本発明の一実施形態によると、下記式(1)または式(2)で示される2-アミノエチルメタクリレートのケチミン化合物を水および塩化水素と反応させ、下記式(3)で示される2-アミノエチルメタクリレート塩酸塩を含む混合物を得る工程(A)を含む、2-アミノエチルメタクリレート塩酸塩の製造方法が提供される。ここで、前記工程(A)において、2-アミノエチルメタクリレートのケチミン化合物の全量に対する塩化水素の全量の割合は、当量比で1.0~1.5の範囲であり、2-アミノエチルメタクリレートのケチミン化合物の全量に対する水の全量の割合は、当量比で1.0~2.0の範囲である。
(式(1)中、R1およびR2は、それぞれ独立に、炭素数1~12のアルキル基もしくはアリール基を示す。)
(式(2)中、R3は、それが結合している炭素原子と共に環を形成する炭素数3~10の2価の炭化水素基であり、その一部に二重結合を有していてもよい。)
式(2)において、R3は、それが結合している炭素原子と共に環を形成する炭素数3~10の2価の炭化水素基であり、その一部に二重結合を有していてもよい。炭素数は4~7であることが好ましく、5~6であることがより好ましい。二重結合の数は、0~2個、好ましくは0~1個、より好ましくは0個である。具体的には、R3がそれが結合している炭素原子と共に形成する環としては、シクロペンタン、シクロヘキサン、2-シクロペンテン、2-シクロヘキセン、2,4-シクロペンタジエン、2,5-シクロヘキサジエンが挙げられ、これらのうちシクロペンタン、またはシクロヘキサンが好ましい。
また、2-アミノエチルメタクリレートのケチミン化合物としては、式(1)および式(2)のうち、式(1)で示される化合物を使用することがより好ましい。

以下、工程(A)について説明する。
工程(A)は、式(1)または(2)で示される2-アミノエチルメタクリレートのケチミン化合物を水および塩化水素と反応させ、式(3)で示される2-アミノエチルメタクリレート塩酸塩を含む混合物を得る工程である。
式(1)または式(2)で示される2-アミノエチルメタクリレートのケチミン化合物を、有機溶剤の存在下あるいは非存在下で塩化水素および水と混合することにより、先ず水との反応でケトンを遊離して2-アミノエチルメタクリレートが得られる。その後直ちに塩化水素との中和反応を行うことにより、目的化合物である式(3)で示される2-アミノエチルメタアクリレート塩酸塩に変換される。上記反応を経て、式(3)で示される2-アミノエチルメタクリレート塩酸塩を主成分として含む混合物が得られる。以下、工程(A)を脱保護/中和反応工程と表記することがある。
上記脱保護/中和反応工程を反応容器を使用して行う場合、原料注加工程は、式(3)で示される2-アミノエチルメタクリレート塩酸塩の合成に供される反応原料の反応容器内への供給を開始した時点から、反応原料の供給が終了する時点までの期間のことを指す。反応原料は、式(1)または式(2)で示される2-アミノエチルメタクリレートのケチミン化合物、塩化水素、および水を含む。反応原料は、反応溶媒として使用される有機溶媒を含んでもよい。反応原料を反応容器内に供給する方法は、上述したとおりである。原料注加工程では、酸と塩基の混合により中和熱が多量に発生するため、冷却に努める。原料注加工程での好ましい反応温度の範囲は、通常-10~60℃であり、好ましくは-10~40℃、より好ましくは-10~20℃である。
熟成工程は、反応原料の供給が終了した時点から、反応が終了する時点までの期間、反応液を一定の温度下で撹拌する工程のことを指す。熟成工程における反応の終了時点は、反応液をガスクロマトグラフィーで分析し、2-アミノエチルメタクリレートのケチミン化合物の消失が確認された時点である。熟成工程では若干の発熱はあるが、反応温度をほぼ一定に保つことが重要で、結晶が多量に析出するため撹拌も大切になる。熟成工程での望ましい反応温度の範囲は、通常-10~40℃、好ましくは-10~20℃である。原料注加工程および熟成工程のいずれにおいても、反応温度が-10℃を下回る場合、反応速度が遅くなり効率が悪く、40℃を超える場合、重合反応などの副反応が起きやすい傾向にある。
