例えば、ポリグリセリン脂肪酸エステルやショ糖脂肪酸エステル等の界面活性剤は食品添加物であるが、これらの界面活性剤は起泡性が不十分であり、洗浄成分として使用した場合、手洗いが不十分となる可能性があった。そのため、食品添加物である界面活性剤を主成分として含有しながら、高い起泡性を有する洗浄剤が求められていた。また、手洗い以外の感染防止策としては、アルコール製剤等の消毒液をスプレー等で塗付して手に摺りこませる方法等が挙げられる。最近では、スプレー等で塗付した消毒液を手の隅々までむらなく摺りこませることができない乳幼児向けに、泡で手全体を包み込んで消毒できる泡状の消毒液が発売されている。しかしながら、洗浄成分として、上記のような食品添加物である界面活性剤を使用した場合、起泡性が不十分であるばかりでなく、ポリグリセリン脂肪酸エステルやショ糖脂肪酸エステルはアルコールとの相溶性が悪いため、低温保存下で析出する場合があった。したがって、アルコールを多量に配合しても豊かな泡で手全体を包み込むことができ、低温安定性が良好な泡状の消毒液が求められていた。
ポリオキシエチレンソルビタン脂肪酸エステルを使用した洗浄剤の報告例としては、例えば、特許文献1及び2が挙げられる。特許文献1では、(A)低級アルコール、(B)有機酸及びそのアルカリ金属塩、又は無機酸及びそのアルカリ金属塩、(C)ポリオキシエチレンソルビタン脂肪酸エステルを含む1種類以上の非イオン性界面活性剤を含有する便座用除菌洗浄剤組成物が提案されている。この洗浄剤組成物は、低温保存安定性が改善され、除菌効果には優れるものの、起泡性の点で不十分であった。
また、特許文献2では、(A)ポリオキシエチレンソルビタンモノ脂肪酸エステル、(B)HLB8以下の親油性多価アルコールモノ脂肪酸エステルを含有する食品用洗浄剤が提案されている。この食品用洗浄剤は、洗浄力は優れているものの、起泡性の点で不十分であった。
さらに、低温保存下で親油性多価アルコールモノ脂肪酸エステルが析出し、保存安定性が不十分となる場合があった。
このように、主たる洗浄成分が食品又は食品添加物でありながら、起泡性が良好で、優れた除菌効果を有し、しかも低温安定性にも優れる除菌洗浄剤は未だ提供されていないのが実情であった。
すなわち、本発明は、下記(a)、(b)及び(c)成分を含有し、組成物全量に対するa成分とb成分の含有量の和[a+b]が0.1~30質量%であり、a成分とb成分の質量比[a/b]が1/2~4/1であり、組成物全量に対するc成分の含有量が60~99.5質量%である、除菌洗浄剤組成物に関する。
(a) ポリオキシエチレンモノラウリン酸ソルビタン
(b) アシル基の炭素数が8~12である脂肪酸グリセライドであって、b成分全量に対するモノグリセライドの含有量が85質量%以上であり、かつ、モノグリセライド全量に対する1-モノグリセライドの質量分率が0.9~1.0である脂肪酸グリセライド
(c) 水及び/又はエタノール
本発明のa成分であるポリオキシエチレンモノラウリン酸ソルビタンは、ソルビトール又はソルビタンとラウリン酸との部分エステルであるソルビタン脂肪酸エステルに、エチレンオキシドを付加させた非イオン性界面活性剤である。
ポリオキシエチレンソルビタン脂肪酸エステルの構造は特に限定されるものではないが、Griffinの算定式に基づくHLBとしては14~19、好ましくは15~18、より好ましくは16~17である。HLBが14未満では、起泡性を阻害する場合があり、HLBが19を超えると除菌効果が発揮されない場合がある。
本発明のa成分の脂肪酸残基は、ラウリン酸である。炭素数が12未満の脂肪酸の場合、起泡性が低下する傾向がある。炭素数が12を超える脂肪酸の場合、起泡性がやや低下し、十分な除菌効果が得られにくい傾向がある。ただし、全脂肪酸量に対するラウリン酸の比率が50質量%以上であれば混合脂肪酸を用いても構わない。また、ラウリン酸を主成分とするヤシ油・パーム核油等の天然油脂由来の脂肪酸でもよい。
