Abstract
Cryタンパク質をバチルス属細菌の菌体内で高生産するための方法を提供する。 Cryタンパク質をコードする遺伝子とσA依存性プロモーター又はσH依存性プロモーターを含む制御領域とが作動可能に連結して組み込まれてなる発現プラスミドでバチルス属細菌を形質転換し、当該形質転換細胞を培養することを含むCryタンパク質又はそれを含有する培養産物の製造方法であって、前記発現プラスミドが、配列番号9で示されるアミノ酸配列からなる複製タンパク質又はこれとアミノ酸配列において80%以上の同一性を有し、且つ複製開始に関与するタンパク質をコードするポリヌクレオチドを含むプラスミドである、方法。
Description
本発明はバチルス属細菌を用いたCryタンパク質の製造方法に関する。
一般的に、大腸菌を用いて、その菌体内にてタンパク質を大量発現させるとインクルージョンボディ(inclusion body、封入体)と呼ばれる不溶で不活性な凝集体(変性タンパク質)として回収されることが多く、タンパク質が生物活性を回復するためには、凝集体の可溶化及び活性化(リフォールディング)の操作が必要となる(非特許文献1)。それでも活性型のタンパク質が得られない場合はインクルージョンボディを形成しないように、発現量を調節することが行われる(非特許文献2、3)。その他、異種タンパク質を菌体内にて生産させる場合は、異種タンパク質の宿主への影響等々より、プロモーターの発現やプラスミドコピー数を調整する必要もある(非特許文献2、3)。
一方、菌体外タンパク質の発現においては、インクルージョンボディの形成や宿主内の該タンパク質濃度の影響を考慮する必要性が少なく、従来、高発現プロモーターと高コピー数プラスミドを組み合わせた高発現化やプロモーターの抑制を解除する検討 (特許文献1)等が行われてきた。また、このような菌体外タンパク質の生産に適する宿主として、バチルス属細菌、特に、枯草菌が優れていることも報告されている(非特許文献4、5)。さらに、枯草菌は特に安全性が高い微生物として認識されている点においても、宿主として有利である。
バチルス・チューリンゲンシス(Bacillus thuringiensis)は、各種殺虫性結晶タンパク質(crystal protein、以下「Cryタンパク質」と称する)を生産し、生物農薬として使用されている微生物である(非特許文献6)。一般に、バチルス・チューリンゲンシスが有するCryタンパク質遺伝子(cry遺伝子)をコードするプラスミドは、シータ型複製を行う低コピー数の巨大なプラスミドであるのに対して、枯草菌における複製に機能する配列番号9で示される複製タンパク質(RepB)をコードするプラスミドの様にローリングサークル型の複製を行うプラスミドは、一般に高コピー数であることが知られ、前述したシータ型複製を行う低コピー数プラスミドと異なる(非特許文献7)。斯かるバチルス・チューリンゲンシスを用いて、そのプラスミド上のCryタンパク質遺伝子(cry遺伝子)から当該Cryタンパク質を菌体内生産する場合においては、cry遺伝子のコピー数は飽和に達しており、コピー数の増加により生産量が増加しないことが示唆されている(非特許文献6)。また、高コピー数プラスミドへcry遺伝子をクローニングすると生理的平衡が崩れ、胞子形成が阻害されることも観察されている(非特許文献6)。また、Cryタンパク質の結晶構造の形成や可溶性には、2次構造や付加的な構成成分などの様々な因子が関係していることが示唆されており(非特許文献6)、単純なcry遺伝子発現の増加により当該タンパク質を高生産できるとは云えないと考えられていた。例えば、バチルス・チューリンゲンシス由来のCry5Bタンパク質をバチルス・ズブチリス PY79にて発現させた場合に、バチルス・チューリンゲンシスの該タンパク質の生産性75mg/Lに対して、バチルス・ズブチリス PY79での生産性は10mg/Lに過ぎないことが報告されている(非特許文献8、特許文献2)。また、ラクトコッカス・ラクティスにて、高コピー数プラスミドを用いて高発現プロモーターの制御下でCry5Bタンパク質遺伝子を発現させた場合に、バチルス・チューリンゲンシスの該タンパク質の生産性はウエスタンブロットレベルしか検出できないことも報告されている(非特許文献9)。
斯様に、一般的に菌体外タンパク質の発現向上のために検討されてきた手法が、菌体内タンパク質の生産に適用可能であるとは云えず、特にCryタンパク質を高発現するための効率な手法は見出されていない。

