上記ハロゲン原子としては、例えばフッ素原子、塩素原子、臭素原子が挙げられる。
X1、X2、X3およびX4としては、それぞれ独立に、ハロゲン原子が好ましく、フッ素原子および塩素原子がより好ましく、フッ素原子が更に好ましい。X1、X2、X3およびX4は、全てハロゲン原子であることが好ましい。
化合物(1)の例として、2−フルオロテレフタル酸ジアミド、2−クロロテレフタル酸ジアミド、2,5−ジフルオロテレフタル酸ジアミド、2,6−ジフルオロテレフタル酸ジアミド、2,3−ジフルオロテレフタル酸ジアミド、2、6−ジクロロテレフタル酸ジアミド、2,3−ジクロロテレフタル酸ジアミド、2,3,5−トリフルオロテレフタル酸ジアミド、2,3,5−トリクロロテレフタル酸ジアミド、2,3,5,6−テトラフルオロテレフタル酸ジアミド、2,3,5,6−テトラクロロテレフタル酸ジアミド、2,3,5−トリフルオロ−6−クロロテレフタル酸ジアミドが挙げられる。なかでも2,3,5,6−テトラフルオロテレフタル酸ジアミドが好ましい。
化合物(1)は、例えば、対応する酸ハライドとアンモニアガスとを反応させる(例えば、Zhurnal Obshchei Khimii,34,2953−2958(1964)参照。)等の方法に準じて製造することができる。
式(2)において、Rは、置換されていてもよいアルコキシカルボニル基、すなわちアルコキシカルボニル基または置換されたアルコキシカルボニル基を表す。
Rにより示されるアルコキシカルボニル基としては、炭素数2~20のアルコキシカルボニル基が挙げられる。かかるアルコキシカルボニル基は、直鎖状、分枝鎖状および環状の何れであってもよく、例えば、メトキシカルボニル基、エトキシカルボニル基、プロポキシカルボニル基、イソプロポキシカルボニル基、n−ブトキシカルボニル基、tert−ブトキシカルボニル基、イソブトキシカルボニル基、ペンチルオキシカルボニル基、ヘキシルオキシカルボニル基、ドデシルオキシカルボニル基、オクタデシルオキシカルボニル基、シクロヘキシルオキシカルボニル基が挙げられる。
該アルコキシカルボニル基の置換基としては、例えば、フェニル基、ナフチル基等のアリール基が挙げられる。置換されたアルコキシカルボニル基としては、ベンジルオキシカルボニル基等の炭素数8~20のアリール置換アルコキシカルボニル基が挙げられる。
式(2)において、Rとしては、炭素数2~10のアルコキシカルボニル基が好ましく、炭素数2~5のアルコキシカルボニル基がより好ましく、tert−ブトキシカルボニル基が更に好ましい。
化合物(2)としては、例えばピロカルボン酸ジメチルエステル、ピロカルボン酸ジエチルエステル、ピロカルボン酸ジイソプロピルエステル、ピロカルボン酸ジ−tert−ブチルエステル、ピロカルボン酸ジベンジルエステル等が挙げられる。 化合物(2)として、例えば特開平4−211634号公報等に記載の公知の方法により製造したものや、市販のものを用いてもよい。
化合物(2)の量は、通常、化合物(1)1モルに対して4モル以上であればよい。該量の上限は、特に限定されないが、経済的には、化合物(1)1モルに対して10モル以下であることが好ましい。
化合物(1)と化合物(2)との反応は、一般に、4−ジアルキルアミノピリジンの存在下で行われる。
4−ジアルキルアミノピリジンは、ジアルキルアミノ基が炭素数1~5のアルキルを有することが好ましく、炭素数1~3のアルキルを有することがより好ましい。
4−ジアルキルアミノピリジンとしては、4−ジメチルアミノピリジン、4−ジエチルアミノピリジン、4−ジ(n−プロピル)アミノピリジン、4−ジ(n−ブチル)アミノピリジン等が挙げられ、入手の容易さから、4−ジメチルアミノピリジンが好ましい。
4−ジアルキルアミノピリジンとして、市販のものを用いることができる。
4−ジアルキルアミノピリジンは、通常、化合物(1)1モルに対して0.001モル以上、好ましくは0.001~0.1モル、より好ましくは0.005~0.1モルの範囲で用いられる。
