Abstract
Description
〔1〕 可溶性トロンボモジュリンをコードする塩基配列を包含するDNAを宿主細胞にトランスフェクトして得られた形質転換細胞にて産生される可溶性トロンボモジュリンであって、該宿主細胞由来たん白質の含有率が、可溶性トロンボモジュリン10,000Uに対して10ng未満の比率である高純度可溶性トロンボモジュリン。
〔1-2〕 高純度可溶性トロンボモジュリンが、総たん白質中純度99%以上である前記〔1〕に記載の高純度可溶性トロンボモジュリン。
〔1-3〕 高純度可溶性トロンボモジュリンが水溶液の形態である前記〔1〕
又は〔1-2〕に記載の高純度可溶性トロンボモジュリン。
〔1-4〕 高純度可溶性トロンボモジュリン水溶液中の可溶性トロンボモジュリン濃度が8mg/mL以上の濃度である前記〔1-3〕に記載の高純度可溶性トロンボモジュリン。
なお、前記〔1〕~〔1-4〕のように引用する項番号が範囲で示され、その範囲内に〔1-2〕等の枝番号を有する項が配置されている場合には、〔1-2〕等の枝番号を有する項も引用されることを意味する。以下においても同様である。
(a)可溶性トロンボモジュリンをコードする塩基配列を包含するDNAを宿主細胞にトランスフェクトして得られた形質転換細胞、又はその宿主細胞のいずれかを無血清培養して得られる培養上清から宿主細胞由来たん白質を調製する工程、
(b)上記(a)の該宿主細胞由来たん白質をウサギに感作して得られる抗血清から抗宿主細胞由来たん白質抗体を精製する工程、
(c1)上記(b)の該抗宿主細胞由来たん白質抗体を固相に吸着させる工程、
(c2-1)該抗宿主細胞由来たん白質抗体を吸着させた該固相に、該宿主細胞由来たん白質の混入が疑われる可溶性トロンボモジュリン含有被検溶液を接触させる工程、及び該抗宿主細胞由来たん白質抗体を吸着させた該固相に濃度が既知の宿主細胞由来たん白質含有溶液を接触させる工程、
(c3)該固相にビオチン化した抗宿主細胞由来たん白質抗体を添加する工程、
(c4)該固相にアビジン化したペルオキシダーゼ溶液を添加する工程、
(c5)酵素基質溶液を添加し、発色させる工程、及び
(c6)発色を停止し、その吸光度を測定する工程、
を含む測定系を構築する工程、
(d)上記測定系にて該宿主細胞由来たん白質の混入が疑われる可溶性トロンボモジュリン含有被検溶液の宿主細胞由来たん白質濃度を測定し、該可溶性トロンボモジュリン含有被検溶液の宿主細胞由来たん白質濃度が、上記測定系にて濃度が既知の宿主細胞由来たん白質含有溶液を用いて測定することにより予め確認した上記測定系における定量可能範囲におさまるか否かを判断する工程、
(e-1)上記(d)において、該宿主細胞由来たん白質の混入が疑われる可溶性トロンボモジュリン含有被検溶液中の宿主細胞由来たん白質濃度が定量可能範囲におさまる場合、該濃度を該宿主細胞由来たん白質濃度とする工程、
(e-2-1)上記(d)において、該宿主細胞由来たん白質の混入が疑われる可溶性トロンボモジュリン含有被検溶液中の宿主細胞由来たん白質濃度が定量可能範囲におさまらない場合、上記測定系における定量可能範囲におさまる測定可能な濃度とするために必要に応じて可溶性トロンボモジュリン含有被検溶液を濃縮又は希釈し、その濃縮率又は希釈率を記録する工程、
(e-2-2)上記(e-2-1)において濃縮又は希釈した可溶性トロンボモジュリン含有被検溶液中の宿主細胞由来たん白質濃度を、上記(c1)~(c6)で示される測定系のうち(c2-1)を以下に示す(c2-2)に置き換えた測定系にて測定し、濃縮率又は希釈率を考慮して該宿主細胞由来たん白質濃度を求める工程、
(c2-2)該抗宿主細胞由来たん白質抗体を吸着させた該固相に、必要に応じて濃縮又は希釈した可溶性トロンボモジュリン含有被検溶液を接触させる工程、及び該抗宿主細胞由来たん白質抗体を吸着させた該固相に濃度が既知の宿主細胞由来たん白質含有溶液を接触させる工程、及び
(f)別途測定した可溶性トロンボモジュリン含有被検溶液単位体積あたりのトロンボモジュリンのAPC活性に対する、(e-1)又は(e-2-2)で求めた宿主細胞由来たん白質濃度の割合を算出する工程。
〔2-3〕 前記〔2-2〕(a)における宿主細胞がチャイニーズハムスター卵巣細胞DXB11株である前記〔2-2〕に記載の高純度可溶性トロンボモジュリン。
〔4〕 該可溶性トロンボモジュリンが、下記の(1)~(5)の性質を有する可溶性トロンボモジュリンである前記〔1〕~〔3〕のいずれかに記載の高純度可溶性トロンボモジュリン;
(1)トロンビンと選択的に結合する作用、
(2)トロンビンによるプロテインCの活性化を促進する作用、
(3)トロンビンによる凝固時間を延長する作用、
(4)トロンビンによる血小板凝集を抑制する作用、及び
(5)抗炎症作用。
〔4-2〕 該可溶性トロンボモジュリンが、下記の(1)~(4)の性質を有する可溶性トロンボモジュリンである前記〔1〕~〔3〕のいずれかに記載の高純度可溶性トロンボモジュリン;
