본 발명은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중의 예방 또는 치료용 약학 조성물, 식품 조성물 및 사료 조성물, 상기 약학 조성물을 투여하는 단계를 포함하는 뇌졸중의 예방 또는 치료방법 및 상기 약학 조성물의 제조방법에 관한 것이다.
Description
본 발명은 뇌졸중의 예방 및 치료용 조성물에 관한 것으로, 구체적으로 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중의 예방 또는 치료용 약학 조성물, 식품 조성물 및 사료 조성물, 상기 약학 조성물을 투여하는 단계를 포함하는 뇌졸중의 예방 또는 치료방법 및 상기 약학 조성물의 제조방법에 관한 것이다.
뇌졸중은 뇌혈관 질환으로서, 심장 질환 및 암과 함께 3가지 주요한 사망 원인이며, 빈발수가 점진적으로 증가됨에 따라 평균 수명 전망 및 인구의 나이를 연장에 영향을 주는 가장 일반적인 질환 중 하나이다. 고혈압, 고지방혈증, 죽상동맥경화증, 고콜레스테롤혈증은 뇌혈성 뇌졸중에 대한 다양한 중요한 위험인자들로 알려져있다.
뇌졸중은 크게 2가지로 나누는데, 뇌조직으로 가는 혈액 공급의 감소 혹은 차단으로 뇌조직의 허혈상태로 발생하는 허혈성 뇌졸중(ischemic stroke)과 혈관이 터져 뇌조직으로 출혈을 일으키는 출혈성 뇌졸중(hemorrhagic stroke)으로 구분하고 있으며, 허혈성 뇌졸중이 전체 뇌졸중 환자의 약 80% 이상을 차지하고 있다. 이러한 뇌졸중으로 인한 사망률은 국내뿐만 아니라 세계적으로 매년 증가하고 있는 추세이며, 오늘날 뇌졸중 치료 약물 개발에 많은 관심이 모이고 있다. 예를 들어, 천연물 추출물 또는 천연물에서 추출한 화합물을 유효성분으로 함유하는 뇌졸중의 예방 또는 치료용 조성물 개발과 같은 천연물을 활용하는 접근 방법이 주목받고 있다(KR 10-1182199 B2).
뇌졸중이나 신경변성 질환 등의 난치성 신경질환은 공통으로 뇌세포 또는 신경세포가 사멸하게 되는데, 그 결과 비가역적인 고차 신경기능장애를 가져온다. 현재 세계적으로 여러 종류의 화합물, 줄기세포 등을 이용한 치료를 동물 모델에서 시도하고 있으나, 임상시험에 적용되기엔 많은 문제가 수반되는 상황이다. 따라서, 뇌세포 또는 신경세포의 사멸 전에 이들 세포를 유효하게 보호할 수 있는 방안의 고려가 요구되고 있다.
이러한 배경하에, 본 발명자들은 본 발명의 최적의 조성비로 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 조성물이 신경 세포 보호 효과를 가지고, 뇌졸중으로 인한 행동 장애를 개선하며, 뇌졸중으로 인한 뇌 조직의 손상을 감소시키는 효과를 나타냄을 확인하여, 뇌졸중의 예방 및 치료에 유용하게 활용될 수 있음을 확인함으로써 본 발명을 완성하였다.
본 발명자들은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물의 우수한 뇌졸중 예방 또는 치료 효과를 최초로 규명하고, 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중 예방 또는 치료용 약학 조성물; 상기 약학 조성물을 이용한 뇌졸중의 예방 또는 치료 방법; 및 상기 약학 조성물의 제조방법을 개발하여 본 발명을 완성하였다.
본 발명의 하나의 목적은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중 예방 또는 치료용 약학 조성물을 제공하는 것이다.
본 발명의 다른 목적은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중 예방 또는 개선용 식품 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중 예방 또는 개선용 사료 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 약학 조성물을 투여하는 단계를 포함하는 뇌졸중 예방 또는 치료 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 약학 조성물의 제조방법을 제공하는 것이다.
본 발명의 또 다른 하나의 목적은 상기 뇌졸중 예방 또는 치료용 약학 조성물을 제조하기 위한, 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 포함하는 조성물의 용도를 제공한다.
본 발명의 또 다른 하나의 목적은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 포함하는 조성물의 뇌졸중 예방 또는 치료 용도를 제공한다.
본 발명의 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물은 우수한 뇌졸중 예방 또는 치료 효과를 나타내므로, 약학 조성물 또는 식품에 유효성분으로 포함되어 뇌졸중의 예방제, 치료제 또는 개선제 등의 개발에 이용될 수 있다.

이를 구체적으로 설명하면 다음과 같다. 본 발명에서 개시된 각각의 설명 및 실시형태는 각각의 다른 설명 및 실시 형태에도 적용될 수 있다. 즉, 본 발명에서 개시된 다양한 요소들의 모든 조합이 본 발명의 범주에 속한다. 또한, 하기 기술된 구체적인 서술에 의하여 본 발명의 범주가 제한된다고 볼 수 없다.
상기 목적을 달성하기 위한 본 발명의 하나의 양태는 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중 예방 또는 치료용 약학 조성물을 제공하는 것이다.
본 발명에서 용어, "구기자(Lycium chinense)"는 구기자나무의 열매를 의미한다. 구기자는 보통 달걀모양이나 긴 타원형의 형태를 가지고 있다. 상기 구기자는 강장제, 해열제로 쓰이며 간기능 보호 작용이 뛰어난 것으로 알려져 있다.
본 발명에서 용어, "숙지황(Rehmannia glutinosa)"은 지황의 뿌리를 쪄서 말린 한약재이다. 지황은 현삼과에 딸린 여러해살이 약용식물로 그 뿌리가 한방에서 약재로 이용된다.
본 발명에서 용어, "산수유(Cornus officinalis)"는 산수유나무의 열매를 말한다.
본 발명에서 용어, "산약(Dioscorea rhizome)"은, 마과(科: Dioscoreaceae)에 속한 디오스코레아 바타타스(Dioscorea batatas(=Dioscorea opposita)), 디오스코레아 오포시타(Dioscorea opposite), 디오스코레아 자포니카(Dioscorea japonica), 디오스코레아 테누이페스(Dioscorea tenuipes)의 뿌리 줄기를 말한다.
본 발명에서 용어, "복령(Poria cocos)"은 소나무 뿌리에 외생균근이 공생하여 생긴 것을 말한다. 상기 복령은 구체적으로 소나무 뿌리에 기생하는 잔나비걸상과의 복령(Poria cocos Wolf)의 균핵으로, 바깥 층을 거의 제거하여 사용될 수 있다.
본 발명에서 용어, "택사(Alisma rhizome)"는 질경이 택사(Alisma orientale Juzepczuk) 또는 기타 동속 근연식물(Alismataceae)의 덩이 뿌리줄기를 말한다.
본 발명에서 용어, "목단피(Paeonia suffruticosa)"는 작약과의 모란의 뿌리껍질로 만든 약재를 말한다. 상기 목단피는 진통, 진정, 해열, 항경련작용, 항염증작용, 혈전형성 억제 등의 약리작용을 가지는 것으로 알려져 있다.
본 발명의 용어, “추출물”은 어떤 물질을 추출 처리하여 얻어지는 물질을 의미하는 것으로, 구체적으로 상기 본 발명의 추출처리에 의하여 얻어지는 추출액, 상기 추출액의 희석액이나, 농축액, 상기 추출액을 건조하여 얻어지는 건조물, 상기 추출액의 조정제물이나 정제물 또는 이들의 혼합물 등, 추출액 자체 및 추출액을 이용하여 형성 가능한 모든 제형의 추출물을 포함한다.
