Abstract
본 발명은 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 및 치료용 조성물에 관한 것으로, 본 발명에 따른 댕댕이나무열매 추출물은 신경세포 보호효과를 가져 허혈성 뇌혈관 질환에 의한 신경행동학적 결손 및 경색 용적을 현저하게 감소시켜 허혈성 뇌혈관 질환의 예방 또는 치료용 약학적 조성물로 제공될 수 있고, 허혈성 뇌혈관 질환의 예방 또는 개선용 건강식품으로 제공될 수 있다.
Description
본 발명은 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 및 치료용 약학적 조성물 또는 건강식품에 관한 것이다.
WHO의 통계에 의하면 매년 1,700만 명이 혈관질환으로 사망하고 있으며, 이는 전체 사망인구의 3분의 1에 해당하여 사망원인 1위이다. 혈관질환은 미국에서 65세 이상 인구에서는 사망원인 1위이며, 45 내지 64세 인구에서는 암에 이어 사망원인 2위이다. 또한, 노령인구의 증가와 함께 혈관질환에 의한 사망은 계속 증가 추세이다. 또한, 한국인의 사망원인은 1위 암, 2위 뇌혈관질환, 3위가 심장질환으로 혈액순환기 계통의 질환이 주요 사망원인의 2, 3위를 차지하고 있으며, 혈액순환관련 환자는 80년대 초와 비교할 때 7배 정도로 급증하고 있는 상황이다.
뇌졸중(stroke)은 흔히 '중풍'으로 알려져 있으며, 뇌에 혈액을 공급하고 있는 혈관이 막히거나 터짐으로써 그 부분의 뇌가 손상되어 급작스런 뇌혈류 장해에 의한 의식소실, 반신마비, 언어장애 등의 국소적 신경장애 증상을 일으킨 상태를 일컫는다. 뇌졸중은 크게 허혈성 뇌졸중(ischemic stroke)과 출혈성 뇌졸중 (hemorrhagic stroke)으로 구분된다. 허혈성 뇌졸중은 뇌조직으로 가는 혈액 공급의 감소 혹은 차단으로 뇌조직이 허혈상태가 되어 발생하며, 출혈성 뇌졸중은 혈관이 터져 뇌조직으로 출혈을 일으켜 발생한다. 즉, 원인에 따라 뇌혈관이 막혀서 생기는 뇌경색(cerebral infarction)과 반대로 뇌혈관이 터져서 발생하는 뇌출혈 (cerebral hemorrhage)로 구분되며, 뇌출혈은 다시 고혈압성 뇌내출혈 (intracerebral hemorrhage)과 뇌혈관이 꽈리모양의 변형을 일으켜 발생하는 뇌동맥류(cerebral aneurysm) 파열에 의한 뇌지주막하 출혈(subarachnoid hemorrhage, SAH)로 구분된다. 또한, 뇌경색은 뇌혈전증(뇌혈관에 혈전이 형성되어 혈관을 막는 것)과 뇌색전증(뇌혈관의 혈전 조각이나 심장이상으로 인한 피떡이 머리 혈관으로 흘러들어가 혈관을 막는 것)으로 구분된다. 뇌졸중은 뇌의 어느 부위에서나 발생할 수 있으므로 신체의 거의 모든 기능에 장애를 초래할 수 있다. 또한 발생하는 뇌손상은 손상의 정도와 위치에 따라 결정되는데, 뇌졸중에 의한 심각한 뇌손상의 경우 운동, 감각기능의 손상 및 기억, 학습, 연산, 추리 등 고차원적 기능을 저해하며, 중풍, 지능 박약, 학습 장애, 간질 등과 같은 심각한 후유증을 유발시킨다. 또한, 뇌졸중에서 회복된다 하여도 대부분의 경우 후유장해를 남기기 때문에 이에 관계되는 사회적, 경제적 손실은 지대하여 연간 70억 달러에 이른다(Davenport et al. Journal of Neurology, 2000, vol.68, p.277).
허혈성 뇌손상은 혈액순환에 의존적인데 뇌로 혈액을 공급하는 혈관에 여러 가지 병리학적 이상이 발생되어 정상적 뇌혈류(50 ml/gram/minute)의 장애가 초래되면 일과성 뇌허혈증(transient cerebral ischemia)으로부터 완전 뇌졸중(complete stroke)으로 급속히 발전, 사망한 것으로 보고되고 있다. 허혈성 뇌졸증은 갑자기 발생하고, 신경학적 이상을 동반하며 단시간내 급격하게 질병의 경과가 진행되어 응급 치료법인 색전용해술에 의해서도 완전 치유가 불가능하며 사망에 이르지 않더라도 부작용 및 후유증이 초래되어 직접간접적으로 심각한 사회, 경제적인 손실까지 야기되고 있으므로 예방이 매우 중요하게 인식되고 있다.
허혈성 뇌졸중은 주로 혈청 콜레스테롤의 증가, 고혈압, 심장질환 등 혈류량의 변화를 야기시키는 위험인자들의 제거에 주력하는 일차적 예방과 질환의 발생 후 악화를 예방하는 이차적 예방으로 나누어 볼 수 있는 바, 허혈성 뇌졸중을 예방할 수 있는 식품 관련 선행연구들은 특정 위험인자들에 의한 위해를 저감시키는데 주로 초점이 맞추어져 왔다. 특히, 뇌로 혈액의 공급감소 등에 기인된 뇌허혈(brain ischemia)의 발생시 글루타메이트(glutamate)에 의한 칼슘이온 채널의 개방에 따른 과도한 Ca 이온의 유입으로 과흥분에 의한 흥분성세포독성(exitotoxicity)이 야기되거나, 재관류(reperfusion)시 유발되는 산소 라디칼에 의해 신경세포내 효소군들이 파괴되는데 이러한 신경세포의 손상기전과 관련한 타겟 특이적인(target-specific) 연구가 미흡한 실정이므로 관련연구가 필요한 실정이다(Hagberg H et al., Neurosci Lett. 78:311-317, 1987).
