本发明涉及抗肥胖剂和抗肥胖食品,更详细地说,本发明涉及可以抑制脂质从消化管的摄入、预防肥胖的抗肥胖剂和抗肥胖食品。
背景技术
肥胖是以体脂肪过剩为特征的体重控制体系疾病。现代社会中,由于运动不足和卡路里过剩的饮食导致中性脂肪在体内蓄积并被判定为肥胖的人不断增加,成为很大的问题。
为了预防肥胖,首要的是进行对消耗所摄取的脂肪来说充分的运动,但实际上进行运动较为困难,需要减少脂肪的摄取。
但是,想通过过度的节食来抑制肥胖时,其它必需营养的摄取量不足或平衡被破坏,结果还会对身体造成不良影响。可以说这与摄取被称为减肥食品的营养少但可以带来饱涨感的食品的情况相同。
发明内容
发明要解决的问题
因此,强烈要求开发摄取正常饮食的同时可防止成为肥胖较大原因的脂质(三酰甘油)被吸收到体内的方法。
用于解决问题的方法
本发明人着眼于脂质被吸收到体内的机理,对于肥胖的抑制方法进行了研究,结果注意到,如果肠内的微生物群落(菌群)将脂质摄取分解,则人摄取的脂质减少,自然可以预防肥胖。于是,对于这样的可摄取分解脂质的微生物进行了深入研究,结果发现属于乳酸菌的微生物中具有这种作用的菌,进而完成了本发明。
即,本发明为抗肥胖剂,其以属于罗伊氏乳杆菌的微生物为有效成分,该微生物产生由以下(1)~(3)的氨基酸序列或该氨基酸序列中1个或多个氨基酸缺失、置换或增加的氨基酸序列所表示的脂肪酶。这里所说的1个或多个氨基酸缺失、置换或增加的氨基酸序列是指与这些原本的序列等价的序列、依然保持脂肪酶活性的序列。作为这种氨基酸序列可以举出显示80%以上同源性的序列。

发明效果
通过摄取本发明的抗肥胖剂和抗肥胖饮食品,可以进行正常饮食但防止脂质由肠道的吸收。而且,乳酸菌自古就用于酸奶或发酵乳等发酵食品的制造中,安全性极高,因此可以放心地摄取。

具体实施方式
作为本发明的抗肥胖剂和抗肥胖食品(以下有时称为“抗肥胖剂等”)的有效成分的微生物,属于乳酸菌的罗伊氏乳杆菌(Lactobacillus reuteri),是可以产生由上述(1)~(3)的氨基酸序列(分别对应于序列表的1、3、5)或在该氨基酸序列中1个或多个氨基酸缺失、置换或增加的氨基酸序列所表示的脂肪酶(以下称为“本发明脂肪酶类”)的乳酸菌(以下称为“本发明乳酸菌”)。
该本发明的脂肪酶类将存在于肠道粘膜细胞附近的脂质(三酰甘油)分解为脂肪酸和甘油,摄入到本发明乳酸菌中。而且,如图1所示,所摄入的脂肪酸作为菌体构成要素被利用,甘油经过各种代谢途径成为二氧化碳、丙酮、乙醇、乳酸盐或罗伊氏素(reuterin)。
本发明乳酸菌除了具有产生上述脂肪酶类的能力,即具有编码上述脂肪酶的核苷酸序列(基因)之外,优选具有以下(4)所示的核苷酸序列(对应于序列表的8)作为编码运载蛋白的基因。另外,本发明还包括与下述(4)所示核苷酸序列的同源性为80%以上、且能在严格条件下与编码具有甘油转运活性的蛋白质的核苷酸序列和下述(4)所示核苷酸序列杂交、且编码具有甘油转运活性的蛋白质的核苷酸序列。

该基因编码将甘油摄入到本发明乳酸菌的细胞内的运载蛋白,如图1所示,所摄入的甘油通过乳酸菌内的酶代谢。
进而,本发明乳酸菌优选具有由上述式(5)~(7)所示的氨基酸序列(分别对应于序列表的序列号9、11、13)或该氨基酸序列中1个或多个氨基酸缺失、置换或增加的氨基酸序列所表示的亚基所构成的甘油分解酶。该亚基(5)~(7)所构成的甘油分解酶为在pdu(propanediol utilization)操纵子中发挥作用的甘油脱水酶(glycerol dehydratase),可以高效地代谢通过本发明脂肪酶的作用所产生的甘油。
另外,可通过基因工程学方法将编码该亚基(5)~(7)的核苷酸序列(9)~(11)(分别对应于序列表的序列号10、12、14)整合到其它乳酸菌等中,由此获得甘油分解性高的乳酸菌。本发明还包括与下述(9)~(11)所示的核苷酸序列的同源性为80%以上、且在严格条件下与编码具有甘油分解活性的蛋白质的核苷酸序列和下述(9)~(11)所示的核苷酸序列杂交、且编码具有甘油分解活性的蛋白质的核苷酸序列。

