化合物(I)は、当業者に周知な方法により製造されたものであってもよい。例えば、化合物(I)は、後述する参考例2に記載の方法で合成することができる。
化合物(I)の塩は、通常の塩を製造する方法により得ることができる。具体的には、例えば、化合物(I)を溶媒に、必要に応じて加温して、懸濁または溶解させ、次いで、得られる懸濁液または溶液に、無機酸塩、有機酸塩、酸性アミノ酸塩、無機塩基塩、有機塩基塩および塩基性アミノ酸塩からなる群から選択される一つの酸または塩基を加え、室温下あるいは氷浴で冷却しながら数分から数日間撹拌することにより、または室温下あるいは氷浴で冷却しながら数分から数日間放置することにより、製造することができる。これらの製造方法により、化合物(I)の塩を、結晶または非晶質として得ることができる。ここで使用する溶媒としては、例えばエタノール、1-プロパノール、イソプロパノール等のアルコール系溶媒;アセトニトリル;アセトン、2-ブタノン等のケトン系溶媒;酢酸エチル等のエステル系溶媒;ヘキサン、ヘプタン等の飽和炭化水素系溶媒;t-ブチルメチルエーテル等のエーテル系溶媒または水を挙げることができる。これらの溶媒は単独で使用してもよく、2種以上を混合して使用してもよい。
以下に化合物(I)またはその塩の結晶の製造方法について説明する。化合物(I)またはその塩の結晶は、化合物(I)またはその塩を、溶媒中で加熱溶解し、攪拌下冷却して晶析することにより製造することもできる。
本発明の他の実施形態は、化合物(I)の塩またはその結晶、および薬剤学的に許容される添加物を含有する医薬組成物である。医薬組成物は、薬剤学的に許容される添加物を化合物(I)の塩またはその結晶と混和することにより製造することができる。本発明に係る医薬組成物は例えば第十六改正日本薬局方の製剤総則に記載の方法など既知の方法に従って製造することができる。

室温で窒素雰囲気下、ジエチルホスホノ酢酸エチル(16.56g)のTHF(340mL)溶液に水素化ナトリウム(約60%、2.95g)を加え、15分間攪拌した後、3-メトキシ-4-オキソピペリジン-1-カルボン酸t-ブチルエステル(14.11g)(国際公開第2012/080735号)のテトロヒドロフラン(50mL)溶液を滴下して加え、室温で16時間攪拌した。反応液に酢酸エチル(1000mL)と飽和炭酸水素ナトリウム水溶液(400mL)を加えた。有機層を無水硫酸マグネシウムで乾燥した。乾燥剤を濾去し、濾液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(14.65g)を得た。

4-(2-エトキシ-2-オキソエチリデン)-3-メトキシピペリジン-1-カルボン酸t-ブチルエステル(1.20g)のジクロロメタン(20mL)溶液にトリフルオロ酢酸(4mL)を加え、混合物を室温で5時間攪拌した。減圧濃縮し、トルエン(100mL)を加え、減圧濃縮した。再度トルエン(10mL)を加え、減圧濃縮した後、残渣に酢酸エチル(10mL)、トルエン(10mL)および4N塩化水素酢酸エチル溶液(5mL)を加え、減圧濃縮し、標記化合物(0.95g)を得た。

2-(3-メトキシピペリジン-4-イリデン)酢酸エチルエステル塩酸塩(0.95g)、エタノール(10mL)および酢酸エチル(10mL)の混合物にパラジウム-活性炭素(Pd10%)(0.21g)を加え、水素雰囲気下、室温で17時間攪拌した。雰囲気を窒素に戻し、混合物をセライト濾過し、セライトをエタノールで洗浄した。濾液と洗浄液を合わせ、減圧濃縮して、標記化合物(1.02g、主にシス異性体)を得た。

窒素雰囲気下、-20℃にて、2-(3-メトキシピペリジン-4-イル)酢酸エチルエステル塩酸塩(2.098g)、DMF(80mL)、トリエチルアミン(1.35mL)および炭酸カリウム(2.44g)の混合物にブロモジフェニルメタン(2.62g)のDMF(5mL)溶液を加え、室温で終夜攪拌した。t-ブチルメチルエ-テル(400mL)および水(200mL)を加えた。有機層を水(200mLx2)および飽和塩化ナトリウム水溶液(200mL)で順次洗浄し、無水硫酸マグネシウムで乾燥した。乾燥剤を濾去して、濾液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(1.127g)を無色液体として得た。

