Claims (36)
- acyl coenzyme A dehydrogenase를 코딩하는 유전자 및 DNA-binding transcriptional dual regulator를 코딩하는 유전자가 결실 또는 약화되어 있고, fatty acyl-acp를 free fatty acid로 전환시키는 효소를 코딩하는 유전자, free fatty acid를 fatty acyl-CoA로 전환시키는 효소를 코딩하는 유전자, fatty acyl-CoA를 fatty aldehyde로 전환시키는 효소를 코딩하는 유전자 및 fatty aldehyde를 alkane으로 전환시키는 효소를 코딩하는 유전자가 도입 또는 증폭되어 있는 것을 특징으로 하는 알칸, 알켄, 알킨 및 방향족 탄화수소로 구성된 군에서 선택되는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, acyl-CoA synthetase를 코딩하는 유전자의 native promoter 및 long-chain fatty acid outer membrane transporter를 코딩하는 유전자의 attenuator를 포함하는 native promoter가 strong promoter로 추가적으로 치환되어 있고, short-chain fatty acid outer membrane transporter를 코딩하는 유전자가 추가적으로 도입되어 있는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, fabDHG operon의 native promoter 및 fabA operon의 native promoter가 strong promoter로 추가적으로 치환되어 있는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제 1항에 있어서, acetate kinase A 효소를 코딩하는 유전자(ackA)가 추가적으로 결실되어 있는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 미생물은 박테리아, 효모 및 곰팡이로 구성된 군에서 선택되는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제 5항에 있어서, 상기 박테리아는 지방산 대사회로를 갖는 박테리아인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제 6항에 있어서, 상기 지방산 대사회로를 갖는 박테리아는 코리네박테리움 (Corynebacterium) 속, 브레비박테리움(Brevibacterium) 속 및 대장균으로 구성된 군에서 선택되는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 acyl coenzyme A dehydrogenase를 코딩하는 유전자는 fadE 유전자인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 DNA-binding transcriptional dual regulator를 코딩하는 유전자는 fadR 유전자인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 fatty acyl-acp를 free fatty acid로 전환시키는 효소는 acyl-CoA thioesterase 인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 free fatty acid를 fatty acyl-CoA로 전환시키는 효소는 Escherichia coli 유래의 acetyl-CoA : acetoacetyl-CoA transferase 또는 acyl-coA synthetase 인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 fatty acyl-CoA를 fatty aldehyde로 전환시키는 효소는 fatty acyl-CoA reductase인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 fatty aldehyde를 alkane으로 전환시키는 효소는 fatty aldehyde decarbonylase인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제2항에 있어서, 상기 acyl-CoA synthetase를 코딩하는 유전자는 fadD 이고, short-chain fatty acid outer membrane transporter를 코딩하는 유전자는 atoE인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제2항에 있어서, 상기 long-chain fatty acid outer membrane transporter를 코딩하는 유전자는 fadL 인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 fatty acyl-acp를 free fatty acid로 전환시키는 효소를 코딩하는 유전자는 tesA(acyl-CoA thioesterase 를 코딩하는 유전자)인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 free fatty acid를 fatty acyl-CoA로 전환시키는 효소를 코딩하는 유전자는 atoDA(acetyl-CoA : acetoacetyl-CoA transferase를 코딩하는 유전자) 또는 fadD(acyl-coA synthetase를 코딩하는 유전자)인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 fatty acyl-CoA를 fatty aldehyde로 전환시키는 효소를 코딩하는 유전자는 luxC(long chain fatty acyl-CoA reductase를 코딩하는 유전자)인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항에 있어서, 상기 fatty aldehyde를 alkane으로 전환시키는 효소를 코딩하는 유전자는 CER1(fatty aldehyde decarbonylase 1을 코딩하는 유전자)인 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- 제1항 내지 제19항 중 어느 한 항에 있어서, 상기 탄화수소는 펜타데칸, 헵타데칸, 옥테인, 노네인 및 노닌으로 이루어진 군에서 선택된 것을 특징으로 하는, 탄화수소 생성능을 가지는 변이 미생물.
- 제2항 또는 제3항에 있어서, 상기 strong promoter는 trc promoter, tac promoter, T7 promoter, lac promoter 및 trp promoter로 구성된 군에서 선택되는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물.
- acyl coenzyme A dehydrogenase를 코딩하는 유전자(fadE) 및 DNA-binding transcriptional dual regulator를 코딩하는 유전자(fadR)가 결실 또는 약화되어 있고, acyl-CoA synthetase를 코딩하는 유전자(fadD)의 native promoter 및 long-chain fatty acid outer membrane transporter를 코딩하는 유전자(fadL)의 attenuator를 포함하는 native promoter가 strong promoter로 치환되어 있으며, fatty acyl-acp를 free fatty acid로 전환시키는 효소(acyl-CoA thioesterase), free fatty acid를 fatty acyl-CoA로 전환시키는 효소, fatty acyl-CoA를 fatty aldehyde로 전환시키는 효소(fatty acyl-CoA reductase) 및 fatty aldehyde를 alkane으로 전환시키는 효소(fatty aldehyde decarbonylase)를 코딩하는 유전자가 도입 또는 증폭되어 있는 알칸, 알켄, 알킨 및 방향족 탄화수소로 구성된 군에서 선택되는 탄화수소 생성능을 가지는 변이 대장균.