上記脱保護/中和反応工程で生成した式(3)で示される2-アミノエチルメタクリレート塩酸塩は、反応終了時、2-アミノエチルメタクリレート塩酸塩を主成分とする混合物中に白色結晶として存在する。ここで説明する工程(B)は、上記混合物から、目的とする式(3)で示される2-アミノエチルメタクリレート塩酸塩の固体を分離精製する工程であり、工程(B)は、工程(A)で得られた混合物から固体析出物を分離する工程(b1)と、前記固体析出物を溶媒により洗浄する工程(b2)と、洗浄後の固体析出物を乾燥する工程(b3)とを含む。
洗浄工程において、洗浄に使用する溶媒に混合する場合、洗浄に使用する溶媒に対する重合禁止剤の割合は、0.0001~1wt%であることが好ましい。
(ケチミン化工程)
マグネチックスターラ―、サイズ6mmのマクマホンパッキンを高さ300mmになるまで充填した内径25mmφの充填塔を充填した精留塔、ディーンスタークトラップ、および還流冷却管を備えた内容積1000mlの三つ口フラスコに、2-アミノエタノール100.0g(1.6モル)、メチルイソブチルケトン443g(4.4モル)、およびシクロヘキサン84g(1.0モル)を仕込み、釜液温度90.7~105.9℃で加熱還流し、ディーンスタークトラップにたまった水を系外に抜き出しながらケチミン合成を行った。2時間後、2-アミノエタノールの転化率は100%、2-アミノエタノールメチルイソブチルケチミンの収率は99%となった。
得られた反応液から、余剰のメチルイソブチルケトンとシクロヘキサンを減圧蒸留により留去した。残った釜液に、亜鉛アセチルアセトン錯体20.1g、メチルメタクリレート317g(3.2モル)、およびp-メトキシフェノール0.32gを加え、ディーンスタークトラップを通常の蒸留ヘッド(還流量調整可能なもの)に置き換えて、釜液温度107.3~126.4℃で加熱還流し、エステル交換反応を行った。副生したメタノールは、メタノール-メチルメタクリレートの共沸混合物として系外に抜き出した。重合防止措置として、精留塔の塔頂より1%-p-メトキシフェノール-メチルメタクリレート溶液を0.8g/時間で供給し、反応液には空気を6ml/分で供給した。5時間後、反応液をガスクロマトグラフィーで分析したところ、2-アミノエタノールメチルイソブチルケチミンの転化率は100%であった。仕込んだ2-アミノエタノール基準の2-アミノエチルメタクリレートメチルイソブチルケチミン収率は62%であった。得られた反応液を減圧蒸留に供して、まず余剰のメチルメタクリレートを留去し、次いで生成物の2-アミノエチルメタクリレートメチルイソブチルケチミンを分離した。2-アミノエチルメタクリレートメチルイソブチルケチミンの沸点は110℃/0.53kPa、ガスクロマトグラフィー純度90%以上の留分の合計収量は146g(回収率は69%)であった。
(ケチミン化工程)
マグネチックスターラー、ディーンスタークトラップ、サイズ6mmのマクマホンパッキンを高さ300mmになるまで充填した内径25mmφの充填塔を充填した精留塔、および還流冷却管を備えた内容積1000mlの三つ口フラスコに、2-アミノエタノール49.0g(0.80モル)、メチルエチルケトン173g(2.4モル)、およびヘキサン162g(1.9モル)を仕込み、釜液温度63.7~69.8℃で加熱還流し、ディーンスタークトラップにたまった水を系外に抜き出しながらケチミン合成を行った。6時間後、2-アミノエタノールの転化率は100%、2-アミノエタノールメチルエチルケチミンの収率は99%となった。
