模式对反应进程测定5分钟。使用SoftMaxPro数据分析软件(购自Molecular Devices,Inc.)将每个孔的数据点拟合成线,然后计算线的斜率。将每种测试化合物孔的斜率与板中载体对照物的平均斜率进行比较,使用下文公式(1)确定胰蛋白酶活性百分比。由于胰蛋白酶水解基质,7-酰氨基香豆素组的荧光增加,造成了信号增加,这反映了胰蛋白酶活性水平。抑制剂使荧光增加衰减,从而获得较小的斜率。

一般采用上述方法,对每个化合物采用n=8孔的样本尺寸,测定1%N-十一碳烯酰-L-苯丙氨酸、3%肌醇、5%烟酰胺和5%己基癸醇的1:900稀释液的胰蛋白酶活性百分比。所得胰蛋白酶活性百分比示于下表1中。

一般采用上述方法,对每种组合采用n=8的样本尺寸,还测定了1%N-十一碳烯酰-L-苯丙氨酸、3%肌醇、5%烟酰胺和5%己基癸醇的四种组合的1:900稀释液的胰蛋白酶活性百分比。所述组合为:
组合#1=N-十一碳烯酰-L-苯丙氨酸和肌醇。
组合#2=N-十一碳烯酰-L-苯丙氨酸;肌醇;和烟酰胺。
组合#3=N-十一碳烯酰-L-苯丙氨酸;肌醇;和己基癸醇。
组合#4=N-十一碳烯酰-L-苯丙氨酸;肌醇;烟酰胺和己基癸醇。
所得胰蛋白酶活性百分比示于下表2中。

大于100%的百分比值代表胰蛋白酶活性的增量调节,而小于100%的百分比值代表胰蛋白酶活性的减量调节(抑制)。由表1和2中意识到,与各个化合物(例如A、B)相比,N-十一碳烯酰-L-苯丙氨酸和肌醇的组合(组合#1)具有令人惊奇地意料不到的或优异的胰蛋白酶抑制度。将烟酰胺加入组合#1中,致使与组合#1相比,胰蛋白酶活性增加,而将己基癸醇加入组合#1中,获得与组合#1相类似的胰蛋白酶活性。然而令人惊奇的是,将烟酰胺和己基癸醇加入组合#1中(组合#4),获得最低水平的胰蛋白酶活性(即最高的胰蛋白酶抑制水平)。
III.示例性组合物
以下是本发明组合物的非限制性实例。实例的给出仅是为了举例说明之目的,不应被解释为是对本发明的限制,因为在不背离本发明的实质和范围的情况下,其许多改变是可能的,这会被本领域的普通技术人员所认识到。在实例中,除非另外指明,所有浓度均按重量百分比列出,并且可排除次要材料,如稀释剂、填料等。因此,所列的制剂包括所列的组分以及与该组分有关的任何次要材料。对本领域的普通技术人员来说显而易见的是,这些微量物质的选择将根据选择制备如本文所述发明的具体成分的物理和化学特性而不同。
下文提供一些组合物的实例。
本发明组合物通常由常规方法制备,如本领域已知的制备组合物和局部用组合物的方法。上述方法通常涉及在一个或多个步骤中将成分混合至比较均一的状态,可使用或不使用加热、冷却、施加真空等。通常,通过首先将水相材料与脂肪相材料分开混合,然后视情况混合两相以获得所期望的连续相来制得乳液。优选制备所述组合物,以使活性物质的稳定性(物理稳定性、化学稳定性、光稳定性)和/或递送最优化。该最优化可包括合适的pH(如小于7)、排除可与活性物质复合并因而负面影响稳定性或递送的物质(如排除杂质铁)、使用预防复合物形成的方法(如合适的分散剂或双层包装)、使用合适的光稳定性方法(如加入防晒油/防晒霜、使用不透明的包装)等。
IV.治疗方法
各种治疗方法可利用本发明的组合物。在一个实施例中,所述方法包括用过组合物鉴定皮肤表面的步骤,所述皮肤表面包含一个或多个老年斑或不均匀的皮肤色调。皮肤表面可由使用者或第三方如皮肤科医师、美容师或其他护理人员进行鉴定。鉴定可通过基于尺寸和/或颜色来目视检查需要治疗的皮肤表面来完成。鉴定也可通过可商购获得的成像装置如V(购自Astron Clinica,Ltd.,UK)或肤色分析系统(购自Canfield Scientific,Inc.,Fairfield,NJ)来完成。两个装置均能够采集皮肤的图像并鉴定老年斑。
最令人关切的皮肤表面大多是那些通常未被衣服覆盖的部分,例如面部皮肤表面、手和臂皮肤表面、脚和腿部皮肤表面、以及颈和胸部皮肤表面(例如(经常穿)坦胸露背的衣服的)。具体地,面部皮肤表面的一个或多个老年斑和/或不均匀皮肤色调的鉴定包括前额、口周、下巴、眶周、鼻部、和/或面颊皮肤表面。
所述方法可包括向皮肤表面施用所述组合物的步骤,所述皮肤表面可能之前已被鉴定。存在许多方案来施用所述组合物。在治疗阶段,至少每天一次、每天两次,或以每天更频繁的方式施用所述组合物。治疗阶段可不同。在一个实施例中,治疗阶段介于约1周和约12周之间。在另一个实施例中,治疗阶段介于约4周和约12周之间。在另一个实施例中,治疗阶段介于约4周和约8周之间。在某些实施例中,治疗阶段将延续多个月(即3-12个月)或多年。在一个实施例中,在至少约4周或至少约8周的治疗阶段期间,至少每天一次施用所述组合物。在另一个实施例中,在至少约4周或8周的治疗阶段期间,每天两次施用所述组合物。当每天两次施用时,第一次和第二次施用可相隔至少1至约12小时。通常,所述组合物可在早上和/或晚上临睡前施用。
所述组合物可在包装件中提供,所述包装件的尺寸设定成贮存足量的组合物以用于治疗阶段。包装件的尺寸、形状和设计可广泛地变化。一些包装件例子描述于USPN D570,707;D391,162;D516,436;D535,191;D542,660;D547,193;D547,661;D558,591;D563,221;2009/0017080;2007/0205226;和2007/0040306中。
治疗阶段应为足够的时间以提供皮肤表面的改善。改善可为可察觉的老年斑尺寸减小、老年斑变淡(例如颜色更浅)、黑素减少、或黑素均匀度改善。

