Abstract
これまでに報告されていない新規の免疫チェックポイント抑制剤を提供する。 次の(a)ないし(g) (a)大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワおよびキビよりなる群から選ばれる1種又は2種以上の豆・穀物類の麹菌発酵物 (b)ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビおよびスモモよりなる群から選ばれる1種又は2種以上の果実類の酵母および/又は乳酸菌発酵物 (c)紫イモ、菊芋、ニンジン、タマネギ、サツマイモ、里芋、自然薯、大根、赤カブ、ゴボウ、レンコン、ヤーコン、ユリ根、クワイ、しょうが、にんにくおよびウコンよりなる群から選ばれる1種又は2種以上の根菜類・イモ類の酵母および/又は乳酸菌発酵物 (d)キャベツ、紫蘇、桑葉、どくだみ、ヨモギ、クマザサおよびタンポポよりなる群から選ばれる1種又は2種以上の花・葉菜類の酵母および/又は乳酸菌発酵物 (e)コンブ、ワカメおよびモズクよりなる群から選ばれる1種又は2種以上の海藻類の酵母および/又は乳酸菌発酵物 (f)黒ゴマ、クルミおよびギンナンよりなる群から選ばれる1種又は2種以上の種子類の酵母および/又は乳酸菌発酵物 (g)マイタケおよびシイタケよりなる群から選ばれる1種又は2種のキノコ類の酵母および/又は乳酸菌発酵物 の混合物である植物発酵物を有効成分として含有することを特徴とする免疫チェックポイント抑制剤。
Description
本発明は、植物発酵物を利用した免疫チェックポイント抑制剤に関する。
がんの治療法については、外科切除をはじめ各種細胞毒を中心とした抗がん剤、ホルモン剤、放射線療法に加え、免疫療法が知られている。
これらのがんの治療法の中でも、従来、免疫療法は補助的手段であった。免疫療法に関するメカニズムとしてはマクロファージや樹状細胞等による自然免疫に属するものと各種Tリンパ球が関与するキラーTリンパ球が殺がん細胞効果を発揮する獲得免疫が強力である。しかしながら、キラーT細胞が効果を十分発揮しえないメカニズムが分かってきた。
実際、抑制性Tリンパ球の表面にあるProgramed Cell Death-1受容体(PD-1)にがん細胞などが生産するリガンド(PD-L1)が結合すると、抑制性T細胞が増殖してキラーT細胞のがん細胞障害効果を抑制することが報告されている(非特許文献1)。このメカニズムを免疫チェックポイントと言う。
この免疫チェックポイントを阻害する免疫チェックポイント阻害剤は、従来治療が難しかった各種のがんに対して有効性を発揮する新たな抗がん剤として注目されている。医学系専門雑誌でも免疫チェックポイント阻害剤に関する特集が出版されている(非特許文献2~3)。
これまでに多くの免疫チェックポイントが発見され、これらに対する特異抗体が医薬品として次々に承認され、がん患者の広く投与されるようになったが、医薬品として開発された免疫チェックポイント阻害剤および開発中の物質も全て特異抗体のみである。その他の物質については報告されていない。

従って、本発明の課題は、これまでに報告されていない新規の免疫チェックポイント抑制剤を提供することである。
本発明者らは、上記課題を解決するために鋭意研究した結果、本発明者らがすでに有意に寿命延長と老化の抑制をすることを証明し、老化抑制剤として特許を取得している植物発酵物(特許第6013670号公報)が、従来の免疫チェックポイント阻害剤とは異なる仕組みの免疫チェックポイント抑制剤となることを見出し、本発明を完成させた。
すなわち、本発明は、次の(a)ないし(g)
(a)大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワおよびキビよりなる群から選ばれる1種又は2種以上の豆・穀物類の麹菌発酵物
(b)ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビおよびスモモよりなる群から選ばれる1種又は2種以上の果実類の酵母および/又は乳酸菌発酵物