得られた反応液から余剰のメチルエチルケトンとヘキサンを留去し、残った釜液に亜鉛アセチルアセトン錯体10.6g、p-メトキシフェノール0.16g、およびメチルメタクリレート160g(1.6モル)を加え、ディーンスタークトラップを通常の蒸留ヘッド(還流量調整可能なもの)に置き換えて、釜液温度97.9~113.9℃で加熱還流し、エステル交換反応を行った。副生したメタノールは、メタノール-メチルメタクリレートの共沸混合物として系外に抜き出した。重合防止措置として、精留塔の塔頂より4%-p-メトキシフェノール-メチルメタクリレート溶液を2g/時間で供給し、反応液には空気を10ml/分で供給した。3時間後、反応液をガスクロマトグラフィーで分析したところ、2-アミノエタノールメチルエチルケチミンの転化率は96%であった。仕込んだ2-アミノエタノール基準の2-アミノエチルメタクリレートメチルエチルケチミン収率は、45%であった。得られた反応液を減圧蒸留に供して、まず余剰のメチルメタクリレートを留去し、次いで生成物の2-アミノエチルメタクリレートメチルエチルケチミンを分離した。2-アミノエチルメタクリレートメチルエチルケチミンの沸点は86.1℃/0.47kPa、ガスクロマトグラフィー純度90%以上の留分の合計収量は44g(回収率は67%)であった。
撹拌翼および滴下チューブを備えた内容積2000ml三つ口フラスコに、メチルイソブチルケトン770g(7.7モル、水200ppm含有)および水16.1g(0.894モル)を仕込み、メタノール-氷浴で冷却した。ここで仕込んだ水の合計は、メチルイソブチルケトン中に含まれる水と併せて0.903モルであった。ここに撹拌下、塩化水素ガス32.5g(0.890モル)を供給しながら、同時に合成例1により得られた2-アミノエチルメタクリレートメチルイソブチルケチミン181g(0.858モル)を40分かけて滴下した。ここで、系内は0℃以下に保ち、2-アミノエチルメタクリレートメチルイソブチルケチミンと塩化水素ガスの混合を開始した時点から混合を終了した時点まで、2-アミノエチルメタクリレートメチルイソブチルケチミンの累積供給量に対する塩化水素ガスの累積供給量の割合は当量比で1.04/1.00を保つように制御した。添加終了後、20℃水浴で23時間撹拌しながら熟成した。反応に供された2-アミノエチルメタクリレートメチルイソブチルケチミンに対する塩化水素の割合は、当量比で1.04であり、2-アミノエチルメタクリレートメチルイソブチルケチミンに対する水の割合は、当量比で1.05であった。熟成後、系内には目的化合物である2-アミノエチルメタクリレート塩酸塩が白色結晶として大量に析出した。この白色結晶をガラスフィルターによりろ別し、メチルイソブチルケトン400gで5回洗浄後、減圧乾燥を行い、純度99.4%の2-アミノエチルメタクリレート塩酸塩を139g得た。2-アミノエチルメタクリレートメチルイソブチルケチミン基準の単離収率は、97%であった。
撹拌翼および滴下チューブを備えた内容積2000ml三つ口フラスコに、合成例1により得られた2-アミノエチルメタクリレートメチルイソブチルケチミン195g(0.924モル)、メチルイソブチルケトン770g(7.7モル、水分88ppm含有)および水17.4g(0.967モル)を仕込み、氷水で冷却した。ここで仕込んだ水の合計は、メチルイソブチルケトン中に含まれる水と併せて0.970モルであった。ここに撹拌下、塩化水素ガス35.1g(0.962モル)を20℃以下に保ちながら、60分かけて供給し、供給終了後、30℃水浴で6時間撹拌しながら熟成した。反応に供された2-アミノエチルメタクリレートメチルイソブチルケチミンに対する塩化水素の割合は、当量比で1.04であり、2-アミノエチルメタクリレートメチルイソブチルケチミンに対する水の割合は、当量比で1.05であった。熟成後、系内には目的化合物である2-アミノエチルメタクリレート塩酸塩が白色結晶として大量に析出した。この白色結晶をガラスフィルターによりろ別し、メチルイソブチルケトン300gで5回洗浄後、減圧乾燥を行い、純度99.0%の2-アミノエチルメタクリレート塩酸塩を148g得た。2-アミノエチルメタクリレートメチルイソブチルケチミン基準の単離収率は、96%であった。