(1)トロンビンと選択的に結合する作用、
(2)トロンビンによるプロテインCの活性化を促進する作用、
(3)トロンビンによる凝固時間を延長する作用、及び
(4)トロンビンによる血小板凝集を抑制する作用。
〔6〕 該高純度可溶性トロンボモジュリンが以下の工程を含む方法により製造される前記〔1〕~〔5〕のいずれかに記載の高純度可溶性トロンボモジュリン;
(a)可溶性トロンボモジュリンをコードする塩基配列を包含するDNAを宿主細胞にトランスフェクトして形質転換細胞を取得する工程、
(b)該形質転換細胞を培養して可溶性トロンボモジュリンを含む溶液を取得する工程、及び
(c)該可溶性トロンボモジュリンを含む溶液をナイロン及び/又はポリエーテルスルホンに接触させ、宿主細胞由来たん白質の含有率が可溶性トロンボモジュリン10,000Uに対して10ng未満の比率である高純度可溶性トロンボモジュリンを得る工程。

また、配列番号9もしくは配列番号11のそれぞれにおける第514位、第515位、及び第516位で終わるペプチドの混合割合としては、(0:0:100)~(0:90:10)が例示され、場合によっては、(0:70:30)~(10:90:0)、(10:0:90)~(20:10:70)が例示される。
これらペプチドの混合割合は、通常の方法により求めることができる。
さらに、配列番号1、3、5、7、9、及び11のそれぞれにおける第1~18位の配列は、全て同一の配列である。
動物細胞としては、チャイニーズハムスター卵巣(CHO)細胞、COS-1細胞、COS-7細胞、VERO(ATCC CCL-81)細胞、BHK細胞、イヌ腎由来MDCK細胞、ハムスターAV-12-664細胞、NS0細胞等が、またヒト由来細胞としてHeLa細胞、WI38細胞、ヒト293細胞、PER.C6細胞が挙げられる。CHO細胞が極めて一般的であり好ましく、CHO細胞においては、ジヒドロ葉酸還元酵素(DHFR)欠損CHO細胞がさらに好ましい。
本実施の形態の高純度可溶性トロンボモジュリンとしては、可溶性トロンボモジュリン以外のたん白質を実質的に含まない高い純度の可溶性トロンボモジュリンが挙げられる。具体的には、例えば、HCPを実質的に含まない可溶性トロンボモジュリンが挙げられる。好ましくは、HCP、マウスIgG、及びウシ血清たん白質を実質的に含まない可溶性トロンボモジュリンが挙げられる。
本実施の形態の高純度可溶性トロンボモジュリンはヒト由来のたん白質は含まない。
可溶性トロンボモジュリンを含む溶液をナイロン及び/又はポリエーテルスルホンに接触させる工程を含む(a)~(g)からなる製造工程。
(a)形質転換細胞を培養して培養液(生産液)を回収する工程、
(b)生産液をろ過してろ過後生産液を得る工程、
(c)ろ過後生産液を陰イオン交換カラムクロマトグラフィーに供し、粗精製液を得る工程、
(d)粗精製液を抗トロンボモジュリンモノクローナル抗体を担持させたアフィニティーカラムクロマトグラフィーに供し、精製液1を得る工程、
(e)精製液1を陽イオン交換カラムクロマトグラフィー工程に供し、精製液2を得る工程、
(f)濃縮した精製液2をゲルろ過カラムクロマトグラフィーに供し、その溶出液を濃縮して精製液3を得る工程、及び
(g)精製液3をウイルス除去膜及び無菌ろ過膜で濾過する工程。
また、ウシ血清たん白質の混入を0とするために、(a)の形質転換細胞の培養が無血清培養であることがより好ましい。
可溶性トロンボモジュリンを含む溶液をナイロン及び/又はポリエーテルスルホンに接触させる工程を含む(a)~(g)からなる製造工程。
(a)形質転換細胞を培養して培養液(生産液)を回収する工程、
(b)生産液をろ過してろ過後生産液を得る工程、
(c)ろ過後生産液を陰イオン交換カラムクロマトグラフィーに供し、粗精製液1を得る工程、
(d)粗精製液1を疎水カラムクロマトグラフィーに供し、粗精製液2を得る工程、
(e)粗精製液2を抗トロンボモジュリンモノクローナル抗体を担持させたアフィニティーカラムクロマトグラフィーに供し、精製液1を得る工程、

(g)精製液2を無菌ろ過膜で濾過する工程。
また、ウシ血清たん白質の混入を0とするために、(a)の形質転換細胞の培養が無血清培養であることがより好ましい。