化合物(1)と化合物(2)との反応は、通常、溶媒の存在下に実施される。溶媒としては、例えば、メチルtert−ブチルエーテル、テトラヒドロフラン、ジグライム等のエーテル溶媒;アセトニトリル、プロピオニトリル等のニトリル溶媒;トルエン、キシレン等の芳香族炭化水素溶媒;が挙げられる。該反応において、該溶媒は1種類のみ用いられてもよいし、2種以上用いられてもよい。
該溶媒としては、ニトリル溶媒が好ましく、アルキルニトリル溶媒がより好ましく、炭素数1~5のアルキルニトリル溶媒が更に好ましい。該溶媒を2種以上用いる場合、ニトリル溶媒と芳香族炭化水素溶媒との混合物を用いることが好ましい。
溶媒を使用する場合、その量は制限されないが、実用的には、化合物(1)1重量部に対して、通常、1重量部以上、100重量部以下である。
反応温度は、通常−20~150℃、好ましくは30~100℃の範囲である。また、常圧条件下で反応を実施してもよいし、加圧条件下で反応を実施してもよい。
反応の進行は、例えばガスクロマトグラフィー、高速液体クロマトグラフィー、薄層クロマトグラフィー、NMR、IR等の通常の分析手段により確認することができる。
化合物(1)と化合物(2)との反応は、これらの化合物を混合し、必要に応じて加熱することにより実施される。化合物(1)および化合物(2)の混合順序は特に制限されない。溶媒や4−ジアルキルアミノピリジンの存在下で行う場合であっても、化合物(1)、化合物(2)、溶媒や4−ジアルキルアミノピリジンの混合順序は特に制限されない。
化合物(1)と化合物(2)との反応について、好ましい実施態様としては、溶媒と4−ジメチルアミノピリジンと化合物(1)の混合物に化合物(2)を反応温度下で加えていく態様が挙げられる。
化合物(1)と化合物(2)との反応により、式(3)
で示されるジカルボキサミド化合物(以下、この化合物を化合物(3)と称することがある。)を含む反応混合物が得られる。
式(3)において、R、X1、X2、X3およびX4は、それぞれ上記定義と同じである。
式(3)において、X1、X2、X3およびX4の好ましい範囲は、式(1)と同様である。
式(3)において、Rの好ましい範囲は、式(2)と同様である。

化合物(1)と化合物(2)との反応により得られる反応混合物は、そのまま後述する第2工程に供してもよいし、晶析、濃縮、分液等を行うことにより該反応混合物から化合物(3)を単離して後述する第2工程に供してもよい。単離された化合物(3)は、例えば晶析、カラムクロマトグラフィー等の一般的な方法により精製した後に第2工程に供してもよい。
化合物(3)は、好ましくは、有機溶媒と酸水溶液とを用いた分液により上記反応混合物から単離される。より好ましくは、有機溶媒と酸水溶液とを該反応混合物に加え、これらを攪拌し更に静置し、得られる有機溶媒層から化合物(3)を回収することができる。該反応混合物が有機溶媒を含む場合、酸水溶液のみを該反応混合物に加えることにより分液することができる。該有機溶媒は、水と混和しない溶媒であればよい。該有機溶媒としては、トルエン等の芳香族炭化水素化合物;酢酸エチル等の脂肪族炭化水素化合物;アセトニトリル等のニトリル化合物が挙げられる。該酸水溶液としては、塩酸、硫酸等の無機酸の水溶液が好ましく、塩酸がより好ましい。
本発明のハロゲン置換ベンゼンジメタノールの製造方法において、第2工程は化合物(3)を水素化ホウ素化合物で還元して、式(4)
(式中、R、X1、X2、X3およびX4は、それぞれ上記と同じ意味を表す。)で示されるハロゲン置換ベンセンジメタノール(以下、この化合物を化合物(4)と称することがある。)を得る工程である。
水素化ホウ素化合物としては、水素化ホウ素アルカリ金属塩や水素化ホウ素アルカリ土類金属塩等が挙げられる。上記水素化ホウ素アルカリ金属塩としては、例えば、水素化ホウ素ナトリウム、水素化ホウ素リチウム、水素化ホウ素カリウムが挙げられる。上記水素化ホウ素アルカリ土類金属塩としては、水素化ホウ素カルシウム、水素化ホウ素マグネシウム等が挙げられる。