撹拌翼および滴下チューブを備えた内容積2000ml三つ口フラスコに、メチルイソブチルケトン770g(7.7モル、水分200ppm含有)および水17.1g(0.950モル)を仕込み、メタノール-氷浴で冷却した。ここで仕込んだ水の合計は、メチルイソブチルケトン中に含まれる水と併せて0.959モルであった。ここに撹拌下、塩化水素ガス40.3g(1.10モル)を供給しながら、同時に合成例1により得られた2-アミノエチルメタクリレートメチルイソブチルケチミン193g(0.915モル)を40分かけて滴下した。ここで、系内は0℃以下に保ち、2-アミノエチルメタクリレートメチルイソブチルケチミンと塩化水素ガスの混合を開始した時点から混合を終了した時点まで、2-アミノエチルメタクリレートメチルイソブチルケチミンの累積供給量に対する塩化水素ガスの累積供給量の割合は当量比で1.2/1.0を保つように制御した。添加終了後、20℃水浴で93時間撹拌しながら熟成した。反応に供された2-アミノエチルメタクリレートメチルイソブチルケチミンに対する塩化水素の割合は、当量比で1.2であり、2-アミノエチルメタクリレートメチルイソブチルケチミンに対する水の割合は、当量比で1.05であった。熟成後、系内には目的化合物である2-アミノエチルメタクリレート塩酸塩が白色結晶として大量に析出した。この白色結晶をガラスフィルターによりろ別し、メチルイソブチルケトン400gで5回洗浄後、減圧乾燥を行い、純度98.9%の2-アミノエチルメタクリレート塩酸塩を136g得た。2-アミノエチルメタクリレートメチルイソブチルケチミン基準の単離収率は、89%であった。ここで熟成23時間後の転化率は81%、熟成93時間後の転化率は97%であった。転化率とは、2-アミノエチルメタクリレートメチルイソブチルケチミンの消費された割合を意味する。
撹拌翼および滴下チューブを備えた内容積2000ml三つ口フラスコに、メチルイソブチルケトン770g(7.7モル、水200ppm含有)および水17.1g(0.950モル)を仕込み、メタノール-氷浴で冷却した。ここで仕込んだ水の合計は、メチルイソブチルケトン中に含まれる水と併せて0.959モルであった。ここに撹拌下、塩化水素ガス50.4g(1.38モル)を供給しながら、同時に合成例1により得られた2-アミノエチルメタクリレートメチルイソブチルケチミン193g(0.915モル)を40分かけて滴下した。ここで、系内は0℃以下に保ち、2-アミノエチルメタクリレートメチルイソブチルケチミンと塩化水素ガスの混合を開始した時点から混合を終了した時点まで、2-アミノエチルメタクリレートメチルイソブチルケチミンの累積供給量に対する塩化水素ガスの累積供給量の割合は当量比で1.5/1.0を保つように制御した。添加終了後、20℃水浴で182時間撹拌しながら熟成した。反応に供された2-アミノエチルメタクリレートメチルイソブチルケチミンに対する塩化水素の割合は、当量比で1.5であり、2-アミノエチルメタクリレートメチルイソブチルケチミンに対する水の割合は、当量比で1.05であった。熟成後、系内には目的化合物である2-アミノエチルメタクリレート塩酸塩が白色結晶として大量に析出した。この白色結晶をガラスフィルターによりろ別し、メチルイソブチルケトン400gで5回洗浄後、減圧乾燥を行い、純度96.5%の2-アミノエチルメタクリレート塩酸塩を112g得た。2-アミノエチルメタクリレートメチルイソブチルケチミン基準の単離収率は、71%であった。ここで熟成24時間後の転化率は60%、熟成182時間後の転化率は90%であった。転化率とは、2-アミノエチルメタクリレートメチルイソブチルケチミンの消費された割合を意味する。

撹拌翼および滴下チューブを備えた内容積1000ml三つ口フラスコに、合成例2により得られた2-アミノエチルメタクリレートメチルエチルケチミン98.4g(0.54モル)、メチルエチルケトン324g(4.5モル、水525ppm含有)および水10.0g(0.55モル)を仕込み氷水で冷却した。ここで仕込んだ水の合計は、メチルエチルケトン中に含まれる水と併せて0.57モルであった。ここに撹拌下、塩化水素ガス20.3g(0.56モル)を20℃以下に保ちながら、30分かけて供給した。供給終了後、30℃水浴で6時間撹拌しながら熟成した。反応に供された2-アミノエチルメタクリレートメチルエチルケチミンに対する塩化水素の割合は、当量比で1.04であり、2-アミノエチルメタクリレートメチルエチルケチミンに対する水の割合は、当量比で1.05であった。熟成後、系内には目的化合物である2-アミノエチルメタクリレート塩酸塩が白色結晶として大量に析出した。この白色結晶をガラスフィルターによりろ別し、メチルエチルケトン200gで5回洗浄後、減圧乾燥を行い、純度99.7%の2-アミノエチルメタクリレート塩酸塩を75g得た。2-アミノエチルメタクリレートメチルエチルケチミン基準の単離収率は、84%であった。
