Abstract

"MéTODOS DE PRODUZIR UM PANTO-COMPOSTO, PANTOTENATO E BETA-ALANINA, DE AUMENTAR A PRODUçãO DE UM PANTO-COMPOSTO E DE IDENTIFICAR COMPOSTOS QUE MODULAM A ATIVIDADE DA PANTOTENATO CINASE, MICROORGANISMO RECOMBINANTE, VETOR, MOLéCULA DE áCIDO NUCLEICO ISOLADA, POLIPETìDEO E PROTEìNA DA PANTOTENATO CINASE ISOLADA". A presente invenção retrata métodos de produzir panto-compostos (por exemplo, pantotenato) usando microorganismos em que o caminho biossintétíco do pantotenato e/ou o caminho biossintético da isoleucina-valina e/ou o caminho biossintético da coenzimaA tenha sido manipulado. Métodos que retratam microorganismos que supraexpressam a cetopantoato reductase bem como microorganismos que supraexpressam a aspartato <244>-descarboxilase são fornecidos. Métodos de produzir panto-compostos em uma maneira independente de precursor e em alto rendimento são descritos. Microorganismos recombinantes, vetores, moléculas de ácido nucleico isoladas, genes e produtos de gene úteis na prática das metodologias acima também são fornecidos. A presente invenção também retrata um gene da pantotenato cinase microbiana não identificado anteriormente, coaX, bem como métodos de produzir panto-compostos que utilizam microorganismos tendo atividade modificada da pantotenato cinase. Microorganismos recombinantes, vetores, moléculas de ácido nucleico coaX isoladas e proteínas CoaX purificadas são retratados. Também são retratados métodos para identificar os moduladores da pantotenato cinase que utilizam os microorganismos recombinantes e/ou as proteínas CoaX purificadas da presente invenção.
