이에, 본 발명자들은 KGF1의 안정성을 높이고자 예의 노력한 결과, KGF1 단백질의 안정성을 증가시키기 위하여 단백질의 아미노산 서열에 변형을 주는 방식의 분자설계 방식을 적용하여, 열 안정성이 향상된 후보군을 도출하고, 도출된 KGF1 변이체가 수용액 상태에서의 우수한 안정성을 가지는 것을 확인하고, 본 발명을 완성하게 되었다.

본 발명에서 사용되는 용어 "각질세포 성장인자-1" 또는 "KGF1"은 140개 아미노산으로 구성된 단백질로, 'FGF7(fibroblast growth factor 7)'으로도 알려져 있으며, 상처 치유의 상피화 단계에서 나타나는 성장인자로, 이 단계에서 각질 세포는 상처를 덮고 상피를 형성하게 된다. KGF는 섬유아세포 성장 인자 수용체 2b(FGFR2b)에 결합하는 작은 신호 분자로, 신호가 발생하려면 헤파린 분자에 의해 함께 연결된 두 FGF:FGFR 복합체 사이에 이량체가 필요하며, 23개의 알려진 FGF와 4개의 FGF 수용체가 있다. FGF:FGFR 결합은 복잡하고 조직 특이적 방식으로 다양한 메커니즘에 의해 조절된다.
KGF1은 진피에서 발현되고 주변 분비 기전을 통해 표피 증식 및 분화를 조절하여 상처 치유 및 모발 성장을 자극하는 것으로 확인되고 있으며, 실제로, 누드 마우스에 KGF1을 주사하면 주사 부위에서의 모발 성장을 직접 자극하는 것으로 알려져 있다(Guo et al.,EMBO J. 12:973, 1993).
본 발명의 변이체는 KGF1의 3차 구조와 종간 상동성정렬(homology alignment) 방법 및 컴퓨터를 이용한 단백질 분자 모델링을 통해, KGF1의 활성부위와 관련 없는 부위를 선정하고, 변이실험을 통해 제조된 변이체로서, KGF1의 아미노산 잔기를 시스테인(cysteine)으로 치환시켜 이황화결합을 추가적으로 생성시킴으로서 루프 엔트로피를 감소시키는 방법으로 안정성이 증가된 것을 특징으로 한다.

본 발명의 일 구현예에 있어서 상기 유전자는 서열번호 2 내지 46으로 구성된 군으로부터 선택된 하나의 DNA 염기 서열로 이루어진 것이 바람직하나 상기 서열에 하나 이상의 치환, 결손, 역위, 전좌 등의 돌연변이를 수행하여 본 발명에서 목적하고자 하는 효과를 달성하는 모든 돌연변이체도 본 발명의 범위에 포함된다.
본 발명의 고안정성 KGF1 변이체는 단백질 활성은 유지하면서 열에 대한 안정성이 천연형에 비해 증가한다. 본 발명의 일 시험예에서는 고안정성 KGF1 변이체는 천연형과 동등한 활성을 지니고 있으며, 열에 대한 안정성이 역시 현저히 증가되었음을 확인하였다.
또한, 본 발명은 상기 유전자를 포함하는 발현벡터를 제공한다.
또한, 본 발명은 상기 유전자 또는 상기 발현 벡터에 의해 형질전환된 형질전환체를 제공한다.
본 발명은 또한, 상기 고안정성 KGF1 변이체를 유효성분으로 함유하는는 탈모의 예방 또는 치료용 약학적 조성물을 제공한다.

본 발명의 고안정성 KGF1 변이체는 부위특이 돌연변이 유도법 등에 의해 제조한 고안정성 KGF1 변이체를 코딩하는 유전자를 함유하는 벡터로 숙주세포를 형질 전환시켜 고안정성 KGF1 변이체를 발현시키는 방법으로 제조할 수 있으며, 또한 화학적 아미노산 합성 방법에 의해 제조될 수 있다.
