Description
本発明はトリフルオロメチルピリジン類の精製方法、具体的には塩基性化合物の水溶液で処理することを特徴とするトリフルオロメチルピリジン類の精製方法に関する。
トリフルオロメチルピリジン類、特に塩素原子を有するトリフルオロメチルピリジン類は、医薬や農薬(例えば、除草剤、殺虫剤、殺菌剤等)の原料として有用な化合物である。
塩素原子を有するトリフルオロメチルピリジン類の製造方法としては、例えば、β-ピコリン類を、触媒の存在下、塩素及び無水弗化水素と気相で反応させる方法が知られている(特許文献1~4)。この方法は、原料化合物から1工程でかつ短時間で塩素原子を有するトリフルオロメチルピリジン類が得られることから、工業的に有利な方法である。
しかしながら、上記方法で得られるトリフルオロメチルピリジン類は複数の物質の混合物であり、医薬や農薬の原料として使用するためには、分離、精製操作が必要である。
工業的な分離、精製方法としては、例えば、蒸留が挙げられる。しかしながら、混合物に含まれる物質の沸点が近い場合、医薬や農薬の原料としての使用に求められる純度のトリフルオロメチルピリジン類を得ることが難しい場合もある。

本発明者らは、前記課題を解決すべく検討した結果、塩素原子を有するトリフルオロメチルピリジン類の混合物を塩基性化合物の水溶液で処理することにより、特定の目的外の物質が簡便に除去できることを見出し、本発明を完成した。

本発明によれば、工業的に実施できる簡便な方法で、式(I)で表される化合物及び式(II)で表される化合物を含む混合物中の、不純物である式(II)で表される化合物の含有量を減少させることができ、医薬や農薬の原料として有用な式(I)で表される化合物を精製することができる。
1.式(I)で表される化合物の精製方法
本発明の精製方法は、医薬や農薬の原料として有用な式(I)で表される化合物と、不純物である式(II)で表される化合物と、場合によって更に他の化合物とを含む混合物を、塩基性化合物の水溶液で処理する工程を含むことを特徴とする。

本発明における式(I)で表される化合物は、具体的には、式(Ia)で表される化合物(2-クロロ-5-トリフルオロメチルピリジン)、又は式(Ib)で表される化合物(2,3-ジクロロ-5-トリフルオロメチルピリジン)であることが好ましい。
本発明において、塩基性化合物の水溶液で処理される混合物(以下、単に混合物ともいう。)は、式(I)で表される化合物に加えて、式(II)で表される化合物、および場合によって更に他の化合物を不純物として含む。
当該混合物としては、特許文献1~4に記載の製造方法に準じる製造方法、より具体的には、後述する製造方法によって得られる混合物が好適に使用できるが、これに限定されず、例えば、他の製造方法によって得られる混合物を使用することもできる。また、当該混合物としては、後述する製造方法によって得られる生成物を、本技術分野において公知の精製方法(例えば、ろ過、蒸留、抽出、再結晶、クロマトグラフィー等)によって精製して得られる混合物を使用してもよい。
式(I)で表される化合物が式(Ia)で表される化合物である場合、不純物として含有され得る式(II)で表される化合物としては、

