Abstract
"PROTEìNA, DNA, E, MéTODO PARA A PRODUçãO DE L-AMINOACIDO". Uma pluralidade de grupos iniciadores são designados com base em uma região onde a conservação no nível de aminoácido é observada entre vários microorganismos para as seq³ências de gene conhecidas correspondendo a um gene que codifica uma enzima da via biossintética de L-aminoácido derivado de Corynebacterium thermoaminogenes, preferivelmente uma que funciona em uma temperatura mais elevada comparada com aquela do Corynebacterium glutamicum. O PCR é executado mediante o uso dos iniciadores e DNA cromossómico de Corynebacterium thermoaminogenes como um modelo. Os iniciadores com os quais um fragmento de amplificação foi obtido são usados como iniciadores de peneiramento para selecionar um clone contendo um fragmento de DNA alvo a partir de uma livraria de plasmídeo de DNA cromossómico de Corynebacterium thermoaminogenes.
