しかし、再生可能資源を原料としたバイオフェノール生産は、乳酸やエタノールの生産と比較して、原料となる糖類から代謝反応段数が大変多いために生産性が低く、また生産物であるフェノールにより菌の増殖が阻害されたり、フェノールによる細胞毒性がある等の理由により、これまで工業的生産が不可能とされていた。
これまで自然界においてフェノール生産菌は報告されていない。
また、特許文献1は、エンテロバクター クロアカエ(Enterobacter cloacae)の4-ヒドロキシベンゾエート デカルボキシラーゼ遺伝子を導入した形質転換菌を用いて4-ヒドロキシベンゾエートからフェノールを製造する技術を開示している。
しかし、非特許文献1及び特許文献1の方法は、実用上十分に効率よくフェノールを製造することができない。
(i) コリネバクテリウム グルタミカムに4-ヒドロキシベンゾエート デカルボキシラーゼ遺伝子を導入した形質転換体は、効率よく、4-ヒドロキシベンゾエートからフェノールを生産できる。
(ii) この形質転換体において、宿主のコリネバクテリウム グルタミカムの染色体上に存在するフェノール2-モノオキシゲナーゼ遺伝子が破壊又は欠損しているときは、一層効率よくフェノールを生産できる。
(iii) この形質転換体は、還元条件下の反応液中で実質的に増殖しない状態で反応させる場合、特に、フェノール生産効率が高い。

一般に微生物はフェノールのような溶剤の細胞毒性により生育が阻害されるため、微生物を用いてフェノールを製造することは困難であったが、本発明方法によれば、微生物を用いて、実用上十分に効率良くフェノールを製造することができる。
(I)フェノール生産能を有する形質転換体
本発明のフェノール生産能を有する形質転換体は、4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有する酵素をコードする遺伝子が、宿主のコリネバクテリウム グルタミカムに導入された形質転換体である。
宿主として用いられるコリネバクテリウム グルタミカムとは、バージーズ・マニュアル・デターミネイティブ・バクテリオロジー〔Bergey’s Manual of Determinative Bacteriology、Vol. 8、599(1974)〕に定義されている一群の微生物である。
具体的には、コリネバクテリウム グルタミカム(Corynebacterium glutamicum)R(FERM P-18976)、ATCC13032、ATCC13869、ATCC13058、ATCC13059、ATCC13060、ATCC13232、ATCC13286、ATCC13287、ATCC13655、ATCC13745、ATCC13746、ATCC13761、ATCC14020、ATCC31831、MJ-233(FERM BP-1497)又はMJ-233AB-41(FERM BP-1498)等の菌株が挙げられる。中でも、R(FERM P-18976)株、ATCC13032株、及びATCC13869株が好ましく、R(FERM P-18976)株がより好ましい。
なお、分子生物学的分類により、ブレビバクテリウム フラバム(Brevibacterium flavum)、ブレビバクテリウム ラクトファーメンタム(Brevibacterium lactofermentum)、ブレビバクテリウム ディバリカタム(Brevibacterium divaricatum)、コリネバクテリウム リリウム(Corynebacterium lilium)等のコリネ型細菌もコリネバクテリウム グルタミカム(Corynebacterium glutamicum)に菌名が統一されている〔Liebl, W. et al., Transfer of Brevibacterium divaricatum DSM 20297T, "Brevibacterium flavum" DSM 20411, "Brevibacterium lactofermentum" DSM 20412 and DSM 1412, and Corynebacterium glutamicum and their distinction by rRNA gene restriction patterns. Int J Syst Bacteriol. 41:255-260. (1991)、駒形和男ら, コリネフォルム細菌の分類, 発酵と工業, 45:944-963 (1987)〕ため、本発明に含まれる。
また、コリネバクテリウム グルタミカムは、野生株の他に、その変異株や人為的な遺伝子組換え体であってもよい。例えば、ラクテート(乳酸)デヒドロゲナーゼ(lactate dehydrogenase:LDH)、フォスフォエノールピルベートカルボキシラーゼ(phosphoenolpyrvate carboxylase)、マレートデヒドロゲナーゼ(malate dehydrogenase)などの遺伝子の破壊株が挙げられる。このような遺伝子破壊株を宿主として用いることにより、フェノールの生産性を向上させたり、副生成物の生成を抑制したりすることができる。
中でも、ラクテートデヒドロゲナーゼ遺伝子の破壊株が好ましい。この遺伝子破壊株は、乳酸デヒドロゲナーゼ遺伝子が破壊されていることにより、ピルビン酸から乳酸への代謝経路が遮断されている。中でも、コリネバクテリウム グルタミカムR(FERM P-18976)のラクテートデヒドロゲナーゼ遺伝子の破壊株が特に好ましい。
このような遺伝子破壊株は、遺伝子工学的手法により常法に従い作製できる。例えば、WO2005/010182A1に、乳酸デヒドロゲナーゼ破壊株、及びその作製方法が記載されている。
4-ヒドロキシベンゾエート デカルボキシラーゼは、4-ヒドロキシベンゾエートの脱炭酸によるフェノールの生成反応を触媒する酵素である。