ナイロン及び/又はポリエーテルスルホンに接触させる工程の後に存在してもよい精製工程としては、陰イオン交換カラムクロマトグラフィー、アフィニティカラムクロマトグラフィー、陽イオン交換カラムクロマトグラフィー、ゲルろ過カラムクロマトグラフィー、又は疎水カラムクロマトグラフィー等のカラムクロマトグラフィー工程、膜濃縮工程、ウイルス除去、無菌ろ過等の膜ろ過工程、又はこれら2種以上の組み合わせが挙げられ、陽イオン交換カラムクロマトグラフィー、ゲルろ過カラムクロマトグラフィー、膜濃縮、ウイルス除去、無菌ろ過、又はこれらの2種以上の組み合わせが好ましい。ナイロン及び/又はポリエーテルスルホンに接触させる工程の後に、陽イオン交換カラムクロマトグラフィー、ゲルろ過カラムクロマトグラフィー、膜濃縮、ウイルス除去、及び無菌ろ過が存在することが好ましい場合がある。また、ナイロン及び/又はポリエーテルスルホンに接触させる工程の後に、陽イオン交換カラムクロマトグラフィー、ゲルろ過カラムクロマトグラフィー、膜濃縮、ウイルス除去、及び無菌ろ過の精製工程が、陽イオン交換カラムクロマトグラフィー、膜濃縮、ゲルろ過カラムクロマトグラフィー、膜濃縮、ウイルス除去、無菌ろ過の順番で存在することがより好ましい例として挙げられる。なお、陽イオン交換カラムクロマトグラフィーとしては、SP Sepharose Fast Flow、DEAE Sepharose Fast Flow、Capto S、Capto DEAE(GEヘルスケア社)、S HyperCel(ポール社)、TOYOPEARL GigaCap S-650(東ソー社)が例示され、SP Sepharose Fast Flowが好ましい。濃縮膜としては、MICROZA UF(旭化成ケミカルズ社)、Kvick Flow 10KD(GEヘルスケア社)、Pellicon 2 (ミリポア社)が例示され、MICROZA UFが好ましい。ゲルろ過カラムクロマトグラフィーとしては、Sephacryl S-300 HR、Superose 12 pg(GEヘルスケア社)、TOYOPEARL HW(東ソー)が例示され、Sephacryl S-300 HRが好ましい。ウイルス除去膜としては、PLANOVA 15N(旭化成メディカル)、Biresolve NFP(ミリポア社)、Ultipor VF(ポール社)が例示され、PLANOVA 15Nが好ましい。無菌ろ過膜としては、Millipak、Millidisk(ミリポア社)、Supore EVA(ポール社)、Sartopore 2(サルトリウス ステディム社)が例示され、Millipakが好ましい。

(e-1)上記(d)において、該宿主細胞由来たん白質の混入が疑われる可溶性トロンボモジュリン含有被検溶液中の宿主細胞由来たん白質濃度が定量可能範囲におさまる場合、該濃度を該宿主細胞由来たん白質濃度とする工程、

HCPは、可溶性トロンボモジュリンを産生する遺伝子組換え細胞を作製する際に用いた宿主細胞由来のたん白質であり、少なくとも上記(a)~(f)の工程を含む方法にて測定されるものとして定義されるが、HCPの構成要素としては、試験例6に示すようにヒストンH2B(Biochimie, 61(1), 61-69(1979))が挙げられる。
HCP濃度測定系を構築する際には、その定量可能範囲を明確にする必要があり、トロンボモジュリン10,000U当たりのHCP含量が10ng未満であることが測定できるのであれば、定量可能範囲は限定されないが、低い濃度まで測定できるほどよい。定量可能範囲としては、下限として100ng/mL以上が例示され、50ng/mL以上が好ましく、25ng/mL以上がより好ましく、上限として500ng/mLが例示される。
トロンボモジュリン10,000U当たりのHCP含量は、以下の式に従って算出される。

Biologicals(2002)30,69-76に従い、プロテインCの活性化を指標としたトロンボモジュリンのAPC活性を測定する。
2 0mM塩化カルシウム溶液75μLに0.05%ポリソルベート20を含むトリス・イミダゾール緩衝液で希釈した試料溶液25μLを添加し氷冷した後、40U/mLのヒトトロンビン(米国、シグマ)溶液25μLを添加撹拌し、37℃で加温する。ヒトトロンビン溶液を添加してから10分後に12U/mLのヒトプロテインC(米国、エンザイムリサーチ)溶液25μLを添加撹拌し、37℃で加温する。ヒトプロテインC溶液を添加してから10分後に、ヘパリン-アンチトロンビンIII溶液100μLを添加撹拌し、37℃で加温する。ヘパリン-アンチトロンビンIII溶液を添加してから10分後に、あらかじめ37℃に加温した合成基質S-2366(スウェーデン国、クロモジェニックス)溶液250μLを添加撹拌し、37℃で加温する。基質溶液を添加してから10分後に、50%酢酸1.5mLを添加攪拌し、水を対照として405nmにおける吸光度を測定する。

B液:2-アミノ-2-ヒドロキシメチル-1,3-プロパンジオール4.04g、イミダゾール2.27g及び塩化ナトリウム1.95gを水に溶解し100mLとし、塩化ナトリウム11.7g加えて溶解する。

60mM塩化カルシウム溶液1mLにトリス・イミダゾール緩衝液2mLを加える。
<ヘパリン-アンチトロンビンIII溶液>
2U/mLのアンチトロンビンIII(日本国、三菱ウェルファーマ)溶液7.5μL、トリス・イミダゾール緩衝液42.5μL及び30U/mLのヘパリン(日本国、持田製薬)溶液50μLを加えて振り混ぜる。用時調製し、使用直前まで氷冷する。