상기 추출물은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피로 이루어진 군에서 선택된 둘 이상의 복합 추출물 일 수 있으나, 이에 제한되는 것은 아니다. 보다 구체적으로는, 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물일 수 있다.
상기 추출물을 추출하는 방법은 특별히 제한되지 않으며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 추출할 수 있다. 상기 추출물 추출 방법의 비제한적인 예로는, 용매 추출법, 초음파 추출법, 여과법, 환류 추출법 등을 들 수 있으며, 이들은 단독으로 수행되거나 2종 이상의 방법을 병용하여 수행될 수 있다.
본 발명에서 상기 추출물에 사용되는 추출용매의 종류는 특별히 제한되지 않으며, 당해 기술 분야에서 공지된 임의의 용매를 사용할 수 있다. 상기 추출용매의 비제한적인 예로는 물, 탄소수 1 내지 4의 알코올 또는 이들의 혼합 용매 등을 들 수 있으며, 이들은 단독으로 사용되거나 1종 이상 혼합하여 사용될 수 있다. 구체적으로, 상기 추출용매는 열수일 수 있다.
본 발명의 용어, “분획물”은 여러 다양한 구성 성분들을 포함하는 혼합물로부터 특정 성분 또는 특정 성분 그룹을 분리하기 위하여 분획을 수행하여 얻어진 결과물을 의미한다.
본 발명에서 상기 분획물을 얻는 분획 방법은 뇌졸중을 억제하는 효과를 나타낼 수 있는 한 특별히 이에 제한되지 않으나, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 수행될 수 있다. 예를 들어, 다양한 용매를 처리하여 수행하는 용매 분획법, 일정한 분자량 컷-오프 값을 갖는 한외 여과막을 통과시켜 수행하는 한외여과 분획법, 다양한 크로마토그래피(크기, 전하, 소수성 또는 친화성에 따른 분리를 위해 제작된 것)를 수행하는 크로마토그래피 분획법 및 이의 조합 등이 될 수 있다.
본 발명에서 상기 분획물을 얻는 데에 사용되는 분획 용매의 종류는 특별히 제한되지 아니하며, 당해 기술 분야에서 공지된 임의의 용매를 사용할 수 있다. 상기 분획 용매의 비제한적인 예로는 물, 증류수, 알코올 등의 극성 용매; 헥산(Hexan), 에틸 아세테이트(Ethyl acetate), 클로로포름(Chloroform), 디클로로메탄(Dichloromethane) 등의 비극성 용매 등을 들 수 있다. 이들은 단독으로 사용되거나 2 종 이상 혼합하여 사용될 수 있다. 상기 분획 용매 중 알코올을 사용하는 경우에는 구체적으로는 C1 내지 C4의 알코올을 사용할 수 있다.
본 발명에서 “뇌졸중(stroke)”이란, 뇌혈류 이상으로 인해 갑작스레 유발된 국소적인 신경학적 결손 증상을 의미한다. 이러한 뇌졸중에는 혈전 등으로 뇌혈관이 막혀 뇌세포의 사멸이 유도되는 뇌경색, 또는 뇌혈관이 터져서 발생하는 뇌출혈이 포함될 수 있다. 구체적인 뇌졸중의 증세로서는 갑작스러운 두통 및 구토가 있고, 반신 마비나 신체 일부의 마비, 신체 일부의 감각마비와 소실이 있을 수 있으며, 언어장애(실어증 혹은 발음장애), 안면신경장애, 운동실조 등이 나타날 수 있다. 본 발명에서 뇌졸중은 중풍을 의미하는 것일 수 있다.
본 발명에서 상기 뇌졸중은 구체적으로는 허혈성 뇌졸중인 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 “허혈성 뇌졸중”이란 뇌혈관의 폐색으로 인해 뇌혈류가 감소되어 뇌조직이 기능을 하지 못하는 상태로, 뇌경색과 일과성 허혈성 발작 모두를 통틀어서 일컫는 용어이다.
본 발명의 상기 약학 조성물은 뇌졸중으로 인해 손상된 운동 기능 회복을 촉진하거나, 또는 경색 크기를 감소시키는 것일 수 있다.
본 발명의 구체적인 일 실시예에서는, 본 발명의 조성물을 처리한 신경세포배양에서 신경세포 독성 물질로 인한 신경세포 손상이 대조군에 비하여 감소됨을 확인함으로써, 뇌졸중 예방에 효과를 보임을 확인하였다(도 1 및 도 2).
또한, 본 발명의 구체적인 일 실시예에서는, 본 발명의 조성물을 처리한 신경세포배양에서 신경세포 독성 물질로 인한 신경세포의 세포자멸사 (apoptosis)가 대조군에 비하여 감소됨을 확인함으로써, 뇌졸중 예방에 효과를 보임을 확인하였다 (도 3 및 도 4).
상기 결과를 통해, 본 발명의 조성물이 뇌졸중의 예방 또는 치료 용도로 우수한 효과가 있음을 확인하였다.
본 발명의 용어, “예방”은 본 발명에 따른 약학 조성물의 투여에 의해 뇌졸중의 성장을 억제시키거나 또는 지연시키는 모든 행위를 의미한다.
본 발명의 용어, “치료”는 상기 약학 조성물의 투여에 의해 뇌졸중의 의심 및 발병 개체의 증상이 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명의 약학 조성물에 포함되는 상기 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물은 구기자 1 중량 대비하여, 숙지황 0.7 내지 1.3, 산수유 0.2 내지 0.8, 산약 0.2 내지 0.8, 복령 0.01 내지 0.55, 택사 0.01 내지 0.55 및 목단피 0.01 내지 0.55의 중량비로 혼합된 것일 수 있고, 구체적으로 구기자 1 중량 대비하여, 숙지황 0.9 내지 1.1, 산수유 0.4 내지 0.6, 산약 0.4 내지 0.6, 복령 0.15 내지 0.35, 택사 0.15 내지 0.35 및 목단피 0.15 내지 0.35의 중량비로 혼합된 것일 수 있고, 보다 구체적으로 구기자 1 중량 대비하여, 숙지황 0.95 내지 1.05, 산수유 0.45 내지 0.55, 산약 0.45 내지 0.55, 복령 0.2 내지 0.3, 택사 0.2 내지 0.3 및 목단피 0.2 내지 0.3의 중량비로 혼합된 것일 수 있다.
예를 들어, 본 발명의 약학 조성물에 포함되는 상기 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물은 구기자 1 중량 대비하여, 숙지황 1, 산수유 0.5, 산약 0.5, 복령 0.25, 택사 0.25 및 목단피 0.25의 중량비로 혼합된 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 약학 조성물에 포함된 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물의 함량은 특별히 이에 제한되지 않으나, 최종 조성물 총 중량을 기준으로 0.0001 내지 80 중량%, 0.0001 내지 50 중량%, 보다 구체적으로 0.01 내지 20 중량%의 함량으로 포함될 수 있다.
본 발명의 약학 조성물은, 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 추가로 포함할 수 있는데, 상기 담체는 비자연적 담체(non-naturally occuring carrier)를 포함할 수 있다.
보다 구체적으로, 상기 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트, 폴리카프로락톤(polycaprolactone), 폴리락틱액시드(Poly Lactic Acid), 폴리-L-락틱액시드(poly-L-lactic acid), 광물유 등을 들 수 있다.