2-((3R*,4S*)-1-ベンズヒドリル-3-メトキシピペリジン-4-イル)酢酸エチルエステル(411mg)、1N塩酸(1.118mL)、パラジウム-活性炭素(Pd5%)(0.238g)、シクロヘキセン(20mL)およびエタノール(80mL)の混合物を、90℃で18時間攪拌した。反応液をセライト濾過し、セライトをエタノール(50mLx2)にて洗浄した。濾液と洗浄液を合わせ、減圧濃縮した。残渣にヘプタンとトルエンの混合溶媒(10mL、1:1)および2N塩酸(20mL)を加え、分液した。有機層を2N塩酸(20mL)で抽出した。合わせた水層に5N水酸化ナトリウム水溶液を加え、アルカリ性にし、ジクロロメタン(50mLx2)で抽出した。合わせた有機層を無水硫酸マグネシウムで乾燥した。乾燥剤を濾去して、濾液に4N塩化水素酢酸エチル溶液(2mL)を加え、減圧濃縮して、標記化合物(119mg)を得た。

2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸の合成
5,7-ジクロロピラゾロ[1,5-a]ピリミジン-2-カルボン酸エチルエステル(CAS No.1232224-62-2)(5g)および2-フルオロ-6-メチルアニリン(CAS No.443-89-0)(2.41g)のNMP(3mL)溶液を120℃で4時間撹拌した。別途に5,7-ジクロロピラゾロ[1,5-a]ピリミジン-2-カルボン酸エチルエステル(0.50g)および2-フルオロ-6-メチルアニリン(0.27g)とNMP(0.20mL)を120℃で4時間撹拌した反応液と合わせて酢酸エチルで希釈し、水および飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥した。乾燥剤を濾去後、濾液を減圧濃縮して残渣にジクロロメタンを加えて不溶の固体を濾取し、標記化合物(2.88g)を淡黄色固体として得た。濾液を減圧濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン-酢酸エチル系)で精製し、標記化合物(2.28g)を得た。

5-クロロ-7-((2-フルオロ-6-メチルフェニル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボン酸エチルエステル(5.16g)のDMF(80mL)溶液に炭酸セシウム(7.23g)とヨウ化メチル(4.61mL)を加え、50℃で3時間撹拌した。反応液を酢酸エチルで希釈して水および飽和食塩水で順次洗浄し、無水硫酸ナトリウムで乾燥した。乾燥剤を濾去して、濾液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(4.64g)を淡黄色固体として得た。

窒素雰囲気下、5-クロロ-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボン酸エチルエステル(0.73g)および1,4-ジオキサン(16mL)の混合物に水(2mL)、炭酸ナトリウム(0.53g)、4-クロロ-3,5-ジフルオロフェニルボロン酸(0.44g)およびテトラキス(トリフェニルホスフィン)パラジウム(0)(0.12g)を加え、100℃で3時間撹拌した。室温に戻した後、反応液に酢酸エチル(200mL)および水(50mL)を加え、有機層を分離し、無水硫酸マグネシウムで乾燥した。乾燥剤を濾去して、濾液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(0.96g)を淡黄色固体として得た。

5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボン酸エチルエステル(2.35g)および1,4-ジオキサン(100mL)の混合物に水(10mL)および4N水酸化リチウム水溶液(2.47mL)を加え、室温で17時間撹拌した。5N塩酸(2mL)を加え、減圧下、容積が約15mLになるまで濃縮した。超音波処理し、混合物を室温で30分間撹拌した後、固体を濾取し、水(5mL)で洗浄し、減圧下で乾燥して、標記化合物(2.24g)を淡黄色固体として得た。

5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボン酸(112mg)のDMF(6mL)溶液にHOBT(57mg)、WSC(72mg)、2-((3R*,4S*)-3-メトキシピペリジン-4-イル)酢酸エチルエステル塩酸塩(77mg)(参考例1)、およびトリエチルアミン(101mg)を順次加え、室温で15時間撹拌した。反応液に酢酸エチル(50mL)と水(20mL)を加え、有機層を水(20mLx2)で洗浄し、無水硫酸マグネシウムで乾燥した。乾燥剤を濾去し、濾液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(137mg)を得た。

