Vは、単結合、メチレン又は-C(CH3)2O-を示し、
R3は、置換基を有していてもよいC3-6アルキル基、架橋環式炭化水素基、縮合多環式炭化水素基、置換基を有していてもよいアリール基又は置換基を有していてもよい複素環式基を示し、
R4及びR5は同一又は異なって、水素原子、ハロゲン原子、シアノ基、置換基を有していてもよいC1-6アルキル基、置換基を有していてもよいC1-6アルコキシ基を示すか、又はR4とR5は一緒になって置換基を有していてもよい非芳香族複素環を形成してもよく、
R2は、アシル基、置換基を有していてもよいC1-6アルキル基、置換基を有していてもよいアリール基又は置換基を有していてもよい複素環式基を示し、
Xは、水素原子、酸素原子、ヒドロキシル基、メチル基又はメチレン基を示し、
Aは、-NR6-、-NHCONH-、-O-CH2-又は-S-CH2-を示し、
R6は、水素原子、置換基を有していてもよいC1-6アルキル基を示すか、又はR6とR2が一緒になって置換基を有していてもよい非芳香族複素環を形成してもよく、
nはメチレン鎖の数を示し、0、1又は2の整数であり、点線部は単結合又は二重結合を示し、

〔4〕T型カルシウムチャネルが作用する疾患の予防治療薬製造のための、上記〔1〕記載の一般式(I)で表される化合物、その薬学的に許容される塩又はそれらの溶媒和物の使用。
〔5〕T型カルシウムチャネルが作用する疾患を予防又は治療するための、上記〔1〕記載の一般式(I)で表される化合物、その薬学的に許容される塩又はそれらの溶媒和物。
〔6〕上記〔1〕記載の一般式(I)で表される化合物、その薬学的に許容される塩の有効量を投与することを特徴とする、T型カルシウムチャネルが作用する疾患の予防又は治療方法。
一般式(I)において、R2、R3で示される置換基を有していてもよい複素環式基のうち、置換基を有していてもよいヘテロアリール基の「ヘテロアリール基」として好ましくは、ピリジル基、ピラゾリル基、オキサゾリル基、チアゾリル基、チエニル基、インダゾリル基、キノリル基、インドリル基、ベンゾフラニル基であり、より好ましくは、ピラゾリル基、チエニル基、インドリル基、ベンゾフラニル基である。
一般式(I)において、R2、R3で示される置換基を有していてもよい複素環式基のうち、置換基を有していてもよい非芳香族複素環式基の「非芳香族複素環」として好ましくは、ピロリジン環、ピペリジン環、ヘキサメチレンイミン環、オキセタン環、オキサアダマンタン環であり、より好ましくは、ピペリジン環、オキサアダマンタン環である。
R4とR5が一緒になって置換基を有していてもよい非芳香族複素環を形成している場合、当該「非芳香族複素環」として好ましくは、ジオキソラン環、ジオキサン環、テトラヒドロフラン環であり、より好ましくはジオキサン環である。R6とR2が一緒になって置換基を有していてもよい非芳香族複素環を形成している場合、当該「非芳香族複素環」として好ましくは、ピロリジン環、ピペリジン環、モルホリン環であり、より好ましくはモルホリン環である。
<1>R1としては、-NH(C=O)-V-R3、-(C=O)NH-V-R3又は下式:
Vとしては、単結合又はメチレンが好ましく、単結合がより好ましい。R3としては、C3-6アルキル基、アダマンチル基、ノルアダマンチル基、置換基を有していてもよいアリール基、置換基を有していてもよいヘテロアリール基(ここで、アリール基又はヘテロアリール基上の置換基としては、直鎖状又は分岐鎖状のC1-6アルキル基、C3-6環状アルキル基、ハロゲン原子、シアノ基、ヒドロキシ基及びC1-6アルコキシ基から選ばれる1~3個が好ましい)又はオキサアダマンチル基が好ましく、アダマンチル基、フェニル基、ベンゾフラニル基、インドリル基、ピラゾリル基、オキサゾリル基、チエニル基、チアゾリル基(ここで、フェニル基又は前記ヘテロアリール基上の置換基としては、直鎖状又は分岐鎖状のC1-6アルキル基、C3-6環状アルキル基、ハロゲン原子、シアノ基、ヒドロキシ基及びC1-6アルコキシ基から選ばれる1~3個が好ましい)又はオキサアダマンチル基がより好ましく、アダマンチル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたフェニル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたベンゾフラニル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたインドリル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたピラゾリル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたオキサゾリル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メチル基で置換されたチエニル基、メトキシ基又はシアノ基で置換されたチアゾリル基、オキサアダマンチル基がさらに好ましい。

化合物(3)は、化合物(2)の還元反応によって製造できる。還元反応は、例えば、金属触媒の存在下、水素雰囲気下において不活性溶媒中で反応させることによって行うことができる。金属触媒としては、パラジウム、水酸化パラジウム等のパラジウム触媒、ロジウム、ウィルキンソン触媒等のロジウム触媒、クラブトリー触媒等のイリジウム触媒、ルテニウム、野依触媒等のルテニウム触媒又は白金、酸化白金等の白金触媒等の単体あるいは炭素、炭化水素、金属等への担持物が使用できる。不活性溶媒としては、メタノール、エタノール、2-プロパノール等のアルコール類、酢酸エチル、酢酸ブチル等のエステル類、テトラヒドロフラン(THF)、1,4-ジオキサン等のエーテル類、ベンゼン、トルエン等の芳香族炭化水素類、酢酸等の有機酸類、塩酸等の無機酸類、水又はこれらの混合物が使用できる。反応温度は室温~加熱還流の範囲、反応時間は0.5~168時間が好ましい。
化合物(4)は、化合物(3)のトロパン環窒素原子の保護基の脱保護によって製造できる。脱保護は、一般に有機合成化学の分野において公知の方法、例えば、T.W.Greene及びP.G.Wuts著、「Greene's Protective Groups in Organic Synthesis(Fourth Edition)」(John Wiley & Sons、Inc.)に記載の方法から適宜選択して用いることができる。例えば、保護基Pがtert-ブトキシカルボニル(Boc)基である場合、化合物(3)を酸触媒の存在下又は非存在下、不活性溶媒の存在又は非存在下、反応させることによって行うことができる。酸触媒としては、トリフルオロ酢酸、酢酸等のカルボン酸類、塩酸、硫酸等の無機酸、塩化水素不活性溶媒溶液等が使用できる。不活性溶媒としては、エーテル類、エステル類、アルコール類、ジクロロメタン、クロロホルム等のハロゲン化炭化水素類、アセトニトリル、プロピオニトリル、ベンゾニトリル等のニトリル類、芳香族炭化水素類、水又はこれらの混合物が使用できる。反応温度は0℃~加熱還流の範囲、反応時間は0.5~24時間が好ましい。
化合物(5)は、化合物(4)のアルキル化反応によって製造できる。アルキル化反応は、例えば、化合物(4)を、塩基及び/又は添加剤の存在下又は非存在下、不活性溶媒中でハロゲン化アルキル化合物または置換エポキシド化合物と反応させることによって行うことができる。塩基としては、1,8-ジアザビシクロ[5,4,0]-7-ウンデセン(DBU)等のアミジン類、トリエチルアミン、N-エチルジイソプロピルアミン等の第三級有機アミン、金属水素化物、金属アルコキシド塩、アルカリ金属炭酸塩、アルカリ金属炭酸水素塩又はアルカリ金属水酸化物が使用できる。添加剤としては、アルカリ金属ヨウ化物、ヨウ化テトラブチルアンモニウム等の四級アンモニウム塩、クラウンエーテル等の相間移動触媒が使用できる。不活性溶媒としては、エーテル類、エステル類、ハロゲン化炭化水素類、ニトリル類、芳香族炭化水素類、ケトン類、水又はこれらの混合物が使用できる。反応温度は0℃~加熱還流の範囲、反応時間は0.5~168時間が好ましい。
化合物(6)は、化合物(5)の加水分解反応によって製造できる。加水分解反応は、通常の条件で行えばよく、例えば、化合物(5)を水酸化ナトリウム、水酸化カリウム等のアルカリ金属水酸化物の存在下、不活性溶媒中で反応させることによって行うことができる。不活性溶媒としては、アルコール類、エーテル類、アセトン、ジエチルケトン等のケトン類、水又はこれらの混合物が使用できる。反応温度は0℃~加熱還流の範囲、反応時間は15分間~168時間が好ましい。