の混合物である植物発酵物を有効成分として含有することを特徴とする免疫チェックポイント抑制剤である。
また、本発明は、更に、がんの治療薬および/またはがんの代替治療薬を含有する上記免疫チェックポイント抑制剤である。
更に、本発明は、上記免疫チェックポイント抑制剤を含有する飲食品である。
また更に、本発明は、上記免疫チェックポイント抑制剤を含有する放射線療法の副作用低減剤である。
本発明により、特定の植物発酵物に免疫チェックポイントの抑制効果があることが明瞭に示された。植物発酵物では初めての証明である。
本発明の免疫チェックポイント抑制剤は免疫チェックポイントに関連する疾患の治療、予防、改善等をすることができる。
また、本発明の免疫チェックポイント抑制剤は、これまでも食経験の豊かな特定の植物発酵物を有効成分としているため飲食品、医薬品等としても好適である。
本発明の免疫チェックポイント抑制剤は、下記の植物発酵物を有効成分として含有するものである。この植物発酵物は、以下の(a)~(g)の植物発酵物の混合物である。
(a)大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワおよびキビよりなる群から選ばれる1種又は2種以上の豆・穀類に麹菌を作用させることにより得られる発酵物。
(b)ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビおよびスモモよりなる群から選ばれる1種又は2種以上の果実類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(c)紫イモ、菊芋、ニンジン、タマネギ、サツマイモ、里芋、自然薯、大根、赤カブ、ゴボウ、レンコン、ヤーコン、ユリ根、クワイ、しょうが、にんにくおよびウコンよりなる群から選ばれる1種又は2種以上の根菜類・イモ類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(d)キャベツ、紫蘇、桑葉、どくだみ、ヨモギ、クマザサおよびタンポポよりなる群から選ばれる1種又は2種以上の花・葉菜類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(e)コンブ、ワカメおよびモズクよりなる群から選ばれる1種又は2種以上の海藻類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
(f)黒ゴマ、クルミおよびギンナンよりなる群から選ばれる1種又は2種以上の種子類に酵母および/又は乳酸菌を発酵させることにより得られる発酵物。
(g)マイタケおよびシイタケよりなる群から選ばれる1種又は2種のキノコ類に酵母および/又は乳酸菌を作用させることにより得られる発酵物。
本発明の免疫チェックポイント抑制剤は、上記(a)~(g)の発酵物を混合した植物発酵物をそのまま有効成分としてもよいが、さらに多段階発酵させることにより、呈味性や製剤性を向上させることができる。
酵母としては、サッカロマイセス属、ジゴサッカロマイセス属等に属する酵母が挙げられ、中でもサッカロマイセス・セレビシエ(S. cervisiae)、ジゴサッカロマイセス・ロキシー(Z. rouxii)、サッカロマイセス・エキシグス(S.exiguus)等が好ましく用いられる。