4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有する酵素をコードする遺伝子の由来は特に限定されないが、バチラス サブチリス(Bacillus subtilis)、バチラス メガテリウム(Bacillus megaterium)、バチラス リッケニフォーミス(Bacillus licheniformis)、バチラス アトロファエウス(Bacillus atrophaeus)、バチラス サブチリス 亜種 スピジゼニイ(Bacillus subtilis subsp. spizizenii)のようなバチラス属細菌由来の遺伝子;シトロバクター コセリ(Citrobacter koseri)のようなシトロバクター属細菌由来の遺伝子;エンテロバクター アエロゲネス(Enterobacter aerogenes)、エンテロバクター クロアカエ(Enterobacter cloacae)、エンテロバクター ホルメシェイ(Enterobacter hormaechei)、エンテロバクター サカザキ(Enterobacter sakazakii)のようなエンテロバクター属細菌由来の遺伝子;エシェリヒア コリ(Escherichia coli)、エシェリヒア フェルグソニー(Escherichia fergusonii)のようなエシェリヒア属細菌由来の遺伝子;パエニバチラス ポリミキサ(Paenibacillus polymyxa)のようなパエニバチラス属細菌由来の遺伝子;パントエア アナナティス(Pantoea ananatis)のようなパントエア属細菌由来の遺伝子などが挙げられる。中でも、バチラス属細菌、特にバチラス サブチリス由来の遺伝子、エンテロバクター属細菌、特にエンテロバクター クロアカエ由来の遺伝子、エシェリヒア属細菌、特にエシェリヒア コリ由来の遺伝子が好ましい。
なお、4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有する酵素をコードする遺伝子は、由来により異なる種々の略称が用いられている。例えば、バチラス サブチリス由来の4-ヒドロキシベンゾエート デカルボキシラーゼ遺伝子はbsdBCDと略称されている。本明細書では、4-ヒドロキシベンゾエート デカルボキシラーゼ遺伝子を、由来を問わず「dca」と略称することがある。
本発明において「ストリンジェントな条件」は、一般的な条件、例えば、Molecular Cloning, A Laboratory Manual, Second Edition,1989,Vol2,p11.45に記載された条件を指す。具体的には、完全ハイブリッドの融解温度(Tm)より5~10℃低い温度でハイブリダイゼーションが起こる場合を指す。
また、本発明では、配列番号16、配列番号23、配列番号26、配列番号29、配列番号32、配列番号35、配列番号38、配列番号41、配列番号44、配列番号47、配列番号50、又は配列番号53の塩基配列と同一性が90%以上、好ましくは95%以上、より好ましくは98%以上の塩基配列からなり、かつ4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有するポリペプチドをコードするDNAも使用できる。
本発明において、塩基配列の同一性は、GENETYX ver.8(GENETYX 株式会社ゼネティックス製)により算出した値である。
PCRで増幅した4-ヒドロキシベンゾエート デカルボキシラーゼ酵素をコードするDNAは、それぞれ、宿主で増幅できる適切なベクターにクローニングすればよい。
プラスミドベクターとしては、コリネバクテリウム グルタミカム内で自律複製機能を司る遺伝子を含むものであれば良い。その具体例としては、ブレビバクテリウム ラクトファーメンタム(Brevibacterium lactofermentum)2256由来のpAM330〔特開昭58-67699〕、〔Miwa, K. et al., Cryptic plasmids in glutamic acid-producing bacteria. Agric. Biol. Chem. 48:2901-2903(1984)〕 及び 〔Yamaguchi, R. et al., Determination of the complete nucleotide sequence of the Brevibacterium lactofermentum plasmid pAM330 and the analysis of its genetic information. Nucleic Acids Symp. Ser. 16:265-267(1985)〕、コリネバクテリウム グルタミカム ATCC13058由来のpHM1519 〔Miwa, K. et al., Cryptic plasmids in glutamic acid-producing bacteria. Agric. Biol. Chem. 48:2901-2903(1984)〕及びpCRY30 〔Kurusu, Y. et al., Identification of plasmid partition function in coryneform bacteria. Appl. Environ. Microbiol. 57:759-764 (1991)〕、コリネバクテリウム グルタミカム T250由来のpCG4〔特開昭57-183799〕、〔Katsumata, R. et al., Protoplast transformation of glutamate-producing bacteria with plasmid DNA. J. Bacteriol.、159:306-311 (1984)〕、pAG1、pAG3、pAG14、pAG50〔特開昭62-166890〕、pEK0、pEC5、pEKEx1 〔Eikmanns, B.J. et al., A family of Corynebacterium glutamicum/Escherichia coli shuttle vectors for cloning, controlled gene expression, and promoter probing. Gene, 102:93-98 (1991)〕などが挙げられる。
好ましいターミネーターとしては、大腸菌rRNAオペロンのrrnB T1T2 ターミネーター、大腸菌のtrpA ターミネーター、ブレビバクテリウム ラクトファーメンタム(Brevibacterium lactofermentum)のtrp ターミネーターなどが挙げられ、中でも、rrnB T1T2 ターミネーターが好ましい。