水素化ホウ素化合物としては、入手しやすので、水素化ホウ素アルカリ金属塩が好ましく、水素化ホウ素ナトリウムがより好ましい。
水素化ホウ素化合物は、市販のものを用いてもよいし、調製して用いてもよい。例えば、水素化ホウ素ナトリウムは、ホウ酸エステルと水素化ナトリウムとから容易に調製可能である。また、他の水素化ホウ素化合物は、水素化ホウ素ナトリウムと対応する金属ハロゲン化物との反応により調製可能であり、例えば水素化ホウ素カルシウムは、水素化ホウ素ナトリウムと塩化カルシウムとの反応により得られる。水素化ホウ素化合物を調製して用いる場合には、予め調製した水素化ホウ素化合物を用いてよいし、化合物(3)の還元反応と同時に該化合物(3)の存在下で調製されてもよい。
水素化ホウ素化合物は、化合物(3)1モルに対して、通常1モル以上の量で用いればよい。該水素化ホウ素化合物の量は、経済性の点において、化合物(3)1モルに対して、通常5モル以下、好ましくは1~2.5モルの範囲である。
化合物(3)の還元は、好ましくはモノアルコールの存在下で実施される。化合物(3)の還元をモノアルコールの存在下で実施した場合、化合物(3)が化合物(4)に効率よく変換される。上記モノアルコールは溶媒として用いることができる。
上記モノアルコールとしては、メタノール、エタノール、n−プロパノール、イソプロパノール、n−ブタノール、tert−ブタノール等の炭素数1~4の脂肪族モノアルコールが挙げられ、メタノールが好ましい。
該モノアルコールの量は、制限されないが、化合物(3)1モルに対して、通常1~100モルの範囲である。
化合物(3)の還元は、通常、溶媒の存在下で実施される。かかる溶媒としては、例えば、ジエチルエーテル、メチルtert−ブチルエーテル、テトラヒドロフラン、ジオキサン、ジイソプロピルエーテル等のエーテル溶媒や、トルエン、キシレン、クロロベンゼン等の芳香族炭化水素溶媒が挙げられる。上記溶媒は、好ましくはエーテル溶媒である。
溶媒の量は、制限されないが、化合物(3)1質量部に対して、通常1質量部以上、100重量部以下である。反応温度は、通常0~100℃、好ましくは20~70℃の範囲である。
化合物(3)の還元は、通常、溶媒の存在下、化合物(3)と水素化ホウ素化合物とを、反応温度条件下で混合することにより実施され、それらの混合順序は特に限定されない。好ましい実施態様としては、化合物(3)と水素化ホウ素化合物とを含む混合物にモノアルコールを該混合物に徐々に加えていく態様が挙げられる。モノアルコールを該混合物に徐々に加えていくことにより、化合物(3)の水素化ホウ素化合物による還元反応を促進することができる。
本反応は、通常、常圧条件下で実施されるが、加圧条件下に実施してもよい。反応の進行は、例えばガスクロマトグラフィーや液体クロマトグラフィー等の通常の分析手段により確認することができる。
反応終了後、得られる反応混合物には化合物(4)が含まれており、これに、例えば塩酸、硫酸、リン酸、硝酸等の鉱酸の水溶液を加えた後、必要に応じて、中和、抽出、濃縮等の公知の手段により、化合物(4)を単離することができる。得られた化合物(4)は、例えば晶析、カラムクロマトグラフィー等の一般的な方法により精製されてもよい。
上記化合物(4)は、式(4)で示される化合物である。式(4)において、X1、X2、X3およびX4の好ましい範囲は、式(1)と同様である。
上記化合物(4)としては、2−フルオロ−1,4−ベンゼンジメタノール、2−クロロ−1,4−ベンゼンジメタノール、2,5−ジフルオロ−1,4−ベンゼンジメタノール、2,6−ジフルオロ−1,4−ベンゼンジメタノール、2,3−ジフルオロ−1,4−ベンゼンジメタノール、2,5−ジクロロ−1,4−ベンゼンジメタノール、2,6−ジクロロ−1,4−ベンゼンジメタノール、2,3−ジクロロ−1,4−ベンゼンジメタノール、2,3,5−トリフルオロ−1,4−ベンゼンジメタノール、2,3,5−トリクロロ−1,4−ベンゼンジメタノール、2,3,5,6−テトラフルオロベンゼンジメタノール、2,3,5,6−テトラクロロベンゼンジメタノール、2,3,5−トリフルオロ−6−クロロベンゼンジメタノール等が挙げられる。なかでも、2,3,5,6−テトラハロベンゼンジメタノールが好ましく、2,3,5,6−テトラフルオロベンゼンジメタノール及び2,3,5,6−テトラクロロベンゼンジメタノールがより好ましく、2,3,5,6−テトラフルオロベンゼンジメタノールが更に好ましい。