脂肪酸グリセライドは、脂肪酸とグリセリンのエステル化反応、又は油脂及び脂肪酸エステルとグリセリンとのエステル交換反応によって得られる。反応によって得られたものは反応グリセライドと呼ばれる。
反応グリセライドは、一般に、主成分として1-モノグリセライド、副成分として2-モノグリセライド、1,3-ジグリセライド、1,2-ジグリセライド、トリグリセライド、そしてグリセリンからなる混合物である。反応グリセライドを蒸留することで、モノグリセライド純度を高めることが可能であり、得られたものは蒸留モノグリセライドと呼ばれる。
モノグリセライドの含有量は、常法に従い、前処理として試料をシリルエーテル化した後、ガスクロマトグラフィーで測定することができる。
本発明のb成分において、モノグリセライド全量に対する1-モノグリセライドの質量分率は0.9~1.0であり、好ましくは0.92~1.0、より好ましくは0.94~1.0である。モノグリセライドは、1-モノグリセライドと2-モノグリセライドの2種の構造異性体に分類され、1-モノグリセライドと2-モノグリセライドとの質量の和に対する1-モノグリセライドの質量分率が、モノグリセライド全量に対する1-モノグリセライドの質量分率である。
本発明において、モノグリセライド全量に対する1-モノグリセライドの質量分率が0.9~1.0であるb成分を使用することは、起泡性や除菌効果を発揮する上で決定的に重要な要素である。1‐モノグリセライドの質量分率が0.9未満では、起泡性、除菌効果において不十分である。
モノグリセライド全量に対する1-モノグリセライドの質量分率は、1H-NMRを用いてグリセロールの2位の炭素(下記式(1)及び(2)中、*1及び*2で示す)に位置するプロトンの存在比から、以下の計算式によって算出することができる。
[a/b]が4/1を超えると、起泡性及び除菌効果が不十分となる。一方、[a/b]が1/2未満では、起泡性及び低温安定性が不十分となる。
c成分として、水及びエタノールの混合溶媒を使用する場合、その混合割合は任意であり、製剤のタイプにより適宜に調整する。例えば、ハンドソープタイプの洗浄剤であれば、水を主溶媒とし、溶媒全量中、30質量%までのエタノールを混合することができる。また、泡状消毒剤タイプの洗浄剤であれば、溶媒全量中、30~85質量%のエタノールを混合することが好ましく、40~80質量%を混合することがさらに好ましい。
本発明の洗浄剤組成物には、粘度や泡質を調整するために、食品又は食品添加物に指定されている多糖類又はその誘導体を使用することもできる。例えば、アラビアガム、アルギン酸又はその塩、アルギン酸プロピレングリコール又はその塩、カラギナン、ガディガム、カードラン、キタンサンガム、グァーガム、キトサン、ジェランガム、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロース又はその塩、タマリンドシードガム、タラガム、プルラン、ペクチンなどが挙げられる。これらの中でも、洗浄剤としての基本機能、つまり水への透明溶解性、温度変化に対する保存安定性、泡質の改善効果、増粘効果、すすぎ性等を総合的に考慮すると、アルギン酸又はその塩、アルギン酸プロピレングリコール又はその塩、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロース又はその塩が好ましく、アルギン酸又はその塩、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロース又はその塩がさらに好ましい。