본 발명의 조성물은 약학적으로 허용가능한 담체, 희석제 또는 부형제를 추가로 포함할 수 있으며, 각각의 사용 목적에 맞게 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁제, 에멀젼, 시럽, 에어로졸 등의 경구 제형, 멸균 주사 용액의 주사제 등 다양한 형태로 제형화하여 사용할 수 있으며, 경구 투여하거나 정맥내, 복강내, 피하, 직장, 국소 투여 등을 포함한 다양한 경로를 통해 투여될 수 있다. 이러한 조성물에 포함될 수 있는 적합한 담체, 부형제 또는 희석제의 예로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸셀룰로즈, 미정질셀룰로스, 폴리비닐피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유 등을 들 수 있다. 또한, 본 발명의 조성물은 충전제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
경구 투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 상기 조성물에 적어도 하나 이상의 부형제, 예를 들면 전분, 탄산칼슘, 수크로스, 락토즈, 젤라틴 등을 혼합하여 제형화한다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크와 같은 윤활제가 사용될 수 있다.
경구용 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 예시될 수 있으며, 흔히 사용되는 단순 희석제인 물, 액체 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액제, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜, 폴리에틸렌글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔, 마크로골, 트윈61. 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다. 한편, 주사제에는 용해제, 등장화제, 현탁화제, 유화제, 안정화제, 방부제 등과 같은 종래의 첨가제가 포함될 수 있다.
본 발명의 또 다른 실시 양태는 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중 예방 또는 개선용 식품 조성물을 제공하는 것이다.
이때, 상기 용어 “구기자”, “숙지황”, “산수유”, “산약”, “복령”, “택사”, “목단피”, “추출물”, “분획물” 및 “예방”의 정의는 상기에서 서술한 바와 같다.
본 발명의 용어, "개선"은 상기 조성물의 투여로 치료되는 상태와 관련된 파라미터, 예를 들면 증상의 정도를 적어도 감소시키는 모든 행위를 의미한다.
본 발명에 따른 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물은 우수한 뇌졸중 억제 효과를 나타내므로, 뇌졸중의 예방 또는 개선을 목적으로 식품 조성물에 포함될 수 있으며, 상기 식품 조성물은 일상적으로 섭취하는 것이 가능하기 때문에 뇌졸중의 예방 또는 개선에 대하여 높은 효과를 기대할 수 있다.
본 발명에서 용어, "식품"은 육류, 소시지, 빵, 초콜릿, 캔디류, 스낵류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올음료, 비타민 복합제, 건강기능식품 등의 통상적인 의미에서의 식품을 모두 포함하며, 본 발명의 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 포함할 수 있는 한, 이에 제한되지 않는다.
본 발명의 용어, "건강기능식품"은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, '기능성'은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻는 것을 의미한다. 한편, 건강식품은 일반식품에 비해 적극적인 건강유지나 증진 효과를 가지는 식품을 의미하고, 건강보조식품은 건강 보조 목적의 식품을 의미하는데, 경우에 따라, 건강기능식품, 건강식품, 건강보조식품의 용어는 혼용될 수 있다.
본 발명의 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물은 그대로 첨가되거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다.
본 발명의 식품은 당 업계에서 통상적으로 사용되는 방법에 의하여 제조 가능하며, 상기 제조 시에는 당 업계에서 통상적으로 첨가하는 원료 및 성분을 첨가하여 제조할 수 있다. 구체적으로, 상기 식품 조성물은 생리학적으로 허용 가능한 담체를 추가로 포함할 수 있는데, 담체의 종류는 특별히 제한되지 않으며 당해 기술 분야에서 통상적으로 사용되는 담체라면 어느 것이든 사용할 수 있다. 또한, 상기 식품 조성물은 방부제, 살균제, 산화방지제, 착색제, 발색제, 표백제, 조미료, 감미료, 향료, 팽창제, 강화제, 유화제, 증점제, 피막제, 검기초제, 거품억제제, 용제, 개량제 등의 식품 첨가물을 포함할 수 있다. 상기 첨가물은 식품의 종류에 따라 선별되고 적절한 양으로 사용될 수 있다.
또한, 상기 식품의 제형은 식품으로 인정되는 제형이면 제한 없이 제조될 수 있다. 본 발명의 식품용 조성물은 다양한 형태의 제형으로 제조될 수 있으며, 일반 약품과는 달리 식품을 원료로 하여 약품의 장기 복용 시 발생할 수 있는 부작용 등이 없는 장점이 있고 휴대성이 뛰어나므로, 본 발명의 식품은 뇌졸중의 예방 또는 개선의 효과를 증진시키기 위한 보조제로 섭취가 가능하다.
본 발명의 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피는 뇌졸중의 예방 또는 개선 효과를 나타낼 수 있다면 식품 조성물에 다양한 중량%로 포함될 수 있다. 구체적으로 식품 조성물의 총 중량 대비 0.00001 내지 100 중량% 또는 0.01 내지 80 중량%로 포함될 수 있으나, 이에 제한되지 않는다. 건강 및 위생을 목적으로 장기간 섭취할 경우에는 상기 범위 이하의 함량을 포함할 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
본 발명의 또 다른 실시 양태는 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 뇌졸중 예방 또는 개선용 사료 조성물을 제공하는 것이다.
이때, 상기 용어 “구기자”, “숙지황”, “산수유”, “산약”, “복령”, “택사”, “목단피”, “추출물”, “분획물”, “예방” 및 “개선”의 정의는 상기에서 서술한 바와 같다.
본 발명에 따른 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물은 우수한 뇌졸중 치료 효과를 나타내므로, 뇌졸중의 예방 또는 개선을 목적으로 사료 조성물에 포함될 수 있으며, 상기 사료 조성물은 동물이 일상적으로 섭취하는 것이 가능하기 때문에 뇌졸중의 예방 또는 개선에 대하여 높은 효과를 기대할 수 있다.
본 발명의 용어, "사료"는 동물이 먹고, 섭취하며, 소화시키기 위한 또는 이에 적당한 임의의 천연 또는 인공 규정식, 한끼식 등 또는 상기 한끼식의 성분을 의미한다.
상기 사료의 종류는 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용되는 사료를 사용할 수 있다. 상기 사료의 비제한적인 예로는, 곡물류, 근과류, 식품 가공 부산물류, 조류, 섬유질류, 제약 부산물류, 유지류, 전분류, 박류 또는 곡물 부산물류 등과 같은 식물성 사료; 단백질류, 무기물류, 유지류, 광물성류, 유지류, 단세포 단백질류, 동물성 플랑크톤류 또는 음식물 등과 같은 동물성 사료를 들 수 있다. 이들은 단독으로 사용되거나 2종 이상을 혼합하여 사용될 수 있다.
본 발명의 또 다른 실시 양태는 상기 약학 조성물을 투여하는 단계를 포함하는 뇌졸중 예방 또는 치료 방법을 제공하는 것이다.
이때, 상기 “약학 조성물”, “예방” 및 “치료”에 대한 설명은 상기에서 서술한 바와 같다.
본 발명의 약학 조성물은 뇌졸중의 예방 또는 치료 효과를 나타내므로, 이를 개체에 투여하는 단계를 포함하는 본 발명의 방법은 뇌졸중의 예방 또는 치료에 유용하게 활용될 수 있다.
본 발명의 용어, "투여"는 적절한 방법으로 개체에게 상기 조성물을 도입하는 것을 의미한다.
본 발명의 용어, "개체"는 뇌졸중이 발병하였거나 발병할 수 있는 인간을 포함한 쥐, 생쥐, 가축 등을 포함하는 포유 동물일 수 있으나, 본 발명의 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는 약학 조성물로 뇌졸중의 예방 또는 치료가 가능한 개체는 제한 없이 포함된다. 이에 제한되지 않는다. 또한, 본 발명의 개체에서 인간은 제외될 수 있으나, 이에 제한되지 않는다.