撹拌翼および滴下チューブを備えた内容積5000ml三つ口フラスコに、メチルエチルケトン2454g(34.1モル、水525ppm含有)および水74.5g(4.139モル)を仕込み、メタノール-氷浴で冷却した。ここで仕込んだ水の合計は、メチルエチルケチミン中に含まれる水と併せて4.210モルであった。ここに撹拌下、塩化水素ガス152.3g(4.173モル)を供給しながら、同時に合成例2により得られた2-アミノエチルメタクリレートメチルエチルケチミン734g(4.011モル)を60分かけて滴下した。ここで、系内は20℃以下に保ち、2-アミノエチルメタクリレートメチルエチルケチミンと塩化水素ガスの混合を開始した時点から混合を終了した時点まで、2-アミノエチルメタクリレートメチルエチルケチミンの累積供給量に対する塩化水素ガスの累積供給量の割合は当量比で1.04/1.00を保つように制御した。添加終了後、30℃水浴で6時間撹拌しながら熟成した。反応に供された2-アミノエチルメタクリレートメチルエチルケチミンに対する塩化水素の割合は、当量比で1.04であり、2-アミノエチルメタクリレートメチルエチルケチミンに対する水の割合は、当量比で1.05であった。熟成後、系内には目的化合物である2-アミノエチルメタクリレート塩酸塩が白色結晶として大量に析出した。この白色結晶をガラスフィルターによりろ別し、メチルエチルトン800gで5回洗浄後、減圧乾燥を行い、純度99.9%の2-アミノエチルメタクリレート塩酸塩を617g得た。2-アミノエチルメタクリレートメチルエチルケチミン基準の単離収率は、93%であった。
マグネチックスターラ―および滴下チューブを備えた内容積500ml三つ口フラスコに、濃塩酸83.8g(塩化水素36.3重量%含有)を仕込み、氷浴で冷却した。ここに、合成例2で得られた2-アミノエチルメタクリレートメチルエチルケチミン147g(0.803モル)を撹拌下、2時間かけて滴下した。反応に供された2-アミノエチルメタクリレートメチルエチルケチミンに対する塩化水素の割合は、当量比で1.04であり、2-アミノエチルメタクリレートメチルエチルケチミンに対する水の割合は、当量比で3.69であった。滴下終了後、20℃水浴で2時間撹拌しながら熟成を行い、テトラヒドロフラン832gを加え、20℃で15時間静置した。このとき、結晶の析出は確認されず、固体の2-アミノエチルメタクリレート塩酸塩は得られなかった。
Claims (16)
- 下記式(1)または式(2)で示される2-アミノエチルメタクリレートのケチミン化合物を水および塩化水素と反応させ、下記式(3)で示される2-アミノエチルメタクリレート塩酸塩を含む混合物を得る工程(A)を含む、2-アミノエチルメタクリレート塩酸塩の製造方法であって、
前記工程(A)において、2-アミノエチルメタクリレートのケチミン化合物の全量に対する塩化水素の全量の割合は、当量比で1.0~1.5の範囲であり、2-アミノエチルメタクリレートのケチミン化合物の全量に対する水の全量の割合は、当量比で1.0~2.0の範囲である、2-アミノエチルメタクリレート塩酸塩の製造方法。
(式(1)中、R1およびR2は、それぞれ独立に、炭素数1~12のアルキル基もしくはアリール基を示す。)
(式(2)中、R3は、それが結合している炭素原子と共に環を形成する炭素数3~10の2価の炭化水素基であり、その一部に二重結合を有していてもよい。)
