そして、シリンジ6によってこの試料を採取し、試料をカラムに保持させた後、
1)試料の前処理、2)本発明の検出装置への試料の添加(コリンエステラーゼ阻害反応)、3)発色基質の添加、の工程を行うことによって、試料中のコリンエステラーゼ阻害性物質の存在の有無を目視判定することができる。
図3に示す工程に従って、有機リン系農薬の検出を行った。
なお、サンプル試料にダイアジノンを含まない場合には、青色の発色が確認されたことから、本発明の方法によって、有機リン系農薬を確実に検出できることが示された。
実施例1の酸化処理(NBS溶液の吸引処理)の効果を調べるため比較実験を行った。実施例1と同様に、カラムにサンプル中のダイアジノンを吸着させた後、酸化処理(NBS溶液の吸引処理)を行わず、カラムからダイアジノンを溶出させた。この溶出液を、検出装置中に添加し、酢酸インドキシル溶液を装置内に添加した。
その結果、ダイアジノンの検出値は、5ppm(5000ppb)に低下したことから、酸化処理の有効性が示された。
検出方法は、図3に示す工程に従うが、カーバメート系農薬が検出対象であるため、実施例1で行ったNBS溶液の吸引、排出操作は行わなかった。
5分後、発色基質となる酢酸インドキシル溶液を装置内に添加し、5分間発色反応を行った。その結果、NACによりコリンエステラーゼの酵素活性は阻害され、発色は生じず、NACは10ppbの濃度で検出が可能であった。
その他の農薬物質についても、実施例1および実施例2の方法と同様に試験したところ、以下の結果が得られた。

- 容器内底部の基底層の上にコリンエステラーゼを含有する反応層が固定されていることを特徴とするコリンエステラーゼ阻害性物質検出装置。
- 反応層には、空隙が形成されていることを特徴とする請求項1のコリンエステラーゼ阻害性物質検出装置。
- 基底層及び反応層が親水性光硬化性樹脂からなることを特徴とする請求項1または2のコリンエステラーゼ阻害性物質検出装置。
- 請求項1から3のいずれかの検出装置と、シリンジと、シリンジの先端部に取り付け可能であるとともにカラム粒子が内部に充填されているカラムチップと、コリンエステラーゼとの反応によって発色を生じる基質と
を含むことを特徴とするコリンエステラーゼ阻害性物質検出キット。 - コリンエステラーゼとの反応によって発色を生じる基質は、酢酸インドキシルである請求項4のコリンエステラーゼ阻害性物質検出キット。
- 試料中のコリンエステラーゼ阻害性物質の存在を判定する方法であって、
1)試料を請求項1から3のいずれかの検出装置内に添加し、
2)コリンエステラーゼとの反応によって発色を生じる基質を添加し、
3)発色が生じなかった場合に、試料中にコリンエステラーゼ阻害性物質が存在すると判定する
ことを特徴とするコリンエステラーゼ阻害性物質検出方法。 - 試料をカラムによって捕捉して、酸化処理を行った後に、請求項1から3のいずれかの検出装置内に添加することを特徴とする請求項6のコリンエステラーゼ阻害性物質検出方法。
- 試料中の有機リン系農薬の存在を判定する方法であって、
1)試料をカラムによって捕捉して、酸化処理を行い、
2)この試料を請求項1から3のいずれかの検出装置内に添加し、
3)コリンエステラーゼとの反応によって発色を生じる基質を添加し、
4)発色が生じなかった場合に、試料中にコリンエステラーゼ阻害性物質が存在すると判定する
ことを特徴とする有機リン系農薬の検出方法。 - コリンエステラーゼとの反応によって発色を生じる基質は、酢酸インドキシルであることを特徴とする請求項6から8のいずれかの検出方法。