乳酸菌としては、例えば、ぺディオコッカス(Pediococcus)属、ロイコノストック(Leuconostoc)属、ラクトバチルス(Lactobacillus)属、ラクトコッカス(Lactococcus)属等に属する乳酸菌が挙げられ、中でも、ぺディオコッカス・アシディラクティ(P.acidilacti)、ラクトバチルス・ブレビス(L. brevis)、ロイコノストック・メセンテロイデス(L. mesenteroides)、ラクトバチルス・プランタラム(L. plantarum)、ラクトコッカス・ラクチス(L. lactis)、ラクトバチルス・サケイ(L. sakei)、ペディオコッカス・ペントサセウス(P. pentosaceus)、ラクトバチルス・カゼイ(L. casei)、ラクトバチルス・ロイテリ(L.reuteri)およびラクトバチルス・クルバタス(L. curvatus)等が好ましく用いられ、これらの1種または2種以上を使用することができる。麹菌としては、アスペルギルス・オリゼー(Aspergillus oryzae)、アスペルギルス・ニガー(Aspergillus niger)、アスペルギルス・カワチ(Aspergillus kawachii)等が例示でき、これらの1種又は2種以上を使用できる。これらは市販されているものも使用することができる。
発酵に供する豆・穀類、果実等の植物は、そのまま使用してもよく、あるいは必要に応じて粉砕、乾燥等の前処理を行ってもよい。また必要に応じて水を添加して希釈してもよい。
上記(a)~(g)の発酵物は、原料となる植物に、乳酸菌、酵母または麹菌を接種し、培養させることによって得られる。培養は、常法によって行えばよく、例えば、1種または2種以上の植物の混合物に対し、乳酸菌、酵母または麹菌を0.001~1質量%程度添加し、20~50℃で70~140時間程度発酵処理させればよい。
このようにして得られた(a)~(g)の発酵物を混合してそのまま有効成分とすることもできるが、必要に応じさらに20~40℃で200~300時間程度培養することが好ましい。さらにこの混合物を後発酵(熟成)させることが好ましい。後発酵は、必要に応じて、酢酸菌を作用させてもよく、例えば、上記(a)~(g)に含まれる植物のうち1種または2種以上に上記酵母を作用させたものに、アセトバクター・アセチ(A.aceti)等の酢酸菌を作用させた酢酸発酵物を添加すればよい。後発酵は、25~35℃で70時間~約1年程度行えばよい。後発酵による熟成過程を経ることより、呈味性や製剤性を改善することができ、また抗酸化活性の増大を図ることもできる。
上記植物発酵物を製造するための好適な方法の例として、多段階複合発酵方式が挙げられる。この方法は、果実類、野菜類、海藻類を主原料とする複合乳酸菌発酵物と、野菜類、根菜類、種子類、キノコ類、果実類を主要原料とする酵母発酵物を混合し、これに穀類、豆類を主原料とする麹菌発酵物を加え、さらに酢酸発酵物を加え混合した後、ろ過濃縮し、さらに約1年程度の後発酵を行うものである。このような多段階複合発酵によって、それぞれの発酵生産物がさらに別種の菌類によって資化・変換されることにより、風味が向上するとともに、抗酸化活性などの効果が高められると考えられる。
本発明に用いる有効成分の植物発酵物は、下記(1)から(4)の性質を有する。
(1)呈味性
果物、野菜等原料由来の甘味や有機酸に加え麹由来の甘味を有する。原料由来のポリフェノール類を含むものの苦味は少ない。
(2)水に対する溶解性
水に対して容易に溶解する。
(3)安定性
熱、酸に対して安定であり、ペーストは1年間室温保存しても腐敗や呈味性に変化はない。
(4)安全性
原料に用いた野菜、果物、ハーブ等は日常食しているものであり、また発酵に用いた酵母、乳酸菌、麹菌はいずれも食品の醸造や漬物等に由来する菌種を用いているため食経験が豊富である。
(1)呈味性

