本発明における化合物の塩としては、製薬上許容される塩が好ましい。製薬上許容される塩としては、例えば、無機酸との塩、有機酸との塩、無機塩基との塩、有機塩基との塩、アミノ酸との塩等が挙げられる。
有機酸との塩として、例えば、シュウ酸、マレイン酸、クエン酸、フマル酸、乳酸、リンゴ酸、コハク酸、酒石酸、酢酸、トリフルオロ酢酸、グルコン酸、アスコルビン酸、メタンスルホン酸、ベンゼンスルホン酸、p-トルエンスルホン酸等との塩が挙げられる。
有機塩基との塩として、例えば、メチルアミン、ジエチルアミン、トリメチルアミン、トリエチルアミン、エタノールアミン、ジエタノールアミン、トリエタノールアミン、エチレンジアミン、トリス(ヒドロキシメチル)メチルアミン、ジシクロヘキシルアミン、N,N’-ジベンジルエチレンジアミン、グアニジン、ピリジン、ピコリン、コリン、シンコニン、メグルミン等との塩が挙げられる。

反応の順序は適宜変更し得る。合理的と思われる工程又は部位から反応を行えばよい。
また、各工程間に適宜置換基変換(置換基の変換又は更なる修飾)工程が挿入されていてもよい。反応性官能基がある場合は、適宜保護、脱保護を行えばよい。また、反応の進行を促進するために、例示した試薬以外の試薬を適宜用いることができる。各工程で得られる化合物は、蒸留、再結晶、カラムクロマトグラフィー等の慣用される常法で精製することができるが、場合によっては、単離精製せず次の工程に進むことができる。
なお、1重量に対して1容量とは、例えば、1kgに対して1Lの比率を意味する。
化合物IIの製造
(1-i)(R)-1-フェニルエタノール(以下、化合物VIと略す場合がある。)から化合物VIのジアニオンを調製した後、当該ジアニオンをホウ酸エステルを用いてボリル化する工程、および(1-ii)当該ボリル化物をtert-ブチル 4-ブロモ-2-メチルベンゾエート(以下、化合物VIIと略す場合がある。)と反応させる工程
により、化合物IIを製造する工程である。
化合物VIからそのジアニオンを調製した後、当該ジアニオンを、ホウ酸エステルを用いてボリル化する。
該ジアニオンの調製は、好ましくは、溶媒中、塩基存在下、及び添加剤存在下又は非存在下で行われる。
ジアニオンの調製に用いる溶媒としては、例えば、ジエチルエーテル、tert-ブチルメチルエーテル、ジ-n-ブチルエーテル、シクロペンチルメチルエーテル等のエーテル系溶媒;n-ヘキサン、トルエン等の炭化水素系溶媒等が挙げられ、これらは単独又は2種以上を混合して使用することができる。本反応における好ましい溶媒はn-ヘキサンである。溶媒の使用量は限定されないが、通常、化合物VIの1重量に対して約1容量乃至約100容量、好ましくは約5容量乃至約20容量である。
該ジアニオンの調製に用いる塩基としては、例えば、n-ブチルリチウム、s-ブチルリチウム等のアルキルリチウム;リチウムジイソプロピルアミド、ナトリウムアミド、リチウムビストリメチルシリルアミド等のアルカリ金属アミド等が挙げられ、好ましくはn-ブチルリチウムである。塩基の使用量は、化合物VIの1モルに対し、通常、約2モル乃至約5モルであり、好ましくは約2モル乃至約3モルである。
該ジアニオンの調製に用いる添加剤としては、例えば、1,4-ジアザビシクロ[2.2.2]オクタン(以下、DABCOと略す場合がある。)、N,N,N’,N’-テトラメチルエチレンジアミン(以下、TMEDAと略す場合がある。)、ヘキサメチルホスホリックトリアミド等の3級の有機アミン類が挙げられ、好ましくはTMEDAである。添加剤の使用量は、化合物VIの1モルに対し、通常、0モル乃至約4モルであり、好ましくは約0.5モル乃至約2モルである。
該ジアニオンの調製のための反応温度は、通常、約-50℃乃至約150℃で、好ましくは室温乃至約80℃である。
該ジアニオンの調製のための反応時間は、通常、約30分乃至約4日間で、好ましくは約1時間乃至約24時間である。