고안정성 KGF1 변이체를 코딩하는 DNA는 천연형 KGF1의 상기 치환되는 부위의 아미노산을 코딩하는 DNA이다.
한편, 코돈의 퇴화(degeneracy)에 의해 하나의 아미노산을 코딩하는 코돈이 다수 존재하므로 동일한 아미노산 서열을 코딩하는 DNA라도 그 뉴클레오티드 서열이 다를 수 있음은 주지된 사실이다.
이와 같은 고안정성 KGF1 변이체를 코딩하는 DNA는 화학적으로 합성하거나 천연형 KGF1 cDNA를 제조하여 이를 바탕으로 부위특이 돌연변이 유도법 등의 방법을 사용하여 제조할 수도 있다.
상기 제조된 본 발명의 고안정성 KGF1 변이체를 코딩하는 DNA는 당 분야에 잘 공지된 적당한 원핵 또는 진핵 발현 시스템중 어느 하나를 사용하여 이를 발현시킬 수 있다(Sambrook et al., Molecular Cloning, A Laboratory Manual, 2nd ed., Cold Spring Habor Laboratory, Cold Spring Habor Laboratory Press, USA, 1989).
발현은 당화되지 않은 고안정성 KGF1 변이체의 경우 바람직하게는 대장균, 예를 들어, 대장균 BL21(DE3), 대장균JM109(DE3), 대장균 NM522 등에서 수행되며, 대장균에서의 발현을 위해 사용될 수 있는 적당한 벡터들은 샘브룩 등의 문헌(상동) 및 피어스(Fiers)등의 논문("Proced. 8th Int. Biotechnology Symposium", Soc. Frac, de Microbiol., Paris,(Durand et al., eds.), pp. 680-697, 1988)에 언급되어 있다.
상술한 벡터에 의한 숙주세포의 형질전환은 통상적인 방법 중 어느 하나에 의해 수행될 수 있다(Sambrooketal., Molecular Cloning, ALaboratory Manual, 1989; Ito et al., J. Bacteriol. 153:263, 1983).
대장균을 형질전환시킬 경우, DNA를 흡수할 수 있는 콤피턴트 세포(competent cell)를 준비하고, 이어서 공지된 방법 등에 따라 처리할 수 있다.
본 발명의 다른 양태에 따르면, 본 발명은 다음 단계를 포함하는 고안정성 KGF1 변이체 생산 방법을 제공한다:(a) 상기 형질전환체를 배양하는 단계; 및 (b) 상기 단계 (a)에서 얻은 배양액으로부터 변이체를 분리하는 단계.
본 발명의 바람직한 구현예에 따르면, 상기 방법의 (b) 단계는, (c) 상기 형질전환체를 세포 파쇄하고 응집체를 분리하는 단계; (d) 상기 분리된 응집체를 제거하는 단계; (e) 상기 응집체가 제거된 상층액을 이온교환수지 크로마토그래피를 이용하여 분리정제 하는 단계; 및 (f) 상기 이온교환 수지 후 고안정성 KGF1 변이체를 헤파린 친화 크로마토그래피를 이용하여 분리 정제하는 단계를 포함한다.
일반적으로, 목적 발현 벡터를 함유하는 숙주 미생물은 목적 단백질의 생산을 최대화하는 범위에서 그들의 최적성장 조건에서 배양된다. 예를 들면, 엠피실린 저항 유전자를 선택 표지로 함유하는 벡터로 형질 전환된 대장균 Top10 세포는 엠피실린이 함유된 LB 배지에서 37℃로 배양한다.
형질전환된 숙주세포를 배양한 후 생산된 고안정성 KGF1 변이체의 회수 및 정제는 당해 분야에 공지된 여러 방법 또는 그들을 조합하여 사용함으로써 수행할 수 있다. 예를 들면, 형질 전환된 대장균 세포에서 발현된 고안정성 KGF1 변이체는 세포 배양물로부터 또는 세포의 파쇄 후에 단백질 화학계에 공지된 적합한 방법에 의해 추출함으로써 회수될 수 있다.