本発明は以下の1)~2)を提供するものである。
1)Cryタンパク質をコードする遺伝子とσA依存性プロモーター又はσH依存性プロモーターを含む制御領域とが作動可能に連結して組み込まれてなる発現プラスミドでバチルス属細菌を形質転換し、当該形質転換細胞を培養することを含むCryタンパク質又はそれを含有する培養産物の製造方法であって、前記発現プラスミドが、配列番号9で示されるアミノ酸配列からなる複製タンパク質又はこれとアミノ酸配列において80%以上の同一性を有し、且つ複製開始に関与するタンパク質をコードするポリヌクレオチドを含むプラスミドである、方法。
2)バチルス属細菌内でCryタンパク質を発現するための発現プラスミドであって、配列番号9で示されるアミノ酸配列からなる複製タンパク質又はこれとアミノ酸配列において80%以上の同一性を有し、且つ複製開始に関与するタンパク質をコードするポリヌクレオチドを含み、Cryタンパク質をコードする遺伝子とσA依存性プロモーター又はσH依存性プロモーターを含む制御領域とが作動可能に連結されてなる発現プラスミド。
1)Cryタンパク質をコードする遺伝子とσA依存性プロモーター又はσH依存性プロモーターを含む制御領域とが作動可能に連結して組み込まれてなる発現プラスミドでバチルス属細菌を形質転換し、当該形質転換細胞を培養することを含むCryタンパク質又はそれを含有する培養産物の製造方法であって、前記発現プラスミドが、配列番号9で示されるアミノ酸配列からなる複製タンパク質又はこれとアミノ酸配列において80%以上の同一性を有し、且つ複製開始に関与するタンパク質をコードするポリヌクレオチドを含むプラスミドである、方法。
2)バチルス属細菌内でCryタンパク質を発現するための発現プラスミドであって、配列番号9で示されるアミノ酸配列からなる複製タンパク質又はこれとアミノ酸配列において80%以上の同一性を有し、且つ複製開始に関与するタンパク質をコードするポリヌクレオチドを含み、Cryタンパク質をコードする遺伝子とσA依存性プロモーター又はσH依存性プロモーターを含む制御領域とが作動可能に連結されてなる発現プラスミド。
本発明は、Cryタンパク質をバチルス属細菌の菌体内で高生産するための方法を提供することに関する。
本発明者らは、バチルス属細菌、好ましくは、枯草菌によるCryタンパク質の菌体内生産について検討したところ、意外にも菌体外タンパク質の生産に用いられる特定の高発現プラスミドを用いることにより、菌体内に著量のCryタンパク質が生産され、当該タンパク質は野生型と同程度の活性を有することを見出した。
本発明によれば、バチルス属細菌、好ましくは、枯草菌(Bacillus subtilis)を用いたCryタンパク質の高発現系が提供され、これを用いることにより、Cryタンパク質又はそれを含有する培養産物を効率よく製造することができる。
本明細書において、アミノ酸配列及びヌクレオチド配列の同一性はLipman-Pearson法(Lipman,DJ.,Pearson.WR.:Science,227,1435-1441,1985)によって計算される。具体的には、遺伝情報処理ソフトウェアGenetyx-Win(ソフトウェア開発)のホモロジー解析(Search homology)プログラムを用いて、Unit size to compare(ktup)を2として解析を行うことにより算出される。
本明細書において、別途定義されない限り、アミノ酸配列又はヌクレオチド配列におけるアミノ酸又は塩基の欠失、置換、付加又は挿入に関して使用される「1又は数個」とは、例えば、1~12個、好ましくは1~8個、より好ましくは1~4個であり得る。また本明細書において、アミノ酸又は塩基の「付加」には、配列の一末端及び両末端への1~数個のアミノ酸の又は塩基の付加が含まれる。
本明細書において、ハイブリダイゼーションに関する「ストリンジェントな条件下」としては、Molecular Cloning-A LABORATORY MANUAL THIRD EDITION(Joseph Sambrook,David W.Russell,Cold Spring Harbor Laboratory Press,2001)に記載の条件が挙げられ、例えば、6×SSC(1×SSCの組成:0.15M塩化ナトリウム、0.015Mクエン酸ナトリウム、pH7.0)、0.5%SDS、5×デンハート及び100mg/mLニシン精子DNAを含む溶液にプローブとともに65℃で8~16時間恒温し、ハイブリダイズさせる条件であり得る。
本明細書において、遺伝子の上流及び下流とは、それぞれ、対象として捉えている遺伝子又は領域の5'側及び3'側に続く領域を示す。別途定義されない限り、遺伝子の上流及び下流とは、それぞれ遺伝子の翻訳開始点からの上流領域及び遺伝子の下流領域には限定されない。
本明細書において、転写開始制御領域はプロモーター及び転写開始点を含む領域であり、翻訳開始制御領域は開始コドンと共にリボソーム結合部位を形成するShine-Dalgarno(SD)配列に相当する部位である(Shine,J.,Dalgarno,L.:Proc.Natl.Acad.Sci.USA.,71,1342-1346,1974)。
本発明において、Cryタンパク質は、バチルス・チューリンゲンシス(Bacillus thuringiensis)が産生する結晶性殺虫タンパク質を意味する。Cryタンパク質は、タンパク質の1次構造より、Cry1からCry75までのクラスに分類されており(Microbiology and Molecular Biology Reviews(1998) 62, 807-813. Revision of the Nomenclature for the Bacillus thuringiensis Pesticidal Crystal Proteins, http://www.lifesci.sussex.ac.uk/Home/Neil#Crickmore/Bt/.(2017年12月7日)、各クラスは配列類似性の程度によりさらにサブクラスに分類されている。例えば、Cry1クラスに属するものは100種以上存在する。主要なCryタンパク質を、以下の表1-1~1-3に示す。なお、該表に示すアクセス番号はGenBank Accession No.である。
このうち、本発明の方法は、Cry1Aタンパク質、Cry1Fタンパク質、Cry2Aタンパク質、Cry34Aタンパク質、Cry35Aタンパク質、Cry3Aタンパク質、Cry3Bタンパク質、Cry21タンパク質、Cry14Aタンパク質、Cry6Aタンパク質、Cry13タンパク質、Cry5Bタンパク質、Cry4Aaタンパク質、Cry4Baタンパク質、Cry11Aaタンパク質の製造に、より好適であり、Cry5Bタンパク質Cry4Aaタンパク質、Cry4Baタンパク質、Cry11Aaタンパク質の製造に、より好適である。
Cry5Bタンパク質は、土壌伝播蠕虫症に対して有効であることが知られている殺線虫タンパク質である(Cappello M et al. Proc. Natl. Acad. Sci. U. S. A. 103:15154-15159, Hu Y et al. PLoS Negl. Trop. Dis. 4:e614)。Cry5Bのアミノ酸配列の例としては、配列表の配列番号2で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号1で示される。
Cry5Bタンパク質は、土壌伝播蠕虫症に対して有効であることが知られている殺線虫タンパク質である(Cappello M et al. Proc. Natl. Acad. Sci. U. S. A. 103:15154-15159, Hu Y et al. PLoS Negl. Trop. Dis. 4:e614)。Cry5Bのアミノ酸配列の例としては、配列表の配列番号2で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号1で示される。
Cry4Aaタンパク質、Cry4Baタンパク質及びCry11Aaタンパク質は、蚊に対して殺虫活性を有するタンパク質として知られている(FEMS Microbiol Lett 266 (2007) 163-169, Mhalakshmi A at al. Advances Microbiol. 2 (2012) 216-226)。
Cry4Aaのアミノ酸配列の例としては、配列表の配列番号4で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号3で示される。
Cry4Baのアミノ酸配列の例としては、配列表の配列番号6で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号5で示される。
Cry11Aaのアミノ酸配列の例としては、配列表の配列番号8で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号7で示される。
Cry4Aaのアミノ酸配列の例としては、配列表の配列番号4で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号3で示される。
Cry4Baのアミノ酸配列の例としては、配列表の配列番号6で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号5で示される。
Cry11Aaのアミノ酸配列の例としては、配列表の配列番号8で示され、当該タンパク質をコードする遺伝子の塩基配列は配列表の配列番号7で示される。
斯かる天然のタンパク質の中には生態型(ecotype)等の違いによる遺伝子の変異、あるいはよく似たアイソザイムの存在などに起因して1から数個のアミノ酸変異を有する変異タンパク質が存在することは周知である。
よって、本発明のCryタンパク質には、表1に示されるCryタンパク質の他に当該タンパク質を構成するアミノ酸配列において、1ないし数個のアミノ酸残基が付加又は置換、或いは1ないし数個のアミノ酸残基が欠失したものであって、同様の殺虫活性を有する、当該Cryタンパク質の変異体も包含される。
よって、本発明のCryタンパク質には、表1に示されるCryタンパク質の他に当該タンパク質を構成するアミノ酸配列において、1ないし数個のアミノ酸残基が付加又は置換、或いは1ないし数個のアミノ酸残基が欠失したものであって、同様の殺虫活性を有する、当該Cryタンパク質の変異体も包含される。
例えば、Cry5Bタンパク質、Cry4Aaタンパク質、タンパク質Cry4Ba、Cry11Aaタンパク質であれば、以下の(A)~(C)が包含される。
(A)配列番号2、4、6又は8に示すアミノ酸配列からなるタンパク質
(B)配列番号2、4、6又は8に示すアミノ酸配列において1又は数個のアミノ酸が欠失、置換、付加又は挿入されたアミノ酸配列からなり、且つ殺虫活性を有するタンパク質(C)配列番号2、4、6又は8に示すアミノ酸配列と80%以上、より好ましくは90%以上、より好ましくは95%以上、より好ましくは96%以上、より好ましくは97%以上、より好ましくは98%以上、より好ましくは99%以上の同一性を有するアミノ酸配列からなり、且つ殺虫活性を有するタンパク質
(A)配列番号2、4、6又は8に示すアミノ酸配列からなるタンパク質
(B)配列番号2、4、6又は8に示すアミノ酸配列において1又は数個のアミノ酸が欠失、置換、付加又は挿入されたアミノ酸配列からなり、且つ殺虫活性を有するタンパク質(C)配列番号2、4、6又は8に示すアミノ酸配列と80%以上、より好ましくは90%以上、より好ましくは95%以上、より好ましくは96%以上、より好ましくは97%以上、より好ましくは98%以上、より好ましくは99%以上の同一性を有するアミノ酸配列からなり、且つ殺虫活性を有するタンパク質
本発明のCryタンパク質をコードする遺伝子(cry遺伝子とも称する)は、その類は特に限定されず、天然由来のDNA、組換えDNA、化学合成DNAの何れでもよく、またゲノムDNAクローン、cDNAクローンの何れでもよい。
本発明のcry遺伝子は典型的には、上記表1で示す各cry遺伝子を指すが、天然の遺伝子の中には、生態型(ecotype)等の違いに起因する少数の変異やよく似たアイソザイムの存在に起因する少数の変異が存在することは当業者に周知である。従って、本発明のcry遺伝子は、表1で示される遺伝子のみに限定されるわけではなく、上記のCryタンパク質をコードする全ての遺伝子が包含される。
例えば、cry5B遺伝子、cry4Aa遺伝子、cry4Ba遺伝子、cry11Aa遺伝子であれば、以下の(a)~(f)が包含される。
(a)配列番号1、3、5又は7に示すヌクレオチド配列からなるポリヌクレオチド