式(I)で表される化合物を、医薬や農薬の原料として使用する場合、混合物中の不純物の含量は0.1質量%以下であることが求められる場合がある。例えば、後述する製造方法や特許文献1~4に記載の製造方法に準じて、塩素原子を有するトリフルオロメチルピリジン類を製造した場合、0.1~50質量%程度の式(II)で表される化合物や他の化合物を不純物として含有し、必要に応じて蒸留等により精製する。好ましくは、このような蒸留等によって精製して得られた混合物に対して、本発明を適用することにより、不純物の含有量を、0.1質量%以下とすることが可能になる。
本発明では、式(I)で表される化合物及び式(II)で表される化合物を含む混合物を塩基性化合物の水溶液で処理する工程を含む。
「塩基性化合物の水溶液で処理する工程」とは、塩基性化合物の水溶液を投入した後、撹拌しながら加熱を行う工程を意味する。
「塩基性化合物の水溶液で処理する工程」とは、塩基性化合物の水溶液を投入した後、撹拌しながら加熱を行う工程を意味する。
上記撹拌の時間は、使用する塩基性化合物の種類や濃度、反応温度に応じて適宜選択すればよく、通常0.1~24時間、好ましくは1~12時間である。
上記加熱は、通常60~110℃、好ましくは80~100℃で行なうことができる。
あるいは、公知の分析方法(例えば、高速液体クロマトグラフィー、ガスクロマトグラフィー等)を用いて、不純物である式(II)で表される化合物の含有量が一定量以下となった時点で撹拌及び加熱を終了することもできる。
上記加熱は、通常60~110℃、好ましくは80~100℃で行なうことができる。
あるいは、公知の分析方法(例えば、高速液体クロマトグラフィー、ガスクロマトグラフィー等)を用いて、不純物である式(II)で表される化合物の含有量が一定量以下となった時点で撹拌及び加熱を終了することもできる。
本発明における塩基性化合物としては、特に限定されないが、アルカリ金属水酸化物、アルカリ金属炭酸塩等が挙げられる。
具体的には、アルカリ金属水酸化物としては、水酸化ナトリウム、水酸化カリウム等が挙げられ、アルカリ金属炭酸塩としては、炭酸ナトリウム、炭酸カリウム等が挙げられる。中でも、水酸化ナトリウム、水酸化カリウム等のアルカリ金属水酸化物が好ましい。
具体的には、アルカリ金属水酸化物としては、水酸化ナトリウム、水酸化カリウム等が挙げられ、アルカリ金属炭酸塩としては、炭酸ナトリウム、炭酸カリウム等が挙げられる。中でも、水酸化ナトリウム、水酸化カリウム等のアルカリ金属水酸化物が好ましい。
本発明において、塩基性化合物の水溶液における塩基性化合物の濃度としては、使用する塩基性化合物に応じて適宜選択すればよいが、通常10~50質量%、好ましくは40~50質量%である。
塩基性化合物の水溶液の使用量は、混合物に含まれる式(II)で表される化合物の含有量に応じて選択すればよいが、混合物に対して、通常0.1~20質量%、好ましくは3~5質量%である。
本発明において、式(II)で表される化合物は塩基性化合物の水溶液での処理によって分解されて、より親水性の高い分解物が生じる。従って、塩基性化合物の水溶液での処理終了後、必要に応じて、水、温水等で洗浄、分液することにより、式(II)で表される化合物の分解物が水相側へ除去されて、式(I)で表される化合物をより簡便に精製することができる。
例えば、式(II)で表される化合物が、R2及びR4が、それぞれ同一又は異なって、水素原子又は塩素原子であり、R3がフッ素原子である化合物の場合(例えば、以下に示す化合物である場合)、当該化合物の沸点は式(I)で表される化合物の沸点と近い。よって、蒸留等の通常用いられる精製手段では、当該化合物を分離及び除去することが困難である。このような場合でも、塩基性化合物の水溶液で処理することによって、以下に示す化合物が、α位のフッ素原子がヒドロキシ基に変換された親水性の高い分解物となることで、水相側へ除去されて、式(I)で表される化合物をより簡便に精製することができる。
2.式(I)で表される化合物及び式(II)で表される化合物を含む混合物の製造方法
本発明において、塩基性化合物の水溶液で処理される混合物としては、式(I)で表される化合物及び式(II)で表される化合物を含有している限り、特に限定されないが、好ましくは、特許文献1~4に記載の製造方法に準じる方法、即ち、式(A)で表されるβ-ピコリン類を、触媒と希釈剤の存在下、塩素及び/又は無水弗化水素と気相で反応させる方法により得られる混合物である。以下、当該製造方法について詳述する。
本発明において、塩基性化合物の水溶液で処理される混合物としては、式(I)で表される化合物及び式(II)で表される化合物を含有している限り、特に限定されないが、好ましくは、特許文献1~4に記載の製造方法に準じる方法、即ち、式(A)で表されるβ-ピコリン類を、触媒と希釈剤の存在下、塩素及び/又は無水弗化水素と気相で反応させる方法により得られる混合物である。以下、当該製造方法について詳述する。

