項3.男性ホルモンがテストステロンである、項1または2に記載の組成物。
項4.前記組成物中、コンパウンドKの含有量が0.05重量%以上である、項1〜3のいずれかに記載の組成物。
項5.前記組成物中、ジンセノシドRb1の含有量が10重量%以下である、項1〜4のいずれかに記載の組成物。
項6.前記発酵物が、薬用人参の微生物発酵物である、項2〜5のいずれかに記載の組成物。
項7.前記酵素分解物が、薬用人参のグリコシダーゼ分解物である、項2〜5のいずれかに記載の組成物。
項8.1日摂取量として、コンパウンドKを0.12〜160mg/60kg体重で含有する、項1〜7のいずれかに記載の組成物。
項9.1日摂取量として、薬用人参の発酵物及び/または酵素分解物を19.2〜24000mg/60kg体重で含有する、項2〜8のいずれかに記載の組成物。
項10.食品組成物、医薬組成物または飼料組成物である、項1〜9のいずれかに記載の組成物。
項11.男性ホルモン量の減少に起因する症状の改善を目的として使用される、項1〜10のいずれかに記載の組成物。
本発明は、コンパウンドKを含有する、男性ホルモン増加促進用組成物に関する。
本発明は、薬用人参の発酵物及び/または酵素分解物を有効成分として含有する、男性ホルモン増加促進用組成物に関する。
次の手順に従い、コンパウンドKによる男性ホルモン分泌に対する影響を評価した。
コンパウンドK(商品名GINSENOSIDE CK、0150S、エキストラシンシース社製)4mg/バイアルに50%エタノールを2 mL添加してコンパウンドKを溶解し、得られた溶液を、0.1%ウシ血清アルブミン(BSA)を添加したDME/F-12培地(ナカライテスク社製)(以下、0.1%BSA-DME/F-12)を用いて適宜希釈し、これを試料とした。
マウス(ddY系、雄、7〜8週令)から精巣を採取し初代培養を行った。具体的には、精巣をコラゲナーゼ処理し(Worthington、Type2、0.05%、37℃、15分)、細胞を分散させ、10%ウシ胎児血清(FBS)を含むDME/F-12培地にて3×105cell/0.5mLに調製し、24ウェルプレートに播種し、一夜、培養した。培養後、PBS(-)(ダルベッコズPBS(塩化カリウム0.2g/L、リン酸一カリウム0.2g/L、塩化ナトリウム8g/L、無水リン酸二ナトリウム1.15g/L、pH 7.4、等張溶液。イオン含有量(mEq)Na+ 148、K+ 4、Cl- 139)で細胞を洗浄し、これに、0.1%BSA-DME/F-12培地と共に、前述のようにして調製した試料を1/10容量となるように添加し、ウェル中のコンパウンドKの濃度を1μg/mL、10μg/mL、100μg/mLとした。
結果を図1に示す(平均値±標準誤差、検体数各4)。図1から明らかなように、コンパウンドKを添加していない培地(図中「0μg/mL」)におけるテストステロン濃度と比較して、コンパウンドKを添加した培地においてテストステロン濃度が高まった。具体的には、コンパウンドKの添加量が10μg/mLあるいは100μg/mLのいずれであっても培地中のテストステロン濃度が明らかに高まった。また、図1には示していないが、コンパウンドKの添加量が1μg/mLであっても培地中のテストステロン濃度が高まった。このことから、コンパウンドKによれば、細胞からのテストステロン分泌を有意に促進できることが確認された。
次の手順に従い、薬用人参による男性ホルモン分泌に対する影響を評価した。
ラクトバチルスカゼイハセガワ菌株(受託番号:FERM BP-10123)を用いて薬用人参の発酵物を得た。具体的には、ラクトバチルスカゼイハセガワ菌株1白金耳を培地(グルコース30g/L、酵母エキス10g/L、大豆ペプチド5g/L、K2HPO4 2g/L、MgSO4・7H2O 0.5g/L)20mLに植菌し、28℃で48時間静置培養した。次いで、得られた培養液2mLを、新たな培地(組成は前述と同じ)120mLに添加し、28℃で48時間静置培養した。得られた培養液20mLを、高圧蒸気滅菌処理(121℃、20分間)した発酵用溶液(オタネニンジン(側根)粉末130g/L 、酵母エキス10g/L、大豆ペプチド5g/L、炭酸カルシウム10g/L)1 Lに添加し、28℃ で10日間発酵した。得られた発酵液を水酸化ナトリウムでpH5.0に調整し、噴霧乾燥することにより、オタネニンジン発酵物を得た。