本発明において、Cryタンパク質又はそれを含有する培養産物は、前述したcry遺伝子とσA依存性プロモーター又はσH依存性プロモーターを含む制御領域とを作動可能に連結して発現プラスミドに組み込み、当該プラスミドでバチルス属細菌を形質転換し、当該形質転換細胞を培養することにより製造され、発現プラスミドとして、配列番号9で示されるアミノ酸配列からなる複製タンパク質又はこれとアミノ酸配列において80%以上の同一性を有し、且つ複製開始に関与するタンパク質をコードするポリヌクレオチドを含むプラスミドが用いられる。
複製タンパク質(Rep)は、プラスミドの複製開始に働くイニシエータータンパク質であるが、本発明で用いられるプラスミドは、枯草菌における複製に必要な配列番号9で示される複製タンパク質を包含する。例えば、配列番号9で示される複製タンパク質は、pAMα1に存在する。pAMα1の複製タンパク質は2種類存在するが、本発明において用いられる複製タンパク質は、配列番号9で示されるアミノ酸からなるタンパク質(RepB)である。また、当該タンパク質とアミノ酸配列において80%以上、より好ましくは90%以上、より好ましくは95%以上、より好ましくは96%以上、より好ましくは97%以上、より好ましくは98%以上、より好ましくは99%以上の同一性を有し、且つ複製開始に関与するタンパク質も当該タンパク質と同様に用いることができる。斯かるプラスミド複製タンパク質としては、例えば、配列番号9で示されるアミノ酸配列において、1若しくは数個のアミノ酸が欠失、置換若しくは付加されたものが包含される。例えば、特許第5361484号に記載の変異タンパク質、すなわち、配列番号9で示されるアミノ酸配列において、(a)48位、(b)262位、(c)149位及び(d)198位から選ばれる1以上のアミノ酸残基が特定のアミノ酸残基に置換してなるタンパク質が挙げられる。
上記プラスミドには、複製タンパク質をコードするポリヌクレオチドの他に、複製開始に関与するタンパク質をコードするポリヌクレオチド等が含まれる。また、当該プラスミドには、これらの他に、薬剤耐性遺伝子、マルチクローニングサイト等を適宜含むことができる。
本発明において用いられる好適なプラスミドとしては、例えばpHY300PLK、特許第5361484号に記載のプラスミド等が挙げられる。
pHY300PLKは、Escherichia coli とBacillus subtilisの両方へDNAを形質転換できるシャトルベクターで、E.coliのプラスミドpACYC177とStreptococcus faecalisのプラスミドpAMα1由来のDNAより構築され、pAMα1の複製領域にはRepBが含まれる。

