・「サイトップM」…主鎖にパーフルオロテトラヒドロフラン環を含む繰り返し単位を有し、末端に式:-CONH~Si(OEt)3で表される部分を含む置換基を有するポリマー(サイトップM、旭硝子(株)製)
(1)シランカップリング剤による前処理
シランカップリング剤(KBE-903、3-アミノプロピルトリエトキシシラン)を精製水で希釈して、シランカップリング剤濃度が0.5重量%の溶液を調製した。この溶液を、バイアル(内容量20mL、硼珪酸ガラス製、以下同じ)の内表面に塗布し、遠心処理して液切りした後、100℃で30分焼き付け処理し、シランカップリング剤で処理されたバイアルを製造した。
(2)フッ素樹脂コーティング
非晶性フッ素系樹脂(テフロンAF)を、フッ素系不活性液体(フロリナート FC-40、住友スリーエム(株)製)で希釈し、非晶性フッ素系樹脂濃度4%の溶液を調製した。
非晶性フッ素系樹脂(サイトップM)を、フッ素系不活性液体(フロリナート FC-40、住友スリーエム(株)製)で希釈し、非晶性フッ素系樹脂濃度4%の溶液を調製した。
(1)シランカップリング剤による前処理
シランカップリング剤(KBMー13、メチルトリメトキシシラン)を精製水で希釈し、これに酢酸を加えてpH4に調整して、シランカップリング剤濃度が2.0重量%の溶液を調製した。この溶液を、バイアルの内表面に塗布し、遠心処理して液切りした後、200℃で30分焼き付け処理し、シランカップリング剤で処理されたバイアルを製造した。
(2)フッ素樹脂コーティング
非晶性フッ素系樹脂(テフロンAF)を、フッ素系不活性液体(フロリナート FC-40、住友スリーエム(株)製)で希釈し、非晶性フッ素系樹脂濃度1%の溶液を調製した。
非晶性フッ素系樹脂(サイトップM)を、フッ素系不活性液体(フロリナート FC-40、住友スリーエム(株)製)で希釈し、非晶性フッ素系樹脂濃度1%の溶液を調製した。
バイアルを、シランカップリング剤の前処理(実施例1(1))をすることなく、直接、実施例1(2)に従い非晶性フッ素系樹脂(テフロンAF)で処理し、内面処理されたバイアルを製造した。
シリコンエマルジョン(KM-740、ジメチルポリシロキサン濃度35%、信越化学(株))を精製水で希釈し、ジメチルポリシロキサン濃度1%の溶液を調製した。
未処理のバイアル(内容量20mL、硼珪酸ガラス)を用いた。
実施例及び比較例で表面処理されたバイアル(以下、コートバイアルと表記する)を、以下の試験に供した。試験結果を表1~3に示す。
(1)外観評価
検査灯下でバイアル内表面を肉眼観察して評価した。
○:コーティングムラ及び不透明な部分が僅かに認められるが問題なし
×:かなりのコーティングムラがあり不透明である(白化あり)
(2)撥水性評価
コートバイアルに精製水を充填し、バイアル内表面の撥水性を肉眼で評価した。
○:バイアル内表面の撥水がほぼ均一である
×:バイアル内表面の撥水にムラがある
(3)撥粉性評価
コートバイアルに微粉末(混合ビタミン微粉末;粒径約1~30μm)を少量充填後、バイアル内表面の微粉末付着を肉眼で評価した。
○:バイアル内表面に粉が僅かに付着している
×:バイアル内表面に粉が多量に付着する
(4)乾熱耐久性評価
コートバイアルを250℃×30分間乾熱加熱試験を行った。試験後の膜の剥離及び撥水性の有無を評価した。
○:膜の剥離及び/又は撥水性不良が僅かに認められるが問題なし
×:膜の剥離及び撥水不良あり
(5)耐超音波洗浄(耐US)評価
コートバイアルを精製水中で25℃×40秒間超音波処理(28KHzの洗浄機を使用)の条件で超音波処理を行った。試験後の膜の剥離及び撥水性の有無を評価した。
○:不溶性微粒子の増加が僅かに認められるが問題なし
×:不溶性微粒子の増加が認められる
(6)耐熱水性評価
コートバイアルに、精製水を充填し121℃×60分間加熱した。試験後の膜の剥離及び撥水性の有無を評価した。