形質転換方法は、公知の方法を制限無く使用できる。このような公知の方法として、例えば塩化カルシウム/塩化ルビジウム法、リン酸カルシウム法、DEAE-デキストラン介在トランスフェクション、電気穿孔法などが挙げられる。中でも、コリネバクテリウム グルタミカムには、電気パルス法が好適であり、電気パルス法は、公知の方法 〔Kurusu, Y. et al., Electroporation-transformation system for Coryneform bacteria by auxotrophic complementation. Agric. Biol. Chem. 54:443-447 (1990)〕 及び 〔Vertes A.A. et al., Presence of mrr- and mcr-like restriction systems in Coryneform bacteria. Res. Microbiol. 144:181-185 (1993)〕により行うことができる。
炭素源としては、グルコース、フルクトース、スクロース、マンノース、マルトース、マンニトール、キシロース、アラビノース、ガラクトース、澱粉、糖蜜、ソルビトール、グリセリン等の糖質又は糖アルコール;酢酸、クエン酵、乳酸、フマル酸、マレイン酸又はグルコン酸等の有機酸;エタノール、プロパノール等のアルコール等が挙げられる。また、所望によりノルマルパラフィン等の炭化水素等も用いることができる。炭素源は、1種を単独で使用でき、又は2種以上を混合して使用してもよい。培地中のこれら炭素源の濃度は、通常、約0.1~10(w/v%)とすればよい。
窒素源としては、塩化アンモニウム、硫酸アンモニウム、硝酸アンモニウム、酢酸アンモニウム等の無機又は有機アンモニウム化合物、尿素、アンモニア水、硝酸ナトリウム、硝酸カリウム等が挙げられる。また、コーンスティープリカー、肉エキス、ベプトン、NZ-アミン、蛋白質加水分解物、アミノ酸等の含窒素有機化合物等も使用できる。窒素源は、1種を単独で使用してもよく、また2種以上を混合して使用してもよい。培地中の窒素源濃度は、使用する窒素化合物によっても異なるが、通常、約0.1~10(w/v%)とすればよい。
無機塩類としては、例えばリン酸第一カリウム、リン酸第二カリウム、硫酸マグネシウム、塩化ナトリウム、硝酸第一鉄、硫酸マンガン、硫酸亜鉛、硫酸コバルト、又は炭酸カルシウム等が挙げられる。これら無機塩は、1種を単独で使用してもよく、また2種以上を混合して使用してもよい。培地中の無機塩類濃度は、使用する無機塩によっても異なるが、通常、約0.01~1(w/v%)とすればよい。
栄養物質としては、例えば肉エキス、ペプトン、ポリペプトン、酵母エキス、乾燥酵母、コーンスティープリカー、脱脂粉乳、脱脂大豆塩酸加水分解物、又は動植物若しくは微生物菌体のエキスやそれらの分解物等が挙げられる。栄養物質の培地濃度は、使用する栄養物質によっても異なるが、通常、約0.1~10(w/v%)とすればよい。さらに、必要に応じて、ビタミン類を添加することもできる。ビタミン類としては、例えば、ビオチン、チアミン(ビタミンB1)、ピリドキシン(ビタミンB6)、パントテン酸、イノシトール、ニコチン酸等が挙げられる。
培地のpHは約5~8が好ましい。
培養温度は約15~45℃とすればよく、培養時間は約1~7日間とすればよい。
宿主のコリネバクテリウム グルタミカムは、その染色体上に存在するフェノール2-モノオキシゲナーゼ活性を有する酵素をコードする遺伝子(poxF)が、破壊され、または欠失していることが好ましく、これにより一層効率良くフェノールを製造することができる。
また、宿主のコリネバクテリウム グルタミカムは、その染色体上に存在する4-ヒドロキシベンゾエート ヒドロキシラーゼ活性を有する酵素をコードする遺伝子(pobA)が破壊され、または欠失していることが好ましく、これにより、一層効率良くフェノールを製造することができる。
poxF及びpobAの双方が破壊され、または欠失していることが特に好ましい。
遺伝子の部分配列を欠失し、正常に機能する酵素タンパク質を産生しないように改変した欠失型遺伝子を作製し、該遺伝子を含むDNAで細菌を形質転換して、欠失型遺伝子と染色体上の遺伝子とで相同組換えを起こさせることにより、染色体上の遺伝子を欠失型又は破壊型の遺伝子に置換することができる。欠失型又は破壊型の遺伝子によってコードされる酵素タンパク質は、生成したとしても、野生型酵素タンパク質とは異なる立体構造を有し、機能が低下又は消失している。このような相同組換えを利用した遺伝子置換による遺伝子欠失又は破壊は既に確立しており、温度感受性複製起点を含むプラスミド、接合伝達可能なプラスミドを用いる方法、宿主内で複製起点を持たないスイサイドベクターを利用する方法などがある(米国特許第6303383号、特開平05-007491号)。
具体的には、実施例1の項目に記載の方法により、poxFが破壊又は欠失したコリネバクテリウム グルタミカムを得ることができる。また、同様の方法でpobAが破壊又は欠失したコリネバクテリウム グルタミカムを得ることができる。
上記説明した本発明の形質転換体を、4-ヒドロキシベンゾエートを含有する反応液中で反応させる工程と、反応液中のフェノールを回収する工程とを含む方法によりフェノールを製造することができる。
反応に先立ち、形質転換体を好気条件下で、温度約25~38℃で、約12~48時間培養して増殖させることが好ましい。
反応に先立つ形質転換体の好気的培養に用いる培地は、炭素源、窒素源、無機塩類およびその他の栄養物質等を含有する天然培地または合成培地を用いることができる。
炭素源として、糖類(グルコース、フルクトース、マンノース、キシロース、アラビノース、ガラクトースのような単糖;スクロース、マルトース、ラクトース、セロビオース、キシロビオース、トレハロースのような二糖;澱粉のような多糖;糖蜜等)、マンニトール、ソルビトール、キシリトール、グリセリンのような糖アルコール;酢酸、クエン酵、乳酸、フマル酸、マレイン酸、グルコン酸のような有機酸;エタノール、プロパノールのようなアルコール;ノルマルパラフィンのような炭化水素等も用いることができる。