当該プラスミドに、目的のCryタンパク質をコードする遺伝子と、σA因子依存性プロモーター又はσH因子依存性プロモーターを含む制御領域とが作動可能に連結され、Cryタンパク質を枯草菌の菌体内に生産させ得る組換えプラスミド(発現ベクター)が構築される。
ここで、「制御領域と作動可能に連結された遺伝子」とは、当該制御領域による制御の下に発現し得るように配置されている遺伝子をいう。
ここで、プロモーターとは、所定の遺伝子におけるコーディング領域の上流に存在し、RNAポリメラーゼが相互作用して当該遺伝子の転写を制御する機能を有する領域と定義される。より具体的には、プロモーターとは、所定の遺伝子におけるコーディング領域の上流200~600塩基程度の領域を意味する。
また、プロモーターを含む制御領域としては、転写開始制御領域及び翻訳開始制御領域が挙げられる。当該制御領域は、好ましくは、宿主内における下流の遺伝子の発現を高める機能を有し、より好ましくは、下流遺伝子を構成的に発現又は高発現させる機能を有する。
ここで、「制御領域と作動可能に連結された遺伝子」とは、当該制御領域による制御の下に発現し得るように配置されている遺伝子をいう。
ここで、プロモーターとは、所定の遺伝子におけるコーディング領域の上流に存在し、RNAポリメラーゼが相互作用して当該遺伝子の転写を制御する機能を有する領域と定義される。より具体的には、プロモーターとは、所定の遺伝子におけるコーディング領域の上流200~600塩基程度の領域を意味する。
また、プロモーターを含む制御領域としては、転写開始制御領域及び翻訳開始制御領域が挙げられる。当該制御領域は、好ましくは、宿主内における下流の遺伝子の発現を高める機能を有し、より好ましくは、下流遺伝子を構成的に発現又は高発現させる機能を有する。
σA因子依存性プロモーター及びσH因子依存性プロモーターは、胞子形成微生物において、胞子形成期におけるスポアコートタンパク沈着期より前において働くプロモーターである。
σA依存性プロモーター又はσH依存性プロモーターを含む制御領域が、Cryタンパク質をコードする遺伝子の起源微生物における当該遺伝子の制御領域とは異なる制御領域、から選択されることが好ましい。
σA因子依存性プロモーターとしては、特に限定されないが、枯草菌ファージSP01プロモーター(Proc. Natl. Acd. Sci. USA. (1984) 81:439-443.)、veg遺伝子、amyE(アミラーゼ)遺伝子、aprE(ズブチリシン)遺伝子及びS237(S237セルラーゼ、特開2000-210081号公報)遺伝子のプロモーターを挙げることができる。σH因子依存性プロモーターとしては、特に限定されないが、citG遺伝子、spoVS遺伝子及びspoVG(Proc. Natl. Acd. Sci. USA. (1986) 83:9438-9442.)遺伝子のプロモーターが挙げられる。
本発明においては、σA因子依存性プロモーターとしてはBacllus sp.KSM-S237株のセルラーゼ遺伝子のプロモーターがより好ましく、σH因子依存性プロモーターとしては枯草菌Bacillus subtilis Marburg No.168(枯草菌168株)のspoVG遺伝子(BG10112)のプロモーターがより好ましい。
σA依存性プロモーター又はσH依存性プロモーターを含む制御領域が、Cryタンパク質をコードする遺伝子の起源微生物における当該遺伝子の制御領域とは異なる制御領域、から選択されることが好ましい。
σA因子依存性プロモーターとしては、特に限定されないが、枯草菌ファージSP01プロモーター(Proc. Natl. Acd. Sci. USA. (1984) 81:439-443.)、veg遺伝子、amyE(アミラーゼ)遺伝子、aprE(ズブチリシン)遺伝子及びS237(S237セルラーゼ、特開2000-210081号公報)遺伝子のプロモーターを挙げることができる。σH因子依存性プロモーターとしては、特に限定されないが、citG遺伝子、spoVS遺伝子及びspoVG(Proc. Natl. Acd. Sci. USA. (1986) 83:9438-9442.)遺伝子のプロモーターが挙げられる。
本発明においては、σA因子依存性プロモーターとしてはBacllus sp.KSM-S237株のセルラーゼ遺伝子のプロモーターがより好ましく、σH因子依存性プロモーターとしては枯草菌Bacillus subtilis Marburg No.168(枯草菌168株)のspoVG遺伝子(BG10112)のプロモーターがより好ましい。
σA因子依存性プロモーターを含む制御領域としては、Bacillus sp.KSM-S237株のセルラーゼ遺伝子の制御領域が好適に挙げられる。当該制御領域は、具体的には当該セルラーゼ遺伝子の転写開始制御領域及び翻訳開始領域であり(配列番号10)、好ましくは配列番号10で示される塩基配列、または当該塩基配列に対して80%以上、より好ましくは90%以上、さらに好ましくは95%以上、さらにより好ましくは98%以上の同一性を有する塩基配列、或いは上記いずれかの塩基配列の一部が欠失した塩基配列である。上記配列番号10で示される塩基配列に対して80%以上、より好ましくは90%以上、さらに好ましくは95%以上、さらにより好ましくは98%以上の同一性を有する塩基配列とは、上記同一性を有しており、且つσA因子依存性プロモーターとして遺伝子の転写に関わる機能、および、翻訳に関わる機能を保持している配列を意味する。また、上記塩基配列の一部が欠失した塩基配列とは、上記塩基配列の一部を欠失しているが、σA因子依存性プロモーターとして遺伝子の転写に関わる機能、および、翻訳に関わる機能を保持している配列を意味する。このうち、より好ましいσA因子依存性プロモーターとして、配列番号11で示される塩基配列からなるプロモーターが挙げられる。当該プロモーターは上記Bacillus sp.KSM-S237株のセルラーゼ遺伝子の制御領域においてCre様配列が削除された配列からなり(特開2011-103875号公報)、配列番号10で示される塩基配列に対して95%の配列同一性を有するプロモーターである。
ここで、「σA因子依存性プロモーターとして機能する」とは、RNAポリメラーゼであるσA因子によって特異的に転写制御されることを意味する。この特異性は、評価対象となるポリヌクレオチドの下流にレポーター遺伝子を結合し、σA因子存在下及び不存在下でのレポーター遺伝子の発現を観察することによって評価することができる。
ここで、「σA因子依存性プロモーターとして機能する」とは、RNAポリメラーゼであるσA因子によって特異的に転写制御されることを意味する。この特異性は、評価対象となるポリヌクレオチドの下流にレポーター遺伝子を結合し、σA因子存在下及び不存在下でのレポーター遺伝子の発現を観察することによって評価することができる。
また、σH因子依存性プロモーターを含む制御領域としては、枯草菌Bacillus subtilis Marburg No.168(枯草菌168株)のspoVG遺伝子の制御領域が好適に挙げられる。当該制御領域は、具体的には当該spoVG遺伝子(BG10112)の転写開始制御領域及び翻訳開始領域であり(配列番号12)、好ましくは配列番号12で示される塩基配列、または当該塩基配列に対して80%以上、より好ましくは90%以上、さらに好ましくは95%以上、さらにより好ましくは98%以上の同一性を有する塩基配列、或いは上記いずれかの塩基配列の一部が欠失した塩基配列である。上記配列番号12で示される塩基配列に対して80%以上、より好ましくは90%以上、さらに好ましくは95%以上、さらにより好ましくは98%以上の同一性を有する塩基配列とは、上記同一性を有しており、且つσH因子依存性プロモーターとして遺伝子の転写に関わる機能、および、翻訳に関わる機能を保持している配列を意味する。また、上記塩基配列の一部が欠失した塩基配列とは、上記塩基配列の一部を欠失しているが、σH因子依存性プロモーターとして遺伝子の転写に関わる機能、および、翻訳に関わる機能を保持している配列を意味する。ここで、「σH因子依存性プロモーターとして機能する」とは、RNAポリメラーゼであるσH因子によって特異的に転写制御されることを意味する。この特異性は、評価対象となるポリヌクレオチドの下流にレポーター遺伝子を結合し、σH因子存在下及び不存在下でのレポーター遺伝子の発現を観察することによって評価することができる。
上記cry遺伝子、及び上記制御領域を含むプラスミドへの挿入は、当該分野における通常の方法に従って行うことができる。例えば、上記cry遺伝子、及び制御領域の断片をPCR等によって増幅し、これらの断片を制限酵素法等によってプラスミドに挿入し、連結すればよい。あるいは、上記遺伝子及び制御領域の断片を予め連結した断片を作製し、これをプラスミドに挿入してもよい。その際、プラスミド上において、上流から制御領域断片、cry遺伝子の断片の順で連結されているようにする。簡便には、市販のライゲーションキット(例えば、タカラバイオ製等)を用いることもできる。
構築されたプラスミドを宿主細胞に導入する方法としては、Sambrook,J.ら,Molecular Cloning, A Laboratory Manual(2nd edition),Cold Spring Harbor Laboratory,1.74(1989)に記載の塩化カルシウム法または塩化カルシウム/塩化ルビジウム法、エレクトロポレーション法、エレクトロインジェクション法、PEGなどの化学的な処理による方法、遺伝子銃などを用いる方法などが挙げられる。
上記発現プラスミドを導入する宿主細胞としては、バチルス属細菌、好ましくは枯草菌(Bacillus subtilis)、バチルス・メガテリウム(Bacillus megaterium)又はバチルス・ブレビス(Bacillus brevis)が使用されるが、これは野生型のものでも変異を施したものでもよい。バチルス属に属する微生物としては、具体的には、Bacillus subtilis、Bacillus cereus、Bacillus thuringiensis、Bacillus anthracis、Bacillus stearotheromophilus、Bacillus coagulans、Bacillus megaterium、Bacillus halodurans、Bacillus brevis(Brevibacillus brevis)、Bacillus choshinensis(Brevibacillus choshinensis)、Bacillus pumilus、Bacillus alcalophilus、Bacilus amylolyticus、Bacilus amyloliquefaciens、Bacilus liqueniformis、Bacillus polymyxa(Paenibacillus polymyxa)、Bacillus sphaericus、Bacillus firmus、Bacillus clausii、Bacillus macerans等を挙げることができる。なお、Bacillus brevisはBrevibacillus属に分類されて、株によってBrevibacillus brevisあるいはBrevibacillus choshinensis等と記載される場合があり、Bacillus polymyxaはPaenibacillus属に分類されてPaenibacillus polymyxaと記載される場合があり、Bacillus stearotheromophilusはGeobacillus属に分類されてGeobacillus stearotheromophilusと記載される場合がある。しかしながら本明細書においては、Bacillus brevis、Bacillus polymyxa、及びBacillus stearotheromophilusは、共にBacillus属に属する細菌として定義する。すなわち、本発明において、Bacillus属に属する微生物といった場合、Brevibacillus brevis、Brevibacillus choshinensisと記載される微生物、Paenibacillus polymyxaと記載される微生物及びGeobacillus stearotheromophilusと記載される微生物を含む意味として解釈する。なお、上記バチルス属に属する微生物は各微生物保存機関より購入可能である。
野生型のバチルス属細菌としては、例えば枯草菌Bacillus subtilis Marburg No.168(枯草菌168株)が挙げられ、枯草菌変異株としては、Cryタンパク質の製造に適するものであれば特に限定されないが、例えば枯草菌野生株のゲノム上に生存、増殖やタンパク質生産にとって不必要な領域が欠失した枯草菌株、特定のプロテアーゼ遺伝子が欠失した枯草菌株、胞子形成期特異的なσ因子遺伝子が欠失した枯草菌株、或いはこれらの変異を組み合わせた枯草菌株等が挙げられる。
野生型のバチルス属細菌としては、例えば枯草菌Bacillus subtilis Marburg No.168(枯草菌168株)が挙げられ、枯草菌変異株としては、Cryタンパク質の製造に適するものであれば特に限定されないが、例えば枯草菌野生株のゲノム上に生存、増殖やタンパク質生産にとって不必要な領域が欠失した枯草菌株、特定のプロテアーゼ遺伝子が欠失した枯草菌株、胞子形成期特異的なσ因子遺伝子が欠失した枯草菌株、或いはこれらの変異を組み合わせた枯草菌株等が挙げられる。
ゲノムの大領域が欠失した枯草菌変異株は、枯草菌野生株(例えば、枯草菌168株)のゲノムと比べて、そのゲノム中の大領域が欠失したゲノムを有するものであり、例えば特許4955358号に記載の変異株が挙げられる。例えば、枯草菌野生株のゲノム中の、prophage6(yoaV-yobO)領域、prophage1(ybbU-ybdE)領域、prophage4(yjcM-yjdJ)領域、PBSX(ykdA-xlyA)領域、prophage5(ynxB-dut)領域、prophage3(ydiM-ydjC)領域、spb(yodU-ypqP)領域、pks(pksA-ymaC)領域、skin(spoIVCB-spoIIIC)領域、pps(ppsE-ppsA)領域、prophage2(ydcL-ydeJ)領域、ydcL-ydeK-ydhU領域、yisB-yitD領域、yunA-yurT領域、cgeE-ypmQ領域、yeeK-yesX領域、pdp-rocR領域、ycxB-sipU領域、prophage7(yrkM-yraK、又はyrkS-yraK)領域、sbo-ywhH領域、yybP-yyaJ領域及びyncM-fosB領域からなる群より選択される領域の1以上が欠失した枯草菌変異株が挙げられ、好ましくは、prophage6(yoaV-yobO)領域、prophage1(ybbU-ybdE)領域、prophage4(yjcM-yjdJ)領域、PBSX(ykdA-xlyA)領域、prophage5(ynxB-dut)領域、prophage3(ydiM-ydjC)領域、spb(yodU-ypqP)領域、pks(pksA-ymaC)領域、skin(spoIVCB-spoIIIC)領域、pps(ppsE-ppsA)領域、prophage2(ydcL-ydeJ)領域、ydcL-ydeK-ydhU領域、yisB-yitD領域、yunA-yurT領域、cgeE-ypmQ領域、yeeK-yesX領域、pdp-rocR領域、ycxB-sipU領域、prophage7(yrkS-yraK)領域、sbo-ywhH領域、yybP-yyaJ領域及びyncM-fosB領域の全てを欠失している枯草菌MGB874株が挙げられる。当該MGB874株は国立遺伝学研究所NBRP(ナショナルバイオリソースプロジェクト)より分譲可能となっている(http://www.shigen.nig.ac.jp/bsub/kaoListAction.do)。さらに、MGB874株の変異株として、例えば特開2017-79639号に記載の、abrB遺伝子又はそれに相当する遺伝子が恒常的に発現するように遺伝子構築され、且つkinA遺伝子が欠失又は不活性化されている、枯草菌変異株(MGB874abrB* ΔkinA)が挙げられる。
特定のプロテアーゼ遺伝子が欠失した枯草菌株としては、例えば特許4485341号に記載の、枯草菌のaprX遺伝子と、枯草菌のaprE、nprB、nprE、bpr、vpr、mpr、epr及びwprAから選ばれる1以上の遺伝子を欠失又は不活性化させた枯草菌、好ましくは、aprX、aprE、nprB、nprE、bpr、vpr、mpr、epr及びwprAの9個の遺伝子を欠失した9重欠失株(Dpr9株)が挙げられる。
また、胞子形成期特異的なσ因子遺伝子が欠失した枯草菌株としては特許4336082号に記載の、sigF、sigG、及び、sigEから選ばれる1以上の遺伝子を欠失又は不活性化した枯草菌株、好ましくはsigE欠失株(ΔsigE株)、sigF欠失株(ΔsigF株)、より好ましくは、sigF欠失株(ΔsigF株)が挙げられる。
Cryタンパク質の生産の点から、上記MGB874株に対してsigFを欠失させたMGB874ΔsigF変異株、及び上記Dpr9株に対してsigFを欠失させたDpr9ΔsigF変異株を用いるのより好ましい。