바람직하게는, 고안정성 KGF1 변이체를 정제하기 위해, 재조합 대장균 세포의 배양액을 원심 분리하여 세포를 수확하고, 수확한 세포를 라이소자임이 첨가된 완충용액에 현탁시키고 초음파로 파쇄 한다. 세포 파쇄 액을 원심 분리하여 불용성 과립형태의 응집체를 분리하고, 상기 분리된 응집체를 제거한다. 상기 응집체가 제거된 상층액을 이온교환수지 크로마토그래피를 이용하여 분리정제하고, 이온교환 수지 후 헤파린 친화 크로마토그래피를 이용하여 분리 정제하여 결과물인 고안정성 KGF1 변이체를 획득한다.
본 발명의 다른 양태에 따르면, 본 발명은 상기 고안정성 KGF1 변이체를 유효성분으로 포함하는 탈모 예방 또는 치료용 약학적 조성물을 제공한다. 하기의 실시예에서 입증된 바와 같이, 본 발명의 고고안정성 KGF1 변이체는 천연 KGF1와 동일한 활성을 갖고, 매우 우수한 열안정성 및 수용액에서의 안정성을 갖는다. 따라서, 본 발명의 조성물은 탈모 예방 또는 치료에 매우 유효하다.
또한, 본 발명은 상기 고안정성 KGF1 변이체를 유효성분으로 함유하는 창상, 구내염 또는 피부염증 치료용 약학적 조성물을 제공한다.
바람직하게는, 본 발명의 조성물은 폐쇄창(closed wound) 및 개방창(open wound)의 치료에 이용된다. 폐쇄창의 예는 좌상(contusion or Burise)을 포함하고, 개방창의 예는 찰과상(abrasion), 열상(laceration), 결출상(Avulsion), 관통상(penetrated wound) 및 총상(gun shot wound)을 포함한다.
본 발명의 조성물은 (a) 상술한 본 발명의 고안정성 KGF1 변이체의 약학적 유효량; 및 (b) 약학적으로 허용되는 담체를 포함하는 약학적 조성물이다.
본 명세서에서 사용되는 용어 "약학적 유효량"은 상술한 고안정성 KGF1 변이체의 효능 또는 활성을 달성하는 데 충분한 양을 의미한다.
본 발명의 약학적 조성물에 포함되는 약학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 트레할로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
본 발명의 약학적 조성물은 경구 또는 비경구, 바람직하게는 비경구로 투여할 수 있고, 비경구 투여인 경우에는 정맥내 주입, 피하 주입, 근육 주입, 복강 주입, 국소 투여, 경피 투여 등으로 투여할 수 있다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 한편, 본 발명의 약제학적 조성물의 바람직한 1일 투여량은 0.0001-10mg/kg 체중이다.

본 발명의 제형이 계면-활성제 함유 샴푸 또는 클린징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
본 발명의 화장품 조성물에 포함되는 성분은 유효 성분으로서의 고안정성 KGF1 변이체와 담체 성분 이외에, 화장품 조성물에 통상적으로 이용되는 성분들을 포함하며, 예컨대 항산화제, 안정화제, 용해화제, 비타민, 안료 및 향료와 같은 통상적인 보조제를 포함할 수 있다.
본 발명은 또한, 상기 고안정성 KGF1 변이체를 투여하는 단계를 포함하는, 탈모의 예방 또는 치료 방법을 제공한다.
본 발명은 또한, 상기 고안정성 KGF1 변이체를 투여하는 단계를 포함하는, 창상, 구내염 또는 피부염증의 치료방법을 제공한다.
본 발명은 또한, 탈모의 예방 또는 치료를 위한 상기 고안정성 KGF1 변이체의 용도를 제공한다.

발명의 상세한 설명 및 바람직한 구현예
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 해당 업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예 1: 고안정성 KGF1 변이체 제작을 위한 분자설계
상동성 모델링(Homology modeling)
상동성 모델링은 Modeller(Andrej Sali)를 이용하였다.