본 발명의 방법은 상기 약학 조성물을 약학적으로 유효한 양으로 투여하는 것을 포함할 수 있다. 적합한 총 1일 사용량은 올바른 의학적 판단범위 내에서 처치의에 의해 결정될 수 있으며, 1회 또는 수회로 나누어 투여할 수 있다.
구체적으로, 본 발명의 용어 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 유효용량 수준은 환자의 건강상태, 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
예컨대, 투여 경로, 질병의 중증도, 성별, 체중, 연령 등에 따라서 증감될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
구체적으로, 본 발명의 약학 조성물의 투여량은 예를 들어, 본 발명의 약학 조성물을 사람을 포함하는 동물에 하루 동안 0.1 내지 200 mg/체중(kg), 구체적으로는 1 내지 200 mg/체중(kg), 10 내지 150 mg/체중(kg) 또는 40 내지 110 mg/체중(kg), 더욱 구체적으로는 45 내지 55 mg/체중(kg), 보다 구체적으로는 약 50 mg/체중(kg)으로 투여할 수 있으나, 이에 제한되지 않는다. 본 발명의 조성물의 투여 빈도는 특별히 이에 제한되지 않으나, 1일 1회 투여하거나 또는 용량을 분할하여 수회 투여할 수 있다. 그러나, 투여 경로, 질병의 중증도, 성별, 체중, 연령 등에 따라서 증감될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 또 다른 실시 양태는 상기 약학 조성물의 제조방법을 제공하는 것이다.
상기 방법은 (a) 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피에 각각 물, 탄소수 1 내지 6의 알코올 또는 이들의 혼합 용매를 가하여 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물을 각각 추출하는 단계;
(b) 상기 (a) 단계에서 추출한 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물을 감압 농축하여 농축물을 제조하는 단계;
(c) 상기 (b) 단계에서 제조된 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 농축물을 건조하여 건조엑스를 제조하는 단계;
(d) 상기 (c) 단계에서 제조된 각 구기자, 숙지황, 산수유, 산약, 택사 및 목단피 건조엑스에 옥수수전분을 혼합하는 단계;
(e) 상기 (c) 단계에서 제조된 복령 건조엑스 또는 상기 (d) 단계에서 제조된 옥수수전분이 혼합된 각 구기자, 숙지황, 산수유, 산약, 택사 및 목단피 건조엑스를 1: 1: 0.5: 0.5: 0.25: 0.25: 0.25 중량비(w/w)로 혼합하여 생약 복합 추출물의 건조엑스를 제조하는 단계; 및
(f) 상기 (e) 단계에서 제조된 생약 복합 추출물의 건조엑스에 물, 탄소수 1 내지 6의 알코올 또는 이들의 혼합 용매를 추가하는 단계;를 포함할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 제조 방법에 있어, 상기 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물은 상기 재료 각각을 건조, 분쇄 또는 추출하여 혼합하거나, 상기 재료 중 두 개 이상을 혼합하여 건조, 분쇄 또는 추출하여 제조하거나, 또는 상기 재료 중 일부는 추출하고 다른 일부는 건조 후 분쇄하여 혼합하여 제조할 수 있다.
이하, 본 발명의 제조 방법의 일 구체예를 설명한다.
먼저, 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피에 각각 물, 탄소수 1 내지 6의 알코올 또는 이들의 혼합 용매를 가하여 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물을 각각 추출한다((a) 단계).
본 발명의 제조 방법에 있어, 상기 (a) 단계 이전에, 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피를 각각 건조 후 파쇄하는 단계를 추가적으로 포함하여 제조할 수 있다.
상기 본 발명의 제조 방법에 있어, 추출하는 경우 상기 재료의 건조물 또는 분쇄물에 물, 탄소수 1 내지 4의 알코올 또는 이들의 혼합 용매를 가하여 추출할 수 있고, 구체적으로는 물을 용매로 가하여 추출할 수 있다.
본 발명의 제조 방법에 있어, 상기 가하는 용매의 양은 상기 추출하고자 하는 재료의 5 내지 15배 중량 또는 부피, 구체적으로 10 내지 12배를 가할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 제조 방법에 있어, 상기 추출은 진탕 추출, Soxhlet 추출 또는 환류추출 방법을 이용할 수 있으나 이에 한정되지 않는다. 또한 추출 온도는 40 내지 100℃, 추출 시간은 0.5 내지 24시간일 수 있으며, 추출회수는 1 내지 5회일 수 있으나, 이에 제한되는 것은 아니다.
구체적으로, 상기 추출은 90 내지 110℃의 온도 범위에서 2 내지 3시간 동안 수행될 수 있으나, 이에 제한되는 것은 아니다.
이어서, 상기 (a) 단계에서 추출한 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물을 감압 농축하여 농축물을 제조한다((b) 단계).
이어서, 상기 (b) 단계에서 제조된 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 농축물을 건조하여 건조엑스를 제조한다((c) 단계).
본 발명의 제조 방법에 있어, 상기 추출물은 농축 및 건조될 수 있으며, 상기 농축은 감압농축, 진공 농축일 수 있으며, 구체적으로는 감압농축일 수 있으나, 이에 제한되는 것은 아니다. 상기 건조는 동결건조, 감압건조, 진공건조, 비등건조 또는 분무건조 방법일 수 있고, 구체적으로는 동결건조일 수 있으나, 이에 제한되는 것은 아니다.
구체적으로, 상기 감압 농축은 40 내지 45℃의 온도 범위에서 수행 될 수 있으나, 이에 제한되는 것은 아니다.
이어서, 상기 (c) 단계에서 제조된 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 건조엑스를 1: 1: 0.5: 0.5: 0.25: 0.25: 0.25 중량비(w/w)로 혼합하여 생약 복합 추출물의 건조엑스를 제조한다((d) 단계).
본 발명의 제조 방법에 있어, 상기 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물은 구기자 1 중량 대비하여, 숙지황 0.7 내지 1.3, 산수유 0.2 내지 0.8, 산약 0.2 내지 0.8, 복령 0.01 내지 0.55, 택사 0.01 내지 0.55 및 목단피 0.01 내지 0.55의 중량비로 혼합하여 제조될 수 있고, 구체적으로 구기자 1 중량 대비하여, 숙지황 0.9 내지 1.1, 산수유 0.4 내지 0.6, 산약 0.4 내지 0.6, 복령 0.15 내지 0.35, 택사 0.15 내지 0.35 및 목단피 0.15 내지 0.35의 중량비로 혼합하여 제조될 수 있고, 보다 구체적으로 구기자 1 중량 대비하여, 숙지황 0.95 내지 1.05, 산수유 0.45 내지 0.55, 산약 0.45 내지 0.55, 복령 0.2 내지 0.3, 택사 0.2 내지 0.3 및 목단피 0.2 내지 0.3의 중량비로 혼합하여 제조될 수 있다.
이어서, 상기 (d) 단계에서 제조된 생약 복합 추출물의 건조엑스에 물, 탄소수 1 내지 6의 알코올 또는 이들의 혼합 용매를 추가한다((e) 단계).
구체적으로는 물을 용매로 가할 수 있다.
본 발명의 또 다른 하나의 목적은 상기 뇌졸중 예방 또는 치료용 약학 조성물을 제조하기 위한, 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 포함하는 조성물의 용도를 제공한다.
본 발명의 또 다른 하나의 목적은 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 포함하는 조성물의 뇌졸중 예방 또는 치료 용도를 제공한다.