本製造方法により得られた式(I)で表される化合物及び式(II)で表される化合物の混合物には、さらに、不純物として、前記したような各種の化合物を含む場合がある。
式(A)で表されるβ-ピコリン類としては、β-ピコリン、2-クロロ-β-ピコリン、6-クロロ-β-ピコリン等が挙げられる。
特に、式(Ia)で表される化合物を製造する場合は、原料として入手し易いβ-ピコリンを使用することが有利である。
また、式(Ib)で表される化合物を製造する場合、Xが塩素原子であり、Z及びYが水素原子であり、CW3がCF3である式(A)で表される化合物(2-クロロ-5-トリフルオロメチルピリジン、即ち、式(Ia)で表される化合物)を原料として使用することもできる。
特に、式(Ia)で表される化合物を製造する場合は、原料として入手し易いβ-ピコリンを使用することが有利である。
また、式(Ib)で表される化合物を製造する場合、Xが塩素原子であり、Z及びYが水素原子であり、CW3がCF3である式(A)で表される化合物(2-クロロ-5-トリフルオロメチルピリジン、即ち、式(Ia)で表される化合物)を原料として使用することもできる。
触媒としては、アルミニウム、クロム、鉄、ニッケル、マンガン、及びコバルトからなる群から選択される少なくとも一種の金属元素のハロゲン化物を含む触媒が挙げられる。
上記ハロゲン化物としては、より具体的には、水和三弗化アルミニウム(AlF3・3H2O)、三弗化アルミニウム(AlF3)、水和三弗化クロム(CrF3・3H2O)、三弗化クロム(CrF3)、水和弗化第1鉄(FeF2・8H2O)、弗化第1鉄(FeF2)、弗化第2鉄(FeF3)、塩化第1鉄(FeCl2)、塩化第2鉄(FeCl3)、水和弗化第1ニッケル(NiF2・3H2O)、弗化第2ニッケル(NiF3)、弗化第1マンガン(MnF2)、弗化第二マンガン(MnF3)、四弗化マンガン(MnF4)、弗化第1コバルト(CoF2)、弗化第二コバルト(CoF3)等が挙げられる。
触媒の使用量は、反応条件により一概に規定できないが、通常、原料の式(A)で表される化合物1モルに対して0.01~3モルである。
通常、触媒は活性炭、活性アルミナ等の担体と混合し、適当な大きさの粒状、ペレット状に成型してから固定床又は流動床として存在させる。また、触媒として弗化物を使用する場合、触媒は前記した金属元素の弗化物の形態で直接、反応管に入れて存在させる方法もあるが、工業的には、対応する金属元素の酸化物、塩化物、炭酸塩の型態で反応管に入れ、無水弗化水素と反応させて弗化物に変換して存在させる方法が有利である。
希釈剤としては、例えば、四塩化炭素、クロロホルム、塩化メチレン、F-112(CFCl2・CFCl2)、F-113(CF2Cl・CFCl2)などのハロゲン化炭化水素の有機溶媒、窒素、ヘリウム、アルゴンなどの不活性気体が使用でき、これら希釈剤は燃焼、炭化、タール状副生物の生成などを抑制する機能を有する。希釈剤の使用量は、通常、式(A)で表される化合物1モルに対して2~70モルである。
塩素及び無水弗化水素の使用量は原料の式(A)で表される化合物の種類、目的物の種類、反応装置などの違いによって一概に規定できないが、通常式(A)で表される化合物1モルに対して、それぞれ2~15モル及び2~60モルである。
本製造方法において、式(A)で表される化合物、希釈剤、並びに塩素及び/又は無水弗化水素を別々に反応器へ供給してもよいし、これらの混合物として供給してもよい。また、これらを同時に又は順次に、或は一括又は分割して供給することもできる。
反応温度は、通常200~600℃であり、反応混合物の反応帯域における滞留時間は、通常3~60秒である。塩素原子を有するトリフルオロメチルピリジン類の生成率は通常、90%以上である。
通常、反応器からは式(I)で表される化合物を主成分とする生成物、式(II)で表される化合物、中間生成物、塩化水素、未反応の弗化水素及び塩素、希釈剤を含有するガス状物質が排出され、適当な冷却、凝縮装置を経て、反応生成物は液体混合物として得られる。得られた液体混合物中に中間生成物が含まれているときには、これら中間生成物は、未反応原料又は希釈剤と共に分離、回収し、必要に応じて通常の手法による還元によって脱塩素化を行い、式(A)で表される化合物として反応帯域へ循環使用することができる。
このようにして得られた式(I)で表される化合物と、不純物である式(II)で表される化合物と、場合によって更に他の化合物とを含む混合物を、そのまま本発明の精製方法に供してもよいが、好ましくは、本技術分野において公知の精製方法(例えば、ろ過、蒸留、抽出、再結晶、クロマトグラフィー等)によってさらに精製して得られる混合物を本発明の精製方法に供する。
次に本発明の実施例を記載するが、本発明はこれらに限定して解釈されるものではない。
なお、以下の実施例において使用する略称は以下の通りである。