该肠道黏附性蛋白质具有使乳酸菌黏附在肠道粘膜细胞附近的作用,具有该蛋白质的本发明乳酸菌在肠道粘膜附近存在一定期间,可以稳定地摄入脂质。另外,可通过公知的方法将编码该蛋白质的下述(12)所示的基因(对应于序列表的序列号16)整合到其它乳酸菌中,由此获得在肠道内滞留时间长的乳酸菌。另外,本发明还包括与下述(12)所示的核苷酸序列的同源性为80%以上、且在严格条件下与编码具有使乳酸菌黏附在肠道粘膜细胞附近的活性的蛋白质的核苷酸序列和下述(12)所示的核苷酸序列杂交、且编码具有使乳酸菌黏附在肠道粘膜细胞附近的活性的蛋白质的核苷酸序列。

本发明中,所谓的“严格条件”可以列举如下条件:例如在含有6×SSC(1×SSC的组成:0.15M NaCl、0.015M的柠檬酸钠、pH7.0)、0.5%SDS、5×Denhardt、100μg/mL的热变性鲑鱼精子DNA的溶液中,与探针一起在50~65℃的温度下保存一晚使其杂交。
进而,具有运载蛋白基因(4)的本发明乳酸菌,具有编码甘油分解酶亚基的基因(9)~(11)、编码甘油分解酶的基因(32)~(38)、编码醛脱氢酶的基因(39)~(41)、编码甘油酸激酶的基因(42)、编码甘油激酶的基因(43)、编码甘油-3-磷酸脱氢酶的基因(44)、编码磷酸丙糖异构酶的基因(45)~(47)或编码肠道黏附性蛋白质的基因(12)的本发明乳酸菌,也可以与上述同样地获得。
作为上述本发明乳酸菌的代表性菌,可以举出罗伊氏乳杆菌JCM1112T,其为理化学研究所的标准株。
本发明的抗肥胖剂可以通过将上述本发明乳酸菌制成可以口服且以活的状态到达肠道的活菌剂而调制。对于其剂型并无特别限定,例如可以为粉末剂、颗粒、片剂、胶囊剂等固体,凝胶、糊剂等半固体状,悬浮液,糖浆等液体状。这些各剂型可以通过制药领域中公知的方法制得。
配合在上述抗肥胖剂中的本发明乳酸菌可以应用公知的乳酸菌培养方法进行培养。作为该培养方法,可以根据常规方法将本发明乳酸菌进行液体培养,直接利用所得的培养物,或者通过离心分离等方法从该培养物中收集菌体后使用,还可以将培养物冷冻干燥制成粉末状后使用。
作为固体状的本发明抗肥胖剂的通常的制造方法,可以举出将本发明乳酸菌与水、淀粉、微晶纤维素、面粉、砂糖等载体一起配合,制成所需形态的方法。上述载体也是公知的,可以根据使用形态进行适当选择后使用。更具体地说,可以将通过常规方法培养获得的本发明乳酸菌的菌体制成冷冻干燥粉末,将其与砂糖混合,从而调制粉末剂。另外,还可以将本发明乳酸菌的菌体与适当的片剂用载体混合,根据常规方法进行压片,获得片剂。而且,还可以将本发明乳酸菌的湿菌体悬浮在糖浆中制成糖浆剂。调制本发明的抗肥胖剂时,还可以根据需要含有其它成分,例如其它微生物或有效成分、或者甜味料、香料、着色剂等。
如此获得的抗肥胖剂的给予量可以考虑对象的身体状况、例如健康状态、体重、年龄、病史、所使用的其它成分等适当地决定,一般来说,作为本发明乳酸菌的菌数,一个成人大致为108~109CFU/天左右。
使用本发明乳酸菌调制抗肥胖饮食品时,可以利用目前公知的乳酸菌的培养方法制成可以口服摄取的发酵食品。具体地说,可以制成酸奶等发酵乳、乳酸菌饮料、发酵香肠等食品,这些食品的制造可以通过使部分或全部所用乳酸菌采用本发明乳酸菌来进行。另外,还可以形成较多地含有本发明乳酸菌的形态,制成健康食品和功能性食品。在调制该抗肥胖食品时,当然并非仅含有本发明乳酸菌,还可以含有其它的乳酸菌,另外,还可以添加食品添加剂或调味品等。
本发明乳酸菌如后述实施例所示,具有显著的体重增加抑制效果(瘦身效果),认为其理由如下所示。
即,如图1所示,本发明乳酸菌通过3个脂肪酶(脂肪酶(1)~(3))的作用,将消化道内的脂肪成分分解为甘油和脂肪酸。然后,所分解的甘油被本发明乳酸菌所具有的运载蛋白(PduF;(4)的核苷酸编码)摄入到菌体内,进而被菌体内的甘油分解酶基因(PduCDE;由亚基(5)~(7)构成)代谢,产生罗伊氏素或转变为本菌自身的能量源。
另一方面,脂肪酸作为本菌的菌体成分被利用,而不会被吸收入生物体内。而且,(8)的肽具有本发明乳酸菌固定在人或哺乳动物肠道的功能,使本发明乳酸菌可以稳定地在肠道内存在一定期间。
如上所述,本发明乳酸菌由于稳定地存在于肠道内、积极地分解脂肪、且利用或进一步分解其代谢物,因此可以抑制脂质从肠道被吸收入体内、即便摄取通常的饮食也可以预防肥胖、维持及提高瘦身效果。
另外,本发明的一个方式包括本发明乳酸菌用于预防或治疗肥胖症的应用,还包括本发明乳酸菌用于制造抗肥胖剂的应用。
进而,本发明的又一个方式包括以向肥胖症患者给予本发明乳酸菌为特征的肥胖症治疗方法,还包括以给予本发明抗肥胖剂为特征的肥胖症治疗方法。
实施例
以下举出实施例,更加详细地说明本发明,但本发明并不局限于这些实施例。
实施例1
罗伊氏乳杆菌JCM1112T的抗肥胖效果试验:
通过下述材料和方法研究罗伊氏乳杆菌JCM1112T的抗肥胖效果。
使用材料和试验方法:
(1)使用的实验动物
使用体重约180~200g的SPF级Wistar系大鼠(8周龄-雄;日本SLC株式会社),从放入实验室开始设置7天的驯化期,开始实验。饲养环境设定为温度22±1℃、湿度55±5%、照明时间12小时(8点~20点),大鼠为1只/笼,利用给水瓶使其自由摄取灭菌蒸馏水、使用给饵器使其自由摄取大鼠用放射线灭菌固体样品*(CE-2,CLEA Japan Inc.)。所使用的饲养器具均使用通过高压蒸汽灭菌器灭菌的器具。