에너지 미니마이제이션(Energy minimization)
Energy minimization은 Chimera에 포함된 Amber 99FF force field(University of California, San Francisco, CA, USA)를 사용하였다.
이황화결합 예측(Disulfide predict)
이황화 결합의 형성에 따른 예측은 YASARA Web server를 이용하였다.
이황화결합 거리 측정
이황화결합이 가능한 거리를 측정해주는 Protein contact map visualization (Andreas Viklund.)을 이용하였다.
분자 모델링(Molecular modeling)
PDB(Worldwide Protein Data Bank)에 등록된 단백질의 구조를 이용하여 이황화 결합이 가능 후보군을 설정하고, Protein contact map visualization 프로그램(www.bioinformatics.org)을 이용하여 두 개의 아미노산의 C-alpha 탄소의 거리가 6Å 이하, C-beta 탄소의 거리가 5Å인 잔기를 plot을 이용하여 분석하였다. 이후 Yasara energy minimization server를 이용하여 이황화 결합의 생성유무를 분석하였고 chimera의 AMBER force filed FF99를 이용하여 에너지 최소화(energy minimization)를 진행하였다. 이후 생성된 단백질의 구조를 천연형의 KGF1와 align하여 RMSD를 측정하고, 0.5 이하 값을 갖는 구조를 선별하였다.
실시예 2: 선별된 KGF1 변이체들에 대한 클로닝
DNA 컨스트럭션(construction)
일반적인 E.coli와 yeast 단백질 발현 벡터와 일반적인 발현용 Yeast, E.coli 균주를 사용하였으며, 클로닝용 균주는 E.coli Top10(invitrogen사)균주를 사용하였다. 유전자 재조합 시 사용된 제한효소는 모두 NEB(New England Biolabs)의 제품을 사용하였으며, 리가아제(ligase)는 Roche사의 T4 DNA ligase를 사용하였으며, PCR시 사용된 Ex taq DNA polymerase는 Takara사 제품을 사용하였다. 점 돌연변이에 사용된 pfuUltra DNA polymerase는 Agilent사의 제품을 사용하였으며, DNA gel extraction kit, plasmid mini prep kit는 (주)코스모진텍의 제품을 사용하였다. 또한, 프라이머는 (주)코스모진텍에서 제작하였으며, DNA 시퀀싱 또한, (주)코스모진텍에 의뢰하여 수행하였다.
천연형 사람 KGF1 cDNA를 포함하는 플라스미드의 구축
천연형 KGF1 cDNA를 포함하는 플라스미드를 만들기 위하여, keratinocyte growth factor(GenBank: AVZ66187.1)을 주형으로 PrimeSTAR HS DNA 중합효소(TAKARA사 제품)와 아래 프라이머를 이용하여, PCR법에 의해 해당 DNA 절편을 증폭하여 gel&PCR clean-up kit(코스모진텍사) 를 이용하여 순수하게 분리하였다.

깨끗하게 분리한 DNA 절편과 플라스미드를 제한효소(Xho1, Not1)를 사용하여 절단한 후 ligation(EZ-Fusion_HT_Cloning, enzynomics사 제품)하였고, E.coli Top10에 heat shock으로 형질전환 시켰다. 형질전환 후 고체 배지에 생기는 앰피실린에 저항성이 있는 콜로니를 선별하여 10ml의 LB배지(LB/ampicillin)에 접종하였다. 37℃에서 16시간 동안 배양한 후 DNA prep을 실시하고 얻어진 DNA를 시퀀싱을 통하여 천연형 KGF1 플라스미드를 확인하였다.