최근 carnosine, ellagic acid 유도체, 카로틴 유도체, S-cysteinyl 유도체 등의 식품 성분들이 허혈성 뇌손상의 일차적 예방뿐 아니라 이차적 예방에도 효과가 있는 것으로 국외에서 보고되기 시작했다. 뇌졸중 치료제 개발을 위한 연구는 주로 1차(급성) 뇌손상을 막기 위한 시도로서 글루타민산수용체 차단제 개발과 항산화제를 이용한 치료제 개발이 시도되어 왔으나 임상시험 단계에서 효과가 없거나 독성 문제로 실패하였다. NMDA 수용체 길항제는 심각한 독성으로 사용되지 않고 있으며 대부분 임상시험에서 불합격되고 있다.
국내외적으로 식품성분들에 대해서는 신경세포의 손상기전과 관련한 타겟-특이적인(target-specific) 연구가 미흡할 뿐 아니라 식품소재의 효과를 동물의 뇌에서 최종적으로 검증하고 제품개발로 성공한 사례가 거의 없다. 또한 국내의 경우 대부분의 연구들이 실험실적 어세이 정도에서 그치고 있는 실정이며, 기능성 소재의 구명, 체내 생리활성 발현기작, 소재의 개질 기반기술이 취약하다.
현재 국내 시판되고 있는 대부분의 기능성소재는 단순복합제 형태이며, 일부 기능성 식품들은 산소접촉에 의한 산화, 갈변 등에 의하여 유용성이 저해되고 식품소재로서의 외형적 가치도 저급한 수준이므로 코팅에 의한 차단막 형성으로 저장안정성을 확보하고 유용성분의 활성을 유지시킬 수 있는 기본 기술뿐만 아니라 활성 소재를 적정 동반 식품(food vehicle)에 적용하여 활용도를 증진시킬 수 있는 가공기술이 요구되나 이에 관한 시도는 미흡한 수준이다.
현재까지 연구된 약물들은 글루타민산 수용체 길항제, 항산화제, 칼슘 혹은 나트륨, 등의 이온채널 차단제 등을 이용한 것이 대부분이며 효과적인 약물이 개발되지 못하고 있는 실정이다. 따라서, 획기적인 아이디어 전환과 새로운 신개념의 치료제 타겟발굴이 요구되고 있는 실정이다.
한편, 중추신경계 내에서도 염증반응이 나타나며 이는 신경퇴행을 야기하는 주요 원인중의 하나임이 보고되고 있어, 뇌허혈 후의 염증 반응은 새로운 뇌허혈 치료제의 목표가 될 것으로 보이며, 이러한 치료제는 세포 독성 물질들의 유리에 관여하는 효소들을 억제하거나, 호중구(neutrophil)의 침착을 억제하는 약물들로 구성될 것으로 사료된다(Dirnagl, U. et al; TINS., 22 , pp391-397, 1999).
휴에트(Hewette) 등은 활성화된 교세포가 신경세포의 사멸에 영향을 미칠 수 있다는 것을 발견하였다 (Hewett, S.J. et al: Neuron, 13:487-494 1994; Hewett, S.J. et al, Stroke, 27:1586-15911996). 활성화된 교세포는 세포독성을 유발할 수 있는 TNF-알파, 인터루킨 등과 같은 염증 유발 인자인 사이토카인(cytokine)의 생성을 증가시켜 신경세포와 교세포에 대해 독성을 증가시킨다.
뇌염증세포인 대식세포(macrophage)와 뇌신경교세포(microglia)로부터 분비되는 염증물질, 이로 인한 신경세포사멸이 중요하며 특히 신경교세포의 활성조절과 신경세포보호는 뇌졸중의 예방, 조절 및 치료법 개발의 핵심표적이다.
식물을 이용한 새로운 특용 작물 발굴 노력에 힘입어 그 생산량은 해마다 증가하고 있으나, 현재는 착유 공정 등 단순 가공처리만을 거쳐 일반 첨가 소재로 사용되고 있거나, 일부 한방 의약품에 추출물 형태로 첨가되고 있어 그 활용도가 낮은 실정이다. 그러나, 식물에는 허혈성 뇌졸중 예방 활성이 있을 것이라고 기대되는 활성물질의 존재 가능성이 높고 한방에서는 예로부터 수종 식물을 뇌졸중 예방에 사용해 오고 있어, 이를 건강식품 소재로 개발할 경우 식물의 부가가치가 증가될 것으로 사료된다. 따라서, 국내산 식물로부터 허혈성 뇌졸증을 예방할 수 있는 식품선도소재를 탐색하고 동물실험에서 효과를 검증한 다음 기능성 식품으로 응용할 수 있는 기술을 개발함으로써 건강기능성식품 개발에 이르는 체계적인 연구의 수행이 필요하다.
댕댕이나무(Lonicera caerulea L. var. edulis)는 꼭두서니목 인동과의 쌍떡잎식물로 높이가 1.5m에 달하는 낙엽관목으로서 가지가 많이 갈라지고 작은 가지의 마디에 방패 모양의 포엽이 있고 나무줄기의 속부분은 흰색이다. 잎은 마주나며 바소꼴이거나 타원형이고 양 끝이 뭉뚝하거나 날카로우며 톱니가 없고 가장자리 및 표면에 짧은 털이 나고 뒷면에 융모가 많다. 꽃은 대개 꽃자루가 짧고 잎겨드랑이에 달리며 꽃부리는 깔때기 모양으로 엷은 황백색이며 여름에 핀다. 꽃받침은 거치처럼 5개로 갈라지며, 화관은 황백색이고 원통상 종형이며 길이 1.2~1.5cm로서 털이 약간 있다. 수술은 암술대보다 짧고 털이 없으며 자방은 2개가 합쳐지고, 열매는 타원형 또는 거의 원형이며, 7~8월에 자흑색으로 익고 백분으로 덮인다. 시베리아, 사할린, 중국 북부, 티베트, 북한 등지에 분포하고 있는 냉대성 식물이다.