さらに、本発明の洗浄剤組成物には、発明の効果を損なわない範囲でその他の成分を添加することができ、その他添加成分としては、ポリグリセリン脂肪酸エステル、ショ糖脂肪酸エステル等の、本発明において規定した界面活性剤以外の界面活性剤、植物油脂、動物油脂などの油脂類、スクワレン、流動パラフィンなどの炭化水素油、プロピレングリコール、グリセリンなどの溶剤、クエン酸又はその塩、コハク酸又はその塩、リンゴ酸又はその塩、酒石酸又はその塩、エチレンジアミン四酢酸又はその塩、メタリン酸又はその塩などのビルダー、トコフェロール類などの抗酸化剤、多糖類、アミノ酸、ペプチド、本発明の組成物の成分以外の防腐剤、香料、着色剤などを適宜配合することができる。
本発明の洗浄剤組成物の除菌効果は、長期にわたって持続することから、組成物に別途防腐剤を添加しなくても、微生物汚染に対する保存力(防腐効力)を得ることができる。よって、本発明は、防腐剤組成物としても機能する。また、上記各用途を含め、例えば、化粧水、乳液、クリーム、美容液、洗顔料、クレンジング料等のスキンケア製剤や、ヘアシャンプー、ヘアコンディショナー、ヘアトリートメント、ヘアスタイリング料等のヘアケア製剤等、任意の製剤に対し、本発明のaないしc成分を、本発明において規定する量的関係を充足するように配合することによって、各種製剤に対し防腐効果を付与することができるため、本発明は、各種製剤に対し防腐効果を付与するための方法としての側面をも有する。
表2及び3に示す配合からなるハンドソープ、及び表4に示す配合からなる泡状消毒液を調製し、1)起泡性、2)低温安定性、3)除菌効果についてそれぞれ評価した。評価結果を、表2ないし表4に示す。
表2ないし表4中の「a成分」及び「b成分」は、本発明のa成分及びb成分であり、「a’成分」及び「b’成分」は、a成分及びb成分に対する比較成分である。また、「b成分」及び「b’成分」は、いずれも市販品であり、下記表1に示す脂肪酸残基、モノグリセライド含量、及び1-モノグリセライド質量分率を有するものである。

また、1-モノグリセライドの質量分率については、NMRスペクトルから前記した式に基づいて算出した。
・ハンドソープ
表2及び表3中のヒドロキシプロピルメチルセルロース、カルボキシメチルセルロースナトリウム又はアルギン酸ナトリウムを、常温(約25℃)でイオン交換水に分散し、ダマがないことを確認してからウォーターバス中プロペラで攪拌しながら80℃まで加温した。その後、エタノール以外の成分を全て投入し、溶解を確認した後、40℃以下に冷却した。40℃以下でエタノールを投入し、均一に溶解するまで攪拌してハンドソープを得た。
・泡状消毒液
表4中のイオン交換水及びエタノールを常温(約25℃)で仕込み、均一に溶解した後、その他の原料を投入した。均一に溶解するまで常温で攪拌し、泡状消毒液を得た。
・起泡性
ハンドソープの5質量%水溶液をミルサー試験機(Iwatani(株)製、IFM-100)で5秒間攪拌し、1分間静置した後の泡の高さを測定した。判定は下記の基準で行い、AA及びAを合格とした。
(評点) : (評価)
AA : 泡の高さが30mm以上
A : 泡の高さが25mm以上~30mm未満
B : 泡の高さが20mm以上~25mm未満
C : 泡の高さが20mm未満
調製したハンドソープ50gを50mLのスクリュー管に充填し、密閉した後、5℃の恒温槽で1ヶ月間保存した。試験後の試料の性状を確認し、以下の基準で評価を行った。
○ : 試料の外観に変化が確認されない。
× : 沈殿の発生や固化など、試料の外観に変化が確認される。
Association of Official Agricultural Chemists (AOAC)のサニタイザー試験法に準じて行った。詳しくは、各種供試薬剤の原液9.9mLに、Nutrient Broth(NB)培地(MERCK社製)に108~109個の菌数になるよう調整した菌液を0.1mL加える。30秒接触させた後、その1mLを不活化剤入りリン酸緩衝液9mLに加え、その後すぐに段階希釈を行う。Plate Count Agar(PCA)培地(MERCK社製)で混釈し、37℃で培養後48時間及び7日後に生存菌数を測定した。接種菌数と生存菌数の対数差からLog Reductionを算出し、以下の判定基準で評価した。尚、供試菌としては、大腸菌(Escherichia coli IFO-12734)と黄色ブドウ球菌(Staphylococcus aureus IFO-12732)を用いた。