精製例1
SUS製撹拌機を備えたジャケット付タンク(10m3)に、粗DCTF混合物(7500kg(5m3))を仕込んだ。次に粗DCTF混合物の質量に対し4質量%に相当する48質量%NaOH水溶液(300kg)を仕込んだ。その後、80℃で8時間加熱撹拌した。
下記測定条件でのGC分析によりDCFTFのピーク面積率(Peak area%)が0.1PA%以下であることを確認した後、60℃の温水(3750kg)を投入し、30分撹拌した。50℃に冷却し、30分静置し、分液することにより、精製されたDCTFを得た。
なお、「粗DCTF混合物」とは、DCTF、DCFTF及び56CFTFを含有する混合物を意味する。
SUS製撹拌機を備えたジャケット付タンク(10m3)に、粗DCTF混合物(7500kg(5m3))を仕込んだ。次に粗DCTF混合物の質量に対し4質量%に相当する48質量%NaOH水溶液(300kg)を仕込んだ。その後、80℃で8時間加熱撹拌した。
下記測定条件でのGC分析によりDCFTFのピーク面積率(Peak area%)が0.1PA%以下であることを確認した後、60℃の温水(3750kg)を投入し、30分撹拌した。50℃に冷却し、30分静置し、分液することにより、精製されたDCTFを得た。

下記測定条件でのGC分析によりDCFTFのピーク面積率が0.1PA%以下であることを確認した後、精製例1に準じて後処理を行ない、精製されたDCTFを得た。

撹拌機を備えた反応容器に、粗DCTF混合物(150g)を仕込んだ。次に粗DCTF混合物の質量に対し12.5質量%に相当する48質量%NaOH水溶液を仕込んだ。その後、80℃で11時間加熱撹拌した。
下記測定条件でのGC分析によりDCFTFのピーク面積率が0.1PA%以下であることを確認した後、精製例1に準じて、後処理を行ない、精製されたDCTFを得た。
撹拌機を備えた反応容器に、粗DCTF混合物(150g)を仕込んだ。次に粗DCTF混合物の質量に対し12.5質量%に相当する48質量%NaOH水溶液を仕込んだ。その後、80℃で11時間加熱撹拌した。
下記測定条件でのGC分析によりDCFTFのピーク面積率が0.1PA%以下であることを確認した後、精製例1に準じて、後処理を行ない、精製されたDCTFを得た。
精製例1~4についてGC分析の結果を表1に示す。なお、GCの測定条件は以下の通りであった。

精製例5
丸底フラスコ(1L)に粗CTF混合物(300g、CTF:98PA%以上)を仕込み、次に粗CTF混合物の質量に対し5質量%に相当する25質量%NaOH水溶液(15g)を仕込み、100℃で15分加熱撹拌した。
下記測定条件でのHPLCにて分析した結果、粗CTF混合物における26CFTFのピーク面積率は、0.307PA%であったが、処理後は0.167PA%に半減した。26CFTFのピーク面積率を0.1PA%以下とするには、処理時間を更に長くすればよい。
なお、「粗CTF混合物」とは、CTF及び26CFTFを含有する混合物を意味する。
丸底フラスコ(1L)に粗CTF混合物(300g、CTF:98PA%以上)を仕込み、次に粗CTF混合物の質量に対し5質量%に相当する25質量%NaOH水溶液(15g)を仕込み、100℃で15分加熱撹拌した。
下記測定条件でのHPLCにて分析した結果、粗CTF混合物における26CFTFのピーク面積率は、0.307PA%であったが、処理後は0.167PA%に半減した。26CFTFのピーク面積率を0.1PA%以下とするには、処理時間を更に長くすればよい。
なお、「粗CTF混合物」とは、CTF及び26CFTFを含有する混合物を意味する。