변이체 DNA를 포함하는 플라스미드의 구축
천연형의 KGF1 cDNA를 포함하는 플라스미드를 주형으로 PrimeSTAR HS DNA 중합효소와 각각의 변이체들에 해당하는 두 상보적인 프라이머(표 2)를 이용하여 PCR법에 의해 변이체 플라스미드들을 제작하였다, 제작한 변이체 플라스미드를 절단 한 후 E.coli Top10에 heat shock으로 형질전환시켰다. 형질전환 후 고체 배지에 생기는 앰피실린에 저항성이 있는 콜로니를 선별하여 10ml의 LB배지(LB/ampicillin)에 접종하였다. 37℃에서 16시간동안 배양한 후 DNA prep을 실시하고 얻어진 DNA를 시퀀싱을 통하여 KGF1 변이체 플라스미드를 확인하였다. 상기 과정에 사용한 프라이머의 염기서열은 표 2에 나타내었다.
실시예 3: KGF1 변이체 단백질의 생산 및 정제
실시예 2에서 제작한 KGF1 변이체의 발현 플라스미드 각각을 sal1 효소로 절단한 후 electroporation system을 이용하여 Pichia pastoris (GS115)에 형질전환 켜 발현 균주를 만들고, 발현 균주를 25ml의 BMGY 배지에 접종한 후 30℃에서 250rpm 속도로 12시간 이상 배양하였다. 그 후 흡광도 O.D600가 2~6일 때 3000xg에서 5분간 원심 분리하여 효모 펠렛(pellet)을 얻어 흡광도 O.D600가 1이 되도록 BMMY 배지에 넣어 주었다(배지 적정량 100-200ml). 단백질 발현이 유지될 수 있도록 100% 메탄올을 최종 농도 0.5%가 되도록 24시간 주기로 넣어 준다. 정해진 시간마다 배지를 채취하여 단백질 발현량을 확인하였다. 그 후 FPLC를 이용하여 컬럼정제하였다. 각 분획에 대하여 SDS-PAGE 분석법을 통하여 KGF1 변이체가 포함된 분획을 확인 후, Bradford assay 법으로 정량을 진행하였다. 이 때 얻은 변이체의 양은 변이체에 따라 그 차이가 있었으며 변이체에 따라 약 4~12mg의 KGF1 변이체를 얻을 수 있었으며 순도는 98% 이상이었다.
KGF1 변이체 단백질의 정제과정에 사용되는 시약은 최대한 순도가 높은 제품을 사용하였으며, 정제 과정에서 사용한 시약은 아래에 나타내었다
Sodium phosphate monobasic(Sigma), Sodium phosphate dibasic(Sigma), Sodium chloride(Sigma).
FPLC는 GE UPC-800(GE healthcare사)을 사용하였으며, 칼럼은 SP-sepharose, heparin affinity column을 사용하였다.
실시예 4: CD(circular dichroism)를 이용한 KGF1 변이체의 Tm 확인
CD(Circular Dichroism)
J-810 spectrometer(JASCO)를 사용하여 CD 분석을 통하여 실시예 3의 정제된 KGF1 변이체의 구조와 열 안정성을 측정하였다. 천연형 KGF1와 변이체의 구조분석 및 Tm 측정을 위하여 천연형 KGF1과 변이체를 각각 20mM sodium phosphate(pH 7.0)에 녹여 최종농도가 0.2mg/ml이 되도록 일정하게 제작하였다. 그리고 0.1cm cell에 넣어 190nm ~ 250nm 영역으로 band width 1nm, response 0.25sec, data pitch 0.1 nm, scanning speed 20nm/min, cell length 1cm, accumulation 8번, 온도는 20℃ 조건으로 구조를 분석하였다.
온도 안정도를 분석하기 위하여 Tm(melting temperature)은 20℃와 95℃에서의 205nm 파장에서 0.1cm cell에 0.2mg/ml 농도로 진행하였다. 조건은 1℃/min의 조건으로 20℃ ~ 95℃까지 측정하였다.