窒素雰囲気下(S)-2-(ブテン-3-エン-1-イル(1-フェニルエチル)アミノ)酢酸エチルエステル(CAS No.186586-65-2)(60.29g)のTHF(750mL)溶液を-75℃に冷却し、カニューレを用いてこの溶液にエタノール・ドライアイスで冷却したリチウムジイソプロピルアミドのn-ヘキサン・THF溶液(1.13M,245mL)を加えた。-20℃まで昇温して15分間攪拌した後、再びエタノール・ドライアイスを冷媒とした冷却槽を用いて反応液を冷却し、-72℃以下で臭化亜鉛(156g)のジエチルエーテル(800mL)溶液を加えた。冷却槽を外して反応液を室温に戻し、1時間攪拌した。食塩を含む氷水を冷媒とした冷却槽を用いて反応液を0℃以下に冷却し、シアン化銅(I)(41.3g)、塩化リチウム(39.1g)およびTHF(750mL)の混合物を加えた。続いて、(4-メチルフェニル)スルホニルシアニド(CAS No.19158-51-1)(50.2g)のTHF(200mL)溶液を加え、室温で14時間攪拌した。攪拌しながら、水(1000mL)、アンモニア水(28%、300mL)および酢酸エチル(1500mL)を加えた。懸濁液をセライト濾過し、沈殿物を除去した。濾液を分液ロートに移し、有機層と水層を分離し、有機層をアンモニア水(10%、750mL)、水(750mL)および飽和塩化ナトリウム水溶液(750mL)で順次洗浄した。セライトと沈殿物を酢酸エチル(500mL)で4回洗浄し、これらの洗浄液を合わせた。合わせた洗浄液を用いて、上記4つの水層(最初に分離した水層、洗浄に使用したアンモニア水、洗浄に使用した水、洗浄に使用した飽和塩化ナトリウム水溶液)を順次抽出した。有機層を全部合わせ、無水硫酸マグネシウムで乾燥した。乾燥剤を濾去し、濾液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(42.98g)を得た。

氷冷下で窒素の雰囲気下、(2S,3S)-3-(シアノメチル)-1-((S)-1-フェニルエチル)ピロリジン-2-カルボン酸エチルエステル(44.74g)のTHF(700mL)溶液にテトラヒドロほう酸リチウム(20g)を加え、5時間加熱還流した。反応液を室温に冷却し、1N塩酸(1000mL)および酢酸エチルの混合物に注いだ。30分間攪拌した後、炭酸水素ナトリウム(約100g)を加え、酢酸エチルで3回、ジクロロメタンで2回抽出した。有機層を合わせ、乾燥し、乾燥剤を濾去した後、濾液を減圧濃縮した。残渣をNHシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(28.98g)を得た。

窒素雰囲気下2-((2S,3S)-2-ヒドロキシメチル-1-((S)-1-フェニルエチル)ピロリジン-3-イル)アセトニトリル(52.44g)のTHF(2000mL)溶液を-74℃以下に冷却し、攪拌しながらトリフルオロ酢酸無水物(36.4mL)を内温が-73℃を超えないように滴下して加えた。更に3時間攪拌した後、トリエチルアミン(120mL)を滴下して加えた。15分間攪拌した後、内温を室温に戻し、15時間加熱還流した。溶液を減圧除去し、残渣に酢酸エチル(5000mL)と2N水酸化ナトリウム水溶液(500mL)を加え、分液した。有機層を無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮した。残渣をNHシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(51.25g)を得た。

水冷下で窒素雰囲気下、2-((3R,4S)-3-ヒドロキシ-1-((S)-1-フェニルエチル)ピペリジン-4-イル)アセトニトリル(51.01g)とTHF(750mL)の混合物に、攪拌しながら水素化ナトリウム(60%、9.19g)を15分かけて少しずつ加えた。更に10分間攪拌した後、硫酸ジメチル(22.94mL)を滴下して加えた。5時間攪拌した後、飽和塩化アンモニウム水溶液(50mL)、アンモニア水(28%、50mL)および酢酸エチル(1000mL)を順次加え、30分間攪拌した。1N水酸化ナトリウム水溶液(500mL)を加え、分液した。有機層を飽和塩化ナトリウム水溶液(250mL)で洗浄し、合わせた水層を酢酸エチル(500mL)で抽出した。有機層を合わせ、無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮した。残渣をNHシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(47.9g)を得た。