このように調製されたジアニオンは、通常、反応終了後の反応液のままか、または濃縮後に次のボリル化に供される。好ましくは、反応終了後の反応液のままで次のボリル化に供される。
該ボリル化に用いる溶媒としては、例えば、ジエチルエーテル、tert-ブチルメチルエーテル、ジ-n-ブチルエーテル、シクロペンチルメチルエーテル等のエーテル系溶媒;n-ヘキサン、トルエン等の炭化水素系溶媒等が挙げられ、これらは単独又は2種以上を混合して使用することができる。本反応における好ましい溶媒はn-ヘキサンである。溶媒の使用量は限定されないが、通常、化合物VIの1重量に対して約1容量乃至約100容量、好ましくは約5容量乃至約20容量である。なお、上記工程で調製されたジアニオンを反応終了後の反応液のままで使用する場合は、新たに溶媒を加えてもよく、この場合、ジアニオンの調製に用いた溶媒と異なる溶媒でもよい。また、上記工程で調製されたジアニオンを濃縮後に使用する場合も、ジアニオンの調製に用いた溶媒と異なる溶媒を加えてもよい。
該ボリル化に用いるホウ酸エステルとしては、例えば、ホウ酸トリメチル、ホウ酸トリエチル、ホウ酸トリイソプロピル、ホウ酸トリ-n-ブチル等が挙げられ、好ましくは、ホウ酸トリイソプロピルまたはホウ酸トリ-n-ブチルである。ホウ酸エステルの使用量は、化合物VIの1モルに対し、通常、約0.5モル乃至約20モルであり、好ましくは約0.5モル乃至約5モルである。
ボリル化の反応温度は、通常、約-100℃乃至約150℃で、好ましくは約-50℃乃至約80℃である。
ボリル化の反応時間は、通常、約30分乃至約4日間で、好ましくは約1時間乃至約12時間である。
反応終了後、常法の操作にて反応液を処理し、必要により精製して、次の工程に供される。
(1-i)工程で得られたボリル化物を化合物VIIと反応させることにより、化合物IIを製造する。
該反応は、好ましくは、Suzukiカップリングに準じて行われ、溶媒中、塩基及び金属触媒存在下で行われる。
該反応に用いる溶媒としては、例えば、ジエチルエーテル、テトラヒドロフラン、1,2-ジメトキシエタン、ジグリム、アニソール等のエーテル系溶媒;ヘキサン、ベンゼン、トルエン等の炭化水素系溶媒;メタノール、エタノール、1-プロピルアルコール、tert-ブタノール等のアルコール系溶媒;酢酸エチル、酢酸tert-ブチル等のエステル系溶媒;アセトン、N,N-ジメチルホルムアミド、ジメチルスルホキシド、水等の極性溶媒等が挙げられ、これらは単独又は2種以上を混合して使用することができる。本反応における好ましい溶媒は水とテトラヒドロフランとの混合溶媒である。溶媒の使用量は限定されないが、通常、化合物VIIの1重量に対して約1容量乃至約100容量、好ましくは約5容量乃至約20容量である。
該反応に用いる塩基としては、例えば、Suzukiカップリングで使用されている公知の塩基であればよく、例えば、トリエチルアミン、ピリジン等の有機塩基;水酸化ナトリウム、水酸化カリウム等のアルカリ金属及びアルカリ土類金属の水酸化物;炭酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、炭酸水素カリウム等のアルカリ金属及びアルカリ土類金属の炭酸塩;リン酸三ナトリウム、リン酸三カリウム、リン酸二ナトリウム、リン酸二カリウム等のアルカリ金属及びアルカリ土類金属のリン酸塩;酢酸ナトリウム、酢酸カリウム等のアルカリ金属及びアルカリ土類金属のカルボン酸塩;ナトリウムメトキシド、カリウム tert-ブトキシド等のアルカリ金属及びアルカリ土類金属のアルコキシド等が挙げられ、好ましくはリン酸三カリウムである。塩基の使用量は、化合物VIIの1モルに対し、通常、約0.3乃至約20モルであり、好ましくは約1モル乃至約5モルである。