本発明のCryタンパク質は、上記の如く調製された発現ベクターを含む形質転換枯草菌を栄養培地で培養することによって発現(生産)することができる。栄養培地は、枯草菌(形質転換体)の生育に必要な炭素源、無機窒素源もしくは有機窒素源を含んでいることが好ましい。炭素源としては、たとえばグルコース、デキストラン、可溶性デンプン、ショ糖、メタノールなどが、例示される。無機窒素源もしくは有機窒素源としては、例えばアンモニウム塩類、硝酸塩類、アミノ酸、コーンスチープ・リカー、ペプトン、カゼイン、肉エキス、大豆粕、バレイショ抽出液などが例示される。また、所望により他の栄養素(例えば無機塩(例えば、塩化ナトリウム、塩化カルシウム、リン酸二水素ナトリウム、塩化マグネシウム)、ビタミン類、抗生物質(例えばテトラサイクリン、ネオマイシン、カナマイシン、スペクチノマイシン、エリスロマイシン等)など)を含んでいてもよい。培養は、当業界において知られている方法により行われる。培養条件、例えば温度、通気撹拌条件、培地のpH及び培養時間等は、本発明のタンパク質が大量に生産されるように適宜選択される。
本発明のCryタンパク質を含む上記培養により得られる培養物は、遠心分離やろ過などの操作により宿主細胞を集め、これを適当な緩衝液(例えば濃度が10M~100mM程度のトリス緩衝液、リン酸緩衝液、HEPES緩衝液、MES緩衝液などの緩衝液(pH5.0~9.0の範囲が望ましい)や水に懸濁することで得られる。さらに、公知の細胞破砕法、例えばリゾチーム、凍結融解、超音波処理、フレンチプレス、ビーズ破砕等を適宜組み合わせることにより細胞を破壊し、遠心分離によりCryタンパク質を回収することもできる。また、上記培養物を、カルバクロールなどの殺菌効果のある物質を加えてインキュベーションすることにより殺菌処理することもできる(特許文献3)。
回収されたCryタンパク質は、ショ糖密度勾配法、再結晶法、イオン交換クロマトグラフィー、ゲルろ過、疎水性クロマトグラフィー、等電点クロマトグラフィー、Cryタンパク質に対するポリクローナル抗体等をリガンドとしたアフィニテイーカラム等を利用して適宜精製することができる。
上述した実施形態に関し、本発明においてはさらに以下の態様が開示される。

実施例1 Cry5Bタンパク質を含む培養物の製造
1.人工遺伝子の合成
Bacillus thuringiensis YBT-1518由来の殺虫タンパク質遺伝子cry5B(GenBank:CP005935.1、配列番号1)をGenScript社(アメリカ)により人工合成した。合成した遺伝子の3738 bpはpUC57のKpnIとHindIIIサイトにクローニングし、pUC57-cry5Bを得た。
1.人工遺伝子の合成
Bacillus thuringiensis YBT-1518由来の殺虫タンパク質遺伝子cry5B(GenBank:CP005935.1、配列番号1)をGenScript社(アメリカ)により人工合成した。合成した遺伝子の3738 bpはpUC57のKpnIとHindIIIサイトにクローニングし、pUC57-cry5Bを得た。
2.発現プラスミドの構築
全ての発現プラスミドの構築において、ベクターとしてpHY300PLK(タカラバイオ)、およびターミネターとしてS237セルラーゼ遺伝子由来配列を用いた。プロモーターはS237セルラーゼ遺伝子[Hakamada et al, Biosci. Biotechnol. Biochem., 64 (2000), 2281-2289]あるいは枯草菌168株のspoVG遺伝子由来、シグナル配列はS237セルラーゼ由来あるいは無し、cry5B遺伝子は全長(cry5B)あるいは短縮型(cry5Bt)の組み合わせで、8種のプラスミドの作製を行った(図1-A~D)。cry5B遺伝子の全長(3738bp)は、配列番号1で示されるが、塩基番号2095-3738番の塩基配列は線虫の腸内において分解される領域で、塩基番号1-2094番の塩基配列は分解された後の活性化Crystal toxinをコードする配列[Hui et al, 2012, Biochemistry, vol 11, p 9911-21]であり、全長型は全配列(1-3738番)、短縮型は塩基番号1-2094番の配列である。
全ての発現プラスミドの構築において、ベクターとしてpHY300PLK(タカラバイオ)、およびターミネターとしてS237セルラーゼ遺伝子由来配列を用いた。プロモーターはS237セルラーゼ遺伝子[Hakamada et al, Biosci. Biotechnol. Biochem., 64 (2000), 2281-2289]あるいは枯草菌168株のspoVG遺伝子由来、シグナル配列はS237セルラーゼ由来あるいは無し、cry5B遺伝子は全長(cry5B)あるいは短縮型(cry5Bt)の組み合わせで、8種のプラスミドの作製を行った(図1-A~D)。cry5B遺伝子の全長(3738bp)は、配列番号1で示されるが、塩基番号2095-3738番の塩基配列は線虫の腸内において分解される領域で、塩基番号1-2094番の塩基配列は分解された後の活性化Crystal toxinをコードする配列[Hui et al, 2012, Biochemistry, vol 11, p 9911-21]であり、全長型は全配列(1-3738番)、短縮型は塩基番号1-2094番の配列である。
構築方法はIn-Fusion(登録商標) HD EcoDryTM Cloning Kitのプロトコールに従った。各プラスミドの作製過程は図1-A~Dに示した。
2.1 pPsScry5BとpPsScry5Btの構築
表2に示すvect+psFとvect+tRのプライマーセットを用いて、pHYS237 DNAを鋳型にS237のプロモーター、シグナル及びターミネター領域を含むベクターを増幅した。次に、pUC57-cry5B DNAを鋳型にcry5BpssFとcry5BtR(あるいはT237-cry5BatRN)のプライマーセットを用いてcry5B遺伝子全長(あるいは短縮型)のインサートをPCRで増幅した。続いて、In-Fusion(登録商標) HD EcoDryTM Cloning Kit(Clontech社)を用いてベクターとインサートを連結した後、大腸菌HB101コンピテントセル(タカラバイオ)に形質転換した。テトラサイクリン耐性により選抜した形質転換体をコロニーPCRにより確認し、pPsScry5B(あるいはpPsScry5Bt)と命名した(図1-A)。抽出したプラスミドについて、さらにPCR確認、また、制限酵素EcoRI、SpeIとXbaIを用いてプラスミドの消化パターンの確認を行った。