현재 댕댕이나무 추출물을 이용한 발명으로 한국등록특허 제 10-0699790호에는 댕댕이나무 추출물을 포함하는 간 질환 예방 및 치료 효과를 가지는 약제학적 조성물에 관한 것으로, 간염, 간경화 또는 지방간 등의 질환에 대한 예방 및 치료 효과가 기재되어 있고, 한국등록특허 제 10-0454150호에는 현호색, 산해박, 댕댕이덩굴 등으로 구성되는 그룹으로 선택되는 1종의 약초 추출물을 조성물 총 중량에 대하여 0.001~10 중량% 포함하여 이루어지는 저자극성 화장료 조성물에 관한 것으로, 피부 발진, 피부 알러지 등의 부작용이 적어 가려움증등을 예방할 수 있다고 기재되어 있다. 그러나, 본 발명에서와 같이 댕댕이나무열매 추출물을 허혈성 뇌혈관 질환에 사용한 발명은 없었다.
이에, 본 발명자들은 허혈성 뇌질환 예방 또는 치료에 유용하게 사용될 수 있는 물질을 탐색하던 중, 댕댕이나무열매 추출물이 신경행동학적 결손의 억제와 뇌경색 용적이 감소하는 효과를 나타냄으로써 허혈성 뇌질환 예방 또는 치료제의 유효성분으로 사용될 수 있음을 알아내고 본 발명을 완성하였다.
본 발명의 목적은 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 또는 개선용 건강식품을 제공하는 것이다.
본 발명의 또 다른 목적은 약학적으로 유효한 양의 댕댕이나무열매 추출물을 개체에 투여하는 단계를 포함하는 허혈성 뇌혈관 질환의 치료 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 약학적으로 유효한 양의 댕댕이나무열매 추출물을 개체에 투여하는 단계를 포함하는 산화적 스트레스로부터 신경세포를 보호하는 방법을 제공하는 것이다.
본 발명에 따른 댕댕이나무열매 추출물은 천연식물 추출물로 부작용이 적으며 허혈성 뇌혈관 질환 실험동물 모델에서 신경세포 보호효과를 가져 신경행동학적 결손의 억제와 뇌경색 용적을 감소시키므로 이를 유효성분으로 함유하는 조성물은 뛰어난 허혈성 뇌혈관 질환의 예방 및 치료효과를 가진다.
도 1은 랫트 중간대뇌동맥의 바닥부위를 90분간 폐쇄하여 허혈을 유발하고 24시간 후에, 신경행동학적 검사를 실시한 점수 그래프이다. saline은 대조군이고 SH-1013은 본 발명의 댕댕이나무열매 추출물이다. 0점은 신경행동학적 결손이 관찰되지 않는 정상 상태(normal), 1점은 랫트의 전체 몸을 꼬리로 들었을 때 왼쪽 앞발의 신장이 일어나지 않는 상태(mild), 2점은 손상이 일어난 부위의 반대 방향으로 도는 상태(moderate), 3점은 손상이 일어난 부위의 반대 방향으로 휴식시에 쓰러지거나 자발적인 운동 활동이 일어나지 않는 상태(severe)이다.
도 2는 MCAO(middle cerebral artery occlusion)를 이용하여 본 발명의 댕댕이나무열매 추출물의 경색 용적에 대한 효과를 나타내는 그래프이다.
도 3은 MCAO(middle cerebral artery occlusion)를 이용하여 본 발명의 댕댕이나무열매 추출물의 경색 용적에 대한 효과를 나타낸 대표적인 TTC-염색된 조직 사진이다: 각 실험군에 대하여 6개의 절편(2mm의 두께)으로 뇌의 두정부위를 전두극(frontal pole)으로부터 4mm 및 16mm 사이로 절단하였다.
도 4는 MCAO(middle cerebral artery occlusion)를 이용하여 본 발명의 댕댕이나무열매 추출물의 신경세포 손상에 대한 효과를 나타낸 대표적인 크레실 바이올렛(cresyl violet)으로 염색된 조직 사진이다. 각 실험군에 대하여 대뇌피질(cerebral cortex)과 선조체(striatum)에서 크레실 바이올렛(cresyl violet)에 염색된 피라밋 모양의 세포를 확인하였다.
이하, 본 발명에 대하여 상세히 설명한다.
상기 목적을 달성하기 위한 일 실시양태에 의하면, 본 발명은 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 및 치료용 조성물을 제공한다.
본 발명에서 '댕댕이나무(Lonicera caerulea L. var. edulis)'는 천연, 잡종 또는 변종 댕댕이나무의 모든 기관, 예를 들어, 뿌리, 가지, 줄기, 잎, 꽃 및 열매를 모두 포함할 수 있으며, 바람직하게는 댕댕이나무의 열매이다.
본 발명에서 '허혈성 뇌혈관 질환'이란 뇌의 혈관이 어떠한 이유로 막혀 뇌에 필요한 혈액이 공급되지 않아 발생하는 질환을 총칭하는 용어를 의미하며, 뇌허혈 질환이라고도 한다. 상기 허혈성 뇌혈관 질환이란 뇌의 혈류가 감소되어 산소와 포도당의 공급이 정상적으로 이루어지지 않아, 뇌세포 중 뇌허혈에 민감하다고 알려져 있는 조직의 신경세포사가 유발되어 발생 되는 질환을 말한다. 허혈성 뇌혈관 질환은 뇌경색, 뇌졸중, 뇌내출혈, 지주막하 출혈, 백질 이상증, 혈관성 치매 등을 포함할 수 있다.
본 발명의 허혈성 뇌혈관 질환은 바람직하게는 혈관성 치매, 뇌경색, 뇌졸중, 뇌내출혈, 지주막하 출혈 및 백질 이상증으로 이루어진 군으로부터 선택된 어느 하나인 것일 수 있다.
본 발명의 댕댕이나무열매 추출물은 신경세포 보호효과를 가지는 것이 바람직하나 이에 한정되지는 않는다.