化合物(Ia)は、化合物(6)のカルボキシル基をアミド構造へと変換する公知の方法、例えば、アミド縮合反応によって製造できる。アミド縮合反応は、例えば、化合物(6)とアミン化合物を縮合剤の存在下、及び塩基の存在下又は非存在下、不活性溶媒中で反応させることによって製造できる。また、本反応は、1-ヒドロキシベンゾトリアゾ-ル(HOBt)、1-ヒドロキシ-7-アザベンゾトリアゾール(HOAt)等の縮合補助剤を加えて行ってもよい。縮合剤としては、例えば、ジシクロヘキシルカルボジイミド(DCC)、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド塩酸塩(EDC・HCl)、ジイソプロピルカルボジイミド(DIPC),1-エチル-3-(3-ジメチルアミノプロピル)カルボニウム・ヘキサフルオロリン酸塩(BOP)、(ベンゾトリアゾール-1-イルオキシ)トリスピロリジノホスホニウムヘキサフルオロリン酸塩(PyBOP)、ジフェニルホスホリルアジド(DPPA)、カルボニルジイミダゾ-ル(CDI)、(1-シアノ-2-エトキシ-2-オキソエチリデンアミノオキシ)ジメチルアミノ-モルホリノカルベニウムヘキサフルオロリン酸塩(COMU)、2-(1H-7-アザベンゾトリアゾル-1-イル)-1,1,3,3-テトラメチルウロニウムヘキサフルオロリン酸塩(HATU)等が使用できる。不活性溶媒としては、反応が進行する限り特に限定されないが、例えば、ハロゲン化炭化水素類、ニトリル類、エーテル類、エステル類、N,N-ジメチルホルムアミド(DMF)、N,N-ジメチルアセトアミド(DMA)、水又はこれらの混合物が使用できる。塩基としては、ピリジン、4-ジメチルアミノピリジン、第三級有機アミン等が使用できる。反応温度は0℃~加熱還流の範囲、反応時間は15分間~168時間が好ましい。
なお、縮合させるアミン化合物は市販されているか、公知の方法で製造することができる。
化合物(7)は、化合物(3)の加水分解反応によって製造できる。加水分解反応は、工程4と同様の方法で行うことができる。
化合物(8)は、化合物(7)のアミド縮合反応によって製造できる。アミド縮合反応は、工程5と同様の方法で行うことができる。
化合物(9)は、化合物(8)のトロパン環窒素原子の保護基の脱保護によって製造できる。脱保護は、工程2と同様の方法で行うことができる。
化合物(Ia)は、化合物(9)のアルキル化反応によって製造できる。アルキル化反応は、工程3と同様の方法で行うことができる。
化合物(11)は、化合物(10)のアルキル化反応によって製造できる。本工程は、工程3と同様の方法で行うことができる。
化合物(12)は、化合物(11)のアミノ基の保護基の脱保護によって製造できる。アミノ基の脱保護は、工程2と同様の方法で行うことができる。
化合物(Ib)は、化合物(12)のアミノ基をアミド構造へと変換する公知の方法、例えば、アミド縮合反応によって製造できる。アミド縮合反応は、工程5において、用いるアミン化合物を対応するカルボン酸化合物またはカルボン酸塩化物(ただし、本試薬を用いる場合、縮合剤は必要ない)に置き換えることで、同様の方法で行うことができる。
なお、カルボン酸化合物及びカルボン酸塩化物は、市販されているか、公知の方法で製造することができる。
化合物(14)は、化合物(13)のアミド縮合反応によって製造できる。本工程は、工程12と同様の方法で行うことができる。
化合物(15)は、化合物(14)の脱ベンジル化によって製造できる。本工程は、工程2と同様の方法で行うことができる。
化合物(Ib)は、化合物(15)のアルキル化反応によって製造できる。本工程は、工程3と同様の方法で行うことができる。
化合物(17)は、化合物(16)の脱メチル化反応によって製造できる。本工程は、工程2と同様の方法で行うことができる。