この植物発酵物を有効成分とする免疫チェックポイント抑制剤は、免疫チェックポイント、例えば、PD-1、PD-L1およびCTLA4から選ばれる1種以上、好ましくはこれらの全て、より好ましくはPD-1、PD-L1の発現を抑制する。なお、本発明の免疫チェックポイント抑制剤は、既存の免疫チェックポイント阻害剤とは異なり、レセプターとリガンドの結合を阻害するものではなく、上記免疫チェックポイントの発現を抑制するものであり、更には、経口で効果が得られる新たなタイプのものである。
本発明の免疫チェックポイント抑制剤は、上記の通り、既存の免疫チェックポイント阻害剤とは異なり、レセプターとリガンドの結合を阻害するものではなく、上記免疫チェックポイントの発現を抑制するという異なった仕組みを利用することにより、免疫チェックポイントに関連する疾患の治療、予防、改善等をすることができる。具体的には、免疫チェックポイントのうち、PD-1の発現を抑制することにより、例えば、悪性黒色腫、非小細胞肺がん、腎細胞がん、ホジキンリンパ腫、頭頸部がん、胃がん等の治療、予防、改善、悪性胸膜中皮腫と悪性黒色腫の術後補助療法等に用いることができる。CTLA4の発現を抑制することにより、例えば、未治療の進行期メラノーマ、切除不能や転移性のメラノーマ等のメラノーマ、非小細胞肺がん、小細胞肺がん等の治療、予防、改善に用いることができる。PD-L1の発現を抑制することにより、例えば、切除不能な進行・再発の非小細胞肺癌等の治療、予防、改善に用いることができる。なお、本発明の免疫チェックポイント抑制剤には、同じ植物発酵物を有効成分とする自然発がんの発生を抑制する自然発がん予防剤は含まない(自然発がん予防剤は国際特許出願済(PCT/JP2018/ 34617))。
本発明の免疫チェックポイント抑制剤は、かくして得られた植物発酵物に、公知の製剤学的製法に準じて、薬理学的に許容され得る担体、賦形剤、水分活性調整剤などを加え、製剤化することにより得られる。植物発酵物は、必要に応じ濃縮して濃度を調整したり、噴霧または凍結乾燥等により粉末化してもよい。製剤化において用いられる担体や賦形剤としては、たとえば乳糖、ブドウ糖、蔗糖、デンプン、糖類混合物などが用いられる。最終的形態としては、溶液、ペースト、ソフトカプセル、チュアブル、カプセルが用いられる。用量・用法としては、例えば、有効成分の植物発酵物として大人1日当たり、0.1g以上、好ましくは0.1~12g、より好ましくは0.6~10g(固形分換算)程度経口摂取すればよい。
また、本発明の免疫チェックポイント抑制剤は医薬品、医薬部外品等の他、公知の食品素材とともに配合することにより飲食品の形態とすることもできる。上記植物発酵物はそのまま摂取することも可能であるが、日持ち性向上のために殺菌後にろ過、濃縮してもよく、あるいは必要に応じて賦形剤を添加して噴霧または凍結乾燥した粉末状としてもよい。さらに、流通過程におけるシェルフライフ向上のためには、濃縮して水分活性を低下させたものが望ましい。かかる飲食品の形態としては、ペースト、ソフトカプセル、錠剤、ドリンク剤等が例示できる。大人1日当たり植物発酵物として0.1g以上、好ましくは0.1~12g、より好ましくは0.6~10g(固形分換算)程度経口摂取することにより、優れた免疫チェックポイント抑制効果等を得ることができる。このような上記植物発酵物を製剤化、食品の形態とした市販品として、天生酵素金印、天生酵素カプセル(日本自然発酵社製)等が挙げられる。
また、本発明の免疫チェックポイント抑制剤を含有する飲食品は、上記のものの他に、この有効成分である植物発酵物を、調味料等の一種として用いて、みそ等の調味料、食パン等のパン類、せんべい、クッキー、チョコレート、飴、饅頭、ケーキ等の菓子類、ヨーグルト、チーズ等乳製品、たくわん等の漬物類、ラーメン、そば、うどん等の麺類、コーンポタージュ、わかめスープ等のスープ類、清涼飲料、健康飲料、炭酸飲料、果汁飲料等の飲食品に含有させたものであってもよい。
更に、本発明の免疫チェックポイント抑制剤には、更にがんの治療薬および/またはがんの代替治療薬を含有させて、効果を増強させてもよい。がんの治療薬としては、特に限定されないが、例えば、代謝拮抗剤、アルキル化剤、抗がん性抗生物質、微小管阻害薬等の細胞障害性抗がん剤、分子標的治療薬、白金製剤、トポイソメラーゼ阻害剤、免疫チェックポイント特異的阻害剤等が挙げられる。がんの代替治療薬としては、特に限定されないが、例えば、十全大補湯、大柴胡湯、補中益気湯等の漢方製剤、フコイダン、メシマコブ、冬虫夏草、田七人参、鹿角霊芝、百花蛇舌草、発酵アガリクス等が挙げられる。特に、本発明の免疫チェックポイント抑制剤は、既存の免疫チェックポイント阻害剤とは異なる仕組みで免疫チェックポイントの発現を抑制するため、自己免疫増強の副作用が強いがんの治療薬、例えば、CTLA4の特異的阻害剤であるイピリムマブ等との併用が好ましい。また、本発明の免疫チェックポイント抑制剤は、その有効成分である植物発酵物が胸腺萎縮抑制作用もあるため、免疫毒性が強いがんの治療薬、例えば、アルキル化剤等との併用や、更には免疫毒性が強い放射線療法との併用も好ましい。こうすることにより、前記がんの治療薬および/またはがんの代替治療薬や、放射線療法の副作用を低減できる。これらがんの治療薬および/またはがんの代替治療薬を本発明の免疫チェックポイント抑制剤に含有させる場合、含有量は目的とする効果にあわせて適宜設定すればよい。
以下、本発明を実施例を挙げて詳細に説明するが、本発明はこれら実施例に何ら限定されるものではない。
製 造 例 1
植物発酵物の製造:
以下の植物を原料として使用した。
(a)豆・穀類(大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワ、キビ)
(b)果実類(ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビ、スモモ)