본 발명에서 '추출'이란 추출물을 얻기 위한 일련의 과정을 의미하며, 추출액이란 생약을 적절한 침출액으로 짜내고 침출액을 증발시켜 농축한 제제를 의미하는 것으로, 이에 제한되지는 않으나, 추출처리에 의해 얻어지는 추출액, 추출액의 희석액 또는 농축액, 추출액을 건조하여 얻어지는 건조물, 이들의 조정제물 또는 정제물일 수 있다. 본 발명의 추출물은 추출용매로 추출하거나 추출용매로 추출하여 제조한 추출물에 분획용매를 가하여 분획함으로써 제조할 수 있다. 상기 추출용매는 이에 제한되지는 않으나, 물, 유기용매 또는 이들의 혼합용매 등을 사용할 수 있으며, 상기 유기용매는 탄소수 1 내지 4의 알코올이나, 에틸아세테이트 또는 아세톤 등의 극성용매, 헥산 또는 디클로로메탄의 비극성용매 또는 이들의 혼합용매를 사용할 수 있다. 또한, 바람직하게는 물, 탄소수 1 내지 4의 알코올 또는 이들의 혼합용매를 사용할 수 있다. 상기 댕댕이나무열매 추출물은 당업계에 공지된 일반적인 추출방법, 분리 및 정제방법을 이용하여 제조할 수 있다. 상기 추출방법으로는, 이에 제한되지는 않으나, 바람직하게 열탕 추출, 열수 추출, 냉침 추출, 환류 냉각 추출 또는 초음파 추출 등의 방법을 사용할 수 있다.
본 발명의 댕댕이나무열매 추출물은 하기 단계들을 포함하는 제조방법에 의해 제조되는 것이 바람직하나 이에 한정되지는 않는다.
본 발명의 댕댕이나무열매추출물은 1) 댕댕이나무열매에 알콜 또는 알콜수용액을 가해 교반한 후, 80 내지 100℃를 유지하는 추출온도에서 3 내지 5시간 동안 환류 추출하는 단계; 2) 상기 단계 1)에서 환류 추출한 용액의 여액을 분리하고, 40 내지 70℃로 감압농축한 후, 동결 건조시키는 단계를 포함하는 방법으로 제조될 수 있다.
상기 단계 1)에서 추출 용매로 물, 알코올 또는 이들의 혼합물, 바람직하게는 C1 내지 C4의 저급 알코올 또는 이들의 혼합 용매로부터 선택된 용매, 1,3-부틸렌 글리콜(1,3-Butylene glycol)로 이루어진 군으로부터 선택된 어느 하나를 사용하는 것이 바람직하나, 이에 한정되지 않고, 1,3-부틸렌 글리콜을 이용하여 추출하는 것이 더욱 바람직하나, 이에 한정되지 않는다. 가장 바람직하게는 추출용매는 1 내지 100%의 에틸 알콜 또는 1 내지 100%의 메틸 알콜이나 이에 한정되지는 않는다.
상기 단계 1)에서 추출 용매의 양은 댕댕이나무열매 건조 중량의 10 중량% 내지 40 중량%로 함이 바람직하고, 30 중량%로 하는 것이 더욱 바람직하나, 이에 한정되지 않는다.
상기 단계 1)에서 추출시 온도는 10℃ 내지 100℃ 이나, 이에 한정되지 않고, 80 내지 100℃가 바람직하고 더욱 바람직하게는 90 내지 95℃이나 이에 한정되지 않는다.
상기 단계 1)에서 추출 시간은 1시간 내지 7시간인 것이 바람직하고, 3시간 내지 5시간인 것이 더욱 바람직하나 이에 한정되지 않으며, 3시간인 것이 가장 바람직하나, 이에 한정되지 않는다. 본 발명에 따른 방법에 있어서, 추출 회수는 1회 내지 5회인 것이 바람직하나 이에 한정되는 것은 아니다.
상기 추출물을 제조하는 방법은 열수 추출, 침지 추출, 환류 냉각 추출, 초임계추출, 아임계추출, 고온추출, 고압추출 또는 초음파추출법 등의 추출장치를 이용한 방법 또는 XAD 및 HP-20을 포함한 흡착 수지를 이용하는 방법 등 당업계의 통상적인 추출방법을 사용할 수 있으며, 저온에서 초음파 추출하는 것이 바람직하나, 이에 한정하는 것은 아니다.
본 발명에서 용어, '예방'은 상기 추출물 또는 상기 추출물을 유효성분으로 포함하는 조성물의 투여로 허혈성 뇌혈관 질환의 발병을 억제 또는 지연시키는 모든 행위를 의미하며, '치료'는 상기 추출물 또는 상기 추출물을 유효성분으로 포함하는 조성물의 투여로 허혈성 뇌혈관 질환의 증세가 호전되거나 이롭게 되는 모든 행위를 의미한다.
본 발명의 일 실시예에 있어서, 랫트를 마취시켜 중간대뇌동맥의 바닥부위를 폐쇄하여 뇌허혈을 유발시킨 허혈성 뇌혈관 질환 실험동물 모델에 본 발명의 댕댕이나무열매 추출물을 처치하여 허혈성 뇌혈관 질환 치료에의 용도를 평가하였다.
본 발명의 일 실시예에 있어서, 실험동물의 신경행동학적 결손을 신경학적 점수를 매겨 평가한 결과 신경학적 점수가 대조군에 비해 현저히 낮음을 확인하였다.
본 발명의 일 실시예에 있어서, 경색 용적에 대한 본 발명의 댕댕이나무열매 추출물의 효능을 평가한 결과, 경색 용적은 100mg/kg의 댕댕이나무열매 추출물에서 현저히 줄어듬을 확인하였다.
또한, 본 발명의 일 실시예에 있어서, 신경세포를 염색하여 효능을 측정하는 실험에서 댕댕이나무열매 추출물에서 현저하게 개선된 뇌허혈에 의한 신경세포사 억제 효과가 있는 것으로 확인되었고, 현저하게 우수한 신경세포 보호효능을 가지고 있는 것으로 확인되었다.
따라서, 본 발명에 따른 댕댕이나무열매 추출물은 신경세포 보호효과를 가져 허혈성 뇌혈관 질환에 있어서 신경행동학적 결손 및 경색 용적을 현저하게 감소시키므로 본 발명은 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 및 치료용 조성물을 제공한다.
본 발명의 구체적인 실시예에서, 본 발명에 따른 추출물은 천연식물 추출물로 부작용이 없으며 허혈성 뇌혈관 질환 실험동물 모델에서 신경세포 보호효과를 가져 신경행동학적 결손의 억제와 뇌경색 용적을 감소시키므로 이를 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 또는 치료용 약학적 조성물로 유용하게 이용될 수 있다.
