光学分割法:3-置換グルタル酸モノアミドのラセミ体を光学活性メチルベンジルアミンや光学活性フェニルエチルアミンなどで処理してジアステレオ性の塩を形成し、分別結晶することで前記ラセミ体を光学分割する方法(特許文献1、非特許文献1など)。
ニトリルヒドラターゼ法:3-置換グルタロジニトリルの一方のニトリル基をニトリルヒドラターゼで不斉水解し、1)得られたモノニトリルモノカルボン酸をCurtius転位してから酸加水分解するか、2)モノニトリルカルボン酸のニトリル基を加水分解してからHoffmann転位する方法(非特許文献2)。
一方、ニトリルヒドラーゼ法では片方の光学異性体を捨てる必要はないものの、不斉水解の収率は67%程度であり、Curtius転位・酸加水分解の収率がそれぞれ43%、78%であるから、3工程でのトータル収率は22%と低い。また不斉水解後に、ニトリル基の酸加水分解・Hoffmann転位をする場合も、それぞれの収率は79%、71%であるから、3工程でのトータル収率は38%程度と低い。また不斉水解の立体選択性にも改善の余地があった。加えて、医薬中間体の原料として用いるには、更に精製を行う必要があった。
また光学活性3-置換グルタル酸モノアミドの簡便な光学純度向上法も知られていない。例えば、光学活性3-置換グルタル酸モノアミドを塩基性水溶液に溶解し、酸を加えて3-置換グルタル酸モノアミドを析出させても、その光学純度は全く向上しなかった。光学活性3-置換グルタル酸モノアミドの簡便な光学純度向上法が強く望まれる。
本発明の別の目的は、医薬品中間体として有用である高光学純度の光学活性3-置換グルタル酸モノアミドを高収率にて製造する方法を提供することにある。

[15] [7]~[14]のいずれかの製造法で得られた光学活性3-置換グルタル酸モノアミドを、塩基性化合物、水及び有機溶媒と混合して溶液とし、次いで酸を混合することで、前記光学活性3-置換グルタル酸モノアミドを析出させることを特徴とする光学活性3-置換グルタル酸モノアミドの製造法。
また本発明によれば、3-置換グルタル酸無水物(3)を不斉触媒存在下でアルコールと作用させるか、3-置換グルタル酸ジエステル(7)に所定の酵素を作用させることで3-置換グルタル酸モノエステル(4)を製造し、これをアミノリシスしているため、高い光学純度を有する3-置換グルタル酸モノアミド(2)を高収率で製造することができる。
1.第1のルート
1-1.不斉加水分解
第1のルートでは、下記式(1):
で表される3-置換グルタル酸ジアミド(1)の不斉加水分解活性を有する酵素(以下、アミダーゼ系酵素という)を作用させることにより、下記式(2):
で表される光学活性3-置換グルタル酸モノアミド(2)を製造する。
本発明者らは、光学活性モノアミド(2)の光学純度を簡便に向上し得る方法を開発した。この方法は、種々の方法で得られる光学活性モノアミド(2)に対して、その光学純度を向上するのに使用できるが、上記第1のルートで得られる光学活性モノアミド(2)の光学純度改善に適用することができる。本発明者らが開発した方法で光学純度を向上させる場合、光学活性モノアミド(2)、塩基性化合物、水、および有機溶媒の混合液と、酸とを混合して、前記光学活性モノアミド(2)を固体として析出させればよい。
例えば、水は、モノアミド(2)1質量部に対して、0.5質量部以上、好ましくは1質量部以上、さらに好ましくは3質量部以上であって、50質量部以下、好ましくは30質量部以下、さらに好ましくは10質量部以下程度であってもよい。
(a)前記モノアミド(2)、塩基性化合物、水、および有機溶媒の混合液と酸とを混合した後、更に貧溶媒を添加することにより固体を析出させる方法。
(b)前記モノアミド(2)、塩基性化合物、水、および有機溶媒の混合液と酸とを混合した後、濃縮する事により固体を析出させる方法。
(c)前記モノアミド(2)、塩基性化合物、水、および有機溶媒の混合液と酸とを混合した後、冷却する事により固体を析出させる方法。
(d)前記モノアミド(2)、塩基性化合物、水、および有機溶媒の混合液と酸とを混合した後、貧溶媒に濃縮置換する事により固体を析出させる方法。
(e)上記(a)~(d)で例示の貧溶媒添加、濃縮、冷却、貧溶媒置換を適宜組み合わせる方法。
(f)前記モノアミド(2)と有機溶媒を混合後、更に水を添加することにより固体を析出させる方法。
(g)前記モノアミド(2)と有機溶媒と水を混合後、更に貧溶媒を添加することにより固体を析出させる方法。
(h)前記モノアミド(2)と有機溶媒と水を混合後、濃縮する事により固体を析出させる方法。
(i)前記モノアミド(2)と有機溶媒と水を混合後、冷却する事により固体を析出させる方法。
(j)前記モノアミド(2)と有機溶媒と水を混合後、貧溶媒に濃縮置換する事により固体を析出させる方法。

で表される3-置換グルタル酸をジエステル化することで製造できる。
(式中、R1及び*は前記と同じ)