窒素雰囲気下、メタノール(1500mL)に塩化チオニル(352mL)を氷冷下滴下して加えた後、2-((3R,4S)-3-メトキシ-1-((S)-1-フェニルエチル)ピペリジン-4-イル)アセトニトリル(49.8g)のメタノール(50mL)溶液を加えた。混合物を20時間加熱還流した後、氷冷し、更に塩化チオニル(352mL)を滴下して加え、22時間加熱還流した。反応液を再度氷冷し、更に塩化チオニル(50mL)を滴下して加え、62時間加熱還流した。反応液を室温に戻した後、減圧濃縮した。残渣を水(250mL)および酢酸エチル(250mL)に溶解し、アンモニア水(28%、50mL)を加えた後、pH11になるまで2N水酸化ナトリウム水溶液を加えた。分液し、水層を酢酸エチル(500mL)で抽出し、有機層を合わせ、無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮し、残渣Aを得た。
次に水層を減圧濃縮し、残渣にメタノール(1000mL)および濃硫酸(50mL)を加え、22時間加熱還流した。反応液を室温に戻し、炭酸水素ナトリウムでpHを8に調整した。溶媒を減圧除去し、残渣を酢酸エチル(1000mL)および水(250mL)に溶解し、2N水酸化ナトリウム水溶液を用いてpHを11に調整し、分液した。水層を酢酸エチル(500mL)で抽出し、有機層を合わせ、無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮し、残渣Bを得た。
窒素雰囲気下、メタノール(1500mL)に塩化チオニル(352mL)を氷冷下滴下して加えた後、残渣Aのメタノール(100mL)溶液を加えた。混合物を22時間加熱還流した。反応液を室温に戻し、溶媒を減圧除去し、残渣を酢酸エチル(1000mL)および水(250mL)に溶解し、アンモニア水(28%、50mL)を加え、2N水酸化ナトリウム水溶液を用いてpHを11に調整し、分液した。水層を酢酸エチル(500mL)で抽出し、有機層を合わせ、無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮し、残渣Cを得た。
2回目の反応後処理で得た水層を減圧濃縮し、残渣にメタノール(1000mL)および濃硫酸(50mL)を加え、22時間加熱還流した。反応液を室温に戻し、炭酸水素ナトリウムでpHを8に調整した。溶媒を減圧除去し、残渣を酢酸エチル(1000mL)および水(250mL)に溶解し、2N水酸化ナトリウム水溶液を用いてpHを11に調整し、分液した。水層を酢酸エチル(500mL)で抽出し、有機層を合わせ、無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮し、残渣Dを得た。
残渣B、残渣Cおよび残渣Dを合わせ、NHシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、粗2-((3R,4S)-3-メトキシ-1-((S)-1-フェニルエチル)ピペリジン-4-イル)酢酸メチルエステル(52.35g)を得た。
粗2-((3R,4S)-3-メトキシ-1-((S)-1-フェニルエチル)ピペリジン-4-イル)酢酸メチルエステル(52.35g)と別途同様の方法で得た粗2-((3R,4S)-3-メトキシ-1-((S)-1-フェニルエチル)ピペリジン-4-イル)酢酸メチルエステル(1.45g)を合わせた酢酸エチル(400mL)溶液に、攪拌しながら4N塩化水素酢酸エチル溶液(100mL)を加えた。室温で一晩攪拌した後、沈殿物を濾取し、酢酸エチル(100mL)で洗浄し、減圧乾燥した。得られた固体に、攪拌しながら水(250mL)、酢酸エチル(250mL)、炭酸水素ナトリウム飽和水溶液(250mL)を順次加えた。さらに酢酸エチル(750mL)を加えた後、炭酸カリウムを用いてpHを12に調整し、分液した。水層を酢酸エチル(250mL)で洗浄し、この洗浄液と分液した有機層を合わせ、無水硫酸マグネシウムで乾燥後、乾燥剤を濾去し、濾液を減圧濃縮した。得られた残渣(46.3g)を酢酸エチル(400mL)に溶解し、攪拌しながら4N塩化水素酢酸エチル溶液(80mL)を加えた。室温で一晩攪拌した後、沈殿物を濾取し、酢酸エチル(40mL)で洗浄し、減圧乾燥して得られた固体に、水(250mL)および酢酸エチル(250mL)を順次加え、攪拌しながら炭酸水素ナトリウムを加えた。水(750mL)および酢酸エチル(750mL)を加え、分液した。水層に5N水酸化ナトリウム水溶液(50mL)を加え、酢酸エチル(1000mL)で抽出した。有機層を合わせ、無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮し、45.2gの残渣を得た。これを、別途同様の操作で得た16.2gの残渣と合わせ、酢酸エチル(500mL)に溶解し、攪拌しながら4N塩化水素酢酸エチル溶液(100mL)を加えた。室温で一晩攪拌した後、沈殿物を濾取し、酢酸エチル(60mL)で2回洗浄し、減圧乾燥した。得られた固体に水(800mL)および酢酸エチル(800mL)を順次加え、攪拌しながら炭酸水素ナトリウムを加え、分液した。水層に5N水酸化ナトリウム水溶液(50mL)を加え、酢酸エチル(800mL)で抽出した。有機層を合わせ、無水硫酸マグネシウムで乾燥し、乾燥剤を濾去し、濾液を減圧濃縮し、標記化合物(60.7g)を得た。