상기 본 발명의 추출물을 포함하는 조성물은, 조성물 총 중량에 대하여 상기 추출물을 0.1 내지 50 중량%로 포함하는 것이 바람직하나 이에 한정되지 않는다.
본 발명의 조성물은 약제의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다.
본 발명에 따른 조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명의 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 본 발명의 조성물에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 조성물은 경구 또는 비경구로 투여될 수 있으며, 비경구 투여법이라면 어느 것이나 사용 가능하고, 전신 투여 또는 국소 투여가 가능하나, 전신 투여가 더 바람직하며, 정맥 내 투여가 가장 바람직하다.
본 발명의 치료용 조성물의 바람직한 투여량은 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 본원발명이 속하는 기술분야에서 통상의 지식을 가진 자에 의해 적절하게 선택될 수 있다. 바람직하게는 본 발명의 치료용 조성물은 댕댕이나무열매 추출물의 양을 기준으로 1일 0.0001 내지 1000mg/kg으로, 보다 효과적이기 위해서는 0.01 내지 100mg/kg으로 투여하는 것이 바람직하다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량과 투여횟수는 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 및 치료용 약학적 조성물은 거환(bolus) 형태 혹은 상대적으로 짧은 기간 동안 확산(infusion) 등에 의해 단일 투여량(single dose)으로 환자에게 투여될 수 있으며, 다중 투여량(multiple dose)이 장기간 투여되는 분할 치료 방법(fractionated treatment protocol)에 의해 투여될 수 있다. 본 발명의 약학적 조성물의 투여 농도는 약의 투여 경로 및 치료 횟수 뿐만 아니라 환자의 나이 및 건강상태 등 다양한 요인들을 고려하여 환자의 유효 투여량이 결정되는 것이므로, 이러한 점을 고려할 때, 이 분야의 통상적인 지식을 가진 자라면 적절한 유효 투여량을 결정할 수 있다.
본 발명의 조성물은 댕댕이나무열매 추출물 외에 영양제, 비타민, 전해질, 풍미제, 착색제, 중진제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 추가로 함유할 수 있다. 상기 조성물에는 상기 성분들이 독립적으로 또는 조합하여 추가될 수 있다. 상기 추가성분의 함량은 바람직하게는 상기 댕댕이나무열매 추출물 100 중량부 당 0.1 내지 20 중량부 범위에서 추가할 수 있다
본 발명의 약학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 산제, 과립제, 정제, 캅셀제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 연고, 크림등의 외용제, 좌제 및 멸균 주사용액 등을 비롯하여 약학적 제제에 적합한 어떠한 형태로든 사용할 수 있다.
또 하나의 양태로서, 본 발명은 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 또는 개선용 건강식품을 제공한다.
본 발명에서 댕댕이나무열매 추출물, 허혈성 뇌혈관 질환 등은 상기에서 설명한 바와 같다.
본 발명의 구체적인 실시예에서, 본 발명에 따른 추출물은 허혈성 뇌혈관 질환 실험동물 모델에서 신경세포 보호효과를 가져 신경행동학적 결손의 억제와 뇌경색 용적을 감소시키므로 이를 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 또는 개선용 건강식품으로 유용하게 이용될 수 있다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 물질을 첨가할 수 있는 식품의 예로는 드링크제, 육류, 소세지, 빵, 비스켓, 떡, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 알콜 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강 식품을 모두 포함한다.
본 발명의 댕댕이나무열매 추출물은 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합양은 그의 사용 목적(예방 또는 개선용)에 따라 적합하게 결정될 수 있다. 일반적으로, 건강식품 중의 상기 추출물의 양은 전체 식품 중량의 0.1 내지 90 중량부로 가할 수 있다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
본 발명의 건강 기능성 음료 조성물은 지시된 비율로 필수 성분으로서 상기 추출물을 함유하는 외에는 다른 성분에는 특별한 제한이 없으며 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 상술한 것 이외의 향미제로서 천연 향미제(타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100 ㎖당 일반적으로 약 1 내지 20g, 바람직하게는 약 5 내지 12 g이다.
상기 외에 본 발명의 추출물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 추출물은 천연 과일 쥬스 및 과일 쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 그렇게 중요하진 않지만 본 발명의 추출물 100 중량부 당 0.1 내지 약 20 중량부의 범위에서 선택되는 것이 일반적이다.
또 하나의 양태로서, 본 발명은 약학적으로 유효한 양의 댕댕이나무열매 추출물을 유효성분으로 함유하는 조성물을 개체에 투여하는 단계를 포함하는 허혈성 뇌혈관 질환의 예방 또는 치료 방법을 제공한다.
본 발명에서 용어, '투여'는 어떠한 적절한 방법으로 환자에게 소정의 물질을 도입하는 것을 의미하며, 상기 조성물의 투여 경로는 특별히 이에 제한되지 않으나, 상기 조성물이 생체내 표적에 도달할 수 있는 어떠한 일반적인 경로를 통하여 투여될 수 있으며, 예를 들어 복강 내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비내 투여, 폐내 투여, 직장 내 투여 등이 될 수 있다. 그러나 경구 투여 시, 펩타이드는 소화가 되기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화하는 것이 바람직하다.
또 하나의 양태로서, 본 발명은 약학적으로 유효한 양의 댕댕이나무열매 추출물을 유효성분으로 함유하는 조성물을 개체에 투여하는 단계를 포함하는 산화적 스트레스로부터 신경세포를 보호하는 방법을 제공한다.
본 발명에서 '산화적 스트레스'란 활성산소 생산계 및 소거계의 균형이 약제, 방사선, 허혈 등 여러 가지 요인에 의해 이러한 균형이 무너진 상태를 말한다. 세균에서는 활성산소에 의해 유도되는 일군의 효소가 있으며, 전사인자가 직접 산화에 의해 활성화 되는 것이 원인이 된다. 고등생물에서도 많은 유전자가 산화적 스트레스에 의해 유도되지만, 세포 내 신호전달계가 활성화한 결과로 야기되는 것으로 생각되고 있다. 산화적 스트레스는 결과적으로 세포상해(특히 세포막 지질과 산화), DNA염기 수식에 의한 돌연변이, 이폽토시스(apotosis) 또는 괴사의 유도 등으로 병적상태를 유발하기도 한다.