炭素源は、1種を単独で、又は2種以上を混合して使用できる。
無機塩類としては、リン酸第一カリウム、リン酸第二カリウム、硫酸マグネシウム、塩化ナトリウム、硝酸第一鉄、硫酸マンガン、硫酸亜鉛、硫酸コバルト、炭酸カルシウム等が挙げられる。無機塩は、1種を単独で、又は2種以上を混合して使用できる。無機塩類の培地中の濃度は、使用する無機塩によっても異なるが、通常、約0.01~1(w/v%)とすればよい。
さらに、必要に応じて、ビタミン類を添加することもできる。ビタミン類としては、ビオチン、チアミン(ビタミンB1)、ピリドキシン(ビタミンB6)、パントテン酸、イノシトール、ニコチン酸等が挙げられる。
培地のpHは約6~8が好ましい。
反応液は、フェノール前駆体(フェノール原料)を含有する水、緩衝液、無機塩培地などを用いることができる。
前駆体としては4-ヒドロキシベンゾエートを用いる。4-ヒドロキシベンゾエートとしては、ナトリウム塩、カリウム塩のような塩;炭素数1~4のアルコールとのエステルなどが挙げられる。中でも、塩が好ましく、ナトリウム塩がより好ましい。前駆体は、1種を単独で、又は2種以上を混合して使用できる。
反応液中の4-ヒドロキシベンゾエートの濃度は、約0.5~20(w/v%)が好ましく、約1~10(w/v%)がより好ましく、約2~5(w/v%)がさらにより好ましい。4-ヒドロキシベンゾエートは、芳香族化合物であるため細胞生存阻害作用を有するが、4-ヒドロキシベンゾエート濃度が上記範囲であれば、効率良く、フェノールを製造できる。
緩衝液としては、リン酸バッファー、トリスバファー、炭酸バッファーなどが挙げられる。緩衝液の濃度は、約10~150 mMが好ましい。
無機塩培地としては、リン酸第一カリウム、リン酸第二カリウム、硫酸マグネシウム、塩化ナトリウム、硝酸第一鉄、硫酸マンガン、硫酸亜鉛、硫酸コバルト、炭酸カルシウム等の無機塩の1種又は2種以上を含む培地が挙げられる。中でも、硫酸マグネシウムを含む培地が好ましい。無機塩培地として、具体的には、BT培地〔Omumasaba, C.A. et al., Corynebacterium glutamicum glyceraldehyde-3-phosphate dehydrogenase isoforms with opposite, ATP-dependent regulation. J. Mol. Microbiol. Biotechnol. 8:91-103 (2004)〕等が挙げられる。無機塩類の培地中の濃度は、使用する無機塩によっても異なるが、通常、約0.01~1(w/v%)とすればよい。
反応液のpHは約6~8が好ましい。反応中は、アンモニア水溶液、水酸化ナトリウム水溶液などを用いて、pHコントローラー (例えば、エイブル株式会社製、型式:DT-1023)で、反応液のpHを中性付近、特に約7にコントロールしながら反応させることが好ましい。
反応温度、即ち反応中の形質転換体の生存温度は、約20~50℃が好ましく、約25~47℃がより好ましい。上記温度範囲であれば、効率良くフェノールを製造できる。
また、反応時間は、約1~7日間が好ましく、約1~3日間がより好ましい。
培養は、バッチ式、流加式、連続式の何れでもよい。中でも、バッチ式が好ましい。
反応は、好気的条件で行ってもよく、還元条件で行ってもよいが、還元条件で行うことが好ましい。還元条件では、コリネバクテリウム グルタミカムは実質的に増殖せず、一層効率的にフェノールを生産させることができる。
還元条件は、反応液の酸化還元電位で規定される。反応液の酸化還元電位は、約-200mV~-500mVが好ましく、約-250mV~-500mVがより好ましい。
反応液の還元状態は簡便にはレサズリン指示薬(還元状態であれば、青色から無色への脱色)で推定できるが、正確には酸化還元電位差計(例えば、BROADLEY JAMES社製、ORP Electrodes)を用いて測定できる。
具体的には、蒸留水などを加熱処理や減圧処理して溶解ガスを除去することにより、還元条件の反応液用水溶液を得ることができる。この場合、約10 mmHg以下、好ましくは約5 mmHg以下、より好ましくは約3 mmHg以下の減圧下で、約1~60分程度、好ましくは約5~40分程度、蒸留水などを処理することにより、溶解ガス、特に溶解酸素を除去して還元条件下の反応液用水溶液を作成することができる。
また、適当な還元剤(例えば、チオグリコール酸、アスコルビン酸、システィン塩酸塩、メルカプト酢酸、チオール酢酸、グルタチオン、硫化ソーダ等)を添加して還元条件の反応液用水溶液を調整することもできる。
これらの方法を適宜組み合わせることも有効な還元条件の反応液用水溶液の調整方法である。
上記のようにして培養することにより、反応液中にフェノールが生産される。反応液を回収することによりフェノールを回収できるが、さらに、公知の方法でフェノールを反応液から分離することもできる。そのような公知の方法として、蒸留法、膜透過法、有機溶媒抽出法等が挙げられる。
実施例1 フェノール生産遺伝子のクローニングと発現
(1) 微生物からの染色体DNAの抽出
コリネバクテリウム グルタミカム (Corynebacterium glutamicum) R (FERM P-18976)からの染色体DNA抽出は、A培地 [(NH2)2CO 2 g、(NH4)2SO4 7 g、KH2PO4 0.5 g、K2HPO4 0.5 g、MgSO4.7H2O 0.5 g、0.06% (w/v) Fe2 SO4.7H2O + 0.042% (w/v) MnSO4.2H2O 1 ml、0.02% (w/v) biotin solution 1 ml、0.01% (w/v) thiamin solution 2 ml、yeast extract 2 g、vitamin assay casamino acid 7 gを蒸留水1 Lに溶解] に、炭素源として、最終濃度4%になるように50% (w/v)グルコース溶液を添加し、白金耳を用いて植菌後、対数増殖期まで33℃で振盪培養し、菌体を集菌後、DNAゲノム抽出キット(商品名:GenomicPrep Cells and Tissue DNA Isolation Kit、アマシャム社製)を用いて、取扱説明書に従い、集めた菌体から染色体DNAを回収した。