이하 본 발명을 하기 예를 통하여 보다 상세하게 설명한다. 그러나 이들 예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1: 생약 추출물 건조엑스의 제조
실시예 1-1: 구기자 건조엑스의 제조
생약을 정선하여 대한약전 통칙[절도 및 분말도] 항목에 따라 조절로 한 구기자 1kg에 정제수를 5 내지 10배 부피로 넣고, 90 내지 110℃에서 2 내지 3시간 추출한 다음, 여과한 액을 저온(40 내지 45℃) 감압 농축하여 건조한 다음 건조엑스 약 367.6g을 얻은 후 옥수수전분(USP) 약 211.8g을 혼합하여 제조하였다.
실시예 1-2: 숙지황 건조엑스의 제조
생약을 정선하여 대한약전 통칙[절도 및 분말도] 항목에 따라 조절로 한 숙지황 1kg에 정제수를 5 내지 10배 부피로 넣고, 90 내지 110℃에서 2 내지 3시간 추출한 다음 여과한 액을 저온(40 내지 45℃) 감압 농축하여 건조한 다음 건조엑스 약 857.7g을 얻은 후 옥수수전분(USP) 약 403.6g을 혼합하여 제조하였다.
실시예 1-3: 산수유 건조엑스의 제조
생약을 정선하여 대한약전 통칙[절도 및 분말도] 항목에 따라 조절로 한 산수유 1kg에 정제수를 5 내지 10배 부피로 넣고, 90 내지 110℃에서 2 내지 3시간 추출한 다음 여과한 액을 저온(40 내지 45℃) 감압 농축하여 건조한 다음 건조엑스 약 270.2g을 얻은 후 옥수수전분(USP) 약 180.1g을 혼합하여 제조하였다.
실시예 1-4: 산약 건조엑스의 제조
생약을 정선하여 대한약전 통칙[절도 및 분말도] 항목에 따라 조절로 한 산약 1kg에 정제수를 5 내지 10배 부피로 넣고, 90 내지 110℃에서 2 내지 3시간 추출한 다음 여과한 액을 저온(40 내지 45℃) 감압 농축하여 건조한 다음 건조엑스 약 149.0g을 얻은 후 옥수수전분(USP) 약 73.3g을 혼합하여 제조하였다.
실시예 1-5: 복령 건조엑스의 제조
생약을 정선하여 대한약전 통칙[절도 및 분말도] 항목에 따라 조절로 한 산수유 1kg에 정제수를 5 내지 10배 부피로 넣고, 90 내지 110℃에서 2 내지 3시간 추출한 다음 여과한 액을 저온(40 내지 45℃) 감압 농축하여 건조한 다음 건조엑스 약 980.3g을 얻었다.
실시예 1-6: 택사 건조엑스의 제조
생약을 정선하여 대한약전 통칙[절도 및 분말도] 항목에 따라 조절로 한 택사 1kg에 정제수를 5 내지 10배 부피로 넣고, 90 내지 110℃에서 2 내지 3시간 추출한 다음 여과한 액을 저온(40 내지 45℃) 감압 농축하여 건조한 다음 건조엑스 약 247.5g을 얻은 후 옥수수전분(USP) 약 165g을 혼합하여 제조하였다.
실시예 1-7: 목단피 건조엑스의 제조
생약을 정선하여 대한약전 통칙[절도 및 분말도] 항목에 따라 조절로 한 목단피 1kg에 정제수를 5 내지 10배 부피로 넣고, 90 내지 110℃에서 2 내지 3시간 추출한 다음 여과한 액을 저온(40 내지 45℃) 감압 농축하여 건조한 다음 건조엑스 약 373.1g을 얻은 후 옥수수전분(USP) 약 183.7g을 혼합하여 제조하였다.
실시예 2: 생약 추출물의 건조엑스의 제조
상기 실시예 1-1 내지 실시예 1-7에서 제조한 건조 엑스를 1: 1: 0.5: 0.5: 0.25: 0.25: 0.25 중량비(w/w)로 포함하도록 혼합하여 제조하였다.
실험예 1: 본 발명의 생약 추출물 처리로 인한 in vitro 신경세포 보호 효과 확인
실험예 1-1: 실험 준비
일차배양한 신경세포는 임신한 쥐의 태아에서 얻은 배아(E14-15)에서 후두근, 선조체, 해마를 제거하여 피질 조직을 분리하였다. 상온에서 20분 동안 트립신 용액에 두었다. 미리 데워진 DMEM 배양액으로 트립신을 씻어낸 후, 세포를 분리하였고 폴리에틸렌아민으로 사전 코팅된 96-well 세포 배양 플레이트에서 배양을 시작하였다. 2~3일 간격으로 항산화 물질이 첨가된 배지(antioxidant, B27;+AO)를 교환하고, 배양 7일차부터 항산화효과를 배제하기 위해 항산화 물질이 배제된 배지(-AO)로 교환하였다.
본 발명의 생약 추출물의 뇌졸중 신경세포 손상에 대해 신경세포 보호효과를 측정하기 위하여 배양 10일차에 신경독성 물질(glutamate, Glu, G, 100uM), 상기 실시예 2에서 제조한 생약 건조엑스를 10% DMSO가 포함된 식염수(saline)에 용해한 생약 추출물(PM, 0.1mg/ml 또는 1mg/ml) 또는 대조군(10% DMSO가 포함된 식염수, Vehicle, Veh)을 배지와 함께 세포에 투여하였으며, 48시간 후 세포들은 면역세포화학(immunocytochemistry)을 위해 4% paraformaldehyde을 이용하여 고정하였다.
실험예 1-2: MAP2-
면역반응성 (MAP2-ir) 확인
도 2에 나타난 바와 같이, 신경독성 물질인 글루타메이트를 배양중인 신경세포에 첨가하였을 때, 대조군인 그룹(도 2의 A)에 비해 신경독성물질인 글루타메이트를 처리한 그룹(도 2의 B)에서 세포수가 현저히 감소되는 것을 확인하였다.
반면, 본 발명의 생약 추출물 처리군(도 2의 C 및 도 2의 D)은 신경독성 물질인 글루타메이트 처리 조건에서 글루타메이트에 의한 세포수의 감소를 방지할 수 있음을 확인하였다.
또한, 이를 Neuronal 세포들의 마커인 microtubulin-associated protein 2 (MAP2)를 이용하여, 면역세포화학 실험을 수행한 결과, 도 2의 E에 나타난 바와 같이, 본 발명의 생약 추출물 처리군은 신경 세포의 마커인 MAP2의 면역반응성(immunoreactivity)이 대조군(40%)에 비하여 저농도에서 약 20%, 고농도에서 약 40% 증가된 것을 확인하였다.
그에 따라, 본 발명의 생약 추출물이 신경세포 보호 효과로 인한 뇌졸중 예방 효과가 있음을 확인하였다.
실험예 1-3: TUNEL activity 확인
도 3에 나타난 바와 같이, 세포 자멸의 측면에서도 글루타메이트를 배양중인 신경세포에 첨가하였을 때, 대조군인 그룹(도 3의 A)에 비해 신경독성물질인 글루타메이트를 처리한 그룹(도 3의 B)에서는 세포 자멸이 유도되는 것을 확인하였다. 그러나, 본 발명의 생약 추출물을 투여한 저농도 그룹(도 3의 C) 및 고농도 그룹(도 3의 D)에서는 독성 물질인 글루타메이트에 의한 세포 자멸을 방지할 수 있음을 확인하였다.
한편, TUNEL은 apoptotic DNA fragmentation을 검출하는 방법으로, apoptotic cell을 식별 및 정량화 하거나 개별 세포에서 과도한 DNA 파손을 검출하는데 사용한다. 구체적으로, TdT(terminal deoxynucleotidyl transferase) 효소를 이용하여, UTP(uridine triphosphate)를 DNA 말단에 붙여 DNA fragmentation이 일어났는지 여부를 확인하였다(In Situ Cell Death Detection Kit; Roche, Indianapolis, IN).