2-((3R,4S)-3-メトキシ-1-((S)-1-フェニルエチル)ピペリジン-4-イル)酢酸メチルエステル(4.37g)のメタノール(50mL)溶液および塩化水素のメタノール溶液(5-10%、30mL)の混合物にパラジウム-活性炭素(Pd10%)(0.639g)を加え、水素雰囲気下、室温で16時間攪拌した。さらに、パラジウム-活性炭素(Pd10%)(0.639g)を加え、水素雰囲気下、室温で24時間攪拌した。パラジウム-活性炭素(Pd10%)(0.639g)を加え、水素雰囲気下、室温で更に24時間攪拌した。混合物をセライト濾過し、セライトをメタノールで洗浄した。濾液と洗浄液を合わせ、減圧濃縮して、標記化合物(3.81g)を得た。

5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボン酸(6.03g)(参考例2-(2d))のDMF(100mL)溶液にHOBT(2.280g)およびWSC(3.23g)を加え、15分間攪拌した後、2-((3R,4S)-3-メトキシピペリジン-4-イル)酢酸メチルエステル塩酸塩(3.36g)のDMF(50mL)溶液、およびトリエチルアミン(5.64mL)を順次加え、室温で3時間撹拌した。反応液に酢酸エチル(700mL)と水(500mL)を加え、有機層を少量の塩化ナトリウムを含む水(750mL)で2回洗浄した。水層と洗浄液を酢酸エチル(750mL)で順次抽出し、有機層を合わせ、無水硫酸マグネシウムで乾燥した。乾燥剤を濾去し、濾液を減圧濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘプタン-酢酸エチル系)で精製し、標記化合物(5.83g)を得た。

2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸のX線回折実験
参考例2-(2n)で得られた2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸の固体をメタノールに溶解し、アセトニトリルをリザーバーとする蒸気拡散法により再結晶化した。得られた単結晶を用いてX線回折実験を行った。結晶学的データおよび構造解析結果を表1に、原子座標データを表2に示す。かかる結果より、標記化合物の絶対構造を特定した。

参考例2に記載の方法と同様の方法で得られた2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸(63.5g)、テトラヒドロフラン(156mL)、アセトニトリル(496mL)および水(65mL)の混合物を内温95℃で撹拌しながら、水(550mL)を加えた。30分間撹拌した後、アセトニトリル(25mL)および水(10mL)を加え、沈殿物を溶解させた。水(3mL)を滴下し、30分間撹拌した後、再び透明な溶液になるまでアセトニトリル(3mL)を滴下した。30分間撹拌した後、内温を50℃まで下げ、更に2時間撹拌した。参考例2に記載の方法と同様の方法で得られた標記化合物の固体(0.05g)を加え、加熱を止め、内温が室温に戻った後3日間室温で撹拌した。混合物を内温-5℃まで冷却し、固体を濾取し、氷水を用いて冷却したアセトニトリル:テトラヒドロフラン:水 4:1:5混液(3x200mL)で洗浄し、窒素気流下で減圧乾燥し、標記化合物の結晶(46.0g)を得た。

参考例2に記載の方法と同様の方法で得られた2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸(10mg)に酢酸エチル1mLを加えた。70℃の水浴にて、固体を完全に溶解させた。濃塩酸3μLを加え、終夜室温で撹拌した。固体を濾取し、減圧乾燥し、標記化合物の結晶(9mg)を得た。
参考例2に記載の方法と同様の方法で得られた2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸(1.204g)および酢酸エチル(120mL)を固体が完全に溶解するまで窒素雰囲気下70℃で攪拌した。溶液を室温に戻した後、4N塩化水素酢酸エチル溶液(4mL)および上記で得られた標記化合物の結晶数mgを加えた。終夜室温で撹拌した後、固体を濾取し、酢酸エチル(5mL)で洗浄し、減圧乾燥し、標記化合物の結晶(1.202g)を得た。