トロンボモジュリン遺伝子を導入した遺伝子組換えCHO細胞を無血清培養する。培養上清を抗トロンボモジュリン抗体カラムに供し非吸着画分を得る。この非吸着画分について参考例1に従ってトロンボモジュリンのAPC活性を測定し、活性が検出されないことを確認した後、限外ろ過膜にて濃縮したものをHCPとする。HCPを抗原としてウサギに感作して得た抗HCP抗血清を硫安塩析及びプロテインAカラムにより精製した後、トロンボモジュリンをリガンドとしたアフィニティカラムに供し非吸着画分を得る。このようにしてトロンボモジュリンを認識しないウサギ抗HCP抗体を得る。

<酵素基質溶液>
オルト-フェニレンジアミン二塩酸塩10mgをクエン酸・リン酸塩緩衝液(クエン酸一水和物2.56g及びリン酸水素二ナトリウム十二水和物9.12gを水に溶解し500mLとする)20mLに加えて溶解し、使用する直前に過酸化水素水10μLを加える。
マウスIgGを抗原としてウサギに感作して得た抗マウスIgG抗血清を硫安塩析及びプロテインAカラムにより精製し、ウサギ抗マウスIgG抗体を得る。
予想されるマウスIgG濃度が0~25ng/mLとなるように0.05%ポリソルベート80を含むPBSで希釈し、試料溶液とする。なお、試料溶液のIgG濃度が低い場合は、限外ろ過膜等を使用して適切な濃度まで濃縮する。別にマウスIgGに0.05%ポリソルベート80を含むPBSを加えて、その1mL中に25、20、15、10、5、2.5、1.25、0.63及び0ngを含む9種類の液を調製し、標準溶液とする。

特開平11-341990号公報の実施例1に従って、配列番号9に記載のアミノ酸配列をコードするDNAを遺伝子操作技術により組み込んだ遺伝子組換えCHO細胞を作製した後、150mg/LのL-プロリン(日本国、味の素)、60mg/Lのカナマイシン硫酸塩(日本国、明治製菓)、1mg/Lの酒石酸タイロシン(日本国、メルシャン)、及び10%ウシ血清(米国、ハイクロン)を含むDMEM培地(米国、インビトロジェン)に播種し、CO2インキュベーター内、37℃で培養した。培養液を遠心分離して得られた細胞を150mg/LのL-プロリン(日本国、味の素)、60mg/Lのカナマイシン硫酸塩(日本国、明治製菓)、1mg/Lの酒石酸タイロシン(日本国、メルシャン)、10%ジメチルスルフォキシド(独国、メルク)、及び10%ウシ血清(米国、ハイクロン)を含むDMEM培地(米国、インビトロジェン)にけん濁した後、バイアルに分注し(4×107 cells/本)、液体窒素中に凍結保存した。
同様の操作により、3ロット(A1、A2、A3)の精製品を取得した。
A1、A2、A3のトロンボモジュリンのAPC活性は、それぞれ69000U/mL、68000U/mL、72000U/mLであった。
A1、A2、A3の溶液における可溶性トロンボモジュリン濃度は、それぞれ10.5mg/mL、10.2mg/mL、10.3mg/mLであった。
特開平11-341990号公報の成分表2に示された培地を基本培地として使用した。増殖培地は基本培地に60mg/Lのカナマイシン硫酸塩(米国、インビトロジェン)、1mg/Lの酒石酸タイロシン(日本国、塩野義製薬)、及び8%ウシ血清(米国、ハイクロン)を添加して用いた。また、生産培地は増殖培地の血清濃度を4%とした。
回収した生産液は、孔径0.7μm及び0.22μmのろ過フィルター(米国、ポール)を用いて澄明化し、ろ過後生産液として2~10℃で保存した。
B1のトロンボモジュリンのAPC活性は、79000U/mLであった。
B1の溶液における可溶性トロンボモジュリン濃度は、12.6mg/mLであった。
DMEM培地(米国、インビトロジェン)を基本培地として使用した。増殖培地は基本培地に150mg/LのL-プロリン(日本国、味の素)、60mg/Lのカナマイシン硫酸塩(日本国、明治製菓)、1mg/Lの酒石酸タイロシン(日本国、メルシャン)、及び10%ウシ血清(米国、ハイクロン)を添加して用いた。また、生産培地は増殖培地の血清濃度を1~3%とした。
B2のトロンボモジュリンのAPC活性は、28000U/mLであった。
B2の溶液における可溶性トロンボモジュリン濃度は、3.8mg/mLであった。
比較例1で凍結保存された細胞のバイアルを1本融解し、8mMのL-グルタミン(米国、インビトロジェン)、50μMのヒポキサンチン(米国、インビトロジェン)、及び8μMのチミジン(米国、インビトロジェン)を含む無血清培地IS CHO-CD(米国、アーバイン サイエンティフィック)に播種し、CO2インキュベーター内、37℃で培養した。培養液を遠心分離して得られた細胞を8mMのL-グルタミン(米国、インビトロジェン)、50μMのヒポキサンチン(米国、インビトロジェン)、8μMのチミジン(米国、インビトロジェン)、及び10%ジメチルスルフォキシド(米国、シグマ-アルドリッチ)を含む無血清培地IS CHO-CD(米国、アーバイン サイエンティフィック)にけん濁した後、バイアルに分注し(2×107 cells/本)、液体窒素中に凍結保存した。