作为待测菌,使用罗伊氏乳杆菌JCM1112T(理化学研究所的标准菌、从该所购买)和鼠李糖乳杆菌(L.rhamnosus)ATCC53103(GG株)。这些待测菌接种于MRS液体培养基(Oxid),在37℃下培养一晚,制成预培养液。将该预培养液添加到新鲜的MRS液体培养基中达到1%量,在37℃下培养18小时,将其作为待测菌液。
(3)待测菌液的给予
将上述实验动物分为各待测菌给予组和对照组(1组各5只),对待测菌给予组,将上述待测菌液通过口服给予。待测菌的给予使用探针强制进行。给予菌液在使用时调制,给予菌量为每只大鼠109CFU。另外,对对照组给予相同容量的PBS。
(4)体重测定和一般症状的观察
使用体重计每天测定实验动物的体重。每天均观察一般症状。对每个个体记录症状,用表现个体数来表示观察到显著变化的症状项目。
(5)结果
图2示意出分别给予上述乳杆菌属细菌的大鼠组和未给予的大鼠组的体重随时间增加的状况。由该图可知,与未给予组相比,罗伊氏乳杆菌JCM1112T给予组显著抑制了体重的增加,其程度大于鼠李糖乳杆菌ATCC53103给予组。
另外,实验动物的健康状态良好是由良好的体重增加的进展来显示的同时,由各组大鼠的诊察来确认的。
实施例2
罗伊氏乳杆菌JCM1112T的基因组解析:
使用下述药品,通过以下方法从罗伊氏乳杆菌JCM1112T中获取DNA,进行基因组解析。
DNA获取方法:
(药品)
核裂解液(Wizard基因组DNA纯化试剂盒;Promega公司生产)
生理盐水
50mM EDTA(pH7.0)
50mg/ml溶菌酶溶液(在使用当日,用TE缓冲液、0.25MTris-HCl(pH8.0)或10mM Tris-HCl(pH8.0)/10mMEDTA/0.5%SDS溶解规定量的溶菌酶进行调制)
2mg/ml EDTA
苯酚·氯仿·异戊醇(25∶24∶1)混合液(以下简称为“PCI”)
氯仿·异戊醇(24∶1)混合液(以下简称为“CIA”)
99%乙醇
70%乙醇
TE缓冲液
(方法)
1.在静置条件、37℃下培养罗伊氏乳杆菌JCM1112T 24小时,在3500rpm下离心分离所得培养液(50ml)15分钟后,利用生理盐水悬浮。
2.在3500rpm下离心分离上述1所得的悬浮液15分钟,舍去上清,利用5ml 50mM EDTA悬浮。
3.在上述2所得的悬浮液中加入200μl 50mg/ml溶菌酶溶液,在37℃下培养60分钟。(此时使用空气培养箱时,培养时间为2~3小时)
4.培养后,在3500rpm下离心分离15分钟,舍去上清。
5.在4所得的沉淀物中加入核裂解液5ml,在80℃下培养10分钟。
6.加入1μl 2mg/ml RNase A,在37℃下培养45分钟。
7.加入10ml PCI并混合,在3500rpm下离心分离15分钟。
8.将上清移至新的试管中,再加入10ml PCI并混合。
9.共增加3次同样的操作。(直至变得不能识别蛋白质层)
10.加入10ml CIA,缓缓混合后,在3500rpm下离心分离15分钟。(除去苯酚)
11.将上清移至新的试管中,进行乙醇沉淀。
12.将11所得的沉淀物溶解在1ml TE缓冲液中,获得乳酸菌DNA。
基因组解析方法:
对于如上获得的DNA进行结构基因预测和解析。联合使用GENOMEGAMBLER(Sakiyama,T.Takami,H.Ogasawara,N.Kuhara.S.Kozuki,T.Doga,K.Ohyama,A.Horikoshi,K.Anauromated system for genome analysis to support microbial whole-genome shotgun sequencing.Biosci.Biotechnol.Bichem.64:670-673.2000.)、GLIMMER 2.0(Salzberg,SL.Delcher,AL.Kasif,S.and White,O.Microbial gene identification usinginterpolated Markov models.Nucleic.Acid.Res.26:544-548.1998.)和BLAST program blastp(Altschul,SF.Gish,W.Miller,W.Myers,EW.and Lipman,DJ.Basic local alignment searchtool.J.Mol.Biol.215:403-410.1990)的结果来进行该结构基因预测等。