該反応に用いる金属触媒としては、例えば、Suzukiカップリングの触媒作用を有する公知の金属触媒であればよく、一般的にはパラジウム触媒が用いられる。例えば、塩化パラジウム(PdCl2)、酢酸パラジウム(Pd(OAc)2)、ビス(トリフェニルホスフィン)パラジウムジクロリド(PdCl2(PPh3)2)、ビス(ジベンジリデンアセトン)パラジウム(Pd(dba)2)、トリス(ジベンジリデン)ジパラジウム(Pd2(dba)3)、テトラキス(トリフェニルホスフィン)パラジウム(Pd(PPh3)4)、パラジウム炭素(Pd-C)などが挙げられ、これらは単独又は2種以上を混合して使用してもよく、また、反応系中で調整したものを、精製せずにそのまま用いてもよい。本反応における好ましい金属触媒はビス(トリフェニルホスフィン)パラジウムジクロリド(PdCl2(PPh3)2)である。金属触媒の使用量は、化合物VIIの1モルに対し、通常、約0.01%モル乃至約20%モルであり、好ましくは約0.05%モル乃至約1%モルである。

化合物IIIの製造
該グリシジル化合物はグリシジル基を有する化合物を意味し、例えば、3-ニトロベンゼンスルホン酸(R)-グリシジル(別名(R)-グリシジルノシレート)、4-メチルベンゼンスルホン酸(R)-グリシジル(別名(R)-グリシジルトシレート)、(S)-エピクロロヒドリン等が挙げられ、3-ニトロベンゼンスルホン酸(R)-グリシジル、(S)-エピクロロヒドリンが好ましい。グリシジル化合物又はその溶媒和物の使用量は、化合物IIの1モルに対し、通常、約1乃至約5モルであり、好ましくは約1モル乃至約2モルである。
該反応に用いる溶媒としては、例えば、ジエチルエーテル、テトラヒドロフラン、ジオキサン、1,2-ジメトキシエタン、ジグリム、アニソール等のエーテル系溶媒;ベンゼン、トルエン、ヘキサン、キシレン等の炭化水素系溶媒;酢酸エチル、酢酸tert-ブチル等のエステル系溶媒;アセトン、N,N-ジメチルホルムアミド、ジメチルスルホキシド、水等の極性溶媒等が挙げられ、これらは単独又は2種以上を混合して使用することができる。ここで、グリシジル化合物又はその溶媒和物として(S)-エピクロロヒドリンを用いる場合は、水と上記有機溶媒との混合溶媒が好ましく、水とトルエンとの混合溶媒がさらに好ましい。3-ニトロベンゼンスルホン酸(R)-グリシジルを用いる場合は、上記有機溶媒が好ましく、1,2-ジメトキシエタン又はジグリムがさらに好ましい。溶媒の使用量は限定されないが、通常、化合物IIの1重量に対して約1容量乃至約50容量を、好ましくは約2容量乃至約10容量である。
該反応に用いる塩基としては、例えば、トリエチルアミン、ピリジン等の有機塩基;水酸化ナトリウム、水酸化カリウム等のアルカリ金属及びアルカリ土類金属の水酸化物;炭酸ナトリウム、炭酸カリウム、炭酸水素ナトリウム、炭酸水素カリウム等のアルカリ金属及びアルカリ土類金属の炭酸塩;水素化ナトリウム、水素化カリウム等のアルカリ金属及びアルカリ土類金属の水素化物;n-ブチルリチウム、s-ブチルリチウム等のアルキルリチウム等が挙げられ、好ましくは水素化ナトリウムまたは水酸化ナトリウムである。塩基の使用量は、化合物IIの1モルに対し、通常、約1乃至約20モルであり、好ましくは約1モル乃至約5モルである。
該反応の反応温度は、通常、約0℃乃至約150℃で、好ましくは約15℃乃至約100℃である。
該反応の反応時間は、通常、約1時間乃至約4日間で、好ましくは約5時間乃至約2日間である。
反応終了後、常法の操作にて反応液を処理し、必要により精製して、化合物IIIが得られる。
化合物Vの製造
該反応で溶媒を用いる場合には、例えば、ジエチルエーテル、テトラヒドロフラン、1,2-ジメトキシエタン、シクロペンチルメチルエーテル等のエーテル系溶媒;ヘキサン、ベンゼン、トルエン等の炭化水素系溶媒;塩化メチレン、クロロホルム等のハロゲン化炭化水素系溶媒;メタノール、エタノール、1-プロピルアルコール、tert-ブタノール等のアルコール系溶媒;酢酸エチル、酢酸tert-ブチル等のエステル系溶媒;アセトン、N,N-ジメチルホルムアミド、ジメチルスルホキシド、水等の極性溶媒等が挙げられ、これらは単独又は2種以上を混合して使用することができる。本反応における好ましい溶媒はメタノールまたはトルエンである。溶媒の使用量は限定されないが、通常、化合物IIIの1重量に対して0容量乃至約100容量、好ましくは約2容量乃至約10容量である。