化合物(20)は、化合物(17)と保護基導入剤を塩基の存在下又は非存在下、不活性溶媒中で反応させることによって製造できる。保護基導入剤としては、例えば、二炭酸ジ-tert-ブチル(Boc2O)又はクロロギ酸ベンジル(Cbz-Cl)等であり、T.W.Greene及びP.G.Wuts著、「Greene's Protective Groups in Organic Synthesis(Fourth Edition)」(John Wiley & Sons、Inc.)に記載の方法から適宜選択して用いることができる。例えば、保護基PとしてBoc基を導入する場合、塩基としては、ピリジン、トリエチルアミン等の有機アミン、アルカリ金属水酸化物、アルカリ金属炭酸塩が使用できる。不活性溶媒としては、エーテル類、ニトリル類、DMF、アルコール類、DMSO、ジクロロメタン、1,2-ジクロロエタン等のハロゲン化炭化水素類、水又はこれらの混合物が使用できる。反応温度は0℃~加熱還流の範囲、反応時間は0.5~72時間が好ましい。
化合物(21)は、化合物(20)の加水分解反応によって製造できる。加水分解反応は、工程4と同様の方法で行うことができる。
化合物(22)は、化合物(21)のアミド縮合反応によって製造できる。本工程は、工程5と同様の方法で行うことができる。
化合物(23)は、化合物(22)のトロパン環窒素原子の保護基の脱保護によって製造できる。本工程は、工程2と同様の方法で行うことができる。
化合物(Ic)は、化合物(23)のアルキル化反応によって製造できる。本工程は、工程3と同様の方法で行うことができる。
化合物(24)は、化合物(19)または化合物(21)のクルチウス転位反応によって製造できる。クルチウス転位反応は、例えば、化合物(19)または化合物(21)を活性化剤、塩基及び不活性溶媒の存在下で活性化した後、アジド基導入剤と反応させ酸アジドを得、イソシアネートへの変換後、不活性溶媒の存在下または非存在下、対応するアルコールで処理することによって行うことができる。活性化剤としては、塩化チオニル、塩化オキサリル又はクロロギ酸エチル等が使用できる。塩基としては、トリエチルアミン、N-エチルジイソプロピルアミン等の有機アミンが使用できる。アジド基導入剤としては、ナトリウムアジド、トリメチルシリルアジド、ジフェニルホスホリルアジド(ただし、本試薬を用いる場合は活性化の段階が必要ない)等が使用できる。溶媒としては、エーテル類、DMF、ハロゲン化炭素類、ニトリル類、芳香族炭化水素類、水又はこれらの混合物が使用でき、対応するアルコールとしては、例えば、メタノール、エタノール、tert-ブチルアルコール、ベンジルアルコール等が使用できる。反応温度は0℃~加熱還流の範囲、反応時間は1~168時間が好ましい。
化合物(25)は、化合物(24)のアミノ基の保護基の脱保護によって製造できる。本工程は、工程2と同様の方法で行うことができる。
化合物(Id)または化合物(26)は、化合物(25)のアミド縮合反応によって製造できる。本工程は、工程12と同様の方法で行うことができる。
R8がPである場合、化合物(27)は、化合物(26)のトロパン環窒素原子の保護基の脱保護によって製造できる。本工程は、工程2と同様の方法で行うことができる。
化合物(Id)は、化合物(27)のアルキル化反応によって製造できる。アルキル化反応は、工程3と同様の方法で行うことができる。
化合物(Ie)または化合物(29)は、化合物(6)または化合物(7)または化合物(19)または化合物(21)と化合物(28)のアミド縮合反応と、それに続く環化反応によって製造できる。
アミド縮合反応は、工程5と同様の方法で行うことができる。
続く環化反応は、例えば、酸触媒の存在下、不活性溶媒の存在下又は非存在下で反応させることによって行うことができる。反応は、加熱撹拌あるいはマイクロウエ-ブを照射する方法等により行われる。酸触媒としては、酢酸、ギ酸、トリフルオロ酢酸等のカルボン酸類、p-トルエンスルホン酸等のスルホン酸類又は塩酸、硫酸等の無機酸類が使用できる。不活性溶媒としては、トルエン、キシレン等の芳香族炭化水素、エーテル類、塩化炭化水素類、DMF、水又はこれらの混合物が使用できる。反応温度は40~200℃の範囲、反応時間は0.5~48時間が好ましい。