前記試験例と同様にして、精巣をコラゲナーゼ処理し、細胞を分散させ、10%FBSを含むDME/F-12培地を用いて、得られた細胞をウェルプレートに播種(1.5×105cells/well)し、一夜、培養した。培養後、PBS(-)で細胞を洗浄し、0.1%BSA-DME/F-12培地と共に、前述のようにして調製した試料E、試料W、比較試料Eまたは比較試料Wをオタネニンジン発酵物またはオタネニンジン(側根)粉末1 mg相当量/mL(ウェル最終濃度)となるように添加し、2時間培養後、培地を採取し、各培地中のテストステロン濃度をEIAにより前述と同様に測定した。
結果を図2に示す(平均値±標準誤差、検体数各4)。なお、試料E、W、比較試料E、Wのいずれも用いない以外は同様にして得た培地中のテストステロン濃度を、図中「無添加」にて示す。
次の手順に従い、薬用人参による血中男性ホルモン濃度に対する影響を評価した。
前記試験例2と同様にして、オタネニンジン発酵物を得た。このようにして得た発酵物(粉末)を水と混合し、該発酵物を2000mg/10mL、400mg/10mL、80mg/10mL、16mg/10mLで含有する液体試料(順に試料1〜4とする)を得た。なお、コントロールは液体試料に代えて水とした。
ラット(Wistar系、雄、13週齢)を、ラット用ゲージ(ステンレス金網製蓋、プラスチック製ケージ、1匹/ケージ)内で、市販固型飼料(商品名CE-2、日本クレア社製)及び水の自由給餌にて7日間飼育した。試料投与前体重に基づいて各試験間で差がないように振り分けた(1群あたり4匹)。ラットの体重100g当り1.0mLとして、試料投与時に体重換算して必要量を算出し、調製した試料1〜4をラットに強制経口投与した。強制投与は一日午前と午後の2回行い、投与間隔として6時間以上の間を設けた。なお、コントロールとして水を投与する群では、ラット体重100g当り0.5mLにて同様に強制投与を行った。試料または水の投与は21日間連続して行った。
結果を表1に示す。表1では、ストレス負荷前の各群における血中の遊離テストステロン濃度を1として示す。表1から明らかなように、コントロール群では、ラットにストレスを与えることにより血中の遊離テストステロン濃度が0.36へ大きく低下していることが分かる。これに対して、前記試料1〜4を投与した群ではいずれも、ストレス負荷後の値が、コントロール群におけるストレス負荷後の値を上回っており、ストレス負荷による血中の遊離テストステロン濃度の低下が有意に抑制された。
次の手順に従い、薬用人参による男性ホルモン濃度に対する影響を評価した。
ストレス無通常食群と比較して、ストレス有通常食群ではテストステロン分泌量は有意に低下したのに対して、ストレス有発酵人参食群では分泌量の低下が抑制された。このことは、薬用人参の発酵物の摂取が、ストレス環境下におけるテストステロン分泌の増加に役立つことを示す。
Claims (9)
- コンパウンドKを含有する、男性ホルモン増加促進用組成物。
- 薬用人参の乳酸菌発酵物を有効成分として含有する、男性ホルモン増加促進用組成物(但し、シカの角(deer antler)、水、ブドウ、茶及びジンセン(ginseng)を含む混合物に酵母を加えて発酵させた組成物を除く)。
- 男性ホルモンがテストステロンである、請求項1または2に記載の組成物。
- 前記組成物中、コンパウンドKの含有量が0.05重量%以上である、請求項1〜3のいずれかに記載の組成物。
- 前記組成物中、ジンセノシドRb1の含有量が10重量%以下である、請求項1〜4のいずれかに記載の組成物。
- 1日摂取量として、コンパウンドKを0.12〜160mg/60kg体重で含有する、請求項1〜5のいずれかに記載の組成物。
- 1日摂取量として、薬用人参の乳酸菌発酵物を19.2mg〜24000mg/60kg体重で含有する、請求項2〜6のいずれかに記載の組成物。
- 食品組成物、医薬組成物または飼料組成物である、請求項1〜7のいずれかに記載の組成物。
- 男性ホルモン量の減少に起因する症状の改善を目的として使用される、請求項1〜8のいずれかに記載の組成物。