상기의 산화적 스트레스는 활성산소종 또는 활성질소종으로부터 야기될 수 있다. 상기에서 활성산소종이란 삼중항산소(3O2)의 단계적 환원으로부터 생성되는 슈퍼옥시드(O2-), 과산화수소(H2O2),히드록시라디칼(OH), 및 일중항산소(1O2)를 가리키며, 활성질소종에는 일산화질소 라디칼(NO), 및 퍼옥시 아질산염(ONOO) 등이 있다.
또 다른 양태로서, 본 발명은 허혈성 뇌혈관 질환의 예방 또는 치료용 의약의 제조에 있어서, 댕댕이나무열매 추출물의 용도를 제공한다.
본 발명에서 댕댕이나무열매 추출물, 허혈성 뇌혈관 질환 등은 상기에서 설명한 바와 같다.
이하, 실시예, 실험예, 및 제조예에 의해 본 발명을 상세히 설명한다.
단, 하기 실시예, 실험예 및 제조예는 본 발명을 예시하는 것일 뿐, 본 발명 내용이 하기 실시예, 실험예, 및 제조예에 한정되는 것은 아니다.
<실시예 1> 댕댕이나무열매 추출물의 제조
<1-1> 물 용매를 이용한 열수추출법
중국의 연변지역 및 백두산에서 직접 수확한 야생 댕댕이나무열매를 건조시켜 댕댕이나무열매 추출물의 제조에 사용하였다. 분쇄한 상태의 댕댕이나무열매 100 g을 1 리터의 증류수에 가하여 잘 교반한 다음, 90 내지 95℃를 유지하는 추출온도에서 3시간 동안 환류 추출한 후 여액을 분리하였고 55~65℃로 생약추출물을 감압농축한 후, 동결 건조시켜 생약조성물 분말 엑스 21.2g을 얻었다.
<1-2> 물-알콜 혼합용매를 이용한 열수추출법
상기 실시예 1-1과 같이 분쇄된 댕댕이나무열매 100 g에 1 리터의 25% 에틸 알콜을 가해 잘 교반한 다음, 열을 가해 80 내지 90℃를 유지하는 추출온도에서 3시간 동안 환류 추출한 후 여액을 분리하였고 55~65℃에서 생약추출물을 감압농축한 후, 동결 건조시켜 생약조성물 분말 엑스 19.5g을 얻었다.
<실시예 2> 뇌허혈 모델 제작 (Focal ischemic model)
실험동물로는 Sprague-Dawley(SD) 랫트(250-300 g)를 사용하였다. SD랫트를 24 시간 절식시킨 후, 질소와 산소가 7:3으로 혼합된 가스에 2.5% 이소플루란(isoflurane)으로 마취 상태에서 온목동맥과 바깥목동맥은 결찰하고, 속목동맥의 분지점으로부터 17 mm의 프루브(probe)를 삽입하여 90 분간 중간대뇌동맥의 바닥부위를 폐쇄하여 허혈을 유발하였다. 허혈을 유발시키는 동안 직장 내 체온계를 삽입하여 체온을 측정하고 정상체온인 37±0.3℃로 일정하게 유지하였다. 일정시간 동안 중간대뇌동맥을 폐쇄한 후, 속목동맥에 삽입된 프루브를 밖으로 뽑아내어 재관류를 수행하였다.
<실시예 3> 허혈성 뇌손상 정도 평가
<3-1> 2,3,5-Triphenyltetrazolium chloride (TTC) staining
일시적 뇌허혈에 의해 유발되는 뇌조직 손상을 일차적으로 관찰하기 위하여 TTC 염색을 시행하였다. 실험군의 뇌를 적출하여 뇌 매트릭스(brain matrix)를 이용하여 2mm 두께의 절편을 만들고, 2% 2,3,5-Triphenyltetrazolium chloride (TTC) 용액으로 37℃에서 60 분간 반응시킨 후, TTC 용액으로 염색된 뇌 절편을 10% 중성 완충 포르말린(neutral buffered formalin)용액에 고정하여 경색용적(infarct volume)을 측정하였다(Lee et al., Glia, 50:168-181, 2005)
<3-2>조직학적 염색
정상군과 각 실험군은 허혈 후 정해진 시간에 클로랄 히드레이트(chloral hydrate) (400 mg/)를 복강 (intraperitoreum)내 주사하여 전신마취 시킨 후 실험동물의 흉곽을 열고 좌심실을 통해 카눌라(cannula)를 오름대동맥(ascending aorta)에 삽입한 다음, 우심방을 통하여 방혈시킴과 동시에 주입기(Cole Parmer, USA)를 이용하여 생리식염수(pH 7.4)로 분당 10 ml의 속도로 관류 세척하였다. 이어서 4% 파라포름알데하이드(paraformaldehyde)[0.1 M phosphate buffer (PB), pH 7.4] 용액으로 전신을 관류 고정한 후 골절단기를 이용하여 머리뼈를 절개하여 뇌를 적출한다. 고정된 뇌 조직을 통상적인 조직처리 과정을 거쳐서 파라핀 포매를 한 다음 박절기(microtome, Reichert-Jung, Germany)를 이용하여 5㎛ 두께의 파라핀 연속절편을 제조하였다. 자일렌(Xylene)으로 파라핀을 제거하고 100%, 95%, 90%, 80%, 70% 및 50% 에탄올을 거쳐 정상 세포의 분포 및 변성 여부를 확인하기 위하여 크레실 바이올렛(cresyl violet)과 산성 푸신(acid fuchsin) 염색을 시행한 다음 통상적인 탈수 및 투명과정을 거쳐 퍼마운트(permount)로 봉입하였다.
