도 2는 본 발명의 변이 대장균 제조를 위한 발현 벡터의 개열지도를 나타낸 것이다.
도 3은 본 발명의 변이 대장균에서 생산된 카프로산의 양을 그래프로 나타낸 것이다.
Claims (5)
- (1) 야생형 대장균의 fadE 및 fadR 유전자를 넉 아웃(knock out)시킨 후, pox2 및 tesB 유전자를 포함하는 발현 벡터로 상기 대장균을 형질 전환시켜 변이 대장균을 제조하는 단계; 및
(2) 상기 변이 대장균을 배양하고 휴지 단계의 세포를 수확하고, 배지에 C10, C12, C14, C16, C18, C18:1 및 C18:2로 이루어진 군으로부터 선택되는 포화 지방산 또는 불포화 지방산을 첨가하는 단계를 포함하는, 변이 대장균에서 베타 산화를 통해 카프로산을 생산하는 방법.
- 제1항에 있어서,
상기 pox2 및 tesB 유전자가 각각 서열번호 9 및 10으로 표시되는 염기서열로 구성되는 것을 특징으로 하는 방법.
- 제1항에 있어서,
상기 발현 벡터가 도 2에 도시된 개열지도를 갖는 것을 특징으로 하는 방법.
- 야생형 대장균의 fadE 및 fadR 유전자가 넉 아웃되고, pox2 및 tesB 유전자를 발현하여 베타 산화를 통해 카프로산을 생산하는 변이 대장균.
- 제4항에 있어서,
상기 pox2 및 tesB 유전자가 각각 서열번호 9 및 10으로 표시되는 염기서열로 구성되는 것을 특징으로 하는 변이 대장균.
