本発明のエクオール産生促進剤は酵母マンナンを含有するものであり、腸内でのエクオールの産生を促進する作用のための有効成分として、酵母マンナンのみを含む(すなわち、酵母マンナンからなる)ものであってもよいし、酵母マンナンに加えて同じくエクオール産生促進作用を有する他の有効成分を含むものであってもよい。

酵母マンナンの調製方法としては、公知のいずれかの方法を用いることができる。例えば、酵母からその細胞壁を調製する工程として、ビール、清酒、及び、パン等の製造に用いられた酵母から酵母エキスを抽出し、残滓である酵母の細胞壁から酵母マンナンを調製する工程により、酵母マンナンを得ることができる。酵母マンナンの原料とする酵母の細胞壁としては、例えば「酵母細胞壁」の商品名でアサヒグループ食品株式会社から販売されている酵母の細胞壁や、その他の同様の商品を用いてもよい。

20~40代の男女33名を3群に分け、酵母マンナン調製物X1gまたは2gを含有する麦茶飲料、またはプラセボ(マルトデキストリン1g)を含有する麦茶飲料(以下、各々「試験飲料」ともいう。)を、各群に対して1日1本ずつ4週間摂取させ、摂取前後で、[A]尿中エクオール濃度の測定および[B]肌の不調スコアの記録を行った(実施例1及び2、比較例1)。また、20~40代の女性18名を3群に分け、上記[A]及び[B]と同様に、酵母マンナン調製物X1gまたは2gを含有する麦茶飲料、またはプラセボ(マルトデキストリン1g)を含有する麦茶飲料を、1日1本ずつ4週間摂取させ、摂取前後で、[C]月経前不定愁訴スコアを記録した(実施例3及び4、比較例2)。
エクオール産生促進作用は、酵母マンナン調製物Xの摂取前後の各被験者の尿中エクール濃度をWilcoxon符号順位検定により評価した。尿中エクオール濃度は、室温下で解凍した尿を使用して、イムノクロマト法により測定した。尿中エクオール濃度の測定結果及びWilcoxon符号順位検定結果を表1に示す。酵母マンナン調製物X2g摂取群では、試験飲料の摂取後に尿中エクオール濃度が有意に増加した。また、酵母マンナン調製物X1g摂取群では、試験飲料の摂取後に尿中エクオール濃度が増加する傾向を示した。一方、プラセボ摂取群では、試験飲料の摂取前後で尿中エクオール濃度に有意な変化は認められなかった。
肌の状態改善作用は、以下の質問に対する、4件法による回答のスコア(なかった=スコア1、弱くあった=スコア2、中程度にあった=スコア3、強くあった=スコア4)をWilcoxon符号順位検定により評価した。
質問:最近1か月の様子を思い出し、肌の不調はありましたか。
月経前症候群改善作用は、下記の全17項目の質問に対する、4件法による回答のスコア(なかった=スコア1、弱くあった=スコア2、中程度にあった=スコア3、強くあった=スコア4)をWilcoxon符号順位検定により評価した。これらの質問は、精神的症状、身体的症状、及び、社会的機能の低下(以下、「月経前不定愁訴」ともいう。)に関する質問であり、アメリカ精神医学会の精神疾患の診断基準の手引きであるDSM-IV-TRの月経前不快気分障害(PMDD)診断基準案(American Psychiatric: Association :Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition Text Revision: DSM-IV-TR. Washington DC: American Psychiatric Association, 2000)の日本語訳(高橋三郎, 大野裕, 染谷俊幸(訳): DMS-IV-TR精神疾患の診断・統計マニュアル新訂版. 東京: 医学書院, 2003)に基づき作成された質問紙(宮岡佳子、秋元世志枝、上田嘉代子、加茂登志子: PMDD評価尺度の開発と妥当性および信頼性の検討 (The reliability and validity of the newly developed PMDD scale): 日本女性心身医学会雑誌 (JJp Soc Psychosom ObsLet Gynecol) Vol.14, No. 2, pp.194-201. 平成21年10月))に準じて作成された。
質問1:下記のような症状が月経の始まる1、2週間前から始まり、しかもその症状は月経が始まると2、3日で消失するということが、最近の月経周期にありましたか?
(1)抑うつ気分になる。または絶望的な気分になる。
(2)不安になる。または緊張する。
(3)涙もろくなる。または突然悲しくなる。
(4)怒りっぽくなる。またはイライラする。または人にあたる。
(5)興味がなくなる(仕事、学校、趣味など)。
(6)集中力が低下する。
(7)疲れやすくなる。または気力がなくなる。
(8)食欲が増す。または特定の食べ物(例:甘いものなど)が欲しくなる。
(9)いつもより眠りすぎる。眠気が強い。
(10)いつもより眠れなくなる。
(11)自分をコントロールできない感じになる。
(12)下記の身体症状のどれかが現れる。乳房の痛みや張り、腹部の張る感じ、腹痛、頭痛、関節痛、筋肉痛、身体がむくんだ感じ、体重増加、便秘。
質問2:上記に挙げた症状が1つでもあった人(弱くあった~強くあった)に伺います。
その症状が出現している間、下記の日常活動に支障がでたかお答えください。
(13)仕事の能率に支障がでた(職場または学校)。
(14)家事に支障がでた。
(15)職場の人との関係に支障が出た(関係に支障がでる例:つきあいを避ける、人にあたる、けんかをするなど)。
(16)家族との関係に支障がでた。
(17)友人・知人との関係に支障がでた。
(酵母マンナン調製物A:サッカロマイセス・パストリアヌス由来)
市販の酵母の細胞壁(商品名「酵母細胞壁」、アサヒグループ食品株式会社)10kgを蒸留水に懸濁し、水酸化ナトリウムを添加してpH11.0に調製した後、90℃で12時間煮沸した。遠心分離により上清を回収し、塩酸を添加してpH5.5に調製した。凍結乾燥操作により2kgの酵母マンナン調製物Aを得た。得られた酵母マンナン調製物Aの酵母α-マンナンの含有率を上述の算出方法により算出したところ、酵母α-マンナン含有率は50.5%であった。
市販の試薬(商品名「マンナン from Saccharomyces cerevisiae M7504(シグマ アルドリッチ ジャパン合同会社)を酵母マンナン調製物Bとした。酵母マンナン調製物Bの酵母α-マンナンの含有率を上述の算出方法により算出したところ、酵母α-マンナン含有率は、78.8%であった。
キャンディダ・ユーティリス(IFO0626)を8.0LのYPD培地に植菌し、0.50Lずつ2.0L容の三角フラスコに分注して、25℃、120rpmで24時間振とう培養した。培養液を集菌して蒸留水に懸濁し、100℃で10分間加熱後に遠心分離により上清を取り除いて細胞壁画分を調製した。得られた細胞壁画分を蒸留水に懸濁し、水酸化ナトリウムを添加してpH11.0に調製した後、90℃で12時間煮沸した。遠心分離により上清を回収し、塩酸を添加してpH5.5に調製した。凍結乾燥操作により3.7gの酵母マンナン調製物Cを得た。使用したYPD培地の組成は以下であり、pHは調製しなかった。
酵母エキス: 10g
ペプトン : 10g
グルコース: 20g
蒸留水 : 1L
肌の乾燥を自覚する30~40代の女性25名を2群に分け、酵母マンナン調製物Aを1230mg含有する錠剤、または麦芽糖を1230mg含有する対照錠剤(以下、各々「試験食」及び「対照食」ともいう。)を、各群に対して1日1回8週間摂取させた。試験食及び対照食の摂取前後で、[D]経皮水分蒸散量の測定及び[E]尿中エクオール濃度からのエクオール変換効率を算出した(実施例5、比較例3)。なお、1日に摂取する試験食中の酵母α―マンナン量は0.62gに相当する。
経皮水分蒸散量は、体内から無自覚のうちに角層を通じて揮散する水分量であり、肌の保湿レベルを表す指標として広く用いられている。経皮水分蒸散量は、温度が20~22℃、湿度が50±5%に管理された恒温恒湿室で被験者を30分間安静にさせた後、側臥位で、Tewameter TM300MP(Courage+Khazaka(CK)社)を使用して前腕内側を測定した。試験食及び対照食の肌保湿効果は、試験食摂取群(実施例5)と対照食摂取群(比較例3)について、試験食及び対照食の摂取8週後の経皮水分蒸散量、試験食及び対照食の摂取前の経皮水分蒸散量及び両者の経皮水分蒸散量の変化量を各々t検定により評価した。各群の水分蒸散量およびその変化量と検定結果を表8に示す。
尿中のダイゼイン、ジヒドロダイゼイン、エクオール濃度を後述の酵素HPLC(High Performance Liquid Chromatography)法で測定し、以下の算出式を用いてエクオール変換効率を算出した。