R8がPである場合、化合物(30)は、化合物(29)のトロパン環窒素原子の保護基の脱保護によって製造できる。本工程は、工程2と同様の方法で行うことができる。
化合物(Ie)は、化合物(30)のアルキル化反応によって製造できる。アルキル化反応は、工程3と同様の方法で行うことができる。
経口投与用製剤としては、例えば錠剤(糖衣錠、フィルムコーティング錠を含む)、丸剤、顆粒剤、散剤、カプセル剤(ソフトカプセル剤を含む)、シロップ剤、乳剤、懸濁剤等が挙げられる。この経口投与用製剤は製剤分野において通常用いられる添加剤を配合し、公知の方法に従って製造することができる。このような添加剤としては、例えば乳糖、マンニトール、無水リン酸水素カルシウム等の賦形剤;ヒドロキシプロピルセルロース、メチルセルロース、ポリビニルピロリドン等の結合剤;でんぷん、カルボキシメチルセルロース等の崩壊剤、ステアリン酸マグネシウム、タルク等の滑沢剤等が挙げられる。
非経口的には、注射剤、直腸投与製剤、局所投与剤等として投与することができ、なかでも注射剤が好ましい。注射剤としては、例えば無菌の溶液又は懸濁液等が挙げられる。これらの注射剤は、例えば本発明化合物又はその薬学的に許容しうる塩を日局注射用水に溶解又は懸濁することにより製造される。必要により塩化ナトリウム等の等張化剤;リン酸二水素ナトリウム、リン酸一水素ナトリウム等の緩衝剤;溶解補助剤等を配合してもよい。また、用時溶解型(粉末充填、凍結乾燥)の注射剤とすることができ、この場合、マンニトール、乳糖などの賦形剤を添加して、通常の方法で製造することができる。
局所投与製剤は液剤、点眼剤、クリーム、軟膏、ゲル製剤、スプレー剤、粉剤等が挙げられる。液剤は、本発明化合物又はその薬学的に許容しうる塩を水に加え、安定化剤、溶解剤、増粘剤、分散剤、懸濁化剤等を必要に応じて加えて製造することができる。この増粘剤としては、ゼラチン、ヒアルロン酸ナトリウム、高分子デキストラン、アルギン酸ナトリウム、コンドロイチン硫酸ナトリウムなどを用いることができる。点眼剤は、緩衝剤、pH調整剤、等張化剤のほかに防腐剤を加えて製造することができる。クリーム及び軟膏は、水性又は油性の基剤、例えば水、流動パラフィン、植物油(ピーナッツ油、ひまし油等)、マクロゴールなどを用いて製造することができる。ゲル製剤は、公知の方法により、ゼラチン、ペクチン、カラゲナン、寒天、トラガント、アルギン酸塩、セルロースエーテル(メチルセルロース、ナトリウムカルボキシメチルセルロースなど)、ペクチン誘導体、ポリアクリレート、ポリメタクリレート、ポリビニルアルコールおよびポリビニルピロリドンなどを用いて製造することができる。スプレー剤は本発明化合物又はその薬学的に許容しうる塩を水などに溶解又は懸濁した後、スプレー容器に入れて製造することができる。粉剤とする場合は、本発明化合物又はその薬学的に許容しうる塩をそのまま使用することもできるが、適当な賦形剤と混合して製造することができる。
[(エキソ)-8-tert-ブトキシカルボニル-8-アザビシクロ[3.2.1]オクタン-3-イル]酢酸エチル
3-エトキシカルボニルメチレン-8-アザビシクロ[3.2.1]オクタン-8-カルボン酸tert-ブチル(WO2007/079239)(50.3g)をエタノール/水(7:2)(567mL)に溶解し、5%ロジウムアルミナ粉末(12.6g)を加え、水素雰囲気下、室温で27時間撹拌した。不溶物をセライトで濾過し、濾液を減圧留去した後、飽和食塩水を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムで乾燥した後、溶媒を減圧留去し、表題化合物(51.7g)を粗生成物として得た。

参考例3の化合物(2.14g)を1,4-ジオキサン(以下、ジオキサン)(37mL)に溶解し、2-クロロ-N-tert-ブチルアセトアミド(1.37g)、DBU(3.01mL)を加え、50℃加温下で17時間撹拌した。反応溶媒を減圧留去した後、水を加え、酢酸エチルで抽出した。有機層を1mol/L塩酸で抽出し、水層を1mol/L水酸化ナトリウム水溶液でpH9~10に調整し、酢酸エチルで再度抽出した。有機層を飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥し、溶媒を減圧留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(1~10% メタノール/クロロホルム)にて精製し、表題化合物(2.45g)を得た。