5.一种用于抑制或降低皮肤表面中的胰蛋白酶活性的美容方法,包括向皮肤表面局部施用前述权利要求中任一项所述的化妆品组合物。

8.根据前述权利要求中任一项所述的美容方法,其中所述化妆品组合物在包装件中提供,所述包装件的尺寸设定成贮存足量的所述化妆品组合物以用于所述治疗阶段。
9.根据前述权利要求中任一项所述的美容方法,其中所述化妆品组合物作为洗剂或霜膏提供。
10.根据前述权利要求中任一项所述的美容方法,其中所述皮肤表面为面部皮肤表面。
Claims (10)
1.一种配制用于向皮肤局部施用的化妆品组合物,包含:
安全且有效量的按所述化妆品组合物的重量计优选介于2%和3%之间的环己-1,2,3,4,5,6-六醇;
安全且有效量的N-酰基氨基酸化合物,优选十一碳烯酰苯丙氨酸,更优选地,其中所述十一碳烯酰苯丙氨酸的浓度按所述化妆品组合物的重量计介于0.2%和1%之间;和
皮肤病学可接受的载体。
2.根据权利要求1所述的化妆品组合物,还包含维生素B3化合物,优选烟酰胺,还更优选地,其中所述维生素B3化合物的浓度按所述化妆品组合物的重量计介于3%和5%之间。
3.根据前述权利要求中任一项所述的化妆品组合物,还包含2-己基癸-1-醇,优选地,其中所述2-己基癸-1-醇的浓度按所述化妆品组合物的重量计介于2.5%和5%之间。
4.一种用于改善、减少或治疗不均匀皮肤色调或老年斑的美容方法,包括:
a)鉴定需要此类治疗的皮肤表面;
b)向所述皮肤表面局部施用前述权利要求中任一项所述的化妆品组合物;以及
在治疗阶段期间将步骤b)重复足够的次数,以改善、减少、或治疗
所述皮肤表面的不均匀皮肤色调或老年斑。
5.一种用于抑制或降低皮肤表面中的胰蛋白酶活性的美容方法,包括向皮肤表面局部施用前述权利要求中任一项所述的化妆品组合物。
6.根据前述权利要求中任一项所述的美容方法,还包括在治疗阶段期间,重复向所述皮肤表面局部施用所述化妆品组合物的步骤。
7.根据前述权利要求中任一项所述的美容方法,其中所述治疗阶段介于4和12周之间,优选介于4和10周之间,并且其中在所述治疗阶段期间,将局部施用所述化妆品组合物的步骤重复至少一次/天,优选在所述治疗阶段期间重复至少两次/天。
8.根据前述权利要求中任一项所述的美容方法,其中所述化妆品组合物在包装件中提供,所述包装件的尺寸设定成贮存足量的所述化妆品组合物以用于所述治疗阶段。
9.根据前述权利要求中任一项所述的美容方法,其中所述化妆品组合物作为洗剂或霜膏提供。
10.根据前述权利要求中任一项所述的美容方法,其中所述皮肤表面为面部皮肤表面。