Redruello, B. et al.の酵素HPLC法(Redruello, B. et al., A novel UHPLC method for the rapid and simultaneous determination of daidzein, genistein and equol in human urine, Journal of Chromatography B (2015), Volume 1005 (15 November 2015),pages 1-8)に基づいて、尿中のダイゼイン、ジヒドロダイゼイン、エクオールを定量した。

ICRマウスの新鮮排泄便を30倍容のGAM培地に懸濁して100μm孔径のフィルター(FALCON社製)で濾過した。濾過液3mlを7mlのGAM培地に植菌し、嫌気チャンバー内で37℃、24時間培養した。培養後、半量の80%グリセロール溶液と混合して、マウス糞便懸濁液として-80℃で凍結保存した。

- 請求項1に記載のエクオール産生促進剤を含む、エクオール産生促進用食品組成物または飲料組成物。
- 請求項1に記載のエクオール産生促進剤を含む月経前症候群改善剤。
- 請求項3に記載の月経前症候群改善剤を含む、月経前症候群改善用食品組成物または飲料組成物。
- 請求項1に記載のエクオール産生促進剤を含む肌の状態改善剤。
- 請求項5に記載の肌の状態改善剤を含む、肌の状態改善用食品組成物または飲料組成物。
- エクオールの産生を促進するために使用される酵母マンナン。
- エクオール産生促進用食品組成物または飲料組成物の製造のための酵母マンナンの使用。
- 月経前症候群を改善するために使用される酵母マンナン。
- 月経前症候群改善用食品組成物または飲料組成物の製造のための酵母マンナンの使用。