で表されるアミノカルボン酸化合物を製造できる。前記モノアミド(2)が、3-(4-クロロフェニル)グルタル酸モノアミドの場合、得られるアミノカルボン酸は4-アミノ-3-(4-クロロフェニル)ブタン酸(即ちバクロフェン)であって、中枢性筋弛緩作用,鎮痛作用などを有する医薬として有用である。
第2のルートでは、不斉有機触媒存在下、下記式(3):
で表される3-置換グルタル酸無水物(3)にアルコールを作用(特に付加)させることにより、下記式(4):
で表される光学活性3-置換グルタン酸モノエステル(4)を製造し、これを更にアミノリシスすることで、前記モノアミド(2)を製造する。
なお酸無水物(3)とアルコールとを作用させてモノエステル(4)を製造する場合、R2は、このアルコールからOHを除いた残基に相当する。
で表される化合物などが挙げられる。

第3のルートでは、下記式(7):
(式中、R1及びR2は、前記に同じ)
で表される3-置換グルタル酸ジエステルに不斉加水分解活性を有する酵素(以下、エステラーゼ系酵素という)を作用させることにより、前記モノエステル(4)を製造し、更にアミノリシスすることよって、前記モノアミド(2)を製造する。なお式(7)の化合物(ジエステル(7))は、一般に、光学純度が低く、通常はラセミ体である。
前記エステラーゼ(好ましくはリパーゼ)は、固定化されているのが好ましい。
3-(4-クロロフェニル)グルタル酸(ラセミ体、10.0g、41.2mmol)をメタノール(60ml)に溶解させ、塩化チオニル(12.3g、103mmol)を滴下した。滴下終了後、2時間攪拌した後、減圧下にて溶媒を留去することにより、3-(4-クロロフェニル)グルタル酸ジメチルの白色固体(11.1g、41.2mmol)を得た。
3-n-プロピルグルタル酸(ラセミ体、8.0g、45.9mmol)をメタノール(50ml)に溶解させ、塩化チオニル(13.7g、115mmol)を滴下した。滴下終了後、2時間攪拌した後、減圧下にて溶媒を留去することにより、3-n-プロピルグルタル酸ジメチルの油状物(9.3g、45.9mmol)を得た。
表1に示した各微生物を、滅菌した8mlの培地(グリセロール1.0%、ペプトン0.5%、モルトエキス0.3%、イーストエキス0.3%、ε-カプロラクタム0.1%、pH7.0)に植菌して、試験管内で30℃で72時間、振とう培養した。培養終了後、遠心分離により菌体を集め、2mlの100mMリン酸緩衝液(pH7.0)に懸濁した。
変換率(%)=生成物量/(原料基質量+生成物量)×100
光学純度(%ee)=(A-B)/(A+B)×100 (A及びBは対応する鏡像異性体量を表し、A>Bである)

30%水酸化ナトリウム水溶液を加えることにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミド0.49g(光学純度:90.5%ee。参考例4で得られたモノアミドと、ラセミ体のモノアミドとを混合して調製。)を含むpH13.4の水溶液5.0gとし、これにメタノール5.0gを添加した。その後、これに濃塩酸をpH2.3になるまでゆっくり添加した。添加するにつれて3-(4-クロロフェニル)グルタル酸モノアミドの固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、3-(4-クロロフェニル)グルタル酸モノアミドの白色固体0.25gを得た(収率:51%、光学純度:99.6%ee)。
30%水酸化ナトリウム水溶液を加えることにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミド0.49g(光学純度:90.5%ee)を含むpH13.4の水溶液5.0gとし、これにイソプロパノール2.0gを添加した。その後、これに濃塩酸をpH1.8となるまでゆっくり添加した。添加するにつれて(R)-3-(4-クロロフェニル)グルタル酸モノアミドの固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、3-(4-クロロフェニル)グルタル酸モノアミドの白色固体0.35gを得た(収率:71%、光学純度:99.5%ee)。
30%水酸化ナトリウム水溶液を加えることにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミド0.49g(光学純度:90.5%ee)を含むpH13.4の水溶液5.0gとし、これにイソプロパノール5.0gを添加した。その後、これに濃塩酸をpH2.5となるまでゆっくり添加した。その後、水20.0gをゆっくり添加した。水の添加と共に(R)-3-(4-クロロフェニル)グルタル酸モノアミドの固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミドの白色固体0.35gを得た(収率:71%、光学純度:99.8%ee)。