得られた粗生成物(333mg)を酢酸(3mL)に溶解し、90℃加温下で2時間撹拌した。反応溶媒を減圧留去し、得られた残渣をシリカゲルカラムクロマトグラフィー(0~12%メタノール/クロロホルム)にて精製し、表題化合物(302mg)を得た。

参考例17の化合物(2.10g)をメタノール(40mL)に溶解し、1mol/L水酸化ナトリウム水溶液(20mL)を加え、70℃加温下で2時間撹拌した。メタノールを減圧留去し、水層をジエチルエーテルで洗浄した後、1mol/L塩酸を加えて酸性とし、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムで乾燥し、溶媒を減圧留去し、表題化合物(2.00g)を得た。

ヒトCav3.2が安定発現したHEK293細胞は37℃で10%(v/v)ウシ胎仔血清(FBS)、ペニシリン(100U/mL)、ストレプトマイシン(100μg/mL)、G418(250μg/mL)を加えたAlpha-MEMにて培養した。細胞は培養液で懸濁し、96穴プレ-トに播種した後、48時間培養した。培養液を除き、5%(v/v)FBS、塩化カルシウム(0.5mmol/L)、L-グルタミン(2mmol/L)、L-アラニン(8.9ng/mL)、L-アスパラギン(13.2ng/mL)、L-アスパラギン酸(1.33ng/mL)、L-グルタミン酸(14.7ng/mL)、グリシン(7.5ng/mL)、L-プロリン(11.5ng/mL)、L-セリン(10.5ng/mL)、ペニシリン(100U/mL)、ストレプトマイシン(100μg/mL)を加えたS-MEMに交換し、さらに24時間培養した。再び培養液を除き、37℃に保温したアッセイバッファー(140mmol/L塩化ナトリウム、5mmol/L塩化カリウム、0.5mmol/L塩化マグネシウム、0.5mmol/L塩化カルシウム、10mmol/Lグルコ-ス、0.4mmol/L硫酸マグネシウム、10mmol/L HEPES、250μmol/Lスルフィンピラゾン、pH7.4)にて洗浄した後、蛍光Ca2+指示薬であるFura2-AMを5μMに溶解させたアッセイバッファーを加え、37℃で30分間静置した。Fura2が溶解しているアッセイバッファーを除き、アッセイバッファーにて洗浄した後、試験化合物を添加したアッセイバッファーを加え、15分間静置した。蛍光測定装置(FLex Station II、Molecular Devices社)にプレートを設置し、20秒間ベースラインを測定した後、100mmol/L塩化カルシウムを加えたアッセイバッファーを添加した際に誘起される細胞内カルシウム濃度の変化を測定し(340nm、380nm励起、510nm検出)、各波長より得られる蛍光強度比を算出した。

HC:コントロール溶液処理後のCa2+含有アッセイバッファー添加後25~30秒間(測定開始後45~50秒間)のRFU値の平均
LC:コントロール溶液処理後のCa2+非含有アッセイバッファー添加後25~30秒間(測定開始後45~50秒間)のRFU値の平均式中のRFUは相対的蛍光強度(Relative Fluorescence Unit)を表す。
本発明化合物の遺伝毒性の有無について、細菌を用いる復帰突然変異試験(Ames試験)により検討した。試験は、ネズミチフス菌(Salmonella typhimurium)TA98及びTA100の2菌株を使用し、代謝活性化系の非存在下(S9mix(-))および存在下(S9mix(+))について、プレインキュベーション法により実施した。化合物はジメチルスルホキシド(DMSO)に溶解し、試験液を調製した。陰性対照には溶媒であるDMSOを用いた。陽性対照には、S9mix(-)では2-(2-フリル)-3-(5-ニトロ-2-フリル)アクリルアミド(AF-2)及びS9mix(+)では2-アミノアントラセン(2-AA)を用いた。