<실험예 1> 신경행동학적 관찰 점수 분석
뇌허혈 유발 후 3 시간이 경과 했을 때 수술한 SD 랫트의 신경행동학적 결손을 관찰하였다. 신경행동학적 검사는 공중에서 랫트의 꼬리를 완전히 들었을 때, 왼쪽의 국부마비(hemiparesis)가 일어나는 정도와 꼬리를 위로 들었을 때, 또는 자발적으로 왼쪽으로의 선회(circling)가 일어나는 지를 관찰하여 확인하였다. 신경학적 점수에서 0점은 신경행동학적 결손이 관찰되지 않는 정상 상태(normal), 1점은 랫트의 전체 몸을 꼬리로 들었을 때, 왼쪽 앞발의 신장이 일어나지 않는 상태(mild), 2점은 손상이 일어난 부위의 반대 방향으로 도는 상태(moderate), 3점은 손상이 일어난 부위의 반대 방향으로 휴식시에 쓰러지거나 자발적인 운동 활동이 일어나지 않는 상태(severe)이다.
뇌허혈 유발 후 약 30분 및 3시간 경과 했을 때, 랫트의 꼬리를 완전히 들었을 때 반시계 방향으로의 선회가 일어나는지를 확인하였다. 95% 이상의 랫트에서 반시계 방향의 선회가 관찰되었고, 신경학적 점수(neurological score)는 2.8±0.28로 나타났으나, 댕댕이나무열매 추출물 투여군에서 자발적인 선회가 감소하였으며, 신경학적 점수는 댕댕이나무열매 추출물(SH-1013)(100 mg/kg) 투여군에서는 1.48±0.23으로 유의성 있는 신경행동학적 결손이 억제되었다(도 1).
이상의 결과로 본 발명의 댕댕이나무열매 추출물은 허혈성 뇌혈관 질환 실험동물 모델에서 신경행동학적 결손을 현저하게 감소시키는 것을 알 수 있다.
<실험예 2> 허혈성 뇌손상에 대한 보호약물의 효과
일시적인 (transient) MCAO(middle cerebral artery occlusion)에 의한 뇌손상을 일차적으로 평가하기 위하여 TTC 용액으로 염색하고, 경색용적(total infacrt volume)을 측정하였다. 정상조직은 미토콘드리아 효소들이 TTC를 환원시킴으로써 진한 적색으로 염색되나, 손상된 조직은 미토콘드리아 손상에 의해 환원력을 소실함으로써 정상조직과 구분되어 진다. 각 실험군 뇌의 후각망울을 제외하고, 앞쪽으로부터 2 mm 두께로 연속 절단한 6개 절편을 TTC 염색한 결과 2, 3 및 4번째 조직에서 뚜렷한 경색부위가 관찰되었다. 전체 경색용적은 허혈 24시간 군에서 25.38±2.85% 으로 측정되었으나, 댕댕이나무열매 추출물(SH-1013) 투여군에서 전체 경색용적(8.59±4.68%)이 유의성 있게 줄어들었다. 이는 60%이상 뇌경색 용적이 댕댕이나무 열매추출물 투여군에서 감소됨을 확인하였다(도 2, 3).
또한, 두 시간 중간대뇌동맥의 결찰 후 24 시간동안 재관류 후 크레실 바이올렛(cresyl violet) 염색 결과 허혈 손상부위의 대뇌피질 및 선조체에서 크레실 바이올렛(cresyl violet)에 염색된 세포들의 수가 감소하였으며, 특히, 피라밋 모양의 신경세포들은 관찰되지 않았다. 본 발명의 댕댕이나무열매 추출물 투여군에서는 많은 수의 피라밋 모양의 신경세포들이 허혈 손상부위의 대뇌피질 및 선조체에서 관찰되었다(도 4).
이상의 결과로 본 발명의 댕댕이나무열매 추출물은 허혈성 뇌혈관 질환 실험동물 모델에서 경색 용적을 현저하게 감소시키고 신경세포 보호효과가 현저하다는 것을 알 수 있다.
<제조예 1> 약학적 제제의 제조
<1-1> 산제의 제조
댕댕이나무열매 추출물 20 ㎎
유당 20 ㎎
상기의 성분을 혼합한 후, 기밀포에 충진하여 산제를 제조하였다.
<1-2> 정제의 제조
댕댕이나무열매 추출물 10 ㎎
옥수수전분 100 ㎎
유 당 100 ㎎
스테아린산 마그네슘 2 ㎎
상기의 성분을 혼합한 후, 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조하였다.
<1-3> 캡슐제의 제조
댕댕이나무열매 추출물 10 ㎎
결정성 셀룰로오스 3 ㎎
락토오스 14.8 ㎎
스테아린산 마그네슘 0.2 ㎎
상기의 성분을 혼합한 후, 통상의 캡슐제의 제조방법에 따라서 젤라틴 캡슐에 충전하여 캡슐제를 제조하였다.
<1-4> 액제의 제조
댕댕이나무열매 추출물 20 ㎎
이성화당 10 g
만니톨 5 g
정제수 적량
통상의 액제의 제조방법에 따라 정제수에 각각의 성분을 가하여 용해시키고 레몬향을 적량 가한 다음 상기의 성분을 혼합한 다음 정제수를 가하여 전체를 정제수를 가하여 전체 100 ㎖로 조절한 후 갈색병에 충진하여 멸균시켜 액제를 제조한다.
<1-5> 주사제의 제조
댕댕이나무열매 추출물 10 ㎍/㎖
묽은 염산 BP pH 7.6로 될 때까지
주이용 염화나트륨 BP 최대 1 ㎖
적당한 용적의 주이용 염화나트륨 BP 중에 댕댕이나무열매 추출물을 용해시키고, 생성된 용액의 pH를 묽은 염산 BP를 이용하여 pH 7.6로 조절하고, 주이용 염화나트륨 BP를 이용하여 용적을 조절하고 충분히 혼합하였다. 용액을 투명 유리로 된 5 ㎖ 타입 I 앰플 중에 충전시키고, 유리를 용해시킴으로써 공기의 상부 격자하에 봉입시키고, 120℃에서 15 분 이상 오토클래이브(autoclave)시켜 살균하여 주사액제를 제조하였다.
<제조예 2> 식품의 제조
본 발명의 댕댕이나무열매 추출물을 포함하는 식품들을 다음과 같이 제조하였다.
