Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates. Gegenstand des vorliegenden Patentes, ist ein Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates, das dadurch ge kennzeichnet ist, dass man ein 2-Benzolsulfon- amido-1,3,5-triazin, das in p-Stellung einen durch Hydrolyse in die Aminogruppe über führbaren Substituenten aufweist, mit einem hydrolysierenden Mittel behandelt.
Das so erhaltene 2-(p-Amino-benzolsul- fonamido)-1,3,5-triazin ist ein kristallines Pulver. Die neue Verbindung soll als Arznei mittel sowie als Zwischenprodukt Verwen dung finden.
Das 2-Benzolsulfonamido-1;3,5-triazin, das in p-Stellung zur Sulfonamidgruppe einen durch Hydrolyse in die Aminogruppe über führbaren Substituenten enthält, kann auf verschiedene Art und Weise gewonnen wer den.
Besonders geeignet ist die Umsetzung der entsprechenden reaktionsfähigen Benzol- sulfon3äurederivate, insbesondere der Benzol- sulfonsäurehalogenide mit 1,3,5-Triazinver- bindungen, die in 2-Stellung eine Gruppe ent- halten, die mit dem Benzolsudfonsäurederivat ein 2-Benzolsulfonamido-1,3,5-triazin zu bil- i den vermag, wie mit 2-Amino-1,3"5-triazin. Man kann auch entsprechende Sulfonamide der Formel R - SOZNHY,
in der Y einen bei der nachfolgenden Reaktion sich abspalten den Rest bedeutet, mit 2'-Halogen-1,3,5-tri- ; azinen umsetzen. Auch können andere dem Fachmann geläufige Ilerstellungsmethoden benutzt werden.
<I>Beispiel 1:</I> 29,2 g 2-(p-Acetylamino-benzolsulfon-; amido)-1,3,5-triazin, das durch Kondensation von p - Acetylamino - benzolsulfoehlori.d mit 2-Amino-1,3,5-triazin erhalten wurde, werden mit einer Lösung von 12 g Natriumhydrogyd in 150 em3 Wasser zwei Stunden unter Rück fluss gekocht. Die Lösung wird gegebenen falls unter Zusatz von Tierkohle filtriert und mit konz. Salzsäure neutralisiert. Dabei fällt das 2-(p-Amino-benzolsulfonamido)-1;3"5-tri- azin als kristallines Pulver aus.
Die Verbin- dun, kann durch Umkristallisation aus ver dünnter Essigsäure gereinigt werden.
<I>Beispiel 2:</I> 32,2 g 2 - (p- Ca.rbäthoxyamino-benzolsul- fonamido)-1,3,5-triazin, das durch Konden sation von p-Carbäthoxyamino-benzolsulfo- chlorid mit 2-Amino-1,3,5-triazin erhalten wurde, werden, wie im Beispiel 1, mit einer wässerigen Lösung von 12 ,
Natriumhydro- xyd in 150 ein' Wasser unter Kochen ver- s <B>x</B> eift. Nach der Neutralisation mit Salzsäure wird das gebildete 2-(p-Aminobenzolsulfon- amido)-1,3,5-tria.zin isoliert und durch Um- kristallisieren gereinigt.
Die Hydrolyse kann auch mit andern alkalischen Mitteln, beispielsweise Erdalkali- hydroxyden, wie Caleiumhydroxyd, oder mit sauren Mitteln, z. B. Salzsäure in Gegenwart von Wasser oder organischen Lösungsmitteln, wie Alkohol, durchgeführt werden.
Das gebildete p-Aminobenzolsulfona.mid- derivat lässt sich ferner in Form seiner Salze, z. B. des Natriums oder des Calciums, iso lieren.