化合物IVの使用量は、化合物IIIの1モルに対し、通常、約1乃至約5モルであり、好ましくは約1モル乃至約2モルである。
該反応の反応温度は、通常、約15℃乃至約200℃で、好ましくは約50℃乃至約150℃である。
該反応の反応時間は、通常、約2時間乃至約4日間で、好ましくは約5時間乃至約24時間である。
反応終了後、常法の操作により反応液を処理し、必要により精製して、化合物Vが得られる。あるいは反応液をそのまま次の工程に供することもできる。
化合物Iの製造
該アルカリ加水分解は、好ましくは、水又は水と有機溶媒の混合溶媒中、塩基存在下で行われる。
該アルカリ加水分解に用いる溶媒としては、アルカリ加水分解で一般的に用いる溶媒であればよく、通常、水、又は水と有機溶媒との混合溶媒が用いられる。有機溶媒としては、例えば、ジエチルエーテル、テトラヒドロフラン、1,2-ジメトキシエタン、シクロペンチルメチルエーテル等のエーテル系溶媒;ヘキサン、ベンゼン、トルエン等の炭化水素系溶媒;メタノール、エタノール、1-プロピルアルコール等のアルコール系溶媒;N,N-ジメチルホルムアミド、ジメチルスルホキシド等の極性溶媒等が挙げられ、これらは単独又は2種以上を、水と混合して加水分解反応に使用することができる。本反応における好ましい有機溶媒はアルコール系溶媒であり、特にメタノールが好ましい。有機溶媒の使用量は、通常、化合物Vの1重量に対して約1容量乃至約100容量、好ましくは約2容量乃至約10容量であり、水の使用量は、用いる有機溶媒の1容量に対して約0.05容量乃至約2容量である。
該アルカリ加水分解反応に必要な塩基としては、例えば、水酸化リチウム、水酸化ナトリウム、水酸化カリウム等の水酸化アルカリ金属が挙げられ、好ましくは水酸化ナトリウムまたは水酸化カリウムである。塩基の使用量は、化合物Vの1モルに対し、通常、約1モル乃至約20モルであり、好ましくは約1モル乃至約5モルである。
アルカリ加水分解の反応温度は、通常、室温乃至約200℃で、好ましくは約50℃乃至約100℃である。
アルカリ加水分解の反応時間は、通常、約3時間乃至約7日間で、好ましくは約10時間乃至約3日間である。
(ii)エステル交換した後アルカリ加水分解する方法
該エステル交換は、好ましくは、アルカリ金属アルコキシドを用いて、有機溶媒中で行われる。
該エステル交換に用いるアルカリ金属アルコキシドとしては、例えば、ナトリウムメトキシド、カリウムイソプロポキシド等の一級アルコール又は二級アルコールから誘導されるアルカリ金属アルコキシドが挙げられる。好ましくは、一級アルコールから誘導されるアルカリ金属アルコキシドであり、特にナトリウムメトキシドが好ましい。アルカリ金属アルコキシドの使用量は、化合物Vの1モルに対し、通常、約0.1モル乃至約20モルであり、好ましくは約1モル乃至約5モルである。該アルカリ金属アルコキシドは、反応系中で調製したものを用いてもよい。
該エステル交換に用いる有機溶媒としては、例えば、ジエチルエーテル、テトラヒドロフラン、1,2-ジメトキシエタン、シクロペンチルメチルエーテル等のエーテル系溶媒;ヘキサン、ベンゼン、トルエン等の炭化水素系溶媒;メタノール、エタノール、1-プロピルアルコール等のアルコール系溶媒;N,N-ジメチルホルムアミド、ジメチルスルホキシド等の極性溶媒等が挙げられ、これらは単独又は2種以上を混合して使用することができる。本反応における溶媒及びアルカリ金属アルコキシドとの組み合わせとして、好ましくは、メタノール及びナトリウムメトキシドの組み合わせ、並びにエタノール及びナトリウムエトキシドの組み合わせである。本反応における好ましい溶媒はメタノールである。有機溶媒の使用量は、通常、化合物Vの1重量に対して約1容量乃至約100容量、好ましくは約2容量乃至約10容量である。