上記(c)、(d)および(g)の原料(730g)に対し、乳酸菌類(P. acidilacti, L. brevis, L. mesenteroides, L. plantarum, L. lactis, L. sakei, L. casei)が約1.0×105cfu/gの菌濃度となるように調製した培養液を0.2質量%接種し、30℃で50時間培養を行った。また上記(b)、(e)および(f)の原料(900kg)に対し、酵母類(S. cervisiae 5種、Z. rouxii 2種)が約1.0×105cfu/gの菌濃度となるように調製した培養液を0.4質量%接種し30℃で50時間培養を行った。(a)豆類および穀類系原料(1000kg)には、麹菌類(黄麹菌、黒麹菌、白麹菌)を0.1質量%接種して35℃で70時間培養を行った。次いでそれぞれの培養物を混合後、30℃で200時間培養を行った。発酵を終了したモロミについて固液分離操作を行い、得られた濾液を濃縮してペースト状にしたものを容器に分注し、更に1年間後発酵(熟成)させ植物発酵物を得た。
製造例1で得られた植物発酵物は以下の性質を有するものであった。

胸腺の萎縮抑制試験:
(1)マウス
SAM-P1(♀)マウスは各群6匹で8週齢のものを三協ラボサービス株式会社から購入して使用した。飼育は、SPF仕様のマウス飼育施設(23±2℃、湿度50±10%、20:00~8:00の照明の明暗サイクル)を用い、マウスは各サンプル1群6匹を用い日本クレア製のポリカーボネートケージを用いSPF条件下で単独飼育した。餌は三協ラボサービスから購入した500N(γ線滅菌済)を自由摂取させた。床敷きは日本クレア製の高温殺菌済みを用いた。ケージ交換は週1回行った。なお、飼育中、マウスの摂餌量が減ることはなかった。
胸腺の萎縮抑制試験:
(1)マウス

(3)解剖

老齢マウスの胸腺のみが加齢により萎縮を示し、若齢マウスの胸腺重量の約1/2となった。植物発酵物の0.2%群は0%群とほぼ同程度の胸腺重量であったが、2%群の結果を考慮すると濃度依存的に胸腺萎縮を抑制したが、10週齢の胸腺重量には達しなかった。このことは植物発酵物が免疫器官の高齢化による萎縮を抑制できることを示した。

飼育2週目(10週齢)の若齢マウスを、3種混合麻酔薬(メデトミジン(0.3mg/0.3ml/Kg)、ミダゾラム(4mg/0.8ml/Kg)、プトルファノール(5mg/1ml/Kg):最終調整液量0.05ml/10g)を腹腔投与後、開腹し、下行動脈から全血、胸腺、脾臓を採取した。全血はそのまま冷蔵し、胸腺および脾臓はGibco社製の保存培地(AIM-V培地:血清フリー)に浸漬し、アイスパックとともに測定機関(株式会社鎌倉テクノサイエンス 生物試験事業部)に輸送した。
<1回目>
飼育2週目(10週齢)の若齢マウスを、3種混合麻酔薬(メデトミジン(0.3mg/0.3ml/Kg)、ミダゾラム(4mg/0.8ml/Kg)、プトルファノール(5mg/1ml/Kg):最終調整液量0.05ml/10g)を腹腔投与後、開腹し、下行動脈から全血、胸腺、脾臓を採取した。全血はそのまま冷蔵し、胸腺および脾臓はGibco社製の保存培地(AIM-V培地:血清フリー)に浸漬し、アイスパックとともに測定機関(株式会社鎌倉テクノサイエンス 生物試験事業部)に輸送した。
免疫関連細胞を広く網羅した抗体およびPD-1抗体について行った。フローサイトメーターはBD FACSCanto IIを用いた。胸腺と脾臓はRPMI1640を加え、gentleMACS C tube 中でgentleMACS Dissociator で細胞を分散させた。細胞を回収後WBC数を計測後、HBSSを用いて約1x107 cells/mlに調整した。FACS Tubeに全血または細胞浮遊液50μlを分注した。抗体混合液を加え、室温暗所で30分インキュベートした。FACS装置に掛け、CD45/SSC プロットにて展開後、CD45陽性細胞にゲートを掛けた。続いて、FSC/SSCプロットにてWBCを展開し、Lymphocyteゲートに掛けた。取り込み細胞はLymphocytes を10,000個として全てのサンプルを測定した。解析はBD FACS Diva ソフトウエア Ver. 6.1.3.使用し解析結果を印刷し、これを生データとした。全血、胸腺および脾臓中の以下の細胞の割合について、群ごとにまとめた。免疫臓器リンパ球のフローサイトメトリーによるPD-1+T細胞比率(%)を表1に示した。

PD-1+T細胞比率は、水投与群(サンプル0%)内では全血、胸腺、脾臓の全ての臓器で担癌体の方が高かった。2%投与(サンプル2%)では、担癌体でもPD-1+T細胞比率は、1例を除いては低値を示した。この1例は大腿部の骨肉腫であった。
<2回目>
PD-1および周辺のTregに注目した抗体(CD45, CD3e, CD4, CD8a, CD25、FoxP3およびCD279(PD-1))について行った。これらの細胞群を植物発酵物の濃度0%、0.4%、2.0%に分類し、更に担癌体と非担癌体に仕分けて解析した。担癌体におけるフローサイトメトリーによるPD-1+T細胞比率を表2に示した。