CTFおよびDCTFの製造例
製造例(1)
反応器として、反応部が内径97.1mm、高さ1570mmの触媒流動床を有するインコネル製竪型反応管を設置し、原料物質及び不活性希釈剤用に内径30mm、長さ1000mmのインコネル製予熱管を2本接続したものを使用し、反応管及び予熱管を温度制御できるように電熱器及び断熱材で覆った。
製造例(1)
反応器として、反応部が内径97.1mm、高さ1570mmの触媒流動床を有するインコネル製竪型反応管を設置し、原料物質及び不活性希釈剤用に内径30mm、長さ1000mmのインコネル製予熱管を2本接続したものを使用し、反応管及び予熱管を温度制御できるように電熱器及び断熱材で覆った。
無水塩化第2鉄277gを粒径105~250μmの三弗化アルミニウム2.2kgに含浸させたものを触媒充填部へ入れ、200℃に加熱されて無水弗化水素を2.3L/分で1時間導入して触媒の活性化を行った。
反応器を400℃に加熱し、β-ピコリンを6.8g/分及び窒素ガスを9.9L/分の割合になるよう予熱管を通じ、また塩素ガスを7.4L/分及び無水弗化水素7.4L/分の割合になるよう予熱管を通じ、それぞれ約200℃の混合ガスとして反応管に導入し、約30時間にわたって反応させた。この間活性化触媒は300g/時間の割合で連続的に供給、排出された。反応混合物の管内滞留時間は約3.4秒であった。
反応器より排出するガスは水洗塔及びアルカリ洗浄塔に通され、凝縮生成物が分離され、アンモニア水溶液で中和されて水素気蒸留によって油状物19.11kgを得た。この油状物にDCFTFが1PA%程度、56CFTFが2PA%程度含まれる。
この油状物を蒸留してβ-トリフルオロメチルピリジンを主成分とする初留分1.53kg、CTFを主成分とする主留分9.56kg及び後留分7.62kgを得た。なお、後留分にはCTF3.7PA%、2-CTF14.5PA%、2,6-DCTF47.7PA%及びその他34.1PA%が含まれていた。
製造例(2)
前記製造例(1)において、β-ピコリンを4.6g/分、β-トリフルオロメチルピリジンを3.4g/分、塩素ガスを5.8L/分及び無水弗化水素を5L/分の割合で導入させた以外は同様にして反応させた。反応混合物の滞留時間は約4秒であり、反応は約8時間連続的に行なわれた。反応生成物は同様にして後処理されて油状物5.47kgを得た。
前記製造例(1)において、β-ピコリンを4.6g/分、β-トリフルオロメチルピリジンを3.4g/分、塩素ガスを5.8L/分及び無水弗化水素を5L/分の割合で導入させた以外は同様にして反応させた。反応混合物の滞留時間は約4秒であり、反応は約8時間連続的に行なわれた。反応生成物は同様にして後処理されて油状物5.47kgを得た。
この油状物の主な組成は、β-トリフルオロメチルピリジン8.2PA%、CTF57.8PA%、2-CTF9.5PA%、DCTF16.1PA%及びその他8.4PA%であった。
この油状物を蒸留することにより、CTFを主成分とする留分を得ることができる。

製造例(3)
反応器として、反応部が内径30mm、高さ500mmの触媒流動床を有するインコネル製竪型反応管であり、これに内径20mm、長さ300mmのインコネル製予熱管を2本接続し、これらを温度制御できるように電熱器及び断熱材で覆ったものを使用した。

塩化第2鉄24gをエタノール200mlに溶解した溶液と、活性炭(80~200メッシュ)120gとを混合し、乾燥して得られたもの60gを反応管の触媒充填部に入れ、約200℃に昇温した後、塩素ガスを1.3L/分の割合になるよう約3時間にわたって導入し、触媒を活性化した。
CTFと窒素ガスとの混合ガスを約200~250℃に予熱して前者が0.5g/分及び後者が1.0L/分の割合になるよう一方の予熱管を通じ、また、同温度に予熱した塩素ガスと窒素ガスとの混合ガスを前者が0.25L/分及び後者が0.6L/分の割合になるよう他方の予熱管を通じ、それぞれ反応管に導入し、250℃の温度で約4時間にわたって反応させた。
反応器より排出するガスを水洗塔及びアルカリ洗浄塔に通じて凝縮させ、油状物を分液して採取し、水洗後芒硝で乾燥して油状物110gを得た。
この油状物の主な組成は、DCTF74.1PA%、256TCTF7.1PA%、CTF16.0PA%、その他2.7PA%であった。
この油状物を蒸留することにより、DCTFを主成分とする留分を得ることができる。

本発明を詳細にまた特定の実施態様を参照して説明したが、本発明の精神と範囲を逸脱することなく様々な変更や修正を加えることができることは当業者にとって明らかである。本出願は2017年4月4日出願の日本特許出願(特願2017-74229)に基づくものであり、その内容はここに参照として取り込まれる。
Claims (9)
- 前記式(II)中、R2及びR4が、それぞれ同一又は異なって、水素原子又は塩素原子であり、R3がフッ素原子である、請求項1~3のいずれか1項に記載の精製方法。
- 前記塩基性化合物が、アルカリ金属水酸化物、及びアルカリ金属炭酸塩からなる群から選択される少なくとも一種である、請求項1~4のいずれか1項に記載の精製方法。
- 前記塩基性化合物が、アルカリ金属水酸化物である、請求項1~4のいずれか1項に記載の精製方法。
- 前記塩基性化合物の水溶液における前記塩基性化合物の濃度が、40~50質量%である、請求項1~6のいずれか1項に記載の精製方法。
- 前記塩基性化合物の水溶液で処理する工程が、60~110℃で行なわれる、請求項1~7のいずれか1項に記載の精製方法。