2.3 pPvcry5BとpPvcry5Btの構築
表2に示すvectRとvect+tRのプライマーセットを用いて、pHYS237 DNAを鋳型にS237のターミネター領域を含むベクターを増幅した。次に、pUC57-cry5B DNAを鋳型にcry5BaFとcry5BtR(あるいはT237-cry5BatRN)のプライマーセットを用いてcry5B遺伝子全長(あるいは短縮型)のインサートをPCRで増幅した。また、Vect-PvgFとcry5Ba-PvgRのプライマーセットを用いて、枯草菌168株のゲノムDNAを鋳型にspoVG遺伝子のプロモーター領域を2つ目のインサートとして増幅した。続いて、In-Fusion(登録商標) HD EcoDryTM Cloning Kit(Clontech社)を用いてベクターと2つのインサートを連結した後、大腸菌HB101コンピテントセル(タカラバイオ)に形質転換した。テトラサイクリン耐性により選抜した形質転換体をコロニーPCRにより確認し、pPvcry5B(あるいはpPvcry5Bt)と命名した(図1-C)。抽出したプラスミドについて、さらにPCR確認、また、制限酵素EcoRI、SpeIとXbaIを用いてプラスミドの消化パターンの確認を行った。

2.4 pPvScry5BとpPvScry5Btの構築

3.プラスミドのシーケンス確認
構築した8種のプラスミドについてシーケンス確認を行った。シーケンスの鋳型をPCRにより調製し、全長cry5Bの4種のプラスミドは、表2示したfrag1-Fとfrag1-Rまたは frag2-Fとfrag2-Rのプライマーを用いてそれぞれ5'側と3'側の断片を調製した。短縮型cry5Bの4種のプラスミドは、表2に示したfrag1-Fとfrag2-Rのプライマーを用いて断片の調製を行った。それらのPCR産物について表2に示したSEQ-P1~SEQ-P10の10個のプライマーを用いたシーケンス解析を行った。解析の結果、すべてのプラスミドについて、変異がなかったことから、全てのプラスミドが設計通りに構築できたことが確認された。
構築した8種のプラスミドについてシーケンス確認を行った。シーケンスの鋳型をPCRにより調製し、全長cry5Bの4種のプラスミドは、表2示したfrag1-Fとfrag1-Rまたは frag2-Fとfrag2-Rのプライマーを用いてそれぞれ5'側と3'側の断片を調製した。短縮型cry5Bの4種のプラスミドは、表2に示したfrag1-Fとfrag2-Rのプライマーを用いて断片の調製を行った。それらのPCR産物について表2に示したSEQ-P1~SEQ-P10の10個のプライマーを用いたシーケンス解析を行った。解析の結果、すべてのプラスミドについて、変異がなかったことから、全てのプラスミドが設計通りに構築できたことが確認された。
4.分泌シグナルを持つプラスミドによる枯草菌野生株でのCry5Bタンパク質、および、短縮型Cry5B(Cry5Bt)タンパク質の発現
構築した4種の分泌シグナルを持つプラスミド(図1-A、D)を、トリプトファン栄養要求性を解除した枯草菌168株(168T株)(特開2017-79640号公報)に形質転換し、得られた形質転換体をテトラサイクリン含有2xL/mal培地(バクトトリプトン2%(w/v)、酵母エキス1.0%(w/v)、塩化ナトリウム1.0%(w/v)、硫酸マンガン五水和物(和光純薬工業株式会社製)0.00075%(w/v)、マルトース一水和物(和光純薬工業株式会社製)7.5%(w/v)、テトラサイクリン塩酸塩0.0015%(w/v))で3日間、30℃、250rpm振盪培養した。培養液1mLを15000rpm、4℃で遠心分離し、培養上清と細胞に分けた。調製した培養上清のCry5Bの発現についてSDS-PAGEで確認を行った結果、Cry5Bのバンドが認められなかった(図2)。

構築した分泌シグナルを持たないプラスミド(図1-B、C)のうち、Cry5Bタンパク質を発現するための2種類のプラスミド<5>、<6>(図3)を、トリプトファン栄養要求性を解除した枯草菌168株(168T株)に形質転換し、得られた形質転換体を、2xL/mal培地で3日間、30℃、250rpm振盪培養した。培養液1mLを15000rpm、4℃で遠心分離し、培養上清と細胞に分けた。細胞ペレットは1xPBSを用いて洗浄した後、1mL 1xPBSに懸濁し、1mg/mLリゾチームを添加して、37℃で1hr保温した。続いて、BIORUPTOR(Cosmo Bio)を用いて、30秒x20回ソニケーションにより細胞破砕を行った後に、15000rpm、4℃で30分遠心分離し、上清を捨てて沈殿を1mL 1xPBSに懸濁し、細胞破砕液とした。調製した細胞破砕液のCry5Bの発現についてSDS-PAGEで確認を行った結果、pHY-Pscry5B(<5>)およびpHY-Pvcry5B(<6>)による高発現が確認された。pHY-Pscry5BとpHY-Pvcry5Bは全長型の140kDで、いずれも推定サイズと一致した(図3)。

6.枯草菌変異株でのCry5Bタンパク質の発現(1)
枯草菌野生株ゲノムの大領域が欠失した枯草菌変異株(MGB874株;特許4955358)、プロテアーゼの9重欠失株(Dpr9株;特許4485341)、sigF欠失株(ΔsigF株;特許4336082)、MGB874ΔsigF株、Dpr9ΔsigF株を宿主にそれぞれpHY-Pscry5Bの形質転換を行った。MGB874ΔsigF株とDpr9ΔsigF株、MGB874abrB*ΔkinA株(特開2017-79639号公報)は、ΔsigF株の構築と同様な方法で、MGB874とDpr9株をベースにそれぞれsigFを削除することにより作成した。上記4と同様に2xL/mal培地で培養し、調製した細胞破砕液のCry5B発現をSDS-PAGEで解析した。その結果、野生株の168T株に比べ、MGB874株、Dpr9株、sigF欠失株、MGB874ΔsigF株、Dpr9ΔsigF株で発現したCry5Bのバンドが明らかに太いことが分かった(図4-A)。