若齢(10週齢)マウスのPD-1発現率は全血、胸腺、脾臓とも低値(0.7~1.1)であった。71週齢では、全血、胸腺、脾臓ともPD-1発現率は高値を示した。脾臓におけるPD-1率は全血や胸腺の約1/2であった。胸腺では、サンプルの全ての濃度群において、担癌体の方が高値を示し、サンプルは濃度依存的にPD-1発現率が低かった。全血も類似の傾向をしめしたが、脾臓では胸腺とは異なる傾向を示した。高齢化により免疫チェックポイントの発現は増加し、担癌体ではさらに高値を示すが、植物発酵物摂取したSAMP-1マウスでは発現は減少した。
試 験 例 3
DNAマイクロアレイ(免疫チェックポイントの遺伝子発現の解析):
植物発酵物0%(イオン交換水)あるいは2.0%を飲水として与えたSAMP-1(♂)マウス各群6匹について10週齢および56週齢まで長期飼育したマウスをソムノペンチル麻酔後、開腹し胸腺および腎臓を採取し、氷冷したRNA Later溶液に浸漬して、測定機関(フィルジェン株式会社)に冷蔵輸送した。RNA抽出からDNAマイクロアレイでの測定も測定機関によって行われた。
DNAマイクロアレイ(免疫チェックポイントの遺伝子発現の解析):
植物発酵物0%(イオン交換水)あるいは2.0%を飲水として与えたSAMP-1(♂)マウス各群6匹について10週齢および56週齢まで長期飼育したマウスをソムノペンチル麻酔後、開腹し胸腺および腎臓を採取し、氷冷したRNA Later溶液に浸漬して、測定機関(フィルジェン株式会社)に冷蔵輸送した。RNA抽出からDNAマイクロアレイでの測定も測定機関によって行われた。
上記機関から入手した分析データから、先ず、寿命延長に関係すると言われているサーチュインの7分子について確認した。10週齢、56週齢および胸腺・腎植物発酵物の0%および2%の全ての組合せについてサーチュインの遺伝子発現の増減は認められず、サーチュインは寿命延長作用には関係が無いと判断した。
また、遺伝子発現の増減の大きさ、頻度から見て免疫グロブリンが分子の様々な部分で動きがあったが、抗体の特性までは検出不能であったが、免疫が関係していることは明らかであった。またsmall Nuclear RNAの動きも目立ったものの、引用文献が全くなく、核内での信号伝達への関係が寿命延長現象に関係すると予測されたが、詳細は解析し得なかった。この結果から、植物発酵物には、サーチュイン等の従来から知られている遺伝子の動きはないことが分かった。
試 験 例 4
DNAマイクロアレイによる免疫チェックポイントの解析:
フローサイトメトリーの結果から免疫チェックポイントであるPD-1の関与が浮かび上がってきた。そこで、DNAマイクロアレイのプローブの中から免疫チェックポイントに関係するものを抽出する事を試みた。その結果、3種類が含まれていた。即ち、Ctla4(CTLA-4に相当)、Pdcd1(PD-L1に相当)およびCd274 (PD-1に相当)がデータベースに含まれていた。試験例3で得たデータの中から異なった視点からの再解析を行った。そこで胸腺並びに腎臓における10週齢vs56週齢、植物発酵物の濃度0%vs2.0%についての上記3遺伝子の遺伝子発現を比較した。胸腺のみ増減が見られ、腎臓は全く変化が無かった。胸腺の結果を表3に示した。