約250Lのろ過後生産液を、150mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で平衡化したQ-Sepharose Fast Flow(米国、GEヘルスケア)カラム(直径25cm、高さ25cm)に供した。次に6CVの180mM 塩化ナトリウムを含む20mM酢酸緩衝液(pH5.6)で洗浄し、更に4CVの180mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で洗浄を行ない、290mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で溶出を開始し、溶出液の吸光度280nmのピーク立ち上がりからの0.5カラムボリューム容量の溶出液を粗精製液として取得した。なお、温度は2~10℃、クロマト流速は18L/時で実施した。

A4のトロンボモジュリンのAPC活性は、60000U/mLであった。
A4の溶液における可溶性トロンボモジュリン濃度は、9.3mg/mLであった。
実施例1で取得した約2,000Lのろ過後生産液を、150mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で平衡化したQ-Sepharose Fast Flow(米国、GEヘルスケア)カラム(直径63cm、高さ25cm)に供した。次に6CVの170mM 塩化ナトリウムを含む20mM酢酸緩衝液(pH5.45)で洗浄し、更に4CVの170mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で洗浄を行ない、300mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で溶出を開始し、溶出液の吸光度280nmのピーク立ち上がりからの0.5カラムボリューム容量の溶出液を粗精製液として取得した。同様の操作を2回繰り返し実施し、2ロットの粗精製液を取得した。なお、温度は2~10℃、クロマト流速は109L/時で実施した。
A5のトロンボモジュリンのAPC活性は、69000U/mLであった。
A5の溶液における可溶性トロンボモジュリン濃度は、10.9mg/mLであった。

A6のトロンボモジュリンのAPC活性は、81000U/mLであった。
A6の溶液における可溶性トロンボモジュリン濃度は、11.9mg/mLであった。
増殖培地は20.78gのIS CHO-CD-A3(米国、アーバイン サイエンティフィック)、4.06gの塩化ナトリウム(日本国、富田製薬)、及び2.20gの炭酸水素ナトリウム(日本国、和光純薬工業)を1Lの水に溶解して作製した。また、生産培地は1Lの水に対し、20.78gのIS CHO-CD-A3(米国、アーバイン サイエンティフィック)、2.63gの塩化ナトリウム(日本国、富田製薬)、及び4.40gの炭酸水素ナトリウム(日本国、和光純薬工業)を溶解して作製した。
約1,400Lのろ過後生産液を、150mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で平衡化したQ-Sepharose Fast Flow(米国、GEヘルスケア)カラム(直径63cm、高さ25cm)に供した。次に6CVの180mM 塩化ナトリウムを含む20mM酢酸緩衝液(pH5.6)で洗浄し、更に4CVの180mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で洗浄を行ない、290mM 塩化ナトリウムを含む20mMトリス塩緩衝液(pH7.7)で溶出を開始し、溶出液の吸光度280nmのピーク立ち上がりからの0.5カラムボリューム容量の溶出液を粗精製液として取得した。約900Lのろ過後生産液についても同様の操作を行い、2ロットの粗精製液を取得した。なお、温度は2~10℃、クロマト流速は109L/時で実施した。
A7のトロンボモジュリンのAPC活性は、69000U/mLであった。
A7の溶液における可溶性トロンボモジュリン濃度は、10.4mg/mLであった。
比較例1で得られた精製液1(HCP濃度:462ng/ml)を異なる材質のろ過膜に通液した後のHCP濃度を比較した。すなわち、(1)PVDF(ポリフッ化ビニリデン)製(米国、ミリポア、Millex GV)、(2)CA(セルロースアセテート)製(独国、ザルトリウス、Minisart)、(3)PES(ポリエーテルスルホン)製(独国、ザルトリウス、Minisart High-Flow)、(4)ナイロン製(米国、サーモフィッシャサイエンティフィック、NALGENE Syringe Filter)、(5)CA+GF(セルロースアセテート+グラスファイバー)製(ザルトリウス、Minisart Plus)の各ろ過膜に5mlの精製液1を1ml/分の流速で通液し、全量をろ過液として回収した。