표 3은 천연형 KGF1과 KGF1 변이체에 대한 구조 변화 정도와 열 안정성 측정 실험인 CD분석 중 205nm의 파장에서 온도 별 풀림 구조비율(fraction unfolded)을 측정한 결과를 나타낸 것이다. 접힘-풀림 현상이 일어날 때에는 205nm 부근의 구간에서 구조의 변화를 보이게 되는데 이를 이용하여 20~95℃ 범위 내에서 Tm을 측정하여 정확한 Tm값을 분석하였다.
실시예 5: 2차 분자 모델링에 의한 변이체 제작 및 Tm 분석
실시예 3에서 선택된 22번 변이체 단백질에 추가적으로 이황화 결합을 삽입하는 2차 분자설계를 진행하였다. 시스테인 변이 위치는 표 4에 나타내었다.
실시예 2의 방법으로 변이체를 클로닝한 후, 실시예 3의 방법으로 변이체 단백질을 생산 및 정제하였다.
정제된 변이체 단백질을 실시예 4와 동일한 방법을 CD를 이용한 Tm 값을 측정한 결과를 표 5에 나타내었다.
그 결과, 표 5 및 도 1에 나타난 바와 같이, 38번 변이체와 39번 변이체가 22번 변이체보다 Tm 값이 더 증가한 것을 확인하였다.
실시예 6: KGF1 변이체의 활성평가
세포 증식 분석(Cell proliferation assay)
천연형 KGF1와 KGF1 변이체가 실제로 활성을 나타내는지 확인하기 위하여 세포 증식 능력을 이용한 실험을 진행하였다.
실험에 사용한 BALB-3T3 세포주(주)다윈바이오사)는 10% heat-inactivated FBS, 100units/ml의 페니실린, 100mg/ml의 스트렙토마이신이 포함된 DMEM complete 배지를 이용하여 유지시켰다. 96웰 배양플레이트에 1x104 세포/웰의 BALB-3T3 세포를 분주(seeding)하고, 2시간 배양 후, 무혈청 DMEM 배지로 기아(starvation)화한 후에, 0.5% FBS가 포함된 DMEM 배지에 천연형 KGF1와 KGF1 변이체를 포함하는 샘플용액을 각각의 농도별(1pg~1ug)로 처리하고, 72시간 배양하였다.
배양 후 10 ㎕의 MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide] 용액을 첨가하고 2 시간 반응시킨 후, 형성된 formazan crystal을 100 ㎕의 DMSO로 용해시킨 후, 540 nm 파장에서 흡광도를 측정하였다. 약제에 대한 감수성은 약제를 처리하지 않은 웰(control)의 흡광도에 대한 약제처리 웰에서의 백분율로 비교하였다.
그 결과, 표 6에 나타난 바와 같이, Tm 값이 증가한 KGF1 변이체들의 세포증식 활성이 천연형 KGF1와 비교하여 큰 차이가 없는 것을 확인하였다.
실시예 7: KGF1 변이체 단백질의 안정성 확인
시스테인으로 치환된 KGF1 변이체들의 장기 보관시 수용액내에서의 안정성을 확인하기 위하여, 25℃ incubation test를 수행하였다. 인체의 상태와 가장 유사한 PBS(Phosphate buffer saline)상태에서 천연형 KGF1과 그 변이체들을 0.5mg/ml 농도로 용해시킨 후 25℃ 수조에서 incubation을 하였다. 7일 단위로 샘플링한 후, 13000rpm으로 15분간 4℃에서 원심 분리하여 상층액만 얻어 실시예 6의 방법으로 세포증식 실험을 실시하였다.
그 결과, 도 2 및 표 7에 나타난 바와 같이, 천연형 KGF1 단백질이 7일 후부터 급격한 활성저하를 나타내는 반면, KGF1 변이체 단백질은 대부분의 단백질이 7일 이후에도 높은 활성을 나타내고, 28일 이후에도 높은 활성을 나타내는 것을 확인하였으며, 28일 이후에도 90% 이상의 활성을 가지는 변이체도 있어 변이체의 안정성이 천연형에 비하여 현저히 개선된 것을 확인할 수 있었다.