フローサイトメトリーの結果から免疫チェックポイントであるPD-1の関与が浮かび上がってきた。そこで、DNAマイクロアレイのプローブの中から免疫チェックポイントに関係するものを抽出する事を試みた。その結果、3種類が含まれていた。即ち、Ctla4(CTLA-4に相当)、Pdcd1(PD-L1に相当)およびCd274 (PD-1に相当)がデータベースに含まれていた。試験例3で得たデータの中から異なった視点からの再解析を行った。そこで胸腺並びに腎臓における10週齢vs56週齢、植物発酵物の濃度0%vs2.0%についての上記3遺伝子の遺伝子発現を比較した。胸腺のみ増減が見られ、腎臓は全く変化が無かった。胸腺の結果を表3に示した。
受容体であるPD-1と同時にそのリガンドであるPD-L1が類似の挙動を示したのは興味深く、両者の特異抗体はいずれも免疫チェックポイント阻害剤として医薬品として承認されている。若齢マウス(10週齢)に比べ、高週齢化マウスでは3種類すべての遺伝子発現がLog1.3倍程度増加し、その増加を2%群では0%群に比べて減少(log0.9程度)に抑えられていることが特にPD-1とPD-L1では明瞭であった。Ctla4も同様の傾向は示したものの程度はPD-1系に比べて大きくなかった。腎臓では上記3遺伝子については増減は観察されず、免疫調節の中心である胸腺とは異なった挙動を示した。
また、56週齢は0%、2%でも3種の免疫チェックポイント発現は増加した。PD-1 及びPD-L1(Cd274)は2%投与群でのRatio は低い傾向にあるが、CTLA-4についてはあまり変わらなかった。56週齢では0%に比べて2%投与群では免疫チェックポイントが低下した。腎臓では、この三種の遺伝子の発現の増減は観察されず、上記の増減は胸腺特異的であった。即ち、胸腺リンパ球の免疫チェックポイントに関わる2種の受容体およびリガンドは加齢によって増加する事が分かった。植物発酵物2%を摂取したマウスは増加率が低かった。さらに。高週齢(56週齢)間では2%植物発酵物摂取群は明らかに減少した。PD-1およびPD-L1の増減率はCTLA4に較べて明瞭であった。この結果から、通常高週齢化によって胸腺のPD-1、PD-L1、Ctla4の遺伝子発現は増えるのに対し、植物発酵物ではいずれも減ることが分かった。
試 験 例 5
PD-1受容体とリガンド(PD-L1およびPD-L2)の結合阻害活性:
マウス由来のPD-1受容体に対するリガンド2種類、PD-L1およびPD-L2の結合試験が可能なビーピーエス バイオサイエンス社製の結合アッセイキットを使用し、in vitroの結合試験を実施した。検出方法は、パーキンエルマー製のAlphaLISA法で測定した。試験に用いた製造例1で製造した植物発酵物のペーストは茶褐色のためペルオキシダーゼによる発色方法は避けた。植物発酵物の10%水溶液をオートクレーブ殺菌し、エッペンドルフ遠心分離機で上清を回収した。なお、本サンプルは微量のビオチンを含むことから、PBSで洗浄したStrptoavidin Mag Sepharoseと混和することでサンプル中のビオチンを除去した。得られた水溶液を10%サンプル溶液と規定した。この溶液をmiliQ水で段階希釈して結合試験に供した。発色はパーキンエルマー製のOptiplate-384を用いて測定した。陽性コントロールとしてPD-1特異抗体を用いて結合阻害活性を確認した。
PD-1受容体とリガンド(PD-L1およびPD-L2)の結合阻害活性:
マウス由来のPD-1受容体に対するリガンド2種類、PD-L1およびPD-L2の結合試験が可能なビーピーエス バイオサイエンス社製の結合アッセイキットを使用し、in vitroの結合試験を実施した。検出方法は、パーキンエルマー製のAlphaLISA法で測定した。試験に用いた製造例1で製造した植物発酵物のペーストは茶褐色のためペルオキシダーゼによる発色方法は避けた。植物発酵物の10%水溶液をオートクレーブ殺菌し、エッペンドルフ遠心分離機で上清を回収した。なお、本サンプルは微量のビオチンを含むことから、PBSで洗浄したStrptoavidin Mag Sepharoseと混和することでサンプル中のビオチンを除去した。得られた水溶液を10%サンプル溶液と規定した。この溶液をmiliQ水で段階希釈して結合試験に供した。発色はパーキンエルマー製のOptiplate-384を用いて測定した。陽性コントロールとしてPD-1特異抗体を用いて結合阻害活性を確認した。
PD-1特異抗体は確かに濃度依存的な直接阻害を示したため本試験は正常に評価できることを示した。この試験におけるIC50%は約2.5nMであった。一方、植物発酵物は、全く阻害活性は示さなかった。植物発酵物は従来知られている免疫チェックポイント阻害剤とは全く異なる挙動を示し。免疫チェックポイントの結合反応に対してこのような態度を示し、しかもin vivoでPD-1発現を抑制する物質は知られていない。

製造例1で製造したペースト状の植物発酵物とみそを混合し、調味料で味を調整してみそ様食品を製造した。

ソフトカプセル:
製造例1で製造したペースト状の植物発酵物と、植物油、レシチンを混合したものを常法でソフトカプセルに充填してソフトカプセルを製造した。

製造例1で製造したペースト状の植物発酵物と、植物油、レシチンを混合したものを常法でソフトカプセルに充填してソフトカプセルを製造した。
実 施 例 4
チュアブル錠:
製造例1で製造したペースト状の植物発酵物と、結晶セルロース、白糖を混合したものを常法で打錠してチュアブル錠を製造した。