試験例1において、ナイロン製ろ過膜とPES製ろ過膜は高いHCP除去能を有することが判明したため、これらのろ過膜について通液量によるHCP除去能の変化を評価した。なお、ろ過膜は各材質について2種類のメーカーの商品を準備し、通液する精製液1については、試験例2で使用したロットとは異なるロットを用いた(HCP濃度:303ng/ml)。ろ過膜は、コントロールとしてPVDF製(米国、ミリポア、Millex GV:孔径0.22μm、膜径25mm、有効膜面積3.9cm2)を使用し、ナイロン製として(1)米国、ポール社のAcrodisc AP-4436T、:孔径0.2μm、膜径25mm、有効膜面積3.9cm2、及び(2)独国、ザルトリウス社のMinisart NY25:孔径0.2μm、膜径25mm、有効膜面積4.8cm2を、PES製として(3)米ポール社のAcrodisc PN4612:孔径0.2μm、膜径25mm、有効膜面積2.8cm2、及び(4)ザルトリウス社のMinisart High-Flow:孔径0.2μm、膜径26mm、有効膜面積5.3cm2を用いた。ろ過膜当りの精製液1の通液量は10ml/分の流速で100mlとし、20ml通液毎にろ過液をサンプリングして参考例2に示した方法に従ってHCP濃度を、280nmの吸収からたん白質濃度を測定した。
緩衝液組成及び可溶性トロンボモジュリン濃度の違いがナイロン製ろ過膜のHCP除去能に与える影響を評価した。比較例4で得られた精製液1、精製液2、精製液2濃縮後、精製液3、及び精製品について各5mLを膜径25mm、有効膜面積4.8cm2、孔径0.2μmのナイロン製ろ過膜(独国、ザルトリウス社、Minisart NY25)に1mL/分の流速で通液し、全量をろ過液として回収した。各サンプルに含まれるHCP1mgに対するろ過膜面積は、精製液1:0.77m2、精製液2:1.7m2、精製液2濃縮後:0.43m2、精製液3:0.72m2、精製品:0.81m2であった。得られたろ過液について、参考例2に示した方法に従ってHCP濃度を測定し、たん白質濃度を280nmの吸収から求めた。その結果、ナイロン製膜は全てのサンプルに対して高いHCP除去効果が認められ、たん白質濃度回収率は90%以上の高い値が得られた(表3)。
異なった製造方法により取得した可溶性トロンボモジュリンを、さらにナイロン製ろ過膜に通液することによりHCPが除去されるかどうか検討した。比較例1~3で取得した可溶性トロンボモジュリン精製品(それぞれ、A1、B1及びB2)について各5mLを膜径25mm、有効膜面積4.8cm2、孔径0.2μmのナイロン製ろ過膜(独国、ザルトリウス社、Minisart NY25)に1mL/分の流速で通液し、全量をろ過液として回収した。各可溶性トロンボモジュリン精製品に含まれるHCP1mgに対するろ過膜面積は各可溶性トロンボモジュリン精製品に含まれるHCP1mgに対するろ過膜面積は、A1:0.34m2、B1:0.46m2、B2:1.7m2であった。ろ過前後の液について、参考例2~4に示した方法に従ってHCP含量、マウスIgG含量及びウシ血清たん白質含量を測定した。全ての精製品においてナイロン製ろ過膜はHCPに対してのみ強い除去効果が認められた(表4)。このことからナイロン製ろ過膜はたん白質を非特異的に吸着するのではなく、HCPを特異的に吸着する作用を有すると考えられた。
ナイロン製ろ過膜未使用の工業化レベル製造(比較例1)及びナイロン製ろ過膜を使用した工業化レベル製造(実施例1~4)で得られたトロンボモジュリン精製品について、参考例2~4に示した方法に従ってHCP含量、マウスIgG含量及びウシ血清たん白質含量を測定した。
ナイロン製ろ過膜を通液していない比較例1の3ロット(A1、A2及びA3)は10ng/10,000U以上の高いHCP含量を有していたが、ナイロン製ろ過膜を通液した実施例1~4の4ロット(A4、A5、A6及びA7)のHCP含量は定量限界未満であった。その他の不純物のマウスIgG含量及びウシ血清たん白質含量についてはナイロン製ろ過膜の使用の有無でその含量に際は認められなかった(表5)。このように工業化レベルでの製造においてもナイロン製ろ過膜は、HCPに特異的な除去能を有し、HCP含量が10ng/10,000U未満という高純度可溶性トロンボモジュリンの製造を可能にした。