化合物(1)と化合物(2)を反応させて化合物(3)を得る該化合物(3)の製造方法もまた、本願発明の1つである。
化合物(3)は、ハロゲン置換ベンゼンジメタノールの製造方法における中間体として有用である。化合物(3)は、容易にハロゲン置換ベンゼンジメタノールに変換することができる。化合物(3)の製造方法は、ハロゲン置換ベンゼンジメタノールの製造に好適な化合物(3)を容易に得る方法として有用である。
化合物(3)を水素化ホウ素化合物で還元して化合物(4)を得る該化合物(4)の製造方法もまた、本願発明の1つである。該化合物(4)の製造方法は、化合物(3)を用いるので、ハロゲン置換ベンゼンジメタノールを容易に得ることができる。
実施例1
(1)N,N,N’,N’−テトラキス(tert−ブトキシカルボニル)−(2,3,5,6−テトラフルオロベンゼン)−1,4−ビスカルボキサミドの調製
還流冷却管を付した100mlフラスコに、室温(約25℃)で2,3,5,6−テトラフルオロテレフタル酸ジアミド500mg、トルエン10g、アセトニトリル10g、4−ジメチルアミノピリジン10mgを仕込み、内温を80℃に調整した。内容物を同温度で攪拌しながら、そこに、ピロカルボン酸ジ(tert−ブチルエステル)3.0gを2時間かけて滴下した。滴下終了後、内容物を同温度で2時間攪拌し、その後室温まで冷却した。得られた反応混合物に5重量%塩酸水20gを加えて攪拌、静置したところ、2層に分離したので、分液して上層の有機溶媒層を得た。該有機溶媒層を水30gで2回洗浄し、得られた有機溶媒層から溶媒を留去することにより、淡黄色の油状物として、{N,N,N’,N’−テトラキス(tert−ブトキシカルボニル)−(2,3,5,6−テトラフルオロベンゼン)−1,4−ビスカルボキサミド1.3gを得た。収率は96%であった。
N,N,N’,N’−テトラキス(tert−ブトキシカルボニル)−(2,3,5,6−テトラフルオロベンゼン)−1,4−ビスカルボキサミド

(2)2,3,5,6−テトラフルオロベンゼンジメタノールの調製
還流冷却管を付した100mlフラスコに、室温で水素化ホウ素ナトリウム125mg、テトラヒドロフラン5gを仕込んだ。そこに、上記実施例1(1)で得た油状物700mgを加え、内温を50℃に調整し、内容物を同温度で2時間攪拌した。得られた反応混合物を室温まで冷却し、そこに10重量%塩酸水10gと酢酸エチル10gを加えて攪拌、静置したところ、2層に分離したので、分液して上層の有機溶媒層を得た。該有機溶媒層を液体クロマトグラフィー内部標準法により分析したところ、2,3,56−テトラフルオロベンゼンジメタノールの得量は95mgであり、収率は41%であった。
実施例2
(1)N,N,N’,N’−テトラキス(tert−ブトキシカルボニル)−(2,3,5,6−テトラフルオロベンゼン)−1,4−ビスカルボキサミドの調製
還流冷却管を付した100mlフラスコに、室温で2,3,5,6−テトラフルオロテレフタル酸ジアミド500mg、アセトニトリル10g、4−ジメチルアミノピリジン10mgを仕込み、内温を80℃に調整した。得られた混合物を同温度で攪拌しながら、そこに、ピロカルボン酸ジ(tert−ブチルエステル)3.0gを2時間かけて滴下した。滴下終了後、得られた混合物を同温度で2時間攪拌し、その後室温まで冷却した。得られた反応混合物に5重量%塩酸水20gと酢酸エチル20gを加えて攪拌、静置したところ、2層に分離したので、分液して上層の有機溶媒層を得た。該有機溶媒層を水30gで2回洗浄し、得られた有機溶媒層から溶媒を留去することにより、N,N,N’,N’−テトラキス(tert−ブトキシカルボニル)−(2,3,5,6−テトラフルオロベンゼン)−1,4−ビスカルボキサミドを主成分とする淡黄色の油状物を得た。