ウシ胎児血清3%(3%FBS)添加DMEM培養液(インビトロジェン社)を用いて培養したヒト胎児腎臓由来細胞株HEK293細胞を2×105cells/mLに調製し、1型コラーゲンでコートした384穴黒色プレート(透明底)(グライナー社)に25μL/wellとなるように播いて、炭酸ガス培養装置で一晩培養した。Hanks-20mM Hepes緩衝液(pH7.4)で調製したFLIPR Calcium Assay Kit(Molecular Devices社)を細胞に25μL/wellとなるように添加して、1時間炭酸ガス培養装置にて培養した。Hanks-20mM Hepes緩衝液(pH7.4)で、終濃度が5U/mL(BAEE unit)になるように調製したトリプシン(SIGMA-ALDRICH社、カタログ番号:T8816、Enzyme Commission Number 3.4.21.4)を、384穴deep wellポリプロピレンプレート(グライナー社)に添加してアゴニスト試薬プレートとした。FDSS6000(浜松ホトニクス社)で測定する30分前に、Hanks-20mM Hepes緩衝液(pH7.4)で調製した被検化合物を、細胞に8μL/well添加した。被検化合物を添加した細胞プレートと試薬プレートをFDSS6000にセットし、試薬プレートからアゴニスト溶液を22μl/well添加し、その際の細胞内カルシウム濃度の変化をCCDカメラにて検出した。測定は37℃で120秒間行い、試薬プレートから細胞プレートへの試薬の添加は、FDSS6000本体内蔵の384well自動分注機で行った。
被検化合物の細胞内カルシウム上昇抑制活性(IC50(nM))を、表3に示した。
被検化合物約1mgを試験管に秤量し、試験液1mLを添加した。試験液は、0.1M塩酸水溶液(pH1)、およびブリトン-ロビンソン緩衝液(pH3、pH5およびpH7;イオン強度0.3)を用いた。室温で試験液を24時間振とう後、フィルターろ過した。ろ液中の被検化合物の濃度を高速液体クロマトグラフィーにより測定し、溶解度を評価した。結果を表5に示す。また、高速液体クロマトグラフィー(HPLC)条件を以下に示す。

グラジエント条件
溶出試験は、FaSSIF(3mMタウロコール酸ナトリウム、0.75mMレシチン、29mMリン酸二水素ナトリウムおよび103mM塩化カリウムを含有する水溶液で、水酸化ナトリウム水溶液の滴下によりpH6.5に調整した水溶液)を試験液とし、パドル法にて行った。試験液100mL中に、被検化合物約10mgを添加した。37℃、50rpmの条件で、パドルを用いて試験液を撹拌した。撹拌開始後、表6に示す各時点で、試験液1mLを採取し、フィルターろ過した。ろ液中の被検化合物の濃度を高速液体クロマトグラフィー(HPLC条件は、試験例2に記載した条件と同じ条件)により測定した。結果を表6に示す。

粉末X線回折において、回折角度(2θ±0.2°)8.3°および13.1°に回折ピークを有する、2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸の塩酸塩の結晶。 - 請求項2記載の結晶であって、
粉末X線回折において、回折角度(2θ±0.2°)8.3°、13.1°および15.7°に回折ピークを有する、2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸の塩酸塩の結晶。 - 請求項2記載の結晶であって、

粉末X線回折において、回折角度(2θ±0.2°)8.3°、11.4°、13.1°、15.2°、15.7°、17.3°、18.8°、19.7°、22.3°および25.0°に回折ピークを有する、2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸の塩酸塩の結晶。 - 請求項2記載の結晶であって、
図1で示される粉末X線回折パターンと実質的に同一の粉末X線回折パターンを有する、2-((3R,4S)-1-(5-(4-クロロ-3,5-ジフルオロフェニル)-7-((2-フルオロ-6-メチルフェニル)(メチル)アミノ)ピラゾロ[1,5-a]ピリミジン-2-カルボニル)-3-メトキシピペリジン-4-イル)酢酸の塩酸塩の結晶。 - 請求項2記載の結晶であって、