エシェリヒア コリ(Escherichia coli) W NBRC 13500からの染色体DNA抽出は、NBRC Medium No.802培地 [polypeptone 10 g, yeast extract 2 g, MgSO4・7H2O 1 gを蒸留水1 Lに溶解]に、白金耳を用いて植菌後、対数増殖期まで30℃で振盪培養し、菌体を集菌後、DNAゲノム抽出キット(商品名:GenomicPrep Cells and Tissue DNA Isolation Kit、アマシャム社製)を用いて、取扱説明書に従い、集めた菌体から染色体DNAを回収した。
クローニングベクターpCRB22の構築
コリネバクテリウム カゼイ JCM12072由来のプラスミドpCASE1のDNA複製起点(以降、pCASE1-oriと記す)配列、及びクローニングベクターpHSG298(タカラバイオ株式会社製)をそれぞれ含むDNA断片を以下のPCR法により増幅した。
PCRに際して、pCASE1-ori配列、クローニングベクターpHSG298をそれぞれクローン化するべく、配列番号1(pCASE1-ori配列)、配列番号2(クローニングベクター-pHSG298)を基に、それぞれ下記の一対のプライマーを合成し、使用した。

実際のPCRは、サーマルサイクラー GeneAmp PCR System 9700(アプライド・バイオシステムズ社製)を用い、反応試薬としてTaKaRa LA Taq(タカラバイオ株式会社製)を用いて下記の条件で行った。
得られたライゲーションA液を、塩化カルシウム法〔Journal of Molecular Biology, 53, 159 (1970)〕によりエシェリヒア コリJM109を形質転換し、カナマイシン50μg/mlを含むLB寒天培地〔1% ポリペプトン、0.5% 酵母エキス、0.5% 塩化ナトリウム、および1.5% 寒天〕に塗布した。
培地上の生育株を常法により液体培養し、培養液よりプラスミドDNAを抽出、該プラスミドを制限酵素BglIIでそれぞれ切断し、挿入断片を確認した。この結果、クローニングベクターpHSG298約2.7-kbのDNA断片に加え、pCASE-ori配列の約1.4-kb DNA断片が認められた。
pCASE1-ori配列を含むクローニングベクターをpCRB22と命名した。
コリネバクテリウム グルタミカムR由来のグリセルアルデヒド3リン酸デヒドロゲナーゼ(glyceraldehyde-3-phosphate dehydrogenase)をコードするgapA遺伝子のプロモーター配列(以降、PgapAと記す)を含むDNA断片、及びクローニングベクターpKK223-3(ファルマシア社製)由来rrnBT1T2双方向ターミネーター配列(以降、ターミネーター配列と記す)を含むDNA断片を以下の方法により増幅した。
PCRに際して、PgapA配列及びターミネーター配列をそれぞれクローン化するべく、配列番号7(PgapA配列)、配列番号8(ターミネーター配列)を基に、それぞれ下記の一対のプライマーを合成し、使用した。

培地上の生育株を常法により液体培養し、培養液よりプラスミドDNAを抽出、該プラスミドを制限酵素SalIでそれぞれ切断し、挿入断片を確認した。この結果、クローニングベクターpCRB22約4.1-kbのDNA断片に加え、PgapA配列の約0.6-kb DNA断片が認められた。
PgapA配列を含むクローニングベクターをpCRB206と命名した。
得られたライゲーションC液を、塩化カルシウム法〔Journal of Molecular Biology, 53, 159 (1970)〕によりエシェリヒア コリJM109を形質転換し、カナマイシン50μg/mlを含むLB寒天培地〔1% ポリペプトン、0.5% 酵母エキス、0.5% 塩化ナトリウム、および1.5% 寒天〕に塗布した。
培地上の生育株を常法により液体培養し、培養液よりプラスミドDNAを抽出、該プラスミドを制限酵素で切断し、挿入断片を確認した。この結果、クローニングベクターpCRB206約4.7-kbのDNA断片に加え、ターミネーター配列の約0.4-kb DNA断片が認められた。
rrnBT1T2ターミネーター配列を含むクローニングベクターをpCRB207と命名した。

この培地上の生育株を常法により液体培養し、培養液よりプラスミドDNAを抽出、該プラスミドを制限酵素NdeIで切断し、制限酵素サイトの挿入を確認した。
PgapA配列及びrrnBT1T2ターミネーター配列を含むクローニングベクタ-をpCRB209と命名した。
バチラス サブチリス由来のフェノール生産遺伝子のクローニング
バチラス サブチリス由来の4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有する遺伝子をコードするbsdBCD遺伝子を含むDNA断片を以下のPCR法により増幅した。
PCRに際して、bsdBCD遺伝子をクローン化するべく、配列番号16(バチラスサブチリス bsdBCD遺伝子)を基に、それぞれ下記の一対のプライマーを、アプライド・バイオシステムズ(Applied Biosystems)社製「394 DNA/RNAシンセサイザー(synthesizer)」を用いて合成し、使用した。

コリネバクテリウム グルタミカム株由来poxF欠失遺伝子を含むプラスミドをpCRA725-poxF/CGと命名した。

コリネバクテリウム グルタミカム株poxF遺伝子破壊株の構築
マーカーレス染色体遺伝子導入用ベクターpCRA725は、コリネバクテリウム グルタミカムR内で複製不能なプラスミドである。pCRA725-poxF/CGを用いて、電気パルス法 [Agric. Biol. Chem.、Vol. 54、443-447(1990) 及びRes. Microbiol.、Vol. 144、181-185(1993)] により、コリネバクテリウム グルタミカムRを形質転換し、カナマイシン 50μg/mlを含むA寒天培地〔A液体培地、および1.5% 寒天〕に塗布した。上記の培地で得られた一重交叉株を、10%(W/V)スクロース含有BT寒天培地[(NH2)2CO 2g、(NH4)2SO4 7g、KH2PO4 0.5 g、K2HPO4 0.5 g、MgSO4.7H2O 0.5 g、0.06% (w/v) Fe2 SO4.7H2O + 0.042% (w/v) MnSO4.2H2O 1 ml、0.02% (w/v) biotin solution 1 ml、0.01% (w/v) thiamin solution 2 mlを蒸留水1Lに溶解、および1.5% 寒天]に塗付した。