30%水酸化ナトリウム水溶液を加えることにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミド0.46g(光学純度:90.4%ee)を含むpH14.2の水溶液5.0gとし、これにテトラヒドロフラン1.5gを添加した。その後、これに濃塩酸をpH1.6となるまでゆっくり添加した。添加するにつれて(R)-3-(4-クロロフェニル)グルタル酸モノアミドの固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミドの白色固体0.26gを得た(収率:57%、光学純度:99.5%ee)。
30%水酸化ナトリウム水溶液を加えることにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミド0.46g(光学純度:90.4%ee)を含むpH14.2の水溶液5.0gとし、これにアセトニトリル1.5gを添加した。その後、これに濃塩酸をpH1.5となるまでゆっくり添加した。添加するにつれて(R)-3-(4-クロロフェニル)グルタル酸モノアミドの固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミドの白色固体0.38gを得た(収率:83%、光学純度:98.2%ee)。
30%水酸化ナトリウム水溶液を加えることにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミド0.46g(光学純度:90.4%ee)を含むpH14.2の水溶液5.0gとし、これにアセトン1.5gを添加した。その後、これに濃塩酸をpH1.3となるまでゆっくり添加した。添加するにつれて(R)-3-(4-クロロフェニル)グルタル酸モノアミドの固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミドの白色固体0.38gを得た(収率:83%、光学純度:97.6%ee)。
30%水酸化ナトリウム水溶液を加えることにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミド0.46g(光学純度:90.4%ee)を含むpH14.2の水溶液5.0gとし、これに濃塩酸をpH1.4となるまでゆっくり添加した。添加するにつれて(R)-3-(4-クロロフェニル)グルタル酸モノアミドの固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミドの白色固体0.41gを得た(収率:89%、光学純度:91.0%ee)。
3-(4-クロロフェニル)グルタル酸無水物(151mg、0.668mmol)をテトラヒドロフラン(2ml)に溶解させ、前記式(5)で表される触媒(40.0mg、0.067mmol)を加えた後、-20℃に冷却した。更に、メタノール(214mg、6.68mmol)を添加して、22時間攪拌した。室温まで昇温し、濃塩酸、水(5ml)及びトルエン(10ml)を加えた後、水層を除去した。得られた有機層に28%アンモニア水(1.13g、18.6mmol)を加え、48時間攪拌した。有機層を除去した後、濃塩酸を添加することで、固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミドの白色固体112.5mgを得た(収率:70%、光学純度:92.0%ee)。
3-(4-クロロフェニル)グルタル酸ジメチルエステル(15.0g、0.0554mol)に、水(150ml)と固定化酵素であるNovozym435(1.5g)を加えて50℃に昇温した。30%水酸化ナトリウム水溶液を添加して、pH=7とした後、pH=7に維持しながら24時間攪拌した。濾過により固定化酵素を除去した後、濃塩酸にてpH=2としてトルエンにて抽出した。得られた有機層に28%アンモニア水(67.3g、1.11mol)を加え、48時間攪拌した。有機層を除去した後、濃塩酸を添加することで、固体が析出した。1時間熟成後、析出した固体を濾別して、減圧下にて乾燥することにより、(R)-3-(4-クロロフェニル)グルタル酸モノアミドの白色固体9.73gを得た(収率:73%、光学純度:96.3%ee)。
Claims (15)
- R1がn-プロピル基、又は4-クロロフェニル基である、請求項1に記載の製造法。
- 前記有機溶媒が、メタノール、エタノール、イソプロパノール、テトラヒドロフラン、アセトニトリル、およびアセトンからなる群より選択される少なくとも1つである、請求項1または2に記載の製造法。
- 有機溶媒の量が、光学活性3-置換グルタル酸モノアミド1質量部に対して、0.5~50質量部である請求項1~3のいずれかに記載の製造法。
- 前記塩基性化合物が、アルカリ金属の塩、アンモニア、及びトリエチルアミンから選択される少なくとも1つである請求項1~4のいずれかに記載の製造法。
- 前記混合液のpHが6以上である請求項1~5のいずれかに記載の製造法。
- 不斉有機触媒存在下、下記式(3):
(式中、R1は炭素数1~8のアルキル基、炭素数2~8のアルケニル基、炭素数2~8のアルキニル基、炭素数4~20のアリール基、または炭素数5~20のアラルキル基を示し、これらアルキル基、アルケニル基、アルキニル基、アリール基、またはアラルキル基は、置換基を有していてもよい。)