実施例1~4で得られた高純度可溶性トロンボモジュリンの総たん白質中純度をゲルろ過液体クロマトグラフィー及びイオン交換液体クロマトグラフィーにて測定した。ゲルろ過液体クロマトグラフィーについては、TOSOH TSKgel G3000SWXL(日本国、東ソー)を用いて、0.1M硫酸ナトリウムを含む50mMリン酸塩緩衝液(pH7.3)、温度40℃、流速0.9mL/分という条件下で測定を行った結果、全ての精製品(A4、A5、A6及びA7)の純度は99%以上であった。また、イオン交換液体クロマトグラフィーについては、TOSOH DEAE 5PW(日本国、東ソー)を用いて、50mM塩化ナトリウムを含む20mMピペラジン塩酸緩衝液(pH5.6)から350mM塩化ナトリウムを含む20mMピペラジン塩酸緩衝液(pH5.6)への30分間直線グラジエント溶出、温度40℃、流速0.9mL/分という条件下で測定を行った結果、全ての精製品(A4、A5、A6及びA7)の純度は99%以上であった。
さらに、日本薬局方一般試験法のエンドトキシン試験法<4.01>のゲル化法に従って試験を実施した結果、エンドトキシン含量が0.004~0.03EU/10,000Uであり、非常に低いレベルであった(表6)
実施例3と同様の方法により高純度可溶性トロンボモジュリンを製造した後、製造で使用したナイロン製ろ過膜(独国、ザルトリウス、SARTOLON Maxi Caps 5101307H3、孔径0.4μm+0.2μm、膜面積1.8m2)をハウジングから取り出し、膜を約3gの断片になるように切断した。5枚の断片を50mM 塩化ナトリウム含む20mMリン酸緩衝液(pH7.3)で十分洗浄した後、40mLの0.5%CHAPS、200mM 塩化ナトリウムを含む50mM トリス塩酸緩衝液(pH8.0)が入った試験管に断片を1枚ずつ入れた。室温で一晩振とうすることにより、膜に吸着したたん白質を緩衝液中に抽出した。抽出液全量を限外ろ過膜のVivaspin20(独国、ザルトリウス)とAmiconUltra-0.5mL(米国、ミリポア)を用いて、15μLになるまで濃縮した。この濃縮液のうち約1/5量をSDS-PAGE(日本国、アトー、e-PAGEL5/20)に供し、CBB染色(日本国、和光純薬工業、Quick-CBB)した。分子量10,000付近に認められたバンドを切り出して、そのゲル断片をジチオトレイトールで還元後、ヨードアセトアミドによりカルバミドメチル化し、トリプシンによる酵素消化を終夜で実施した。酵素消化物をLC/MS/MSに供し、得られたマススペクトルデータから、NCBIのデータベースを用いてMascot検索を行い、酵素消化物のアミノ酸配列を解析した。

Claims (22)
- 可溶性トロンボモジュリンをコードする塩基配列を包含するDNAを宿主細胞にトランスフェクトして得られた形質転換細胞にて産生される可溶性トロンボモジュリンであって、宿主細胞由来たん白質の含有率が可溶性トロンボモジュリン10,000Uに対して10ng未満の比率である高純度可溶性トロンボモジュリン。
- 可溶性トロンボモジュリンが該形質転換細胞を無血清培養することにより産生される可溶性トロンボモジュリンである請求項1に記載の高純度可溶性トロンボモジュリン。
- 該宿主細胞がチャイニーズハムスター卵巣細胞である請求項1又は2に記載の高純度可溶性トロンボモジュリン。
- 該可溶性トロンボモジュリンが、下記の(1)~(5)の性質を有する可溶性トロンボモジュリンである請求項1~3のいずれかに記載の高純度可溶性トロンボモジュリン;
(1)トロンビンと選択的に結合する作用、
(2)トロンビンによるプロテインCの活性化を促進する作用、
(3)トロンビンによる凝固時間を延長する作用、
(4)トロンビンによる血小板凝集を抑制する作用、及び

(c)該可溶性トロンボモジュリンを含む溶液をナイロン及び/又はポリエーテルスルホンに接触させ、宿主細胞由来たん白質の含有率が可溶性トロンボモジュリン10,000Uに対して10ng未満の比率である高純度可溶性トロンボモジュリンを得る工程。 - 該可溶性トロンボモジュリンが、
(i)配列番号9又は配列番号11のいずれかに記載のアミノ酸配列における第367~480位のアミノ酸配列を含み、かつ下記(ii-1)又は(ii-2)のいずれかのアミノ酸配列を含むペプチドであって、該ペプチドが下記の(1)~(5)の性質を有する可溶性トロンボモジュリンである請求項1~6のいずれかに記載の高純度可溶性トロンボモジュリン;

(5)抗炎症作用。 - 該可溶性トロンボモジュリンが、
(i-1)配列番号9又は配列番号11のいずれかに記載のアミノ酸配列における第19~516位のアミノ酸配列、又は
(i-2)上記(i-1)のアミノ酸配列の1つ又は複数のアミノ酸が置換、欠失、又は付加されたアミノ酸配列
のいずれかのアミノ酸配列を有するペプチドであって、該ペプチドが下記の(1)~(5)の性質を有する可溶性トロンボモジュリンである請求項1~6のいずれかに記載の高純度可溶性トロンボモジュリン;
(1)トロンビンと選択的に結合する作用、
(2)トロンビンによるプロテインCの活性化を促進する作用、
(3)トロンビンによる凝固時間を延長する作用、