- 제22항 또는 제23항에 있어서, 상기 탄화수소는 펜타데칸, 헵타데칸, 옥테인, 노네인 및 노닌으로 이루어진 군에서 선택된 것을 특징으로 하는, 탄화수소 생성능을 가지는 변이 대장균.

- 제25항에 있어서, acyl-CoA synthetase를 코딩하는 유전자의 native promoter 및 long-chain fatty acid outer membrane transporter를 코딩하는 유전자의 attenuator를 포함하는 native promoter를 strong promoter로 추가적으로 치환시키고, short-chain fatty acid outer membrane transporter를 코딩하는 유전자를 추가적으로 도입시키는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물의 제조방법.
- 제25항에 있어서, fabDHG operon의 native promoter 및 fabA operon의 native promoter를 strong promoter로 추가적으로 치환시키는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물의 제조방법.
- 제 25항에 있어서, acetate kinase A 효소를 코딩하는 유전자(ackA)를 추가적으로 결실시키는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물의 제조방법.

- 제 30항에 있어서, 상기 지방산 대사회로를 갖는 박테리아는 코리네박테리움 (Corynebacterium) 속, 브레비박테리움(Brevibacterium) 속 및 대장균으로 구성된 군에서 선택되는 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물의 제조방법.
- 제25항에 있어서, 상기 탄화수소는 펜타데칸, 헵타데칸, 옥테인, 노네인 및 노닌으로 이루어진 군에서 선택된 것을 특징으로 하는 탄화수소 생성능을 가지는 변이 미생물의 제조방법.
- 제1항 내지 제19항 중 어느 한 항의 탄화수소 생성능을 가지는 변이 미생물을 배양하여 알칸, 알켄, 알킨 및 방향족 탄화수소로 구성된 군에서 선택되는 탄화수소를 생성시킨 다음, 배양액으로부터 탄화수소를 회수하는 것을 특징으로 하는 알칸, 알켄, 알킨 및 방향족 탄화수소로 구성된 군에서 선택되는 탄화수소의 제조방법.
- 제33항에 있어서, 상기 탄화수소는 펜타데칸, 헵타데칸, 옥테인, 노네인 및 노닌으로 이루어진 군에서 선택된 것을 특징으로 하는 탄화수소의 제조방법.
- 제22항 또는 제 23항의 탄화수소 생성능을 가지는 변이 대장균을 배양하여 알칸, 알켄, 알킨 및 방향족 탄화수소로 구성된 군에서 선택되는 탄화수소를 생성시킨 다음, 배양액으로부터 탄화수소를 회수하는 것을 특징으로 하는 알칸, 알켄, 알킨 및 방향족 탄화수소로 구성된 군에서 선택되는 탄화수소의 제조방법.
- 제35항에 있어서, 상기 탄화수소는 펜타데칸, 헵타데칸, 옥테인, 노네인 및 노닌으로 이루어진 군에서 선택된 것을 특징으로 하는 탄화수소의 제조방법.