本発明によれば、免疫チェックポイント抑制することができるので、これに関連する疾患の治療、予防、改善等に役立つ。
Claims (10)
- 次の(a)ないし(g)
(a)大麦、黒大豆、赤米、黒米、小豆、はと麦、ヒエ、アワおよびキビよりなる群から選ばれる1種又は2種以上の豆・穀物類の麹菌発酵物
(b)ミカン、ブドウ、リンゴ、ヤマブドウ、モモ、カキ、パパイヤ、ナシ、スイカ、ウメ、イチジク、カリン、カボチャ、キンカン、ユズ、ビワ、アンズ、ナツメ、クリ、マタタビおよびスモモよりなる群から選ばれる1種又は2種以上の果実類の酵母および/又は乳酸菌発酵物
(c)紫イモ、菊芋、ニンジン、タマネギ、サツマイモ、里芋、自然薯、大根、赤カブ、ゴボウ、レンコン、ヤーコン、ユリ根、クワイ、しょうが、にんにくおよびウコンよりなる群から選ばれる1種又は2種以上の根菜類・イモ類の酵母および/又は乳酸菌発酵物
(d)キャベツ、紫蘇、桑葉、どくだみ、ヨモギ、クマザサおよびタンポポよりなる群から選ばれる1種又は2種以上の花・葉菜類の酵母および/又は乳酸菌発酵物
(e)コンブ、ワカメおよびモズクよりなる群から選ばれる1種又は2種以上の海藻類の酵母および/又は乳酸菌発酵物
(f)黒ゴマ、クルミおよびギンナンよりなる群から選ばれる1種又は2種以上の種子類の酵母および/又は乳酸菌発酵物
(g)マイタケおよびシイタケよりなる群から選ばれる1種又は2種のキノコ類の酵母および/又は乳酸菌発酵物
の混合物である植物発酵物を有効成分として含有することを特徴とする免疫チェックポイント抑制剤。 - 植物発酵物が、植物発酵物100g当たり以下の組成のアミノ酸を含有するものである請求項1に記載の免疫チェックポイント抑制剤。
アルギニン 0.2~0.6g
リジン 0.1~0.7g
ヒスチジン 0.1~0.4g
フェニルアラニン 0.2~0.8g
チロシン 0.1~0.6g
ロイシン 0.3~1.2g
イソロイシン 0.2~0.8g
メチオニン 0.05~0.30g
バリン 0.2~0.9g
アラニン 0.2~0.9g
グリシン 0.2~0.7g
プロリン 0.4~1.2g
グルタミン酸 1.2~3.0g
セリン 0.2~0.8g
スレオニン 0.2~0.7g
アスパラギン酸 0.4~1.5g
トリプトファン 0.03~0.15g
シスチン 0.05~0.40g - 植物発酵物が、植物発酵物100g当たり以下の組成の有機酸を含有するものである請求項1又は2に記載の免疫チェックポイント抑制剤。
クエン酸 0.5~1.2g
リンゴ酸 0.05~0.5g
コハク酸 0.04~0.3g
乳酸 0.5~6.0g
ギ酸 0.01~0.1g
ピルビン酸 0.005~0.05g
遊離γ-アミノ酪酸 0.01~0.05g - 乳酸菌が、ぺディオコッカス・アシディラクティ(P.acidilacti)、ラクトバチルス・ブレビス(L. brevis)、ロイコノストック・メセンテロイデス(L. mesenteroides)、ラクトバチルス・プランタラム(L. plantarum)、ラクトコッカス・ラクチス(L. lactis)、ラクトバチルス・サケイ(L. sakei)、ペディオコッカス・ペントサセウス(P. pentosaceus)、ラクトバチルス・カゼイ(L. casei)およびラクトバチルス・クルバタス(L. curvatus)よりなる群から選ばれた1種または2種以上である請求項1~3のいずれかの項記載の免疫チェックポイント抑制剤。
- 酵母が、サッカロマイセス・セレビシエ(S. cervisiae)および/又はジゴサッカロマイセス・ロキシー(Z. rouxii)である請求項1~4のいずれかの項記載の免疫チェックポイント抑制剤。
- PD-1、PD-L1およびCTLA4から選ばれる1種以上の発現を抑制するものである請求項1~5のいずれかの項記載の免疫チェックポイント抑制剤。
- 更に、がんの治療薬および/またはがんの代替治療薬を含有する請求項1~6のいずれかの項記載の免疫チェックポイント抑制剤。
- 請求項1~7のいずれかの項記載の免疫チェックポイント抑制剤を含有する飲食品。
- 医薬品である請求項1~7のいずれかの項記載の免疫チェックポイント抑制剤。
- 請求項1~7のいずれかの項記載の免疫チェックポイント抑制剤を含有する放射線療法の副作用低減剤。