滅菌した試験管に、試験液を0.1mL入れ、次いでS9mix(-)の場合は0.1mol/Lナトリウム・リン酸緩衝液(PH7.4)を0.5mL、S9mix(+)の場合はラット肝S9mixを0.5mL添加した。さらに、前培養した試験菌株懸濁液0.1mLを添加した後、37℃で20分間振盪培養した(プレインキュベーション)。プレインキュベーション終了後、トップアガー(ヒスチジン及びビオチンをそれぞれ0.05mMの濃度で含有する軟寒天)を2mL添加し、泡をたてないように混合した。その後、試験管の内容物を最少グルコース寒天培地プレート上に重層し、一様に拡げた。プレートは恒温器で37℃、48時間培養した。培養終了後、プレート上に生育した復帰突然変異コロニー数をコロニーカウンターを用いて計測した。判定は、復帰突然変異コロニー数が陰性対照のコロニー数の2倍以上に増加し、かつ濃度依存性が認められた場合を陽性とした。
N-tert-ブチル-2-[4-(4-メチル-1H-ベンズイミダゾール-2-イル)メチルピペリジン-1-イル]アセトアミド(対照化合物1)
[1-(tert-ブチルカルバモイル)メチルピペリジン-4-イル]酢酸塩酸塩(1.50g)をアセトニトリル(20mL)に懸濁し、2,3-ジアミノトルエン(657mg)、PyBOP(2.93g)を加えた後、トリエチルアミン(3.03mL)を加え、室温で5時間撹拌した。反応溶媒を減圧留去し、水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄した後、無水硫酸ナトリウムで乾燥し、溶媒を減圧留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(0~10% メタノール/酢酸エチル)にて精製し、粗生成物を得た。
得られた粗生成物を酢酸(14mL)に溶解し、80℃加温下で4時間撹拌した。反応溶媒を減圧留去し、1mol/L塩酸を加え、クロロホルムで洗浄した後、水層を2mol/L水酸化ナトリウム水溶液でpH10~11に調整し、クロロホルムで抽出した。有機層を飽和食塩水で洗浄した後、無水硫酸ナトリウムで乾燥し、溶媒を減圧留去した。得られた残渣をシリカゲルカラムクロマトグラフィー(0~11%メタノール/クロロホルム)にて精製し、表題化合物(1.25g)を得た。
1H-NMR(CDCl3)δ:7.37(1H,d,J=7.9Hz),7.16-6.98(3H,m),2.86-2.70(6H,m),2.57(3H,s),2.12-1.98(2H,m),1.94-1.76(1H,m),1.73-1.58(2H,m),1.40-1.19(2H,m),1.35(9H,s).
ESI+APCI-MS Found:m/z 343(M+H)+
N-tert-ブチル-2-[4-(4-メトキシ-1H-ベンズイミダゾール-2-イル)メチルピペリジン-1-イル]アセトアミド(対照化合物2)
対応する原料を用いて、参考例31と同様の方法により表題化合物を得た。
1H-NMR(CDCl3)δ:7.23-7.00(3H,m),6.72-6.63(1H,m),3.96(3H,s),2.84(2H,s),2.84-2.72(4H,m),2.14-2.02(2H,m),1.98-1.78(1H,m),1.76-1.64(2H,m),1.42-1.24(2H,m),1.35(9H,s).
ESI+APCI-MS Found:m/z 359(M+H)+
N-tert-ブチル-2-[4-(5,6-ジヒドロ-1H-[1,4]ジオキシノ[2,3-e]ベンズイミダゾール-2-イル)メチルピペリジン-1-イル]アセトアミド二塩酸塩(対照化合物3)
[1-(tert-ブチルカルバモイル)メチルピペリジン-4-イル]酢酸塩酸塩(5.10g)と2,3-ジヒドロベンゾ[1,4]ジオキシン-5,6-ジアミン二塩酸塩(4.37g)を用いて、実施例149と同様の方法により、表題化合物(3.90g)を得た。
1H-NMR(DMSO-d6)δ:9.62(1H,brs),8.24(1H,brs),7.15(1H,d,J=9.2Hz),6.98(1H,d,J=9.2Hz),4.46-4.30(4H,m),3.76(2H,brs),3.43-2.88(6H,m),2.20-1.98(1H,m),1.85-1.48(4H,m),1.27(9H,s).