<2-1> 밀가루 식품의 제조
본 발명의 댕댕이나무열매 추출물 0.5 ~ 5.0 중량부를 밀가루에 첨가하고, 이 혼합물을 이용하여 빵, 케이크, 쿠키, 크래커 및 면류를 제조하여 건강 증진용 식품을 제조하였다.
<2-2> 유제품(dairy products)의 제조
본 발명의 댕댕이나무열매 추출물 5 ~ 10 중량부를 우유에 첨가하고, 상기 우유를 이용하여 버터 및 아이스크림과 같은 다양한 유제품을 제조하였다.
<2-3> 선식의 제조
현미, 보리, 찹쌀, 율무를 공지의 방법으로 알파화시켜 건조시킨 것을 배전한 후 분쇄기로 입도 60 메쉬의 분말로 제조하였다.
검정콩, 검정깨, 들깨도 공지의 방법으로 쪄서 건조시킨 것을 배전한 후 분쇄기로 입도 60 메쉬의 분말로 제조하였다.
본 발명의 댕댕이나무열매 추출물을 진공 농축기에서 감압농축하고, 분무, 열풍건조기로 건조하여 얻은 건조물을 분쇄기로 입도 60 메쉬로 분쇄하여 건조분말을 얻었다.
상기에서 제조한 곡물류, 종실류 및 댕댕이나무열매 추출물의 건조분말을 다음의 비율로 배합하여 제조하였다.
곡물류(현미 30 중량부, 율무 15 중량부, 보리 20 중량부),
종실류(들깨 7 중량부, 검정콩 8 중량부, 검정깨 7 중량부),
댕댕이나무열매 추출물(3 중량부),
영지(0.5 중량부), 및
지황(0.5 중량부).
<2-4> 건강보조식품의 제조
댕댕이나무열매 추출물 100 ㎎
비타민 혼합물 적량
비타민 A 아세테이트 70 ㎍
비타민 E 1.0 ㎎
비타민 B1 0.13 ㎎
비타민 B2 0.15 ㎎
비타민 B6 0.5 ㎎
비타민 B12 0.2 ㎍
비타민 C 10 ㎎
비오틴 10 ㎍
니코틴산아미드 1.7 ㎎
엽산 50 ㎍
판토텐산 칼슘 0.5 ㎎
무기질 혼합물 적량
황산제1철 1.75 ㎎
산화아연 0.82 ㎎
탄산마그네슘 25.3 ㎎
제1인산칼륨 15 ㎎
제2인산칼슘 55 ㎎
구연산칼륨 90 ㎎
탄산칼슘 100 ㎎
염화마그네슘 24.8 ㎎
상기의 비타민 및 미네랄 혼합물의 조성비는 비교적 건강식품에 적합한 성분을 바람직한 실시예로 혼합 조성하였지만, 그 배합비를 임의로 변형 실시하여도 무방하며, 통상의 건강식품 제조방법에 따라 상기의 성분을 혼합한 다음, 과립을 제조하고, 통상의 방법에 따라 건강식품 조성물 제조에 사용할 수 있다.
<제조예 3> 건강음료의 제조
댕댕이나무열매 추출물 100 ㎎
구연산 100 ㎎
올리고당 100 ㎎
매실농축액 2 ㎎
타우린 100 ㎎
정제수를 가하여 전체 500 ㎖
통상의 건강음료 제조방법에 따라 상기의 성분을 혼합한 다음, 약 1시간 동안 85℃에서 교반 가열한 후, 만들어진 용액을 여과하여 멸균된 1 ℓ용기에 취득하여 밀봉 멸균한 뒤 냉장 보관한 다음 본 발명의 건강음료 조성물 제조에 사용한다.
상기 조성비는 비교적 기호 음료에 적합한 성분을 바람직한 실시예로 혼합 조성하였지만, 수요계층, 수요국가, 사용 용도 등 지역적, 민족적 기호도에 따라서 그 배합비를 임의로 변형 실시하여도 무방하다.
이상의 본 발명은 상기에 기술된 실시예, 실험예 및 제조예들에 의해 한정되지 않고, 당업자들에 의해 다양한 변형 및 변경을 가져올 수 있으며, 이는 첨부된 청구항에서 정의되는 본 발명의 취지와 범위에 포함된다.
Claims (8)
- 댕댕이나무(Lonicera caerulea L. var. edulis)열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 허혈성 뇌혈관 질환은 혈관성 치매, 뇌경색, 뇌졸중, 뇌내출혈, 지주막하 출혈 및 백질 이상증으로 이루어진 군으로부터 선택된 어느 하나인 허혈성 뇌혈관 질환의 예방 또는 치료용 약학적 조성물.
- 댕댕이나무열매 추출물을 유효성분으로 함유하는 허혈성 뇌혈관 질환의 예방 또는 개선용 건강식품.
- 제3항에 있어서, 상기 허혈성 뇌혈관 질환은 혈관성 치매, 뇌경색, 뇌졸중, 뇌내출혈, 지주막하 출혈 및 백질 이상증으로 이루어진 군으로부터 선택된 어느 하나인 허혈성 뇌혈관 질환의 예방 또는 개선용 건강식품.
- 약학적으로 유효한 양의 댕댕이나무열매 추출물을 유효성분으로 함유하는 조성물을 개체에 투여하는 단계를 포함하는 허혈성 뇌혈관 질환의 치료 방법.
- 제5항에 있어서, 상기 허혈성 뇌혈관 질환은 혈관성 치매, 뇌경색, 뇌졸중, 뇌내출혈, 지주막하 출혈 및 백질 이상증으로 이루어진 군으로부터 선택된 어느 하나인 허혈성 뇌혈관 질환의 치료 방법.
- 약학적으로 유효한 양의 댕댕이나무열매 추출물을 유효성분으로 함유하는 조성물을 개체에 투여하는 단계를 포함하는 산화적 스트레스로부터 신경세포를 보호하는 방법.
- 제7항에 있어서, 상기 산화적 스트레스는 활성산소종 또는 활성질소종으로부터 야기되는 것인 신경세포를 보호하는 방법.