아울러, 동일한 방식으로 50℃ incubation test를 진행하였으며, 1일 단위로 샘플링하여 13,000rpm으로 15분간 4℃에서 원심 분리하여 상층액만 얻어 세포증식 실험을 실시하였다.
그 결과, 도 3 및 표 8에 나타난 바와 같이, 50℃에서 반응 시, 천연형 KGF1 단백질은 1일 후에 30%의 활성을 나타내었으나, M7, M22, M37, M38, M39를 비롯한 다수의 변이체 단백질들은 50℃에서 1일 반응시켰을 때 90% 이상의 활성을 유지하였으며, 7일을 경과하여도 높은 활성을 유지하는 것을 확인하였다.
실시예 8: 3차 분자 모델링에 의한 변이체 제작 및 Tm 분석
변이체에 새로 도입된 이황화 결합을 방해할 가능성이 높다고 판단되는 79번째 시스테인(Cys)을 세린(Ser)으로 치환하여 하기 6종의 변이체를 추가로 선정하고, Tm 변화를 확인하였다(표 9).
추가 변이체에 대한 TM 결과는 기존 변이체와 비슷하거나 약간 상향된 결과를 나타내었다.
추가 변이체에 대한 활성평가는 실시예 6과 동일한 방법으로 수행하였으며, 표 10에 나타난 바와 같이, 추가변이체는 야생형과 비슷한 활성을 나타내었다.
실시예 9: KGF1 추가 변이체 단백질의 안정성 확인
실시예 8에서 선정된 추가 KGF1 변이체들의 장기 보관시 수용액내에서의 안정성을 확인하기 위하여, 실시예 7과 동일한 방법으로, 25℃ incubation test를 수행하였다.
그 결과, 도 4 및 표 11에 나타난 바와 같이, 천연형 KGF1 단백질이 7시간 후부터 급격한 활성저하를 나타내는 반면, KGF1 변이체 단백질은 7시간 이후에도 높은 활성을 나타내고, 28시간 이후에도 높은 활성을 나타내는 것을 확인하였으며, 6종의 추가 변이체 모두 28시간 이후에도 90% 이상의 활성을 가지는 것을 확인하여, 변이체의 안정성이 천연형에 비하여 현저히 개선된 것을 확인할 수 있었다.
아울러, 동일한 방식으로 50℃ incubation test를 진행하였으며, 1일 단위로 샘플링하여 13,000rpm으로 15분간 4℃에서 원심 분리하여 상층액만 얻어 세포증식 실험을 실시하였다.
그 결과, 도 5 및 표 12에 나타난 바와 같이, 50℃에서 반응 시, 천연형 KGF1 단백질은 7일 후에 30%의 활성을 나타내었으나, 추가 변이체는 시간이 경과하여도 높은 활성을 유지하였으며, 특히, M40, M42, M43, M44 및 M45는 50℃에서 28일 후 반응시켰을 때 90% 이상의 활성을 유지하였다.
본 발명에 따른 고안정성 KGF1 변이체는 열 안정성과 수용액 상태에서의 안정성이 천연형 KGF1에 비하여 현저히 우수하면서도, 치료활성은 동일하므로, 고안정성 KGF1 변이체를 함유하는 치료용 의약품 또는 기능성 화장품은 유통과 보관과정 중에서도 활성을 유효하게 오랫동안 유지될 수 있다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시태양일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

- 제3항의 유전자를 포함하는 발현 벡터.
- 제3항의 유전자 또는 제5항의 발현 벡터에 의해 형질전환된 형질전환체.
- 제1항의 고안정성 KGF1 변이체를 유효성분으로 함유하는 탈모의 예방 또는 치료용 약학적 조성물.
- 제1항의 고안정성 KGF1 변이체를 유효성분으로 함유하는 창상, 구내염 또는 피부염증 치료용 약학적 조성물.
- 제1항의 고안정성 KGF1 변이체를 유효성분으로 함유하는 탈모 방지 또는 개선용 모발 세정용 조성물.