○:膜の剥離及び/又は撥水性不良が僅かに認められるが問題なし
×:膜の剥離及び撥水不良あり
(7)耐アルカリ性評価
精製水に0.05mol/Lの水酸化ナトリウム水溶液を加えてpH9に調整したものをコートバイアルに充填し121℃×60分間加熱した。試験後の膜の剥離及び撥水性の有無を評価した。
○:膜の剥離及び/又は撥水性不良が僅かに認められるが問題なし
×:膜の剥離及び撥水不良あり
(8)ガラスからの溶出金属濃度評価
コートバイアルに、精製水、緩衝液(フタル酸塩緩衝液、リン酸塩緩衝液及びホウ酸塩緩衝液)を充填し121℃×60分間加熱した。バイアルの内表面から試験液中に溶出した金属イオン(Na、B、Al、Si、Ca及びBa)の濃度を測定した。Naは原子吸光分析(AAS)装置を用いて、その他の金属は誘導結合プラズマ発光分析(ICP-AES)装置を用いて測定した。その結果を表3に示す。単位はいずれもppmである。
(9)ガラスからの溶出フッ素濃度評価
コートバイアルに、精製水を充填し120℃×60分間加熱した。バイアルの内表面から水中に溶出したフッ素イオン(F-)の濃度を測定した。フッ素イオン(F-)はイオンクロマトグラフ法を用いて測定した。その結果を表4に示す。単位はいずれもppmである。
実施例1で表面処理されたバイアル(コートバイアル1)、これを試験例1における「(4)乾熱耐久性評価」に供したバイアル(コートバイアル2)、又は「(4)乾熱耐久性評価」に供したバイアルと同じ処理をしさらに121℃×20分の湿熱滅菌を施したバイアル(コートバイアル3)について、コーティング被膜の密着強度を評価した。
実施例1で表面処理されたバイアル(コートバイアル1)について、コーティング被膜の耐衝撃強度を評価した。
実施例1、比較例2及び比較例3のバイアルについて、コーティング被膜のタンパク吸着性を評価した。
これより、実施例1のバイアルでは、比較例2及び3のバイアルに比べて、タンパクの吸着量が低減されていることが確認された。
Claims (12)
- ガラス容器の製造方法であって、
(1)ガラス容器の内表面をシランカップリング剤及び/又はその部分加水分解物で処理する工程、及び
(2)工程(1)で得られた処理表面を非晶性フッ素系樹脂で処理する工程
を含むことを特徴とする製造方法。 - 前記シランカップリング剤が、一般式(A):
(R1O)4-nSiR2 n (A)
(式中、nは1、2又は3を示し、R1は低級アルキル基を示し、R2は置換基としてアミノ基又はアミノ低級アルキルアミノ基を有していてもよい低級アルキル基を示す)
で表される化合物である請求項1に記載の製造方法。 - 前記シランカップリング剤が、一般式(A)において、nが1を示し、R1がC1~3アルキル基を示し、R2が置換基としてアミノ基又はアミノC2~4アルキルアミノ基を有していてもよいC2~6アルキル基を示す、化合物である請求項2に記載の製造方法。
- 前記非晶性フッ素系樹脂が、含フッ素環状エーテル構造の繰り返し単位を有する非晶性フッ素系樹脂である請求項1~3のいずれか1項に記載の製造方法。
- 前記式(C)及び/又は(D)で表される繰り返し単位を有する非晶性フッ素系樹脂が、カルボキシル基を含む置換基、又は、式(E):
-CONH-R3-Si(OR4)3 (E)
(式中、R3はリンカーを示し、R4は低級アルキル基を示す)
で表される部分を含む置換基を有する請求項7に記載の製造方法。 - 前記式(C)及び/又は(D)で表される繰り返し単位を有する非晶性フッ素系樹脂の末端に、式(E)で表される部分を含む置換基を有する請求項8に記載の製造方法。
- 前記請求項1~9のいずれか1項に記載の製造方法により得られるガラス容器。
- 医療用ガラス容器である請求項10に記載のガラス容器。
- 前記請求項11に記載の医療用ガラス容器に医薬品又は検査用薬品を収納してなる薬品が収納された医療用ガラス容器。