그 결과, 도 4에 나타난 바와 같이, 본 발명의 생약 추출물을 투여한 그룹에서 세포 자멸도가 현저하게 줄어든 것을 확인하였다.
그에 따라, 본 발명의 생약 추출물이 신경세포 보호 효과로 인한 뇌졸중 예방 효과가 있음을 확인하였다.

비가역성 뇌손상은 Ca2+의 세포 내 축적으로 인한 염기성 탈분극화(anoxic depolarization of the cell membrane) 후에 일어난다. N-메틸-D 아스파르트산염 수용체(N-methyl-D-aspartate receptor, NMDAR)는 Ca2+ 매개의 neuronal death에 밀접하게 관계가 있기 때문에, NMDAR의 활성화를 통해 유발된 Ca2+ 축적을 gCaMP5라는 유전적으로 암호화된 칼슘 지표 단백질의 형광을 통해 확인하였다.
그 결과, 도 5에 나타난 바와 같이, NMDAR을 자극하기 위해 NMDA 첨가하였을 때, 대조군인 그룹(도 5의 A)에서 신경세포내 Ca2+ 유입으로 세포 내 Ca2+ 농도가 증가하는 것을 확인하였다. 그러나, NMDA를 첨가하기 3분(180초) 전에 본 발명의 생약 추출물을 투여한 그룹(도 5의 B)에서는 신경세포 내 Ca2+ 유입이 현저하게 적은 것을 확인하였다.
구체적으로, 도 6의 A는 신경세포 배양 중 NMDA 물질을 처리하기 3초 전의 대조군(control)이며, 도 6의 A1은 신경세포 배양 중 NMDA 물질 처리 5초 후의 대조군(control)이고, 도 6의 B는 신경세포 배양 중 NMDA 물질을 처리하기 3초 전의 본 발명의 생약 추출물을 투여한 그룹(PM012)이며, 도 6의 B1은 NMDA 물질 처리 5초 후의 발명의 생약 추출물을 투여한 그룹(PM012)이다 (NMDA 실험).
이어서, 상기 NMDA 실험 이후, 세포들을 고정하여 면역세포화학법을 이용하여 각각의 marker들에 대한 발현양을 분석하여 도 6의 C에 나타내었다.
구체적으로, 도 6의 C의 상단 부분은 대조군(control), 도 6의 C의 하단 부분은 각각 본 발명의 생약 추출물을 투여한 그룹(PM012)이다.
이어서, 상기 NMDA 실험의 각각의 실험군에서 각각 대조군 44개(control), 본 발명의 생약 추출물을 투여한 그룹 33개(PM012)의 세포들에 대해 칼슘유입 지표인 녹색 (GFP)의 발현량을 시간에 따라 비교한 결과를 도 6의 D에 나타내었다.
그 결과, 도 6에 나타난 바와 같이, gCaMP5로 확인된 Ca2+은 NMDA 5μM를 투여한 시점에서 2초(s)이내에 세포 내 유입이 증가하였지만, 본 발명의 생약 추출물 투여군은 NMDA로 인한 Ca2+ 유입이 억제되고, Ca2+ 수준이 유지되는 것을 확인하였다.
그에 따라, 본 발명의 생약 추출물이 칼슘대사 조절에 의한 신경세포 보호 효과로 인한 뇌졸중 예방 효과가 있음을 확인하였다.
실험예 1-5: 신경세포 손상에 대한 특이적인 보호 효과 확인
신경독성 물질(glutamate, Glu, G, 100uM), 상기 실시예 2에서 제조한 생약 추출물(PM, 0.1mg/ml 또는 1mg/ml) 또는 대조군(Vehicle, Veh)을 DMSO를 용매로 이용하여 제조한 것을 제외하고는, 상기 실험예 1-2과 동일한 방법으로 상기 실시예 2에서 제조한 생약 추출물의 신경세포 보호 효과를 확인하였다. 그 결과는 도 7에 나타내었다.
도 7에 나타난 바와 같이, DMSO 독성 또는 본 발명의 생약 추출물 자체로는 MAP2의 면역반응성(immunoreactivity)을 변화시키지 않음을 확인하였다. 반면, 신경독성 물질인 글루타메이트 및 본 발명의 생약 추출물 처리군에서는 MAP2의 면역반응성(immunoreactivity)이 글루타메이트 단독 처리군 또는 글루타메이트 및 DMSO 처리군에 비하여 증가하는 것을 확인하였다.
그에 따라, 본 발명의 생약 추출물이 신경세포 손상에 대한 특이적인 보호 효과를 보여 뇌졸중 예방 효과가 있음을 확인하였다.
실험예 2: 본 발명의 생약 추출물 처리로 인한 in vivo 뇌졸중 예방 효과 확인
실험예 2-1: 실험동물의 준비
실험동물로 300g 내외의 8 ~ 9주령의 수컷 스프라그 돌리(Sprague Dawley, BioLASCO, Taiwan) 랫트(rat)를 구입하여 사료와 물을 충분히 공급하면서 실험환경에 적응하도록 하였다. 본 발명의 생약 추출물의 국소 뇌허혈에 의한 신경세포 손상에 대하여 신경세포 보호 효과를 측정하기 위하여 허혈성 뇌졸중 동물실험 모델(중대뇌동맥 결찰, Middle Cerebral Artery occlusion; MCAo)을 사용하였다. 뇌졸중 수술 2일 전부터 대조군에는 10% DMSO가 포함된 식염수(saline), 상기 실시예 2에서 제조한 생약 건조엑스는 10% DMSO가 포함된 식염수(saline)에 용해하여 복강주사로 주입한 후, 2일차에 중대뇌동맥폐쇄술(MCAo)을 진행하였다 (도 9)(Yu SJ, Reiner D, Shen H, Wu KJ, Liu QR, Wang Y. Time-dependent protection of CB2 receptor agonist in stroke. PLoS One. 2015;10: e0132487).
구체적으로, 먼저 상기 랫트를 클로랄 하이드레이트(chloral hydrate, 400mg/kg, i.p.) 마취를 한 후, 오른쪽 두개골에서 약 2 ~ 4㎜의 개두술을 진행하였다. 오른쪽 MCA는 10-0으로 봉합하고, 경동맥은 클립으로 혈류를 막았다. 그 후, 60분 후에 MCA의 봉합과 경동맥의 클립을 제거함으로써 혈류의 재관류를 시행하였다. 수술은 체온은 37℃에서 모니터링 및 유지하였고, 마취에서 회복된 동물들은 온도 조절 인큐베이터안에서 체온을 37℃로 유지시켰다.
실험예 2-2: TTC(2,3,5-triphenyltetrazolium chloride) 염색에 의한 생약 추출물의 뇌졸중 예방 효과 확인
뇌혈류 감소가 일정 시간 이상 지속되면 뇌조직의 괴사(조직이나 세포의 일부가 죽는 것)가 시작된다. 뇌조직이 괴사되어 회복 불가능한 상태에 이르렀을 때 이를 뇌경색(cerebral infarction)이라고 하며, 이를 미토콘드리아의 디하이드로게네이즈(dehydrogenase) 반응을 이용한 TTC(2,3,5-Triphenyltetrazolium Chloride) 염색을 하면 죽은 조직은 흰색으로, 살아있는 조직은 붉게 염색할 수 있다.