判定は、培養7日後の生菌数が検出限界以下で、かつ培養48時間後のLog Reductionが下記の基準でAA及びAである試料を合格とした。
(評点): (評価)
AA : 供試菌の Log Reductionが5以上
A : 供試菌のLog Reductionが4以上~5未満
B : 供試菌のLog Reductionが3以上~4未満
C : 供試菌のLog Reductionが3未満
* 2種類の供試菌のLog Reductionの評価が異なる場合は、評点の低い方で判定した。
* 培養48時間後のLog ReductionがAA及びAであっても、培養7日後に菌の生存が確認された場合は不合格(判定:C)とした。
・起泡性
泡状消毒液を100mLフォーマー容器(吐出量;0.8±0.5g)に充填し、300mLのメスシリンダーに10プッシュ吐出し、1分間静置した後の泡の体積を測定した。判定は下記の基準で行い、AA及びAを合格とした。
(評点) : (評価)
AA : 泡の体積が90mL以上
A : 泡の体積が80mL以上~90mL未満
B : 泡の体積が60mL以上~80mL未満
C : 泡の体積が60mm未満
調製した泡状消毒液40gを50mLのスクリュー管に充填し、密閉した後、5℃の恒温槽で1ヶ月間保存した。試験後の試料の性状を確認し、以下の基準で評価を行った。
○ : 試料に外観の変化が確認されない。
× : 沈殿の発生など、試料の外観に変化が確認される。
上記のAssociation of Official Agricultural Chemists (AOAC)のサニタイザー試験法に準じて行った。
これに対し、表3の比較例中、比較例1は[a/b]が4/1を超える値であるため、起泡性、除菌効果、及び防腐効果が劣り、比較例2はa/bが1/2より低い値であるため、起泡性及び低温安定性が劣る。比較例3は[a+b]が0.1より小さい値であるため、起泡性、除菌効果、及び防腐効果が劣る。比較例4~6はa成分が配合されず、代替品として、a成分とは脂肪酸の炭素数が異なるソルビタン脂肪酸エステル、ポリグリセリンモノ脂肪酸エステル、ショ糖モノ脂肪酸エステルを配合しているため、起泡性、低温安定性、除菌効果、及び防腐効果のいずれかの性能が劣り、比較例7~10は、b成分が配合されず、代替品として、モノグリセライドの含有量、1-モノグリセライドの質量分率、あるいは脂肪酸の炭素数が本発明の規定から外れるか、非グリセライドを代替品として使用しているため、起泡性、低温安定性、除菌効果、及び防腐効果のいずれかの性能が劣る結果となった。
表4の比較例11~18も、本発明のa成分及びb成分の両方を含まない(比較例11)か、a成分又はb成分の一方を含まず(比較例12、13)、あるいは、a成分又はb成分の代替品を使用している(比較例14~18)ため、起泡性、低温安定性、除菌効果、及び防腐効果のいずれかの性能が劣る結果となった。
Claims (1)
- 下記(a)、(b)及び(c)を含有し、組成物全量に対するa成分とb成分の含有量の和[a+b]が0.1~30質量%であり、a成分とb成分の質量比[a/b]が1/2~4/1であり、組成物全量に対するc成分の含有量が60~99.5質量%である、除菌洗浄剤組成物。
(a)ポリオキシエチレンモノラウリン酸ソルビタン
(b)アシル基の炭素数が8~12である脂肪酸グリセライドであって、b成分全量に対するモノグリセライドの含有量が85質量%以上であり、かつ、モノグリセライド全量に対する1-モノグリセライドの質量分率が0.9~1.0である脂肪酸グリセライド
(c)水及び/又はエタノール