根据以上信息可知,(13)~(15)所示的基因分别为编码脂肪酶的基因、(4)所示的基因为编码运载蛋白的基因、(9)、(10)和(11)所示的基因为编码甘油分解酶的基因、(12)所示的基因为黏附性基因。另外可知,(32)~(38)所示的基因为编码甘油分解酶的基因、(39)~(41)所示的基因为编码醛脱氢酶的基因、基因(42)所示的基因为编码甘油酸激酶的基因、(43)所示的基因为编码甘油激酶的基因、(44)所示的基因为编码甘油-3-磷酸脱氢酶的基因、(45)~(47)所示基因为编码磷酸丙糖异构酶的基因。

甘油分解基因的扩增:
将实施例2中精制的DNA作为模板,将以下核苷酸序列作为引物,使用下述反应液进行PCR。PCR条件也示于下。

产业实用性
由使用了作为本发明乳酸菌代表的罗伊氏乳杆菌JCM1112T的上述实施例的结果可知,通过该微生物的给予,可以抑制体重的增加,而不损害实验动物的健康、尤其是对营养摄取没有限制。
因此,仅摄取利用了本发明乳酸菌的抗肥胖剂和抗肥胖饮食品,不需要特殊治疗、处置即可预防肥胖、获得瘦身效果。
通过利用公知的方法将编码由本发明乳酸菌发现的甘油分解酶亚基的基因和编码肠道黏附性蛋白质的基因整合在其它乳酸菌中,可以获得甘油分解性高的乳酸菌和在肠道内的停留时间长的乳酸菌,可以有效地用于其它有用的乳酸菌改性中。