(2)2,3,5,6−テトラフルオロベンゼンジメタノールの調製
還流冷却管を付した100mlフラスコに、室温で水素化ホウ素ナトリウム240mg、テトラヒドロフラン10gを仕込んだ。そこに、実施例2(1)で得た油状物の全量を加え、内温を50℃に調整し、得られた混合物を同温度で2時間攪拌した。そこに、メタノール5gを1時間かけて滴下し、得られた混合物を同温度で1時間攪拌した。得られた反応混合物を室温まで冷却し、そこに10重量%塩酸水20gと酢酸エチル20gを加えて攪拌、静置したところ、2層に分離したので、分液して上層の有機溶媒層を得た。該有機溶媒層を液体クロマトグラフィー内部標準法により分析したところ、2,3,5,6−テトラフルオロベンゼンジメタノールの得量は360mgであった。実施例2−1からの通算収率は81%であった。
実施例3
還流冷却管を付した100mlフラスコに、室温で2,3,5,6−テトラフルオロテレフタル酸ジアミド500mg、アセトニトリル5g、4−ジメチルアミノピリジン10mgを仕込み、内温を60℃に調整した。得られた混合物を同温度で攪拌しながら、そこに、ピロカルボン酸ジ(tert−ブチルエステル)3.0gを1時間かけて滴下した。滴下終了後、得られた混合物を同温度で4時間攪拌した後、室温まで冷却した。冷却すると結晶が析出した。該結晶をろ過し、アセトニトリル1mlで洗浄し、その後乾燥して、白色結晶450mgを得た。この結晶は、NMRによりN,N,N’,N’−テトラキス(tert−ブトキシカルボニル)−(2,3,5,6−テトラフルオロベンゼン)−1,4−ビスカルボキサミドと同定した。

Claims (19)
- 式(1)
(式中、X1、X2、X3およびX4は、それぞれ独立に、水素原子またはハロゲン原子を表す。ただし、X1、X2、X3およびX4の少なくとも一つはハロゲン原子である。)
で示される化合物を、式(2)
(式中、Rは置換されていてもよいアルコキシカルボニル基を表す。)
で示される化合物と反応させて、式(3)
(式中、R、X1、X2、X3およびX4は、それぞれ上記と同じ意味を表す。)
で示されるジカルボキサミド化合物を得る第1工程と、
前記ジカルボキサミド化合物を水素化ホウ素化合物で還元して、式(4)
(式中、X1、X2、X3およびX4は、それぞれ上記と同じ意味を表す。)
で示されるハロゲン置換ベンゼンジメタノールを得る第2工程とを含む
該ハロゲン置換ベンゼンジメタノールの製造方法。 - X1、X2、X3およびX4は、全てハロゲン原子である請求項1記載の方法。
- Rは炭素数2~5のアルコキシカルボニル基である請求項1記載の方法。
- 式(1)で示される化合物と式(2)で示される化合物とを4−ジアルキルアミノピリジンの存在下で反応させる請求項1記載の方法。
- ジカルボキサミド化合物をモノアルコールの存在下で還元する請求項1記載の方法。
- モノアルコールは炭素数1~5の脂肪族モノアルコールである請求項5記載の方法。
- 水素化ホウ素化合物の量はジカルボキサミド化合物1モルに対して1~3モルである請求項1記載の方法。
- 水素化ホウ素化合物は水素化ホウ素アルカリ金属塩である請求項1記載の方法。
- X1、X2、X3およびX4は、全てハロゲン原子である請求項9記載の方法。
- Rは炭素数2~5のアルコキシカルボニル基である請求項9記載の方法。
- 式(1)で示される化合物と式(2)で示される化合物とを4−ジアルキルアミノピリジンの存在下で反応させる請求項9記載の方法。
- ジカルボキサミド化合物をモノアルコールの存在下で還元する請求項13記載の方法。
- X1、X2、X3およびX4は、全てハロゲン原子である請求項13記載の方法。