Claims (15)
- 下記の一般式(I)
Vは、単結合、メチレン又は-C(CH3)2O-を示し、
R3は、置換基を有していてもよいC3-6アルキル基、架橋環式炭化水素基、縮合多環式炭化水素基、置換基を有していてもよいアリール基又は置換基を有していてもよい複素環式基を示し、
R4及びR5は同一又は異なって、水素原子、ハロゲン原子、シアノ基、置換基を有していてもよいC1-6アルキル基、置換基を有していてもよいC1-6アルコキシ基を示すか、又はR4とR5は一緒になって置換基を有していてもよい非芳香族複素環を形成してもよく、
R2は、アシル基、置換基を有していてもよいC1-6アルキル基、置換基を有していてもよいアリール基又は置換基を有していてもよい複素環式基を示し、
Xは、水素原子、酸素原子、ヒドロキシル基、メチル基又はメチレン基を示し、
Aは、-NR6-、-NHCONH-、-O-CH2-又は-S-CH2-を示し、
R6は、水素原子又は置換基を有していてもよいC1-6アルキル基を示すか、又はR6とR2が一緒になって置換基を有していてもよい非芳香族複素環を形成してもよく、
nはメチレン鎖の数を示し、0、1又は2の整数であり、点線部は単結合又は二重結合を示し、

- R3が、C3-6アルキル基、アダマンチル基、ノルアダマンチル基、置換基を有していてもよいアリール基又は置換基を有していてもよいヘテロアリール基(ここで、アリール基又はヘテロアリール基上の置換基は、直鎖状又は分岐鎖状のC1-6アルキル基、C3-6環状アルキル基、ハロゲン原子、シアノ基、ヒドロキシ基及びC1-6アルコキシ基から選ばれる1~3個である)である請求項1~3のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- R3が、アダマンチル基、フェニル基、ベンゾフラニル基、インドリル基、ピラゾリル基、オキサゾリル基又はチアゾリル基(ここで、フェニル基又は前記ヘテロアリール基上の置換基は、直鎖状又は分岐鎖状のC1-6アルキル基、C3-6環状アルキル基、ハロゲン原子、シアノ基、ヒドロキシ基及びC1-6アルコキシ基から選ばれる1~3個である)である請求項1~4のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- R3が、アダマンチル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたフェニル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたベンゾフラニル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたインドリル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたピラゾリル基、無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたオキサゾリル基又は無置換、若しくは1~2個のハロゲン原子、メチル基、エチル基、シクロプロピル基、メトキシ基又はシアノ基で置換されたチアゾリル基である請求項1~5のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- R2が、ハロゲン原子、シアノ基、ヒドロキシ基及びC1-6アルコキシ基から選ばれる1~3個が置換していてもよい直鎖状又は分岐鎖状C1-6アルキル基又はハロゲン原子、シアノ基、ヒドロキシ基及びC1-6アルコキシ基から選ばれる1~3個が置換していてもよい環状C3-6アルキル基である請求項1~6のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- Xが酸素原子である請求項1~7のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- Aが、-NR6-であり、R6が水素原子又はメチル基である請求項1~8のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- nが1である請求項1~9のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- 請求項1~10のいずれか1項記載の化合物、その薬学的に許容される塩又はそれらの溶媒和物を含有する医薬。
- 請求項1~10のいずれか1項記載の一般式(I)で表される化合物、その薬学的に許容される塩又はそれらの溶媒和物、及び薬学的に許容される担体を含有する医薬組成物。
- T型カルシウムチャネルが作用する疾患の予防治療薬製造のための、請求項1~10のいずれか1項記載の一般式(I)で表される化合物、その薬学的に許容される塩又はそれらの溶媒和物の使用。
- T型カルシウムチャネルが作用する疾患を予防又は治療するための、請求項1~10のいずれか1項記載の一般式(I)で表される化合物、その薬学的に許容される塩又はそれらの溶媒和物。
- 請求項1~10のいずれか1項記載の一般式(I)で表される化合物、その薬学的に許容される塩の有効量を投与することを特徴とする、T型カルシウムチャネルが作用する疾患の予防又は治療方法。