7.枯草菌変異株でのCry5Bタンパク質の発現(2)
sigE欠失株(ΔsigE株;特許4336082)を宿主にpHY-Pscry5Bの形質転換を行った。上記4と同様に、2xL/mal培地で培養し、調製した細胞破砕液のCry5B発現をSDS-PAGEで解析した。その結果、sigE欠失株においてもCry5Bのバンドが明らかに太いことが分かった(図4-B)。
sigE欠失株(ΔsigE株;特許4336082)を宿主にpHY-Pscry5Bの形質転換を行った。上記4と同様に、2xL/mal培地で培養し、調製した細胞破砕液のCry5B発現をSDS-PAGEで解析した。その結果、sigE欠失株においてもCry5Bのバンドが明らかに太いことが分かった(図4-B)。
8.バチルス・メガテリウム(Bacillus megaterium)でのCry5Bタンパク質の発現
Cry5Bタンパク質を発現するためのプラスミドpHY-Pscry5Bを、プロトプラスト法によってBacillus megaterium ATCC 14581株(以下14581株)に導入した。得られた組換え株を実施例1の5と同様に、2xL/mal培地で培養し、調製した細胞破砕液のCry5B発現をSDS-PAGEで解析した。その結果、pHY-Pscry5Bを導入した形質転換体において、Cry5Bの太いバンドを確認した。
Cry5Bタンパク質を発現するためのプラスミドpHY-Pscry5Bを、プロトプラスト法によってBacillus megaterium ATCC 14581株(以下14581株)に導入した。得られた組換え株を実施例1の5と同様に、2xL/mal培地で培養し、調製した細胞破砕液のCry5B発現をSDS-PAGEで解析した。その結果、pHY-Pscry5Bを導入した形質転換体において、Cry5Bの太いバンドを確認した。
9.発現したCry5Bタンパク質の定量
米国国立衛生研究所(National Institute of Health, NIH)が開発した画像ソフトImageJ(http://rsb.info.nih.gov/ij/download.html)を用いてSDS-PAGE上のバンドの定量を行った。標準タンパク質としてのウシ血清アルブミン(BSA)(和光純薬工業株式会社製)およびpHY-Pscry5B形質転換体で発現したCry5BのSDS-PAGEを行い、Bio-SafeTM Coomassie (BIO-RAD)で1時間振盪しながら染色した。イオン交換水にて脱色後、ゲルの撮影を行った。ImageJを用いてSDS-PAGEの画像に対して各バンドの輝度を解析し、BSAの検量線を作成した。この検量線からCry5Bタンパク質量を計算した(表3、表4、表5)。表3に示すように、枯草菌野生株でのCry5B生産性が1.1g/Lであり、組換え生産の文献値(75mg/L)より15倍高いことが分かった。MGB874ΔsigF株を用いることにより生産性が更に2倍以上向上した。

10.Cry5Bの活性評価
10.1 抱卵成虫の調製
大腸菌(E.coli OP50-1)溶液(1g湿重/6mL S培地)750μL,ストレプトマイシン(1mg/mL)溶液100μL、L1幼虫(Caenorhabditis elegans)1000頭を含んだ液をNGMプレートに滴下し、これを20℃のインキュベーター内で3~4日培養し、抱卵成虫を得た。
10.1 抱卵成虫の調製
大腸菌(E.coli OP50-1)溶液(1g湿重/6mL S培地)750μL,ストレプトマイシン(1mg/mL)溶液100μL、L1幼虫(Caenorhabditis elegans)1000頭を含んだ液をNGMプレートに滴下し、これを20℃のインキュベーター内で3~4日培養し、抱卵成虫を得た。
10.2 第一期(L1)幼虫の調製
線虫を培養しておいたNGMプレート(表6)をS basalで3回ほど洗い、全て15mL遠心管に移した。上澄みを捨て、沈殿している線虫を1.5mLエッペンチューブに移した。しばらく放置した後,1mLまで上澄みを除き,そこへ4M NaOH 250μL,次亜塩素酸ナトリウム溶液(Wako)250μLを加え,軽く撹拌した。3分静置した後、卓上ミニ遠心分離機で30秒程度スピンダウンを行った。沈殿物を残して上澄みを取り除き、1.5mLまでS basalを加え、軽く撹拌した後30秒程度スピンダウンを行った。この操作を3回繰り返した。これにより得られた卵の沈殿全量を35mm径シャーレに移し、20℃で約24時間培養し、ふ化した第一期幼虫(L1幼虫)を得た。

10.3 L1 growth アッセイ
Bischofらの方法(Methods in Molecular Biology, vol. 331, pp139-154, C. elegans: Methods and applications, Edited by: K. Strange, Humana Press)に従い、L1 growthアッセイを行った。48穴平底プレートを用い、各ウェルに1g/35mLの大腸菌溶液を20μL、1mg/mLストレプトマイシンを20μL、L1幼虫10匹、枯草菌で発現したCry5Bタンパク質を適量、S培地(表7)で総量を200μLに調整した。Cry5Bのタンパク質量は1.25、2.5、5μg/mLを使用した。コントロールはベクター(pHY300PLK)を導入した枯草菌を使用した。20℃で3日間培養した後、各ウェルの線虫の様子をカメラで撮影した(図5)。撮影した画像をImage Jを用いて、線虫の輪郭を書き、面積を計算した。その結果を図6に示した。Cry5Bを使用することにより、線虫の生育が顕著に抑制され、コントロールに比べ1.25μg/mLの濃度でも生育が約80%抑制された。この結果から、枯草菌で発現したCry5Bが線虫に対する毒性を有することを証明した。

実施例2 殺蚊タンパク質(Cry4Aa、Cry4Ba、Cry11Aa)の製造
1.人工遺伝子の合成
Bacillus thuringiensis serovar israelensis由来の殺虫タンパク質遺伝子cry4Aa(GenBank: YP_001573833、配列番号3)、cry4Ba(GenBank: NC_010076、配列番号5)およびcry11Aa(GenBank: NC_010076、配列番号7)をGenScript社(アメリカ)により人工合成した。合成した遺伝子はpUC57のKpnIとHindIIIサイトにクローニングし、それぞれpUC57-cry4Aa、pUC57-cry4BaとpUC57-cry11Aaのプラスミドを得た。
1.人工遺伝子の合成

2.発現プラスミドの構築
全ての発現プラスミドの構築において、ベクターとしてpHY300PLK、およびターミネターとしてS237セルラーゼ遺伝子由来配列を用いた。プロモーターは当研究室で実績のあるS237セルラーゼ遺伝子[Hakamada et al, Biosci. Biotechnol. Biochem., 64 (2000), 2281-2289]あるいは枯草菌168株のspoVG遺伝子由来、cry4Aa、cry4Ba、cry11Aaの3つの遺伝子との組み合わせで、図8に示した6種のプラスミドの作製を行った。pHY-Pscry4Aa、pHY-Pscry4Ba、pHY-Pscry4AaはS237セルラーゼ遺伝子のプロモーター、pHY-Pvcry4Aa、pHY-Pvcry4Ba、pHY-Pvcry4Aaは枯草菌168株のspoVG遺伝子のプロモーターを使用した。構築方法はIn-Fusion(R) HD EcoDryTM Cloning Kitのプロトコールに従った。
全ての発現プラスミドの構築において、ベクターとしてpHY300PLK、およびターミネターとしてS237セルラーゼ遺伝子由来配列を用いた。プロモーターは当研究室で実績のあるS237セルラーゼ遺伝子[Hakamada et al, Biosci. Biotechnol. Biochem., 64 (2000), 2281-2289]あるいは枯草菌168株のspoVG遺伝子由来、cry4Aa、cry4Ba、cry11Aaの3つの遺伝子との組み合わせで、図8に示した6種のプラスミドの作製を行った。pHY-Pscry4Aa、pHY-Pscry4Ba、pHY-Pscry4AaはS237セルラーゼ遺伝子のプロモーター、pHY-Pvcry4Aa、pHY-Pvcry4Ba、pHY-Pvcry4Aaは枯草菌168株のspoVG遺伝子のプロモーターを使用した。構築方法はIn-Fusion(R) HD EcoDryTM Cloning Kitのプロトコールに従った。
2.1 pHY-Pscry4Aa、pHY-Pscry4BaとpHY-Pscry11Aaの構築
表8に示すvect+pFとvect+tRのプライマーセットを用いて、pHYS237 DNAを鋳型にS237のプロモーター、及びターミネター領域を含むベクターを増幅した。次に、pUC57-cry4Aa DNAを鋳型にcry4AFPSとcry4ARTのプライマーセットを用いてcry4Aa遺伝子のインサートをPCRで増幅した。続いて、In-Fusion(登録商標) HD EcoDryTM Cloning Kit(Clontech社)を用いてベクターとインサートを連結した後、大腸菌HB101コンピテントセル(タカラバイオ)に形質転換した。テトラサイクリン耐性により選抜した形質転換体をコロニーPCRにより確認し、pHY-Pscry4Aaと命名した。抽出したプラスミドについて、さらにPCR確認、また、制限酵素を用いてプラスミドの消化パターンの確認を行った。
上記と同様に、pUC57-cry4Ba DNAを鋳型にcry4BFPSとcry4BRTのプライマーセットを用いてcry4Ba遺伝子、pUC57-cry11Aa DNAを鋳型にcry11AFPSとcry11ARTのプライマーセットを用いてcry11Aa遺伝子のインサートをそれぞれPCRで増幅し、pHY-Pscry4BaとpHY-Pscry11Aaの構築を行った。