생약 추출물의 뇌졸중 예방 효과를 확인하기 위하여, 실험동물에 50mg/㎏, 100mg/㎏로 용량을 달리하여 생약 추출물을 2일간 복강 투여한 후, 상기 실험예 2-1의 방법으로 뇌손상을 유도하였다. 이어서, 뇌졸중 유발 2일 경과 후, 실험동물(실험군 및 대조군)의 뇌를 적출하였다.
적출된 뇌는 2mm 간격으로 브레인 매트릭스(brain matrix)를 사용하여 절편을 만든 후, 2% TTC(2,3,5-triphenyltetrazolium chloride) 용액에서 5분간 배양한 후, 4% PFA 용액으로 고정시켰다. 염색이 끝난 뇌조직 절편은 디지털 스캐너와 이미지 도구 프로그램(텍사스 대학교 보건 과학 센터)으로 뇌손상 부위를 측정하였다. 그 결과는 도 9(도 9의 A: 염색된 뇌 절편 사진, 도 9의 B: A의 손상부분을 수치화한 그래프)에 나타내었다.
실험에서 사용된 TTC 염색은 정상세포에서는 미토콘드리아 내의 디하이드로제나제(dehydrogenase)와 반응하여 붉게 보이나, 뇌경색의 경우는 디하이드로제나제가 소실되어 염색되지 않으므로 백색으로 보여 세포의 비가역적 손상의 유무를 알 수 있는 지표로 널리 사용된다.
그 결과, 도 9의 A에 나타난 바와 같이, 뇌조직 중에서 손상으로 인하여 조직이 자멸한 부위는 염색이 되지 않아 흰색, 정상 부위 조직은 염색이 되어 흑적색을 나타내는 것을 확인하였다. MCAo 수술이 뇌 조직의 손상을 유발하는 것을 확인하였고(Veh), 본 발명의 생약 추출물을 사전 투여한 경우, 대조군에 비해 뇌손상이 현저히 적어 흑적색 부위의 면적이 넓고, 신경세포가 자멸한 흰색 부위의 면적이 적음을 확인하였다.
또한, 도 9의 B에 나타난 바와 같이, MCAo 수술이 뇌 조직의 손상을 유발하는 것을 확인하였고(Veh), 생약 추출물을 사전 투여한 경우, 50㎎/㎏(p=0.017), 100㎎/㎏(p=0.034)로 유의적인 뇌 조직의 손상 감소 효과를 보임을 확인하였다. 그 이상의 용량인 200mg/kg에서는 뇌 손상의 감소를 보이기는 하였지만, 100mg/kg 투여와는 달리 통계적으로 유의한 뇌 손상의 감소를 보이지는 않았다.
따라서, 본 발명의 생약 추출물의 뇌경색 감소로 인한 뇌졸중 예방 효능을 확인하였다. 즉, 본 발명은 상기에서 사용한 본 발명의 중대뇌동맥 결찰(middle cerebral artery occlusion; MCAo)에 의한 뇌손상 유도 모델을 통해 생약 추출물의 뇌졸중 예방 효과를 명확히 확인한 것으로, 이는 치료 효능을 확인한 것과는 또 다른 측면에서 생약 추출물의 뇌졸중 예방 효능을 명확히 확인한 것이다.
실험예 2-3: 행동테스트 분석을 통한 생약 추출물의 뇌졸중 예방 효과 확인
뇌졸중 유발모델처럼 신경 손상이 유발된 모델은 근육과 신경의 기능이 쇠퇴하여 자발적 행동량이 감소하는 경향을 보인다. 본 실험에서는 뇌졸중이 유발된 실험동물의 행동에 생약 추출물의 투여가 미치는 영향을 확인하기 위하여, 13마리의 실험군(PM012)에 50mg/㎏ 용량으로 본 발명의 생약 추출물을, 9마리의 비히클군(veh)에 비히클을 각각 2일간 복강 투여한 후, 상기 실험예 2-1의 MCAo를 사용하여 뇌손상을 유도하였다. 5마리의 대조군(naive)의 경우, MCAo를 받지 않고 순수 대조군으로 사용되었다. 이어서, 각각의 실험동물을 외부에 바닥에서 10cm 떨어진 곳에 8개의 적외선 센서가 설치된 42×42×31㎤ 사이즈의 투명한 아크릴 상자에 두고 2일(D2, 뇌졸중 유발 2일경과)차에 2시간 동안 활동성을 측정하였다. 투명상자 외부에는 바닥에서 10cm 떨어진 곳에 8개의 적외선 센서가 포함되어 동물들의 움직임을 기록할 수 있다 (Acccuscan, Columbus, OH). 구체적으로, 수직 활동(수직 센서에서 발생한 총 빔 중단 수, VACTV), 총 이동 거리(센티미터로 이동한 거리, TOTDIST), 수평 활동(수평 센서에서 발생한 총 빔 중단 수, HACTV)는 Versamax 프로그램(Accus, Columbus, O)에 의해 분석되었다. 그 결과는 도 10에 나타내었다.
도 10에 나타난 바와 같이, 비히클군(veh)에서 실험동물의 움직임이 최소화 된 반면, 본 발명의 생약 추출물(50mg/㎏)을 미리 투여한 실험군(PM012)에서는 뇌졸중의 유도로 감소된 HACTV (도 10의 A), TOTDIST (도 10의 B) 및 VACTV (도 10의 C)를 뇌졸중이 유도되지 않은 대조군(naive) 수준으로 회복함을 확인하였다. 즉, 본 발명의 생약 추출물의 처리는 뇌졸중 쥐에서 전체적인 운동량을 현저하게 증가시킴을 확인하였다.
그에 따라, 본 발명의 생약 추출물이 뇌졸중 예방 효능이 있음을 확인할 수 있었다. 즉, 본 발명은 상기에서 사용한 본 발명의 중대뇌동맥 결찰 (middle cerebral artery occlusion; MCAo)에 의한 뇌손상 유도 모델을 통해 생약 추출물을 미리 투여한 경우, 뇌졸중 후 동반되는 행동장애의 개선이 나타남을 확인하여 뇌졸중 예방 효과를 명확히 확인한 것으로, 이는 치료 효능을 확인한 것과는 또 다른 측면에서 생약 추출물의 뇌졸중 예방 효능을 명확히 확인한 것이다.
실험예 2-4: 염증 관련 유전자 확인
염증 관련 유전자를 확인하기 위하여, 실험동물에 50mg/㎏ 용량으로 본 발명의 생약 추출물을 2일간 복강 투여한 후, 상기 실험예 2-1의 방법으로 뇌손상을 유도하였다. 상기 랫트는 MCAo 수술 2일차에 염증 관련 특이 유전자 발현 분석을 위해 대뇌피질 조직을 채취했다. 이는 PCR 실험을 위해 사용되었으며, 소교세포 마커(IBA1; Anti-ionized Ca2+ binding adapter molecule 1), 염증 촉진 사이토카인(IL-6; Interleukin-6), M1 표시인자(CD86; pro-inflammatory), M2 표지인자(CD206; anti-inflammatory)를 통해 유전자 분석을 하였다(각각, 도 11의 A 내지 도 11의 D). 구체적으로, lesioned는 뇌졸중이 유발된 뇌 반구를 나타내는 것이며, Non-lesioned는 뇌졸중이 유발되지 않은 뇌 반구를 나타내는 것이다.
그 결과, 도 11에 나타난 바와 같이, IBA1과 IL-6의 마커들을 이용하여 확인한 결과, 본 발명의 생약 추출물 그룹에서 발현된 소교세포 마커(IBA1)와 염증 촉진 사이토카인(IL-6)은 대조군에 비해 유의하게 감소한 것을 확인하였다. 이를 통해 본 발명의 생약 추출물이 소교세포 활성 등 염증을 억제하여 신경세포 자멸을 억제하는 것을 확인하였다.