そこで、カナマイシン感受性及びスクロース含有培地生育性を示した株を選択した。このコリネバクテリウム グルタミカムR株poxF遺伝子マーカーレス破壊株をコリネバクテリウム グルタミカム(Corynebacterium glutamicum)ΔpoxFと命名した。
マーカーレス染色体遺伝子導入用ベクターpCRA725は、コリネバクテリウム グルタミカムR内で複製不能なプラスミドである。pCRA725-pobA/CGを用いて、電気パルス法 [Agric. Biol. Chem., Vol. 54, 443-447(1990) 及びRes. Microbiol., Vol. 144, 181-185(1993)] により、コリネバクテリウム グルタミカムΔpoxFを形質転換し、カナマイシン50μg/mlを含むA寒天培地〔A液体培地、および1.5% 寒天〕に塗布した。上記の培地で得られた一重交叉株を、10%(W/V)スクロース含有BT寒天培地[(NH2)2CO 2g、(NH4)2SO4 7g、KH2PO4 0.5 g、K2HPO4 0.5 g、MgSO4.7H2O 0.5 g、0.06% (w/v) Fe2 SO4.7H2O + 0.042% (w/v) MnSO4.2H2O 1 ml、0.02% (w/v) biotin solution 1 ml、0.01% (w/v) thiamin solution 2 mlを蒸留水1Lに溶解、および1.5% 寒天]に塗付した。

上述のプラスミド12種類(pCRB209-bsdBCD/BS、pCRB209-dca/BAE、pCRB209-dca/BSS、pCRB209-dca/CKO、pCRB209-dca/EAE、pCRB209-dca/ECL、pCRB209-dca/EHO、pCRB209-dca/ESA、pCRB209-dca/ECK、pCRB209-dca/EFE、pCRB209-dca/PPY及びpCRB209-dca/PAM)をそれぞれ用いて、電気パルス法 [Agric. Biol. Chem.、Vol. 54、443-447(1990) 及びRes. Microbiol.、Vol. 144、181-185(1993)] により、コリネバクテリウム グルタミカムΔpoxF株を形質転換し、カナマイシン 50μg/mlを含むA寒天培地に塗布した。
この培地上の生育株を常法により液体培養し、培養液よりプラスミドDNAを抽出、該プラスミドを制限酵素で切断し、挿入断片を確認した。この結果、上記で作製のプラスミドpCRB209-bsdBCD/BS、pCRB209-dca/BAE、pCRB209-dca/BSS、pCRB209-dca/CKO、pCRB209-dca/EAE、pCRB209-dca/ECL、pCRB209-dca/EHO、pCRB209-dca/ESA、pCRB209-dca/ECK、pCRB209-dca/EFE、pCRB209-dca/PPY及びpCRB209-dca/PAMの導入が認められた。

Biol. Chem., Vol. 54, 443-447(1990) 及びRes. Microbiol. Vol. 144, 181-185(1993)] により、コリネバクテリウム グルタミカムΔpoxFΔpobA株を形質転換し、カナマイシン 50μg/mlを含むA寒天培地に塗布した。

上述のプラスミド12種類(pCRB209-bsdBCD/BS、pCRB209-dca/BAE、pCRB209-dca/BSS、pCRB209-dca/CKO、pCRB209-dca/EAE、pCRB209-dca/ECL、pCRB209-dca/EHO、pCRB209-dca/ESA、pCRB209-dca/ECK、pCRB209-dca/EFE、pCRB209-dca/PPY及びpCRB209-dca/PAM)をそれぞれ用いて、電気パルス法 [Agric. Biol. Chem., Vol. 54、443-447(1990) 及びRes. Microbiol., Vol. 144、181-185(1993)] により、コリネバクテリウム グルタミカム R株を形質転換し、カナマイシン 50μg/mlを含むA寒天培地に塗布した。
この培地上の生育株を常法により液体培養し、培養液よりプラスミドDNAを抽出、該プラスミドを制限酵素で切断し、挿入断片を確認した。この結果、上記で作製のプラスミドpCRB209-bsdBCD/BS、pCRB209-dca/BAE、pCRB209-dca/BSS、pCRB209-dca/CKO、pCRB209-dca/EAE、pCRB209-dca/ECL、pCRB209-dca/EHO、pCRB209-dca/ESA、pCRB209-dca/ECK、pCRB209-dca/EFE、pCRB209-dca/PPY及びpCRB209-dca/PAMの導入が認められた。
pCRB209-bsdBCD/BSの導入が認められた株をコリネバクテリウム グルタミカムPHE23-1、pCRB209-dca/BAEの導入が認められた株をコリネバクテリウム グルタミカムPHE23-2、pCRB209-dca/BSSの導入が認められた株をコリネバクテリウム グルタミカムPHE23-3、pCRB209-dca/CKOの導入が認められた株をコリネバクテリウム グルタミカムPHE23-4、pCRB209-dca/EAEの導入が認められた株をコリネバクテリウム グルタミカムPHE23-5、pCRB209-dca/ECLの導入が認められた株をコリネバクテリウム グルタミカムPHE23-6、pCRB209-dca/EHOの導入が認められた株をコリネバクテリウム グルタミカムPHE23-7、pCRB209-dca/ESAの導入が認められた株をコリネバクテリウム グルタミカムPHE23-8、pCRB209-dca/ECKの導入が認められた株をコリネバクテリウム グルタミカムPHE23-9、pCRB209-dca/EFEの導入が認められた株をコリネバクテリウム グルタミカムPHE23-10、pCRB209-dca/PPYの導入が認められた株をコリネバクテリウム グルタミカムPHE23-11、pCRB209-dca/PAMの導入が認められた株をコリネバクテリウム グルタミカムPHE23-12と命名した。尚、本株の遺伝子組換えの概要を、表3にまとめて示した。