- 前記式(1)におけるR1およびR2が、それぞれ独立に、炭素数1~4のアルキル基である請求項1に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記式(1)におけるR1がメチル基であり、R2がエチル基またはイソブチル基である、請求項2に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記式(1)におけるR2がイソブチル基である、請求項3に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記式(1)におけるR2がエチル基である、請求項3に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記塩化水素が塩化水素ガスである、請求項1~5のいずれかに記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記式(1)で示される2-アミノエチルメタクリレートのケチミン化合物と水および塩化水素との反応は、2-アミノエチルメタクリレートのケチミン化合物に対して塩化水素の混合を開始した時点から塩化水素の全量の混合が終了する時点までの全期間において、2-アミノエチルメタクリレートのケチミン化合物の累積供給量に対する塩化水素の累積供給量の割合が、当量比で1.0以上を維持するように行われる、請求項1~6のいずれか1項に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記2-アミノエチルメタクリレートのケチミン化合物と水および塩化水素との反応は、テトラヒドロフラン、メチルイソブチルケトン、メチルエチルケトン、アセトン、およびジオキサンからなる群より選ばれる少なくとも1つの有機溶媒の存在下で行われる、請求項1~7のいずれかに記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記有機溶媒が、メチルイソブチルケトンおよびメチルエチルケトンからなる群より選ばれる少なくとも1つである、請求項8に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記有機溶媒が、前記式(1)または式(2)で示される2-アミノエチルメタクリレートのケチミン化合物に対して重量比で4倍以下の量で使用される、請求項8または9に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記工程(A)は、反応容器内に反応原料の供給を開始した時点から反応原料の供給が終了する時点までの間、-10~60℃の範囲の温度に制御される、請求項1~10のいずれかに記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記工程(A)は、反応原料の供給が終了した時点から反応終了時点までの間、-10~40℃の範囲の温度に制御される、請求項1~11のいずれかに記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- さらに工程(B)を含み、前記工程(B)は、前記工程(A)で得られた2-アミノエチルメタクリレート塩酸塩を含む混合物から固体析出物を分離する工程(b1)と、前記固体析出物を溶媒により洗浄する工程(b2)と、洗浄後の固体析出物を乾燥する工程(b3)を含む、請求項1~12のいずれかに記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記工程(b2)における溶媒が、アセトン、メチルエチルケトン、メチルイソブチルケトン、テトラヒドロフランおよび1,4-ジオキサンからなる群より選ばれる少なくとも1つである、請求項13に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記工程(b2)における溶媒が、重合禁止剤を含む、請求項13または14に記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
- 前記2-アミノエチルメタクリレート塩酸塩が固体である、請求項1~15のいずれかに記載の2-アミノエチルメタクリレート塩酸塩の製造方法。