エステル交換の反応温度は、通常、室温乃至約200℃で、好ましくは約50℃乃至約100℃である。
エステル交換の反応時間は、通常、約2時間乃至約4日間で、好ましくは約5時間乃至約2日間である。
アルカリ加水分解の反応温度は、通常、室温乃至約200℃で、好ましくは約50℃乃至約100℃である。
アルカリ加水分解の反応時間は、通常、約10分乃至約4日間で、好ましくは約1時間乃至約24時間である。
当該工程の好適な態様は、(ii)エステル交換した後アルカリ加水分解する方法である。
反応終了後、常法の操作により反応液を処理し、必要により精製して、化合物Iが得られる。また、必要に応じて、化合物Iの種晶を用いて晶析してもよい。

2’-((1R)-1-{(2R)-3-[1-(4-クロロ-3-フルオロフェニル)-2-メチルプロパン-2-イルアミノ]-2-ヒドロキシプロポキシ}エチル)-3-メチルビフェニル-4-カルボン酸 1/2硫酸塩 1/2水和物(化合物I-aと略す場合がある。)の製造
化合物IIの製造
窒素雰囲気下、得られた濃縮液(959g)に、THF(4.8L)、水(1.8L)、化合物VII(1.00kg)を添加し、室温で10分間激しく攪拌した。次いで、ビス(トリフェニルホスフィン)パラジウムジクロリド(13.0g)を添加し、リン酸三カリウム(1.18kg)の水(3.0L)溶液を、65℃で1時間かけて滴下した。同温にてさらに1.5時間攪拌し、室温まで冷却後、分液した。トルエン(5.0L)を添加し、ジエチレントリアミン(381g)と水(5.0L)との混合液で2回洗浄し、酢酸(0.25L)と水(5.0L)との混合液、水(5.0L)で順次洗浄した。得られた有機層を減圧濃縮し、さらに、メタノール(5.0L)を加えて減圧濃縮する操作を2回行った。濃縮液に、メタノールを加えて全体量を5.0Lとし、Si-Thiol(15.3g)を加え、室温で終夜攪拌した。この溶液を濾過し、濾取物をメタノール(3.4L)で洗浄した。濾液と洗液を合わせた溶液に、水(1.8L)を室温で滴下し、種晶(0.15g)を添加した後に、同温で70分間攪拌した。さらに、水(3.0L)を、同じく室温で滴下し、同温で終夜攪拌した。析出した結晶を濾取し、メタノール(2.2L)と水(1.4L)との混合液で洗浄し、減圧乾燥して表題化合物(結晶、1.10kg,化合物VIIからの収率95.5%)を得た。

ここで種晶として使用した化合物IIの結晶は、例えば、実施例1の方法により得た結晶であってもよく、上記工程1により別途得られた結晶であってもよい。
化合物IIを結晶として得ることができるため、品質のコントロール容易性の観点から改善された製造方法を提供することができる。
化合物IIIの製造
同様にして得られた粗生成物を、シリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=5:1)にて精製し、表題化合物を得た。

化合物IIを用いることにより、グリシジル化の反応進行度の観点で改善された製造方法を提供することができる。
化合物Vの製造
同様にして得られた粗生成物を、シリカゲルカラムクロマトグラフィー(n-ヘキサン:酢酸エチル=2:1)にて精製し、表題化合物を得た。

化合物Iの製造

ここで、種晶として使用した化合物Iの結晶は、例えば、下記方法により得た結晶であっても、上記工程4により別途得られた結晶であってもよい。
国際公開公第WO2004/094362号に記載の化合物I(500mg)に、メタノール(5 ml)を加えて、2時間加熱還流した後、反応液を室温に戻し、析出した結晶を濾過して表記化合物(400 mg)を得た。
