Description
Verfahren zur Herstellung von Vitamin-A-Estern Es wurde gefunden, dass die bisher unbe- kannten Vitamin-A-Ester sowohl des natür- lichen eis-als auch des durch Umlagerung er hältlichen, stabilen All-trans-Bixins wertvolle Eigenschaften besitzen. Sie eignen sich zum gleichzeitigen Färben und Vitaminieren von Nahrungsmitteln. Gegenstand der Erfindung ist ein Verfahren zur Herstellnng von Vitamin-A-Estern, welches dadurch gekennzeiehnet ist, dass man ein Bixin mit Vitamin A verestert. Die Ver esterung erfolgt zum Beispiel in der Weise, dass die Bixine mit Oxalylehlorid in die Säure- chloride umgewandelt und letztere in an sich bekannter Weise mit Vitamin A umgesetzt werden. Die neuen Vitamin-A-Ester zeichnen sich im Vergleich zu andern Carotinfarbstoffen durch Loslichkeit in Ölen und Fetten aus. Die Färbungen, mit welchen gleichzeitig eine Vitaminierung erzielt wird, sind stabil. Beispiel 1 40 Gewichtsteile All-trans-Bixin werden in 1000 Raumteilen wasserfreiem Benzol mit 40 Raumteilen Oxalylchlorid während einer Stunde unter Rückflu13 gekocht. Nun werden Benzol und überschüssiges Oxalylchlorid im Vakuum eingedampft. Den verbleibenden, violett gefärbten Rückstand lost man in 1000 Raumteilen Benzol, versetzt unter gutem Kühlen mit einer Lösung von 40 Gewichtsteilen Vitamin A in 1000 Raumteilen Benzol und 60 Raumteilen Pyridin. Nach 3stündigem Stehen wird kurz aufgekocht und auf Zimmertemperatur abgekühlt. Man versetzt mit 2000 Raumteilen Äther und wäscht die tiefrot ge färbte Losung zweimal mit je 3000 Raum- teilen 0,5n-Schwefelsäure aus. Die gewaschene Lösung wird mit Natriumsulfat getrocknet und unter Verwendung von Aluminiumoxyd alsAdsorptionsmittel chromatographiert. Hier- bei verbleiben kleine Mengen nicht umgesetzten All-trans-Bixins in der obersten Alumi niumoxydschieht, während das im Überschuss zugesetzte Vitamin A zuerst in das Filtrat übergeht. Die Aluminiumoxydsäule wird mit Äther gewaschen und die vereinigten, tiefrot gefärbten Lösungen im Vakuum auf 600 Raumteile konzentriert. Man versetzt mit 1000 Raumteilen Äthylalkohol und konzern- triert erneut auf 500 Raumteile. Schon während des Eindampfens kristallisiert der neue Vitamin-A ; Ester aus. Durch Stehen in der Kälte wird die Ausscheidung vervollständigt. Die Ausbeute an rohem Ester beträgt 60 Ge- wichtsteile, entsprechend 90i 0/o der Theorie. Durch Umkristallisieren aus Petroläther erhält man den Vitamin-A-Ester des All-trans Bixins in feinen, tiefroten Rosette, die bei 140 C schmelzen. Im'Gitterspektrbskop zeigt der Ester, in Schwefelkohlenstoff gelöst, folgende Absorptionsbanden : 527, 492, 457 m . Die spektrophotometrische Aufnahme zeigt bei 326 m die für Vitamin A charakteristische Absorptionsbande. Beispiel 2 50 Gewichtsteile cis-Bixin werden in 1600 Raumteilen Benzol mit 50 Raumteilen Oxalyl chlorid während einer Stunde unter Rückfluss gekocht. Nun werden Benzol und übersehüssi- ges Oxalylehlorid imValiuum abgedampft. Der Rückstand wird in 1500 Raumteilen Benzol gelost und unter gutem Kühlen mit einer Lösung von 50 Gewichtsteilen Vitamin A in 500 Raumteilen Benzol + 75 Raumteilen Pyridin versetzt. Man lässt über Nacht stehen, kocht kurz auf und kühlt wieder auf Zimmertemperatur ab. Man versetzt mit 2000 Raumteilen Äther und wäscht die tiefrot gefärbte Lösung zweimal mit je 3000 Baumteilen 0, 5n-Schwe- felsäure aus. Die gewaschene Losung wird mit Natriumsulfat getrocknet und unter Verwen- dung von Aluminiumoxyd als Adsorptionsmittel chromatographiert. Kleine Mengen nicht umgesetzten Bixins verbleiben in der obersten Aluminiumoxydschicht, während überschüssiges Vitamin A zuerst ins Filtrat übergeht. Die Aluminiumoxydsäule wird mit Äther gewaschen und die vereinigten roten Losungen im Vakuum auf'200 Baumteile konzentriert. Man versetzt mit 200 Raumteilen Äther und giesst die Lösung unter Rühren in 3000 Raumteile tiefsiedenden Petroläther. Man lässt bei-10 C stehen, wobei die an fänglich schmierige Fällung fest wird. Die Mutterlaugen werden auf 700 Raunteile kon- zentriert und über Nacht bei tiefer Temperatur stehengelassen, wobei sich eine zweite Fraktion des Vitamin-A-Esters abscheidet. Die Ausbeute an rohem Ester beträgt 70 Ge wichtsteile, entsprechend 85 /o der Theorie. Durch Umkristallisieren aus Petroläther erhält man den Vitamin-A-Ester des cis-Bixins in Form glänzender braunroter Plättchen, die bei 124 C schmelzen. Im Gitterspektroskop zeigt der Ester, in Schwefelkohlenstoff gelöst, folgende Absorptionsbanden : 521, 487, 456 m . Die spektrophotometrische Aufnahme zeigt bei 326 my die für Vitamin A charakteristische Absorptionsbande.

- PATENTANSPRUCH Verfahren zur Herstellung von Vitamin A-Estern, dadurch gekennzeichnet, dass man ein Bixin mit Vitamin A verestert.UNTERANSPBUCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass Vitamin A mit dem Säurechlorid eines Bixins umgesetzt wird.2. Verfahren nach Patentanspruch und Unteransprueh l, dadurch gekennzeichnet, dass man als Ausgangsmaterial das Säure- chlorid des cis-Bixins verwendet.3. Verfahren nach Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass man als Ausgangsmaterial das Säure- chlorid des All-trans-Bixins verwendet.