(4)トロンビンによる血小板凝集を抑制する作用、及び
(5)抗炎症作用。 - 該可溶性トロンボモジュリンをコードする塩基配列を包含するDNAが、配列番号9又は配列番号11のいずれかに記載のアミノ酸配列をコードするDNAである請求項1~6のいずれかに記載の高純度可溶性トロンボモジュリン。
- 請求項1~9のいずれかに記載の高純度可溶性トロンボモジュリン及び薬学上許容される担体を含有する医薬組成物。
- 宿主細胞由来たん白質の含有率が可溶性トロンボモジュリン10,000Uに対して10ng未満の比率である高純度可溶性トロンボモジュリンの製造方法であって、可溶性トロンボモジュリンをコードする塩基配列を包含するDNAを宿主細胞にトランスフェクトして得られた形質転換細胞が産生する可溶性トロンボモジュリンを含む溶液をナイロン及び/又はポリエーテルスルホンに接触させる工程を含む製造方法。
- 該形質転換細胞を無血清培養して可溶性トロンボモジュリンを産生する請求項11に記載の製造方法。
- 該可溶性トロンボモジュリンが、下記の(1)~(5)の性質を有する可溶性トロンボモジュリンである請求項11又は12に記載の高純度可溶性トロンボモジュリンの製造方法;
(1)トロンビンと選択的に結合する作用、
(2)トロンビンによるプロテインCの活性化を促進する作用、
(3)トロンビンによる凝固時間を延長する作用、
(4)トロンビンによる血小板凝集を抑制する作用、及び
(5)抗炎症作用。 - 該宿主細胞がチャイニーズハムスター卵巣細胞である請求項11~13のいずれかに記載の製造方法。
- 該可溶性トロンボモジュリンの分子量が50,000から80,000である請求項11~14のいずれかに記載の製造方法。
- 該可溶性トロンボモジュリンが、
(i)配列番号9又は配列番号11のいずれかに記載のアミノ酸配列における第367~480位のアミノ酸配列を含み、かつ下記(ii-1)又は(ii-2)のいずれかのアミノ酸配列を含むペプチドであって、該ペプチドが下記の(1)~(5)の性質を有する可溶性トロンボモジュリンである請求項11~15のいずれかに記載の製造方法;
(ii-1)配列番号9又は配列番号11のいずれかに記載のアミノ酸配列における第19~244位のアミノ酸配列、又は
(ii-2)上記(ii-1)のアミノ酸配列の1つ又は複数のアミノ酸が置換、欠失、又は付加されたアミノ酸配列、
(1)トロンビンと選択的に結合する作用、
(2)トロンビンによるプロテインCの活性化を促進する作用、
(3)トロンビンによる凝固時間を延長する作用、
(4)トロンビンによる血小板凝集を抑制する作用、及び
(5)抗炎症作用。 - 該可溶性トロンボモジュリンが、
(i-1)配列番号9又は配列番号11のいずれかに記載のアミノ酸配列における第19~516位のアミノ酸配列、又は
(i-2)上記(i-1)のアミノ酸配列の1つ又は複数のアミノ酸が置換、欠失、又は付加されたアミノ酸配列
のいずれかのアミノ酸配列を有するペプチドであって、該ペプチドが下記の(1)~(5)の性質を有する可溶性トロンボモジュリンである請求項11~15のいずれかに記載の製造方法;
(1)トロンビンと選択的に結合する作用、
(2)トロンビンによるプロテインCの活性化を促進する作用、
(3)トロンビンによる凝固時間を延長する作用、
(4)トロンビンによる血小板凝集を抑制する作用、及び
(5)抗炎症作用。 - 該可溶性トロンボモジュリンをコードする塩基配列を包含するDNAが、配列番号9又は配列番号11に記載のアミノ酸配列をコードするDNAである請求項11~15のいずれかに記載の製造方法。
- ナイロン及び/又はポリエーテルスルホンの形態がろ過膜の形態である請求項11~18のいずれかに記載の製造方法。
- ろ過膜の膜面積が宿主細胞由来たん白質1mgに対して0.01から0.5m2である請求項19に記載の製造方法。
- ナイロン及び/又はポリエーテルスルホンがナイロンである請求項11~20のいずれかに記載の製造方法。
- 宿主細胞由来たん白質の含有率が可溶性トロンボモジュリン10,000Uに対して10ng未満の比率である高純度可溶性トロンボモジュリンの製造方法であって、
(a)配列番号9又は配列番号11に記載のアミノ酸配列をコードするDNAを宿主細胞にトランスフェクトして形質転換細胞を取得する工程、
(b)該形質転換細胞を培養して可溶性トロンボモジュリンを含む溶液を取得する工程、
(c)該可溶性トロンボモジュリンを含む溶液を総たん白質中トロンボモジュリン純度99%以上に精製する工程、及び
(d)該可溶性トロンボモジュリンを含む溶液をナイロンに接触させ、宿主細胞由来たん白質の含有率が可溶性トロンボモジュリン10,000Uに対して10ng未満の比率である高純度可溶性トロンボモジュリンを単離する工程
を含む製造方法であって、前記宿主細胞がチャイニーズハムスター卵巣細胞である請求項11~21のいずれかに記載の製造方法。