実施例1において作製したコリネバクテリウム グルタミカムΔpoxF/フェノール生産遺伝子導入株(表1参照)を、カナマイシン50μg/mlを含むA寒天培地[(NH2)2CO 2g、(NH4)2SO4 7g、KH2PO4 0.5 g、K2HPO4 0.5 g、MgSO4.7H2O 0.5 g、0.06% (w/v) Fe2 SO4.7H2O + 0.042% (w/v) MnSO4.2H2O 1 ml、0.02% (w/v) biotin solution 1 ml、0.01% (w/v) thiamin solution 2 ml、yeast extract 2 g、vitamin assay casamino acid 7 g、glucose 40 g、寒天 15 gを蒸留水1Lに懸濁] に塗布し、28℃、20時間暗所に静置した。
上記のプレートで生育したコリネバクテリウム グルタミカムΔpoxF/フェノール生産遺伝子導入株を、カナマイシン50μg/mlを含有したA液体培地10mlの入った試験管に一白金耳植菌し、28℃にて15時間、好気的に振盪培養を行った。
上記条件で生育したコリネバクテリウム グルタミカムΔpoxF/フェノール生産遺伝子導入株を、カナマイシン50μg/mlを含有したA液体培地500mlの入った容量2Lの三角フラスコに植菌し、28℃にて15時間、好気的に振盪培養を行った。
サンプリングした反応液を遠心分離 (4℃、15,000×g、10分) し、得られた上清液を用いてフェノールの定量を行った。
この結果、コリネバクテリウム グルタミカムPHE21株は、還元条件下における反応において、1時間後に65 mM(6 g/L)、4時間後に180 mM (17 g/L)のフェノールを生成していた(表4)。同条件下におけるコリネバクテリウム グルタミカムPHE21-2~PHE21-12株の結果も表4に併せて示した。

この結果より、poxA遺伝子を破壊することによりフェノール生産性の向上が観察された。

この結果より、コリネバクテリウム グルタミカムR株(野生株)よりも、ΔpoxF株(表4)、さらにはΔpoxFΔpobA株(表5)にフェノール生産遺伝子を導入株した方が、フェノール生産性が高いことより、poxF遺伝子の破壊及びpobA遺伝子の破壊によりフェノール生産性に正の効果があることが明らかとなった。

フェノールによる好気増殖への影響
コリネバクテリウム グルタミカム、エシェリヒア コリおよびシュードモナス プチダについて、好気培養におけるフェノールの生育阻害試験を行った。尚、本試験に用いたシュードモナス プチダS12は、溶媒耐性菌として報告されており、これまでに唯一フェノール生産の宿主として用いられた技術が開示されている。
コリネバクテリウム グルタミカムRをA寒天培地[(NH2)2CO 2g、(NH4)2SO4 7g、KH2PO4 0.5 g、K2HPO4 0.5 g、MgSO4.7H2O 0.5 g、0.06% (w/v) Fe2 SO4.7H2O + 0.042% (w/v) MnSO4.2H2O 1 ml、0.02% (w/v) biotin solution 1 ml、0.01% (w/v) thiamin solution 2 ml、yeast extract 2 g、vitamin assay casamino acid 7 g、glucose 40 g、寒天 15 gを蒸留水1Lに懸濁]に塗布し、33℃、15時間暗所に静置した。
上記のプレートで生育したコリネバクテリウム グルタミカムRを、A液体培地[(NH2)2CO 2g、(NH4)2SO4 7g、KH2PO4 0.5 g、K2HPO4 0.5 g、MgSO4.7H2O 0.5 g、0.06% (w/v) Fe2 SO4.7H2O + 0.042% (w/v) MnSO4.2H2O 1 ml、0.02% (w/v) biotin solution 1 ml、0.01% (w/v) thiamin solution 2 ml、yeast extract 2 g、vitamin assay casamino acid 7 g、glucose 40 gを蒸留水1Lに溶解] 10mlの入った試験管に一白金耳植菌し、33℃にて13時間、好気的に振盪培養を行った。
上記条件で生育したコリネバクテリウム グルタミカムRを、A液体培地100mlに初期菌体濃度OD610=0.05となるように植菌し、同時にフェノールが終濃度0、0.16、0.2、0.24、0.32mMとなるように添加し、33℃にて好気的に振盪培養を行った。菌体の生育はOD610の吸光度を測定することにより行った。
上記プレートで生育したエシェリヒア コリJM109を、LB液体培地〔1% ポリペプトン、0.5% 酵母エキスおよび0.5% 塩化ナトリウム〕10mlの入った試験管に一白金耳植菌し、37℃にて13時間、好気的に振盪培養を行った。
上記条件で生育したエシェリヒア コリJM109をLB液体培地100mlに初期菌体濃度OD610=0.05となるように植菌し、同時にフェノール濃度が終濃度0、0.16、0.20mMとなるように添加し、37℃にて好気的に振盪培養を行った。菌体の生育はOD610の吸光度を測定することにより行った。
上記プレートで生育したシュードモナス プチダF1およびS12を、LB(+グルコース)液体培地〔1% ポリペプトン、0.5% 酵母エキス、0.5% 塩化ナトリウムおよび0.4%グルコース〕10mlの入った試験管に一白金耳植菌し、30℃にて13時間、好気的に振盪培養を行った。
上記条件で生育したシュードモナス プチダF1およびS12株をLB(+グルコース)液体培地100mlに初期菌体濃度OD610=0.05となるように植菌し、同時にフェノール濃度が終濃度0、0.10、0.20mMとなるように添加し、30℃にて好気的に振盪培養を行った。菌体の生育はOD610の吸光度を測定することにより行った。