또한, 구기지황탕 추출물을 처리한 그룹에서 MCAo 수술로 인해 증가된 M1 표시인자(proinflammatory marker; CD86)의 발현을 억제하는 것을 확인하였고, 활성화된 M2표시인자(anti-inflammatory marker; CD206)가 대조군에 비해 유의적으로 증가하는 것을 확인하였다. 이는 본 발명의 생약 추출물 처리가 염증 촉진 인자들을 분비하여 신경 손상을 유발하는 M1 type을 대식작용에 의해 손상된 세포 잔해 제거 및 신경보호를 촉진하는 M2 type으로의 변화를 유도하고, M2 type의 증가는 허혈성 뇌졸중을 회복하는 동안 뇌 조직 복구에 기여하는 것을 시사한다.
그에 따라, 본 발명의 생약 추출물이 항염증 효능에 의한 신경세포 보호 효과로 인한 뇌졸중 예방 효과가 있음을 확인하였다.
실험예 2-5: ER stress 및 세포 자멸 관련 유전자 확인
소포체(endoplasmic reticulum, ER)에 존재하는 ATF6, BIP, CHOP, IRE1, PERK, SIGMAR1 등 유전자들은 신호전달 과정은 세포의 생존에 관련되어 있을 뿐만 아니라 세포자멸에도 관여하는데, 세포에 대한 스트레스 상황이 매우 강하거나 지속적으로 일어나면 소포체의 기능이 비 정상적으로 작용하여 소포체 스트레스에 의한 세포자멸사를 유도한다.
소포체 스트레스 및 세포 자멸에 관여하는 유전자의 발현 변화를 확인하기 위하여, 실험동물에 50mg/㎏ 용량으로 본 발명의 생약 추출물을 2일간 복강 투여한 후, 상기 실험예 2-1의 방법으로 뇌손상을 유도하였다. 상기 랫트는 MCAo 수술 2일차에 소포체 스트레스와 세포자멸에 대한 특이 유전자 발현 분석을 위해 대뇌피질 조직을 채취했다. 이는 PCR 실험을 사용하였으며, 소포체 막통과 신호전달 물질 ARF6(activating transcription factor 6), BIP(immunoglobulin-binding protein), CHOP(C/EBP homologous protein), IRE1(inositol requiring 1), PERK(protein kinase PKR-like ER kinase), SIGMAR1(Sigma Non-Opioid Intracellular Receptor 1); 및 세포자멸사의 마커인 Caspase-3 발현을 측정하였다(각각, 도 12의 A 내지 도 12의 G). 구체적으로, lesioned는 뇌졸중이 유발된 뇌 반구를 나타내는 것이며, Non-lesioned는 뇌졸중이 유발되지 않은 뇌 반구를 나타내는 것이다.
그 결과, 도 12의 A 내지 도 12의 F에 나타난 바와 같이, 본 발명의 생약 추출물 처리 그룹(빨강)에서 소포체 스트레스에 의해 발현이 증가되는 유전자들은 대조군(검정)에 비해 모두 유의적으로 감소하는 것을 확인하였다.
아울러, 도 12의 G에 나타난 바와 같이, 본 발명의 생약 추출물 처리 그룹(빨강)에서는 대조군(검정)과 비교하였을 때, 세포자멸사의 마커인 Caspase-3의 발현량이 감소하는 것을 확인하였다.
그에 따라, 본 발명의 생약 추출물이 소포체 스트레스에 의한 자멸에 대한 신경세포 보호 효과로 인한 뇌졸중 예방 효과가 있음을 확인하였다.
실험예 2-6: 신경영양인자 관련 유전자 확인
신경영양인자 관련 유전자의 발현 변화를 확인하기 위하여, 실험동물에 50mg/㎏ 용량으로 본 발명의 생약 추출물을 2일간 복강 투여한 후, 상기 실험예 2-1의 방법으로 뇌손상을 유도하였다. 상기 랫트는 MCAo 수술 2일차에 신경영양인자 발현 분석을 위해 대뇌피질 조직을 채취했다. 이는 PCR 실험을 사용하였으며, 신경영양인자인 BDNF(brain-derived neurotrophic factor)와 GDNF(glial cell line-derived neurotrophic factor) 발현을 측정하였다(각각, 도 13의 A 및 도 13의 B). 신경영양인자는 신경세포의 성장 및 분화, 생존에 중요한 역할을 하는 것으로 알려져 있다.
그 결과, 도 13에 나타난 바와 같이, 본 발명의 생약 추출물 처리 그룹(빨강)에서는 대조군(검정)과 비교하였을 때, 신경인자인 BDNF 및 GDNF의 발현량이 증가하는 것을 확인하였다.
그에 따라, 본 발명의 생약 추출물이 신경인자를 통한 신경세포 보호 효과로 인한 뇌졸중 예방 효과가 있음을 확인하였다.
이상의 설명으로부터, 본 발명이 속하는 기술 분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시 예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로서 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (12)
1: 1: 0.5: 0.5: 0.25: 0.25: 0.25 중량비(w/w)의 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는, 뇌졸중 예방 또는 치료용 약학 조성물.
제1항에 있어서, 상기 뇌졸중은 허혈성 뇌졸중인 것인, 약학 조성물.
제1항에 있어서, 상기 추출물은 물, 탄소수 1 내지 4의 알코올, 및 이들의 혼합용매로 구성되는 군으로부터 선택되는 용매로 추출하여 제조되는 것인, 약학 조성물.
제1항에 있어서, 상기 추출물은 열수 추출물인 것인, 약학 조성물.
제1항에 있어서, 상기 조성물은 약학적으로 허용되는 담체, 부형제 또는 희석제를 추가로 포함하는 것인, 약학 조성물.
제1항 내지 제5항 중 어느 하나의 항의 조성물을 인간을 제외한 개체에 투여하는 단계를 포함하는 뇌졸중의 예방 또는 치료 방법.
1: 1: 0.5: 0.5: 0.25: 0.25: 0.25 중량비(w/w)의 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물 또는 이의 분획물을 유효성분으로 포함하는, 뇌졸중 예방 또는 개선용 식품 조성물.

(b) 상기 (a) 단계에서 추출한 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 추출물을 감압 농축하여 농축물을 제조하는 단계;
(c) 상기 (b) 단계에서 제조된 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 농축물을 건조하여 건조엑스를 제조하는 단계;
(d) 상기 (c) 단계에서 제조된 각 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피 건조엑스를 1: 1: 0.5: 0.5: 0.25: 0.25: 0.25 중량비(w/w)로 혼합하여 생약 복합 추출물의 건조엑스를 제조하는 단계; 및
(e) 상기 (d) 단계에서 제조된 생약 복합 추출물의 건조엑스에 물, 탄소수 1 내지 6의 알코올 또는 이들의 혼합 용매를 추가하는 단계;
를 포함하는, 뇌졸중 예방 또는 치료용 약학 조성물의 제조방법.
제9항에 있어서, 상기 (a) 단계 이전에, 구기자, 숙지황, 산수유, 산약, 복령, 택사 및 목단피를 각각 건조 후 파쇄하는 단계를 추가적으로 포함하는 것인, 방법.
제9항에 있어서, 상기 (a) 단계에서 상기 추출은 90 내지 110℃의 온도 범위에서 2 내지 3시간 동안 수행되는 것인, 방법.
제9항에 있어서, 상기 (b) 단계에서 상기 감압 농축은 40 내지 45℃의 온도 범위에서 수행되는 것인, 방법.