2.2 pHY-Pvcry4Aa、pHY-Pvcry4Ba、pHY-Pvcry11Aaの構築
表8に示すvect+pvgFとvect+tRのプライマーセットを用いて、pHY-Pscry5B(Cry5Bの実施例参考)を鋳型にspoVG遺伝子のプロモーター、及びS237セルラーゼ遺伝子のターミネター領域を含むベクターを増幅した。次に、pUC57-cry4Aa DNAを鋳型にcry4AFPVとcry4ARTのプライマーセットを用いてcry4Aa遺伝子のインサートをPCRで増幅した。続いて、In-Fusion(登録商標) HD EcoDryTM Cloning Kit(Clontech社)を用いてベクターとインサートを連結した後、大腸菌HB101コンピテントセル(タカラバイオ)に形質転換した。テトラサイクリン耐性により選抜した形質転換体をコロニーPCRにより確認し、pHY-Pvcry4Aaと命名した。抽出したプラスミドについて、さらにPCR確認、また、制限酵素を用いてプラスミドの消化パターンの確認を行った。

3.枯草菌での殺蚊タンパク質の発現
構築した6種のプラスミド(図8)をトリプトファン栄養要求性を解除した枯草菌168株(168T株)に形質転換し、得られた形質転換体を2xL/mal培地で3日間、30℃、250rpm振盪培養した。培養液1mLを15000rpm、4℃で遠心分離し、培養上清と細胞に分けた。細胞ペレットは1xPBSを用いて洗浄した後、1mL 1xPBSに懸濁し、1mg/mLリゾチームを添加して、37℃で1hr保温した。続いて、Biorupter(Cosmo Bio)を用いて、30秒x20回ソニケーションにより細胞破砕を行った後に、15000rpm、4℃で30分遠心分離し、上清を捨てて沈殿を1mL 1xPBSに懸濁し、細胞破砕液とした。調製した細胞破砕液中のタンパク質の発現についてSDS-PAGEで確認を行った。結果は図9に示したように、すべてのプラスミドにおいて、高発現が認められた。

4.発現した殺蚊タンパク質の定量
米国国立衛生研究所(National Institute of Health, NIH)が開発した画像ソフトImageJ(http://rsb.info.nih.gov/ij/download.html)を用いてSDS-PAGE上のバンドの定量を行った。標準タンパク質としてのウシ血清アルブミン(BSA)を使用した。ImageJを用いてSDS-PAGEの画像に対して各バンドの輝度を解析し、BSAの検量線を作成した。この検量線から各殺蚊タンパク質の量を計算した。表9に示すように、0.6-0.7g/Lの生産性が確認された。

5.殺蚊タンパク質の活性評価
5.1 ボウフラの調製
ヒトスジシマカ(Aedes albopictus)は住化テクノサービス株式会社より購入した卵を孵化させたものを使用した。プラスチックパンに水を1cm程度張り、卵が産みつけられているろ紙を入れた。毎日孵化後のボウフラ用餌として熱帯魚用のエサ(テトラミンベビー)を与えた。孵化後5日目のボウフラを活性評価に用いた。
5.1 ボウフラの調製

Leetachewaらの方法(BMB reports, 47 (2014), 546-551)に従い、枯草菌で発現したCry4Aa、Cry4BaとCry11Aaの殺蚊活性評価を行った。24穴平底プレートを用い、各ウェルに5日齢ボウフラ10匹、殺蚊タンパク質濃度は50μg/mL、水で総量を1mLに調整した。25℃で24h放置した後、死滅したボウフラの数を数え、死亡率を算出した。その結果は表10に示した。対照の死亡率が0%に対して、Cry4Aa、Cry4Ba、Cry11Aaはそれぞれ93.3%、93.3%と76.7%を示した。この結果から、枯草菌で発現した殺蚊タンパク質は高い殺蚊効果が認められた。

Claims (33)
- Cryタンパク質をコードする遺伝子とσA依存性プロモーター又はσH依存性プロモーターを含む制御領域とが作動可能に連結して組み込まれてなる発現プラスミドでバチルス属細菌を形質転換し、当該形質転換細胞を培養することを含むCryタンパク質又はそれを含有する培養産物の製造方法であって、前記発現プラスミドが、配列番号9で示されるアミノ酸配列からなる複製タンパク質又はこれとアミノ酸配列において80%以上の同一性を有し、且つ複製開始に関与するタンパク質をコードするポリヌクレオチドを含むプラスミドである、方法。
- σA依存性プロモーター又はσH依存性プロモーターを含む制御領域が、Cryタンパク質をコードする遺伝子の起源微生物における当該遺伝子の制御領域とは異なる請求項1記載の方法。
- σA依存性プロモーターを含む制御領域が、Bacillus sp.KSM-S237株のセルラーゼ遺伝子の制御領域である請求項1又は2記載の方法。
- σH依存性プロモーターを含む制御領域が、spoVG遺伝子の制御領域である請求項1又は2記載の方法。
- Bacillus sp.KSM-S237株のセルラーゼ遺伝子の制御領域が、当該セルラーゼ遺伝子の転写開始制御領域及び翻訳開始領域である請求項3記載の方法。
- spoVG遺伝子の制御領域が、当該spoVG遺伝子遺伝子の転写開始制御領域及び翻訳開始領域である請求項4記載の方法。
- バチルス属細菌が、枯草菌、バチルス・メガテリウム(Bacillus megaterium)又はバチルス・ブレビス(Bacillus brevis)である請求項1~6のいずれか1項記載の方法。

- 枯草菌が、aprX、aprE、nprB、nprE、bpr、vpr、mpr、epr及びwprAから選択される遺伝子を欠失又は不活性化させた枯草菌株である請求項7~12のいずれか1項記載の方法。

- σA依存性プロモーター又はσH依存性プロモーターを含む制御領域が、Cryタンパク質をコードする遺伝子の起源微生物における当該遺伝子の制御領域とは異なる請求項22記載の発現プラスミド。
- σA依存性プロモーターを含む制御領域が、Bacillus sp.KSM-S237株のセルラーゼ遺伝子の制御領域を含む請求項22又は23記載の発現プラスミド。
- σH依存性プロモーターを含む制御領域が、spoVG遺伝子の制御領域を含む請求項求項22又は23記載の発現プラスミド。
- Bacillus sp.KSM-S237株のセルラーゼ遺伝子の制御領域が、当該遺伝子の転写開始制御領域及び翻訳開始領域である請求項24記載の発現プラスミド。