培地中へのフェノール添加による好気増殖への影響の解析結果を図2に示す。
シュードモナス プチダF1及び溶剤耐性菌として報告されていたシュードモナス プチダS12は、ほぼ同じ傾向を示し、0.10%フェノール存在下で著しく増殖阻害を受け0.20%フェノールでは完全に増殖が阻害された。
これに対して、コリネバクテリウム グルタミカムは、エシェリヒア コリが顕著な増殖阻害を受けた0.16%のフェノール存在下においても増殖への影響はほとんどなく、エシェリヒア コリやシュードモナス プチダでは完全に増殖を阻害された0.20%のフェノール存在下においても良好な生育を示した。さらに0.24%のフェノール存在下において増殖が可能であった。
このように、コリネバクテリウム グルタミカムはエシェリヒア コリおよびシュードモナス プチダと比較して、フェノールに対し高い耐性を有し、フェノール生産の宿主として高い適性を有することが示された。
Claims (12)
- 4-ヒドロキシベンゾエート デカルボキシラーゼ(4-hydroxybenzoate decarboxylase)活性を有する酵素をコードする遺伝子が、宿主のコリネバクテリウム グルタミカム(Corynebacterium glutamicum)に導入された、フェノール生産能を有する形質転換体。
- 4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有する酵素をコードする遺伝子が、バチラス サブチリス(Bacillus subtilis)、バチラス アトロファエウス(Bacillus atrophaeus)、バチラス サブチリス 亜種 スピジゼニイ(Bacillus subtilis subsp. spizizenii)、シトロバクター コセリ(Citrobacter koseri)、エンテロバクター アエロゲネス(Enterobacter aerogenes)、エンテロバクター クロアカエ(Enterobacter cloacae)、エンテロバクター ホルメシェイ(Enterobacter hormaechei)、エンテロバクター サカザキ(Enterobacter sakazakii)、エシェリヒア コリ(Escherichia coli)、エシェリヒア フェルグソニー(Escherichia fergusonii)、パエニバチラス ポリミキサ(Paenibacillus polymyxa)、又はパントエア アナナティス(Pantoea ananatis)由来の遺伝子である請求項1に記載の形質転換体。
- 4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有する酵素をコードする遺伝子が下記の(a)又は(b)のDNAである請求項1に記載の形質転換体。
(a) 配列番号16、配列番号23、配列番号26、配列番号29、配列番号32、配列番号35、配列番号38、配列番号41、配列番号44、配列番号47、配列番号50、又は配列番号53の塩基配列からなるDNA
(b) (a)の何れかの塩基配列と相補的な塩基配列からなるDNAとストリンジェントな条件でハイブリダイズし、かつ4-ヒドロキシベンゾエート デカルボキシラーゼ活性を有するポリペプチドをコードするDNA - 宿主のコリネバクテリウム グルタミカムが、その染色体上に存在するフェノール2-モノオキシゲナーゼ(phenol 2-monooxygenase)活性を有する酵素をコードする遺伝子が破壊され、又は欠損したものである、請求項1~3の何れかに記載の形質転換体。
- 宿主のコリネバクテリウム グルタミカムが、その染色体上に存在する4-ヒドロキシベンゾエート ヒドロキシラーゼ(4-hydroxybenzoate hydroxylase)活性を有する酵素をコードする遺伝子が破壊され、又は欠損したものである、請求項1~4の何れかに記載の形質転換体。
- 宿主のコリネバクテリウム グルタミカムが、コリネバクテリウム グルミカムR(FERM P-18976)、ATCC13032、又はATCC13869である請求項1~3の何れかに記載の形質転換体。
- 宿主のコリネバクテリウム グルタミカムが、コリネバクテリウム グルミカムR(FERM P-18976)、ATCC13032、又はATCC13869の染色体上に存在するフェノール2-モノオキシゲナーゼ活性を有する酵素をコードする遺伝子が破壊され、又は欠損したものである、請求項1~3の何れかに記載の形質転換体。
- 宿主のコリネバクテリウム グルタミカムが、コリネバクテリウム グルミカムR(FERM P-18976)、ATCC13032、又はATCC13869の染色体上に存在する4-ヒドロキシベンゾエート ヒドロキシラーゼ活性を有する酵素をコードする遺伝子が破壊され、又は欠損したものである、請求項1~3の何れかに記載の形質転換体。

コリネバクテリウム グルタミカム PHE21(受託番号:NITE BP-996)、PHE21-2、PHE21-3、PHE21-4、PHE21-5、PHE21-6、PHE21-7、PHE21-8、PHE21-9、PHE21-10、PHE21-11、PHE21-12、PHE22-1、PHE22-2、PHE22-3、PHE22-4、PHE22-5、PHE22-6、PHE22-7、PHE22-8、PHE22-9、PHE22-10、PHE22-11、PHE22-12、PHE23-1、PHE23-2、PHE23-3、PHE23-4、PHE23-5、PHE23-6、PHE23-7、PHE23-8、PHE23-9、PHE23-10、PHE23-11、又はPHE23-12の形質転換体。 - 請求項1~9の何れかに記載の形質転換体を、還元条件下、4-ヒドロキシベンゾエート又はその塩を含有する反応液中で反応させる工程と、反応液中のフェノールを回収する工程とを含むフェノールの製造方法。
- 反応工程において、形質転換体が実質的に増殖しない請求項10に記載のフェノールの製造方法。
- 還元条件下の反応液の酸化還元電位が-200~-500ミリボルトである請求項10又は11に記載のフェノールの製造方法。
